Abstract:The goal of this study was to design Cefpodoxime Proxetil SMEDDS (self-microemulsifying drug delivery system), to improve solubility and permeability which could improve therapeutic performance and drug loading capacity. Castor oil, Tween 80, PEG 400 were used as the oil, surfactant, and, co-surfactant respectively. A ternary phase diagram was used to choose the best formulations. Selected formulations were evaluated for various parameters. According to the findings, all SMEDDS formulations had nano-sized globules, good stability, and rapid dispersibility of microemulsions, which were clear and slightly bluish in colour, and no symptoms of phase separation, creaming or, cracking Intestinal permeability studies of SMEDDS formulations show that the drug diffused through a biological membrane is more when given in form of SMEDDS. The present investigation has shown that it is possible to enhance the solubility and permeability of poorly soluble drugs.
Efavirenz Self-Nano-Emulsifying Drug Delivery System: In Vitro and In Vivo Evaluation

Abstract:Self-emulsifying drug delivery system (SEDDS) is the isotropic and thermodynamically stable mixture of oil, surfactant, co‐solvent/surfactant, and drug. It emulsifies spontaneously when introduced into an aqueous phase under a mild agitation. The current study was aimed to prepare SNEDDS to augment solubility, release rate, and oral bioavailability of BCS class II drug, efavirenz (EFV). A series of oil, surfactant, and co-surfactant was screened out by a ternary phase diagram to locate a better homogenous mixture. The prepared SNEDDS was evaluated regarding its appearance, mean droplet size, phase separation, in vitro drug release, and oral bioavailability. Among the screened oil, surfactant, and co-surfactant, Labrafil M 2125 CS, Tween 80, and Transcutol®P mixture exhibited superior solubilizing capacity, respectively. Optimized SNEDDS exhibits 98.39% drug release. SNEDDS dissolution behavior was attributed to oil/surfactant ratios and properties of the surfactant phase. It also demonstrates threefold increments in the area under curve (AUC) in comparison to neat EFV. Furthermore, the optimized SNEDDS does not show any vitrification during its 3-month storage. In the present study, better performance of SNEDDS is explained by various factors like (i) improved surface area of droplets, (ii) superior solubilization potential for hydrophobic drugs due to Labrafil M 2125 CS, and (iii) result of surfactant on mucosal permeability. This study demonstrated that SNEDDS may be an alternative approach for the poorly soluble drugs to improve their solubility and oral bioavailability.

Physical Characterization and In Vitro Evaluation of Dissolution Rate from Cefpodoxime Proxetil Loaded Self Solidifying Solid SNEDDS

Abstract:Background: Cefpodoxime Proxetil (CPD) is a broad-spectrum cephalosporin indicated in respiratory and urinary tract infections. CPD is a BCS class IV drug with pH-dependent solubility and has poor bioavailability. This study investigated the challenges of developing ternary components based on solid SNEDDS of CPD for in vitro dissolution rate enhancement and self-solidifying behaviour. Methods: Tween 80, Transcutol and PEG6000 were employed as surfactants, solvents and solidifiers for a base of ternary components to develop self-solidifying solid SNEDDS, respectively. Ternary phase diagrams were used to characterize solidifying behaviour of ternary components in different proportions. S-SNEDDS formulations were drawn on the solidification areas available in the phase diagram and characterized for IR, XRD, DSC and in vitro drug release in various pH media. Results: Ternary components for the preparation of self-solidifying solid SNEDDS were selected based on drug solubility. FTIR and DSC characterization studies ruled out any drug interaction between CPD and components chosen to prepare S-SNEDDS. CPD was transformed from a crystalline into an amorphous state in ternary dispersions as revealed from XRD data. Optimized formulation (S-S 1) demonstrated more than 95% of drug release irrespective of the pH environments of the medium. Calculation of dissolution efficiency and similarity factors indicate that S SNEDDS resulted in a higher drug dissolution rate over binary dispersion (p<0.01). The stability studies showed that the S SNEDDS were stable in performances and CPD assay. Conclusion: The present investigation provides an alternative approach for enhancing the CPD dissolution rate using self-solidifying solid SNEDDS exhibited solidification behaviour at ambient temperature conditions and drug loading, which could be exploited over conventional dosage form.

