Abstract:The impact of deficit and excess of soil water on plant growth, morphological plant features, N and P plant nutrition, soil properties, Rhizobium nodulation and the symbiosis between arbuscular mycorrhizal (AM) fungi and Lotus tenuis Waldst. & Kit. were studied in a saline-sodic soil. Water excess treatment decreased root growth by 36% and increased shoot growth by 13% whereas water deficit treatment decreased both root and shoot growth (26 and 32%, respectively). Differences between stress conditions on shoot growth were due to the ability of L. tenuis to tolerate low oxygen concentration in the soil and the sufficiency of nutrients in soil to sustain shoot growth demands. Water excess treatment decreased pH, and increased available P and labile C in soil. Water deficit treatment decreased available P and also increased labile C. In general, N and P acquisition were affected more by water excess than water deficit. The number of nodules per gram of fresh roots only increased in water excess roots (97%). Under both stress conditions there was a significant proportion of roots colonized by AM fungi. Compared to control treatment, arbuscule formation decreased by 55 and 14% under water excess and water deficit, respectively. Vesicle formation increased 256% in water excess treatment and did not change under water deficit treatment. L. tenuis plants subjected to water deficit or excess treatments could grow, nodulated and maintained a symbiotic association with AM fungi by different strategies. Under water excess, L. tenuis plants decreased root growth and increased shoot growth to facilitate water elimination by transpiration. Under water deficit, L. tenuis plants decreased root growth but also shoot growth which in turn significant decreased the shoot/root ratio. In the present study, under water excess conditions AM fungi reduced nutrient transfer structures (arbuscules), the number of entry points and spore, and hyphal densities in soil, but increased resistance structures (vesicles). At water deficit, however, AM fungi reduced external hyphae and arbuscules to some extent, investing more in maintaining a similar proportion of vesicles in roots and spores in soil compared to control treatment.

Abstract:The present study aimed to investigate the effect of defoliation frequency (low and high) and water stress (excess or deficit) on biomass production, P and N nutrition, and symbiosis with native soil microorganisms on a Lotus tenuis and Schedonorus arundinaceus co-culture in a pot experiment. Combined effects of defoliation frequency and water stress affected plant accumulated shoot biomass. L. tenuis root biomass decreased in response to defoliation and water stress, while S. arundinaceus root biomass was similar between non-defoliated and defoliated plants, at all water levels. Low and high frequencies of defoliation in a waterlogged soil can be considered the most stressful scenario for L. tenuis and S. arundinaceus co-culture. Colonization of arbuscular mycorrhizal fungi in L. tenuis roots and dark septate endophytes colonization in S. arundinaceus roots were affected by both factors, whereas arbuscular mycorrhizal colonization in S. arundinaceus was affected only by water stress. Both plants tolerated defoliation and water stress due to the interaction between the translocation of nutrients and carbon compounds from roots to shoots, and P and N absorption (plus N2 fixation in L. tenuis). Highlights: Both plants tolerated defoliation and water stress due to the interaction between the translocation of nutrients and carbon compounds from roots to shoots, and P and N absorption (plus N2 fixation in tenuis). Abstract:Defoliation often has little effect on the percent plant root length colonised by arbuscular mycorrhizal (AM) fungi, and this has been interpreted as a lack of support for the carbon limitation hypothesis. We performed an experiment with three levels of repeated defoliation (none, every 3 weeks and weekly) of Lolium rigidum growing in pasture soil, and assessed colonisation of roots by naturally occurring AM fungi over 4 months in the glasshouse. Surprisingly, the percent root length colonised by AM fungi increased with defoliation. We also assessed root mass and the length of colonised root to obtain an estimate of the quantity of mycorrhizal root. As expected, both root mass and length of mycorrhizal root decreased with defoliation, as did soluble sugars in the roots. Thus, increasing the frequency of defoliation reduced mycorrhizas consistent with the carbon-limitation hypothesis. Such a decline in mass of mycorrhizal root could contribute to reducing soil biological fertility in pastures over time.
Inhibitory effects of a synthetic atrial peptide on contractions and 45Ca fluxes in vascular smooth muscle.

