Abstract:Modern lifestyle has profoundly modified human sleep habits. Sleep duration has shortened over recent decades from 8 to 6.5 hours resulting in chronic sleep deprivation. Additionally, irregular sleep, shift work and travelling across time zones lead to disruption of circadian rhythms and asynchrony between the master hypothalamic clock and pacemakers in peripheral tissues. Furthermore, obstructive sleep apnea syndrome (OSA), which affects 4 - 15% of the population, is not only characterized by impaired sleep architecture but also by repetitive hemoglobin desaturations during sleep. Epidemiological studies have identified impaired sleep as an independent risk factor for all cause of-, as well as for cardiovascular, mortality/morbidity. More recently, sleep abnormalities were causally linked to impairments in glucose homeostasis, metabolic syndrome and Type 2 Diabetes Mellitus (T2DM). This review summarized current knowledge on the metabolic alterations associated with the most prevalent sleep disturbances, i.e. short sleep duration, shift work and OSA. We have focused on various endocrine and molecular mechanisms underlying the associations between inadequate sleep quality, quantity and timing with impaired glucose tolerance, insulin resistance and pancreatic β-cell dysfunction. Of these mechanisms, the role of the hypothalamic-pituitary-adrenal axis, circadian pacemakers in peripheral tissues, adipose tissue metabolism, sympathetic nervous system activation, oxidative stress and whole-body inflammation are discussed. Additionally, the impact of intermittent hypoxia and sleep fragmentation (key components of OSA) on intracellular signaling and metabolism in muscle, liver, fat and pancreas are also examined. In summary, this review provides endocrine and molecular explanations for the associations between common sleep disturbances and the pathogenesis of T2DM.

Connecting insufficient sleep and insomnia with metabolic dysfunction

Abstract:The global epidemic of obesity and type 2 diabetes parallels the rampant state of sleep deprivation in our society. Epidemiological studies consistently show an association between insufficient sleep and metabolic dysfunction. The goal of this review is to establish an updated framework for the causal links between insufficient sleep and insomnia and risks for type 2 diabetes and obesity. The global epidemic of obesity and type 2 diabetes parallels the rampant state of sleep deprivation in our society. Epidemiological studies consistently show an association between insufficient sleep and metabolic dysfunction. Mechanistically, sleep and circadian rhythm exert considerable influences on hormones involved in appetite regulation and energy metabolism. As such, data from experimental sleep deprivation in humans demonstrate that insufficient sleep induces a positive energy balance with resultant weight gain, due to increased energy intake that far exceeds the additional energy expenditure of nocturnal wakefulness, and adversely impacts glucose metabolism. Conversely, animal models have found that sleep loss–induced energy expenditure exceeds caloric intake resulting in net weight loss. However, animal models have significant limitations, which may diminish the clinical relevance of their metabolic findings. Clinically, insomnia disorder and insomnia symptoms are associated with adverse glucose outcomes, though it remains challenging to isolate the effects of insomnia on metabolic outcomes independent of comorbidities and insufficient sleep durations. Furthermore, both pharmacological and behavioral interventions for insomnia may have direct metabolic effects. The goal of this review is to establish an updated framework for the causal links between insufficient sleep and insomnia and risks for type 2 diabetes and obesity.

Abstract:Animals must continually respond dynamically as they move through complex environments, and slopes are a common terrain on which legged animals must move. Despite this, non-level locomotion remains poorly understood. In this study, we first review the literature on locomotor mechanics, metabolic cost, and kinematic strategies on slopes. Using existing literature we then performed scaling analyses of kinematic variables, including speed, duty factor, and stride-length across a range of body sizes from ants to horses. The studies that examined locomotion on inclines vastly outnumbered those focusing on declines. On inclines, animals tend to reduce speed and increase duty factor, but a similar consensus could not be reached for declines. Remarkably, stride-length did not differ between locomotion on inclines and on level terrain, but this may have resulted from data only being available for low slopes (<30°). On declines there appears to be a shift in locomotor strategy that is size-dependent. At masses <1-10 kg, animals tended to use shorter strides than on level terrain, and the opposite occurred at larger body masses. Therefore, possibly due to stability issues, body mass plays a significant role in the locomotor strategy used when traveling downhill. Although we currently lack sufficient data, differential leg function is likely to be critical for locomotion on slopes, with mechanical demands differing on limbs during movement on level, inclined, and declined surfaces. Our scaling analysis not only highlights areas that require future work, but also suggests that body size is important for determining the mechanics and strategies animals use to negotiate non-level terrain. It is clear that selection has resulted in an incredible range of body size among animals, both extant and extinct, and it is likely that the ability to move up and down slopes has constrained or relaxed these mechanical pressures. Given the lack of integration of ecological data with laboratory experiments, future work should first determine which inclines animals actually use in nature, as this likely plays a key role in behaviors such as predator-prey interactions.

