Abstract:Different sources of nitrogen pose diverse effects to algal community, but the mechanism of inhibitory effects of nitrogen sources on freshwater diatoms is not fully understood. The purpose of this study was to compare biomass, photosynthetic activity, and morphological structure of three common freshwater diatoms ( Cyclotella meneghiniana , Nitzschia sp., and Gomphonema parvulum ) under different nitrogen sources (NO 3 − or NH 4 + ). The sorption characteristic of each diatom was investigated, and chlorophyll a (Chl-a) content and oxygen evolution rate were analyzed to investigate stress of different nitrogen sources on each diatom in the batch experiments. Ammonium lowered the growth rate of C. meneghiniana and Nitzschia sp. when it was supplied in addition to growth-saturating nitrate concentrations, suggesting a combined effect of inhibition of nitrate uptake and direct ammonium stress. Oxygen evolution rate of Nitzschia sp. showed that the direct ammonium stress on the photosynthetic activity can be alleviated by coexistence of nitrate in the nitrogen enriched treatment, but not for C. meneghiniana and G. parvulum , which may be caused by a different nitrate transporter system within algal cells. Transmission electron microscopy was used to assess the toxicity of ammonium on ultrastructural chloroplast of each diatom. Ultrastructural changes in chloroplasts showed undefined electron-dense granules and lipid droplets, but the membrane integrity of cell was maintained, suggesting an adaptation to adjustment to ammonia stress. Results showed that Cyclotella meneghiniana and Nitzschia sp. were more sensitive to ammonium stress than Gomphonema parvulum on growth, but the mechanism remains unclear.
Physiological Effect of Vallisneria Natans L.under Different Concentrations of Ammonia

Abstract:The effects of biomass,nitrogen in free amino acids,chlorophyll,soluble proteins and malondialdehyde(MDA),intensities of O2-and antioxidant enzyme activities in the leaves of Vallisneria natans L.exposed to different concentrations of NH4+-N(0.02,0.05,0.10,0.30,0.60,1.00,3.00mg/L) after 14 days were investigated.No significant changes were observed for the biomass of leaves in the studied exposing level,while the physiological signals showed different trends.Intensities of O2-increased at the lowest concentration of ammonia(0.02mg/L).The nitrogen contents in the free amino acids started to increase significantly from 0.30mg/L NH4+-N.When the NH4+-N reached 0.60mg/L,SOD activities were significantly induced,indicating oxidative stress to the plant without oxidative damage.When the ammonia concentrations were higher than 1.00mg/L,the signal intensities of free radicals and activities of SOD and POD significantly increased,while the contents of chlorophyll and soluble protein decreased,indicating oxidative damage might happen.Generally,the plants grew well when the NH4+-N concentration was below 0.60mg/L and stress and growth disturbance happened when it was above 1.00mg/L.Among all those physiological parameters measured,the nitrogen content in the free amino acids in the plant leaves was the most sensitive indicator.Results also suggested that deficiency of nutrient at a low level of ammonia may cause the induction of free radicals in submerged plant leaves.

Abstract:Although excess ammoniacal-nitrogen (NH4+-N) results in the disturbance of various important biochemical and physiological processes, a detailed study on the effects of NH4+-N stress on the photosynthesis and global changes in protein levels in submerged macrophytes is still lacking. Here, the changes of excess NH4+-N on physiological parameters in Hydrilla verticillata (L.f.) Royle, a submerged macrophyte were investigated, including the contents of photosynthetic pigments, soluble sugars, net photosynthesis and respiration, glutamine synthetase (GS) and glutamate synthase (GOGAT) activities, chloroplast ultrastructure, chloroplast reactive oxygen species (ROS) accumulation and protein levels. Our results showed that the net photosynthetic rate and pigment content reached maximum values when the plants were treated with 1 and 2 mg L-1 NH4+-N, respectively, and decreased at NH4+-N concentrations at 5, 10, 15 and 20 mg L-1. This decrease might be caused by ROS accumulation. Compared that in 0.02 mg L-1 NH4+-N as a control, ROS generation in chloroplasts significantly increased in the presence of more than 2 mg L-1 NH4+-N. Consistently, the damages caused by over-accumulated ROS were observed in chloroplast ultrastructure, showing a loose thylakoid membranes and swollen grana/stroma lamellae. Furthermore, through proteomic analysis, we identified 91 differentially expressed protein spots. Among them, six proteins involved in photosynthesis decreased in abundance in response to excess NH4+-N. Surprisingly, the abundance of all the identified proteins that were involved in nitrogen assimilation and amino acid metabolism tended to increase under excess NH4+-N compared with the control, suggestive of the imbalanced carbon and nitrogen (C-N) metabolisms. In support, activated GS and GOGAT cycle was observed, evidenced by higher activities of GS and GOGAT enzymes. To our knowledge, this work is the first description that excess NH4+-N results in chloroplast ultrastructural damages and the first proteomic evidence to support that excess NH4+-N can lead to a decline in photosynthesis and imbalance of C-N metabolism in submerged macrophytes.
Effects of the ammonium stress on photosynthesis and ammonium assimilation in submerged leaves of Ottelia cordata - an endangered aquatic plant

