Abstract:Effective pathogen detection is an essential prerequisite for the prevention and treatment of infectious diseases. Despite recent advances in biosensors, infectious diseases remain a major cause of illnesses and mortality throughout the world. For instance in developing countries, infectious diseases account for over half of the mortality rate. Pathogen detection platforms provide a fundamental tool in different fields including clinical diagnostics, pathology, drug discovery, clinical research, disease outbreaks, and food safety. Microfluidic lab-on-a-chip (LOC) devices offer many advantages for pathogen detection such as miniaturization, small sample volume, portability, rapid detection time and point-of-care diagnosis. This review paper outlines recent microfluidic based devices and LOC design strategies for pathogen detection with the main focus on the integration of different techniques that led to the development of sample-to-result devices. Several examples of recently developed devices are presented along with respective advantages and limitations of each design. Progresses made in biomarkers, sample preparation, amplification and fluid handling techniques using microfluidic platforms are also covered and strategies for multiplexing and high-throughput analysis, as well as point-of-care diagnosis, are discussed.

Abstract:The emergence of nanofluidic lab‐on‐a‐chip systems heralds new opportunities in molecular biosensing. This review provides a holistic overview of the principles and construction of functional nanofluidic devices, focusing on their applications in single‐molecule analysis, ultrasensitive detection, and subcellular analysis of single cells. The review aims to provide the inspiration and guidance to accelerate the clinical translation of nanofluidic technologies. Biosensing plays a vital role in healthcare monitoring, disease detection, and treatment planning. In recent years, nanofluidic technology has been increasingly explored to be developed into lab‐on‐a‐chip biosensing systems. Given now the possibility of fabricating geometrically defined nanometric channels that are commensurate with the size of many biomolecules, nanofluidic‐based devices are likely to become a key technology for the analysis of various clinical biomarkers, including DNA (deoxyribonucleic acid) and proteins in liquid biopsies. This review summarizes the fundamentals and technological advances of nanofluidics from the purview of single‐molecule analysis, detection of low‐abundance molecules, and single‐cell analysis at the subcellular level. The extreme confinement and dominant surface charge effects in nanochannels provide unique advantages to nanofluidic devices for the manipulation and transport of target biomarkers. When coupled to a microfluidic network to facilitate sample introduction, integrated micro‐nanofluidic biosensing devices are proving to be more sensitive and specific in molecular analysis compared to conventional assays in many cases. Based on recent progress in nanofluidics and current clinical trends, the review concludes with a discussion of near‐term challenges and future directions for the development of nanofluidic‐based biosensing systems toward enabling a new wave of lab‐on‐a‐chip technology for personalized and preventive medicine.

Abstract:Over the last thirty years, lab-on-a-chip (LOC) technology has proven to be a valuable tool for biomedical research, particularly for mammalian cells. With recent advances in nanofabrication capability, nanostructures or nanodevices have been incorporated in LOC devices to enable the development of bacterial research. In this work, we review the recent research progress pertaining to LOC devices involving integrated nanostructures fabricated using the top-down nanofabrication methods, targeted at bacteria. First, we review the widely used top-down nanofabrication technologies to fabricate nanostructures. Next, we survey the capture, lysing, and detection of bacteria via nanofabrication enabled LOC technology. The synergy of LOC and nanofabrication could help in precisely isolating, lysing, and detecting bacteria, thereby enabling the rapid diagnoses of pathogens with a high sensitivity. Moreover, the synergy can help dissect the complex life of bacteria and clarify any perceptions pertaining to bacteria. Finally, we discuss the major challenges in combining nanofabrication and LOC technology, such as in terms of the process compatibility and material choices, as well as scope for future work.

