Abstract:Electrospun fibers provide a promising platform for wound healing; however, they lack requisite characteristics for wound repair, including antibacterial and anti-inflammatory properties and angiogenic ability. Sodium alginate (SA) is being used for different types of applications. However, the poor spinnability of SA restricts its applications. The objectives of this study were three-fold: a) to synthesize oxidized sodium alginate (OSA) to improve its spinnability, b) to fabricate composite fibrous membranes by blending OSA along with zinc oxide nanoparticles (ZnO-NPs), and c) to decipher antibacterial and anti-inflammatory properties as well as biocompatibility of membranes in vitro and in vivo. OSA displaying different oxidation degrees (Dox (%)) was synthesized by varying the molar ratio of sodium periodate to SA. OSA (Dox, ∼48 %) afforded smooth and uniform fibers; 0.5 wt% of adipic dihydrazide (ADH) evolved into structurally stable and water-insoluble membranes. Composite fibrous membranes containing 2 wt% of ZnO-NPs displayed good biocompatibility and bactericidal effect against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) in vitro. In addition, composite membranes showed remarkable epithelialization, neovascularization, and anti-inflammatory response than that of the membranes devoid of ZnO-NPs. Conclusively, these composite fibrous membranes may have broad implications for wound healing applications.
Physical characterization, biocompatibility, and antimicrobial activity of polyvinyl alcohol/sodium alginate blend doped with TiO2 nanoparticles for wound dressing applications

Abstract:The ability of wound dressing materials to tackle skin pathogens colonization that is associated with open wound infections is limited. Recently, green-synthesized metal oxide nanoparticles has received a lot of attention to overcome this limitation. However, titanium dioxide nanoparticles (TiO 2 -NPs) exhibit exceptional antibacterial properties. In this work, several concentrations (0, 1, 3, and 5 wt.%) of TiO 2 NPs prepared using Aloe vera leaf extract were added to a blend of polyvinyl alcohol and sodium alginate (PVA:SA). This nanocomposite was designed to enhance the healing process of wounds. The interaction between the PVA:SA composite and the TiO 2 NPs was confirmed by FTIR. The thermal behavior of the nanocomposite films was investigated using DSC and TGA. The experimental results indicate that the glass transition temperatures of the nanocomposites increased by increasing the added amount of TiO 2 NPs to be 53.7 °C (1 wt.%), 55.8 °C (3 wt.%), and 60.6 °C (5 wt.%), which were consistently lower than the glass transition temperature of the matrix material (69.6 °C). The Dynamic Mechanical Analysis was examined. The nanocomposite doped with 5 wt.% of TiO 2 NPs detected a high storage modulus (21.6 × 10 8 ). Based on swelling and degradation studies, the prepared PVA:SA:TiO 2 nanocomposite films have an excellent swelling rate, and the inclusion of TiO 2 NPs increases the stability of the polymeric matrix. The PVA:SA:TiO 2 nanocomposite films exhibited a superior antibacterial efficacy against Gram-positive bacteria such as Bacillus cereus and Staphylococcus aureus , compared to their effectiveness against Gram-negative bacteria like Escherichia coli . Moreover, the nanocomposite films were biocompatible with Human Skin Fibroblast. Therefore, the developed PVA:SA:TiO 2 nanocomposite films suit wound dressing applications.

Abstract:This study aims to develop sodium alginate (SA) based ZnO nanoparticles as antibacterial film for wound healing applications. As natural materials, sodium alginate has been widely investigated and used in wound healing. The ZnO nanoparticles were incorporated as antibacterial into sodium alginate with different compositions (2, 4, 6, 8, 10 ml). Solvent casting method was performed to prepare a film. The film was characterized by using Fourier Transform Infrared Spectroscopy (FT-IR), Atomic Force Microscopy (AFM) and Field Emission Scanning Electron Microscopy (FESEM). Effectiveness of antibacterial activity was carried out against Gram-negative (Escherichia coli) and Gram-positive (Staphylococcus aureus) to characterize the sample. The results from microstructure analysis indicated that as ZnO nanoparticle compositions increased, the film surface became rough. It was found that sodium alginate incorporated with ZnO had slightly higher antibacterial activities against S. aureus than E. coli. Thus, sodium alginate (SA)-Zinc oxide (ZnO) nanoparticle has the potential to be used as a wound healing material in biomedical applications.
Characterization of Polyvinyl Alcohol/ZnO Nanocomposite Hydrogels for Wound Dressings

