Clarifying the interplay between OT/ VP and sex hormones in the regulation of social cognition is necessary for further applications.

Abstract:The neuropeptides oxytocin (OT) and vasopressin (VP) are known to mediate social cognition and behaviors in a sex-dependent manner. This chapter reviews the sex-dependent influence of OT and VP on social behaviors, focusing on (1) partner preference and sexual orientation, (2) memory modulation, (3) emotion regulation, and (4) trust-related behaviors. Most studies suggest that OT promotes familiar (opposite-sex) partner preference, strengthens memory, relieves anxiety, and increases trust. However, VP-regulated social cognition has been studied less than OT. VP facilitates familiar (opposite-sex) partner preference, enhances memory, induces anxiety, and influences happiness/anger perception. Detailed sex differences of these effects are reviewed. There is a male preponderance in the use of animal models and many study results are too complex to draw firm conclusions. Clarifying the complex interplay between the OT/VP system and sex hormones in the regulation of social behaviors is needed.

Abstract:Conducting randomized controlled trials (RCTs) of potentially effective treatments is time consuming and expensive, and for industry sponsored studies, the information is commercially sensitive. The notion that information from RCTs can be shared to ensure openness and transparency has been proposed by a number of groups, but at the present time there is no consensus on how data sharing should be carried out or the balance of benefit and risk inherent in this process.1–3 Naci et al .4 argue that providing original datasets from clinical trials in an open access anonymized format will avoid publication bias where ‘negative’ trials (which do not show a difference between treatment and control) have less chance of being published, resulting in important information not being available. Oxytocin enhances inter-brain synchrony during social coordination in male adults

Abstract:Recent brain imaging research has revealed oxytocin (OT) effects on an individual's brain activity during social interaction but tells little about whether and how OT modulates the coherence of inter-brain activity related to two individuals' coordination behavior. We developed a new real-time coordination game that required two individuals of a dyad to synchronize with a partner (coordination task) or with a computer (control task) by counting in mind rhythmically. Electroencephalography (EEG) was recorded simultaneously from a dyad to examine OT effects on inter-brain synchrony of neural activity during interpersonal coordination. Experiment 1 found that dyads showed smaller interpersonal time lags of counting and greater inter-brain synchrony of alpha-band neural oscillations during the coordination (vs control) task and these effects were reliably observed in female but not male dyads. Moreover, the increased alpha-band inter-brain synchrony predicted better interpersonal behavioral synchrony across all participants. Experiment 2, using a double blind, placebo-controlled between-subjects design, revealed that intranasal OT vs placebo administration in male dyads improved interpersonal behavioral synchrony in both the coordination and control tasks but specifically enhanced alpha-band interbrain neural oscillations during the coordination task. Our findings provide first evidence that OT enhances inter-brain synchrony in male adults to facilitate social coordination.
Intranasal Oxytocin Selectively Modulates Large-Scale Brain Networks in Humans

Abstract:A growing body of evidence indicates that the neuropeptide oxytocin (OT) alters the neural correlates of socioemotional and salience processing. Yet the effects of OT over important large-scale networks involved in these processes, such as the default mode (DM), ventral attention (VA), and cingulo-opercular (CO) networks, remain unknown. Therefore, we conducted a placebo-controlled crossover study with intranasal 24 IU OT in 38 healthy male subjects using a resting-state functional magnetic resonance imaging paradigm to investigate its impact over these three networks candidates. To understand the underlying mechanisms of the neuropeptide, we compared the intranetwork connectivity for each network candidate and also the internetwork connectivity across all networks between both treatment conditions. Based on the relevance of interindividual factors for OT effects, we correlated individual network changes with behavioral performance in a decision-making task and with impulsivity scores. Our results show that OT mainly alters connectivity in the VA, on one side reducing the coupling to regions that typically form the nodes of DM, an introspective and self-referential network, and on the other side increasing the coupling to the edges of the CO, which is involved in salience processing. The results of the internetwork analyses confirmed the specificity of the OT effects. Indeed, we observed significant correlations with the erroneous performance during decision-making but not with the obtained impulsivity scores. Overall, our data support that the modulation of functional connectivity within the VA is a basic mechanism by which OT directs attentional resources from internal to external cues, preparing the brain for context-dependent salience processing.
Atmospheric degradation of 2-butanol, 2-methyl-2-butanol, and 2,3-dimethyl-2-butanol: OH kinetics and UV absorption cross sections.

