Abstract:A new analytical method was developed to simultaneously determine residues of 109 pesticides (including isomers) in the foods of animal origin. Acetonitrile was selected for accelerated solvent extraction (ASE) for effectively extracting the pesticides from the fatty samples. The cleanup was performed with an automated gel permeation chromatography (GPC) cleanup system. The prepared samples were analysed with GC–MS in the selected ion monitoring mode (SIM) using one target and two qualitative ions for each analyte. Chlorpyrifos-d10 was used as an internal standard. The lowest limit of detection was 0.3μgkg−1 for some pesticides. The recoveries and relative standard deviations (RSDs) were checked by spiking untreated samples with pesticides at 0.05, 0.1 and 0.2mgkg−1. The average recoveries of most pesticides were from 62.6% to 107.8%. The precision values expressed as RSD were all ⩽20.5% (n=6). Good linearity (r⩾0.99) was observed between 0.05 and 1.5μgmL−1.

Abstract:A rapid method has been developed to determine the multi-residues of 36 organophosphorus pesticides in the products of animal origin using capillary gas chromatography with flame photometric detector (GC-FPD (P)). The organophosphorus pesticides were extracted with acetonitrile by accelerated solvent extraction, and cleaned up by auto gel permeation chromatography and primary secondary amine (PSA) packing material. The collected solution was analyzed by the GC-FPD (P) and quantified by internal standard method. The 36 organophosphorus pesticides were separated efficiently from impurity in high sensitivity and reproducibility by GC-FPD (P). The limits of detection (LODs) ranged from 0.0012 mg/kg (phorate) to 0.014 mg/kg (pyraclofos), and the limits of quantitation (LOQs) ranged from 0.004 mg/kg (phorate) to 0.047 mg/kg (pyraclofos). The recoveries ranged from 58.2% to 106.3% in blank samples spiked with 0.05, 0.1 and 0.2 mg/kg of 36 organophosphorus pesticides. The LODs, LOQs and the recoveries of the method all satisfy the requirement of pesticide residue analysis.

Abstract:Objective To determine the multi-residues of 12 kinds of carbamate pesticides in animal-derived foods.Method The samples were extracted with acetonitrile by accelerated solvent extraction(ASE)and cleaned up by auto gel permeation chromatography(GPC)and primary secondary amine(PSA)packing material.The collected solution was analyzed by high performance liquid chromatography-fluorescence detector(HPLC-FLD)with post-column derivation and quantified by external standard method.The wavelengths of excitation and emission were set at 330 nm and 465 nm,respectively.Results The limits of detection(S/N=3)of 12 kinds of carbamate pesticides ranged from 0.24 μg/kg(isoprocarb)to 1.02 μg/kg(aldicarb sulfoxide).The recoveries ranged from 62.1%±8.8% to 104.0%±5.6% by fortified with 0.05 mg/kg,0.1 mg/kg and 0.2 mg/kg of 12 kinds of carbamate pesticides into the blank treated samples.The RSD was less than 20%.Conclusion The limits of detection and the recoveries of 12 kinds of carbamate pesticides both meet the analysis requirements of pesticide residues.

Abstract:A liquid chromatography-mass spectrometry (LC-MS) method was established for the simultaneous determination of carbamate and organophosphorus pesticides (OPPs) in fruits and vegetables by dispersive solid phase extraction using acetonitrile as extraction solvent and primary secondary amine (PSA) as sorbent instead of octadecyl bonded silica (ODS). Four common representative samples (tomato, apple, carrot and cabbage) were selected from the supermarket in order to validate the method by recovery and precision. The obtained recoveries were, with a few exceptions, in the range of 80%-110% with RSDs less than 8%; and limits of detection ranged from 0.5 to 35 μg/kg in tomato. In addition, the method was applied to pesticides residue detection in vegetables and fruits obtained from local markets.
Simultaneous Determination of Multi-Component Mycotoxin Contaminants in Foods and Feeds by Ultra-Performance Liquid Chromatography Tandem Mass Spectrometry.