Abstract:Although ammonium (NH4+-N) is an important nutrient for plants, increases in soil nitrogen (N) input and atmospheric deposition have made ammonium toxicity a serious ecological problem. In this study, we explored the effects of NH4+-N stress on the ultrastructure, photosynthesis, and NH4+-N assimilation of Ottelia cordata (Wallich) Dandy, an endangered heteroblastic plant native to China. Results showed that 15 and 50 mg L-1 NH4+-N damaged leaf ultrastructure and decreased the values of maximal quantum yield (Fv/Fm), maximal fluorescence (Fm), and relative electron transport rate (rETR) in the submerged leaves of O. cordata. Furthermore, when NH4+-N was ≥ 2 mg L-1, phosphoenolpyruvate carboxylase activity (PEPC) and soluble sugar and starch contents decreased significantly. The content of dissolved oxygen in the culture water also decreased significantly. The activity of the NH4+-N assimilation enzyme glutamine synthetase (GS) significantly increased when NH4+-N was ≥ 10 mg L-1 and NADH-glutamate synthase (NADH-GOGAT) and Fd-glutamate synthase (Fd-GOGAT) increased when NH4+-N was at 50 mg L-1. However, the activity of nicotinamide adenine dinucleotide-dependent glutamate dehydrogenase (NADH-GDH) and nicotinamide adenine dinucleotide phosphate-dependent glutamate dehydrogenase (NADPH-GDH) did not change, indicating that GS/GOGAT cycle may play an important role in NH4+-N assimilation in the submerged leaves of O. cordata. These results show that short-term exposure to a high concentration of NH4+-N is toxic to O. cordata.
Combined Effects of Light Reduction and Ammonia Nitrogen Enrichment on the Submerged Macrophyte Vallisneria Natans

Abstract:The decline of submerged plants resulting from low light and high ammonia nitrogen (ammonia-N) has become a serious problem worldwide. In the present study, three levels of ammonia-N concentrations (0, 3 and 6mgL–1) and four levels of light intensity (control, 15, 2.5 and 0.75% underwater light) were designed to investigate the combined effects of low light and high ammonia-N stress on the submerged plant Vallisneria natans. The effects of low light and ammonia-N were examined by measuring the relative growth rate (RGR), chlorophyll content and superoxide dismutase (SOD) and peroxidase (POD) activity in response to the stressors. The decline in RGR and increase in SOD and POD activity in high ammonia-N water were more significant than under low light conditions, indicating that the stress imposed on submerged plants due to ammonia-N enrichment is stronger. Moreover, the combination of ammonia-N enrichment and low light had a greater effect on submerged plants. This study indicates that V. natans were tolerant to ammonia-N concentrations <6mgL–1. Moreover, low light intensity (0.75% underwater light) amplified the toxic effects of ammonia-N, reducing ammonia-N tolerance from <6 to <3mgL–1.

