Abstract:Objective To analyze the effects of norepinephrine(NE) on the proliferation and migration of endothelial progenitor cells(EPCs) and the related mechanism.Methods NE,adrenoceptor antagonist,and MAPK signal pathway blocker of various concentrations were used to treat peripheral EPCs derived from healthy adults.The proliferation potential,migration capacity and activation of ERK1/2 were assessed after different treatments.Results NE increased the proliferation potential of EPCs in a dose-dependent manner.The number of EPCs increased by(48.3±23.3)%,(70.5±35.6)%,(82.4±14.9)% and(100.3±48.1)% after treatment with NE at 0.01 μmol/L,0.1 μmol/L,1 μmol/L and 10 μmol/L,respectively.Addition of alpha adrenoceptor antagonist phentolamine,selective beta 2 adrenoceptor antagonist I127,JNK blocker SP600125 and ERK1/2 blocker A6355 could block the above effects of NE,and beta 1 adrenoceptor antagonist metoprolol and p38 blocker PD169318 failed to block the effects of NE.NE at 10 μmol/L significantly promoted the migration of EPCs(P0.05).These effects could be blocked by addition of phentolamine(10 μmol/L) and I127(10 μmol/L),but not by addition of metoprolol.NE(0.1,1 and 10 μmol/L) activated ERK1/2 pathway in a dose-dependent manner,which could also be blocked by phentolamine and I127,but not by metoprolol.Conclusion NE can increase the proliferation potential and migration capacity of EPCs via activating ERK1/2 pathway with alpha and beta 2 adrenoceptor.

Norepinephrine Stimulates Mobilization of Endothelial Progenitor Cells after Limb Ischemia

Abstract:OBJECTIVE:During several pathological processes such as cancer progression, thermal injury, wound healing and hindlimb ischemia, the mobilization of endothelial progenitor cells (EPCs) mobilization was enhanced with an increase of sympathetic nerve activity and norepinephrine (NE) secretion, yet the cellular and molecular mechanisms involved in the effects of NE on EPCs has less been investigated.METHODS AND RESULTS:EPCs from BMs, peripheral circulation and spleens, the VEGF concentration in BM, skeletal muscle, peripheral circulation and spleen and angiogenesis in ischemic gastrocnemius were quantified in mice with hindlimbs ischemia. Systemic treatment of NE significantly increased EPCs number in BM, peripheral circulation and spleen, VEGF concentration in BM and skeletal muscle and angiogenesis in ischemic gastrocnemius in mice with hind limb ischemia, but did not affair VEGF concentration in peripheral circulation and spleen. EPCs isolated from healthy adults were cultured with NE in vitro to evaluate proliferation potential, migration capacity and phosphorylations of Akt and eNOS signal moleculars. Treatment of NE induced a significant increase in number of EPCs in the S-phase in a dose-dependent manner, as well as migrative activity of EPCs in vitro (p<0.05). The co-treatment of Phentolamine, I127, LY294002 and L-NAME with NE blocked the effects of NE on EPCs proliferation and migration. Treatment with NE significantly increased phosphorylation of Akt and eNOS of EPCs. Addition of phentolamine and I127 attenuated the activation of Akt/eNOS pathway, but metoprolol could not. Pretreatment of mice with either Phentolamine or I127 significantly attenuated the effects of NE on EPCs in vivo, VEGF concentration in BM, skeletal muscle and angiogenesis in ischemic gastrocnemius, but Metoprolol did not.CONCLUSION:These results unravel that sympathetic nervous system regulate EPCs mobilization and their pro-angiogenic capacity via α adrenoceptor, β 2 adrenoceptor and meanwhile Akt/eNOS signaling pathway.
Effects of Astragalus on Number and Activity of Peripheral Endothelial Progenitor Cells