Abstract:The present study developed an improved analytical method for the simultaneous quantification of 17 kinds of Aspergillus, Fusarium and Penicillium mycotoxin contaminants in foods and feeds by ultra-performance liquid chromatography combined with electrospray ionization triple quadrupole tandem mass spectrometry (UPLC–MS/MS) under the multiple reaction monitoring (MRM) mode, and especially focused on the optimization of extraction, clean-up, UPLC separation and MS/MS parameters of analytes. Homogenized samples were sequentially extracted by 84% (v/v) of acetonitrile aqueous solution with the selected internal standard (zearalanone) spiking, SPE clean-up with Mycosep 226 Aflazon+ Multifunctional cartridges, filtration, concentration and secondary filtration. Using double sample injection method, the analytes were separated by UPLC BEH C18 column (100mm×2.1mm I.D., 1.7μm), and eluted with ammonium acetate/methanol and aqueous ammonia/methanol for the ESI+ and ESI− analysis, respectively. The 10 positive ions and 7 negative ions of mycotoxins were separated by gradient elution with the retention time of 6.5 and 4min, respectively. The LOQ of selected analytes ranged from 0.01 to 0.70μgkg−1, which was lower than the criteria of EU, USA and other countries on the determination of the minimum limiting level of various mycotoxins in foods including baby foods and feed stuffs. Meanwhile, high correlation coefficients (r2>0.99) of 17 mycotoxins were obtained within their respective linear ranges (0.05–20μgkg−1 for 10 positive ions and 0.5–50μgkg−1 for 7 negative ions) and reasonable recoveries (70.6–119.0%) of them were also demonstrated in different spiked levels. This quantitative method has many advantages including simple pretreatment, rapid determination and high sensitivity, which could be applied to the determination and quantification of multi-component mycotoxin contaminants in complex matrixes. Meanwhile, the method successfully fulfilled the minimum limiting level requests from various countries.

Abstract:An ultra-high-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method based on selective accelerated solvent extraction and magnetic material purification was established to analyze the residues of various veterinary antibiotics and agricultural fungicides and insecticides in livestock and poultry excrement. Methanol-acetonitrile (4:1, V/V) was used as the extraction solvent and static extraction was conducted three times in 5 min. Preliminary purification was achieved by adding 0.5 g acidic alumina-florisil (1:1, W/W) to the extraction cell while the extraction was conducted. This preliminarily-purified extract was further purified using magnetic material, then analyzed using UPLC-MS/MS. Under optimal conditions, 33 types of antibiotics, including 3 amphenicols, 8 macrolides, 17 sulfonamides and 5 nitroimidazoles, as well as 37 types of pesticides, including 27 insecticides and 10 fungicides, were detected. Recoveries ranged from 60.3% to 110.0% at three spiked concentrations (10 mu g/kg, 20 mu g/kg and 50 mu g/kg), the detection limits ranged from 0.2 to 3.5 mu g/kg and the quantitative limits ranged from 0.5 to 11.5 mu g/kg. This newly-established method was tested using 30 livestock and poultry excrement samples and confirmed its use for determining veterinary drugs and pesticides in practical samples. Determination of 46 Antibiotics Residues in Organic Fertilizer by UPLC-MS/MS with QuEChERS Combined with Column Purification

Abstract:A confirmative method was developed for the simultaneous determination of 46 antibiotics,including sulfonamides,quinolones and macrolides in organic fertilizer by ultrahigh performance liquid chromatography-tandem massspectrometry(UPLC-MS/MS).The samples were extracted with acetonitrile-EDTA buffer solution (pH 10.0),and separated by centrifugation after salting-out.The upper acetonitrile layer was purified by the QuEChERS steps with sorbents.The buffer solution layer was purified with solid-phase extraction cartridge.The separation of target drugs was performed on an ACQUITY UPLC BEH C1scolumn using a mobile phase consisting of 2 mmol/L ammonium acetate with 0.1％ formic acid and methanol by gradient elution.The analysis of target compounds was carried out with positive electrospray ionization (ESI +)source under multiple reaction monitoring (MRM) mode.The matrix-matched external standard calibration was used for quantification.Good linearities for the 46 antibiotics were achieved over the concentration of 1-200 μg/L,with correlation coefficients (r2) of 0.996 6-0.999 9.The recoveries of sulfonamides,quinolones and macrolides at spiked levels of 25,100,400 μg/kg were in the range of 67.8％-95.6％,65.6％-89.4％ and 66.6％-107.8％,respectively,with relative standard deviations(RSDs) of 0.4％-11.9％.The limits of detection (LOD,S/N =3) were 0.6-4.6 μg/kg,and the limits of quantitation(LOQ,S/N=10) were 2.1-15.4 μg/kg.

