Abstract:—The allelic composition of genes encoding high molecular weight glutenin subunits was studied in a collection composed of 236 winter and 98 spring wheat samples of Belarussian and foreign breeding. We have identified 13 alleles among winter and 11 alleles among spring wheat samples. This is generally less than the data for European, Asian, and African countries. In total, 36 different genotypes have been identified among the analyzed samples. The most frequent cultivars and lines among the winter wheat samples possessed the following genotypes: Glu-A1c , Glu-B1c , Glu-D1d (16.2% of the total number of the analyzed samples), Glu-A1a , Glu-B1c , Glu-D1d (13.7%), and Glu-A1b , Glu-B1c , Glu-D1d (12.7%). The cultivars and lines possessing the Glu-A1a , Glu-B1c , Glu-D1d (22.5%), Glu-A1a , Glu-B1c , Glu-D1a (16.4%), and Glu-A1c , Glu-B1f , and Glu-D1d (13.3%) genotypes prevailed among the spring wheat samples. The spring wheat KP-406/11 line genome has been found to carry a Bx subunit-encoding gene allele with the maximum identity degree (99%) relative to the nucleotide sequences of the Bx subunits represented under the GenBank database. This gene allele contains an open reading frame with the length of 2367 bp and has the highest degree of identity with the nucleotide sequence of the Bx14 subunit. The 789-long amino acid sequence encoded by this allele differs from the Bx14 by replacements of amino acid residues in three positions: 662, 780, and 788. This subunit was designated Bx14.1 (its Genbank accession number is MH108092).

Assessment of the genetic diversity of the alleles of gliadin-coding loci in common wheat (Triticum aestivum L.) collections in Kazakhstan and Russia

Abstract:The study of genetic resources using prolamin polymorphism in wheat cultivars from countries with different climatic conditions makes it possible to identify and trace the preference for the selection of the alleles of gliadine-coding loci characteristic of specific conditions. The aim of the study was to determine the "gliadin profile" of the collection of common wheat (Triticum aestivum L.) from breeding centers in Russia and Kazakhstan by studying the genetic diversity of allelic variants of gliadin-coding loci. Intrapopulation (μ ± Sμ) and genetic (H) diversity, the proportion of rare alleles (h ± Sh), identity criterion (I) and genetic similarity (r) of common wheat from eight breeding centers in Russia and Kazakhstan have been calculated. It has been ascertained that the samples of common wheat bred in Kostanay region (Karabalyk Agricultural Experimental Station, Kazakhstan) and Chelyabinsk region (Chelyabinsk Research Institute of Agriculture, Russia) had the highest intrapopulation diversity of gliadin alleles. The proportion of rare alleles (h) at Gli-B1 and Gli-D1 loci was the highest in the wheat cultivars bred by the Federal Center of Agriculture Research of the South-East Region (Saratov region, Russia), which is explained by a high frequency of occurrence of Gli-B1e (86 %) and Gli-D1a (89.9 %) alleles. Based on identity criterion (I), the studied samples of common wheat from different regions of Kazakhstan and Russia have differences in gliadin-coding loci. The highest value of I = 619.0 was found when comparing wheat samples originated from Kostanay and Saratov regions, and the lowest I = 114.4, for wheat cultivars from Tyumen and Chelyabinsk regions. Some region-specific gliadin alleles in wheat samples have been identified. A combination of Gli-A1f, Gli-B1e and Gli-Da alleles has been identified in the majority of wheat samples from Kazakhstan and Russia. Alleles (Gli-A1f, Gli-A1i, Gli-A1m, Gli-A1o, Gli-B1e, Gli-D1a, Gli-D1f, Gli-A2q, Gli-B2o, and Gli-D2a) turned out to be characteristic and were found with varying frequency in wheat cultivars in eight regions of Russia and Kazakhstan. The highest intravarietal polymorphism (51.1 %) was observed in wheat cultivars bred in Omsk region (Russia) and the lowest (16.6 %), in Pavlodar region (Kazakhstan). On the basis of the allele frequencies, a "gliadin profile" of wheat from various regions and breeding institutions of Russia and Kazakhstan was compiled, which can be used for the selection of parent pairs in the breeding process, the control of cultivars during reproduction, as well as for assessing varietal purity.
Diversity of alleles of locuses of spare proteins of wheat varieties IFRG NAS of Ukraine

