Abstract:A total of 75 male Awassi (mean BW 23.5 ± 2.0 kg; 3 months old) were used in an 84-day trial to investigate the effects of different feeding regimes on productive performance, carcass characteristics, and meat quality, and the fatty acid profile of growing lambs. Animals were randomly allocated into 3 groups of 25 lambs each. The dietary treatments were as follows: (1) whole barley grain (60%) plus alfalfa hay (40%; GB-AH; the basal diet); (2) a concentrate pelleted diet plus alfalfa hay (CP-AH); and (3) a complete pelleted diet (CPD). Feed intake was measured weekly, and all lambs were weighed every two weeks for an evaluation of the productive parameters. Blood samples were collected from all lambs for the measurement of biochemical and enzymatic variables. At the end of the experiment, 13 lambs from each treatment were slaughtered to evaluate the carcass characteristics, meat quality, and fatty acid composition. The final body weight, body weight gain, average daily gain, and feed efficiency of lambs were lowest (p < 0.05) in lambs on the grain and alfalfa diet compared with the other groups. Feeding lambs either the CP-AH or CPD diets resulted (p < 0.05) in increases in slaughter weight, carcass weights (hot and cold), the percentage of liver and shoulder, carcass length, back fat thickness, and the area of longissimus thoracis muscle compared with those lambs on the GB-AF diet. The proportion of saturated fatty acids in meat was greater (p = 0.04) in lambs fed on the GA-AH diet than in those of lambs fed on the pelleted diets. Lambs on the CP-AH diet had (p < 0.05) the highest ratios of PUFA to SFA and omega 6 to omega 3, and the proportion of omega 6. The atherogenic and thrombogenic indexes were lower (p < 0.05) in the CP-AH group compared with the GB-AH group. In conclusion, the results indicate that feeding growing lambs on concentrate pellets instead of whole barley grain improves the growth rate, traits, meat quality, and fatty acid profile, which have important implications for productivity, efficiency, and profitability in the livestock industry.
Effects of protein grass hay as alternative feed resource on lamb's fattening performance and meat quality

Abstract:Protein grass hay (PGH) was used as a new feed source for lambs to study its effect on fattening performance and meat quality. Fifty-six male lambs were allotted to four experimental groups and fed for eight weeks either alfalfa hay (AH)-based diet (control) or diets in which AH was replaced with 33 %, 66 %, or 99 % PGH. The inclusion of PGH did not affect final body weight, dry matter intake, average daily gain, feed conversion ratio, or carcass weight. Moreover, substituting AH with PGH at any level did not influence the ruminal fermentation or serum biochemical parameters, meat color, water holding capacity, shear force, or amino acid profile. However, relative liver weight was increased with 66 % substitutions. Furthermore, replacing 99 % AH with PGH decreased the meat's pH at 24 h. Higher levels of C18:3n-3, C20:5n-3, and total n-3 PUFA and a lower ratio of n-6: n-3 PUFA were also observed in meat from lambs fed PGH at 99 %. These findings suggest that PGH could be incorporated into the lamb's diet up to 99 % without compromising fattening performance and body health while improving their meat n-3 PUFA deposition.
The carcass characteristics and quality of meat from lambs grazing perennial wheat with different companion legumes (clover, serradella, lucerne) or a mineral supplement

Abstract:This study compared carcasses as well as the quality and mineral concentration of meat from lambs extensively grazing perennial wheat with clover (PW + C), serradella (PW + S), lucerne (PW + L), or a mineral salt supplement (PW + Min). A split-plot design was used, wherein 3 crossbred ewe lambs (n = 72 in total) (sub-plots) grazed each of 4 forage types (plots), that were replicated across 6 locations (blocks). The feeding study concluded after 96 d, when all the lambs were slaughtered. The left longissimus lumborum muscles (LL) were collected and wet aged for either 5 or 56 d post-mortem. Lambs grazing PW + Min were found to produce carcasses with lower dressing percentage values to those grazing the other forage types (P = 0.037). The LL of lambs grazing PW + L had the lowest crude protein values (P = 0.015). Forage type by ageing period interactions did not affect meat quality. The 56 d ageing period resulted in higher purge loss (P < 0.001) and TVB-N values (P < 0.001) and a decline in shear force (P < 0.001) compared to the 5 d ageing period. The other carcass and meat quality parameters were not affected by forage type; including hot carcass weight, pH decline parameters, eye muscle area, cooking loss, intramuscular fat, sarcomere length, colour stability, and concentrations of calcium, iron, magnesium, sodium, and zinc in the LL. These findings confirm that perennial cereal production systems, that include legume forages with contrasting protein, energy, and micronutrient profiles, can deliver comparable lamb carcasses and meat quality.
Improving Shelf Life and Content of Unsaturated Fatty Acids in Meat of Lambs Fed a Diet Supplemented with Grape Dregs