Abstract:Traditional risk factors for obesity and the metabolic syndrome, such as excess energy intake and lack of physical activity, cannot fully explain the high prevalence of these conditions. Insufficient sleep and circadian misalignment predispose individuals to poor metabolic health and promote weight gain and have received increased research attention in the past 10 years. Insufficient sleep is defined as sleeping less than recommended for health benefits, whereas circadian misalignment is defined as wakefulness and food intake occurring when the internal circadian system is promoting sleep. This Review discusses the impact of insufficient sleep and circadian misalignment in humans on appetite hormones (focusing on ghrelin, leptin and peptide-YY), energy expenditure, food intake and choice, and risk of obesity. Some potential strategies to reduce the adverse effects of sleep disruption on metabolic health are provided and future research priorities are highlighted. Millions of individuals worldwide do not obtain sufficient sleep for healthy metabolic functions. Furthermore, modern working patterns, lifestyles and technologies are often not conducive to adequate sleep at times when the internal physiological clock is promoting it (for example, late-night screen time, shift work and nocturnal social activities). Efforts are needed to highlight the importance of optimal sleep and circadian health in the maintenance of metabolic health and body weight regulation.

Abstract:Sleep deficiency is a ubiquitous phenomenon among Americans. In fact, in the United States, ∼78% of teens and 35% of adults currently get less sleep than recommended for their age‐group, and the quality of sleep appears to be getting worse for many. The consequences of sleep disruption manifest in a myriad of ways, including insulin resistance and disrupted nutrient metabolism, dysregulation of hunger and satiety, and potentially increased body weight and adiposity. Consequently, inadequate sleep is related to an increased risk of various cardiometabolic diseases, including obesity, diabetes, and heart disease. Exercise has the potential to be an effective therapeutic to counteract the deleterious effects of sleep disruption listed above, whereas chronic psychosocial stress may causally promote sleep disruption and cardiometabolic risk. Here, we provide a narrative review of the current evidence on the consequences of short sleep duration and poor sleep quality on substrate metabolism, circulating appetite hormones, hunger and satiety, and weight gain. Secondly, we provide a brief overview of chronic psychosocial stress and its impact on sleep and metabolic health. Finally, we summarise the current evidence regarding the ability of exercise to counteract the adverse metabolic health effects of sleep disruption. Throughout the review, we highlight areas where additional interrogation and future exploration are necessary.

Abstract:Introduction and purpose of the work: Sleep is essential for the proper functioning of the body. Adequate quality and quantity of sleep affects metabolic processes, including glucose metabolism, as well as the sensitivity of tissues to insulin, the reduction of which is called insulin resistance. It leads to an increased production of glucose in order to maintain normal blood glucose levels. Insulin resistance is a modifiable risk factor of, inter alia, type 2 diabetes. The aim of this study is to summarize the current knowledge on the relationship between sleep deprivation and an increased risk of developing insulin resistance. State of knowledge: Sleep undoubtedly has a strong influence on glucose metabolism and insulin sensitivity. Numerous studies show that even partial sleep deprivation lowers insulin sensitivity. The available literature shows that shift work and a shifted bedtime, combined with an insufficient amount of it, lead to a significant reduction in insulin sensitivity. The positive effect of increasing the time of night rest on the sensitivity of tissues to insulin may be an element of the prevention of diseases such as obesity or type 2 diabetes. Moreover, disturbances in sleep continuity lead to formation of insulin resistance. An example would be obstructive sleep apnea, the treatment of which affects metabolic processes. Summary: Adequate sleep hygiene is an extremely important part of a healthy lifestyle. More research is needed on the relationship between sleep and the regulation of carbohydrate metabolism, due to the fact that the appropriate sleep-wake rhythm may give hope for improvement in health in people with metabolic disorders and may be an important protective factor for many diseases.

Abstract:This mini-review examines the complex relationship between diet and sleep and explores the clinical and public health implications of the current evidence. Dietary quality and intake of specific nutrients can impact regulatory hormonal pathways to alter sleep quantity and quality. Sleep, in turn, affects the intake of total energy, as well as of specific foods and nutrients, through biological and behavioral mechanisms. Initial research in this field focused primarily on the effects of short sleep duration on nutritional quality. However, more recent studies have explored the dynamic relationship between long sleep duration and diet. Current evidence suggests that extremes of sleep duration alter sleep patterns, hormonal levels, and circadian rhythms, which contribute to weight-related outcomes and obesity, and other risk factors for the development of chronic disease such as type 2 diabetes and cardiovascular disease. These patterns may begin as early as childhood and have impacts throughout the life course. Given that non-communicable diseases are among the leading causes of death globally, deeper understanding of the interactions between sleep and nutrition has implications for both public health and clinical practice.

