Abstract:To develop a method for deleting genes without leaving a selection marker in Escherichia coli, the str~S-kan~R cassette with promoter lac or arabinose were constructed. And then the counter-selection cassette was electroporated into TOP10 strain after flanked by homology arms of flgK gene to produce recombinant strains of TOP10_PlacSK_ΔflgK and TOP10_ParaSK_ΔflgK.Moreover, the minimal inhibitory concentration(MIC) assay was performed to evaluate the sensitivity of streptomycin for recombinant strains. The results showed that the MIC of wild type TOP10 strain, TOP10_PlacSK_ΔflgK strain and TOP10_ParaSK_ΔflgK strain were 8,000μg/mL, 2,000μg/mL and 500μg/mL respectively, which indicated that both recombinant strains were sensitive to streptomycin, and the counter-selection efficiency of the str~S-kan~R cassette with promoter arabinose was much better. This study established a counter-selection system for deleting genes in E.coli and provided foundations for optimization of Red recombination.
Response Distortion on the NEO PI-R among Candidates Taking the Entrance Examination to the National School of Civil Aviation (ENAC-France)

Abstract:This study examined the effect of a selection context on the responses to a five factor personality questionnaire (Revised NEO Personality Inventory) of 974 candidates taking the entrance examination to the ENAC (National School of Civil Aviation, France). A response distortion index was calculated using Schinka, Kinder and Kremer (1997)’s method. The results indicate: 1) lower neuroticism scores but higher conscientiousness, agreeableness and extraversion scores compared to standard conditions; 2) a high estimated effect of the response distortion index on all dimensions of the NEO PI-R except for Openness to experience; 3) substantial stability of scores at a one-year interval (N = 117). The paper discusses the conceptual and practical value of this response distortion index for measuring the dimensions of the NEO PI-R in candidates taking the entrance examination to the ENAC.

Abstract:ABSTRACT Pasteurella multocida is a zoonotic conditional pathogen that infects multiple livestock species, causing substantial economic losses in the animal husbandry industry. An efficient markerless method for gene manipulation may facilitate the investigations of P. multocida gene function and pathogenesis of P. multocida . Herein, a temperature-sensitive shuttle vector was constructed using lacZ as a selection marker, and markerless glgB , opa , and hyaE mutants of P. multocida were subsequently constructed through blue–white colony screening. The screening efficiency of markerless deletion strains was improved by the lacZ system, and the method could be used for multiple gene deletions. However, the fur mutant was unavailable via this method. Therefore, we constructed a pheS m screening system based on mutated phenylalanine tRNA synthetase as a counterselection marker to achieve fur deletion mutant. The transformed strain was sensitive to 20 mM p -chloro-phenylalanine, demonstrating the feasibility of pheS m as a counter-selective marker. The pheS m system was used for markerless deletions of glgB , opa , and hyaE as well as fur that could not be screened by the lacZ system. A comparison of screening efficiencies of the system showed that the pheS m counterselection system was more efficient than the lacZ system and broadly applicable for mutant screening. The methods developed herein may provide valuable tools for genetic manipulation of P. multocida . IMPORTANCE Pasteurella multocida is a highly contagious zoonotic pathogen. An understanding of its underlying pathogenic mechanisms is of considerable importance and requires efficient species-specific genetic tools. Herein, we propose a screening system for P. multocida mutants using lacZ or pheS m screening markers. We evaluated the efficiencies of both systems, which were used to achieve markerless deletion of multiple genes. The results of this study support the use of lacZ or pheS m as counterselection markers to improve counterselection efficiency in P. multocida . This study provides an effective genetic tool for investigations of the virulence gene functions and pathogenic mechanisms of P. multocida .