Abstract:Aim. Investigation of allele frequencies by locus of reserve proteins in groups of wheat varieties created during different selection periods at the leading breeding center of the Institute of Plant Physiology and Genetics of the National Academy of Sciences of Ukraine. Methods. The material for the study were 57 varieties of wheat of the Institute of Plant Physiology and Genetics of the National Academy of Sciences of Ukraine. Separation of gliadins was performed according to the ISTA method in the Poperel modification. High molecular weight glutenin subunits were analyzed by electrophoresis in the presence of sodium dodecyl sulfate (SDS) by the Lemmley method. Results. The allelic composition of loci of spare proteins Glu-A1, Glu-B1, Glu-D1, Gli-1A, Gli-B1, Gli-1D was determined. At the gliad-encoding loci Gli-A1, Gli-B1 6 alleles were detected, at the locus Gli-D1 - 5 alleles. By gluten-encoding loci, the most polymorphic was the Glu-B1 locus, which is represented by 5 alleles. Loci Glu-A1 and Glu-D1 are represented by 3 and 2 allelic variants. Conclusions. The frequency of the Glu-B1al allele, increased from 0% to 18%, this allele is one of the strongest in terms of positive impact on the quality of flour among the identified alleles of loci of high molecular weight glutenins of wheat. It was found that almost 50% of varieties created in the last decade in IFRG NAS of Ukraine have wheat-rye translocations 1AL.1RS, 1BL.1RS.

Abstract:Among a vast number of polymorphisms in the wheat genome, of special interest are those that are markers for loci under selection. Such loci include storage protein loci, which directly determine the level of bread-making quality, and disease resistance genes. In this study we analyzed diversity of functional markers (seven storage protein loci and three disease resistance genes) in the group of East European winter common wheat genotypes (cultivars and lines of Poltava breeding). Using the UPGMA or NJ methods, the cultivars and lines were divided into two clusters differing in the Glu-1 quality score. The cultivars of cluster 1 with the lower mean Glu-1 quality score predominantly carried the combination of alleles Glu-B1c , Glu-D1a , and Gli-B1e , as well as the resistance allele of Lr34 , whereas those of cluster 2 with the higher quality score mainly had the alleles Glu-B1u , Glu-D1d , and Gli-B1b. In the sample of Poltava cultivars, a high correlation between the Glu-1 quality score and the sedimentation value (0.84) was observed. On average for two years, the group of cultivars from cluster 1 showed higher grain protein content and some spike traits in comparison with the means of the sample from cluster 2. Significant associations of Lr34 alleles and alleles at some storage protein loci were revealed, in particular, the association of the resistance allele Lr34R and the allele Glu-D1a in both cultivar and line groups, indicating the adaptive value of such a combination for local soil and climate conditions.

Abstract:Wheat landraces have abundant genetic variation at the Glu-1 loci, which is desirable germplasms for genetic enhancement of modern wheat varieties, especially for quality improvement. In the current study, we analyzed the allelic variations of the Glu-1 loci of 597 landraces and 926 commercial wheat varieties from the four major wheat-growing regions in China using SDS-PAGE. As results, alleles Null, 7+8, and 2+12 were the dominant HMW-GSs in wheat landraces. Compared to landraces, the commercial varieties contain higher frequencies of high-quality alleles, including 1, 7+9, 14+15 and 5+10. The genetic diversity of the four commercial wheat populations (alleles per locus (A) = 7.33, percent polymorphic loci (P) = 1.00, effective number of alleles per locus (Ae) = 2.347 and expected heterozygosity (He) = 0.563) was significantly higher than that of the landraces population, with the highest genetic diversity found in the Southwestern Winter Wheat Region population. The genetic diversity of HMW-GS is mainly present within the landraces and commercial wheat populations instead of between populations. The landraces were rich in rare subunits or alleles may provide germplasm resources for improving the quality of modern wheat.
Identification of QTLs for Grain Protein Content in Russian Spring Wheat Varieties

