Abstract:Monitoring microbial water quality is essential for recreational beaches in order to protect human health. To evaluate the relative importance and impacts of various types of non-point microbial sources at a subtropical beach (Hobie Beach, Miami, USA), we utilized a coastal ocean circulation model (Delft3D) with a microbe transport-fate model. Those non-point sources include beach sediment, dog feces, bather shedding, and rainfall runoff. The hydrodynamic model results agreed well with tidal elevations recorded by a nearby NOAA tidal station and also field data collected by pressure sensors, acoustic Doppler current profiler (ADCP). We modeled enterococci levels from four different types of non-point sources on the beach and Staphylococcus aureus levels from only the bather shedding. Model results suggest that dog feces are spotty sources of enterococci and can result in transient spikes of enterococci levels for hours. Beach sands are pervasive sources of enterococci and may explain observed persistent elevations of enterococci levels at this site. Runoff may also significantly increase enterococci levels during rainfall events while bather shedding contribution of enterococci is almost negligible. Bather is the only Staphylococcus aureus source considered in the study and simulated levels are in the same order as prior field measurements.
Indicator microbes correlate with pathogenic bacteria, yeasts and helminthes in sand at a subtropical recreational beach site

Abstract:Aims: Research into the relationship between pathogens, faecal indicator microbes and environmental factors in beach sand has been limited, yet vital to the understanding of the microbial relationship between sand and the water column and to the improvement of criteria for better human health protection at beaches. The objectives of this study were to evaluate the presence and distribution of pathogens in various zones of beach sand (subtidal, intertidal and supratidal) and to assess their relationship with environmental parameters and indicator microbes at a non-point source subtropical marine beach. Methods and results: In this exploratory study in subtropical Miami (Florida, USA), beach sand samples were collected and analysed over the course of 6 days for several pathogens, microbial source tracking markers and indicator microbes. An inverse correlation between moisture content and most indicator microbes was found. Significant associations were identified between some indicator microbes and pathogens (such as nematode larvae and yeasts in the genus Candida), which are from classes of microbes that are rarely evaluated in the context of recreational beach use. Conclusions: Results indicate that indicator microbes may predict the presence of some of the pathogens, in particular helminthes, yeasts and the bacterial pathogen Staphylococcus aureus including methicillin-resistant forms. Indicator microbes may thus be useful for monitoring beach sand and water quality at non-point source beaches. Significance and impact of the study: The presence of both indicator microbes and pathogens in beach sand provides one possible explanation for human health effects reported at non-point sources beaches.
Intraseptal procaine abolishes hypothalamic stimulation-induced wheel- running and hippocampal theta field activity in rats

Abstract:Rats were implanted chronically with hippocampal recording electrodes, a microinfusion guide cannula aimed at the medial septal nucleus, and an electrode for electrical stimulation of the posterior hypothalamic nucleus (PH). PH stimulation elicited running in rats placed in a wheel and simultaneously occurring hippocampal theta field activity (HPC- theta). In the preprocaine (PRE) testing condition, a positive linear relationship was demonstrated among the intensity of electrical stimulation of the PH, wheel-running speed, and the peak frequency of HPC-theta. HPC-theta amplitude reached an asymptote at the lowest levels of electrical stimulation of the PH. Procaine hydrochloride (1.5 microliters, 20% solution), a local anesthetic, was then infused into the medial septal nucleus (MS). Five minutes after the infusion, PH stimulation no longer induced wheel-running behavior or HPC-theta, and the remaining irregular field activity was significantly reduced in amplitude. Fifteen minutes after the procaine infusion, PH stimulation still did not elicit HPC-theta or running behavior in the majority of animals but did evoke large-amplitude sharp-waves. Thirty minutes after the procaine infusion, PH stimulation again elicited HPC-theta and running behavior, but HPC-theta peak frequency and running speeds were both significantly reduced compared with PRE values. Forty-five minutes after the infusion, HPC-theta amplitude had recovered to PRE values, but HPC-theta frequency and running speeds elicited by PH stimulation were still significantly reduced. By 60 min after procaine administration, the amplitude and frequency of HPC-theta and the running speeds elicited by PH stimulation recovered to PRE values. Multiple regression analysis revealed that the recovery pattern of running behavior reflected the frequency rather than the amplitude of HPC-theta. Neither saline control infusions into the MS nor procaine infusions into the lateral septum and paraventricular thalamic nucleus affected HPC-theta or running behavior. These findings are consistent with the notion that both the locomotor activity and “movement-related” HPC-theta frequency induced by electrically stimulating the PH were attributable to ascending activation of a hypothalamo-septal pathway and not to activation of descending brainstem or peripheral motor systems.