Abstract:The absolute rate coefficients for the reactions of hydroxyl radical (OH) with 2-butanol (k(1)), 2-methyl-2-butanol (k(2)), and 2,3-dimethyl-2-butanol (k(3)) were measured as a function of temperature (263-354 K) and pressure (41-193 Torr of He, Ar, and N(2)) by the pulsed laser photolysis/laser-induced fluorescence technique. This work represents the first absolute determination of k(1)(-)k(3) and their temperature dependence. No pressure dependence of the rate coefficients was observed in the range studied. Thus, k(i)(298 K) values (x10(-12) cm(3) molecule(-1) s(-1) with an uncertainty of +/-2sigma) were averaged over the pressure range studied yielding 8.77 +/- 1.46, 3.64 +/- 0.60, and 9.01 +/- 1.00 for 2-butanol (k(1)), 2-methyl-2-butanol (k(2)), and 2,3-dimethyl-2-butanol (k(3)), respectively. k(1) and k(3) exhibit a slightly negative temperature dependence over the temperature range studied. In contrast, the rate coefficient for the reaction of OH with 2-methyl-2-butanol (k(2)) did not show any temperature dependence. Some deviation of the conventional Arrhenius behavior was clearly observed for k(3). In this case, the best fit to our data was found to be described by the three-parameter expression k(T) = A + B exp(-C/T). The UV absorption cross sections of 2-butanol, 2-methyl-2-butanol, and 2,3-dimethyl-2-butanol have also been measured at room temperature between 208 and 230 nm. The values reported constitute the first determination of the UV cross sections of those alcohols. Our results are compared with previous studies, when possible, and are discussed in terms of the H-abstraction by OH radicals. The atmospheric implications of these reactions and the photochemistry of these alcohols are also discussed.

Abstract:Background: The neuropeptides oxytocin and vasopressin have been repeatedly implicated in social decision making by enhancing social salience and, generally, cooperation. The iterated and sequential version of the prisoner’s dilemma (PD) game is a social dilemma paradigm eliciting strategies of cooperation versus competition. Aims: We aimed to characterise the role of PD players’ sex, game partner type (computer vs. human) and oxytocin or vasopressin inhalation on the player’s strategy preference. Methods: Participants (153 men; 151 women) were randomised to intranasal 24 IU oxytocin, 20 IU vasopressin or placebo, double-blind, and played the PD. We examined main and interactive effects of sex, drug and partner type on strategy preference. Results: We found a pervasive preference for a tit-for-tat strategy (i.e. general sensitivity to the partner’s choices) over unconditional cooperation, particularly when against a human rather than a computer partner. Oxytocin doubled this sensitivity in women (i.e. the preference for tit-for-tat over unconditional cooperation strategies) when playing against computers, which suggests a tendency to anthropomorphise them, and doubled women’s unconditional cooperation preference when playing against humans. Vasopressin doubled sensitivity to the partner’s previous choices (i.e. for tit-for-tat over unconditional cooperation) across sexes and partner types. Conclusions: These findings suggest that women may be more sensitive to oxytocin’s social effects of anthropomorphism of non-humans and of unconditional cooperation with humans, which may be consistent with evolutionary pressures for maternal care, and that vasopressin, irrespective of sex and partner type, may be generally sensitising humans to others’ behaviour.

Abstract:The respective roles of the neuropeptides arginine vasopressin (AVP) and oxytocin (OXT) in modulating social cognition and for therapeutic intervention in autism spectrum disorder have not been fully established. In particular, while numerous studies have demonstrated effects of oxytocin in promoting social attention the role of AVP has not been examined. The present study employed a randomized, double-blind, placebo (PLC)-controlled between-subject design to explore the social- and emotion-specific effects of AVP on both bottom-up and top-down attention processing with a validated emotional anti-saccade eye-tracking paradigm in 80 healthy male subjects (PLC = 40, AVP = 40). Our findings showed that AVP increased the error rate for social (angry, fearful, happy, neutral and sad faces) but not non-social (oval shapes) stimuli during the anti-saccade condition and reduced error rates in the pro-saccade condition. Comparison of these findings with a previous study (sample size: PLC = 33, OXT = 33) using intranasal oxytocin revealed similar effects of the two peptides on anti-saccade errors, although with some difference in effects of specific face emotions, but a significantly greater effect of AVP on pro-saccades. Both peptides also produced a post-task anxiolytic effect by reducing state anxiety. Together these findings suggested that both AVP and OXT decrease goal-directed top-down attention control to social salient stimuli but that AVP more potently increased bottom-up social attentional processing.