Abstract:Objective To investigate the influence of astragalus on the number,activity,and inducible nitric oxide synthase(iNOS) of peripheral endothelial progenitor cells (EPCs). Methods Total mononuclear cells (MNCs) were isolated from peripheral blood by Ficoll density gradient centrifugation,and then the cells were inoculated on fibronectin-coated culture dishes. After 7 days of culturing,attached cells were stimulated with astragalus solution (in a series of final concentrations of 10,20,50 and 100 g/L)for 24 hours. Meanwhile,the attached cells in the control group and astragalus 50 g/L group were cultured for 6,12,24,48 h respectively. EPCs were identified by laser confocal microscopy and were counted under inverted fluorescence microscope. The proliferation and migration of EPCs were assayed with MTT assay and modified Boyden chamber assay,respectively. EPCs adhesion assay was performed by re-inoculation of EPCs on fibronectin-coated dishes,and then adherent cells were counted. In vitro vasculogenesis activity of EPCs was assayed by in vitro vasculogenesis kit. iNOS was assayed by western blot. Results Incubation of isolated human MNCs with astragalus increased the number of EPCs,as well as their proliferative,migratory,adhesive and invitro vasculogenesis capacity in a concentration and time dependent manner. Astragalus also increased iNOS content of EPCs. Conclusion Astragalus increases the number of EPCs,and enhances the proliferative,migratory,adhesive and in vitro vasculogenesis capacity of EPCs. Its mechanism is related to the increase of iNOS by astragalus.
17 Beta-Estradiol Induces The Proliferation Of Hematopoietic Stem Cells By Promoting The Osteogenic Differentiation Of Mesenchymal Stem Cells

Abstract:The process of hematopoiesis is associated with hematopoietic stem cells (HSCs) and the hematopoietic microenvironment. Osteoblasts, derived from mesenchymal stem cells (MSCs), are one of the most important components in the hematopoietic microenvironment. Osteoblasts secrete a variety of cytokines including interleukin-6 (1-6) and granulocyte-macrophage colony-stimulating factor (GM-CSF), thereby regulating the biological activities of HSCs. It has been shown that hematopoiesis dysfunction can be induced by estrogen-deficiency through the exhaustion of HSCs. However, the effect of estrogen on the proliferation of HSCs is not fully understood. The aim of this study was to investigate the role of estradiol in the process of hematopoiesis, especially regarding the proliferation of HSCs in vitro. Bone marrow-derived MSCs and HSCs were isolated from 3-month-old female Sprague-Dawley rats. Mineralization ability and osteocalcin assays demonstrated that treatment with 17 beta-estradiol (E2) significantly enhanced the osteogenic differentiation of MSCs. HSCs and MSCs were then cocultured with or without E2 treatment. Colony forming assays demonstrated that E2 increased the number of colony forming units-granulocyte/ macrophage in a dose-dependent manner when HSCs were co-cultured with MSCs in Osteogenic Medium that is suitable for the in vitro osteogenic differentiation. Further, increased concentrations of GM-CSF and IL-6 were detected by enzyme linked immunosorbent assay (ELISA). These results indicate that E2 induces the proliferation of HSCs, which depends on the promotion of osteogenic differentiation of MSCs, and that process is mediated by both GM-CSF and IL-6.

Abstract:Adiponectin (ADPN) exerts various cellular and metabolic functions by activating signaling pathways, including extracellular signal-regulated protein kinases 1 and 2 (ERK1/2) pathways, the protein kinase B (Akt) pathway, and the p38 mitogen-activated protein kinase (MAPK) pathway. However, generating functional recombinant human adiponectin (hADPN) in bacterial or mammalian cells is challenging. Although ADPN agonist peptides have been developed, problems like stability, solubility, and affinity for receptors remain. Recently, a genome-edited chicken bioreactor system was established, ensuring efficient ADPN production with optimal post-transcriptional modifications. We assessed the ability of egg white (EW)-derived hADPN, commercial hADPN, various ADPN agonist peptides, and globular ADPN on activation of the ERK1/2, Akt, and p38 MAPK pathways. EW-derived hADPN, abundant in hexamers and high molecular weight multimers, significantly phosphorylated ERK1/2 in serum-starved HEK293 cells after 15 min of treatment. Comparative analysis revealed that EW-derived hADPN and commercial hADPN induced greater phosphorylation of ERK1/2, Akt, and p38 MAPK than ADPN agonist peptides and globular ADPN, with EW-derived hADPN showing the highest activation. In summary, the finding that EW-derived hADPN strongly activates the ERK1/2, Akt, p38 MAPK signaling pathways highlights that an ADPN production system based on genome-edited chickens is an advantageous alternative to existing methods.
[High-Dose Neuropeptide Y Inhibits Proliferation and Promotes Differentiation of Human Adipose-Derived Stem Cells].

