Abstract:The central nervous system (CNS) of Chelicerata has remained conserved since the Cambrian, yet few studies have examined its variability within chelicerate orders including Scorpiones. The scorpion CNS comprises the prosomal ganglion and opisthosomal ventral nerve cord. We visualize the scorpion CNS with microCT, explore morphological variation across taxa, compare the scorpion CNS to other arachnids, and create a terminology glossary and literature review to assist future studies. Six scorpion species were microCT scanned. Scan quality varied and most structures in the prosomal ganglion could only be observed in Paruroctonus becki (Vaejovidae). Major nerves and the first opisthosomal ganglion were visible in nearly all taxa. We present the most detailed 3D-rendering of the scorpion prosomal ganglion to date. Our results corroborate existing research and find the scorpion CNS to be conserved. Nearly all structures reported previously in the prosomal ganglion were located in similar positions in P. becki , and nerve morphology was conserved across examined families. Despite similarities, we report differences from the literature, observe taxonomic variation in prosomal ganglion shape, and confirm positional variation for the first opisthosomal ganglion. This study serves as a starting point for microCT analysis of the scorpion CNS, and future work should include more distantly related, size variable taxa to better elucidate these findings.

Abstract:X-ray micro-computed tomography imaging (micro-CT) is valuable for systematic research since it permits the non-destructive scanning and imaging of internal structures of very rare species and/or type specimens. Additionally, micro-CT allows to view the morphology and the functional anatomy of structures in their natural anatomical position, without deformations that typically occur using classical dissection protocols. In this study we provide the description of two new species of lichen moths (Lepidoptera: Erebidae, Lithosiini) from the Atlantic Forest in eastern Brazil: Nodozana heliae Moraes sp. nov. from Rio de Janeiro state and Epeiromulona pataxo Moraes & Aguiar sp. nov. from Bahia state. The male and female genitalia as well as the wing morphology were examined by means of non-destructive micro-CT, subsequent 3D model reconstruction, 360 degree spinning animations, 2D images from different angles, and those were compared against classical genitalia dissections from the same specimens. We conclude that techniques complement each other, micro-CT being particularly useful to study wing venation, sclerotized internal structures and muscles, while classical dissection is useful to study membranous structures, particularly in the female genitalia, abdominal skin and specialised scales on the male 8th sternite.

Abstract:The pulmonary extracellular matrix (ECM) provides structural integrity and essential mechanical properties such as elastic recoil, but also serves as an information-rich signaling template that instructs cell identity and activity in health and disease. The spatial distribution and specificity of ECM proteins to the lungs distinct tissue niches is mostly unknown. In this study, we use spatially resolved mass spectrometry-based proteomics to define global differences in the proteomic composition of anatomically and histologically defined regions of the distal human lung. We characterized the distal airway tree and associated arteries and veins from 12 human donors*, using both 3D surgical microdissection (n=5) and 2D laser-capture microdissection techniques (n=7). Our analysis identified 7594 proteins (including 446 matrisome proteins) from 3D dissections and 4898 proteins (including 341 matrisome proteins) from 2D dissections. We identified gradients of ECM composition along the proximal-distal axis of the airways and vessels and discovered a specific ECM and immune niche in the respiratory bronchioles (RB). Integrative analysis of the spatial proteomes with donor-matched snRNAseq data and the Human Lung Cell Atlas (HLCA) further suggests the presence of a novel RB-associated fibroblast state that can be found across donors also in public datasets. As proof of concept, we identify and validate a subset of fibroblasts characterized by an ECM expression program enriched in RB via interactive imaging method. Given the potential origin of chronic lung diseases in distal airways, our discovery warrants further investigation of this novel RB specific ECM niche.

Abstract:The paper reports on the value of scanning electron microscopy (SEM) in the taxonomy and morphology of Chironomidae. This method has been relatively rarely used in Chironomidae studies. Our studies suggest that the SEM method provides a lot of new information. For example, the plastron plate of the thoracic horn of Macropelopia nebulosa (Meigen) under light microscopy is visible as points, while under SEM we have found that it consists of a reticular structure with holes. By using SEM a more precise picture of the body structure of Chironomidae can be revealed. It allows researchers to explain inconsistencies in the existing descriptions of species. Another advantage of the SEM method is obtaining spatial images of the body and organs of Chironomidae. However, the SEM method also has some limitations. The main problem is dirt or debris (e.g. algae, mud, secretions, mucus, bacteria, etc.), which often settles on the external surface of structures, especially those which are uneven or covered with hair. The dirt should be removed after collection of chironomid material because if left in place it can become chemically fixed to various surfaces. It unnecessarily remains at the surface and final microscopic images may contain artifacts that obscure chironomid structures being investigated. In this way many details of the surface are thus unreadable. The results reported here indicate that SEM examination helps us to identify new morphological features and details that will facilitate the identification of species of Chironomidae and may help to clarify the function of various parts of the body. Fast development of electron microscope technique allows us to learn more about structure of different organisms.