A rapid seamless method for gene knockout in Pseudomonas aeruginosa

Abstract:Background: Pseudomonas aeruginosa is a model organism for the study of quorum sensing, biofilm formation, and also leading cause of nosocomial infections in immune compromised patients. As such P. aeruginosa is one of the most well studied organisms in terms of its genetics. However, the construction of gene deletions and replacements in Pseudomonas aeruginosa is relatively time-consuming, requiring multiple steps including suicide vector construction, conjugation, inactivation with insertion of antibiotic resistance cassettes and allelic exchange. Even employing Gateway recombineering techniques with direct transformation requires a minimum two weeks. METHODS: We have developed a rapid streamlined method to create clean deletion mutants in P. aeruginosa through direct transformation, eliminating the need for the creation of Gateway-compatible suicide vectors. In this method, upstream and downstream sequences of the gene/locus to be deleted are amplified by polymerase chain reaction (PCR) and seamlessly fused with the linearized pEX18Tc sacB suicide plasmid by Gibson assembly. The resulting deletion plasmid is transformed into P. aeruginosa by an electroporation method optimized in this study. The plasmid is then integrated into the chromosome by homologous recombination, and deletion mutants are identified via sacB mediated sucrose counter-selection. RESULTS: The current method was employed to generate clean gene deletions of the heme assimilation system anti-σ factor, hasS and the virulence regulator involving ECF system anti-σ and σ factors vreA and vreI, respectively. The process from plasmid construction to confirmation by DNA sequencing of the gene deletion was completed in one week. Furthermore, the utility of the method is highlighted in the construction of the vreA and vreI deletions, where the start codon of vreA and the stop codon of vreI overlap. Utilizing Gibson assembly deletion mutants were constructed with single base pair precision to generate the respective vreA and vreI deletions, while maintaining the start and stop codon of the respective genes. Overall, this method allows for rapid construction of gene deletions in P. aeruginosa with base pair precision. Conclusion: This method from the construction of the suicide vector to sequence confirmation of the unmarked gene deletion can be performed in one week, without the requirement for expensive proprietary reagents or instruments. The precision of Gibson assembly and the fact the accuracy in generating the desirable construct is 95%, makes this a viable and attractive alternative to previous methods.

Abstract:Staphylococcus aureus (S. aureus) is a major human pathogen that may produce a variety of toxins and cause staphylococcal food poisoning. In the present study, a direct loop-mediated isothermal amplification (LAMP) assay was developed and validated to detect S. aureus in food samples. Without prior cultural enrichment and DNA extraction steps, bacterial DNA was released by heating at 100°C for 5 min and directly subjected to LAMP assay. Using a set of LAMP primers recognizing six distinct regions of nuc gene, the developed direct LAMP assay was highly specific, and the analytical sensitivity was determined to be 7.6 × 102 CFU/mL. Moreover, with the pre-mixed LAMP reagents stored at -20°C, the entire assay should be finished within 40 min. These features greatly simplified the operating procedure and made the direct LAMP a powerful tool for rapid and on-site detection of S. aureus in food samples.

Abstract:We present a protocol for the generation of a gene-deletion allelic-exchange plasmid and its recovery in Escherichia coli for the purpose of constructing an in-frame gene deletion in Staphylococcus aureus Here, we present detailed methodologies for (i) the primer design (using the S. aureus tagO gene as our specific example); (ii) PCR amplification of the required gene fragments; (iii) preparation of the cloning vector (using the S. aureus allelic-exchange vector pIMAY* as an example); (iv) the Gibson assembly cloning method; (v) introduction of the plasmid into E. coli; (vi) confirmation of the plasmid insert in E. coli by colony PCR; and, finally, (vii) confirmation of the insert by sequencing. We also consider the long-term storage of the E. coli strains containing the desired plasmid.

Abstract:Streptomyces bacteria are known for producing important natural compounds by secondary metabolism, especially antibiotics with novel biological activities. Functional studies of antibiotic-biosynthesizing gene clusters are generally through homologous genomic recombination by gene-targeting vectors. Here, we present a rapid and efficient method for construction of gene-targeting vectors. This approach is based on Streptomyces phage phi BT1 integrase-mediated multisite in vitro site-specific recombination. Four 'entry clones' were assembled into a circular plasmid to generate the destination gene-targeting vector by a one-step reaction. The four 'entry clones' contained two clones of the upstream and downstream flanks of the target gene, a selectable marker and an E. coli-Streptomyces shuttle vector. After targeted modification of the genome, the selectable markers were removed by phi C31 integrase-mediated in vivo site-specific recombination between pre-placed attB and attP sites. Using this method, part of the calcium-dependent antibiotic (CDA) and actinorhodin (Act) biosynthetic gene clusters were deleted, and the rrdA encoding RrdA, a negative regulator of Red production, was also deleted. The final prodiginine production of the engineered strain was over five times that of the wildtype strain. This straightforward phi BT1 and phi C31 integrase-based strategy provides an alternative approach for rapid genetargeting vector construction and marker removal in streptomycetes.
Sandwich Fluorimetric Method for Specific Detection of Staphylococcus Aureus Based on Antibiotic-Affinity Strategy