Abstract:The mechanism of a synthetic atrial peptide (APII)-induced inhibition of smooth muscle contractility was investigated by studying its effects on tension development and 45Ca fluxes in isolated rabbit aorta. APII (10(-9) to 10(-7) M) produced a dose-dependent relaxation of contractions produced by alpha adrenoceptor activation with norepinephrine (NE; 10(-6) M). APII was a potent relaxant of NE contraction with an IC50 = 1.1 X 10(-8) M, with 10(-7) M APII causing a 97% relaxation. APII also produced a dose-dependent inhibition of NE contraction when added to the resting muscle before the exposure to NE. The relaxing effects of APII were found to be endothelium independent. In contrast, APII was only marginally effective in relaxing high-K+ contraction, with 10(-7) M APII causing only 17% relaxation. Furthermore, when a NE contraction was obtained on top of a high-K+ contraction, APII was still capable of relaxing the NE component. APII was similarly more effective in inhibiting NE-stimulated 45Ca influx than high-K+-stimulated 45Ca influx, indicating selective action of APII on the receptor-operated Ca++ channels. This was in contrast to D600, a well known Ca++ antagonist, which had a more selective inhibitory effect on the potential-operated Ca++ channels. The data presented indicate that APII is a potent relaxant of contractions produced by receptor-agonists involving 45Ca influx through receptor-operated Ca++ channels. APII may also prove to be a very useful tool to further distinguish and define receptor-operated Ca++ channels and potential-operated Ca++ channels in vascular smooth muscle.

Abstract:Soil salinity often hinders plant productivity in both natural and agricultural settings. Arbuscular mycorrhizal fungal (AMF) symbionts can mediate plant stress responses by enhancing salinity tolerance, but less attention has been devoted to measuring these effects across plant-AMF studies. We performed a meta-analysis of published studies to determine how AMF symbionts influence plant responses under non-stressed vs. salt-stressed conditions. Compared to non-AMF plants, AMF plants had significantly higher shoot and root biomass ( p < 0.0001) both under non-stressed conditions and in the presence of varying levels of NaCl salinity in soil, and the differences became more prominent as the salinity stress increased. Categorical analyses revealed that the accumulation of plant shoot and root biomass was influenced by various factors, such as the host life cycle and lifestyle, the fungal group, and the duration of the AMF and salinity treatments. More specifically, the effect of Funneliformis on plant shoot biomass was more prominent as the salinity level increased. Additionally, under stress, AMF increased shoot biomass more on plants that are dicots, plants that have nodulation capacity and plants that use the C3 plant photosynthetic pathway. When plants experienced short-term stress (<2 weeks), the effect of AMF was not apparent, but under longer-term stress (>4 weeks), AMF had a distinct effect on the plant response. For the first time, we observed significant phylogenetic signals in plants and mycorrhizal species in terms of their shoot biomass response to moderate levels of salinity stress, i.e., closely related plants had more similar responses, and closely related mycorrhizal species had similar effects than distantly related species. In contrast, the root biomass accumulation trait was related to fungal phylogeny only under non-stressed conditions and not under stressed conditions. Additionally, the influence of AMF on plant biomass was found to be unrelated to plant phylogeny. In line with the greater biomass accumulation in AMF plants, AMF improved the water status, photosynthetic efficiency and uptake of Ca and K in plants irrespective of salinity stress. The uptake of N and P was higher in AMF plants, and as the salinity increased, the trend showed a decline but had a clear upturn as the salinity stress increased to a high level. The activities of malondialdehyde (MDA), peroxidase (POD), and superoxide dismutase (SOD) as well as the proline content changed due to AMF treatment under salinity stress. The accumulation of proline and catalase (CAT) was observed only when plants experienced moderate salinity stress, but peroxidase (POD) and superoxide dismutase (SOD) were significantly increased in AMF plants irrespective of salinity stress. Taken together, arbuscular mycorrhizal fungi influenced plant growth and physiology, and their effects were more notable when their host plants experienced salinity stress and were influenced by plant and fungal traits.

