Abstract:A new Schiff-base fluorescence probe CS containing benzimidazole and coumarin fluorophores was synthesized and characterized by NMR, IR and ESI-MS. Fluorescence and absorption spectra show the unique changes in the presence of Hg2+ and Cu2+. Hg2+ induces a remarkable fluorescence enhancement and results in mercury-promoted hydrolysis of a mercury-CS complex to convert the imine group to the aldehyde group. Cu2+ leads to a colorimetric change from colorless to pale yellow and causes formation of a copper-CS complex. Thus, CS can act as a fluorescent chemodosimeter for Hg2+ and a naked-eye chemosensor Cu2+. The cell imaging experiments for Hg2+ show a slow enhancement of a bright blue fluorescence with increasing incubation time, while Cu2+ instantly quenches the fluorescence and then the fluorescence intensity has no further change by extending the incubation time. Therefore, the probe CS shows its potential application for simultaneous differential detection of Hg2+ and Cu2+ in living cells based on the fluorescence response time. (C) 2016 Elsevier B.V. Abstract:Mercury(ii), which is a well-known toxic species, exists in the industrial waste water in many cases. In the present work, CdTe quantum dots (QDs) are studied as a fluorescence probe for Hg2+ detection. Ag ions are induced to QDs to enlarge their detection concentration range. l-cysteine is employed in the QD-based fluorescence probe to connect QDs with Hg2+. X-ray diffraction, transmission electron microscopy and X-ray photoelectron spectroscopy results indicate the formation of zinc blende CdTe QDs with sizes of ∼5 nm and the existence of Ag+ in crystalline CdTe. Photoluminescence (PL) spectra and PL decay spectra were acquired to investigate the emission mechanism of Ag-doped CdTe QDs, revealing multi-emission in QD samples with higher concentrations of Ag+ doping. The highest PL quantum yield of the QD samples was 59.4%. Furthermore, the relationship between the fluorescence intensity and the concentration of Hg2+ has been established. Two linear relationships were obtained for the plot of F/F0 against Hg2+ concentration, enlarging the detection concentration range of Hg2+.

Abstract:Mercury (Hg) is a heavy metal with high toxicity and easy migration; it can be enriched through the food chain, and cause serious threats to the natural environment and human health. So, the development of a method that can be used to detect mercury ions (Hg2+) in the environment, in cells, and in organisms is very important. Here, a new 7-hydroxycoumarin-derived carbonothioate-based probe (CC-Hg) was designed and synthesized for detection of Hg2+. After addition of Hg2+, a large fluorescence enhancement was observed due to the formation of 7-hydroxyl, which reinforced the intramolecular charge transfer process. The CC-Hg probe had good water solubility and selectivity. Moreover, the probe was able to detect Hg2+ quantitatively over the concentration range 0-2 mu M and with a detection limit of 7.9 nM. Importantly, we successfully applied the probe to detect Hg2+ in water samples, in living cells, and in zebrafish. The experimental results demonstrated its potential value in practical applications.

Abstract:A stable silver nanoparticles/carbon quantum dots (Ag/CQDs) composite was prepared by using CQDs as reducing and stabilizing agent. The CQDs synthesized with polyethyleneimine (PEI) showed an extraordinary reducibility. When Hg2+ was presented in the Ag/CQDs composite solution, a color change from yellow to colorless was observed, accompanied by a shift of surface plasmon resonance (SPR) band and decrease in absorbance of the Ag/CQDs composite. On the basis of the further studies on TEM, XPS and XRD analysis, the possible mechanism is attributed to the formation of a silver-mercury amalgam. Hence, a two dimensional sensing platform for Hg2+ detection was constructed upon the Ag/CQDs composite. Based on the change of absorbance, a good linear relationship was obtained from 0.5 to 50μM for Hg2+. And the limit of detection for Hg2+ was as low as 85nM, representing high sensitivity to Hg2+. More importantly, the proposed method also exhibits a good selectivity toward Hg2+ over other metal ions. Besides, this strategy demonstrates practicability for the detection of Hg2+ in real water samples with satisfactory results.
A signal-on fluorescent sensor for ultra-trace detection of Hg2+ via Ag+ mediated sulfhydryl functionalized carbon dots