Abstract:Polymeric hydrogels, as novel soft biomaterials, have found various applications in the medical field; for instance, as wound dressings and wound care management. In this study, in order to achieve wound dressings with improved properties, nanocomposite hydrogels using incorporation of nano zinc oxide (ZnO) in polyvinyl alcohol (PVA), were prepared by a freezing-thawing method. The effect of ZnO loading on the mechanical, structural, physical and antibacterial properties of the samples was investigated and the morphology of the prepared nanocomposite hydrogels was determined by field emission scanning electron microscopy (FESEM). The results of mechanical tests of the nanocomposite hydrogels, using compressive and hardness tests, indicated that by adding increasingly amounts of the ZnO nanoparticles, the mechanical properties of the nanocomposite hydrogels were improved. Structure-related properties of the hydrogels, such as swelling and gel fraction measurements, showed that the increase in ZnO content lead to a decrease in the swelling rate and increase in the gel fraction of the nanocomposites. Antibacterial and biocompatibility analysis revealed that, with increasing nano ZnO content, the antibacterial properties of the nanocomposite significantly increased and the cells viability rose, leading to slightly reduced toxicity. The results of this work indicated that the presence and quantity of added ZnO in the PVA hydrogels were very influential factors to obtain the desirable characteristics of wound dressings, such as absorbing the wound fluids and exudates, being biocompatible, anti-bacterial and nontoxic, and having mechanical stability and flexibility.
Synergistic antibacterial and wound healing effects of chitosan nanofibers with ZnO nanoparticles and dual antibiotics

Abstract:One concern that has been considered potentially fatal is bacterial infection. In addition to the development of biocompatible antibacterial dressings, the screening and combination of new antibiotics effective against antibiotic resistance are crucial. In this study, designing hemostasis electrospun composite nanofibers containing chitosan (CS), polyvinyl pyrrolidone (PVP) and Gelatin (G) as the major components of hydrogel and natural nanofibrillated sodium alginate (SA)/polyvinyl alcohol (PVA) and ZnO nanoparticles (ZnONPs) combination as the nanofiller ingredient, has been investigated which demonstrated significant potential for accelerating wound healing. The hydrogels were developed for the delivery of the amikacin and cefepime antibiotics, along with zinc oxide nanoparticles that were applied to an electrospun layer. Amikacin is a highly effective aminoglycoside antibiotic, particularly for hospital-acquired infections, but its use is limited due to its toxicity. By utilizing it in low concentrations in the form of nanofibers and combining it with cefepime, which exhibits synergistic effects, enhanced efficacy against bacterial pathogens is achieved while potentially minimizing cytotoxicity compared to individual antibiotics. This dressing demonstrated efficient drug release, flexibility, and good swelling properties, indicating its suitable mechanical properties for therapeutic applications. After applying the biocompatible hydrogel to wounds, a significant acceleration in wound closure was observed within 14 days compared to the control group. Furthermore, the notable antibiotic and anti-inflammatory properties underscore its effectiveness in wound healing, making it a promising candidate for medical applications.

Abstract:Antibacterial and cyto-compatible tricomponent composite electrospun nanofibers comprised of polyvinyl alcohol (PVA), copper II oxide nanoparticles (CuONPs), and Momordica charantia (bitter gourd, MC) extract were examined for their potential application as an effective wound dressing. Metallic nanoparticles have a wide range of applications in biomedical engineering because of their excellent antibacterial properties; however, metallic NPs have some toxic effects as well. The green synthesis of nanoparticles is undergoing development with the goal of avoiding toxicity. The aim of adding Momordica charantia extract was to reduce the toxic effects of copper oxide nanoparticles as well as to impart antioxidant properties to electrospun nanofibers. Weight ratios of PVA and MC extract were kept constant while the concentration of copper oxide was optimized to obtain good antibacterial properties with reduced toxicity. Samples were characterized for their morphological properties, chemical interactions, crystalline structures, elemental analyses, antibacterial activity, cell adhesion, and toxicity. All samples were found to have uniform morphology without any bead formation, while an increase in diameters was observed as the CuO concentration was increased in nanofibers. All samples exhibited antibacterial properties; however, the sample with CuO concentration of 0.6% exhibited better antibacterial activity. It was also observed that nanofibrous mats exhibited excellent cytocompatibility with fibroblast (NIH3T3) cells. The mechanical properties of nanofibers were slightly improved due to the addition of nanoparticles. By considering the excellent results of nanofibrous mats, they can therefore be recommended for wound dressing applications.