Abstract:Summary The effect of immunosuppression with the glucocorticosteroid, dexamethasone, on the susceptibility to Haemonchus contortus infection of a highly resistant Merino genotype was investigated. Higher faecal egg counts, larger worm burdens and heavier worms were recorded in immunosuppressed wethers. The characteristic globule leucocyte infiltrate in the abomasum of resistant wethers (12‐month‐old castrated males) was absent in immunosuppressed animals. Treatment with dexamethasone abolished differences between resislanl and susceptible genotypes in faecal egg counts, worm weights, thymus weights and globule leucocyte responses to infection with H. contortus. These results suggest that an immunological basis underlies the high level of resistance to infection in the resistant genotype.

Abstract:Considering its role in prosocial behaviors, oxytocin (OT) has been suggested to diminish levels of aggression. Nevertheless, recent findings indicate that oxytocin may have a broader influence on increasing the salience of social stimuli and may therefore, under certain circumstances, increase antisocial behaviors such as aggression. This controversy led to the following speculations: If indeed oxytocin promotes primarily prosocial behavior, administration of OT is expected to diminish levels of aggression. However, if oxytocin mainly acts to increase the salience of social stimuli, it is expected to elevate levels of aggression following provocation. In order to test this assumption we used the Social Orientation Paradigm (SOP), a monetary game played against a fictitious partner that allows measuring three types of responses in the context of provocation: an aggressive response - reducing a point from the fictitious partner, an individualistic response - adding a point to oneself, and a collaborative response - adding half a point to the partner and half a point to oneself. In the current double-blind, placebo-controlled, within-subject study design, 45 participants completed the SOP task following the administration of oxytocin or placebo. The results indicated that among subjects naïve to the procedure oxytocin increased aggressive responses in comparison with placebo. These results support the saliency hypothesis of oxytocin and suggest that oxytocin plays a complex role in the modulation of human behavior.

Abstract:Oxytocin (OT) is an endogenous neuropeptide that, while originally thought to promote trust, has more recently been found to be context-dependent. Here we extend experimental paradigms previously restricted to de novo decision-to-trust, to a more realistic environment in which social relationships evolve in response to iterative feedback over twenty interactions. In a randomized, double blind, placebo-controlled within-subject/crossover experiment of human adult males, we investigated the effects of a single dose of intranasal OT (40 IU) on Bayesian expectation updating and reinforcement learning within a social context, with associated brain circuit dynamics. Subjects participated in a neuroeconomic task (Iterative Trust Game) designed to probe iterative social learning while their brains were scanned using ultra-high field (7T) fMRI. We modeled each subject's behavior using Bayesian updating of belief-states ("willingness to trust") as well as canonical measures of reinforcement learning (learning rate, inverse temperature). Behavioral trajectories were then used as regressors within fMRI activation and connectivity analyses to identify corresponding brain network functionality affected by OT. Behaviorally, OT reduced feedback learning, without bias with respect to positive versus negative reward. Neurobiologically, reduced learning under OT was associated with muted communication between three key nodes within the reward circuit: the orbitofrontal cortex, amygdala, and lateral (limbic) habenula. Our data suggest that OT, rather than inspiring feelings of generosity, instead attenuates the brain's encoding of prediction error and therefore its ability to modulate pre-existing beliefs. This effect may underlie OT's putative role in promoting what has typically been reported as 'unjustified trust' in the face of information that suggests likely betrayal, while also resolving apparent contradictions with regard to OT's context-dependent behavioral effects.
Opposing Oxytocin Effects on Intergroup Cooperative Behavior in Intuitive and Reflective Minds

Abstract:People often favor ingroup over outgroup members when choosing to cooperate. Such ingroup-favored cooperation is promoted by oxytocin—a neuropeptide shown to facilitate social cognition and that has emerged as a pharmacological target for treatments of social functioning deficits. The current study applied a dual-process model to investigate whether and how intuitive and reflective cognitive styles affect the oxytocin-motivated ingroup favoritism in cooperation. We examined oxytocin effects on ingroup favoritism in a double-blind, placebo-controlled between-subjects design where cognitive processing (intuition vs reflection) was experimentally manipulated in healthy Chinese males ( n =150). We also supplemented this experimental manipulation with an individual difference analysis by assessing participants’ inclination toward intuition or reflection in daily life. Intranasal administration of oxytocin ( vs placebo) increased ingroup favoritism among participants primed to be intuitive or those who preferred intuition in daily life. In contrast, oxytocin decreased ingroup favoritism in participants primed to rely on reflective thinking or those who preferred reflective decision-making in daily life. Our results demonstrate that oxytocin has distinct functional roles when different cognitive styles (ie, intuition vs reflection) are promoted during social cooperation in a group situation. Our findings have implications for oxytocin pharmacotherapy of social dysfunction in that whether the effects of oxytocin on social functioning are facilitative, debilitative, or null, depends on an individual’s cognitive style.