Abstract:PurposeObesity is characterized by increased infiltration of macrophages. Cytokines derived from these macrophages significantly induce inflammatory response and desensitize insulin action. Disturbed sleep pattern is also associated with release of the same kind of cytokines that could play a central role in the development of insulin resistance.Material and MethodsA computer-based search of the literature indexed in PubMed was made using the keywords: glucose metabolism, insulin resistance, adipokines, adiponectin, interleukin-6 (IL-6), leptin, tumor necrosis factor alpha (TNF-α), monocyte chemo-attractant protein-1 (MCP-1), sleep deprivation, and sleep loss. In this review firstly we include the studies that have measured the relationship of cytokines with sleep duration and insulin resistance, and finally, we analyze the relationship of sleep duration with the development of insulin resistance and type 2 diabetes mellitus.DiscussionIL-6, IL-1β, TNF-α, and MCP-1 are secreted by many cell types including macrophages, adipocytes, etc. They are released in response to many stimuli including sleep loss, tissue injury, and infection. Some evidence has suggested that these cytokines are molecular link between obesity, sleep disturbances, and insulin resistance, because similar types of alterations in plasma levels of these cytokines are found in patients with obesity and disturbed sleep having type 2 diabetes mellitus (DM-2) or insulin resistance.ConclusionWe conclude that both sleep loss and obesity contribute to the onset of insulin resistance and DM-2 with the help of the same kind of cytokines. The mechanisms involved in the impaired glucose metabolism seem to act via a decreased efficacy of the hypothalamic–pituitary–adrenal (HPA) axis and due to modified release of cytokines—especially IL-1β, IL-6, TNF-α, and MCP-1 from adipocytes following obesity and poor quality of sleep. The aforementioned biomarkers could provide a minimally invasive means for early detection and specific treatment of these disorders.
Impact of insufficient sleep on dysregulated blood glucose control under standardised meal conditions

Abstract:Aims/hypothesis: Sleep, diet and exercise are fundamental to metabolic homeostasis. In this secondary analysis of a repeated measures, nutritional intervention study, we tested whether an individual's sleep quality, duration and timing impact glycaemic response to a breakfast meal the following morning. Methods: Healthy adults' data (N = 953 [41% twins]) were analysed from the PREDICT dietary intervention trial. Participants consumed isoenergetic standardised meals over 2 weeks in the clinic and at home. Actigraphy was used to assess sleep variables (duration, efficiency, timing) and continuous glucose monitors were used to measure glycaemic variation (>8000 meals). Results: Sleep variables were significantly associated with postprandial glycaemic control (2 h incremental AUC), at both between- and within-person levels. Sleep period time interacted with meal type, with a smaller effect of poor sleep on postprandial blood glucose levels when high-carbohydrate (low fat/protein) (pinteraction = 0.02) and high-fat (pinteraction = 0.03) breakfasts were consumed compared with a reference 75 g OGTT. Within-person sleep period time had a similar interaction (high carbohydrate: pinteraction = 0.001, high fat: pinteraction = 0.02). Within- and between-person sleep efficiency were significantly associated with lower postprandial blood glucose levels irrespective of meal type (both p < 0.03). Later sleep midpoint (time deviation from midnight) was found to be significantly associated with higher postprandial glucose, in both between-person and within-person comparisons (p = 0.035 and p = 0.051, respectively). Conclusions/interpretation: Poor sleep efficiency and later bedtime routines are associated with more pronounced postprandial glycaemic responses to breakfast the following morning. A person's deviation from their usual sleep pattern was also associated with poorer postprandial glycaemic control. These findings underscore sleep as a modifiable, non-pharmacological therapeutic target for the optimal regulation of human metabolic health. Trial registration ClinicalTrials.gov NCT03479866.

Abstract:Research shows that reduced sleep duration is related to an increased risk of obesity. The relationship between sleep deprivation and obesity, type 2 diabetes, and other chronic diseases may be related to the imbalance of appetite regulation. To comprehensively illustrate the specific relationship between sleep deprivation and appetite regulation, this review introduces the pathophysiology of sleep deprivation, the research cutting edge of animal models, and the central regulatory mechanism of appetite under sleep deprivation. This paper summarizes the changes in appetite-related hormones orexin, ghrelin, leptin, and insulin secretion caused by long-term sleep deprivation based on the epidemiology data and animal studies that have established sleep deprivation models. Moreover, this review analyzes the potential mechanism of associations between appetite regulation and sleep deprivation, providing more clues on further studies and new strategies to access obesity and metabolic disease.