The Differing Responses of Two Arabidopsis Ecotypes to Ammonium Are Modulated by the Photoperiod Regime

Abstract:Responses to excessive ammonium (NH4 +) were compared between two Arabidopsis ecotypes (Col-0, JA22) with respect to different photoperiods in hydroponics. In this study, we showed that external extra NH4 + led to severe growth suppression, accumulations of free NH4 + and amino acids and increased the activities of glutamine synthetase (GS) and glutamate dehydrogenase (GDH) in shoots of the two Arabidopsis ecotypes. However, the levels of free NH4 + and total amino acids increased, whereas the activities of GS, NADH-dependent glutamate synthase and GDH decreased under the continuous light when compared with the light (16 h)–dark (8 h) cycle photoperiod. Statistical analyses suggested that strong correlations exist among the growth reduction, accumulations of free NH4 +, total amino acids and levels of GS activity in shoots under the high NH4 + stress regardless of the photoperiod regimes. Interestingly, under the continuous light, Col-0 showed more resistant to such growth reduction and maintained about onefold higher capability of converting excess free NH4 + into amino acids, with onefold higher GS activity induced by the external NH4 + when compared with JA22. In contrast, these differences were abolished between Col-0 and JA22 under the light–dark cycle condition. Taken together, our results conclude that the sensitivity to NH4 + of Col-0 and JA22 is changed between the continuous light and the light–dark cycle photoperiod, which is correlative to the alteration of the GS activity in shoots.
Effects of High Ammonium Loading on Two Submersed Macrophytes of Different Growth Form Based on an 18-Month Pond Experiment

Abstract:Ammonium (NH 4 -N) produces a paradoxical effect on submersed macrophytes because it is not only the preferred nitrogen source for the growth of plants but also threatens the growth of plants at high concentration. Whether short-term and small-scale physiological toxicity experiments at an individual level can reflect the effects of high ammonium on populations of submersed macrophytes in natural conditions is still unclear. In this study, an 18-month experiment was conducted in six 600 m 2 ponds subjected to different levels of ammonium loading. The effects of high ammonium on populations of canopy-forming Myriophyllum spicatum and rosette-forming Vallisneria natans were explored. The results showed that M. spicatum and V. natans populations can develop high cover and height at high ammonium concentration (7 mg/L) at short-term exposures, and V. natans may be tolerant to 18 mg/L ammonium concentration. However, the cover of M. spicatum and the height of both species were inhibited at 2.4 mg/L at long-term exposures. The height of M. spicatum was two to six times higher than that of V. natans across all treatments and control by the end of the experiment, and the cover of M. spicatum was 7–11 times higher than that of V. natans in most NH 4 -N loading treatments, except the cover of M. spicatum in the highest NH 4 -N loading treatment with 18 mg/L NH 4 -N. The rosette-forming V. natans resists ammonium stress by slow growth (shoot elongation) to reduce consumption, while canopy-forming species resist ammonium stress by shoot elongation and canopy development to capture light. Although increasing ammonium concentration may induce severe stress on M. spicatum , the morphological characteristics of this species may, to some extent, release the plants from this stress. Our present study indicates that the negative effects of ammonium stress on the development of populations increased with exposure duration, and the submersed macrophyte community with stronger ability for light capture and dispersal may resist high ammonium stress. Nevertheless, in strongly ammonium-enriched systems, competition and succession cannot be neglected.

Abstract:Background. The decline of submerged plant populations due to high heavy metal (e.g., Cu) levels in sediments and ammonia nitrogen (ammonia-N) accumulation in the freshwater column has become a significant global problem. Previous studies have evaluated the effect of ammonia-N on submerged macrophytes, but few have focused on the influence of sediment Cu on submerged macrophytes and their combined effects. Methods. In this paper, we selected three levels of ammonia-N (0, 3, and 6 mg L(-1)) and sediment Cu (25.75 ± 6.02 as the control, 125.75 ± 6.02, and 225.75 ± 6.02 mg kg(-1)), to investigate the influence of sediment Cu and ammonia-N on submerged Vallisneria natans. We measured the relative growth rate (RGR), above- and below- ground biomass, chlorophyll, non-protein thiol (NP-SH), and free proline. Results and Discussion. The below-ground biomass of V. natans decreased with increasing Cu sediment levels, suggesting that excessive sediment Cu can result in significant damage to the root of V. natans. Similarly, the above-ground biomass significantly decreased with increasing ammonia-N concentrations, indicating that excessive water ammonia-N can cause significant toxicity to the leaf of V. natans. In addition, high ammonia-N levels place a greater stress on submerged plants than sediment Cu, which is indicated by the decline of RGR and chlorophyll, and the increase of (NP-SH) and free proline. Furthermore, high sediment Cu causes ammonia-N to impose greater injury on submerged plants, and higher sediment Cu levels (Cu ≥ 125.75 mg kg(-1)) led to the tolerant values of ammonia-N for V. natans decreasing from 6 to 3 mg L(-1). This study suggests that high sediment Cu restricts the growth of plants and intensifies ammonia-N damage to V. natans.
[Effect of Nutrient Loadings on the Regulation of Water Nitrogen and Phosphorus by Vallisneria natans and Its Photosynthetic Fluorescence Characteristics]