Abstract:Individual plants typically interact with multiple mutualists and enemies simultaneously. Plant roots encounter both arbuscular mycorrhizal (AM) and dark septate endophytic (DSE) fungi, while the leaves are exposed to herbivores. AMF are usually beneficial symbionts, while the functional role of DSE is largely unknown. Leaf herbivory may have a negative effect on root symbiotic fungi due to decreased carbon availability. However, evidence for this is ambiguous and no inoculation-based experiment on joint effects of herbivory on AM and DSE has been done to date. We investigated how artificial defoliation impacts root colonization by AM (Glomus intraradices) and DSE (Phialocephala fortinii) fungi and growth of Medicago sativa host in a factorial laboratory experiment. Defoliation affected fungi differentially, causing a decrease in arbuscular colonization and a slight increase in DSE-type colonization. However, the presence of one fungal species had no effect on colonization by the other or on plant growth. Defoliation reduced plant biomass, with this effect independent of the fungal treatments. Inoculation by either fungal species reduced root/shoot ratios, with this effect independent of the defoliation treatments. These results suggest AM colonization is limited by host carbon availability, while DSE may benefit from root dieback or exudation associated with defoliation. Reductions in root allocation associated with fungal inoculation combined with a lack of effect of fungi on plant biomass suggest DSE and AMF may be functional equivalent to the plant within this study. Combined, our results indicate different controls of colonization, but no apparent functional consequences between AM and DSE association in plant roots in this experimental setup.

Abstract:Soil salinization, especially in arid and semi-arid regions, is one of the major abiotic stresses that affect plant growth. To mediate and boost plant tolerance against this abiotic stress, arbuscular mycorrhizal fungi (AMF) symbiosis is commonly thought to be an effective tool. So, the main purpose of this study was to estimate the role of AMF (applied as a consortium of Claroideoglomus etunicatum Claroideoglomus eutinicatum, Funneliformis mosseae Funliformis mosseae, Rhizophagus fasciculatum, and R. intraradices species) symbiosis in mitigating deleterious salt stress effects on the growth parameters (shoot length (SL), root length (RL), shoot dry weight (SDW), root dry weight (RDW), root surface area (RSA), total root length (TRL), root volume (RV), root diameter (RD), number of nodes and leaves) of Cenchrus ciliaris L. plants through improved accumulations of photosynthetic pigments (chlorophyll a, chlorophyll b, total chlorophyll), proline and phenolic compounds. The results of this experiment revealed that the roots of C. ciliaris plants were colonized by AMF under all the applied salinity levels (0, 75, 150, 225, and 300 mM NaCl). However, the rate of colonization was negatively affected by increasing salinity as depicted by the varied colonization structures (mycelium, vesicles, arbuscules and spores) which were highest under non-saline conditions. This association of AMF induced an increase in the growth parameters of the plant which were reduced by salinity stress. The improved shoot/root indices are likely due to enhanced photosynthetic activities as the AMF-treated plants showed increased accumulation of pigments (chlorophyll a, chlorophyll b and total chlorophyll), under saline as well as non-saline conditions, compared to non-AMF (N-AMF) plants. Furthermore, the AMF-treated plants also exhibited enhanced accumulation of proline and phenolic compounds. These accumulated metabolites act as protective measures under salinity stress, hence explaining the improved photosynthetic and growth parameters of the plants. These results suggest that AMF could be a good tool for the restoration of salt-affected habitats. However, more research is needed to check the true efficacy of different AMF inoculants under field conditions.

Abstract:Soil salinization is one of the most serious abiotic stress factors affecting plant productivity through reduction of soil water potential, decreasing the absorptive capacity of the roots for water and nutrients. A weighted meta-analysis was conducted to study the effects of arbuscular mycorrhizal fungi (AMF) inoculation in alleviating salt stress in C3 and C4 plants. We analyzed the salt stress influence on seven independent variables such as chlorophyll, leaf area, photosynthetic rate (Amax), stomatal conductance (Gs), transpiration rate (E), relative water content (RWC), and water use efficiency (WUE) on AMF inoculated plants. Responses were compared between C3 and C4 plants, AMF species, plant functional groups, level of salinity, and environmental conditions. Our results showed that AMF inoculated plants had a positive impact on gas exchange and water status under salt stress. The total chlorophyll contents of C3 plants were higher than C4 plants. However, C3 plants responses regarding Gs, Amax, and E were more positive compared to C4 plants. The increase in Gs mainly maintained E and it explains the increase in Amax and increase in E. When the two major AMF species (Rhizophagus intraradices and Funnelliformis mosseae) were considered, the effect sizes of RWC and WUE in R. intraradices were lower than those in F. mosseae indicating that F. mosseae inoculated plants performed better under salt stress. In terms of C3 and C4 plant photosynthetic pathways, the effect size of C4 was lower than C3 plants indicating that AMF inoculation more effectively alleviated salt stress in C3 compared to C4 plants.