Abstract:For decades, the risk of exposure to infectious diseases in recreational beaches has been evaluated through the quantification of fecal indicator bacteria in water samples using culture methods. The analyses of sand samples have recently been developed as a complement to the monitoring of recreational waters in beach quality assessments. The growing use of molecular techniques for environmental monitoring allows for the rapid detection of pathogenic genes, thus providing more accurate information regarding the health risk of exposure to contaminated sand. The aim of this work was to determine the relationship between the fecal indicators abundance in water and sand and the presence of Shiga toxin-producer Escherichia coli (STEC) in sand by analyzing samples from touristic beaches using culture-dependent (fecal coliforms assay) and culture-independent (real-time PCR of stx1 , stx2 , and eae genes) techniques. We found a high concentration of coliform bacteria in water and sand in several beaches in eastern Uruguay, with different levels of sanitation networks and levels of urbanization. The presence of STEC virulence genes (mainly stx1 ) was confirmed in 8 out of 20 sand samples. The recreational use of sandy beaches may imply a risk to the health of its users, especially near streams and creek outflows, thus highlighting the need of monitoring sand bacteriological quality and pathogens using molecular tools.

Abstract:Beach sand is a habitat that supports many microbes, including viruses, bacteria, fungi and protozoa (micropsammon). The apparently inhospitable conditions of beach sand environments belie the thriving communities found there. Physical factors, such as water availability and protection from insolation; biological factors, such as competition, predation, and biofilm formation; and nutrient availability all contribute to the characteristics of the micropsammon. Sand microbial communities include autochthonous species/phylotypes indigenous to the environment. Allochthonous microbes, including fecal indicator bacteria (FIB) and waterborne pathogens, are deposited via waves, runoff, air, or animals. The fate of these microbes ranges from death, to transient persistence and/or replication, to establishment of thriving populations (naturalization) and integration in the autochthonous community. Transport of the micropsammon within the habitat occurs both horizontally across the beach, and vertically from the sand surface and ground water table, as well as at various scales including interstitial flow within sand pores, sediment transport for particle-associated microbes, and the large-scale processes of wave action and terrestrial runoff. The concept of beach sand as a microbial habitat and reservoir of FIB and pathogens has begun to influence our thinking about human health effects associated with sand exposure and recreational water use. A variety of pathogens have been reported from beach sands, and recent epidemiology studies have found some evidence of health risks associated with sand exposure. Persistent or replicating populations of FIB and enteric pathogens have consequences for watershed/beach management strategies and regulatory standards for safe beaches. This review summarizes our understanding of the community structure, ecology, fate, transport, and public health implications of microbes in beach sand. It concludes with recommendations for future work in this vastly under-studied area.

Abstract:Highly urban coastal communities in low lying areas and with high water tables are vulnerable to sea-level rise and to corresponding increases in coastal groundwater levels. Stormwater conveyance systems are under increased risk. Rising groundwater levels affect the hydraulics of the stormwater system thereby increasing contaminant transport, for example the fecal indicator bacteria enterococci, to coastal waters. This study offers a unique opportunity to evaluate the impacts of increased contaminant transport on marine coastal environments. Here we assessed historic and recent coastal water quality, stormwater sampling data, groundwater monitoring and tidal elevations near the coastline, in the context of altered hydraulics within the system. Two pathways of enterococci to marine waters were identified. Direct discharge of contaminated stormwater runoff via the stormwater outfalls and tidally driven contaminated groundwater discharge. As sea level continues to rise, we hypothesize that a diminished unsaturated zone coupled with altered hydraulic conditions at the coastal groundwater zone will facilitate the transport of enterococci from urban sediments to the study site (Park View Waterway in Miami Beach, FL USA). We recommend improvements to the stormwater conveyance system, and maintenance of the sanitary sewer system to mitigate these impacts and minimize transport of enterococci, and other stormwater pollutants to coastal waters. The results of this study can be useful to interpret high enterococci levels in low lying coastal areas where groundwater is influenced by rising sea water levels.