Abstract:Most modern breeding programs aim to develop wheat (T. aestivum L.) varieties with a high grain protein content (GPC) due to its greater milling and cooking quality, and improved grain price. Here, we used a genome-wide association study (GWAS) to map single nucleotide polymorphisms (SNPs) associated with GPC in 93 spring bread wheat varieties developed by eight Russian Breeding Centers. The varieties were evaluated for GPC, grain weight per spike (GWS), and thousand-kernel weight (TKW) at six environments, and genotyped with 9351 polymorphic SNPs and two SNPs associated with the NAM-A1 gene. GPC varied from 9.8 to 20.0%, depending on the genotype and environment. Nearly 52% of the genotypes had a GPC > 14.5%, which is the threshold value for entry into high-class wheat varieties. Broad-sense heritability for GPC was moderate (0.42), which is due to the significant effect of environment and genotype × environment interactions. GWAS performed on mean GPC evaluated across six environments identified eleven significant marker-trait associations, of which nine were physically mapped on chromosome 6A. Screening of wheat varieties for allelic variants of the NAM-A1 gene indicated that 60% of the varieties contained the NAM-A1c allele, followed by 33% for NAM-A1d, and 5% for NAM-A1a alleles. Varieties with the NAM-A1d allele showed significantly (p < 0.01) smaller GPC than those with NAM-A1c and NAM-A1a. However, no significant differences between NAM-A1 alleles were observed for both GWS and TKW.

Abstract:Glutenins, the polymeric proteins are composed of high molecular weight (HMW) and low molecular weight (LMW) glutenin subunits. High molecular weight glutenin subunit (HMW-GS) plays a key role in determining bread making quality. The HMW-GS encoded by Glu-1 loci (GluA1, GluB1 and GluD1) are highly polymorphic. Thus it is very important to identify the alleles at Glu-1 loci for all three genomes for wheat quality improvement. In the present study 53 Indian wheat genotypes were evaluated for HMW-GS composition using PCR-based DNA markers. In total, two alleles (Ax2*, Ax1 or Axnull) at Glu-A1, four allele/allelic pairs (Bx7, Bx7 + By8, Bx7 + By9, Bx17 + By18) at Glu-B1 and two allelic pairs (Dx2 + Dy12 and Dx5 + Dy10) at Glu-D1 were identified using diagnostic DNA markers. Allele or allelic pair Ax2* (62.3 %) at Glu-A1, Bx7 + By8 (64 %) at Glu-B1 and Dx2 + Dy12 (58.5 %) at Glu-D1 were present most frequently in wheat genotypes taken for study. Moreover, the allelic combination Ax2*, Bx17 + By18 and Dx5 + Dy10 exhibited highest bread loaf volume in comparison to other allelic combinations studied. The set of markers used here thus represents a useful alternative for screening of HMW-glutenin allele (s) for improvement of bread making quality in Indian wheat genotypes.

Abstract:The baking qualities of 36 varieties of wheat are considered. Storage proteins — high molecular weight glutenin subunits (HMW-GS) — were studied. Electropherograms were processed in the GelPro Analyzer 4.0 computer program. The molecular weight of glutenin subunits was determined according to the method of G. Galili, M. G. Feldman using a marker variety. The quality of glutenin was assessed in points according to the method of O. Lukov. The bread was baked using the straight method, in accordance with the generally accepted method of laboratory baking; the fermentation of the dough and further baking were carried out on Domino and Arianna devices. As a result of the research, it was revealed that only 7 test samples (Askeran, Azemetli 95, Gonen, Gyrmyzy Gul, Nurlu99, Tale38, Ugur) with a null allele, having subunit 1 (Glu-A1a allele), have a fragment of 344 bp in size, in the remaining 29 samples encoded by the Glu-A1 locus, having subunit 2* (Glu-A1b allele), a fragment of 362 bp in size is amplified. The presence of a null allele does not always have a negative effect on baking quality. The reason for this remains to be determined.

Abstract:Background . High-molecular-weight glutenin subunits (HMW-GSs) make the greatest contribution to the formation of baking properties in bread wheat ( Triticum aestivum L.). Glutenin polymorphism of bread wheat relatives is significantly richer than in cultivated varieties. The objective of this work was to identify the HMW-GS composition and assess the gluten quality of bread wheat lines with introgressions of alien genetic material. Materials and methods . We studied the parental varieties of spring bread wheat, accessions of the tetraploid and hexaploid Triticum L . species, and 19 introgressive lines produced with their participation. Glutenins were separated using sodium dodecyl sulfate polyacrylamide gel electrophoresis and HMW-GSs were identified using the Payne nomenclature system. Gluten quality was determined in accordance with GOST 13586.1-68 (the 2017–2019 and 2021–2022 growing seasons). Data processing was performed using the Statistica 10.0 and MS Excel software packages. Results and conclusion . HMW-GSs uncharacteristic of T. aestivum cultivars were identified in the studied accessions of T. dicoccoides (Körn. ex Aschers. et Graebn.) Schweinf., T. spelta L., and T. kiharae Dorof . et Migusch. These HMW-GSs are of interest for enriching the wheat gene pool. HMW-GSs of related species were found in 10 out of 19 introgressive lines. A five-year observation period revealed that wheat relatives on average significantly exceeded common wheat cultivars in gluten quality, and introgressive lines did not differ significantly from the parental genotypes. Lines with high rheological properties of gluten had, as a rule, HMW-GSs of related species in their composition.
Genetic Diversity of Wheat Storage Proteins and Bread Wheat Quality