Abstract:In the current research, synthesized Se-NPs were loaded into sodium alginate/polyvinyl alcohol SA/PVA blend, 1:2 (w:w), with different proportions to form nanocomposite films by using the solution casting method. The main goal is to enhance the physical and antibacterial properties of the polymer blend; therefore, it can be utilized for wound dressing applications. The physical aspects of the prepared films were characterized by X-ray diffraction (XRD), Fourier transform infrared (FTIR), atomic force microscope (AFM), scanning electron microscope (SEM), Energy dispersive X-ray (EDX), UV–Vis spectroscopic technique, and mechanical analysis to show the effect of increasing the concentration of Se-NPs from zero to 0.5 wt% on their properties. Regarding the XRD analysis, the calculated values of lattice strain ( ) increased from 0.005 to 0.013. The FTIR absorption spectrum recorded an intensity reduction and position shift in the main characteristic peaks of SA/PVA. The morphology of Se-NPs revealed a spherical shape with a mean diameter of 131.76 nm, as revealed by transmission electron microscopy (TEM). The surface morphological images showed the appearance of a few cracks; besides, the average roughness increased from 1.59 to 2.08 nm. According to the UV–Vis spectroscopic analysis, the optical band gap reduces (from 5.425 to 5.216 eV) for the direct transition and from (5.07–4.78 eV) for the indirect transition, which is an indication of the interaction between Se-NPs and SA/PVA blend. Young's modulus, tensile strength, and yield strength of the films increased with the addition of selenium nanoparticles. On the other hand, the amount of selenium release significantly increased with enhancing the concentration of Se-NPs in SA/PVA blend film from 0.1 to 0.5 wt%. Additionally, the inclusion of Se-NPs supplied the polymer blend with excellent antimicrobial activity, which would remove the possibility of microbial infection at the wound site. Moreover, the prepared SA/PVA/Se nanocomposite films have good biocompatibility with Human Skin Fibroblast (HSF). The results suggest the potential of this nanocomposite to be used in wound dressing applications.
Electrospun polyvinyl alcohol-polyvinyl pyrrolidone nanofibrous membranes for interactive wound dressing application

Abstract:Cross-linked polyvinyl alcohol (PVA) and polyvinyl pyrrolidone (PVP) composite nanofibrous membranes have been prepared by electrospinning. Mechanical properties of the membranes improved significantly with PVP addition. PVP improved hydrophilicity and sustainable degradation of the membranes. Biocompatibility of the membranes was assessed by in vitro culture of native skin cells (L929 fibroblast and HaCaT keratinocytes). Tests showed sustained release of the antibiotic ciprofloxacin hydrochloride monohydrate by the membranes. Further, zone of inhibition study against Staphylococcus aureus growth demonstrated protective action against external pathogenic microbes. These studies show these simple PVA-PVP nanofibrous membranes are promising interactive antibiotic-eluting wound dressing materials.
A novel electrospun membrane based on moxifloxacin hydrochloride/poly(vinyl alcohol)/sodium alginate for antibacterial wound dressings in practical application