Abstract:Lab-on-a-chip (LOC) biosensors have recently piqued the interest of the research community as a result of its potential utility in personal healthcare and disease diagnostics. LOCs devices have been consistently developed over the last decades as merging microfluidics onto a single chip for performing many lab studies at the same time, such as biochemical and biomolecular detection. Molding, microcontact printing, micromachining, and other techniques were used in the preliminary advancement of miniature sensing platforms known as micro total analysis systems (μTAS). These time-consuming and multi-step processes were utilized to create structures on a micrometer size. As time passes, new approaches and modifications for replacing rigid substrates with flexible substrates were developed. Over the years, paper and plastic substrates have shown unique properties such as durability, flexibility, mobility, cost-effective, and simple manufacturing procedure owing to their compatibility with a wide range of printing equipment. This review discusses the different types of fabrication methods and techniques such as photolithography, soft lithography, screen printing, inkjet printing, laser micromachining, nanoimprinting for designing LOC sensing devices extensively. The types of transduction systems which includes electrochemical, optical and mechanical that play an important role in sensing devices have also been extensively described in this manuscript. Additionally, detection of numerous analytes categorized into small molecules, macromolecules and cell bodies have been comprehensively reviewed in this manuscript with illustrations and tabulated form.

Abstract:Various diseases caused by food-borne or environmental pathogenic microorganisms have been a persistent threat to public health and global economies. It is necessary to regularly detect microorganisms in food and environment to prevent infection of pathogenic microorganisms. However, most traditional detection methods are expensive, time-consuming, and unfeasible in practice in the absence of sophisticated instruments and trained operators. Point-of-care testing (POCT) can be used to detect microorganisms rapidly on site and greatly improve the efficiency of microbial detection. Lab-on-chip (LOC) is an emerging POCT technology with great potential by integrating most of the experimental steps carried out in the laboratory into a single monolithic device. This review will primarily focus on principles and techniques of LOC for detection of microbial nucleic acid in food and environment, including sample preparation, nucleic acid amplification and sample detection.

Abstract:Rapid on-site detection of pathogenic bacteria with high sensitivity and specificity is becoming an urgent need in public health assurance, medical diagnostics, environmental monitoring, and food safety fields. Despite being reliable and widely used, the existing methods of bacteria detection are cumbersome and time-consuming, which is not conducive to field detection. Microfluidic lab-on-a-chip technology has provided a detective tool for various analytes, due to its miniaturization, portability and low reagent consumption. Within this progress report, advances in the bacteria detection using microfluidic biosensors were discussed. Typical methods for pathogenic bacteria capture, separation and detection were introduced respectively in the first part. Then key applications of microfluidic biosensor-based rapid bacteria detection were presented. Finally, we made a conclusion and discussed possible research prospects in aspects of microfluidic biosensors for rapid detection of pathogenic bacteria.

Abstract:The potential threat posed by disease outbreaks to diagnostic instruments demands the development of more effective biosensor technologies to counteract the risks. Diseases like SARS-CoV-2, Ebola, malaria, cholera, and many more have demonstrated beyond the limits of health care that new advancements are required for early detection and diagnosis. The rising number of diseases outbreaks has led to an increasing demand for biosensors that are more effective and quicker to utilize in healthcare settings. A biosensor incorporated with microfluidic chips offers an improved detection compared to traditional or classical biosensors. Microfluidic chips improve the performance of the biosensors by allowing automation, mixing, separation, throughput processing, and transport of the analytes to desired reactors. A biosensor incorporated with a microfluidic chip has improved sensitivity, easy operation and can use small volumes of samples to process the results. The effectiveness of biosensors depends also on the materials used in its fabrication and there are many materials used for fabrication which are reviewed in this work. This paper reviews the potential advantages of the use of microfluidic chips to enhance the performance of biosensors, materials used to fabricate the chips, and potential electrodes incorporated into microfluidic chips which improve the detection time by shortening the processing time for biosensors at the point of care service. This work also reviews new technologies which are not previously addressed other reviews including, integration of cell-imprinted polymers with microfluidic sensors and delved into future technologies outlook.