Abstract:The effects of an arbuscular mycorrhizal (AM) fungus, Glomus mosseae , and a phosphate-solubilizing microorganism (PSM), Mortierella sp., and their interactions, on nutrient (N, P and K) uptake and the ionic composition of different root tissues of the halophyte Kosteletzkya virginica (L.), cultured with or without NaCl, were evaluated. Plant biomass, AM colonization and PSM populations were also assessed. Salt stress adversely affected plant nutrient acquisition, especially root P and K, resulting in an important reduction in shoot dry biomass. Inoculation of the AM fungus or/and PSM strongly promoted AM colonization, PSM populations, plant dry biomass, root/shoot dry weight ratio and nutrient uptake by K. virginica , regardless of salinity level. Ion accumulation in root tissues was inhibited by salt stress. However, dual inoculation of the AM fungus and PSM significantly enhanced ion (e.g., Na + , Cl − , K + , Ca 2+ , Mg 2+ ) accumulation in different root tissues, and maintained lower Na + /K + and Ca 2+ /Mg 2+ ratios and a higher Na + /Ca 2+ ratio, compared to non-inoculated plants under 100 mM NaCl conditions. Correlation coefficient analysis demonstrated that plant (shoot or root) dry biomass correlated positively with plant nutrient uptake and ion (e.g., Na + , K + , Mg 2+ and Cl − ) concentrations of different root tissues, and correlated negatively with Na + /K + ratios in the epidermis and cortex. Simultaneously, root/shoot dry weight ratio correlated positively with Na + /Ca 2+ ratios in most root tissues. These findings suggest that combined AM fungus and PSM inoculation alleviates the deleterious effects of salt on plant growth by enabling greater nutrient (e.g., P, N and K) absorption, higher accumulation of Na + , K + , Mg 2+ and Cl − in different root tissues, and maintenance of lower root Na + /K + and higher Na + /Ca 2+ ratios when salinity is within acceptable limits.
Arbuscular Mycorrhizal Fungi Alleviate Salt Stress Damage by Coordinating Nitrogen Utilization in Leaves of Different Species

Abstract:With the intensification of coastal erosion, damage to coastal shelterbelts has gradually increased. Arbuscular mycorrhizal fungi (AMF) can improve the salinity tolerance and productivity of plants in saline–alkali soils using various strategies including nutrient uptake, osmotic regulation, soil shaping, etc. Thus, the application of AMF to alleviate the impacts of salinization for these shelterbelts has become a research hotspot. For this study, we investigated the effects of inoculation with different AMF strains on the growth and nitrogen (N) utilization of Gleditsia sinensis Lam. and Zelkova serrata (Thunb.) Makino leaves under different salt concentrations. As the salt concentration increased, the growth rates and leaf areas of the autoclaved AMF inoculant (CK) treatment exhibited a decreasing trend for both G. sinensis and Z. serrata, while Funneliformis mosseae (FM) and Corymbiglomus tortuosum (CT) treatments weakened this trend. Between them, on average, FM increased the G. sinensis height growth rate by 396.9%, ground diameter growth rate by 99.0%, and Z. serrata leaf area by 29.1%. At a salt concentration of 150 mM, the chlorophyll content and nitrate reductase activities of leaves under the FM treatment for both tree species were significantly higher than for CK, with an average increase in chlorophyll content of 106.1% and nitrate reductase activities by 74.6%. Moreover, the AMF inoculation significantly reduced the leaf N content and photosynthetic N-use efficiency of G. sinensis in contrast to Z. serrata. Further, in contrast to G. sinensis, the photosynthetic N-use efficiency was significantly positively correlated with the growth rate and leaf area of Z. serrata. Meanwhile, the nitrate reductase activity contributed most to the growth rate and leaf area of Z. serrata. Our results suggest that the issues with coastal shelterbelts might be effectively alleviated through appropriate AMF–plant combinations, which is of great significance for the optimization of forestry production.