Abstract:This study reports on the performance of sodium alginate (SA)/poly(vinyl alcohol) (PVA)/moxifloxacin hydrochloride (MH) nanofibrous membranes (NFM) capable of providing antibacterial agent delivery for wound-dressing applications. The aim of this work was to prepare antibacterial NFM with good permeability properties by employing PVA and SA as carriers. A group of 12% PVA/2% SA solutions blended in various ratios (8:2, 7:3, 6:4, 5:5 and 4:6, v/v) and containing 0.5, 1, 2 or 4 wt% MH were studied for electrospinning into nanoscale fibermats. The optimum ratio found to form smooth fibers with uniform fibrous features was 6:4. The drug release behavior of the electrospun, the antibacterial effects on Pseudomonas aeruginosa and Staphylococcus aureus and the animal wound dressing capabilities were also investigated. As much as 80% of the MH was released from the electrospun after 10 h of incubation at 37 °C. In addition, the NFM with 0.5 MH exhibited less activity, whereas those with higher concentrations of MH exhibited greater antibacterial effect. Furthermore, the MH-loaded electrospun accelerated the rate of wound dressing compared to other groups. The results of the in vitro and in vivo experiments suggest that MH/PVA/SA nanofibers might be an interesting bioactive wound dressing for clinical applications.

Abstract:In the field of wound dressings, hydrogels have become essential due to their biocompatibility and moisture-retention properties. Nevertheless, challenges persist, notably in terms of mechanical strength and antibacterial effectiveness. In this study, we synthesized and characterized polymeric wound dressings comprising sodium alginate and polyethylene glycol, cross-linked using calcium chloride. We incorporated silver and zinc oxide nanoparticles to enhance antibacterial properties. The hydrogels were prepared through solution casting and subjected to comprehensive characterization, including X-ray diffraction (XRD), scanning electron microscopy (SEM), Fourier-transform infrared spectroscopy (FTIR), and thermogravimetric analysis (TGA). XRD revealed a distinct face-centered structure. SEM analysis revealed phase separation and uniform nanoparticle dispersion, and TGA indicated improved thermal stability. The hydrogel exhibited exceptional tensile strength (30.8 MPa), low cytotoxicity (SAPEG/0.01Ag/0.02ZnO), and increased hydrophilicity (contact angle: 27.5°–15.6°) post-nanoparticle addition. Significant antibacterial activity (largest zone of inhibition: 27.25 mm for SA/PEG/0.01Ag/0.02ZnO) aligns these hydrogel membranes with characteristics vital for wound healing applications, making them promising for advanced wound dressings.

Enhanced wound healing with biogenic zinc oxide nanoparticle-incorporated carboxymethyl cellulose/polyvinylpyrrolidone nanocomposite hydrogels

Abstract:Contemporary wound dressings lack antibacterial properties, exhibit a low water vapour transmission rate, and demonstrate inadequate porosity. In order to overcome these limitations, scientists have employed water hyacinth to produce carboxymethyl cellulose (CMC). CMC/PVP nanocomposite films containing biogenic zinc oxide nanoparticles (nZnOs) were synthesised using cost effective solution-casting technique. As the proportion of nZnOs in the film increased, swelling and water permeability decreased, whereas mechanical stability improved. Dynamic light scattering testing and transmission electron microscopy confirmed that the particle size was around 50.7 nm. Field emission scanning electron microscopy (FESEM) images showed that nZnOs were distributed uniformly in the polymer matrix. Cell viability against Vero cells was greater than 94%, and a substantial zone of inhibition against S. aureus and E. coli bacteria was observed. Wounds of albino mice were treated with CMC/PVP and CMC/PVP/nZnO (6%) nanocomposite hydrogels and healed in 20 and 12 days, respectively, as demonstrated by wound healing assay and histological staining. In vitro and in vivo studies revealed that the novel nanocomposite hydrogels exhibit improved cell viability and wound healing features. Therefore, they could be exploited as promising skin wound dressing materials.

Abstract:Biocompatible PVA‐CS‐GO‐ZnO polymer composites with excellent electrical and antibacterial properties. Polymer nanocomposites have been utilized for wound healing applications. Compared to the conventional polymer composites, conducting polymer nanocomposites comprising a blend of biocompatible polymers with electroconductive and antibacterial nanofillers such as graphene‐based nanomaterials and metal/metal oxides, have attracted great attention in this arena due to enhanced physical, electrical and biological characteristics. Herein, polyvinyl alcohol (PVA)/chitosan (CS)/graphene oxide/zinc oxide electroconductive and antibacterial composite hydrogel films were fabricated by solution casting technique, for wound healing applications. The physicochemical and conductivity studies were carried out for all the polymer samples. The successful incorporation of the nanofillers and their even distribution into the PVA‐CS matrix was validated by the scanning electron microscopy‐EDAX, x‐ray diffractometer, Fourier‐transform infrared, and Raman analytical results. An enhancement in the surface roughness of the nanofiller incorporated composite film was elucidated by the topographical images obtained from AFM analysis. The composite with higher concentration of nanofillers exhibited a maximum conductivity of 0.35 × 10−4 Scm−1 compared to the other fabricated composites. A good antibacterial efficacy, minimal hemolytic activity as well as better thrombogenicity were portrayed by the composite films. Additionally, the nanocomposite films exhibited no cytotoxicity and excellent cell attachment and proliferation of RAW 264.7 cells. Therefore, based on the obtained results, it can be concluded that the composite with higher particle concentration, is an ideal candidate for futuristic wound dressing applications with electroconductivity, antimicrobial activity, hemostatic ability, and biocompatibility. Alginate Hydrogels with Embedded ZnO Nanoparticles for Wound Healing Therapy