Abstract:This study was conducted to evaluate the potential effects of dietary grape residue levels on the slaughter indicators, meat quality, meat shelf-life, unsaturated fatty acid content, and expression of fatty acid deposition genes in the muscle of lambs. Sixty 30-month-old male Dorper and Small-Tailed Han F1 hybrid lambs were assigned to a single factor complete randomized trial design and fed with four different diets including 0%, 8%, 16%, and 24% grape dregs, respectively. The findings regarding meat production efficacy in the lambs revealed substantial differences. The control group showed notably lower dressing percentage, carcass weight, net meat weight, meat percentage concerning carcass, meat-to-bone ratio, relative visceral and kidney fat mass, and rib eye area compared to the other groups (p < 0.05). Additionally, the meat shearing force of lambs fed a diet with 16% grape pomace (GP) was significantly higher than that of the 24% GP group (p < 0.05), while the 24 h meat color parameter a* value of the control group was notably higher than that of the 8% GP group (p < 0.05). In addition, compared to the control group, lambs fed with a diet containing 16% GP had higher levels of oleic acid (C18:1n-9c), linoleic acid (C18:2n-6c), behenic acid (C22:0), tricosanoic acid (C23:0), lignoceric acid (C24:0), and conjugated linoleic acid (CLA), at a ratio of ∑CLA/TFA, ∑n-6, ∑MUFA, and ∑PUFA in the longissimus dorsi muscle (p < 0.05), but the reverse case was applicable for Total Volatile Basic Nitrogen (TVB-N) content (p < 0.05). GP supplementation did not substantially affect the expression of stearoyl-CoA desaturase (SCD), peroxisome proliferator activated receptor alpha (PPARα), and peroxisome proliferator-activated receptor gamma (PPARγ) genes (p > 0.05). The findings indicated that incorporating grape dregs in the diets of fattening lambs leads to notable enhancements in meat production and the antioxidant capacity of lamb meat, and effectively extends the shelf life of the meat.

Abstract:Thirty-six Assaf male lambs (29.4 ± 3.10 kg body weight (BW)) were used to study the feasibility of including urea (at 0, 0.6 or 0.95% of dry matter for Control, Urea1, and Urea2 diets, respectively) in substitution of soybean meal in fattening diets. Animals were individually penned and feed intake was recorded daily. Blood samples were taken at days 35 and 63 of the experimental period to determine the acid-base status and the biochemical profile. At the end of the experiment (nine weeks), lambs were slaughtered, ruminal contents were collected and carcass and meat quality were evaluated. There were not differences (p > 0.05) among treatments in dry matter intake, animal performance, ruminal fermentation pattern, and carcass and meat parameters. Serum albumin concentration was higher and concentration of HCO3 and total CO2 in blood were lower in Urea2 compared to Urea1 and Control lambs. These results, together with the tendency to lower (p = 0.065) blood pH in this group might suggest a moderate metabolic acidosis. Partial replacement of soybean meal with urea did not impair growth rate in heavy fattening Assaf lambs (from 29 to 50 kg body weight), reduced feeding costs and had no adverse effects on feed efficiency, rumen fermentation and carcass and meat quality.