Abstract:Submerged macrophytes are an important component of aquatic ecosystems. During the growing period, submerged macrophytes can absorb nitrogen and phosphorus nutrients to reduce pollution loadings. Shoots of submerged macrophytes can also promote the adhesion of suspended substances in water, reducing the turbidity. The release of nutrients in sediments can be suppressed by its root system, and the resuspension of sediments caused by disturbance of winds and waves can also be resisted. The role of submerged macrophytes in ecological restoration of eutrophic lakes has attracted widespread attention. In 1960, the submerged plants Vallisneria natans and Potamogeton malaianus had been the dominant species in East Taihu. However after 2002, Nymphoides peltatum, Elodea nattalii, P. malaianus, etc. have gradually taken over the dominant roles along with significant elevations of nitrogen and phosphorus levels. Nutrients in water are not the only key factors causing eutrophication of water bodies; the nutrient source for submerged plant growth affect both the purification efficiency and the photosynthetic characteristics of submerged macrophytes. Excessive nitrogen and phosphorus concentrations can inhibit the photosynthetic physiological activities of submerged macrophytes, affecting the succession of aquatic vegetation. In addition, under high nutrient conditions, the competition from periphytic algae and planktonic algae may also directly poison submerged macrophytes, leading to its degradation and disappearance. Systematic studies on the regulation and photosynthetic fluorescence response mechanism of submerged macrophytes to varied nutrient loadings are helpful in revealing their relationships. The seedlings of submerged macrophyte V. natans were transplanted in a laboratory mesocosm to study the effect of nutrient loadings on its regulation of water nitrogen and phosphorus. Three nitrogen and phosphorus loadings from low, medium, and high levels derived from nitrate, ammonium, and phosphate were setup as the aquatic medium for the plant growth. Twelve harvests were carried out to determine the evolution of nutrient removal performance of V. natans. Its photosynthetic fluorescence characteristics were measured by a pulse-amplitude modulated fluorometer (Diving-PAM). Results showed that the nitrogen and phosphorus adsorption abilities of V. natans were gradually enhanced with the increase of nutrient concentrations in the range of TN ≤ 12 mg·L-1 and TP ≤ 1.0 mg·L-1. In the treatment of high nutrient concentrations (TN=12 mg·L-1 and TP=1.0 mg·L-1), the removal rates of nitrogen and phosphorus reached more than 95%. V. natans preferentially absorbed ammonium nitrogen when its concentration was high. The medium nutrient concentrations (TN:8-12 mg·L-1 and TP:0.6-1.0 mg·L-1) did not significantly affect the Fv/Fm ratio of leaves. However, the low nutrient concentrations (TN=3 mg·L-1 and TP=0.3 mg·L-1) could improve the Fv/Fm ratio of leaves and were beneficial for the growth of V. natans. The inhibition of photosynthetic activity and light tolerance were enhanced with the increase in nutrient concentrations. The photosynthetic activity of V. natans gradually recovered with no significant changes in the capacity for light harvesting, when the nutrient concentrations gradually decreased in the water. Our results indicate that the high nitrogen and phosphorus loadings indeed hamper the photosynthetic capacity, which may subsequently restrain the maintenance of the dominance of V. natans in the submerged macrophyte communities.
THE PHOTOSYNTHETIC PHYSIOLOGY AND GROWTH RESPONSE OF TWO ALGAE SPECIES, MICROCYSTIS AERUGINOSA AND SCENEDESMUS QUADRICAUDA, TO DIFFERENT NITROGEN FORMS AND CONCENTRATIONS