Abstract:Sulfhydryl functionalized carbon dots (HS-CDs) mediated by Ag+ were used to develop a fluorescent sensor for the determination of Hg2+ in aqueous solutions. FTIR and XPS confirmed the successful modification of -SH on the surface of HS-CDs. Dripping Ag+ into HS-CDs yielded brown agglomerates of silver thiolate leading to strong FL quenching of the HS-CDs. The signal-on fluorescent sensor was acheived by dispersing the precipitates in a PBS buffer (pH 7.4, 0.01 mol/L). Remarkable FL recovery was observed upon the addition of Hg2+ into the sensor owing to the energy trap induced by exposure of -NH2 on the surface of the HS-CDs. Fluorescence intensities of the HS-CDs were linearly recovered in Hg2+ concentrations of 0.01–0.75 nmol/L with an extremely low detection limit of 4.2 pmol/L. The sensor exhibited stronger specificity toward Hg2+ than co-existent metal ions. The detecting results for real water samples obtained by the sensor showed good agreement with ICP-AES (P > 0.05), demonstrating a sensitive and selective fluorescent nanosensor for ultra-trace determination of Hg2+ with acceptable precision and satisfactory accuracy.
An Improved Synthesis of Water-Soluble Dual Fluorescence Emission Carbon Dots from Holly Leaves for Accurate Detection of Mercury Ions in Living Cells.

Abstract:Background Carbon dots (CDs) emitting near-infrared fluorescence were recently synthesized from green leaves. However, the Hg2+ detection of CDs was limited because of the insufficient water solubility, low fluorescence and poor stability. Methods Dual fluorescence emission water-soluble CD (Dual-CD) was prepared through a solvothermal method from holly leaves and low toxic PEI1.8k. PEG was further grafted onto the surface to improve the water solubility and stability. Results The Dual-CD solution can emit 487 nm and 676 nm fluorescence under single excitation and exhibit high quantum yield of 16.8%. The fluorescence at 678 nm decreased remarkably while the emission at 470 nm was slightly affected by the addition of Hg2+. The ratiometric Hg2+ detection had a wide linear range of 0–100 μM and low detection limit of 14.0 nM. In A549 cells, there was a good linear relation between F487/F676 and the concentration of Hg2+ in the range of 0–60 μM; the detection limit was 477 nM. Furthermore, Dual-CD showed visual fluorescence change under Hg2+. Conclusion Dual-CD has ratiometric responsiveness to Hg2+ and can be applied for quantitative Hg2+ detection in living cells.

Abstract:Abstractl-Tryptophan-capped carbon quantum dots (l-CQDs) were facilely synthesized through “green” methodology, and the obtained material was utilized as a sensitive and selective fluorescence sensor for mercury ion (Hg2+) in pure aqueous solutions. Carboxyl-functionalized CQDs were first green synthesized by a one-step hydrothermal route, and l-tryptophan was then attached to CQDs via direct surface condensation reaction in aqueous solution at room temperature. The as-synthesized l-CQDs had an average size of ca. 5 nm with a good dispersity in water, and exhibited a favorable selectivity for Hg2+ ions over a range of other common metal cations in aqueous solution (10 mM PBS buffer, pH 6.0). Upon the addition of Hg2+, a complete fluorescence quenching (ON–OFF switching) of l-CQDs was evident from the fluorescence titration experiment, and the fluorescence detection limit of Hg2+ was calculated to be 11 nM, which indicated that the obtained environmentally friendly l-CQDs had sensitive detection capacity for Hg2+ in aqueous solution.