Abstract:A novel antibiotic-affinity strategy was designed for fluorimetric detection of pathogenic bacteria based on the strong affinity of antibiotic agent to the cell wall of bacteria. In this proof-of-concept work, vancocin, a glycopeptide antibiotic for Gram-positive bacteria, was used as a molecular recognition agent to anchor Staphylococcus aureus (S. aureus) cell. To improve the specificity of this method for S. aureus detection, IgG was adopted as the second recognition agent utilizing the binding between Fc region of IgG and S. aureus protein A in the cell wall, to form a sandwich complex. By using fluorescein isothiocyanate as the signal probe, S. aureus whole cells could be directly assayed within a linear range of 1.0 × 10(3)-1.0 × 10(9) CFU mL(-1) with a detection limit of 2.9 × 10(2) CFU mL(-1). The whole assay process could be completed within 130 min when a ready-for-use microplate was adopted. This proposed strategy for pathogenic bacteria detection possessed some attractive characteristics such as high sensitivity, wide linear range, simple manipulation, short assay time, and low cost. Furthermore, this sandwich mode also showed ideal specificity because vancocin and IgG bound with S. aureus at two distinct sites. It opened up a new pathway for high-throughput screening of pathogenic bacteria in medical diagnosis, food safety, bioterrorism defense, and drug discovery.
Use of a combined synthetic fibre bag method to estimate the true total tract digestion of organic matter and nitrogen of hay and grass silage in cows.

Abstract:A 4 x 4 Latin square experiment was designed to determine and compare the total tract digestion of organic matter (OM) and nitrogen of hay and grass silage by combining the ruminal and intestinal digestion estimates obtained by the rumen bag and mobile bag methods. The diets were composed of forage alone or 30% forage and 70% barley. The rumen degradation (RD) was calculated using the degradation constants a, b, and c obtained to the forages, according to the equation RD = a+bc/(c+k). Intestinal digestion of escape feeds was estimated using the feed residues that resisted rumen degradation during 16 hours. For comparative purposes, the total tract digestion was also calculated based on the actual rumen degradation of forages during 16 hours rumen incubation. The effect of dietary interaction on the digestion of forages was assessed. Rumen degradation values obtained by 16 hours incubation did not provide sufficient basis to estimate reliably the total tract digestion by the combined bag method. Using calculated RD total tract digestion of OM of hay and grass silage were rather close to conventional in vivo digestion coefficient of respective feeds similar in chemical composition, when barley was not included in the diet. With hay, but not with grass silage, barley always markedly reduced the rumen degradation and total digestion of OM. Barley supplementation always increased the proportion of feed digested in the intestine. Nitrogen digestion was always higher with grass silage than with hay, and the combined bag method produced N digestion values 17.6-22.0%-units higher than those in the feed table. Abstract:Glaesserella parasuis is an important bacterial pathogen that affects the swine industry worldwide. Research on the pathogenic mechanism and genetically engineered vaccine remains undeveloped because an effective markerless and multiple-gene knockout system is unavailable for G. parasuis yet. To establish a markerless knockout, deleted allelic genes with kanamycin resistance (Kan R ) cassettes were introduced into the genome of G. parasuis by using natural transformation with suicide plasmids. Then, the Kan R cassette was excised with a thermosensitive plasmid pGF conferring a constitutive Flp expression. To realize the markerless and multiple-gene knockout, plasmid pGAF was constructed by placing the Flp gene under the control of an arabinose-inducible promoter. Firstly, pGAF was introduced into G. parasuis by electroporation, and the marked mutants were produced following natural transformation. Finally, the Kan R cassette was excised from the genome by the inducible expression of Flp upon arabinose action. Based on the natural transformation and the inducible expression of Flp, the markerless single-gene knockout mutants of Δ hsdR , Δ neuA2 , Δ espP2 , Δ apd , and Δ nanH were constructed. In addition, a five-gene knockout mutant of Δ hsdR Δ neuA2 Δ espP2 Δ apd Δ nanH was generated by successive natural transformation with five suicide plasmids. Taken together, a markerless and multiple-gene deletion system was established for G. parasuis in the present study for the first time. This system is simple, efficient, and easy to manipulate for G. parasuis ; thus, our technique will substantially aid the understanding of the etiology, pathogenesis, and genetic engineering of G. parasuis and other bacteria that can be naturally transformed in laboratory conditions.