Abstract:Scanning electron microscopy is the method of choice to visualize the surface structures of animals, fungi, plants, or inorganic objects at the highest resolution and often with impressive appeal. Numerous scanning electron microscope (SEM) images exist of Drosophila melanogaster, one of the most important model organisms in genetics and developmental biology, which have been taken partly for esthetics and often to solve scientific questions. Our work presents a collection of images comprising many prominent anatomical details of D. melanogaster in excellent quality to create a research and teaching resource for all Drosophilists.

Abstract:The Mesobuthus martensii is widely distributed in China.It is recorded by traditional Chinese Pharmacopoeia as a famous medicine.According to the situation in which the systematic study on this scorpion was weak,the morphology and structure of the scorpion species were studied in-depth level by this study.In this paper,we used scanning electron microscope,stereo microscope and anatomical method to systematically describe the morphology and structure features of M..martensii,and improve the basic data of the scorpion biological characteristics,especially for the structure of the respiratory system,reproductive system and venom:1.the structure characteristics of median and lateral eyes,chelicerae,pedipalps and legs including auditory hair on legs (Fig.1-4);2.the morphological structure and function of stemite region including pectines with sensilla on it (Fig.5-6);3.distribution and morphology of book lung in situ and its anatomical structure of lung lamellae (Fig.7);4.the structure in details of venom vesicle and aculeus showing dissection of venom gland and the canals (Fig.8).We explored the relationship between the different expression of different visual proteins,the different function of photoreceptor in scorpion evolution And investigated the sensing function of the tail in mating and predation defense.

Abstract:Representing a basal branch of arachnids, scorpions are known as ‘living fossils’ that maintain an ancient anatomy and are adapted to have survived extreme climate changes. Here we report the genome sequence of Mesobuthus martensii , containing 32,016 protein-coding genes, the most among sequenced arthropods. Although M. martensii appears to evolve conservatively, it has a greater gene family turnover than the insects that have undergone diverse morphological and physiological changes, suggesting the decoupling of the molecular and morphological evolution in scorpions. Underlying the long-term adaptation of scorpions is the expansion of the gene families enriched in basic metabolic pathways, signalling pathways, neurotoxins and cytochrome P450, and the different dynamics of expansion between the shared and the scorpion lineage-specific gene families. Genomic and transcriptomic analyses further illustrate the important genetic features associated with prey, nocturnal behaviour, feeding and detoxification. The M. martensii genome reveals a unique adaptation model of arthropods, offering new insights into the genetic bases of the living fossils.

Abstract:Using serial block-face scanning electron microscopy, we report on the internal 3D structures of the brown planthopper, Nilaparvata lugens (Hemiptera: Delphacidae) at nanometer resolution for the first time. Within the reconstructed organs and tissues, we found many novel and fascinating internal structures in the planthopper such as naturally occurring three four-way rings connecting adjacent spiracles to facilitate efficient gas exchange, and fungal endosymbionts in a single huge insect cell occupying 22% of the abdomen volume to enable the insect to live on plant sap. To understand the muscle and stylet movement during phloem sap-sucking, the cephalic skeleton and muscles were reconstructed in feeding nymphs. The results revealed an unexpected contraction of the protractors of the stylets and suggested a novel feeding model for the phloem sap-sucking.

Abstract:Cavernicolous scorpions are difficult to collect and study due to their often inaccessible habitats. Some have evolved unique morphological adaptations, known as troglomorphies, including reduced sclerotization and pigmentation, reduction and/or loss of eyes, attenuation and elongation of the appendages, which assist them to thrive in dark, humid and low-energy input environments. Cavernicolous scorpions are classified into accidentals, trogloxenes, troglophiles, and troglobites. The Balkans, and particularly the Dinaric Karst region, host a diverse cave-adapted fauna, including scorpions. Despite an 1895 report of a blind scorpion from Bosnia and Herzegovina, the first truly troglobitic European scorpion, Euscorpius studentium Karaman, 2020, was only described a few years ago, based on two immature specimens. In the present contribution, this unique species is redescribed based on the first adult specimens; the ecological classification of all currently known cavernicolous Euscorpiidae Laurie, 1896, is updated; a key to the identification of cavernicolous scorpions occurring in the Dinaric Karst is provided; and the historical and geographical factors affecting the distribution and conservation of cavernicolous scorpions in the Balkans is reviewed.