Abstract:Introduction: In this in-vitro study, we designed a 3D printed composite of zinc oxide (ZnO) nanoparticles (NPs) with photocatalytic activities encapsulated within hydrogel (alginate) constructs, for antibacterial purposes applicable towards wound healing. We primarily sought to confirm the mechanical properties and cell compatibility of these ZnO NP infused scaffolds. Methods: The antibacterial property of the ZnO NPs was confirmed by hydroxyl radical generation using ultraviolet (U.V.) photocatalysis. Titanium dioxide (TiO2), a well-known antibacterial compound, was used as a positive control (1% w/v) for the ZnO NP-based alginate constructs and their antibacterial efficacies compared. Among the ZnO group, 3D printed gels containing 0.5% and 1% w/v of ZnO were analyzed and compared with manually casted samples via SEM, swelling evaluation, and rheological analysis. Envisioning an in-vivo application for the 3D printed ZnO NP-based alginates, we studied their antibacterial properties by bacterial broth testing, cytocompatibility via live/dead assay, and moisture retention capabilities utilizing a humidity sensor. Results: 3D printed constructs revealed significantly greater pore sizes and enhanced structural stability compared to manually casted samples. For all samples, the addition of ZnO or TiO2 resulted in significantly stiffer gels in comparison with the alginate control. Bacterial resistance testing on Staphylococcus epidermidis indicated the addition of ZnO NPs to the gels decreased bacterial growth when compared to the alginate only gels. Cell viability of STO-fibroblasts was not adversely affected by the addition of ZnO NPs to the alginate gels. Furthermore, the addition of increasing doses of ZnO NPs to the alginate demonstrated increased humidity retention in gels. Discussion: The customization of 3D printed alginates containing antibacterial ZnO NPs leads to an alternative that allows accessible mobility of molecular exchange required for improving chronic wound healing. This scaffold can provide a cost-effective and durable antibacterial treatment option.

Abstract:Wound dressing from PVA/NaCAS/AgNP electrospun mats. In this study, nanofibers were produced by the electrospinning method by adding 1%, 3%, 5%, and 7% silver nanoparticles (Ag NPs) into the polyvinyl alcohol/sodium caseinate (60/40, w/w) mixture. Electrospun materials were cross‐linked by immersion in a glutaraldehyde bath. Thus, antibacterial wound dressings that could be used on exuding wounds were developed. According to the scanning electron microscopy images of nanofibers, the cross‐linking process by immersion method did not cause negative effects such as breakage or adhesion on the surface of fibers. The cross‐linking of nanofibers was demonstrated by the presence of acetal linkages by Fourier‐transform infrared spectroscopy analysis. The cross‐linking process significantly improved the thermal properties of the nanofibers. The lowest crystallinity calculated in the differential scanning calorimeter was observed in the 3% Ag NP‐added nanofiber with a value of 1.83%. It exhibited the lowest total soluble matter content with a value of 11.98% owing to its high cross‐linking density. In addition, the 3% Ag NP‐added nanofiber, which had the highest toughness and ductility, displayed the highest tensile stress with 3.5 MPa and the highest tensile strain with 7.53%. Moreover, it showed 78.11% cell viability on the L929 fibroblast cell line at the end of the 24th hour. It was indicated that although the electrospun mats maintained 100% antibacterial effectiveness, they could not be used as a wound dressing for 48 h. It was reported that only 3% Ag NP‐added nanofiber could be used as a 100% effective antibacterial wound dressing for both Escherichia coli and Staphylococcus aureus provided that it was renewed every 24 h. Consequently, it was notified that cross‐linking by the immersion method can be used in biomedical applications. Highlights The antibacterial effects of Ag NP‐doped PVA/NaCAS electrospun mats were significant. The cross‐linking process did not induce toxic effects some of the nanofibers. High cross‐linking density was observed in the nanofibers. The swelling property of nanofibers is a good feature required in an ideal wound dressing.