Abstract:OBJECTIVETo investigate the impact of neuropeptide Y (NPY) at different concentrations on the proliferation and differentiation of human adipose-derived stem cells (hADSCs).METHODSThe hADSCs were separated and incubated with NPY at a range of concentrations 10(-6)-10(-15) mol/L. MTT assay was used to detect cell proliferation. Following the treatment of complete medium, NPY and insulin separately, cell differentiation was observed by Oil red O staining. In addition, Western blotting was performed to exam the levels of peroxisome proliferator-activated receptor-γ (PPARγ), CCAAT/enhancer binding protein alpha (C/EBPα), cell death-inducing dFF45-like effector c (Cidec) and receptor interacting protein 140 (RIP140).RESULTSMTT assay revealed that 10(-11)-10(-15) mol/L NPY stimulated the proliferation of hADSCs, while high-dose NPY (10(-6)-10(-10) mol/L) inhibited the cell proliferation. NPY (10(-7), 10(-9), 10(-11) mol/L) induced the differentiation of hADSCs. The expression levels of related adipocyte markers (PPARγ, C/EBPα), mature white adipose tissue specific markers (Cidec, RIP140) increased at the presence of NPY (10(-7), 10(-9), 10(-11) mol/L).CONCLUSIONLow-dose NPY could stimulate the proliferation of hADSCs, and high-dose NPY could inhibit the proliferation and promote the differentiation of hADSCs.

Abstract:Ethnopharmacological relevance: The roots of Actinidia eriantha Benth have been used clinically to treat a variety of cancers in traditional Chinese medicine. The polysaccharide from this drug (AEPS) was previously reported to be a potential antitumor agent with immunomodulatory activity. However, the mechanisms of its antitumor action in immunomodulation have not yet been well-defined. Aim of the study: To investigate the effects of AEPS on the phenotypic and functional maturation of dendritic cells and to explore the intracellular signaling mechanisms of its antitumor action in the immunomodulation. Materials and methods: The effects of AEPS on the phagocytic activity, expression of surface molecules, mRNA and protein expression levels of cytokines and chemokines in mouse bone -marrow derived dendritic cells (BMDCs) were determined by flow cytometry, qRT-PCR and ELISA, respectively. The transcriptional profile induced by AEPS was established using oligonucleotide microarray, and Ingenuity Pathway Analysis (IPA) was used to identify potential signaling pathways. Western blotting, neutralization experiments and inhibition assay were performed to confirm signaling pathway involved in maturation of DCs induced by AEPS. Furthermore, we discussed the downstream effects of the action of AEPS using clustering, network and pathway mapping approaches. Results: AEPS could significantly reduced phagocytic activity, promoted expression of accessory and co-stimulatory molecules, and up-regulated the mRNA and protein expression levels of cytokines and chemokines in BMDCs. Microarray assay revealed that AEPS induced significantly differential expression of 452 genes including up-regulated cytokines (IL-6, IL-1 beta, TNF-alpha, IL-10, IL-12p40, IFN-beta and IFN-gamma), chemokines (MIP-1 alpha, MIP-1 beta, CCLS, MDC and MCP-1), transcription factors (STAT1, STAT2, STAT5b, IRF1 and IRF7) and pattern recognition receptors (TLR3, DDX58, DHX58 and IFIH1) in the BMDCs. AEPS-induced production of TNF-alpha and IL-12p40 from BMDCs was inhibited by antibodies against TLR2 and TLR4. Furthermore, AEPS induced the phosphorylation of NF-kappa B p65 in a time-dependent manner, and BAY 11-7082, an inhibitor of NF-kappa B, remarkably suppressed the production of cytokines induced by AEPS. Conclusion: AEPS triggers the phenotypic and functional maturation of DCs via TLR2/4 and NF-kappa B signaling pathway, resulting in augmented antitumor immune responses. Our results suggested that AEPS might be helpful in potentiating the efficiency of DC-based cancer immunotherapy. This study further expanded current knowledge on the mechanisms of antitumor action of AEPS.
Epidermal Growth Factor-Induced Proliferation of Chicken Primordial Germ Cells: Involvement of Calcium/Protein Kinase C and NFKB11