Abstract:The neuropeptide oxytocin (OT) regulates mammalian social approach behavior across sexes. Yet most OT studies in humans exclusively investigated men. Here, we studied sex differences in OT's effects on human trust behavior in 144 heterosexual participants (73 women, 71 men). Participants received 24 international units of intranasal OT or placebo treatment and played a trust game in the role of the investor while undergoing electroencephalography. Trustees were represented by photos of the other sex gradually varying in their pre-rated intensities of facial features signaling attractiveness and threat. On a behavioral level, we observed that OT increased trust in men and reduced it in women when trustees showed weak signals of attractiveness and threat. Correspondingly, on the neurophysiological level, we noted that OT intensified the P100 in male participants, but dampened it in female ones. Our findings demonstrate OT's sex- and context-specific effects on social approach behavior and an underlying early visual attention-related brain process. This evidence demonstrates the need to consider psychobiological mechanisms of sexual dimorphism in human OT research.

Abstract:Objective This study extended a classic self-referential learning paradigm by investigating the effects of intranasally-administered oxytocin in high and low socially anxious participants during social learning, as a function of social anxiety levels and sex. Methods In a randomized double-blinded design, 160 participants were either given intranasal oxytocin (24 I·U.) or placebo. Subsequently, while lying in an MR scanner, participants were shown neutral faces that were paired with positively, neutrally, or negatively valenced self-referential sentences, during which we measured self-reported arousal and sympathy of the facial stimuli, pupil dilation, and changes in the brain-oxygen-level dependent signal. Four-factor mixed analyses of variance with the between-subjects factors group (high socially anxious vs. low socially anxious), substance (oxytocin vs. placebo), and sex (male vs. female) and the within-subjects factor sentence valence (positive vs. neutral vs. negative) were conducted for each measure, respectively. Results Administration of intranasal oxytocin yielded (1) an increase in sympathy ratings in high socially anxious compared to low socially anxious individuals and (2) decreased arousal ratings for positively-conditioned faces in low socially anxious participants. As an objective physiological measure of arousal, pupil dilation mirrored the behavioral results. Oxytocin effects on neural activation in the insula interacted with anxiety levels and sex: low socially anxious individuals yielded lower activation under oxytocin than placebo; the converse was observed in high socially anxious individuals. This interaction also differed between sexes, as men yielded higher activation levels than women. These findings were more prominent for positively- and negatively-conditioned faces. Within the amygdala, high socially anxious men yielded higher activation than high socially anxious women in the left hemisphere, and low socially anxious men yielded higher activation than low socially anxious women from positively- and negatively-conditioned faces, though no influence of oxytocin was detected. Conclusion These results suggest oxytocin-induced behavioral, physiological, and neural changes as a function of social learning in socially low and high anxious individuals. These findings challenge the amygdalocentric view of the role of emotions in social learning, instead contributing to the growing body of findings implicating the insula therein, revealing an interaction between oxytocin, sex, and emotional valence. Such discoveries raise an interesting set of questions regarding the computational goals of regions such as the insula in emotional learning and how neural activity can play a diagnostic or prognostic role in social anxiety, potentially leading to new treatment opportunities that may combine oxytocin and neurofeedback differentially for men and women.

Abstract:Oxytocin (OXT) is a neuropeptide which has a critical role in human social behaviour and cognition. Research investigating the role of OXT on functional brain changes in humans has often used task paradigms that probe socioemotional processes. Preliminary evidence suggests a central role of the amygdala in the social cognitive effects of intranasal OXT (IN-OXT), however, inconsistencies in task-design and analysis methods have led to inconclusive findings regarding a cohesive model of the neural mechanisms underlying OXT's actions. The aim of this meta-analysis was to systematically investigate these findings. A systematic search of PubMed, PsycINFO, and Scopus databases was conducted for fMRI studies which compared IN-OXT to placebo in humans. First, we systematically reviewed functional magnetic resonance imaging (fMRI) studies of IN-OXT, including studies of healthy humans, those with clinical disorders, and studies examining resting-state fMRI (rsfMRI). Second, we employed a coordinate-based meta-analysis for task-based neuroimaging literature using activation likelihood estimation (ALE), whereby, coordinates were extracted from clusters with significant differences in IN-OXT versus placebo in healthy adults. Data were included for 39 fMRI studies that reported a total of 374 distinct foci. The meta-analysis identified task-related IN-OXT increases in activity within a cluster of the left superior temporal gyrus during tasks of emotion processing. These findings are important as they implicate regions beyond the amygdala in the neural effects of IN-OXT. The outcomes from this meta-analysis can guide a priori predictions for future OXT research, and provide an avenue for targeted treatment interventions.
A meta-analysis of hormone administration effects on cooperative behaviours: Oxytocin, vasopressin, and testosterone

