Abstract:Matrix metalloproteinase-1 (MMP1) is a member of the matrix metalloproteinases family, and its aberrant expression is implicated in tumor invasion and metastasis. However, the relationship between MMP1 abnormal expression and clinical outcome in breast cancer patients remains to be elucidated. To address this issue, we conducted immunohistochemistry in breast cancer and adjacent normal tissues, and mined the transcriptional and survival data of MMP1 in breast cancer patients through Oncomine, Kaplan-Meier Plotter, bc-GenExMiner, COSMIC and cBioPortal databases. First, we found that both protein and mRNA levels of MMP1 expression were significantly higher in breast cancer tissues. Second, high MMP1 mRNA expression correlated with worse overall survival among grade II (HR = 1.75; p = 0.011), nodal-negative (HR = 2.00; p = 0.00028), ER-positive (HR = 1.61; p = 0.00027) and HER2-negative (HR = 3.17; p = 0.029) patients with breast cancer by using Kaplan-Meier plotter database. Third, the overexpression of MMP1 was associated with unfavorable survival results including overall survival (HR = 1.6; p = 1.6e-05), relapse free survival (HR = 1.78; p < 1e-16) and distant metastasis free survival (HR = 1.65; p = 5.3e-05) in patients with breast cancer. Taken together, the expression status of MMP1 is a significant prognostic indicator and a potential drug target for breast cancer.

Abstract:We investigated the programming of generalization and maintenance of correspondence between verbal and nonverbal behavior in a preschool setting. Four children participated in a series of multiple-baseline designs. In Experiment 1, delayed reinforcement of verbal behavior effectively controlled maintenance of correspondence with previously trained responses and also resulted in generalization of correspondence to one untrained response. As the latter effect was limited, Experiment 2 was a further assessment of the effects of delayed reinforcement of generalization of correspondence to untrained responses, and consistent generalization was shown. Experiment 2 also showed that generalization, if lost, could be recovered through use of "booster training," in which the original contingencies were reinstated for a brief period. Experiment 3 provided replications, with two additional children, of the effects of delayed reinforcement on maintenance of correspondence. Results are discussed in terms of using delayed reinforcement as an indiscriminable contingency.
Association of Matrix Metalloproteinase‐2 and ‐9 Promoter Polymorphisms with Colorectal Cancer in Chinese

Abstract:Matrix metalloproteinase‐2 (MMP‐2) and ‐9 (MMP‐9) play an important role in cancer initiation, invasion, and metastasis. The aim of this study was to investigate whether common genetic variants in these two key MMPs are associated with the development and progression of colorectal cancer in a Chinese population. We detected the MMP‐2‐790T/G, ‐955A/C, ‐1575G/A, and MMP‐9‐1562 C/T polymorphisms in 126 colorectal cancer patients and matched normal controls by PCR‐denaturing high‐performance liquid chromatography (DHPLC) or PCR‐restriction fragment length polymorphism (RFLP), respectively. We found that the G/G genotype in the MMP‐2‐1575G/A polymorphism was significantly increased in colorectal cancer patients than in the controls (odds ratio (OR), 1.96; 95% CI, 1.06–3.64). Colorectal cancers with G/G genotype were more common with serosa/adventitia layer involvement than those with G/A + A/A genotypes (P < 0.05). However, no significant differences were observed in distribution of the MMP‐2‐790T/G, ‐955A/C, and MMP‐9‐1562 C/T polymorphisms or haplotype of MMP‐2 SNPs between patients and controls. Our results suggest that the presence of ‐1575G allele in the MMP‐2 promoter region may be of significance in the assessment of colorectal cancer risk and invasive potential. © 2007 Wiley‐Liss, Inc.
Prognostic Significance of Matrix Metalloproteinase 7 Immunohistochemical Expression in Colorectal Cancer: a Meta-Analysis.

Abstract:Matrix metalloproteinase 7 (MMP-7) was speculated to have a key role in the development and progression of human cancer. Considerable studies investigated the relationship between its expression and survival in colorectal cancer (CRC), but inconsistent results were obtained. The clinical significance of MMP-7 overexpression in CRC remains controversial. Therefore, in this article, we conducted a meta-analysis to analyze the prognostic value of MMP-7 in CRC. We searched studies in PubMed, Medline, and Web of Science databases until August 2014 to find relevant studies. A total of six high-quality studies met the inclusion criteria and 1631 patients were included in our study. Combined hazard ratios (HRs) suggested that MMP-7 overexpression had an unfavorable impact on overall survival (HR = 1.83, 95% CI: 1.24-2.71). Subgroup and sensitivity analyses further validated the role of MMP-7 as a predictor for prognosis. In conclusion, MMP-7 overexpression detected by immunohistochemistry indicated worse prognosis in CRC and may help to guide clinical therapy.
Relationship of MMP-2mRNA Expression with Clinical Pathological Features and Prognosis in Gastric Cancer