Abstract:The field of microfluidics has yet to develop practical devices that provide real clinical value. One of the main reasons for this is the difficulty in realizing low-cost, sensitive, reproducible, and portable analyte detection microfluidic systems. Previous research has addressed two main approaches for the detection technologies in lab-on-a-chip devices: (a) study of the compatibility of conventional instrumentation with microfluidic structures, and (b) integration of innovative sensors contained within the microfluidic system. Despite the recent advances in electrochemical and mechanical based sensors, their drawbacks pose important challenges to their application in disposable microfluidic devices. Instead, optical detection remains an attractive solution for lab-on-a-chip devices, because of the ubiquity of the optical methods in the laboratory. Besides, robust and cost-effective devices for use in the field can be realized by integrating proper optical detection technologies on chips. This review examines the recent developments in detection technologies applied to microfluidic biosensors, especially addressing several optical methods, including fluorescence, chemiluminescence, absorbance and surface plasmon resonance.

Abstract:Improved environmental monitoring of microorganisms is currently an important issue, since traditional detection methods are costly and time consuming (often requiring 2–3 days). In contrast, microfluidic chip-based technologies provide promising alternatives. The presence of environmental contaminants, especially bacterial, viral, and parasitic pathogens and their toxins, is creating a major public health problem. The detection of microbial communities in environmental samples is the first step to recognizing these organisms and insuring health and safety against their infection. This review addresses both the existing and innovative technologies for the detection of pathogens present in the environmental samples. First, impacts and challenges related to pathogens in the environmental samples are discussed, and standard techniques for their identification are addressed. Next, the microfluidic technologies are introduced as an alternative strategy with a particular focus on the detection of environmental contaminants. Challenges that still need to be overcome are also discussed. The review concludes with a section on paper-based microfluidic device and a discussion on future prospects.

Abstract:Advances in molecular biology are enabling rapid and efficient analyses for effective intervention in domains such as biology research, infectious disease management, food safety, and biodefense. The emergence of microfluidics and nanotechnologies has enabled both new capabilities and instrument sizes practical for point-of-care. It has also introduced new functionality, enhanced sensitivity, and reduced the time and cost involved in conventional molecular diagnostic techniques. This chapter reviews the application of microfluidics for molecular diagnostics methods such as nucleic acid amplification, next-generation sequencing, high resolution melting analysis, cytogenetics, protein detection and analysis, and cell sorting. We also review microfluidic sample preparation platforms applied to molecular diagnostics and targeted to sample-in, answer-out capabilities.
Integrated High-Throughput Centrifugal Microfluidic Chip Device for Pathogen Detection On-Site

Abstract:An integrated and high-throughput device for pathogen detection is crucial in point-of-care testing (POCT), especially for early diagnosis of infectious diseases and preventing the spread of infection. We developed an on-site testing platform that utilizes a centrifugal microfluidic chip and automated device to achieve high-throughput detection. The low-power (<32 W), portable (220 mm × 220 mm × 170 mm, 4 kg) device can complete bacterial lysis, nucleic acid extraction and purification, loop-mediated isothermal amplification (LAMP) reaction, and real-time fluorescence detection. Magnetic beads for nucleic acid adsorption can be mixed by applying electromagnetic fields and centrifugal forces, and the efficiency of nucleic acid extraction is improved by 60% compared to the no-mixing group. The automated nucleic acid extraction process achieves equivalent nucleic acid extraction efficiency in only 40% of the time consumed using the kit protocol. By designing the valve system and disc layout, the maximum speed required for the centrifugal microfluidic chip is reduced to 1500 rpm, greatly reducing the equipment power consumption and size. In detecting E. coli, our platform achieves a limit of detection (LOD) of 102 CFU/mL in 60 min. In summary, our active centrifugal microfluidic platform provides a solution for the integration of complex biological assays on turntables, with great potential in the application of point-of-care diagnosis.

Abstract:This report deals with the pathology, diagnosis and therapy of cryptorchidism from the andrological point of view. Hypothalamic-pituitary gonadal failure and a reduced transformation of gonocytes are the key factors in fertility disorders. The aim of all well-timed therapy must be to improve the fertility potential of the sexually mature male. At present, the determination through biopsy of the fertility index during orchiopexy offers the only possibility of prospective diagnosis in this respect. In the future, it will be necessary to have a standardized procedure in order to guarantee the long-term effect of correctly timed orchiopexy on fertility.