Abstract:Wound healing constitutes a formidable challenge within the healthcare system, attributable to infection risks and protracted recovery periods. The pressing need for innovative wound healing methods has spurred the urgency to develop novel approaches. This study sought to advance wound healing by introducing a novel approach employing a composite sponge dressing. The composite sponge dressing, derived from LFL-ZnO (synthesized through the green methodology utilizing Lactobacillus plantarum ZDY2013 fermentation liquid), polyvinyl alcohol (PVA), and sodium alginate (SA) via a freeze–thaw cycle and freeze-drying molding process, demonstrated notable properties. The findings elucidate the commendable swelling, moisturizing, and mechanical attributes of the SA/LFL-ZnO/PVA composite sponge dressing, characterized by a porous structure. Remarkably, the dressing incorporating LFL-ZnO exhibited substantial inhibition against both methicillin-resistant Staphylococcus aureus (MRSA) and Staphylococcus aureus (S. aureus). Hemolysis and cytotoxicity tests corroborated the excellent biocompatibility of the sponge dressing. In vivo evaluation of the therapeutic efficacy of the 1 mg/mL LFL-ZnO composite dressing on scald wounds and S. aureus-infected wounds revealed its capacity to accelerate wound healing and exert pronounced antibacterial effects. Consequently, the composite sponge dressings synthesized in this study hold significant potential for application in wound treatment.

Abstract:Developing a variety of safe and effective functioning wound dressings is a never-ending objective. Due to their exceptional antibacterial activity, biocompatibility, biodegradability, and healing-promoting properties, functionalized chitosan nanocomposites have attracted considerable attention in wound dressing applications. Herein, a novel bio-nanocomposite membrane with a variety of bio-characteristics was created through the incorporation of zinc oxide nanoparticles (ZnONPs) into amine-functionalized chitosan membrane (Am-CS). The developed ZnO@Am-CS bio-nanocomposite membrane was characterized by various analysis tools. Compared to pristine Am-CS, the developed ZnO@Am-CS membrane revealed higher water uptake and adequate mechanical properties. Moreover, increasing the ZnONP content from 0.025 to 0.1% had a positive impact on antibacterial activity against Gram-positive and Gram-negative bacteria. A maximum inhibition of 89.4% was recorded against Escherichia coli, with a maximum inhibition zone of 38 ± 0.17 mm, and was achieved by the ZnO (0.1%)@Am-CS membrane compared to 72.5% and 28 ± 0.23 mm achieved by the native Am-CS membrane. Furthermore, the bio-nanocomposite membrane demonstrated acceptable antioxidant activity, with a maximum radical scavenging value of 46%. In addition, the bio-nanocomposite membrane showed better biocompatibility and reliable biodegradability, while the cytotoxicity assessment emphasized its safety towards normal cells, with the cell viability reaching 95.7%, suggesting its potential use for advanced wound dressing applications.
Preparation of antibacterial ZnO NP-containing schizophyllan/bacterial cellulose nanocomposite for wound dressing