Abstract:Cefpodoxime proxetil (CPD) is a broad-spectrum cephalosporin widely indicated in respiratory tract infections like pneumonia. CPD is a BCS class IV drug with the constraint of low bioavailability attributable to its poor aqueous solubility (10μg/ml) and impaired permeability. Clove oil (CO) acts as a lipophilic additive for lipid-based delivery systems designed to facilitate dissolution characteristics of poorly soluble drugs, modulating drug permeability as well as stability against oxidation. The present work aims to improve CPD solubilization by developing CO-based fully dilutable self nano emulsifying formulation (SNF), and assessment of its antibacterial and antioxidant potential. The existence of nanoemulsion domains was tracked from ternary phase diagrams plotted between CO, Tween 20/PEG400 to act as oil phase, surfactant/co-surfactant (Smix) respectively at weight ratios 1:1, 1:2, and 2:1 whereas aqueous phase acted its tertiary component. Two subsets of SNF formulations coded as (DAO1-DAO5) and (DAM1-DAM3) were prepared and assessed for antioxidant and antibacterial activities respectively, along with their naive CO-counterparts (AO1-AO5) and (AM1- AM5) respectively. Physical characterization revealed SNFs were droplet size range 35-170nm, conductivities in the range 200-500uS/cm, and RI values in 1.361-1.429. The dilution-induced microstructure transition effect was observed in SNF with the addition of an aqueous phase as its oil-continuous domain transformed into water-continuous via bi-continuous structure formation as interpreted from conductivity and refractive index data. In-vitro dissolution characterization of SNF formulation exhibits a quick CPD release pattern in comparison to control in phosphate buffer at pH 6.8 and HCl buffer at pH3.0. SNF produced significant differences in the CPD release characteristics measured at different pH conditions in comparison to controls. These observations are plausible in terms of drug solubilization effect since SNFs possess a high proportion of surfactant and produce interfacial action on CO and CPD. Antioxidant activities in DAO and AO at CO 20, 40 & 100μg/ml levels were significantly different from controls as measured using the DPPH method. Comparison of antibacterial activity of DAMs against E. coli, B. subtilis, Enterobacter species, and S. aureus (scheme-II) vs CPD suspension or its coarse emulsion or even naive CO counterpart were statistically different and minimum inhibitory concentration of CPD was reduced in SNF formulations. These differences were well corroborated in the data exhibited in zones of inhibition studies. CO-based SNFs produced significantly improved dissolution characteristics of poorly soluble drugs like CPD and potentiated the antibacterial and antioxidant activities. Developed SNFs could be employed as Self nano emulsifying drug delivery systems (SNEDDS) for BCS class IV drugs.
Optimization of a Cefuroxime Axetil-Loaded Liquid Self-Nanoemulsifying Drug Delivery System: Enhanced Solubility, Dissolution and Caco-2 Cell Uptake

Abstract:Cefuroxime axetil (CA) is an oral cephalosporin which hydrolyzes rapidly to the active parent compound cefuroxime. CA is known to have incomplete oral bioavailability (30–50%) due to its poor solubility and enzymatic conversion to cefuroxime in the gut lumen. In order to overcome these drawbacks, a lipid-based self-nanoemulsifying drug delivery system (SNEDDS) has been developed and optimized. The SNEDDS formulations were prepared using the aqueous phase titration method. The greatest self-emulsifying area was found in the 2:1 Smix ratio. As a result, different SNEDDS formulations were carefully selected from this phase diagram based on their smaller droplet size 85%. Thermodynamic stability tests were carried out in order to rule out any metastable/unstable SNEDDS formulations. The droplet size, polydispersity index, zeta potential, and entrapment efficiency (% EE) of optimized CA-loaded SNEDDS (C-3) were 18.50 ± 1.83 nm, 0.064 ± 0.008, −22.12 ± 1.20 mV, and 97.62 ± 1.06%, respectively. In vitro release studies revealed that the SNEDDS formulation had increased CA solubility. CA-SNEDDS-C3 increased CA cellular uptake, possibly due to increased CA solubility and the inhibition of enzymatic conversion to cefuroxime. Finally, in terms of the improvement of oral bioavailability, CA-loaded-SNEDDS could be a viable alternative to commercially available CA formulations.