Our results suggested that the use of suitable AMF (e.g., Funneliformis mosseae) might effectively improve the currently unfavorable situation of economic tree species production on land with saline soils, which may greatly optimize the utility of these areas.

Abstract:Mycorrhizal inoculation was widely reported to alleviate the damage resulting from NaCl by various physiological ways. However, the symbiotic benefit under distant NaCl concentrations and the relationship among different responsive physiological processes were elusive. In this study, saline resistant plant Xanthoceras sorbifolium was selected as the experimental material and five concentrations of NaCl in the presence or absence of Arbuscular Mycorrhiza Fungi (AMF) were conducted, in order to understand the differences and similarities on the photosynthesis, antioxidant activity, and osmotic adjustment between arbuscular mycorrhizal (AM) plants and non-arbuscular mycorrhizal (NM) plants. Under low salt stress, X. sorbifolium can adapt to salinity by accumulating osmotic adjustment substances, such as soluble protein and proline, increasing superoxide dismutase (SOD), catalase (CAT) activity, and glutathione (GSH). However, under high concentrations of NaCl [240 and 320 mM (mmol·L −1 )], the resistant ability of the plants significantly decreased, as evidenced by the significant downregulation of photosynthetic capacity and biomass compared with the control plants in both AM and NM groups. This demonstrates that the regulatory capacity of X. sorbifolium was limiting, and it played a crucial role mainly under the conditions of 0–160 mM NaCl. After inoculation of AMF, the concentration of Na + in roots was apparently lower than that of NM plants, while Gs (Stomatal conductance) and Ci (Intercellular CO 2 concentration) increased, leading to increases in Pn (Net photosynthetic rate) as well. Moreover, under high salt stress, proline, soluble protein, GSH, and reduced ascorbic acid (ASA) in AM plants are higher in comparison with NM plants, revealing that mycorrhizal symbiotic benefits are more crucial against severe salinity toxicity. Meanwhile, X. sorbifolium itself has relatively high tolerance to salinity, and AMF inoculation can significantly increase the resistant ability against NaCl, whose function was more important under high concentrations.

Abstract:Salinity is a major abiotic stressor that impedes plant growth and negatively affects crop yield. However, Arbuscular mycorrhizal fungi (AMF) can establish a symbiotic relationship with over 80% of terrestrial plant roots. This relationship ultimately results in increased plant growth, improved plant stress resistance, and, consequently, a promising agricultural production and environmental protection solution. The aim of this experiment was to evaluate the impact of arbuscular mycorrhizal fungi on the photosynthetic physiology of strawberries under salinity stress. The greenhouse experiment involved the strawberry cultivar 'Benihoppe', which was inoculated with Glomus mosseae under three salt stress levels (0 mM, 30 mM, and 60 mM). Subsequently, the results showed that salinity stress led to a significant decline in leaf area, fresh biomass, and photosynthetic characteristics of the strawberries. Under salt stress, especially at the concentration of 60 mM. Pn, Gs, Tr, Ci, Fv/Fm, and NPQ showed significant differences. After inoculation of AMF, arbuscular mycorrhiza established a beneficial symbiotic relationship with strawberry roots, which effectively reduced salt damage and promoted the growth of strawberry plants. Leaf area, fresh biomass, and relative chlorophyll content were significantly increased. Pn, Tr, and Gs of mycorrhizal strawberry were significantly higher than those of control group. In addition, the light energy conversion efficiency of strawberry plants inoculated with AMF was improved, thus increasing the potential photosynthetic capacity and photosynthetic rate of strawberry.