Abstract:Organic nitrogen and inorganic nitrogen were the main nitrogen sources in the north and south of Dianchi Lake, respectively. Thus, the effect of the different concentration (0.5, 1.0, 2.0, 5.0, 10, 20, 50 mg/L) of ammonium nitrogen (NH4-N) and alanine on growth of Microcystis aeruginosa and Scenedesmus quadricauda was investigated. In NH4Cl vitro co-culture, M. aeruginosa grew better than S. quadricauda, and its chlorophyll a increased to 2893.94 ug/L in 2 mg/L NH4Cl group. In vitro alanine treatments, S. quadricauda’s chlorophyll a was measured at 5034.34 ug/L in 50 mg/L alanine, which was higher than M. aeruginosa. M. aeruginosa in NH4Cl vitro monocultures showed better cell structure in 20 mg/L NH4Cl, and its chlorophyll a was higher than that of the corresponding concentration of alanine. The photosynthetic activity (Fv/Fm), the maximum electron transfers rate (ETRmax), and the saturated light intensity point (Ik) of M. aeruginosa increased with the ammonium concentration, and decrease with the alanine concentration, indicating that M. aeruginosa can tolerate higher concentrations of ammonium chloride. In high concentrations of NH4Cl, S. quadricauda’s cell was seriously damaged, but of alanine alone, it was intact. Fv/Fm, ETRmax, and Ik of S. quadricauda increased with the alanine concentrations, showing that S. quadricauda makes better use of organic nitrogen.

Effects of Ammonia Nitrogen and Sediment Nutrient on Growth of the Submerged Plant Vallisneria natans

Abstract:In order to investigate the impacts of ammonia nitrogen concentration and the necessity of dredging, five levels of ammonia nitrogen including 0, 2, 4, 8, and 16 mg L −1 and two levels of sediments nutriments (high and low) were treated. The total biomass, leaf, root, and flower biomasses were selected as the growth indicators, while total chlorophyll (i.e., chl‐a and ‐b), malondialdehyde (MDA), and free proline as subsidiary physiological index. When ammonia‐N contents of the water column were 8–16 mg L −1 , the total biomass of Vallisneria natans in the first harvest is higher than those in the second harvest, and the total chlorophyll concentration, MDA and free proline of the plants became significantly low. It indicates that high ammonia nitrogen (>8 mg L −1 ) of the water column inhibit the growth of V. natans . The total biomass, the total chlorophyll concentration of plants in the high nutrient sediment is higher than those in the low nutrient sediment, suggesting that high sediments nutrient promote the growth of V. natans . Therefore, using V. natans for water ecological restoration, the water should meet the ammonia nitrogen content of <8 mg L −1 . Moreover, dredging eutrophication sediment is probably not necessary.

Abstract:A 24-year-old male Chinese student from southern China presented with sudden onset of generalized weakness. The evening before admission he had played football for 3 hours, and eaten a Chinese takeaway plus two hamburgers. He awoke in the early hours of the following morning unable to move his arms or legs. There was no history of any recurring illness and in particular there were no symptoms suggestive of hyperthyroidism. On examination the only abnormality was a profound symmetrical muscle weakness more marked proximally than distally (Medical Research Council (MRC) classification grade 1–2). Abstract:Ammonia is among the common contaminants in aquatic environments. The present study aimed at evaluation of the toxicity of ammonia at high concentration by detecting its effects on the growth, pigment contents, antioxidant enzyme activities, and DNA damage (comet assay) of a unicellular microalga, Euglena gracilis. Ammonia restrained the growth of E. gracilis, while at higher concentrations, ammonia showed notable inhibition effect, the growth at 2 000 mg x L(-1) was restrained to 55.7% compared with that of the control; The contents of photosynthetic pigments and protein went up with increasing ammonia dosage and decreased when the ammonia concentration was above 1000 mg x L(-1); In addition, there was an obvious increase in SOD and POD activities, at higher concentration (2 000 mg x L(-1)), activities of SOD and POD increased by 30.7% and 49.4% compared with those of the control, indicating that ammonia could promote activities of antioxidant enzymes in E. gracilis; The degree of DNA damage observed in the comet assay increased with increasing ammonia concentration, which suggested that high dose of ammonia may have potential mutagenicity on E. gracilis.
Arabidopsis Plastid AMOS1/EGY1 Integrates Abscisic Acid Signaling to Regulate Global Gene Expression Response to Ammonium Stress