Abstract:Erosion poses a significant threat to oceanic beaches worldwide. To combat this threat, management agencies often utilize renourishment, which supplements eroded beaches with offsite sand. This process can alter the physical characteristics of the beach and can influence the presence and abundance of microbial communities. In this study, we examined how an oceanic beach renourishment project may have impacted the presence and abundance of Escherichia coli ( E . coli ), a common bacteria species, and sand grain size, a sediment characteristic that can influence bacterial persistence. Using an observational field approach, we quantified the presence and abundance of E . coli in sand (from sub-tidal, intertidal, and dune zones on the beach) and water samples at study sites in both renourished and non-renourished sections of Folly Beach, South Carolina, USA in 2014 and 2015. In addition, we also measured how renourishment may have impacted sand grain size by quantifying the relative frequency of grain sizes (from sub-tidal, intertidal, and dune zones on the beach) at both renourished and non-renourished sites. Using this approach, we found that E . coli was present in sand samples in all zones of the beach and at each of our study sites in both years of sampling but never in water samples. Additionally, we found that in comparison to non-renourished sections, renourished sites had significantly higher abundances of E . coli and coarser sand grains in the intertidal zone, which is where renourished sand is typically placed. However, these differences were only present in 2014 and were not detected when we resampled the study sites in 2015. Collectively, our findings show that E . coli can be commonly found in this sandy beach microbial community. In addition, our results suggest that renourishment has the potential to alter both the physical structure of the beach and the microbial community but that these impacts may be short-lived.

Abstract:Coastal water quality assessment is important to maintain a healthy environment for various uses including fisheries and recreation. Microbial populations are used as biological indicators of contamination to monitor water quality and are considered by the government to be one of the critical features for issuing safety guidelines. Different bacterial groups (pathogenic, vibrio and faecal) from five major recreational beaches of Chennai, India, were monitored for the assessment of coastal water quality. Faecal coliforms (FC) were high at all the beaches, with up to 4.2 × 105 CFU/mL and exceeding the normal standard limits of 100 CFU/100 mL set by the Central Pollution Control Board (CPCB) of India. Rainfall was found to have a role in the variability and distribution of indicator and pathogenic bacteria. The seasonal dry period witnessed elevated FC, while dilution in the wet period reduced Escherichia coli–like organisms (ECLO). High microbial counts were detected near the beach situated close to the river mouth, mainly due to discharges of untreated domestic sewage and industrial wastes. Similarly, the biological oxygen demand (BOD) was also high, 0.32 to 10.32 mg/L. Dissolved inorganic nitrogen (DIN) ranged from 2.21 to 134.53 μmol/L and inorganic phosphate (IP) ranged from 0 to 57.53 μmol/L. These values indicated the presence of significant untreated sewage in the coastal water. This study revealed that Chennai coastal waters carry high levels of faecal and pathogenic bacteria, detrimental for recreational and other contact activities. The quantitative and qualitative analyses will be useful for modelling and prediction of coastal water quality and management of other recreational beaches in India.

Abstract:Human population growth, especially in coastal urban cities, increases the potential for fecal pollution of adjacent waterways, requiring continued advances in pollution monitoring and management. Infections remain the largest health risk from contact with fecal- and sewage-polluted waters, and a small number of fecal indicator bacteria (FIB) are used as primary pollution assessment tools. While FIB continue to be useful tools, some of the assumptions about the behavior of FIB in the environment, and the associated pathways for pathogen exposure, have come into question. Research into the extra-enteric ecology of these indicators has identified management-relevant complexities including particle association, prolonged environmental persistence, and multidirectional microbial exchange among water, sediment, and air. These complexities provide opportunities for improving current monitoring and modeling strategies and to better understand exposure pathways for sewage-related infections.

Abstract:Fecal pollution of estuaries and adjacent creeks and streams is of significant concern along the Gulf of Mexico. The prospective threat to human life and water quality impairment via fecal pollution is a substantial danger to the strength and resistance of coastline areas. Pensacola, FL, has a prosperous coastal tourism industry that is utilized for numerous other uses, such as recreational watersports and boating, seafood, and shellfish harvesting. However, the frequency and severity of fecal contamination present possible socio-economic issues, specifically financial hardships. Therefore, understanding the source, abundance, and fate of fecal microbial pollutants in aquatic systems signifies an imperative initial stage for detecting the host sources and techniques to lessen their transport from the landscape. This research aimed to quantify the fecal indicator bacteria (FIB), Escherichia coli, and perform microbiological fecal source tracking to verify if the fecal inputs are of either animal or human host origin. Surface water samples were taken from urban and peri-urban creeks for two sampling periods (February 2021 and January 2022), and IDEXX Colilert-18 (USEPA Standard Method 9223) was used for E. coli enumeration. DNA extractions were obtained from each sample, and quantitative PCR was utilized for fecal microbial source tracking (MST) to detect human, dog, ruminant, and bird host-specific Bacteroides DNA. The result indicates elevated quantities of FIB, E. coli, that surpass the threshold considered safe regarding human health. E. coli at six sites over the two sampling periods exceeded the impairment threshold, reaching as high as 866.4 MPN/100 ml. Fecal source tracking identified human host fecal contamination at four of nine sites, dogs at three of nine, and birds at one site. However, those sites with sources identified via MST all had E. coli levels below impairment thresholds. No sites were determined to be positive for ruminant as a source or for the pathogen Helicobacter pylori. No canine host fecal inputs were found in January 2022, and only one site with human sewage. Our results highlight the utility of MST in assessing bacterial inputs to water bodies and the challenges.