Abstract:Herein, we first reported a facile synthesis method for fabrication of highly photoluminescent carbon quantum dots (CQDs) using sodium alginate as the carbon source and histidine as both the nitrogen source and functional monomer by one-pot hydrothermal synthesis. The as-prepared CQDs gave a high quantum yield of 32%. By employing the new CQDs and rhodamine B (RhB), we demonstrated a simple, facile, sensitive, and accurate ratiometric sensor for detection of Hg2+ and biological thiols. The photoluminescence of CQDs in the ratiometric sensor can be selectively and intensively suppressed by Hg2+ due to strong electrostatic interaction between the surface functional groups of the CQDs and Hg2+. When glutathione (GSH) was introduced into the “Turn Off” CQDs-RhB-Hg2+ sensing system, the fluorescence of the CQDs can be recovered rapidly due to the stronger affinity between thiol and Hg2+, while the fluorescence of the RhB remained constant in this sensing process. Based on the above principle, the ratiometric strategy for detecting Hg2+ and GSH can be achieved readily, and gives satisfactory limit of detections (LODs) of 30 and 20 nM for Hg2+ and GSH, respectively. The dual-emission fluorescent CQDs-RhB sensor does not need the complicated molecular design and the synthesis of dual-emission fluorophores. Meanwhile, the feasibility of the proposed method for analysis of water samples, food samples, and biological samples (plasma from mice oxidative stress study) was investigated. Abstract:Mercury ions (Hg 2+ ) are a class of heavy metal ions that can cause serious harm to the human body due to their toxicity, fluidity, and bioaccumulation. Direct contact with Hg 2+ can cause adverse effects on the human body. In addition, long-term exposure to Hg 2+ can lead to extreme toxicity of enzymes and proteins in the body, disrupting the cell cycle and thus endangering human life. It is becoming increasingly important to develop a method that is reliable, stable, and sensitive for detecting Hg 2+ . In this study, a near-infrared "turn-on" fluorescent probe based on dicyanomethylene-4 H -pyran, DCM-Hg, was designed and synthesized to determine Hg 2+ over other metal ions. Probe DCM-Hg had a detection limit (LOD) of 3.35 μM when used to detect Hg 2+ . The Hg 2+ concentrations and the fluorescence intensities at 695 nm showed a good linear correlation. Additionally, Hg 2+ in HepG2 cells was successfully visualized using the current probe.

Abstract:In this study, a highly selective and sensitive composite fluorescence sensor (Y-CODs @PVDF@PVP) was developed for the quantitative and visual detection of Hg 2+ by electrostatic spinning carbon quantum dots (CQDs) with polyvinylidene fluoride (PVDF) and polyethylene pyrrolidone (PVP). The Y-CODs @PVDF@PVP composite fluorescence sensor exhibited strong yellow fluorescence under ultraviolet light (365 nm), demonstrating stable detection capability for mercury (II) in water samples. The morphology, chemical composition, and optical properties of Y-CODs @PVDF@PVP were thoroughly investigated. Different quenching effects were observed in Y-CODs @PVDF@PVP based on varying concentrations of Hg 2+ , enabling accurate determination of Hg 2+ . The achieved detection limit was 9.27 nM. Both static quenching and dynamic quenching mechanisms contributed to the observed results. Notably, the method successfully detected Hg 2+ in real water samples from Beichuan Lake as well as rice samples, yielding satisfactory outcomes. Furthermore, when combined with software image analysis, quantitative and visual detection of field Hg 2+ levels was accomplished using the composite fluorescence sensor. This research provides valuable insights for developing portable sensors aimed at monitoring harmful substances in water.

Abstract:Nitrogen- and sulfur-codoped carbon dots (N,S-CDs) with a fluorescence quantum yield of 16.1% and good photoluminescent properties were synthesized by a simple hydrothermal method. Cytotoxicity of the N,S-CDs was evaluated by the MTT assay, and human hepatoma HepG2 cells were chosen as the target. The cell viability was more than 85% after 24 h of incubation when its concentration was up to 300 mu g/mL, suggesting low cytotoxicity and good biocompatibility of the N,S-CDs. The fluorescence spectra of the N,S-CDs are excitation-dependent in the excitation-wavelength range of 295-400 nm, and it emits bright blue fluorescence centered at 435 nm. Its selective fluorescence recognition for Hg(2)(+ )ions was found. When Hg(2+ )ions were added to the N,S-CDs solution, its bright blue fluorescence was obviously quenched and could be recovered by the addition of sulfide. Accordingly, a new strategy based on N,S-CDs-Hg(2+ )system as a highly selective and ultrasensitive "turn off-on" fluorescence sensing for the detection of sulfide was fabricated. The limits of detection (S/N = 3) are 83 nM for Hg(2+ )ions and 11 nM for sulfide. Almost no statistically significant interference for Hg2+-ion and sulfide detection was observed among possible coexisting substances in the water samples, including 17 common metal ions and 11 anions. This probe was successfully applied for the cellular imaging of Hg(2+ )ions in HepG2 cells by fluorescence microscopy.