Abstract:<p>The hormones oxytocin, vasopressin, and testosterone have been implicated in cooperative behaviours and have attracted increasing research interest for their potential to regulate human cooperation in both healthy and clinical populations. However, the behavioural effects of the administration of these hormones remain to be verified. The current analysis included 41 studies involving 3,269 participants with a narrow age range. We examined the administration effects of these hormones on cooperative behaviour and the regulatory effects of individual characteristics, hormone interventions, and task structure and context. Results revealed a moderate positive effect size of oxytocin intranasal administration, a large negative effect size of vasopressin intranasal administration, and nonsignificant effects of testosterone administration on cooperative behaviours. Participants with mental dysfunctions were less sensitive to oxytocin and vasopressin administration. Oxytocin administration was effective in an in-group situation and for initial choices, corroborating a Tit-for-Tat strategy.</p>

The effect of oxytocin on group formation and strategic thinking in men

Abstract:Decision-making in groups is a remarkable and decisive element of human societies. Humans are able to organize themselves in groups, engage in collaborative decision-making processes and arrive at a binding agreement, even in the absence of unanimous consent. However, the transfer of decision-making autonomy requires a willingness to deliberately expose oneself to the decisions of others. A lack of trust in the abilities of others or of the underlying decision-making process, i.e. public trust, can lead to a breakdown of organizations in political or economic domains. Recent studies indicate that the biological basis of trust on an individual level is related to Oxytocin, an endogenous neuropeptide and hormone, which is also associated with pro-social behavior and positive conflict resolution. However, little is known about the effects of Oxytocin on the inclination of individuals to form or join groups and to deliberately engage in collaborative decision-making processes. Here, we show that intranasal administration of Oxytocin (n = 60) compared to placebo (n = 60) in males causes an adverse effect on the choice for forming groups in the presence of a competitive environment. In particular, Oxytocin negatively affects the willingness to work collaboratively in a p-Beauty contest game, whereas the effect is most pronounced for participants with relatively high strategic sophistication. Since our data provide initial evidence that Oxytocin has a positive effect on strategic thinking and performance in the p-Beauty contest game, we argue that the adverse effect on group formation might be rooted in an enhanced strategic sophistication of participants treated with Oxytocin.
L-DOPA and oxytocin influence the neural correlates of performance monitoring for self and others

Abstract:Abstract Rationale The ability to monitor the consequences of our actions for others is imperative for flexible and adaptive behavior, and allows us to act in a (pro)social manner. Yet, little is known about the neurochemical mechanisms underlying alterations in (pro)social performance monitoring. Objective The aim of this functional magnetic resonance imaging (fMRI) study was to improve our understanding of the role of dopamine and oxytocin and their potential overlap in the neural mechanisms underlying performance monitoring for own versus others’ outcomes. Method Using a double-blind placebo-controlled cross-over design, 30 healthy male volunteers were administered oxytocin (24 international units), the dopamine precursor L-DOPA (100 mg + 25 mg carbidopa), or placebo in three sessions. Participants performed a computerized cannon shooting game in two recipient conditions where mistakes resulted in negative monetary consequences for (1) oneself or (2) an anonymous other participant. Results Results indicated reduced error-correct differentiation in the ventral striatum after L-DOPA compared to placebo, independent of recipient. Hence, pharmacological manipulation of dopamine via L-DOPA modulated performance-monitoring activity in a brain region associated with reward prediction and processing in a domain-general manner. In contrast, oxytocin modulated the BOLD response in a recipient-specific manner, such that it specifically enhanced activity for errors that affected the other in the pregenual anterior cingulate cortex (pgACC), a region previously implicated in the processing of social rewards and prediction errors. Behaviorally, we also found reduced target sizes—indicative of better performance—after oxytocin, regardless of recipient. Moreover, after oxytocin lower target sizes specifically predicted higher pgACC activity when performing for others. Conclusions These different behavioral and neural patterns after oxytocin compared to L-DOPA administration highlight a divergent role of each neurochemical in modulating the neural mechanisms underlying social performance monitoring.