Abstract:To understand the genetic and biochemical basis of the bread making quality of wheat varieties a large experiment was carried out with a set of 162 hexaploid bread wheat varieties registered in the French or European Wheat Catalogue. This material was used to analyse their allelic composition at the twelve main storage protein loci. A large genetic and biochemical diversity of the gluten proteins was found. Several gliadin encoding loci exhibited the highest allelic diversity whereas the lowest diversity was found for Glu-Al and Glu-D3 loci encoding some high molecular weight glutenin subunits (HMW-GS) and LMW-GS respectively. The varieties were grown in three experimental locations in France. Quality evaluation was carried out from material harvested in each location using seven technological tests: grain protein content (Prot), grain hardness (GH), Zeleny sedimentation test (Zel), Pelshenke test (Pel), water soluble pentosans (relative viscosity: Vr ), mixograph test (giving 11 parameters) and the alveograph test (dough strength W, tenacity P , extensibility L, swelling G, ratio P/L and the elasticity index Ie). Genetic and location effects as well as broad-sense heritability of each of the 22 technological parameters were calculated. GH, corresponding to the major Ha gene, Pel, and MtxW (mixograph parameter) had the highest heritability coefficients, alveograph parameters like W, P, the relative viscosity Vr and several mixograph parameters had medium heritability coefficients whereas Prot and L had the lowest. Variance analysis (using GLM procedure) allowed the effect of the allelic diversity of the storage proteins, on the genetic variations of each quality parameters, to be estimated. Glu-1 and Glu-3 loci had significant additive effects in the genetic variations of many parameters. Gliadin alleles encoded at Gli-1 and Gli-2 were also found to play significant effect on several quality parameters. The major part of the phenotypic variation of the different quality parameters like Zel, Pel, W or mixograph peak time MPT was explained with the GH and alleles encoded at Glu-1 and Glu-3. Allelic variants encoded at Glu3 and Gli-2 had similar contribution to the phenotypic variations of quality parameters and accounted for 4% up to 21% each .

The variability detected could affect gluten quality.AbstractLow-molecular weight glutenin subunits are important in determining the viscoelastic properties of wheat dough. Triticum urartu Thum. ex Gandil., which is related to the A genome of polyploid wheat, has been shown as a good source of variation for these subunits. The present study evaluated the variability of LMW-i genes in this species. High polymorphism was found in the sequences analysed and resulted in the detection of 11 novel alleles, classified into two sets (Group-I and -II) showing unique SNPs and InDels. Both groups were associated with Glu-A3-1 genes from common wheat. In general, deduced proteins from Group-II genes possessed a higher proportion of glutamine and proline, which has been previously suggested to be related with good quality. Moreover, there were other changes compared to common wheat. This novel variation could affect dough quality. Additional epitopes for celiac disease were also detected, suggesting that these subunits could be highly reactive. The results showed that T. urartu could be an important source of genetic variability for LMW-i genes that could enlarge the genetic pool of modern wheat.

Distribution of Low-molecular-weight Glutenin Subunit Glu-A3 Alleles in Mini Core Collections of Chinese Wheat Germplasm

Abstract:Low-molecular-weight glutenin subunit(LMW-GS) plays an important role in the processing quality of wheat for bread and noodle.A total of 208 accessions of common wheat comprising both landraces and improved cultivars collected from all the three growing zones in China were tested for the seven alleles at the Glu-A3 locus using 7 allele-specific PCR markers.The frequency of the Glu-A3d allele(26.4%) was greater than those of the other alleles.The Glu-A3d allele was present in 26.7% of landraces and 26.0% of improved cultivars.The alleles,from most to least frequent,were as Glu-A3dGlu-A3cGlu-A3aGlu-A3bGlu-A3eGlu-A3gGlu-A3f.The Glu-A3e and Glu-A3f allele were only absent among improved cultivars from spring-winter wheat zone.Since the Glu-A3b allele has a more pronounced effect on gluten strength,varieties with the Glu-A3b allele can be considered suitable for the breeding of new,high-quality cultivars,and its frequency was 10.7% in landraces and 18.2% in improved cultivars,this showed that the total quality level of China wheat had significantly improved.