Abstract:Soluble low-molecular-mass protein isoforms were purified from chemosensory organs (antennae, tarsi and labrum) of the desert locust Schistocerca gregaria. Five genes encoding proteins of this group were amplified by PCR from cDNAs of tarsi and sequenced. Their expression products are polypeptide chains of 109 amino acids showing 40-50% sequence identity with putative olfactory proteins from Drosophila melanogaster and Cactoblastis cactorum. Direct structural investigation on isoforms purified from chemosensory organs revealed the presence in the expression products of two of the genes cloned. Two additional protein isoforms were detected and their molecular structure exhaustively characterized. MS analysis of all isoforms demonstrated that the four cysteine residues conserved in the polypeptide chain were involved in disulfide bridges (Cys29-Cys38 and Cys57-Cys60) and indicated the absence of any additional post-translational modifications. Immunocytochemistry experiments, performed with rabbit antiserum raised against the protein isoform mixture, showed selective labelling of the outer lymph in contact sensilla of tarsi, maxillary palps and antennae. Other types of sensilla were not labelled, nor were the cuticle and dendrites of the sensory cells. No binding of radioactively labelled glucose or bicarbonate was detected, in disagreement with the hypothesis that this class of proteins is involved in the CO2-sensing cascade. Our experimental data suggest that the proteins described here could be involved in contact chemoreception in Orthoptera.

Abstract:Over 400 million years old, scorpions represent an ancient group of arachnids and one of the first animals to adapt to life on land. Presently, the lack of available genomes within scorpions hinders research on their evolution. This study leverages ultra-long nanopore sequencing and Pore-C to generate the first chromosome level assembly and annotation for the desert hairy scorpion, . The assembled genome is 2.23 Gb in size with an N50 of 280 Mb. Pore-C scaffolding re-oriented 99.6% of bases into nine chromosomes and BUSCO identified 998 (98.6%) complete arthropod single copy orthologs. Repetitive elements represent 54.69% of the assembled bases, including 872,874 (29.39%) LINE elements. A total of 18,996 protein-coding genes and 75,256 transcripts were predicted, and extracted protein sequences yielded a BUSCO score of 97.2%. This is the first genome assembled and annotated within the family Hadruridae, representing a crucial resource for closing gaps in genomic knowledge of scorpions, resolving arachnid phylogeny, and advancing studies in comparative and functional genomics.

Abstract:Though recent investigations have contributed substantially to our understanding of the Alpine-Dinaric radiation of the genus Zospeum Bourguignat, 1856, its southernmost member, Zospeum troglobalcanicum Absolon, 1916, has remained a taxonomic ghost. The assumed absence of type material, the insufficient original description, and the lack of new samples from its Western Balkan type locality have stymied further clarification. The recent discovery of a single syntype shell housed at the Natural History Museum Vienna now enables the first morphological assessment via 3D X-ray and SEM imaging. Based on this image data, different characters for assessing the southernmost members of the genus are determined and a lectotype is designated. Eleven allied species from 15 Western Balkan populations are described from museum material and recent sampling efforts: Z. amplioscutum Jochum & Ruthensteiner sp. nov., Z. biokovoense Jochum & Ruthensteiner sp. nov., Z. constrictum Jochum & Ruthensteiner sp. nov., Z. dubokidoense Jochum & Ruthensteiner sp. nov., Z. intermedium Jochum & Ruthensteiner sp. nov., Z. kolbae Jochum, Inäbnit, Kneubühler & Ruthensteiner sp. nov., Z. neuberti Jochum & Ruthensteiner sp. nov., Z. njegusiense Jochum & Ruthensteiner sp. nov., Z. njunjicae Jochum, Schilthuizen & Ruthensteiner sp. nov., Z. tortuosum Jochum & Ruthensteiner sp. nov. and Z. tumidum Jochum, Schilthuizen & Ruthensteiner sp. nov. One species, Z. kolbae, is described using DNA sequence data and one species, Z. simplex Inäbnit, Jochum & Neubert, 2021 for which DNA sequence data is already available, is supported by morphological data presented in this study. The DNA sequence dataset (COI, 16S and H3) is included here and implemented in the most recent phylogenetic reconstruction of the genus. A translation of Karel Absolon’s notes from the Balkan scientific expeditions is provided.