Abstract:Coastal sands are important natural recreational facilities that have become hotspots for tourism and economic development. However, these sands harbour diverse microbial assemblages that play a critical role in the balance between public health and ecology. In this study, targeted high-throughput sequencing analysis was used to identify sand-borne bacterial populations at four public beaches in Durban. The effect of heavy metal in shaping the distribution of bacterial metacommunities was determined using canonical correspondence analysis (CCA), while the functional gene profiles were predicted using PICRUSt2 analysis. Sequences matching those of the bacterial phylum Proteobacteria were the most abundant in all samples, followed by those of the phyla Firmicutes, Actinobacteria, Bacteroidetes, and Gemmatimonadetes. Genus-level taxonomic analysis showed the presence of 1163 bacterial genera in all samples combined. The distribution of bacterial communities was shaped by heavy metal concentrations, with the distribution of Flavobacteria, Bacteroidia, and Deltaproteobacteria influenced by Pb and Zn, while B and Cr influenced the distribution of Clostridia and Gammaproteobacteria, respectively. Identified antibiotic resistance genes included the peptidoglycan biosynthesis gene II, III, IV, and V, as well as the polymyxin resistance gene, while the virulence genes included the sitA, fimB, aerobactin synthase, and pilL gene. Our findings demonstrate that beach sand-borne bacteria are reservoirs of virulence and antibiotic resistance genes. Contamination of beach sands with heavy metals selects for both heavy metal resistance and antibiotic resistance in beach sand bacterial communities. Children and immunocompromised people engaging in recreational activities on beaches may be exposed to higher risk of infection.

Assessing multiple fecal sources to surf zone waters of two recreational beaches by bacterial community analysis

Abstract:Fecal sources to recreational surf zone waters should be identified to protect public health. While watershed origins of human and other fecal sources are often discoverable by quantitative polymerase chain reaction (qPCR) of fecal markers using spatially stratified samples, similarly assessing wastewater treatment plant (WWTP) outfall and other offshore contributions to surf zones is challenged by individual marker fate and transport. Here, bacterial communities were assessed for relatedness between all hypothesized fecal sources and surf zone waters for two urban California recreational beaches, by sequencing genes encoding 16S rRNA and analyzing data using SourceTracker and FEAST. Ambient marine bacterial communities dominated the surf zone, while fecal (human, dog, or gull) or wastewater (sewage or treated WWTP effluent) bacterial communities were present at low proportions and those from recycled water were absent. Based on the relative abundances of bacterial genera specifically associated with human feces, the abundances of HF183 in bacterial community sequences, and FEAST and SourceTracker results when benchmarked to HF183, the major sources of HF183 to surf zone waters were human feces and treated WWTP effluent. While surf zone sequence proportions from human sources (feces, sewage and treated WWTP effluent) appeared uncorrelated to previously obtained qPCR HF183 results, the proportions of human fecal and potential human pathogen sequences in surf zone waters were elevated when there were more swimmers (i.e. during weekday afternoons, holidays and busy weekends, and race events), thus confirming previously-published qPCR-based conclusions that bather shedding contributed low levels of human fecal contamination. Here, bacterial community sequencing also showed evidence that treated WWTP effluent from an offshore outfall was entering the surf zone, thereby resolving a prior uncertainty. Thus, bacterial community sequencing not only confirms qPCR HF183-based human marker detections, but further allows for confirming fecal sources for which individual marker quantification results can be equivocal.