Abstract:Maize silage has a high demand for fertilizer and water. As an unconventional feed resource, mulberry silage has the potential to replace most maize silage and to alleviate the shortage of roughage in the mutton sheep industry in China. The purpose of this experiment was to study the effect of the replacement of maize silage and soyabean meal with mulberry silage in the diet of Hu lambs on growth performance, serum biochemical indices, slaughter performance, and meat quality. Ninety-six healthy Hu lambs were randomly divided into four groups with six replicates per group and four lambs per replicate. The amounts of 0, 20, 40, and 60% of maize silage were replaced by mulberry silage in each group (denoted as CON, L, M, and H, respectively). The results showed that replacing maize silage with mulberry silage had no significant effect on the growth performance or the slaughter performance of Hu lambs (p > 0.05). Feeding Hu lambs with mulberry silage significantly reduced serum glucose (GLU) and the blood urea nitrogen (BUN) content (p < 0.05), and it increased the content of ether extract (EE) in the longissimus dorsi muscle (p < 0.05). Meanwhile, the percentage of EAA in the M and H groups was significantly lower than that in the CON and L groups (p < 0.05). In addition, in the fatty acid profile, the percentage of C16:1 in the M group was significantly increased, while the percentage of C18:0 and C20:0 were significantly decreased (p < 0.05). Based on these findings, it was recommended that 20–40% of maize silage be replaced by mulberry silage in the diet of Hu lambs.

Abstract:Abstract Soybean molasses (SBM) is a byproduct of the manufacture of soy protein concentrate and has high energy value. This byproduct has a high potential for use in the nutrition of ruminant animals, mainly in the replacement of other energy feeds such as corn grain. The objective of this study was to evaluate the inclusion of SBM to replace corn grain up to 30% dry matter (DM) in the total diet on growth performance, feeding behavior, carcass characteristics, and meat quality of feedlot lambs (1⁄2 Santa Inês × 1⁄2 Dorper). Forty intact male lambs with an initial average body weight of 20.6 ± 2.5 kg and approximate age of 120 d were used. The animals were distributed in four treatments (0%, 10%, 20%, and 30% SBM), divided into five randomized blocks according to the initial weight and adapted for 16 d, with diets containing increasing concentrations of concentrate and SBM. Feeding behavior was analyzed at the beginning, middle, and final of the finishing period, and when animals reached 42 d on the finishing diet they were slaughtered. Data were evaluated using SAS software (version 9.4), by polynomial orthogonal contrasts, where the growth performance, carcass characteristics, and meat quality values were analyzed as randomized blocks, and the feeding behavior data as randomized blocks with a repeated measure over time. Significant differences were detected for the contrast 0 vs. SBM treatments, which the inclusion of SBM caused an increase (P ≤ 0.05) in ash intake but decreased the ether extract intake. The intake of DM in % body weight was higher for SBM treatments than 0% treatment (P ≤ 0.05). Feeding behavior, ruminating while lying down and drinking water presented a decreasing linear effect (P ≤ 0.05), and for feeding, efficiency increased with the addition of SBM (P ≤ 0.05). Fatty acids C14:0, C17:0, C17:1, C18:2n6c, C20:2, and C20:3n6 showed lower values with the inclusion of SBM (P ≤ 0.05), while fatty acids C22:0 and C22:6n3 increased. The values of n6 polyunsaturated fatty acids and n-6/n-3 ratio were lower (P ≤ 0.05) for SBM treatments. The values of total polyunsaturated fatty acids showed a decreasing linear effect (P ≤ 0.05) with the inclusion of SBM. The use of up to 30% SBM in DM did not impair animal growth performance and feeding behavior did not cause damages to carcass parameters and still made the meat healthier, improving the n-6/n-3 ratio, therefore can be used to feed finishing lambs.