Abstract:Arbuscular mycorrhizal fungi (AMF) enhance salt tolerance of host plants, while it is unclear whether aquaporins are involved in the AMF event. This study analyzed the effects of inoculation with an arbuscular mycorrhizal fungus Paraglomus occultum on plant physiological performance and relative expression of root tonoplast intrinsic proteins (TIPs, a kind of aquaporins) genes in potted trifoliate orange (Poncirus trifoliata) seedlings subjected to 0 and 100 mM NaCl concentrations. AMF inoculation significantly increased plant growth performance, leaf water status (relative water content and water potential) and gas exchange (photosynthetic rate, transpiration rate and stomatal conductance), irrespective of non-salt and salt stress. Mycorrhizal seedlings exhibited significantly higher root salicylic acid concentrations and lower leaf and root jasmonic acid concentrations than non-mycorrhizal seedlings under non-salt and salt stress. Transcript levels of PtTIP1;1, PtTIP1;2 and PtTIP1;3 were down-regulated by AMF inoculation under non-salt and salt stress. PtTIP2;1 and PtTIP2;2 expression was unchanged under non-salt stress whereas was down-regulated by AMF colonization under salt stress. AMF inoculation considerably enhanced PtTIP4;1 transcript levels under non-salt and salt stress and PtTIP5;1 expression under non-salt stress. It is concluded that mycorrhizas mitigated salt stress of trifoliate orange through improving physiological performance and regulating root PtTIPs expression.

Abstract:Alfalfa (Medicago sativa L.) is cultivated in arid and semi-arid regions where salinity is one of the main limiting factors for its production. Thus, this experiment was conducted to evaluate the efficacy of arbuscular mycorrhizal fungus (AMF), Glomus mosseae, alfalfa rhizobia Sinorhizobium meliloti (R) seed inoculation in the development of salinity tolerance of different alfalfa cultivars (Rehnani, Pioneer and Bami) under a variety of salinity levels. The results revealed that under non-stress condition, root mycorrhizal infection, nodulation (the number and weight of nodules per plant), potassium (K), calcium (Ca), phosphorus (P), zinc (Zn), copper (Cu) and magnesium (Mg) contents of the root and shoot, the value of the K/Na ratio, protein [calculated from the nitrogen (N) content] and proline contents of the shoot and the alfalfa yield were found to be the highest while Na contents of the root and shoot were seen to be the lowest when seeds were double inoculated followed by mycorrhizae, rhizobium and control treatments, respectively. Similarly, under salinity condition, the greatest amounts of mycorrhizal infection, nodulation, root and shoot P contents, the value of K/Na ratio, the shoot proline content and the root Ca content were enhanced with the least amount of leaf Na content related to the cases of seeds which were double inoculated, followed by mycorrhizae, rhizobium and control treatments respectively. The results suggested that inoculation of alfalfa seed with AMF or R, especially double inoculation, causes a considerable increase in alfalfa yield under both saline and non-saline conditions by increasing colonization, nodulation and nutrient uptake.

Abstract:Pot experiments were conducted to evaluate the damaging effects of salinity on Sesbania sesban plants in the presence and absence of arbuscular mycorrhizal fungi (AMF). The selected morphological, physiological and biochemical parameters of S. sesban were measured. Salinity reduced growth and chlorophyll content drastically while as AMF inoculated plants improved growth. A decrease in the number of nodules, nodule weight and nitrogenase activity was also evident due to salinity stress causing reduction in nitrogen fixation and assimilation potential. AMF inoculation increased these parameters and also ameliorated the salinity stress to some extent. Antioxidant enzymes like superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX) and glutathione reductase (GR) as well as non enzymatic antioxidants (ascorbic acid and glutathione) also exhibited great variation with salinity treatment. Salinity caused great alterations in the endogenous levels of growth hormones with abscisic acid showing increment. AMF inoculated plants maintained higher levels of growth hormones and also allayed the negative impact of salinity.