Abstract:Ammonium (NH(4)(+)) is a ubiquitous intermediate of nitrogen metabolism but is notorious for its toxic effects on most organisms. Extensive studies of the underlying mechanisms of NH(4)(+) toxicity have been reported in plants, but it is poorly understood how plants acclimate to high levels of NH(4)(+). Here, we identified an Arabidopsis (Arabidopsis thaliana) mutant, ammonium overly sensitive1 (amos1), that displays severe chlorosis under NH(4)(+) stress. Map-based cloning shows amos1 to carry a mutation in EGY1 (for ethylene-dependent, gravitropism-deficient, and yellow-green-like protein1), which encodes a plastid metalloprotease. Transcriptomic analysis reveals that among the genes activated in response to NH(4)(+), 90% are regulated dependent on AMOS1/EGY1. Furthermore, 63% of AMOS1/EGY1-dependent NH(4)(+)-activated genes contain an ACGTG motif in their promoter region, a core motif of abscisic acid (ABA)-responsive elements. Consistent with this, our physiological, pharmacological, transcriptomic, and genetic data show that ABA signaling is a critical, but not the sole, downstream component of the AMOS1/EGY1-dependent pathway that regulates the expression of NH(4)(+)-responsive genes and maintains chloroplast functionality under NH(4)(+) stress. Importantly, abi4 mutants defective in ABA-dependent and retrograde signaling, but not ABA-deficient mutants, mimic leaf NH(4)(+) hypersensitivity of amos1. In summary, our findings suggest that an NH(4)(+)-responsive plastid retrograde pathway, which depends on AMOS1/EGY1 function and integrates with ABA signaling, is required for the regulation of expression of NH(4)(+)-responsive genes that maintain chloroplast integrity in the presence of high NH(4)(+) levels.

Abstract:Erythropalum scandens Bl. is a woody vegetable with high nitrogen demand that inhabits southern China. Ammonium and nitrate are the two main forms of inorganic nitrogen that plants directly absorb. A pot experiment was performed to determine the growth, physiological responses, and preferences of 12-month-old E. scandens seedlings for ammonium and nitrate. Aboveground and underground growth indexes, biomass, physiological and biochemical indexes (chlorophyll [Chl], soluble sugar, soluble protein and free proline contents), and substrate pH and nitrogen contents were determined under different nitrate and ammonium ratios (0 NO3-: 100 NH4+, 25 NO3-: 75 NH4+, 50 NO3-: 50 NH4+, 75 NO3-: 25 NH4+, and 100 NO3-: 0 NH4+), and the control (0 NO3-: 0 NH4+). The results showed that ammonium and nitrate improved the growth and physiological status of E. scandens seedlings in most of the treatments compared to the control. The aboveground growth status and biomass accumulation of E. scandens seedlings were significantly better under the 0 NO3-: 100 NH4+ treatment during fertilization compared with all other treatments. However, the growth status of the underground parts was not significantly different among treatments. Significant differences in osmoregulator content, except for soluble sugars, and Chl content were observed. Soluble sugars and soluble proteins were highest under the 0 NO3-: 100 NH4+ treatment at the end of fertilization (day 175). However, free proline accumulated during fertilization and the increase in NO3- indicated that excessive use of NO3- had a negative effect on the E. scandens seedlings. The order of accumulating nitrogen content was leaves > roots > stems. The highest N accumulation occurred in the aboveground parts under the 0 NO3-: 100 NH4+ treatment, whereas the highest N accumulation occurred in the underground parts under the 50 NO3-: 50 NH4+ treatment. Substrate pH increased at the end of fertilization (day 175) compared with the middle stage (day 75), while total nitrogen, ammonium, and nitrate were highly significantly different among the treatments. Total nitrogen and NH4+ content were the highest under the 0 NO3-: 100 NH4+ treatment, while NO3- content was the highest under the 100 NO3-: 0 NH4+ treatment. In conclusion, 12-month-old E. scandens seedlings grew best, and had better physiological conditions in NH4+ than NO3-. The 0 NO3-:100 NH4+ treatment (ammonium chloride 3.82 g/plant) resulted in the best growth and physiological conditions. Most of the growth and physiological indexes were inhibited with the increase in nitrate.