Abstract:Epidermal growth factor (EGF) has been shown to stimulate survival in diverse cells in vitro. In the present study, the effects of EGF and the EGF-related signaling pathway on proliferation of chicken primordial germ cells (PGCs) were investigated. Results showed that EGF (10-100 ng/ml) increased the number and area of PGC colonies in a time- and dose-dependent manner. EGF also activated PKC, a process that was inhibited by AG1478 (an EGFR tyrosine kinase inhibitor) and ethyleneglycol-bis-(beta-aminoethyl ether)-N,N'-tetraacetic acid (EGTA; an intracellular Ca(2+) chelator). In addition, the degradation of NFKBIA and NFKB1 (p65) translocation was observed after EGF treatment, which was significantly blocked by pretreatment with AG1478, EGTA, H(7), or SN50 (NFKB1-specific inhibitor). Furthermore, we found that EGF-induced cell proliferation was significantly attenuated by AG1478, EGTA, H(7), and SN50, respectively. On the other hand, inhibition of EGFR, Ca(2+)/PKC, or NFKB1 abolished the EGF-stimulated increase in the expression of cyclins CCND1 and CCNE1, cyclin-dependent kinase 6 (CDK6), CDK2, and BCL2, and restored the EGF-induced inhibition of BAX expression and caspase 3/9 activity, indicating that EGFR, PKC, and NFKB1 signaling cascades were involved in EGF-stimulated DNA synthesis and antiapoptosis action. In conclusion, EGF stimulated proliferation of chicken PGCs via activation of Ca(2+)/PKC involving NFKB1 signaling pathway. These observations suggest that EGF signaling is important in regulating germ cell proliferation in the chicken embryonic gonad.
Changing Expression Profiles and Inclination to Competing Endogenous RNA Networks on MAPK Signaling Pathways of Human Adipose-Derived Stem Cells in a Direct Current Electric Field

Abstract:Adipose-derived stem cells (ADSCs) are an abundant cell source and provide an easy way to harvest mesenchymal stem cells, which are the focus of considerable attention in regenerative medicine. Electric fields (EF) play roles in many biological events and have been reported to promote cell proliferation, migration, and differentiation. In this study, ADSCs were treated with a direct current electric field (DCEF) of either 0 (control group) or 300 mV/mm (EF group) for six hours. RNA screening and analysis revealed that 66, 164, 26, and 1310 circRNAs, lncRNAs, miRNAs, and mRNAs, respectively, were differentially expressed in the DCEF-treated ADSCs compared with untreated ADSCs. Differentially expressed mRNAs were enriched in the MAPK signaling pathway, TNF signaling pathway, and some other pathways. ANXA1, ERRFI1, JAG1, EPHA2, PRR9, and H2AFY2 were related to the keratinocyte differentiation process. Competing endogenous RNA (ceRNA) networks were constructed on the basis of genes in the MAPK signaling pathway. Twenty-one RNAs in the above networks were randomly chosen, and their expression was validated using qRT-PCR, which showed the same expression trends as the RNA sequencing analysis. The MAPK signaling pathway is of great importance in the ADSC processes that occur in a DCEF, including keratinocyte differentiation. Several ceRNAs may participate in the regulation of MAPK signaling. This study may give new insight into the proliferation, migration, and differentiation of ADSCs, which will be valuable for tissue engineering and regenerative medicine.
Effects of recombinant human erythropoietin on migration and adhesion of human mesenchymal stem cells