Abstract:Abstract Objective To evaluate which condition of sleep debt has a greater negative impact on insulin resistance: sleep deprivation for 24 hours or 4 hours of sleep restriction for 4 nights. Materials and Methods In total, 28 healthy male subjects aged 18 to 40 years were recruited and randomly allocated to two groups: sleep deprivation (SD) and sleep restriction (SR). Each group underwent two conditions: regular sleep (11 pm to 7 am) and total sleep deprivation for 24 hours (SD); regular sleep (11 pm to 7 am) and 4 nights of sleep restriction (SR) (1 am to 5 am). The oral glucose tolerance test (OGTT) was performed, and baseline glucose, insulin, free fatty acids (FFAs), and cortisol were measured. In addition, the area under the curve (AUC) for glucose and insulin, the homeostasis model assessment of insulin resistance (HOMA-IR), and the Matsuda Index (Insulin Sensitivity Index, ISI) were calculated. Results Glucose and insulin had a similar pattern between groups, except at the baseline, when insulin was higher in the sleep debt condition of the SR when compared with the SD (p < 0.01). In the comparison between regular sleep and sleep debt, the SD had a higher insulin AUC (p < 0.01) and FFAs (p = 0.03) after sleep deprivation, and insulin and the insulin AUC increased (p < 0.01 for both), while the ISI decreased (p = 0.02) after sleep restriction in the SR. In baseline parameters covariate by the condition of regular sleep, insulin (p = 0.02) and the HOMA-IR (p < 0.01) were higher, and cortisol (p = 0.04) was lower after sleep restriction when compared with sleep deprivation. Conclusion Sleep restriction for 4 consecutive nights is more detrimental to energy metabolism because of the higher insulin values and insulin resistance compared with an acute period of sleep deprivation of 24 hours.

Abstract:IntroductionAdipokines are a growing group of secreted proteins that play important roles in metabolism. Accumulating evidence suggests that adipokines may mediate the close association between sleep disturbance and metabolic derangements. Due to the extensive crosstalk between adipokines and metabolic pathways, an integrated approach is required to better understand the significance of adipokines in sleep disorders and associated metabolic dysregulation. In the current study, we employed network analysis, a set of concepts and methods derived from graph theory, to obtain novel insights into the roles of adipokines in sleep disorders and associated metabolic dysregulation.MethodsA network of six adipokines and their molecular targets is constructed based on current understanding of their roles in sleep and metabolic disorders using an adjacency matrix. The network is then visualized and analyzed using an open source platform Gephi to derive network-level metrics, including degree and centrality measures. These metrics are used to explore the relationship between sleep disturbance and associated metabolic dysregulation in several disease processes.Results94 nodes and 264 edges were identified in the network, representing regulatory factors, adipokines, molecular pathways, and disease processes. Two adipokines, leptin and adiponectin, were found to have higher degrees than other adipokines, indicating their central roles in connecting sleep disturbance to metabolic dysregulations. Among the regulatory factors, obesity and obstructive sleep apnea were found to be major drivers for the sleep associated metabolic dysregulation based on their higher degrees. The important pathways adipokines act on in the network included insulin signaling, inflammation, food intake, and energy expenditure. The main disease processes adipokines act on included cardiovascular, reproductive, and autoimmune diseases. Leptin, AMPK, and fatty acid oxidation were found to have global influence in the network based on their high betweenness.ConclusionAdipokines are important players in the metabolic dysregulations associated with sleep disturbance. Adipokines such as leptin and adiponectin act on diverse metabolic pathways in response to sleep disturbance, obesity, insulin resistance, and inflammation.They play important roles in cardiovascular, reproductive, and autoimmune diseases. Abstract:Portable, low cost and real-time gas sensors have a considerable potential in various biomedical and industrial applications. For such applications, nano-photonic gas sensors based on standard silicon fabrication technology offer attractive opportunities. Deposition of high surface area nano-porous coatings on silicon photonic sensors is a means to achieve selective, highly sensitive and multiplexed gas detection on an optical chip. Here we demonstrate selective and reversible ammonia gas detection with functionalized silicon-on-insulator optical micro-ring resonators. The micro-ring resonators are coated with acidic nano-porous aluminosilicate films for specific ammonia sensing, which results in a reversible response to NH(3)with selectivity relative to CO(2). The ammonia detection limit is estimated at about 5 ppm. The detectors reach a steady response to NH(3) within 30 and return to their base level within 60 to 90 seconds. The work opens perspectives on development of nano-photonic sensors for real-time, non-invasive, low cost and light weight biomedical and industrial sensing applications.