Abstract:Loss of submersed macrophytes is a world-wide phenomenon occurring when shallow lakes become eutrophic due to excess nutrient loading. In addition to the well-known effect of phosphorus, nitrogen as a trigger of macrophyte decline has received increasing attention. The precise impact of high nitrogen concentrations is debated, and the role of different candidate factors may well change over the season. In this study, we conducted experiments with Vallisneria natans during the growing season (June–September) in 10 ponds subjected to substantial differences in nitrogen loading (five targeted total nitrogen concentrations: control, 2, 10, 20, and 100mgL−1) and compared the results with those obtained in our earlier published study from the low-growth season (December–April). Like in the low-growth season, growth of V. natans in summer declined with increasing ammonium (NH4) concentrations and particularly with increasing phytoplankton chlorophyll a (ChlaPhyt). Accordingly, we propose that shading by phytoplankton might be of key importance for macrophyte decline, affecting also periphyton growth as periphyton chlorophyll a (ChlaPeri) decreased with increasing ChlaPhyt. Free amino acid contents (FAA) of plants tended to increase with increasing NH4 concentrations, while the relationships between FAA with growth indices were all weak, suggesting that FAA might be a useful indicator of the physiological stress of the plants but not of macrophyte growth. Taken together, the results from the two seasons indicate that although a combination of high nitrogen concentrations (ammonium) and shading by phytoplankton may cause severe stress on macrophytes, active growth in the growing season enabled them to partly overcome the stress.

Abstract:Due to excess nutrient loading, loss of submersed macrophytes is a worldwide phenomenon in shallow lakes. Phosphorus is known to contribute significantly to macrophyte recession, but the role of nitrogen has received increasing attention. Our understanding of how high nitrogen concentrations affect the growth of submersed macrophytes, particularly under natural conditions, is still limited. In this study, we conducted experiments with canopy-forming Potamogeton crispus in 10 ponds subjected to substantial differences in nitrogen loading (five targeted total nitrogen concentrations: control, 2, 10, 20, and 100 mg L-1) and compared the results with those of our earlier published experiments with rosette-forming Vallisneria natans performed 1 year before. Canopy-forming P. crispus was more tolerant than rosette-forming V. natans to exposure to high NH4 concentrations. This is probably because canopy-forming species reach the water surface where there is sufficient light for production of carbohydrates, thereby allowing the plants to partly overcome high NH4 stress. Both the canopy-forming P. crispus and the rosette-forming V. natans showed clear declining trends with increasing chlorophyll a in the water. Accordingly, shading by phytoplankton might be of key importance for the decline in submersed macrophytes in this experiment. Both experiments revealed free amino acids (FAA) to be a useful indicator of physiological stress by high ammonium but is not a reliable indicator of macrophyte growth.
Response of the Growth of Vallisneria Natans under High Zn2+ Stress and Different NH4+/NO3− Ratios

Abstract:In order to evaluate the combined effects of Zn2+ and NH4+/NO3– stress, a cross experiment composed of different concentrations of Zn2+ (0, 2, 4, 8 mg L−1) and ratios of NH4+ and NO3– (1:6, 3:4, and 5:2) was conducted with Vallisneria natans under eutrophication (N = 4.667 mg L−1). Chlorophyll, protein, malondialdehyde (MDA), superoxide dismutase (SOD) activity, and ascorbate peroxidase (APX) activity of V. natans leaves were measured after cultivation for 7 days under stress of NH4+/NO3– and Zn2+. Variance analysis indicated that chlorophyll continuously decreased significantly with increasing Zn2+ concentration and NH4+/NO3– ratio, with the highest value at 0 mg L−1 Zn2+ and NH4+/NO3–, 1:6. However, MDA, soluble protein, SOD activity, and APX activity increased first and decreased subsequently, showing the greatest values at 2 mg L−1 Zn2+ and NH4+/NO3–, 1:6. Physiological responses of V. natans under high Zn2+ stress were obvious. A Zn2+ concentration of <2 mg L−1 could promote the growth of V. natans. Physiological functions of V. natans were partly affected under high NH4+/NO3–. However, it could promote the growth of V. natans at NH4+/NO3– <3:4 and a total nitrogen concentration of 4.667 mg L−1. Meanwhile, this study also showed that Zn2+ and NH4+/NO3– have a synergistic effect on V. natans.