Abstract:Aptamers are oligonucleotides or peptides that are selected from a pool of random sequences that exhibit high affinity toward a specific biomolecular species of interest. Therefore, they are ideal for use as recognition elements and ligands for binding to the target. In recent years, aptamers have gained a great deal of attention in the field of biosensing as the next-generation target receptors that could potentially replace the functions of antibodies. Consequently, it is increasingly becoming popular to integrate aptamers into a variety of sensing platforms to enhance specificity and selectivity in analyte detection. Simultaneously, as the fields of lab-on-a-chip (LOC) technology, point-of-care (POC) diagnostics, and personal medicine become topics of great interest, integration of such aptamer-based sensors with LOC devices are showing promising results as evidenced by the recent growth of literature in this area. The focus of this review article is to highlight the recent progress in aptamer-based biosensor development with emphasis on the integration between aptamers and the various forms of LOC devices including microfluidic chips and paper-based microfluidics. As aptamers are extremely versatile in terms of their utilization in different detection principles, a broad range of techniques are covered including electrochemical, optical, colorimetric, and gravimetric sensing as well as surface acoustics waves and transistor-based detection.

Abstract:Microfluidic chips have the advantages of small sample size, low detection cost, and short detection time. Combined with optical sensors, electrochemical sensors, and microwave sensors, they can be used to construct sensitive, repeatable, and portable detection systems. Optical biosensors are powerful analytical instruments that can measure biomolecular interactions in real-time and unlabeled manner. The combination of affinity and kinetics can be measured through high sensitivity. many optical biosensors have microfluidic control, making them extremely sensitive. Combining the concept of microfluidic technology, this article briefly elucidates the various cutting-edge research fields of microfluidic technology combined with optical biosensors, such as optical detection methods suitable for microfluidic chips, microfluidic biological immunofluorescence detection, the application of microfluidic technology in optical detection methods. It also prospects the future development direction and the challenges as well as strategies for addressing the problems in the development of the new generation of biological microfluidic sensors were proposed.

Abstract:Molecular biological methods such as PCR and LAMP, which are based on nucleic acid amplification, is becoming more and more popular because of their shorter reaction time and real-time detection ability. This study aimed to develop a micro-nanofluidic bio-reaction detecting system for isothermal DNA amplification and sensitive real-time detection of the amplification for sequence-special molecular identification. The identification system contains a plastic micro-nanofluidic chip and a real-time confocal detector. The DNA amplification method is based on loop-mediated isothermal amplification (LAMP). The primary chip was designed with the SolidWorks (TM) and fabricated from the optical disk production line with multiple parallel bioreactors in micro-nano liter in volume. Listeria moncytogenes, a bacterial species that causes diarrhea was chosen as the biological test object. The response time of the micro-nanofluidic system with a volume of 1.45 mu L was reduced to 45 minutes. The detection limit was as low as 1 DNA copy. The detector provided highly sensitive multi-target parallel detection in real time and low background noise. Chip's inert surface, UNG and dUTP in the reaction system were able to improve amplification stability in micro-nanoliter reaction assays. Our system can be developed to a novel high throughput lab-on-a-chip device and is promising to be used for single-molecule droplet amplification. It also has the potential for applications such as nanomedicine, clinical molecular diagnostics, and nanobiotechnology.

Abstract:Conventional microbiological methods for pathogen detection are time consuming methods and require tedious protocol, expensive instruments, and expert personnel, which delays diagnosis time and treatment. To overcome these limitations, a miniature microfluidic system has been introduced for rapid, sensitive, specific, and easy to handle system for microbial pathogen detection. This miniaturized system uses the dynamics of fluid in microchannels for the collection and detection of pathogen. Microfluidics coupled with a different analytical detection system allow the identification and quantification of whole cells, metabolites, and genetic materials, for accurate diagnosis with all details about microbial pathogen. The high throughput efficiency of a microfluidic system is suitable for point-of-care testing of a broad range of pathogens in even resource-limited facilities/areas. Herein, fundamentals of designing and advances in microfluidic chip integrated with analytical system for pathogen detection are discussed. This review will establish microfluidic as a rapid and accurate point-of-care detection system for microbial pathogens.