Abstract:In a previous study we showed that feeding fish meal significantly increased muscle long chain n-3 fatty acids (FA) and hot carcass weight. In this study we compared the effect of fish meal and fish oil on increasing muscle long-chain FA. We also investigated whether the increase in carcass weight was due to the effect of dietary enrichment of muscle long-chain n-3 FA on muscle membrane phospholipids and(or) to rumen by-pass protein provided by fish meal. Forty crossbred ([Merino x Border Leicester] x Poll Dorset) wether lambs between 26 and 33 kg BW were randomly assigned to one of five treatments: I) basal diet of oaten:lucerne chaff (Basal); 2) Basal + fish meal (9% DM) = FM; 3) Basal + fish oil(1.5% DM) with protected sunflower meal (9% DM)= FOSMP; 4) Basal + fish oil (1.5% DM)= FO; or 5) Basal + protected sunflower meal (10.5% DM) = SMP. Daily intake of ME (9.60 - 10.5 MJ ME/d) and CP (150 to 168 g/d) in all treatments was kept similar by varying the ratio of oaten:lucerne chaff and by feeding the animals at 90% ad libitum intake. Blood samples were collected at the start of the experiment and on the day (d 42) prior to slaughter. Lambs were then slaughtered at a commercial abattoir. At 24 h postmortem carcass traits were measured and longissimus thoracis muscle taken for analysis of FA of phospholipid and triglyceride fractions. Lambs fed FO and FOSMP showed a marked increase in muscle long-chain n-3 FA (P < 0.001) and a reduction in magnitude of the rise in insulin concentration (P < 0.001) after feeding compared with lambs fed Basal and SMP diets. Lambs in FM had a moderate increase (P < 0.001) in muscle long-chain n-3 FA content. Compared with Basal diet, both plasma total cholesterol (P < 0.02) and high-density lipoprotein cholesterol (P < 0.001) levels were greater in SMP and less in FO and FOSMP treatments. The i.m. fat content was reduced (P < 0.05) in FM and FO treatments, but carcass weight was increased only with fish meal (P < 0.03). Adding SMP to FO produced muscle with an intermediate level of i.m. fat, whereas muscle long-chain n-3 FA, i.m. fat, and insulin concentration were unchanged with SMP treatment. These results indicate that an increase in carcass weight in FM may be due to the supply of ruminally undegraded protein. They also suggest that fish oil along with fish meal can increase long-chain n-3 FA content in phospholipid of muscle membrane. This may be associated with reduced i.m, fat content and altered insulin action and lipoprotein metabolism.

Abstract:To investigate the effect of adding lactic acid bacteria (LAB) to Sunit lambs’ diet on their protein metabolism, 18 3-month-old Sunit sheep were randomly divided into two groups (Group C (non-feeding of LAB) and Group L (feeding of LAB)) for a 90 d feeding trial. After the animals were slaughtered, we measured several key factors, including basal indices, amino acid composition, protein metabolism enzyme activity, and protein metabolism-related gene expression. There were nine replications of each group. The data show that LAB had a significant effect on the intramuscular fat (IMF) content (P<0.001). The composition of amino acids in the serum and mutton was improved by the addition of LAB. The addition of LAB led to a significant increase in the expression of genes related to AKT serine/threonine kinase 1 (Akt1), mammalian target of rapamycin (mTOR), p70 ribosomal protein S6 kinase (p70S6K1), and eukaryotic initiation factor 4E (eIF4E) protein synthesis (P<0.05), while the expression of all genes related to protein catabolism was reduced. Thus, dietary LAB can increase IMF content, improve amino acid composition, regulate protein metabolism in lambs, and provide a new way and theoretical basis for dietary management to improve meat quality in housed sheep.

Abstract:Our study investigated how different levels of antioxidants and contrasting proportions of native legumes in the diet affect lamb meat quality. Twenty-four male Texel lambs were randomly assigned to three groups: two groups on a natural pasture in southern Brazil (Pampa Biome), each at a different proportion of legumes: Low-legume (LL, 4.37%) and High-legume (HL, 14.01%); the other group was stall-fed (Control) to achieve the same growth rates as the grazing groups. Cold carcass yield from the Control lambs was higher than HL. The meat from pasture-fed animals had a higher deposition of muscle α-tocopherol and lower lipid oxidation (TBARS values) after 9 days of storage. LL lambs had higher subcutaneous fat thickness, which promoted better sensory quality of the meat, as assessed by a trained panel. Pasture-based diets enhanced odd- and branched-chain fatty acids (OBCFAs), trans vaccenic acid, and total conjugated linoleic acids (CLAs), while decreasing elaidic acid. Despite the lower ∆9-desaturase activity, the higher proportion of Desmodium incanum (condensed tannin-rich native legume) in the HL diet did not impact meat nutritional quality. Finishing lambs on the Pampa Biome grasslands is an option for improving the oxidative stability and beneficial fatty acid content of lamb meat, which improves product quality and human consumer health.
Effects of Diets Containing Finger Millet Straw and Corn Straw on Growth Performance, Plasma Metabolites, Immune Capacity, and Carcass Traits in Fattening Lambs