Abstract:Sleep and circadian rhythms modulate or control daily physiological patterns with importance for normal metabolic health. Sleep deficiencies associated with insufficient sleep schedules, insomnia with short-sleep duration, sleep apnea, narcolepsy, circadian misalignment, shift work, night eating syndrome, and sleep-related eating disorder may all contribute to metabolic dysregulation. Sleep deficiencies and circadian disruption associated with metabolic dysregulation may contribute to weight gain, obesity, and type 2 diabetes potentially by altering timing and amount of food intake, disrupting energy balance, inflammation, impairing glucose tolerance, and insulin sensitivity. Given the rapidly increasing prevalence of metabolic diseases, it is important to recognize the role of sleep and circadian disruption in the development, progression, and morbidity of metabolic disease. Some findings indicate sleep treatments and countermeasures improve metabolic health, but future clinical research investigating prevention and treatment of chronic metabolic disorders through treatment of sleep and circadian disruption is needed.

Abstract:Sleep deprivation has become increasingly prevalent in contemporary society, and the consequences of this reality such as cognitive impairment and metabolic disorders, are widely investigated in the scientific scenario. However, the impact of sleep deprivation on the health of future generations is a challenge, and researchers are focusing their attention on this issue. Thus, this review aims to describe the impact of sleep deprivation in early life in animal models, particularly rodents, discussing the molecular physiology impacted by prolonged wakefulness in early life, as well as the changes that interfere with neurodevelopmental processes. Additionally, it explores the changes impacting metabolic mechanisms and discusses both the short- and long-term consequences of these processes on endocrine, behavioral, and cognitive functions. Finally, we briefly address some strategies to mitigate the adverse effects of sleep deprivation.

Abstract:BACKGROUND: Sleep deprivation in individuals with diabetes mellitus type 2 is more prevalent than in ordinary people. At present, the adverse effects of diabetes type 2 people with sleep disorders and sleep deprivation on blood sugar control are irrefutable. Thorough assessments covering the whole structure should be of concern in health-care treatment. It is precipitated and delivered to the physical, mental health, and social environment, but no systematic review or minimal data were published. Furthermore, it may significantly affect the system more than existing research. AIM: An integrative review aims to clarify the results or consequences of sleep disturbance/deprivation or insomnia symptoms among diabetes mellitus type 2 patients. METHODS: The writers implemented a literature search in PubMed, CINAHL, and Medline using the terms DM type 2, insomnia, adult, effect, DM, sleep disturbance, sleep disorder, and a consequence between 2012 and 2022. Inclusion criteria selected through considering the entire article, and providing an abstract, were 20 articles. RESULTS: Integrative data extraction and information were analyzed thematically. Identified were nine ideas: Association with blood sugar control, blood pressure control, risk of CVD, diabetes self-care behavior, weight gained and Obstructive Sleep Apnea (OSA), lifestyle and physical activity, mood/depression and anxiety symptoms, daily calories distribution, cholesterol/triglyceride, and liver enzyme levels. CONCLUSION: The adverse effects of sleep deprivation in type 2 diabetes significantly affect all pathophysiologically, mentally, and lifestyle modifications, including diabetes self-care. Therefore, to highlight the importance of promoting optimum sleep in diabetes type2 patients, a health-care system is inevitably as necessary as diet and exercise management.

Abstract:Three new butenolides (1-3), a new cyclopentenone derivative (4), and a known alcohol (5) were isolated from a marine sponge Homaxinella sp. by bioactivity-guided fractionation. The planar structures were established on the basis of NMR and MS analyses. The stereochemistry of the butenolides and cyclopentenone derivative was defined on the basis of optical rotation and CD spectroscopy. The compounds were tested for cytotoxicity against a panel of five human solid tumor cell lines and displayed marginal to significant activity.

Abstract:This narrative review presents the findings from intervention studies on the effects of sleep deprivation on eating habits, metabolic rate, and the hormones regulating metabolism, and discusses their relevance to weight loss efforts. Disturbed sleeping patterns lead to increased energy intake, partly from excessive snacking, mainly on foods high in fat and carbohydrates. The studies focused mainly on the effects of sleep duration, but also of sleep quality, on dietary intake during weight loss trials, and on weight loss maintenance. It is important to explore sleep routines that could enhance the efforts of obese and overweight people to lose weight, maintain their weight loss, and improve their overall health.