Abstract:Beach health advisories are issued if enterococci (ENT) densities exceed the 30-d geometric mean or singlesample water quality criteria. Current ENT enumeration procedures require 1 day of incubation; therefore, beach managers make policy decisions using 1-day-old data. This is tantamount to using a model that assumes ENT density on day t is equal to ENT density on day t - 1. Research has shown that ENT densities vary over time scales shorterthan a day, calling into question the usefulness of the current model for decision-making. We created Dynamic Partial Least Square Regression (DPLSR) models for ENT at water quality monitoring stations within two adjacent marine recreational sites, Huntington State Beach (HSB) and Huntington City (HCB) Beach, California, using publicly available environmental data and tested whether these models overcome the drawbacks of the current model. The DPLSR models provide a better prediction of ENT than the current models based on comparisons of root mean-square errors of prediction and the numbers of type I and 2 errors. We compared outcomes in terms of predicted illness, swimmers deterred from entering the water, and net benefits to swimmers for hypothetical management scenarios where beach advisories were issued based on (a) the previously collected sample's ENT density in conjunction with the two water quality criteria, and (b) predictions from DPLSR models in conjunction with the singlesample standard. At both HSB and HCB the DPLSR scenario produced a more favorable balance between illness prevention and recreational access. The results call into question the current method of beach management and show that model-informed decision-making and elimination of the geometric mean standard will aid beach managers in achieving more favorable outcomes in terms of illness and access than are presently achieved using 1-day-old measurements, especially at beaches where water quality problems are chronic.
Seasonal dynamics of bacterial community structure and function in the surf zone seawater of a recreational beach in Ostend, Belgium

Abstract:This study involved year‐long, weekly monitoring of bacterial community composition and predicted functions in the surf zone seawater of a recreational beach in Ostend, Belgium, using full‐length 16S rRNA gene sequencing. The data were then correlated with environmental factors to identify potential drivers. Bacterial communities were significantly affected by seasonal variations in chlorophyll a (Chl a), net primary productivity (NPP), and seawater temperature (SWT), with minimal influence from faecal inputs due to beach recreational activities, as indicated by the ratio of Bacilli, Bacteroidota, and Clostridia to Alphaproteobacteria (BBC:A). This study provides a baseline for understanding how surf zone bacterial communities respond to environmental changes and impact health. Despite the importance of bacteria in surf zone water quality, detailed insights into their community composition, functions, and seasonal dynamics at recreational beaches are scarce. This study conducted year‐long, weekly monitoring of bacterial communities and environmental factors at a recreational beach in Ostend, Belgium. Using full‐length 16S rRNA gene sequencing, we correlated bacterial composition and predicted functions with environmental factors to identify potential drivers. Bacterial communities were significantly affected by seasonal variations in chlorophyll a (Chl a), net primary productivity (NPP), and seawater temperature (SWT), with minimal influence from faecal inputs due to human activities. Spring showed distinct abundances of Planktomarina, Amylibacter, and Sulfitobacter, positively correlated with Chl a and related to sulphur oxidation potential. Summer had higher abundances of Cryomorphaceae, likely enhancing chemoheterotrophy. Beginning in mid to late fall and extending into winter, bacterial communities underwent substantial changes. Fall featured a distinctive enrichment of Thioglobaceae, inversely correlated with Chl a. Winter was dominated by Methylophilaceae (OM43 clade), negatively correlated with Chl a, NPP, and SWT. Both seasons exhibited elevated levels of potentially pathogenic phenotypes and predicted functions related to methanol oxidation and methylotrophy. This study provides a baseline for understanding how surf zone bacterial communities respond to environmental changes and impact health.

Abstract:Recreational water activities have become an integral part of society for entertainment, health, and fitness. The water quality for these activities plays an integral role in the health and safety of the participants. This study aimed to monitor the presence of Escherichia coli in selected recreational water bodies, specifically rowing in the Gauteng Province of South Africa. Water samples were collected (upstream, midstream, and downstream) from three recreational water sites, i.e., Klip Rivier, Wemmer Pan, and Germiston Lake, monthly from May to October 2022. The presence of E. coli was determined using the Colilert® Quanti-Tray®/2,000 (IDEXX) assay and ChromoSelect agar. Pathotypes were confirmed using a multiplex PCR. The MPN for E. coli from all the samples exceeded the recommended guidelines of <2,000 microbes/100 mL. Pathogenic E. coli was detected in all three sites, with EPEC, ETEC, EAEC being most prevalent. The E. coli isolates showed 66% ESBL resistance and 94% carbapenemase resistance. Risk assessment showed recreational activities (rowing and swimming) posed a significant health risk as exceeded the annual risk benchmark limit of 1 × 104. The results obtained provide insights into the health risk associated with recreational activities within these water bodies and highlight the need for seasonal monitoring.