Abstract:Background: Previous studies have demonstrated that erythropoietin can promote the proliferation, differentiation and adhesion of endothelial progenitor cells. Objective: To investigate the effects and related cell signaling pathways of recombinant human erythropoietin on migration and adhesion of human mesenchymal stem cells. Methods: After being cultured in vitro, passage 6 human mesenchymal stem cells were treated with recombinant human erythropoietin. Recombinant human erythropoietin, phosphorylation of ERK 1/2, p38MAPK and PI3K/Akt were detected by western blot. After 24 hours of pretreatment with 1U/mL recombinant human erythropoietin, cell migration assay was performed in Transwell chambers, and adhesion assay was performed by plastic dishes. Results and Conclusion: Recombinant human erythropoietin could increase phosphorylation of PI3K/Akt pathway of human MSCs, but reduce phosphorylation of p38MAPK. It had no obvious effect on ERK1/2 pathway and total proteins of Akt and p38MAPK. The number of cells crossing the filter was significantly increased in the recombinant human erythropoietin group (P < 0.01). This effect was completely inhibited by concomitant incubation with Ly294002. After treated with recombinant human erythropoietin, adherent cells increased significantly (P < 0.01). Neither Ly294002 nor anisomycin could inhibit this effect. Recombinant human erythropoietin could increase migratory and adhesive capacities of human mesenchymal stem cells. Activation of PI3K/Akt pathway was involved in the effect of recombinant human erythropoietin on cell migration. The effect of rhEPO on adhesion capacity had nothing to do with PI3K/Akt and p38MAPK pathway.
Effects of enamel matrix derivative on the proliferation and osteogenic differentiation of human gingival mesenchymal stem cells

Abstract:IntroductionGingiva-derived mesenchymal stem cells (GMSCs) have recently been harvested and applied for rebuilding lost periodontal tissue. Enamel matrix derivative (EMD) has been used for periodontal regeneration and the formation of new cementum with inserting collagen fibers; however, alveolar bone formation is minimal. Recently, EMD has been shown to enhance the proliferation and mineralization of human bone marrow mesenchymal stem cells. Because the gingival flap is the major component to cover the surgical wound, the effects of EMD on the proliferation and mineralization of GMSCs were evaluated in the present study.MethodsAfter single cell suspension, the GMSCs were isolated from the connective tissues of human gingiva. The colony forming unit assay of the isolated GMSCs was measured. The expression of stem cell markers was examined by flow cytometry. The cellular telomerase activity was identified by polymerase chain reaction (PCR). The osteogenic, adipogenic and neural differentiations of the GMSCs were further examined. The cell proliferation was determined by MTS assay, while the expression of mRNA and protein for mineralization (including core binding factor alpha, cbfα-1; alkaline phosphatase, ALP; and osteocalcin, OC; ameloblastin, AMBN) were analyzed by real time-PCR, enzyme activity and confocal laser scanning microscopy.ResultsThe cell colonies could be easily identified and the colony forming rates and the telomerase activities increased after passaging. The GMSCs expressed high levels of surface markers for CD73, CD90, and CD105, but showed low expression of STRO-1. Osteogenic, adipogenic and neural differentiations were successfully induced. The proliferation of GMSCs was increased after EMD treatment. ALP mRNA was significantly augmented by treating with EMD for 3 hours, whereas AMBN mRNA was significantly increased at 6 hours after EMD treatment. The gene expression of OC was enhanced at the dose of 100 μg/ml EMD at day 3. Increased protein expression for cbfα-1 at day 3, for ALP at day 5 and 7, and for OC at week 4 after the EMD treatments were observed.ConclusionsHuman GMSCs could be successfully isolated and identified. EMD treatments not only induced the proliferation of GMSCs but also enhanced their osteogenic differentiation after induction.

Abstract:Background: Parkinson's disease (PD) is the second most common neurodegenerative disorder affecting the senile population with manifestation of motor disability and cognitive impairment. Reactive oxygen species (ROS) is implicated in the progression of oxidative stress-related apoptosis and cell death of the midbrain dopaminergic neurons. Its interplay with mitochondrial functionality constitutes an important aspect of neuronal survival in the perspective of PD. Edible bird's nest (EBN) is an animal-derived natural food product made of saliva secreted by swiftlets from the Aerodamus genus. It contains bioactive compounds which might confer neuroprotective effects to the neurons. Hence this study aims to investigate the neuroprotective effect of EBN extracts in the neurotoxin-induced in vitro PD model. Methods: EBN was first prepared into pancreatin-digested crude extract and water extract. In vitro PD model was generated by exposing SH-SY5Y cells to neurotoxin 6-hydroxydopamine (6-OHDA). Cytotoxicity of the extracts on SH-SY5Y cells was tested using MTT assay. Then, microscopic morphological and nuclear examination, cell viability test and ROS assay were performed to assess the protective effect of EBN extracts against 6-OHDA-induced cellular injury. Apoptotic event was later analysed with Annexin V-propidium iodide flow cytometry. To understand whether the mechanism underlying the neuroprotective effect of EBN was mediated via mitochondrial or caspase-dependent pathway, mitochondrial membrane potential (MMP) measurement and caspase-3 quantification were carried out. Results: Cytotoxicity results showed that crude EBN extract did not cause SH-SY5Y cell death at concentrations up to 75 μg/ml while the maximum non-toxic dose (MNTD) of water extract was double of that of crude extract. Morphological observation and nuclear staining suggested that EBN treatment reduced the level of 6-OHDA-induced apoptotic changes in SH-SY5Y cells. MTT study further confirmed that cell viability was better improved with crude EBN extract. However, water extract exhibited higher efficacy in ameliorating ROS build up, early apoptotic membrane phosphatidylserine externalization as well as inhibition of caspase-3 cleavage. None of the EBN treatment had any effect on MMP. Conclusions: Current findings suggest that EBN extracts might confer neuroprotective effect against 6-OHDA-induced degeneration of dopaminergic neurons, particularly through inhibition of apoptosis. Thus EBN may be a viable nutraceutical option to protect against oxidative stress-related neurodegenerative disorders such as PD.