Abstract:ABSTRACT There has been an absence of an efficient method of gene knockdown in the important human pathogen Staphylococcus aureus like RNA interference in eukaryotes. The previously developed antisense RNA technology is mainly applied for forward genetic screening but is rather limited in specific gene knockdown because of the lack of rational antisense RNA design strategies. Here we report an efficient and specific system for gene knockdown in S. aureus based on the type II clustered regularly interspaced short palindromic repeat (CRISPR) system from Streptococcus pyogenes . We can achieve gene silencing with the coexpression of dCas9, an RNA-guided DNA binding protein, and a small guide RNA complementary to the target gene. With this system, we have successfully silenced diverse sets of genes varying in size and expression level in different S. aureus strains. This system exhibited high-efficiency knockdown of both essential and nonessential genes, and its effect is inducible and reversible. In addition, the system can repress the expression of multiple genes simultaneously and silence an entire operon or part of it. This RNA-guided DNA targeting system thus provides a simple, rapid, and affordable method for selective gene knockdown in S. aureus . IMPORTANCE Staphylococcus aureus is an important human and animal pathogen that can cause a diversity of infectious diseases. Molecular genetic study of S. aureus has provided an avenue for the understanding of its virulence, pathogenesis, and drug resistance, leading to the discovery of new therapies for the treatment of staphylococcal infections. However, methodologies developed for genetic manipulation of S. aureus usually involve either low efficiency or laborious procedures. Here we report an RNA-guided system for gene knockdown in S. aureus and show its high efficiency and simplicity for selective gene silencing in different strains of S. aureus . This simple, rapid, and affordable system may serve as a promising tool for functional gene study in S. aureus , especially for the study of essential genes, thus facilitating the understanding of this pathogen and its interaction with its hosts.

Streamlining Marker-less Allelic Replacement in Streptococcus pneumoniae Through a Single Transformation Step Strategy: easyJanus

Abstract:The ability to genetically manipulate bacteria is a staple of modern molecular microbiology. Since the 2000's, marker-less mutants of Streptococcus pneumoniae (Spn) have been made by allelic-exchange predominantly using the kan -rpsLcassette known as "Janus". The conventional Janus protocol involves two transformation steps using multiple PCR-assembled products containing the Janus cassette and the target gene's flanking DNA. We present an innovative strategy to achieve marker-less allelic replacement through a single transformation step. Our approach involves the integration of an additional gene downstream region upstream of the Janus cassette, resulting in a modified genetic arrangement. This single modification reduced the number of required PCR fragments from five to four, lowered the number of assembly reactions from two to one, and simplified the transformation process to a single step. To validate the efficacy of our approach, we implemented this strategy to delete in Spn serotype 4 strain TIGR4 the virulence gene pspA, the entire capsular polysaccharide synthesis locus cps4, and to introduce a single nucleotide replacement into the chromosome. Notably, beyond streamlining the procedure, our method markedly reduced false positives typically encountered during negative selection with streptomycin when employing the traditional Janus protocol. Furthermore, and as consequence of reducing the amount of exogenous DNA required for construct synthesis, we show that our new method is amendable to the use of commercially available synthetic DNA for construct creation, further reducing the work needed to obtain a mutant. Our streamlined strategy, termed easyJanus, substantially expedites the genetic manipulation of Spn facilitating future research endeavors.

Abstract:Targeted gene deletion in mycobacteria remain complicated, requiring expertise and multiple steps. Here we present a single-step, easy to understand and perform method for targeted gene deletion. Using this method, we successfully deleted several genes in both M. smegmatis and M. abscessus . We believe this method will facilitate molecular research of mycobacteria and make it accessible to a greater number of researchers throughout the world.