Abstract:Worldwide, fecal indicator bacteria (FIB) evidence coastal water contamination for which sources are unknown. Here, for two FIB-impacted Santa Barbara recreational beaches, hypothesized fecal sources were investigated over three dry seasons (summers) using nearly 2000 field samples of water (ocean, creek, groundwater), sand, sediments, effluent and fecal sources. In years 1 and 2, gull and dog feces were identified as the probable main FIB sources to surf zone waters, yet HF183 human fecal markers were consistently detected. Determining HF183 sources was therefore prioritized, via year 3 sub-studies. In lower watersheds, human and dog wastes were mobilized by small storms into creeks, but no storm drain outfalls or creeks discharged into surf zones. Beach area bathrooms, sewers, and a septic system were not sources: dye tracing discounted hydraulic connections, and shallow groundwater was uncontaminated. Sediments from coastal creeks and downstream scour ponds, nearshore marine sediments, and sands from inter- and supratidal zones contained neither HF183 nor pathogens. Two nearby wastewater treatment plant (WWTP) outfalls discharged HF183 into plumes that were either deep or distant with uncertain onshore transport. Regardless, local sources were evidenced, as surf zone HF183 detection rates mostly exceeded those offshore and nearshore (around boat anchorages). The presence of swimmers was associated with surf zone HF183, as swimmer counts (on weekdays, holidays, weekends, and during races) significantly correlated (p<0.05, n = 196) to HF183 detections. Besides comprehensively assessing all possible fecal sources, this study provides new explanations of chronic low-level human markers in recreational beach surf zones, suggesting likely lowest achievable HF183 thresholds.

Abstract:Standard methods to measure recreational water quality require at least 24 hr to obtain results, making it impossible to assess the quality of water within a single day. Methods to measure recreational water quality in <or=2 hr have been developed. Application of rapid methods could give considerably more accurate and timely assessments of recreational water quality. We conducted a prospective study of beachgoers at two Great Lakes beaches to examine the association between recreational water quality, obtained using rapid methods, and gastrointestinal (GI) illness after swimming. Beachgoers were asked about swimming and other beach activities and 10-12 days later were asked about the occurrence of GI symptoms. We tested water samples for Enterococcus and Bacteroides species using the quantitative polymerase chain reaction (PCR) method. We observed significant trends between increased GI illness and Enterococcus at the Lake Michigan beach and a positive trend for Enterococcus at the Lake Erie beach. The association remained significant for Enterococcus when the two beaches were combined. We observed a positive trend for Bacteroides at the Lake Erie beach, but no trend was observed at the Lake Michigan beach. Enterococcus samples collected at 0800 hr were predictive of GI illness that day. The association between Enterococcus and illness strengthened as time spent swimming in the water increased. This is the first study to show that water quality measured by rapid methods can predict swimming-associated health effects.

Abstract:Objectives We evaluated the potential impacts from using a rapid same-day quantitative polymerase chain reaction (qPCR) monitoring method for beach posting outcomes at two Toronto beaches. Methods In total, 228 water samples were collected at Marie Curtis Park East and Sunnyside Beaches over the 2021 summer season. Water samples were processed using the USEPA 1609.1 Enterococcus qPCR-based method. Escherichia coli ( E. coli ) culture data and daily beach posting decisions were obtained from Toronto Public Health. Results No significant correlation was observed between previous-day and same-day (retrospective) E. coli enumeration results at any Sunnyside Beach transect, and only relatively low ( R = 0.41–0.56) or no significant correlation was observed at sampling transects for Marie Curtis Park East Beach. Comparing our same-day Enterococcus qPCR data to Toronto's 2-day E. coli geometric mean beach posting decisions, we noted the need for additional postings for 1 (2%) and 3 (8%) missed health-risk days at Sunnyside and Marie Curtis Park East Beaches, respectively. The qPCR data also pointed to incorrect postings for 12 (31%) and 6 (16%) lost beach days at Sunnyside and Marie Curtis Park East Beaches, respectively. Conclusion Application of a rapid Enterococcus qPCR method at two Toronto beaches revealed 5% of beach posting decisions were false negatives that missed health-risk days, while 23% of decisions were false positives resulting in lost beach days. Deployment of the rapid same-day qPCR method offers the potential to reduce both health risks and unnecessary beach postings.