Abstract:As the byproduct of finger millet, millet straw is a new forage resource of ruminants. The effect of the combined utilization of millet straw with corn straw on fattening lamb production is seldom reported. The purpose of this study was to investigate the effect of different proportions of millet straw instead of corn straw on the growth performance, blood metabolites, immune response, meat yield, and quality of fattening lamb. Sixty-three-month-old healthy Small-Tailed Han sheep crossbred rams with an average initial weight of 19.28 ± 2.95 kg were randomly divided into four groups, with three replicates in each group and five lambs in each replicate. The replacement ratio of millet straw of each group (Group I, II, III, IV) was 0%, 25%, 35%, 50% at the first stage (the first two months) and 0%, 20%, 28%, 40% in the second period (final two months), respectively. The experiment lasted 4 months 10 days of the pre-feeding period. The results indicated that the body weight gain and average daily gain of group II were significantly higher than those of group I and group IV (p < 0.05). The concentration of total protein in group IV was significantly increased compared to those of the other three groups at the second stage (p < 0.05), which proved that the protein synthesis metabolism capacity was improved with the addition of millet straw. The concentration of the plasma glutamic-oxalacetic transaminase and lactic dehydrogenase of lambs was significantly decreased in group II (p 0.05). There was no significant difference in the pre-slaughter weight, carcass weight, dressing percentage, loin-eye area, and GR value among the four groups (p > 0.05). Furthermore, the immune response and meat quality were not impacted by the different proportions of millet and corn forage diets. The results showed that the combined utilization of millet straw with corn straw could improve the blood biochemistry metabolism capability of fattening lambs. The replacement of 50% of corn straw with millet straw could improve the growth performance and be an application in fattening lamb production.

Biodiversity and Sustainable Agriculture in the Fertile Crescent

Abstract:The Fertile Crescent is a high biodiversity region where most temperate-zone agricultural species originated and were first domesticated. A favorable environment, a special plant community, and an adaptive population combined to initiate the Neolithic—the New Stone Age—in the Fertile Crescent. However, over time, valuable plant genetic resources of the region are being eroded through degradation of natural habitats, intensification of the cultivation of arable lands, expansion of cultivation into marginal areas, replacement of diverse and widely adapted landraces by new cultivars based on a narrow genetic base, and over-exploitation of natural pastures and grazing lands. There is a grave risk that much of the inherent biodiversity of the Fertile Crescent will be lost unless a holistic approach to the management of ecosystems, based on sustainable agriculture and sustainable development, is implemented. Components of the holistic approach are presented and discussed under the following five themes: (i) assessment of problems, obstacles and needs (ii) land use management, including soils, water and vegetation (iii) the conservation of agricultural biodiversity and its utilization in agricultural production systems (iv) social, political and economic factors, including indigenous knowledge and (v) human resources. It is hypothesized that food security for future generations in the Fertile Crescent will be strengthened by the conservation and utilization of agricultural biodiversity. This can be achieved through the development of productive and sustainable resource management strategies in the agroecosystems of the region. INTRODUCTION The modern territories Iraq, Jordan, Lebanon, Israel, Palestine, Syria, western Iran, and southeast Turkey encompass the region often referred to as the Fertile Crescent. Here, many of the species of temperate-zone agriculture originated and were first domesticated. Their wild relatives and landraces are still found in the region (Harlan 1992; Zohary and Hopf 1993). It is an area of megadiversity of important food, feed, medicinal, horticultural crops, spices, pasture and rangeland species. Abstract:The fatty acid composition for the longissimus dorsi (LD) fat of carcass sheep is a crucial factor impacting meat quality. We performed a 90-day feeding trial of 25 Naemi lambs to investigate the effects of roughage sources (alfalfa or wheat straw) of two sizes (regular and 1 cm chopped) when fed with pelleted total mixed ration (TMR) on the growth performance, fermentation patterns, and fatty acid (FA) composition of longissimus dorsi (LD) fat. Lambs were randomly assigned to individual pens with five treatment diets, as follows: C, control group with TMR; T1, TMR and regular alfalfa hay; T2, TMR and alfalfa hay chopped to 1 cm; T3, TMR and regular wheat straw; and T4, TMR and wheat straw chopped to 1 cm. Four lambs were randomly selected from each treatment (20 total) and sacrificed. LD fat of the carcass was extracted and analyzed for FA using a gas chromatography-mass spectrometry. Significantly increased feed intake was found in T1 and T2. The FA composition of LD fat in T2 had higher unsaturated fatty acid (UFA), omega-6 (n6), and omega-3 (n3) FA content. Conjugated linoleic acid (CLA) and α-linoleic acid were highest in lambs fed T1 and T2. Feeding different types of roughage, especially alfalfa hay, either regular or chopped, with total pelleted mixed ration is crucial to improving feed intake and body weight gain, as it positively enhances the rumen microbial fermentation process by controlling rumen pH. The FA profiles of meat from lambs fed TMR with regular or 1 cm particle size alfalfa hay (T1 and T2) are recommended for human consumption as a source of healthy FAs.