Abstract:Abstract In order to study the allelic variation of Glu-1 gene (High molecular weight glutenin), 100 Iranian wheat cultivars including imported and domestic genotypes were analyzed using ALP-PCR technique. Four specific primer pairs were used based on the genetic loci of Glu-A1, Glu-B1, and Glu-D1 to perform the chain polymerase reactions. PCR reaction products were resolved on 2% agarose gel. Since allele “a” had the largest relative frequency (0.707), two alleles (a 344bp; b 362bp) were identified by P1-P2 primer for Glu-A1 locus. Three alleles (a 800bp; b 500bp; c 300bp) were detected for Glu-B1 locus by P5-P6 primer, and allele “b” was assumed as the highest relative frequency (0.618). Two primer pairs were applied for Glu-D1 locus. Ultimately, four alleles were identified, where allele “c” had the highest relative frequency (0.525). The observed genetic variation value for Glu-D1 locus (H=0.648) exhibits the maximal polymorphism. Using cluster analysis, the relationship between the observed polymorphism and geographical variation was investigated. The results indicated that there exists a remarkable variation in Glu- A1 locus between the Iranian wheat cultivars.

Abstract:High-molecular-weight glutenin subunits (HMW-GS) are an important component of total cereal proteins in wheat. It is closely related to the processing quality of flour. Here, we analyzed allelic variations at the Glu-1 locus in 163 wheat accessions from Hubei Province, China with SDS-PAGE. Among the 15 alleles detected, alleles 1, 7+8, and 2+12 were the major alleles, and 7, 6+8, and 2+10 were rare alleles. The breeding lines had higher genetic diversity than the commercial varieties. Alleles 7 and 6+8 significantly reduced the grain protein content and wet gluten content of wheat. The “1, 7+9, 5+10” and “1, 14+15, and 2+12” allelic combinations significantly increased the grain protein content, hardness index, test weight, and wet gluten content of wheat. Alleles 7+9, 14+15, and 5+10 were identified as alleles related to high wheat quality. The “1, 7, 5+10”, “1, 6+8, 5+10”, “null, 7+9, 2+12”, “1, 14+15, 2+12”, and “1, 7+9, 5+10” allelic combinations had greater effects on wheat grain quality traits. These results demonstrate the effects of HMW-GS on wheat grain quality traits and provide a reference for the genetic improvement of wheat quality.

Novel insights into the composition, variation, organization, and expression of the low-molecular-weight glutenin subunit gene family in common wheat

Abstract:Low-molecular-weight glutenin subunits (LMW-GS), encoded by a complex multigene family, play an important role in the processing quality of wheat flour. Although members of this gene family have been identified in several wheat varieties, the allelic variation and composition of LMW-GS genes in common wheat are not well understood. In the present study, using the LMW-GS gene molecular marker system and the full-length gene cloning method, a comprehensive molecular analysis of LMW-GS genes was conducted in a representative population, the micro-core collections (MCC) of Chinese wheat germplasm. Generally, >15 LMW-GS genes were identified from individual MCC accessions, of which 4-6 were located at the Glu-A3 locus, 3-5 at the Glu-B3 locus, and eight at the Glu-D3 locus. LMW-GS genes at the Glu-A3 locus showed the highest allelic diversity, followed by the Glu-B3 genes, while the Glu-D3 genes were extremely conserved among MCC accessions. Expression and sequence analysis showed that 9-13 active LMW-GS genes were present in each accession. Sequence identity analysis showed that all i-type genes present at the Glu-A3 locus formed a single group, the s-type genes located at Glu-B3 and Glu-D3 loci comprised a unique group, while high-diversity m-type genes were classified into four groups and detected in all Glu-3 loci. These results contribute to the functional analysis of LMW-GS genes and facilitate improvement of bread-making quality by wheat molecular breeding programmes.