Abstract:Arbuscular mycorrhizal (AM) fungi enhance plant salt tolerance. However, physiological mechanisms of enhanced salt tolerance in leaves and roots of trees rarely have been compared. To reveal the different mechanisms, our study utilized comprehensive analyses of leaves and roots to examine the effects of Funneliformis mosseae on the salinity tolerance of Zelkova serrata. Seedlings of Z. serrata were exposed to four salt levels in a greenhouse with and without F. mosseae inoculation. Treatment comparisons revealed that following F. mosseae inoculation, (1) nutrient deficiency caused by osmotic stress was mitigated by the fungus enhancing nutrient contents (K, Ca, and Mg) in roots and (N, P, K, Ca, and Mg) in leaves, with Ca and K contents being higher in both leaves and roots; (2) mycorrhizas alleviated ion toxicity by maintaining a favorable ion balance (e.g., K+/Na+), and this regulatory effect was higher in leaves than that in roots; and (3) oxidative damage was reduced by an increase in the activities of antioxidant enzymes and accumulation of antioxidant compounds in mycorrhizal plants although the increase differed in leaves and roots. In particular, AM fungus–enhanced catalase activity and reduced glutathione content only occurred in leaves, whereas an enhanced content of reduced ascorbic acid was only noted in roots. Growth, root vitality, leaf photosynthetic pigments, net photosynthetic rate, and dry weight were higher in seedlings with AM fungus inoculation. These results suggest that AM fungus inoculation improved salinity tolerance of Z. serrata, but the physiological mechanisms differed between leaves and roots.
Salinity and Defoliation Effects on Soybean Growth

Abstract:An experiment was conducted to determine if salinity stress alters the response and tolerance of soybean to defoliation. Four soybean [Glycine max(L.) Merr.] cultivars ('Tachiutaka,' 'Tousan 69,' 'Dare' and 'Enrei') in a growth chamber were exposed to two salinity treatments (0 and 40 mM NaCl) and two defoliation treatments (with and without defoliation). The interactive effects of salinity stress and defoliation on growth rate, leaf expansion, photosynthetic gas exchange, and sodium (Na+) accumulation were determined. The decrease in growth rate resulting from defoliation was more pronounced in plants grown under salinity stress than in those grown without the stress. Without salinity stress, defoliated plants of all four cultivars had leaf-expansion similar rates to those of the undefoliated ones, but the photosynthetic rates of their remaining leaves were higher than those of undefoliated plants. However, with salinity stress, defoliated 'Tachiutaka' and 'Tousa 69' had lower leaf expansion and photosynthetic rates than undefoliated plants. For cultivars 'Dare' and 'Enrei,' the defoliated plants had leaf-expansion rates similar to undefoliated ones, but the photosynthetic rate of the remaining leaves did not increase. Except for cultivar 'Dare,' defoliated plants grown under salinity stress had higher Na+ accumulation in leaves than undefoliated ones, and this result may be related to slow leaf expansion and photosynthesis. Salinity stress negatively affects soybean response and tolerance of defoliation, and the effects varied according to the salt tolerance of the cultivar.

Abstract:Background and aims Soil salinity negatively affects crop development. Halotolerant nitrogen-fixing bacteria (HNFB) and arbuscular mycorrhizal fungi (AMF) are essential microorganisms that enhance crop nutrient availability and salt tolerance in saline soils. Studying the impact of HNFB on AMF communities and using HNFB in biofertilizers can help in selecting the optimal HNFB-AMF combinations to improve crop productivity in saline soils. Methods We established three experimental groups comprising apple plants treated with low-nitrogen (0 mg N/kg, N0), normal-nitrogen (200 mg N/kg, N1), and high-nitrogen (300 mg N/kg, N2) fertilizer under salt stress without bacteria (CK, with the addition of 1,500 mL sterile water +2 g sterile diatomite), or with bacteria [BIO, with the addition of 1,500 mL sterile water +2 g mixed bacterial preparation (including Bacillus subtilis HG-15 and Bacillus velezensis JC-K3)]. Results HNFB inoculation significantly increased microbial biomass and the relative abundance of beta-glucosidase-related genes in the rhizosphere soil under identical nitrogen application levels ( p < 0.05). High-nitrogen treatment significantly reduced AMF diversity and the relative abundance of beta-glucosidase, acid phosphatase, and urea-related genes. A two-way analysis of variance showed that combined nitrogen application and HNFB treatment could significantly affect soil physicochemical properties and rhizosphere AMF abundance ( p < 0.05). Specifically, HNFB application resulted in a significantly higher relative abundance of Glomus-MO-G17-VTX00114 compared to that in the CK group at equal nitrogen levels. Conclusion The impact of HNFB on the AMF community in apple rhizospheres is influenced by soil nitrogen levels. The study reveals how varying nitrogen levels mediate the relationship between exogenous HNFB, soil properties, and rhizosphere microbes.