Abstract:ABSTRACT Diet modulates the rumen microbiota, which in turn can impact the animal performance. The rumen microbiota is increasingly recognized for its crucial role in regulating the growth and meat quality of the host. Nevertheless, the mechanism by which the rumen microbiome influences the fatty acid and amino acid profiles of lambs in the grass feeding system remains unclear. This study aimed to evaluate the effects of different native grass-based diets on animal performance, meat quality, fatty acid compositions, amino acid profiles, and rumen microbiota of lamb. Seventy-two Ujumqin lambs were randomly assigned into three treatments according to the initial body weight (27.39 ± 0.51 kg) and age (6 months ± 6 days). The lambs received three diets: (i) non-pelleted native grass hay with 40% concentrate diet; the native grass and concentrate were fed individually; (ii) pelleted native grass hay with 40% concentrate diet (PHLC); (iii) pelleted native grass hay with 60% concentrate diet (PHHC). The results showed that among the three groups, the PHHC and PHLC diets had markedly ( P < 0.05) higher average daily gain and pH 45 min , respectively. All amino acid levels were significantly ( P < 0.05) decreased in the PHHC diet than in the PHLC diet. The principal coordinate analysis of the ruminal microbiota indicated the markedly distinct separation ( P = 0.001) among the three groups. In addition, the correlation analysis showed that the Rikenellaceae _RC9_gut_group, Prevotellaceae _UCG-003, Succinivibrio , and Succiniclasticum were significantly ( P < 0.05) associated with most of the fatty acid and amino acid profiles. The correlation analysis of the association of microbiome with the meat quality provides us with a comprehensive understanding of the composition and function of the rumen microbial community, and these findings will contribute to the direction of future research in lamb. IMPORTANCE Diet modulates the gut microbiome, which in turn impact the meat quality, yet few studies investigate the correlation between the rumen microbiome and the fatty acid profile of meat. Here, the current study develops an experiment to investigate the correlation of the rumen microbiome and fatty acid profile of meat: rumen microbiome responses to feed type and meat quality. The results indicated a unique microbiota in the rumen of lamb in response to diets and meat quality. Associations between utilization and production were widely identified among the affected microbiome and meat quality, and these findings will contribute to the direction of future research in lamb.

Effects of dietary energy level on growth performance, blood parameters and meat quality in fattening male Hu lambs

Abstract:This experiment was conducted to investigate the effects of different dietary energy levels on growth performance, slaughter traits, meat quality and blood biochemical parameters in fattening male Hu lambs. Sixty lambs were fed five iso-protein diets which contained different levels of metabolizable energy in a completely randomized design for 70 days. At the end of study, fifteen lambs were harvested for further study. With the increase in dietary energy level, the daily weight gain and dry matter intake extremely increased (p < .001), and feed conversion ratio decreased significantly (p < .01). The live weight before slaughter (LWBS) and carcass weight had a significant increase (p < .05), non-carcass fat ratio and routine indexes of meat quality had no significant difference. Almost no effect was observed for the amino acid profile except for glycine concentration decreased (p < .05) in longissimus dorsi (LD) muscle. The concentrations of C17:0, C18:3n-3 and n-3 PUFA (p < .01) significantly decreased with the increasing dietary energy levels, and the ratio of ∑n-6/∑n-3 (p < .01) increased, whereas the concentrations of C18:1n-9t (p < .05) decreased in LD muscle and C18:1 (p < .01) increased in biceps femoris (BF) muscle. There were no obviously differences for diameter, area and density of muscle fibres. The relative expression of MyHC-IIa and MyHC-IIx decreased significantly (p < .05) in BF muscle, and the relative expression of MyHC-IIa displayed an obviously decreasing trend (p < .10) in LD muscle. These results suggest that increasing the dietary energy level can improve the growth performance and slaughter traits, and influence meat quality and fatty acid profiles in different muscle tissues of fattening male Hu lambs. These results provide a theoretical basis for developing Hu sheep nutritional standards and designing feed formulations.