Abstract:Food quality and safety are important to protect consumers from foodborne illnesses. Currently, laboratory scale analysis, which takes several days to complete, is the main way to ensure the absence of pathogenic microorganisms in a wide range of food products. However, new methods such as PCR, ELISA, or even accelerated plate culture tests have been proposed for the rapid detection of pathogens. Lab-on-chip (LOC) devices and microfluidics are miniaturized devices that can enable faster, easier, and at the point of interest analysis. Nowadays, methods such as PCR are often coupled with microfluidics, providing new LOC devices that can replace or complement the standard methods by offering highly sensitive, fast, and on-site analysis. This review's objective is to present an overview of recent advances in LOCs used for the identification of the most prevalent foodborne and waterborne pathogens that put consumer health at risk. In particular, the paper is organized as follows: first, we discuss the main fabrication methods of microfluidics as well as the most popular materials used, and then we present recent literature examples for LOCs used for the detection of pathogenic bacteria found in water and other food samples. In the final section, we summarize our findings and also provide our point of view on the challenges and opportunities in the field.

Abstract:The rise of antimicrobial resistance (AMR) in infectious pathogens presents a significant global health challenge, necessitating rapid and accurate diagnostic tools for timely intervention. This paper discusses the potential of microfluidic chips as innovative solutions for the on-site detection of AMR. Microfluidic technology, characterized by its ability to manipulate small volumes of fluids within miniaturized devices, offers several advantages in the context of AMR diagnostics, including speed, portability, and the ability to perform multiple assays simultaneously. The integration of microfluidic chips with molecular diagnostics, such as polymerase chain reaction (PCR) and nucleic acid amplification tests (NAATs), enables the rapid identification of resistance genes and mutations directly from clinical samples, often within a matter of hours. This capability is critical in both clinical settings and remote or resource-limited areas, where traditional laboratory-based diagnostics may be inaccessible or too slow to influence treatment decisions effectively. Moreover, microfluidic chips can be designed to detect a wide range of pathogens and resistance mechanisms, offering a versatile platform for comprehensive AMR surveillance. The portability and ease of use of these devices also empower healthcare workers and public health officials to conduct on-site testing, thereby accelerating the decision-making process and improving patient outcomes. Despite these advantages, challenges remain, including the need for standardization, regulatory approval, and ensuring the accuracy and sensitivity of the devices in diverse settings. Abstract:Molecular diagnostics is one of the most important tools currently in use for clinical pathogen detection due to its high sensitivity, specificity, and low consume of sample and reagent is keyword to low cost molecular diagnostics. In this paper, a sensitive DNA isothermal amplification method for fast clinical infectious diseases diagnostics at aM concentrations of DNA was developed using a polycarbonate (PC) microfluidic chip. A portable confocal optical fluorescence detector was specifically developed for the microfluidic chip that was capable of highly sensitive real-time detection of amplified products for sequence-specific molecular identification near the optical diffraction limit with low background. The molecular diagnostics of Listeria monocytogenes with nucleic acid extracted from stool samples was performed at a minimum DNA template concentration of 3.65 aM, and a detection limit of less than five copies of genomic DNA. Contrast to the general polymerase chain reaction (PCR) at eppendorf (EP) tube, the detection time in our developed method was reduced from 1.5 h to 45 min for multi- target parallel detection, the consume of sample and reagent was dropped from 25 mu L to 1.45 mu L. This novel microfluidic chip system and method can be used to develop a micro total analysis system as a clinically relevant pathogen molecular diagnostics method via the amplification of targets, with potential applications in biotechnology, medicine, and clinical molecular diagnostics.

Abstract:Microfluidic chip exhibit a great promising development in clinical diagnosis and disease screening due to their advantages of precise controlling of fluid flow,requirement of mini amount sample,rapid reaction speed and convenient integration.A lot of demonstrations on the diagnostic applications related to genes,proteins,and cells have been reported because of their advantages associated with miniaturization and automation.Here,the applications and developments of on-chip nucleic acid amplification and analysis,protein analysis and detection,cell selection and cell drug screening were discussed.Microfluidic chip can provide an easy integration platform for biomarkers in a high throughput and accurate detection.