Abstract:Edible bird's nest (EBN) is a popular delicacy among the Chinese diaspora associated with various health claims. This study investigated the anti-inflammatory effects of white-EBN acid hydrolysate on tumour necrosis factor-alpha (TNF-α) and nitric oxide (NO) generation, while examining its cytotoxic effect on a lipopolysaccharide-stimulated RAW 264.7 macrophage cell line. Processed commercial EBN and unprocessed raw EBN collected from different geographical zones and at different harvesting seasons were studied. The cytotoxic effect was determined by cell viability assessed by MTS assay, while NO production was determined by the Griess reaction and TNF-α concentration was measured using an ELISA kit. The results showed that both commercial and raw EBN inhibited TNF-α and NO generation, the highest inhibition 58% and 63%, respectively without significant cytotoxic effect was brought about by raw EBN from the South zone. The results suggest that EBN may possess anti-inflammatory properties that should be further studied.

Exendin-4 Promotes Proliferation of Adipose-Derived Stem Cells Through PI3K/Akt-Wnt Signaling Pathways.

Abstract:Adipose-derived stem cell (ADSC) transplantation has emerged as a potential tool for the treatment of cardiovascular disease and skin wounds. However, with a limited renewal capacity and the need for mass cells during the engraftment, strategies are needed to enhance ADSC proliferative capacity. In this study, we explored the effects of Exendin-4, a glucagon-like peptide-1 analog, on the growth of ADSCs, focusing in particular on phosphatidylinositol 3-kinase (PI3K)-protein kinase B (Akt) and Wnt signaling pathways. Firstly, ADSCs were isolated and cultured in vitro. Then, flow cytometry demonstrated that ADSCs were positive for CD44, CD90 and CD29 but negative for CD31, CD34, and CD45. Exendin-4 (0-200 nM) treatment increased ADSC proliferation. In order to examine specific signaling pathways, a western blotting assay was performed. Our results demonstrate that after treated with 50 nM Exendin-4 for 48 h, the phosphorylation of PI3K, Akt, and GSK3 beta were increased and phosphorylation of beta-catenin was decreased. From these results, we concluded that PI3K-Akt and Wnt-beta-catenin signaling pathways mediate Exendin-4 induced ADSC proliferation, the function of which might contribute to the regulation of ADSC proliferation. Our findings provided new insights into the function of the mechanisms underlying Exendin-4 of ADSCs.
Enamel Matrix Derivative Enhances the Odontoblastic Differentiation of Dental Pulp Stem Cells via Activating MAPK Signaling Pathways