Xer-cise in Helicobacter pylori: one-step transformation for the construction of markerless gene deletions

Abstract:Background: Xer-cise is an efficient selectable marker removal technique that was first applied in Bacillus subtilis and Escherichia coli for the construction of markerless gene deletions. Xer-cise marker excision takes advantage of the presence of site-specific Xer recombination in most bacterial species for the resolution of chromosome dimers at the dif site during replication. The identification and functional characterization of the difH/XerH recombination system enabled the development of Xer-cise in Helicobacter pylori. Methods: Markerless deletions were obtained by a single natural transformation step of the Xer-cise cassette containing rpsL and cat genes, for streptomycin susceptibility and chloramphenicol resistance respectively, flanked by difH sites and neighboring homologous sequences of the target gene. Insertion/deletion recombinant H. pylori were first selected on chloramphenicol-containing medium followed by selection on streptomycin-containing medium for clones that underwent XerH mediated excision of the rpsL-cat cassette, resulting in a markerless deletion. Results: XerH-mediated removal of the antibiotic marker was successfully applied in three different H. pylori strains to obtain markerless gene deletions at very high efficiencies. An unmarked triple deletion mutant was also constructed by sequential deletion of ureA, vacA and HP0366 and removal of the selectable marker at each step. The triple mutant had no growth defect suggesting that multiple difH sites per chromosome can be tolerated without affecting bacterial fitness. Conclusion: Xer-cise eliminates the need for multiple passages on non selective plates and subsequent screening of clones for loss of the antibiotic cassette by replica plating.

Abstract:Staphylococcus aureus (S. aureus) , particularly Methicillin-resistant S. aureus (MRSA), poses a significant global public health threat, necessitating advanced methodologies to enhance our understanding of this organism at the omics levels. This study introduces a refined protocol for constructing and curing high-density transposon mutant (tn-mutant) libraries in S. aureus , addressing the challenges associated with low transductant yields, and the complex genetic manipulation mechanism in Gram-positive bacteria. Our methodology employs a Himar1 transposon based on a two-plasmid system, leveraging Himar1's high insertional efficiency in AT-rich organisms. Enhanced transduction efficiency was achieved through chloramphenicol pre-treatment and the use of modified enriched media. Complementing this, an optimized plasmid curing procedure ensured a representative and stable tn-mutant library. The protocol was successfully applied to multiple S. aureus strains, demonstrating an increase in mutant recovery and reduced post-curing impact. The method offers a robust approach for Transposon Insertion Sequencing (TIS) applications in S. aureus , enabling deeper insights into survival, resistance, and pathogenicity mechanisms. This protocol holds a significant potential for accelerating the construction of tn-mutant libraries in various S. aureus strains.

Abstract:OBJECTIVE:We developed a homologous recombination system to make marker-free mutants in Mtb in an easy screening way.METHODS:The plasmid pSL002 harboring recE and recT-like protein gp60 and gp61 was transformed into wt Mtb in order to increase the recombing efficiency. The two homologous arms of target gene were amplified and cloned into pSL001. The purified homologous arms-loxP-gfp-hyg-loxP fragments were further transformed into Mtb (with pSL002 inside) competent cells to obtain the double cross over (DCO) mutants. The plasmid pSL003 was transformed into DCO competent cells to excise the two loxP sites flanked region. The plasmid pSL002 and pSL003 were removed via the counter selection marker sacB.RESULTS:An efficient homologous recombination system was developed. Three different marker free mutants: Rv1364c phosphatase domain, PstP extracellular domain and PstP transmembrane domain were created by the system developed in this study. The efficiency to obtain DCO was varying from 25% to 62.5%, while the efficiency from DCO to KO was close to 100%. The gfp reporter was used to screen SCO, DCO and KO.CONCLUSION:The homologous system shortens the complicated screening process to 3 month in Mtb. This is the fastest allelic exchange system reported so far, providing novel deletion strategy to the repertoire of mycobacterial genetic tools for constructing unmarked mutations.
The IGF‐II receptor system: A G protein‐linked mechanism

Abstract:Based on the finding that stimulation of the IGF‐II, receptor (IGF‐IIR) is capable of activating Gi2 and calcium channels in BALB/c 3T3 fibroblasts, it was found that purified IGF‐IIR can couple directly to purified Gi2 in phospholipid vesicles. IGF‐IIR–Gi2 coupling can be characterized as follows. IGF‐IIR directly couples to Gi2 in response to IGF‐II in a stoichiometrical manner, suggesting that IGF‐IIR works as a transmembrane signaling molecule and that the seven‐transmembrane structure is not essential for receptor‐G protein coupling. The mode of IGF‐IIR–Gi2 interaction is similar to that of conventional receptor–G protein coupling, suggesting that a common G protein recognition mechanism is shared by IGF‐IIR and conventional G‐coupled receptors. The action of IGF‐IIR is specific on Gi2 among various G proteins. Finally, the activity of IGF‐IIR on Gi2 is similarly potent across the species of the proteins. These characteristics led to the discovery of a 14‐amino‐acid region in IGF‐IIR that can directly interact with and activate Gi2, and is located at residues 2410–2423 of the human receptor. Subsequent work has indicated that this region is responsible for Gi‐coupling function of intact IGF‐IIR. The most important extensions of this discovery are the following: (1) The structure–function relationship for the Gi‐activating function of this 14‐amino‐acid sequence, (2) the prediction of G protein‐coupled functions of receptors based on the results obtained from 1), and (3) clarification of the detailed mechanism whereby ligand–receptor complex recognizes G proteins. This paper reviews what we have learned from IGF‐IIR in terms of receptor–G protein interfaces and discusses future prospects. © 1993 Wiley‐Liss, Inc.
An efficient blue-white screening system for markerless deletions and stable integrations in Streptomyces chromosomes based on the blue pigment indigoidine biosynthetic gene bpsA