Many strategies are being investigated to increase the oral absorption of protein. Recently, evaluated the ability of SNEDDS to improve the oral bioavailability of β-lactamase, a model protein. SNEDDS to the solid state can reduce chemical degradation, but in many cases does not eliminate it. Therefore, it is important to identify microenvironment modification strategies to improve the stability of pH-sensitive drugs.

Abstract:Lipid-based drug delivery systems (LBDDS) are the most promising technique to formulate the poorly water soluble drugs. Nanotechnology strongly influences the therapeutic performance of hydrophobic drugs and has become an essential approach in drug delivery research. Self-Nanoemulsifying drug delivery systems (SNEDDS) are a vital strategy that combines benefits of LBDDS and nanotechnology. SNEDDS are now preferred to improve the formulation of drugs with poor aqueous solubility. SNEDDS are isotropic mixtures composed of oils, surfactants, and occasionally cosolvents. The ability of these formulations and methods to produce nanoemulsions or fine oil-in-water (o/w) emulsions after moderate stirring and dilution by water phase along the GI tract. SNEDDS has garnered attention during recent years as it improves oral bioavailability, reduces drug dose, and increases drug protection from unsuitable environment in the gastrointestinal tract. It can solve the problems related to the dissolution and bioavailability of the Biopharmaceutics Classifcation System Class II and IV drugs. This review shortly describes the ambiguity between nanoemulsions and microemulsions, mechanism of self-emulsifications, composition and function of various excipients of SNEDDS. This review discusses characterization of SNEDDS, advantage of SNEEDS over other emulsion, biopharmaceutical aspects, and limitation as well as future views. The SNEDDS is a potential formulation for drug delivery. Owing to its small particle size, large surface area, high encapsulation efficiency, and high drug loading, the SNEDDS can improve the rate and extent of oral absorption by maximizing drug solubility in the intestinal absorption site. Moreover, because of the lipid-based formulation of SNEDDS, it can stimulate and enhance lymphatic transport of drugs to avoid hepatic first-pass metabolism, and thus improve their bioavailability.

Abstract:1) Thirty-eight cases of idiopathic pericarditis in our institutes were analyzed and were classified into 3; group A: clinical picture of pericarditis, group B: clinical picture of peri-myocarditis and group C: dominant clinical picture of chronic pericardial effusion. 2) All cases of group A showed a good clinical course. 3) In group B cases with complete A--V block continued to have heart block with significant myocardial changes, and the prognosis may not be good. 4) Many cases of group C belonged to elderly female and the etiology was unclear. 5) Viral pericarditis was proven in only one case.

Abstract:Approximately one third of newly discovered drug molecules show insufficient water solubility and therefore low oral bio-availability. Self-nano-emulsifying drug-delivery systems (SNEDDSs) are one of the emerging strategies developed to tackle the issues associated with their oral delivery. SNEDDSs are composed of an oil phase, surfactant, and cosurfactant or cosolvent. SNEDDSs characteristics, their ability to dissolve a drug, and in vivo considerations are determinant factors in the choice of SNEDDSs excipients. A SNEDDS formulation can be optimized through phase diagram approach or statistical design of experiments. The characterization of SNEDDSs includes multiple orthogonal methods required to fully control SNEDDS manufacture, stability, and biological fate. Encapsulating a drug in SNEDDSs can lead to increased solubilization, stability in the gastro-intestinal tract, and absorption, resulting in enhanced bio-availability. The transformation of liquid SNEDDSs into solid dosage forms has been shown to increase the stability and patient compliance. Supersaturated, mucus-permeating, and targeted SNEDDSs can be developed to increase efficacy and patient compliance. Self-emulsification approach has been successful in oral drug delivery. The present review gives an insight of SNEDDSs for the oral administration of both lipophilic and hydrophilic compounds from the experimental bench to marketed products.
Development of novel cilostazol-loaded solid SNEDDS using a SPG membrane emulsification technique: Physicochemical characterization and in vivo evaluation