Abstract:A method has been developed for the determination of 33 pesticide residues in tea, including organophosphorous, organochlorine and pyrethroid pesticides. The target analytes were extracted with the solution of acetone/dichloromethane (1:1, v/v) using accelerated solvent extraction (ASE) and then purified using gel permeation chromatography (GPC) to eliminate most of the coextracts, such as pigments, lipids and waxes. They were further purified using Carb-NH2 and Florisil solid phase extraction (SPE) cartridges prior to the identification using gas chromatography-mass spectrometry (GC-MS). The quantitative analysis was performed with flame photometric detector (FPD) for organophosphorous pesticides and electron capture detector (ECD) for organochlorine and pyrethroid pesticides. At the spiked level of 0.05 mg/kg, the recoveries for most pesticides were between 70%–120%; the relative standard deviations were less than 20%; the limits of detection varied from 0.005 to 0.05 mg/kg (defined in terms of 10 times of the baseline noise). This method is precise, sensitive and highly efficient in extraction. After routine applications, the results indicated that this method is suitable for the determination of pesticide residues in the tea for export.

Abstract:The present work developed an analytical method for simultaneous determination of fumonisins B(1), B(2) and B(3) residues in maize by ultra high-performance liquid chromatography combined with electrospray ionization triple quadrupole tandem mass spectrometry (UHPLC-MS/MS) under the multiple reaction monitoring (MRM) mode, and especially focused on the optimization of extraction, clean-up, UHPLC separation and MS/MS parameters. The method involves addition of fumonisins isotope internal standards, extraction with a mixture of acetonitrile and water and clean-up with solid-phase extraction (SPE) cartridges before UHPLC-MS/MS analysis. A single-laboratory method validation was conducted by testing three different spiking levels for repeatability and recovery according to International Union of Pure and Applied Chemistry (IUPAC) guidelines. The LOQ of FB(1), FB(2) and FB(3) were 1.50, 1.65 and 0.4 μg kg(-1), respectively, which were lower than the criteria of EU, USA and other countries regarding minimum residue limits of fumonisins in foods including baby foods and feedstuffs. Recoveries of three fumonisins ranged from 80.9% to 97.0% with RSD values of 2.4-11.1%.The advantages of this method include simple pretreatment, rapid determination and high sensitivity, and it fulfills the requirements for food analysis with respect to minimum residue limits of fumonisins in various countries.

Abstract:In this paper, we developed a sensitive UPLC-MS/MS method to determine pesticide residues in plant matrices (corn, fresh corn, fresh corn stover, old corn stover, and corn silage) and animal matrices (beef, fat, milk, milk fat, kidney, liver, and cow stomach) quantitatively. Twenty-seven pesticides were extracted with acetonitrile from all plant and animal matrices separately and purified with a mixture of primary secondary amine (PSA) and graphitized carbon black (GCB) or octadecylsilane (C 18 ). The average recoveries of these compounds ranged from 60.7% to 118.2%, and the relative standard deviations were less than 20.0%. The limit of quantitation for all compounds was 0.01 mg kg −1 (for cyhalothrin and beta cypermethrin the LOQ was 0.02 mg kg −1 ). The establishment of multi-residue analysis methods for a variety of matrices can be used as a database for future method research. The results of this study are essential for calculating the transfer of pesticide residues from feed to animal products and for monitoring food safety, which will protect people's health and safety.