Abstract:The odontoblastic differentiation of dental pulp stem cells (DPSCs) contributes to pulp-dentin regeneration. Enamel matrix derivative (EMD) is considered to be a critical epithelial signal to induce cell differentiation during odontogenesis and has been widely applied to clinical periodontal tissue regeneration. The purpose of this study was to explore the effect of EMD on DPSCs proliferation and odontoblastic differentiation, as well as the underlying mechanisms. We conducted in vitro and in vivo researches to get a comprehensive understanding of EMD. In vitro phase: cell proliferation was assessed by a cell counting kit-8 (CCK-8) assay; then, alkaline phosphatase (ALP) activity and staining, alizarin red staining, real-time RT-PCR, and western blot analysis were conducted to determine the odontoblastic potential and involvement of MAPK signaling pathways. In vivo phase: after ensuring the biocompatibility of VitroGel 3D-RGD via scanning electron microscopy (SEM), the hydrogel mixture was subcutaneously injected into nude mice followed by histological and immunohistochemical analyses. The results revealed that EMD did not interfere with DPSCs proliferation but promoted the odontoblastic differentiation of DPSCs in vitro and in vivo. Furthermore, blocking the MAPK pathways suppressed the EMD-enhanced differentiation of DPSCs. Finally, VitroGel 3D-RGD could well support the proliferation, differentiation, and regeneration of DPSCs. Overall, this study demonstrates that EMD enhances the odontoblastic differentiation of DPSCs through triggering MAPK signaling pathways. The findings provide a new insight into the mechanism by which EMD affects DPSCs differentiation and proposes EMD as a promising candidate for future stem cell therapy in endodontics.

Abstract:AIM:To evaluate the effects of epidermal growth factor (EGF) on transforming growth factor-beta1 (TGF-β1)-induced epithelial-mesenchymal transition (EMT) in human corneal epithelial cells (HCECs).METHODS:HCECs were cultured and treated with TGF-β1 for establishing the model of EMT in vitro. Biological effect of EGF on TGF-β1-induced EMT was evaluated. Proteins and mRNAs expression changes of E-cadherin, N-cadherin and Fibronectin (EMT-relative markers) after TGF-β1 or TGF-β1 combined EGF treatment were detected by Western blot and RT-PCR, respectively. Viability and migration of HCECs were measured by CCK-8, transwell cell migration assay and cell scratch wound healing assay. Activation of Smad2, ERK, p38, JNK and Akt signaling pathways were evaluated by Western blot. Inhibitors of relevant signaling pathways were added to the HCECs to explore the key signal mechanism.RESULTS:With treatment of TGF-β1 only, three EMT-relative proteins and mRNA expression showed that EMT up-regulated in a concentration-dependent and time-dependent manner, with significantly decreasing cell viability (TGF-β1≥5 ng/mL, P<0.05) and increasing cell migration (TGF-β1≥5 ng/mL, P<0.01). The phosphorylation of Smad2 and p38 was a key process of TGF-β1-induced EMT. Meanwhile, EMT-relative proteins and mRNA expression showed that EGF inhibited TGF-β1-indued EMT, with significantly increasing cell viability (EGF≥10 ng/mL, P<0.01). It was noteworthy that EGF significantly enhanced cell migration although EMT was inhibited (EGF≥10 ng/mL, P<0.01), and the blockage of p38 (by SB202190, a p38 inhibitor) was a potential mechanism of this phenomenon.CONCLUSION:EGF inhibits TGF-β1-induced EMT via suppressive p38, and promotes cells proliferation and migration in a non-EMT process by inhibiting p38 pathway.
Unveiling the Cytotoxic and Apoptosis-Inducing Abilities of An Edible Bird's Nest Extract in Human Hepatocellular Carcinoma HUH-7 Cells

Abstract:Edible bird's nest (EBN) is renowned for diverse pharmacological properties; however, its ability to cause cancer cell death has not been found in recent studies. This study investigates the phytochemical composition of EBN ethanol extract (EEE), the cytotoxic and apoptosis-inducing effects of the extract on human hepatocellular carcinoma Huh-7 cells. Qualitative and quantitative tests showed that proteins and peptides were abundant in the EEE. In the MTT assay, the extract exhibited cytotoxic activity against Huh-7 cells (IC50 = 58.57 ± 2.66 μg/mL, 53.15 ± 3.07 μg/mL, and 55.27 ± 1.85 μg/mL at 24 h, 48 h, and 72 h of incubation, respectively). Annexin-V/PI staining suggested that the extract provoked apoptosis in Huh-7 cells by deregulating the expression patterns of apoptotic markers including caspase 3, caspase 8, caspase 9, and survivin. These findings demonstrate the potential of the EEE as a promising anticancer candidate for further studies.