Abstract:The objective of this study was to develop a novel solid self-nanoemulsifying drug delivery system (SNEDDS) using a membrane emulsification technique involving Shirasu porous glass (SPG) which produced very small and uniform emulsion droplets, resulting in enhanced solubility, dissolution and oral bioavailability of poorly water-soluble cilostazol. The effects of carriers on the drug solubility were assessed, and pseudo-ternary phase diagrams were plotted. Among the liquid SNEDDS formulations tested, the liquid SNEDDS composed of peceol (oil), Tween 20 (surfactant) and Labrasol (cosurfactant) at a weight ratio of 15/55/30, produced the smallest emulsion droplet size. The cilostazol-loaded liquid SNEDDS formulation was suspended in the distilled water and subjected to SPG membrane emulsification. Calcium silicate was added as a solid carrier in this liquid SNEDDS, completely suspended and spray-dried, leading to the production of a cilostazol-loaded solid SNEDDS. The emulsion droplet size, solubility and dissolution of the emulsified solid SNEDDS were assessed as compared to the solid SNEDDS prepared without emulsification. Moreover, the physicochemical characteristics and pharmacokinetics in rats were evaluated with the emulsified solid SNEDDS. The emulsified solid SNEDDS provided significantly smaller and more uniform nanoemulsions than did the non-emulsified solid SNEDDS. The emulsified solid SNEDDS showed significantly higher drug solubility and dissolution as compared to the non-emulsified solid SNEDDS. The crystalline drug in it was converted into the amorphous state. Moreover, in rats, it gave significantly higher initial plasma concentrations and AUC compared to the drug powder, suggesting its improved oral bioavailability of cilostazol. Thus, this novel solid SNEDDS developed using a membrane emulsification technique represents a potentially powerful oral delivery system for cilostazol.

Abstract:Background: The main aim of the current work is to set up a revolutionary Self Nano-emulsifying drug delivery system to improve the necessary bio-availability and to boost clinical efficacy when administered orally. Objective: This study's main target is to develop a novel self-nano emulsifying drug delivery system (SNEDDS) for orally administered, poorly water-soluble medicines. SNEDDS is designed to intensify weakly water dissolvable or lipophilic drugs. SNEDDS is designed to intensify weakly water dissolvable or lipophilic drugs. It contains a micro- or nano-emulsion of oil carrying medicament that spontaneously agitates an aqueous medium. For enhancing the solubility of lipophilic medicines, SNEDDS is the most significantly used in biopharmaceutics classification system [BCS class] II and IV pharmaceuticals. Conclusion: The improved rate of dissolution and prevention of interfacial tension are clearly demonstrated by SNEDDS. In aqueous mediums, such as gastrointestinal fluid, SNEDDS produces a stable emulsion after dilution.

Abstract:Objective: The present study involves the development of SNEDDS employing essential oils for enhancing biopharmaceutical performance. Methods: Preliminary investigations suggested the selection of cinnamon oil as an essential oil, tween 60 as a surfactant, while transcutol HP as a cosolvent for formulating SNEDDS. Formulations evaluated for stability, robustness to dilution, and emulsification time, droplet size, zeta potential (ζ), cloud point, in vitro drug release, drug excipient compatibility, TEM, stability assessment and in vivo pharmacokinetic performance in rats. Results: All formulations were robust, stable, and revealed excellent emulsification time<40 s, with fine droplet size (11.41±2.41 nm), lower PDI (0.028-0.277). Formulation F(FLD)6 exhibited a release of 97.7% within 4h, and TEM photograph confirmed spherical droplets. The bioavailability results revealed a higher rate and extent of absorption, AUC, and Cmax for the formulations found to be 1212.4 and 355.40±13.67 (p<0.05). The results recommend that the developed formulation approach offers bioavailability enhancement of FLD. Conclusion: The study concluded that SNEDDS would be an effective formulation system in increasing the aqueous solubility and potentially bioavailability. Furthermore, it can be applied for other therapeutic categories of drugs belonging to BCS class II and IV that show comparable biopharmaceutical challenges.