Abstract:Objective To establish a method for the rapid determination of 77 kinds of pesticide residues in oil plants and plant oils by ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/ MS). Methods Samples were extracted by QuEChERS with acetonitrile, and purified with PSA+C18 cartridge. A gradient system of a mixture of 0.1% formic acid and acetonitrile with multi-reaction monitoring (MRM) were adopted for quantitative determination. Results Seventy-seven kinds of pesticide residues could be detected within 10 min, the correlation rate (r) was more than 0.99, and the average recoveries ranged from 60.2% to 99.8% with relative standard deviation (RSD) less than 8%. Abstract:A new method for screening and identification 420 pesticide residues in fruits and vegetables by ultra-performance liquid chromatography coupled with quadrupole-time of flight mass spectrometry (UPLC-Q-TOF/MS) were developed. The samples were extracted with acetonitrile/acetic acid (99:1, V/V ), and clean-up by SinChERS-Nano (single-step, cheap, effective, rugged, safe, nano) column, determined by UPLC-Q-TOF/MS. The accurate mass database and MS/MS database which contains 420 pesticides were established, the automatic retrieval of detection results was carried on according to the accurate mass, retention time, isotope ratio, ion fragment information, and so on. Method verification was performed on leeks samples. The results showed that 420 pesticides had good linearity in the range of 0.1–100 μg/L, and the correlation coeffificients ( R 2 ) was greater than 0.990. The limits of detections (LODs) and limits of quantifications (LOQs) of 420 pesticides were in range of 0.05–2.0 and 0.1–5.0 μg/L, respectively. The average spike recoveries at 3 levels were 70.1 % to 119.7 %, and the relative standard deviations (RSD) were lower than 20 % ( n = 6). With this method, a survey of pesticide residues was conducted for 110 samples of 10 different fruits and vegetables, which provided scientific data for ensuring pesticide residue safety of the fruits and vegetables consumed daily by the public. This method was simple, sensitive and accurate, and could be used for rapid screening of 420 pesticide residues in fruits and vegetables.

Abstract:An analytical method for the determination of 34 pesticide residues in plant-derived foods was established using an automated QuEChERS sample preparation system combined with high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). The sample extraction and dispersive solid-phase extraction in the manual QuEChERS method were combined using the vortex vibration and centrifugation functions of the automated QuEChERS sample preparation system. The operating parameters and pretreatment steps were optimized, and the analytes were detected in the multiple reaction monitoring (MRM) mode. The quantification analysis was performed by the matrix-matched external standard method. The automated and the manual QuEChERS methods were compared from the methodological verification standpoint. The calibration curves showed good linearity in a certain range, and the correlation coefficients (R2) were greater than 0.99. The limits of detection and limits of quantification were 0.76-3.60 μg/kg and 2.28-10.80 μg/kg, respectively. Moreover, the recoveries ranged from 53.0% to 125.2%, and the relative standard deviations (RSDs) were less than 1.59% (n=5). The results obtained by the manual QuEChERS method were not significantly different from those obtained by the automated QuEChERS method. This method can effectively reduce the labor intensity and probability of error in the determination of pesticide residues.

Simultaneous determination of triazole fungicides in animal-origin food by ultra-high-performance liquid chromatography coupled with tandem mass spectrometry

Abstract:A method for the simultaneous determination of 21 triazole fungicides in animal-origin foods was established by using UPLC-MS/MS. The dilution solvent, extraction solvent, and QuEChERS purification adsorbent composition, were optimized. The response value of the target compound was the highest and the chromatographic peak shape was optimal under the following conditions: water-acetonitrile as the mobile phase, acetonitrile to extract the target compound, C18 (100 mg) as the adsorbent, and water-acetonitrile as the diluent. Our method was validated under electrospray ionization (ESI) + conditions with six animal-origin foods. The 21 triazole fungicides showed good linear relationships (0.1-20 μg∙L-1, R2 > 0.99). The limits of detection and quantitation ranged from 0.1 to 0.3 μg∙kg-1 and 0.3 to 0.9 μg∙kg-1, respectively. The average recoveries ranged from 72.0% to 114.8% with RSDs < 9.9%. Therefore, our method was suitable for the determination of pesticide residues in commercially available animal-origin samples.
Determination of 44 organophosphorous pesticides residual in chestnut using solid phase extraction and on-line gel permeation chromatography/Gas chromatography-mass spectrometry

Abstract:An analytical method has been developed for the determination of 44 organophosphorous pesticides in chestnut. With acetonitrile-water (4 : 1, V/V) as the extraction solution, the samples were extracted by means of high-speed homogenization. And the crude extract was purified by ENVI-18 solid phase extraction column to remove most fats and sterols in samples, then after on-line GPC-GC/MS analysis which further removed macromolecular interference material, such as pigment and fat, the backgroud interference brought about by the complex matrix in samples was effectively reduced. When standard addition to the samples was 0.05 mg/kg, the recovery rate for most of pesticides was from 65% to 120%, and the relative standard deviation was less then 15%. The limits of detection of 28 organophosphorous pesticides were 0.002 - 0.05 mg/kg. With the application of external standard and gas chromatography-mass spectrometry, the linear relations and the recovery results were satisfactory. Development of a Multiresidue Method for the Analysis of 33 Organochlorine Pesticide Residues in Fatty and High Water Content Foods