Abstract:Mercury ion (Hg2+) is considered a harmful neurotoxin, and real-time monitoring of Hg2+ concentrations in environmental and biological samples is critical. Fluorescent probes are a rapidly emerging visualization tool owing to their simple design and good selectivity. Herein, a novel fluorescence (FL) probe 2-(4-((6-((quinolin-8-yloxy)methyl)pyridin-2-yl)methyl)piperazin-1-yl)anthracene-9,10-dione (QPPA) is designed using piperazine as a linker between the anthraquinone group, which serves as a fluorophore, and N4O as the Hg2+ ligand. The probe exhibits FL "turn-on" sensing of Hg2+ because the complex inhibits the photo-induced electron transfer (PET) process. Moreover, QPPA can overcome the invasion by other possible cations, resulting in a clear color change from orange to colorless with the addition Hg2+. The chelation of QPPA with Hg2+ in a 1:1 ratio. Subsequently, the theoretically determined binding sites of the ligand to Hg2+ are validated through 1H NMR titration. The in situQPPA-Hg2+ complex can be subjected to Hg2+ extraction following the introduction of S2- owing to its robust binding capacity. The exceptional limit of detection values for Hg2+ and S2- are obtained as 63.0 and 79.1 nM (S/N = 3), respectively. Moreover, QPPA can display bright red FL in the presence of Hg2+ in different biological specimens such as HeLa cells, zebrafish, onion root tip tissues, and water flea Daphnia carinata, further providing an effective strategy for environmental monitoring and bioimaging of Hg2+ in living organisms.
Highly selective and sensitive detection of Hg2+ by a novel fluorescent probe with dual recognition sites

Abstract:A novel thionocarbonate-coumarin-thiourea triad-based probe with dual recognition sites for sensing mercury (Hg2+) ion was developed. The synthesized probe possessed both fluorogenic ("off-on") and chromogenic (from colorless to blackish brown) sensing performance towards Hg2+ ions. The fluorescence intensity was increased by 70 fold after the addition of Hg2+. As expected, the probe exhibited excellent selectivity and sensitivity for Hg2+ compared to other common competitive metal ions. The fluorescence intensity of the probe improved linearly with the increase of the concentration of Hg2+ (0-40 μM). Also, the minimum limit of detection (LOD) of the synthesized probe was 0.12 μM. Considering the importance of test feasibility in the harsh environment, the developed probe was applicable for detecting Hg2+ ions over a broad working pH range of 3-11. It is reliable and qualifies for the quantitative determination of Hg2+ concentrations in actual water samples. Finally, the probe achieved the bioimaging performance of Hg2+ in living cells and plants with good biocompatibility.

Abstract:A fluorescent probe (probe C) based on coumarin-thiourea conjugate was developed for selective detection of Hg 2+ . Addition of Hg 2+ to probe C induced obvious enhancement of the fluorescence intensity. Probe C exhibited good selectivity toward Hg 2+ and good anti-interference performance over other common competitive metal ions including Na + , K + , Ag + , Cu 2+ , Zn 2+ , Co 2+ , Ni 2+ , Mn 2+ , Mg 2+ , Fe 2+ , Fe 3+ , Ca 2+ and Al 3+ . The detection limit of probe C for Hg 2+ was calculated to be 0.44 nM. It was capable of detecting Hg 2+ ion in a wide pH range of 1.0–10.0.
Selective detection of mercury ions via single and dual signal by silicon-doped carbon quantum dots

Abstract:Si-CQDs with uniform particle size and good water solubility were synthesised using low-cost citric acid (CA) and aminopropyltriethoxysilane (APTEOS) as precursors. Si-CQDs was used as a novel fluorescence probe for sensitive and selective mercury ions (Hg2+) detection by fluorescence quenching. Following the optimising of the experimental conditions, the fluorescence intensity of Si-CQDs was found to be linearly dependent on the Hg2+ concentration ranging from 0.02 to 5.0 μM with the limit of detection as low as 0.015 μM. A fluorescence resonance energy transfer (FRET) effect between Si-CQDs and Rhodamine B (RhB) was also observed and it was found that Si-CQDs can be used as fluorescence donors and RhB as fluorescence receptors. On this basis, a ratio fluorescent probe (Si-CQDs/RhB) was constructed for Hg2+ detection. When Hg2+ concentration was 1.0-80.0 μM, the fluorescence intensity ratio (I540nm/I440nm) of Si-CQDs/RhB exhibited a good linear relationship with Hg2+ concentration and the detection limit was 0.6 μM. A wide range of Hg2+ detection was ultimately achieved in real samples.