Abstract:We previously developed an efficient deletion system for streptomycetes based on the positive selection of double-crossover events using bpsA, a gene for producing the blue pigment indigoidine. Using this system, we removed interfering secondary metabolite clusters from Streptomyces lividans TK24, resulting in RedStrep strains with dramatically increased heterologous production of mithramycin A (up to 3-g/l culture). This system, however, required a time-consuming step to remove the resistance marker genes. In order to simplify markerless deletions, we prepared a new system based on the plasmid pAMR18A. This plasmid contains a large polylinker with many unique restriction sites flanked by apramycin and kanamycin resistance genes and the bpsA gene for selecting a double-crossover event. The utility of this new markerless deletion system was demonstrated by its deletion of a 21-kb actinorhodin gene cluster from Streptomyces lividans TK24 with 30% efficiency. We used this system to efficiently remove the matA and matB genes in selected RedStrep strains, resulting in biotechnologically improved strains with a highly dispersed growth phenotype involving non-pelleting small and open mycelia. No further increase in mithramycin A production was observed in these new RedStrep strains, however. We also used this system for the markerless insertion of a heterologous mCherry gene, an improved variant of the monomeric red fluorescent protein, under the control of the strong secretory signal sequence of the subtilisin inhibitor protein, into the chromosome of S. lividans TK24. The resulting recombinant strains efficiently secreted mCherry into the growth medium in a yield of 30 mg/l.
Development and Verification of an FLP/FRT System for Gene Deletion in Candida Amazonensis

Abstract:[Objective] To develop an FLP/FRT system for gene disruption in Candida amazonensis that can repeatedly use a single selectable marker,and to verify the effectiveness of this system by deleting the PDC gene encoding pyruvate decarboxylase.[Methods] Four promoters (SpXYLp,SpMAL6p,SpMAL1p and SpGAL1p) from Spathaspora passalidarum and ScGALlp promoter from Saccharomyces cerevisiae were amplified and fused to the reporter gene of green fluorescent protein (gfpm) to study the regulation under corresponding inducible conditions.A strictly inducible promoter was selected to control the expression of the C.amazonensis-adapted FLP gene (caFLP),encoding the site-specific recombinase FLP.The promoter-caFLP fusion fragment was used to ligated with the hphm marker gene that conferred resistance to Hygromycin B,and the ligation product was flanked by direct repeats of the FLP recognition target (FRT).Then with the addition of the homologous arms,we constructed the PDC deletion cassette (PRFgHRP).The cassette was transformed into C.amazonensis CBS 12363 and transformants with hphm were derived.When the transforrnants were incubated into inducible medium,FLP-mediated recombination resulted in the deletion of DNA located between the repeats.[Results] SpMAL1p (induced by maltose) and SpGAL1p (induced by galactose) were identified to be strictly inducible promoters.SpGAL1p was used to regulate the expression of the FLP,and the PDC deletion cassette (PRFgHRP) was constructed and transformed into C.amazonensis successfully.After selection of Hyg-resistant transformant (designated as C.amazonensis PDC01) in which the deletion cassette was inserted into the PDC target gene,FLP expression was induced by growth of the transformant in galactose-containing medium,and Hyg-sensitive transformant in which hphm and caFLP flippers were excised from the genome was obtained,designated as C.amazonensis PDC02.[Conclusion] It is the first time to construct an FLP/FRT system for gene disruption in C.amazonensis,and we obtained a PDC mutant without resistant marker gene successfully through this system.These research results lay a good foundation for further metabolic engineering of C.amazonensis.