Abstract:A new multiresidue method has been developed and validated for the determination of 33 organochlorine pesticides (OCPs) in various fatty and high water content food matrices. The OCP residues in foods were extracted with matrix solid-phase dispersion and cleaned up with gel permeation chromatography and Florisil solid-phase extraction. The instrumental determination was carried out by a gas chromatograph coupled to a single quadrupole mass spectrometer (MS) with runtime of 11 min. Besides, negative chemical ionization mode was also studied and evaluated for OCPs’ sensitivities. The optimized MS was operated in electron ionization mode and acquiring three selected ions per target compound. The method was validated with various food samples, including edible oil, meat, seafood, eggs, coffee, tree nuts, fruits, vegetables, etc. according to SANCO/12571/2013. An adequate linear relationship was obtained in the studied concentration range (0.5–10.0 μg kg−1) in sample; the average spiked recovery values were in the range 70–120 % for the two levels of concentration studied in samples: 0.5 and 2.0 μg kg−1. Precision values, expressed as relative standard deviation, were lower than 18 % at aforementioned spiking levels; detection limits and quantification limits were below or equal to 0.1 and 0.5 μg kg−1, respectively. Moreover, certified reference materials were used to assess the accuracy of the developed method. Finally, the developed method was successfully applied for the OCPs’ determination in real samples for the first Hong Kong Total Diet Study.

Abstract:A multi-residue method for the determination of 70 pesticides in fruits and veg- etables by means of LC-MS/MS is presented.The pesticides were extracted by acetonitrile,pu- rified using SPE and finally detected by LC-MS/MS within 35 minutes.The method is proved to be linear obtaining correlation coefficients from 0.995 to 1.000 over the range 0.01～2.00 μg/mL.The obtained recoveries are in the range 70%～120% and the LOD is 10 μg/kg.The LOD,recovery and RSD can meet the demands of the analysis of pesticide residue.

Abstract:A method was developed for the simultaneous determination of 16 mycotoxins in drug and food homologous products by ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) combined with accelerated solvent extraction (ASE) and QuEChERS. The target mycotoxins in drug and food homologous products were extracted by ASE. After concentration, the extracts were purified by QuEChERS. Then, the target compounds were analyzed by UPLC-MS/MS in both positive and negative electrospray ionization and MRM modes. Aflatoxin B1 and fumonisin B1 were quantified by the internal standard method, and the remaining mycotoxins were quantified by the matrix-matched external standard method. The proposed method showed a good linear relationship, with correlation coefficients greater than 0.99. The limits of detection (LODs) and limits of quantification (LOQs) of the 16 mycotoxins ranged from 0.008 μg/kg to 0.3 μg/kg and from 0.03 μg/kg to 1.0 μg/kg, respectively. The blank samples were spiked at three levels, and the recoveries ranged from 70.8% to 118%, with the RSDs being 2.5% to 10.2%. The developed method was successfully applied to mycotoxin analysis in 30 scutellaria, puerarin and sea buckthorn samples bought from local markets. Different levels of mycotoxins were detected in some of the products. The proposed method is simple, rapid and sensitive, and it can be applied to the simultaneous determination of multi-mycotoxins in drug and food homologous products.

Abstract:A new multi-residue method for determination of pesticide residues in a wide variety of fruit and vegetables, using the National Food Administration (NFA) ethyl acetate extraction and determination by means of LC-MS/MS, is presented. The method includes pesticides normally detected by LC-UV or LC-fluorescence such as benzimidazoles, carbamates, N-methylcarbamates and organophosphorus compounds with an oxidisable sulphide group as well. After extraction with ethyl acetate, the extract is concentrated and an aliquot of the extract is evaporated to dryness and redissolved in methanol before injection on LC-MS/MS. The method has been validated for 57 different pesticides and metabolites. Representative species from different commodity groups were chosen as matrices in order to study the influence from different matrices on recoveries. The fortification levels studied were 0.01-0.5 mg kg(-1). Matrix effects were tested for all matrices by means of standard addition to blank extracts. The matrix effect, expressed as signal in solvent compared to signal in matrix, was in general found to be small. The obtained recoveries are, with a few exceptions, in the range 70-100%. The proposed method is quick and straightforward and no additional clean-up steps are needed. The method can be used for the analysis of all 57 pesticides in one single determination step at 0.01 mg kg(-1).