Abstract:δ-N-(Phosphonacetyl)-l-ornithine [Orn(PAc)], a transition state analogue for ornithine transcarbamylase, is an effective inhibitor of citrulline synthesis in isolated rat liver mitochondria, and urea synthesis in isolated hepatocytes. In intact mitochondria, the efficacy of inhibition of citrulline synthesis by Orn(PAc) was shown to be dependent on the conditions of incubation. The citrulline biosynthetic capacity of intact mitochondria was reduced to 60% of controls by as little as 0.01 mM inhibitor, and to 20% of controls by 0.05 mM inhibitor in mitochondria treated with 0.5% Triton X-100. The greater capacity of Orn(PAc) to inhibit citrulline synthesis after treatment of mitochondria with Triton X-100 was in part due to the elimination of a permeability barrier to reactants of the carbamoyl-phosphate synthetase (ammonia) reaction. Thus, maximum inhibition by Orn(PAc) was dependent on the relative activities of carbamoyl-phosphate synthetase (ammonia) and ornithine transcarbamylase, and inhibition could be completely reversed by stimulating ornithine transcarbamylase using ornithine. When added to isolated hepatocytes, Orn(PAc) inhibited urea biosynthesis by up to 20% at 0.5 mM concentration of inhibitor. This inhibition in the conversion of carbamoyl phosphate to citrulline had a marked effect on the utilization of intra-mitochondrially synthesized carbamoyl phosphate for extramitochondrial synthesis of pyrimidines. There was up to a twofold stimulation in the rate of synthesis of orotate and uridine nucleotides when Orn(PAc) was added to hepatocytes incubated in basal medium (Eagles) supplemented with 10 mM NH4Cl. These data confirm clinical observations that one of the consequences of a reduction in ornithine transcarbamylase activity, such as occurs in certain disease states, is a stimulation in pyrimidine biosynthesis, and further suggests that transition-state analogues, such as Orn(PAc), may be useful in metabolic modelling studies on the effects of enzymic defects in mammalian tissues.

Abstract:Summary Three experiments were conducted to investigate the influence on nitrogen retention of feeding protein supplements to early weaned lambs direct to the abomasum via the oesophageal groove. In Exp. 1 the lambs received a basal diet of barley and a protein mixture consisting of soyabean meal, sunflower meal and fish meal which was given either in a liquid suspension from a bottle or as dry feed incorporated with the barley. The results showed that the regression coefficients of nitrogen retention on intake were 0.48 and 0.33 with liquid and dry feeding respectively. The regressions of urinary nitrogen on intake were with liquid and dry feeding respectively 0.26 and 0.44. In Exp. 2, various sources of protein were compared isonitrogenously as substitute for part of a barley concentrate ration. The nitrogen intakes and retentions were 14·28 and 5·09, 20·16 and 9·39, 20·53 and 8·19, 19·00 and 7·80, and 18·87 and 7·86 g/day for the control diet and fish meal, yeast, soyabean meal and sunflower meal supplemented diets respectively. In Exp. 3 the fish meal supplement was compared with a high lysine, bloodmeal and with casein as liquid supplements to a basal diet of barley concentrate. The intake and retention of nitrogen were for bloodmeal, casein and fish meal respectively 23·41 and 7·48, 22·56 and 9·11, and 21·88 and 8·31 g/day. It appeared that the protein sources high in methionine resulted in the highest nitrogen retention. The implications of these findings in relation to the nutrition of young ruminants is discussed together with an evaluation of the experimental method as a technique to study postruminal digestion.