Abstract:Edoxaban tosylate monohydrate (EDTM) is present in class IV of the biopharmaceutics classification system (BCS), as it is poorly soluble and poorly permeable, which limits its bioavailability. The rationale of this study was to improve the permeability and solubility of EDTM by incorporating it into a self-micro-emulsifying drug delivery system (SMEDDS). Suitable excipients were selected and evaluated for their compatibility. The solubility of EDTM was evaluated for all excipients (oil, surfactant, and co-surfactants) at different ratios. Olive oil, Kolliphor RH40, and PEG-400 were chosen as the oil, surfactant, and co-surfactants, respectively, and were developed into a SMEDDS by adding 15 mg of EDTM. The optimized EDTM-SMEDDS (F1, F2, F3, and F4) were characterized for particle size, polydispersity index (PDI), zeta potential, stability, in vitro release, and ex vivo permeation studies. F2 showed a particle size of 56.43 ± 1.78, PDI of 0.190 and -3.30 ± 0.56 mV of zeta-potential. The enhanced release and permeability of F2 were observed for all other EDTM-SMEDDSs and raw EDTM dispersions. Upon storage under continuous temperature and accelerated stability conditions, F2 showed no signs of phase separation and was visually clear while retaining the %encapsulation efficiency and drug loading, that is, it was stable. The liquid EDTM-SMEDDS improved the bioavailability of EDTM.

Abstract:Biopharmaceutical classification systems (BCS) class III drugs belongs to a group of drugs with high solubility in gastrointestinal (GI) fluids and low membrane permeability result in significantly low bioavailability. Self-emulsifying drug delivery systems (SEDDS) considered a suitable candidate to enhance the bioavailability of poorly soluble drugs by improving their membrane permeability, however, incorporating hydrophilic drugs in to these carriers remained a great challenge. The aim of this study was to develop hydrophobic ion pairs (HIPs) of a model BCS class-III drug tobramycin (TOB) in order to incorporate into SEDDS and improve its bioavailability. HIPs of TOB were formulated using anionic surfactants sodium docusate (DOC) and sodium dodecanoate (DOD). The efficiency of HIPs was estimated by measuring the concentration of formed complexes in water, zeta potential determination and log P value evaluation. Solubility studies of HIPs of TOB with DOC were accomplished to screen the suitable excipients for SEDDS development. Consequently, HIPs of TOB with DOC were loaded into SEDDS and assessed the log DSEDDS/release medium and dissociation of these complexes at different intestinal pH over time. Moreover, cytotoxic potential of HIPs of TOB and HIPs loaded SEDDS formulations was evaluated. HIPs of TOB with DOC exhibited the maximum precipitation efficiency at a stoichiometric ratio of 1:5. Log P of HIPs of TOB improved up to 1500-fold compared to free TOB. Zeta potential of TOB was shifted from positive to negative during hydrophobic ion pairing (HIP). HIPs of TOB with DOC was loaded at a concentration of 1% (w/v) into SEDDS formulations. Log DSEDDS/release medium of loaded complexes in to oily droplets was above 2 and dissociated up to 20% at various pH within 4 h. Finding of this study suggested that improvement of the lipophilic character of BCS class-III drugs followed by incorporation into oily droplets can be deliberated as a promising tool to enhance the permeation across biological membranes.

Abstract:The aim of this study was to prepare an oral administrable SNEDDS for poorly water soluble drugs Bilastine. To choose the SNEDDS excipients, a ternary phase diagram and solubility assessment were conducted. A components of oil (capmul PG-NF) surfactant (Tween 40) and co-surfactant (Capryol 90) were used as the excipients. The optimized formulation was characterized for In-vitro analysis. The prepared S-SNEDDs exhibited good flow characteristics and Particle size (148.5dnm), zeta potential (−28.6), PDI (0.242), drug loading efficiency, % CDR of the optimized S-SNEDDs [TP3PG1 (2:8)] was found to be 103.61 ±0.678, 98.65% respectively. The formulation was stable over a 3-month storage period at 25 ◦C and 4 ◦C in terms of particle size, physical appearance, and drug loading. Overall, a range of lipid based SNEDDS liquid, solid formulations were successfully developed and seem to be a promising alternative to improving the solubility of poorly water-soluble drugs and evaluating In-vitro characterizations.