Abstract:Polyploid wheats include a group of tetraploids known as Timopheevii (AuAuGG), which are represented by two subspecies: Triticum timopheevii ssp. timopheevii (cultivated) and Triticum timopheevii ssp. araraticum (wild). The combined use of electrophoretic (SDS-PAGE) and chromatographic (RP-HPLC) techniques carried out on high-molecular-weight glutenin subunits (HMW-GSs) permitted the association of different x- and y-type subunits to the A and G genomes and the assessment of allelic variation present at corresponding loci. The results also revealed that in both subspecies, accessions are present that possess expressed y-type subunits at the Glu-A1 locus. Genes corresponding to these subunits were amplified and amplicons corresponding to x- and y-type genes associated with the A genome were detected in all accessions, including those without expressed x- and y-type subunits. The comparison with genes of polyploid wheats confirmed the structural characteristics of typical y-type genes, with the presence of seven cysteine residues and with hexapeptide and nonapeptide repeat motifs. The identification of wild and cultivated T. timopheevii with both x- and y-type glutenin subunits at the Glu-A1 and Glu-G1 loci represents a useful source for the modification of the allelic composition of HMW-GSs in cultivated wheats with the ultimate objective of improving technological properties.

Abstract:The high molecular weight glutenin subunits (HMW-GS) are important components of storage proteins in common wheat ( Triticum aestivum L.). Conventional techniques for identifying the HMW-GS apply SDS-PAGE. To study differentiation of the HMW glutenin genes, partial DNA sequences of the concerned alleles were characterized from eight accessions of hexaploid synthetic wheat and common wheat varieties, and compared with the available sequences. SSR markers based on, and generated from, DNA sequences of the D genome of common wheat and Aegilops tauschii were analyzed to estimate genetic relationships of the accessions. Results showed that the HMW-GS D t x2.1 determined by SDS-PAGE could be identified as two types at the DNA sequence level. One type, designated as D t x2.1 a , showed identical SNPs to the D t x 1.5 allele and another type, designated as D t x 2.1 b , had different characteristics. The SSR analysis revealed a relatively close genetic relationship of the two accessions containing the D t x2.1 a allele to the accessions containing the D t x1.5 allele. It was concluded that the conventional SDS-PAGE has limitations in determining some HMW-GS, because the minor changes on the DNA sequence of a particular glutenin subunit cannot be detected at protein level, and that the AS-PCR method based on variation of DNA sequences of particular genes will provide a more powerful tool for identifying the HMW-GS. The differentiation of the genes encoding the HMW-GS is probably more rapid than expected. The close relationship between evolutionary changes of the microsatellite DNA and the HMW-GS genes detected in this study needs to be further investigated.

Abstract:The high molecular weight (HMW) glutenin subunits Dtx1.5+Dty10 from Aegilops tauschii are a novel pair of subunits not detected previously in common wheat (Triticum aestivum). The partial DNA sequences of the x-type HMW glutenin alleles from A. tauschii and synthetic hexaploid wheat samples with different HMW glutenin subunits were charcterised. Five samples were found to contain the HMW glutenin subunit Dtx1.5 that may affect bread-making quality. Polymorphisms of the DNA sequences were detected among alleles encoding different HMW glutenin subunits and within an allele encoding the same HMW glutenin subunit, such as the Dtx1.5 subunit. Three single nucleotide polymorphisms (SNPs) that can distinguish the Dxt1.5 from Dtx2, Dtx5, Dx2 and Dx5 alleles were identified. Allelic specific (AS)-PCR primers were developed based the SNPs located at the non-repetitive N-terminal of the HMW glutenin subunits. The AS-PCR primers can accurately identify accessions containing the Dtx1.5 subunit from those containing other studied subunits by PCR analysis, suggesting the usefulness of AS-PCR for identifying different HMW glutenin subunits of A. tauschii and synthetic hexaploid wheat. The AS-PCR primers developed based on SNPs in this study are valuable in wheat breeding for effective selection of special HMW glutenin subunits.

Abstract:Improved processing quality is an important objective of wheat breeding which is largely associated with the amount and composition of endosperm seed proteins in the wheat kernel. Especially, HMW-GS determines dough strength and visco-elasticity contributing towards end-use product. Glu-B1 is the candidate structural gene, coding for Glu1Bx and associated with the quantity of high-molecular-weight (HMW) glutenin in bread wheat. This study provides detection of GluB-1 alleles on 19 different wheat cultivars by precise AS-PCR marker with the aim to develop marker for HMW-GS 1Bx20 which is assumed to be associated with chapatti making quality (CMQ) in bread wheat. The primer pair gave diagnostic banding pattern in different varieties with 1Bx7, 1Bx20 and 1Bx17+18 GluB-1 alleles. The result of this study will help in detection of varieties with different allelic combinations at their GluB-1 loci in early developmental stage using single marker which reduces the time for varietal selection. It will be also useful in screening of wheat cultivars with good chapatti making quality and introgression of subunit 1Bx20 in high yielding varieties using marker-assisted selection (MAS).