A bright carbon-dot-based fluorescent probe for selective and sensitive detection of mercury ions

Abstract:In this work, we demonstrated a convenient and green strategy for the synthesis of bright and water-soluble carbon dots (CDs) by carbonizing sodium citrate and glutathione together in a hydrothermal method for the first time. Without post surface modification, the as-synthesized CDs display fluorescence quantum yield (QY) as high as 21.03% and show superior stability not only in concentrated salt solutions but also in neutral and alkaline media. The probe exhibits selective and sensitive recognition capability towards mercury ion (Hg2+) in aqueous solution. The fluorescence of CDs can be quenched by Hg2+ through an effective electron energy transfer process. It displays a linear quenching effect toward mercury ion in the concentration range of 0-15μM with a correlation coefficient (R2) of 0.99. The limit of detection is determined to be 25nM at the signal to noise ratio of 3. These attractive merits would enable the extensive applications of this probe in environmental science and analytical chemistry in the future.

Abstract:In this study, carbon quantum dots (CQDs) and rhodamine B (RhB) were physically mixed to construct a highly selective and sensitive ratiometric fluorescent sensor (CQDs@RhB) for the quantitative and visual detection of Hg 2+ . After adding Hg 2+ , the emission peak of CQDs at 445 nm was quenched and the peak of RhB at 575 nm remained unchanged; the ratio of the two emission peaks had a linear relationship with the Hg 2+ concentration. The CQDs@RhB had a detection range of 0–15 μM and the limit of detection (LOD) is 65.93 nM and a color change from blue to pink was visible to the naked eye under ultraviolet light. The study revealed a static quenching mechanism of CQDs@RhB. The established method was successfully applied to the detection of Hg 2+ in real water and rice samples with satisfactory results. In addition, a portable fluorescent paper-based sensor platform was developed using computer software assistance. The relationship with Hg 2+ concentration was constructed by converting the pictures obtained under ultraviolet light to RGB color mode for visualization and quantitative detection of Hg 2+ . This study provides a valuable strategy for constructing a rapid detection system for Hg 2+ in the field.

Abstract:Mercury ions are highly toxic to living organisms and environment, hence Monitoring and quantification of mercury ions remain great significance. Herein, we have designed and synthesized a novel reaction-based fluorescence probe RANS for detection of Hg2+ by using a xanthene derivate as fluorophore and a thiosemicarbazide moiety as the recognition site. By applying mercury-promoted desulfurization reactions, probe RANS exhibited excellent capability of detecting Hg2+, such as rapid response (120 s), high sensitivity (0.019 μM) and good selectivity. Upon reaction with Hg2+, the thiosemicarbazide group of probe RANS formed 1,3,4-oxadiazole along with a significant color change from colorless to light green. Importantly, RANS has been successfully applied to selectively imaging Hg2+ in living cells with low cytotoxicity and quantitative detection of Hg2+ in real water samples.

Abstract:Construction of efficient chemosensors for highly specific and sensitive detection of mercury ions remains a great challenge. In this work a highly selective and sensitive probe CY was designed and synthesized by using coumarin fluorophore as the matrix and thioacetal moiety as the reactive recognition site for Hg 2+ . By virtue of the thiophilicity of Hg 2+ , probe CL could be hydrolyzed to deprotect and the thioacetal was transformed to the acyl group after the addition of Hg 2+ , the blue-green fluorescence was quenched and meanwhile the solution changed from light green to yellow. The detection limit of probe CY for Hg 2+ was as low as 6.8 nM, and it could completely react with Hg 2+ within 3 min. Moreover, probe CY exhibited good resistance against interference from competitive metal ions and biothiols, high stability in pH 1–11 and applicability for fluorogenic and chromogenic dual-modal detection of Hg 2+ in real water samples over a broad range of pH 5–10.