Abstract:This article describes current strategies to enhance aqueous solubility and dissolution rate of poor soluble drugs. Most drugs in the market are lipophilic with low or poor water solubility. There are various methods to enhance solubility: co-solvency, particle size reduction, salt formation and Self Nanoemulsifying drug delivery systems, SEDDS is a novel approach to enhance solubility, dissolution rate and bioavailability of drugs. The study involves formulation and evaluation of solid self-Nano emulsifying drug delivery system (S-SNEDDS) to enhance aqueous solubility and dissolution rate. Oral route is the most convenient route for non-invasive administration. S-SNEDDS has more advantages when compared to the liquid self-emulsifying drug delivery system. Excipients were selected depends upon the drug compatibility oils, surfactants and co surfactants were selected to formulate Liquid SNEDDS these formulated liquid self-nano emulsifying drug delivery system converted into solid by the help of porous carriers, Melted binder or with the help of drying process. Conversion process of liquid to solid involves various techniques; they are spray drying; freeze drying and fluid bed coating technique; extrusion, melting granulation technique. Liquid SNEDDS has a high ability to improve dissolution and solubility of drugs but it also has disadvantages like incompatibility, decreased drug loading, shorter shelf life, ease of manufacturing and ability to deliver peptides that are prone to enzymatic hydrolysis.

Abstract:Objective: The study aims to develop a Self-nanoemulsifying drug delivery system (SNEDDS) of Quercetin to enhance its oral bioavailability. Methods: In the present study, Quercetin was formulated into SNEDDS using various oils, surfactants, and co-surfactants. The developed formulations were subjected to various studies like drug content analysis, droplet size and thermodynamic stability, and in vitro drug release studies. Results: From the screening of oils, surfactant and cosurfactant, the combination of Triacetin as oil phase, Tween 20 as surfactant and Ethanol as co-surfactant was selected for the development of SNEDDS of Quercetin. The composition of the formulation was optimized using pseudo ternary phase diagram. The optimized formulation has been evaluated and found to have good physical stability and improved in vitro drug release. Conclusion: A stable SNEDDS of Quercetin was developed, and results indicated substantial enhancement in the dissolution of the drug when formulated as a self-nano emulsifying drug delivery system, indicating its potential to enhance oral solubility and bioavailability of the drug.
Comparison of two self-nanoemulsifying drug delivery systems using different solidification techniques for enhanced solubility and oral bioavailability of poorly water-soluble celecoxib

Abstract:In this study, we aimed to develop a solid self-nanoemulsifying drug delivery system (S-SNEDDS) and a solid self-nanoemulsifying granule system (S-SNEGS) to enhance the solubility and oral bioavailability of celecoxib. This process involved the preparation of a liquid SNEDDS (L-SNEDDS) and its subsequent solidification into a S-SNEDDS and a S-SNEGS. The L-SNEDDS consisted of celecoxib (drug), Captex® 355 (Captex; oil), Tween® 80 (Tween 80; surfactant) and D-α-Tocopherol polyethylene glycol 1000 succinate (TPGS; cosurfactant) in a weight ratio of 3.5:25:60:15 to produce the smallest nanoemulsion droplet size. The S-SNEDDS and S-SNEGS were prepared with L-SNEDDS/Ca-silicate/Avicel PH 101 in a weight ratio of 103.5:50:0 using a spray dryer and 103.5:50:100 using a fluid bed granulator, respectively. We compared the two novel developed systems and celecoxib powder based on their solubility, dissolution rate, physicochemical properties, flow properties and oral bioavailability in rats. S-SNEGS showed a significant improvement in solubility and dissolution rate compared to S-SNEDDS and celecoxib powder. Both systems had been converted from crystalline drug to amorphous form. Furthermore, S-SNEGS exhibited a significantly reduced angle of repose, compressibility index and Hausner ratio than S-SNEDDS, suggesting that S-SNEGS was significantly superior in flow properties. Compared to S-SNEDDS and celecoxib powder, S-SNEGS increased the oral bioavailability (AUC value) in rats by 1.3 and 4.5-fold, respectively. Therefore, S-SNEGS wolud be recommended as a solid self-nanoemulsifying system suitable for poorly water-soluble celecoxib.