Abstract:Nanocomposite hydrogel is helpful to provide a moist and ideal environment for wound healing. In this research study, a nanocomposite hydrogel was prepared based on schizophyllan (SPG) and amine-functionalized bacterial cellulose (amine-functionalized BC). ZnO-nanoparticles were loaded into amine-grafted BC/SPG hydrogel to improve its antibacterial feature. The successful functionalization of bacterial cellulose by (3-aminopropyl)triethoxysilane was verified by FTIR and XRD analyses. XRD pattern of amine-grafted BC/SPG/ZnO NP showed some new peaks in comparison with XRD pattern of amine-grafted BC/SPG which verified the intercalation of ZnO into polymeric matrix. SEM images showed that the porous spaces of BC have been filled by schizophyllan as a result of the intermolecular attraction between them. Deposition of ZnO nanoparticles on the surface of amine-grafted BC/SPG has also been verified by SEM analysis. Amine-grafted BC/SPG membrane showed the higher swelling degree (850 ± 48%) in comparison with BC (512 ± 29%). The swelling ratio decreased to 725 ± 19% by addition of ZnO nanoparticles. Amine-grafted BC/SPG exhibited the higher mechanical strength (45 MPa) in comparison with pristine BC (10 MPa). The tensile strength of amine-grafted BC/SPG/ZnO NP nanocomposite reached ~ 70 MPa that might be resulted from the formation of hydrogen bonding between hydroxyl groups of polymers and oxygen atoms in the ZnO NPs. Amine-grafted BC/SPG/ZnO showed a more remarkable antibacterial efficiency against E. coli and S. aureus. As compared to BC, amine-grafted BC/SPG could stimulate the proliferation of human fibroblast cell. Cell viability showed a slight decrease after exposure to amine-grafted BC/SPG/ZnO NP nanocomposite.

Abstract:Known for its water solubility, flexibility, strong adhesion, and eco-friendly nature, polyvinyl alcohol (PVA) is widely used in various industries. In the medical field, it is used for applications such as creating bandages and orthopaedic devices. Incorporating sodium alginate (SA) into PVA membranes enhances their structural integrity, breathability, and permeability, thereby minimising the risk of cellular damage in the wound zone. Moreover, the addition of tamanu oil (C alophyllum inophyllum L.) and silver nanoparticles, both of which are known for their antibacterial properties and benefits in traditional wound healing, further enhances the membranes' wound-healing effectiveness. Following production, the membranes undergo a series of tests designed to evaluate their physical properties as well as their antioxidant and antibacterial capabilities. Subsequently, in vitro testing is conducted using human skin cells; experiments on Wistar rats are then performed. Numerous experiments have consistently demonstrated that the performance of polyvinyl alcohol/sodium alginate/tamanu oil (PVA/SA/Oil) membrane is superior to that of polyvinyl alcohol/sodium alginate/tamanu oil/silver nanoparticles (PVA/SA/Oil/Ag NP) membrane. Specifically, the polyvinyl alcohol/sodium alginate (PVA/SA) combination exhibits an impressive wound-healing rate of 98.82% after 15 days, with cells maintaining a high viability of 92% in a nourishing environment. Moreover, these membranes exhibit exceptional resistance to the oxidation of free radicals, surpassing the 70% threshold, and they possess antibacterial activity against Staphylococcus aureus subsp. aureus in vitro. Based on the obtained results, the nanofiber membranes composed of polyvinyl alcohol/ alginate/ tamanu oil, with or without silver nanoparticles, have shown potential as wound dressings in the wound care discipline.

Abstract:To improve the wound dressing characteristics of bacterial cellulose-based materials and address the issue of asymmetric wound dressing with one hydrophilic side and another hydrophobic side, this study developed a new concept for the fabrication of an asymmetric wettable and antibacterial wound dressing by selective drop coating of stearic acid-modified ZnO, chitosan, and AgNPs to form a dot pattern on both surfaces of a bacterial cellulose–alginate composite (BA-ZnS/Ch/Ag). The coated surface was hydrophobic, with a WCA of 150° due to the formation of a low surface energy zinc stearate (C 17 H 35 COO) 2 Zn) monolayer on the ZnO particles and a high degree of hierarchical roughness. The asymmetric wettable BA-ZnS/Ch/Ag wound dressing maintained good water absorptivity (swelling rate 417%) and natural breathability (water vapor transmission rate 792 g.m −2 day −1 ) of the superhydrophilic bacterial cellulose-alginate composite that consisted of dense outer surfaces and porous inner layers and simultaneously possessed the superhydrophobic property of the coating area that can reduce the risk of infection from external fluids and improve the blood repellency and anti-adhesion properties. The BA-ZnS/Ch/Ag wound dressing showed good antibacterial activity against S. aureus and E. coli and non-toxicity to human keratinocyte immortal cells (HaCaT), making it suitable for clinical applications.