Abstract:Objective To explore the relationship of matrix metalloproteinase-2 mRNA(MMP-2mRNA) expression with clinical pathological features and prognosis in human gastric cancer.Methods RT-PCR was used to detect MMP-2mRNA expression of tumor tissues and adjacent tissues(≥5cm) from 67 patients with gastric cancer.Data was analyzed using Kaplan-Meier method.Results The positive expression of MMP-2mRNA in carcinoma tissues was significantly higher than that in adjacent tissues(31 vs 19) and had close relationship with lymphnode metastasis(61.9%).But Kaplan-Meier analysis showed no differences in average survival time between different expressions of MMP-2mRNA.Conclusion MMP-2 plays an important role in tumor formation,invasion and metastasis in gastric cancer,but MMP-2mRNA expression may not be an independent index of the prognosis.

Abstract:OBJECTIVE:To investigate whether the polymorphisms of matrix metalloproteinase-2 (MMP-2) and -9 (MMP-9) promoters contribute to the development and progression of colorectal cancer in Chinese population.METHODS:the PCR-based denaturing high-performance liquid chromatography or PCR-restriction fragment length polymorphism technique respectively was applied to analyze the MMP-2 -1306C/T and MMP-9 -1562C/T polymorphisms in normal group (126 individuals) and colorectal cancer group (126 cases). Genotype frequencies were compared between patients and matched controls, and the association of genotypes with clinical-pathological parameters was studied.RESULTS:The frequency of the CC genotype in the MMP-2 gene polymorphism was significantly increased in colorectal cancer patients when compared with controls (P<0.05), and individuals with the CC genotype had an increased risk of developing colorectal cancer compared to those with CT+TT genotypes (OR: 1.959; 95%CI: 1.055-3.637). Significant correlation was found between the depth of tumor invasion and MMP-2 -1306C/T polymorphism in colorectal cancer patients. However, the genotype frequencies of MMP-9 -1562C/T in colorectal cancer patients were similar to those in control subjects.CONCLUSION:Our results indicate that MMP-2 -1306 C/T polymorphism may be associated with genetic susceptibility to colorectal cancer and the invasive capability of colorectal cancer in Chinese patients. And it is easier for the CC genotype cancer to invade through bowel wall.

Abstract:Matrix metalloproteinase-2 (MMP-2) is an enzyme with proteolytic activity against matrix proteins, particularly basement membrane constituents. A single nucleotide polymorphism C→T transition at −1306, which disrupts an Sp1-type promoter site (CCACC box), displayed a strikingly lower promoter activity with T allele. Our study investigated whether the MMP-2 −1306 C→T polymorphism contributed to the development and progression of colorectal cancer in the Chinese population. One hundred twenty-six colorectal cancer patients and 126 age- and sex-matched controls were included in this study. PCR-based denaturing high performance liquid chromatography analysis and sequencing were used to determine the MMP-2 genotypes. MMP-2 expression of each genotype was analyzed in four colorectal cancer cell lines by semi-quantitative RT-PCR. The correlation between the genotypes and clinicopathological parameters among colorectal cancer cases was investigated. The results showed that the levels of MMP-2 mRNA expression in cell lines containing CC genotype were much higher compared with cell with CT genotype. The frequency of MMP-2 CC genotype was significantly higher in colorectal cancer patients when compared with controls (OR, 1.959; 95% CI, 1.055–3.637). Colorectal cancers with CC genotype were more common with serosa/adventitia layer involvement compared with CT+TT genotypes. Our data suggest that MMP-2 −1306 C→T polymorphism may be associated with colorectal cancer development and invasion in the Chinese population.

Abstract:AIM: To investigate the relationship between matrix metalloproteinase-2 (MMP-2) mRNA expression and clinicopathologic and urokinase-type plasminogen activator (uPA) system parameter and prognosis in human gastric cancer. METHODS: Expression of MMP-2 mRNA, uPA, and uPA-R mRNA in tumor tissues and >= 5 cm adjacent normal tissues from 67 cases of gastric cancer was studied using RTPCR and Northern blot respectively. Survival analyses were done using the Kaplan-Meier method. RESULTS: The expression rates of MMP-2 mRNA, uPA and uPA-R mRNA in tumor tissues (31%, 41%, and 51%, respectively) were significantly higher than those in 5 cm adjacent tissues (19%, 11%, and 9%;chi(2) = 4.59, 43.58, and 53.24 respectively, P<0.05, 0.0001, and 0.0001, respectively). Expression of MMP-2 mRNA was significantly correlated with lymph node metastasis (metastasis: 61.9%, no metastasis: 39.1%, chi(2) = 7.61, P<0.05), Lauren's classification of diffuse/mixed types: 54.2%, intestinal type: 26.3%, chi(2) = 4.25, P<0.05, expression of uPA and uPA-R mRNA (uPA+: 55.1%, uPA-: 22.2% and uPA-R+: 54.9%, uPA-R-: 18.8%, chi(2) = 5.72 and 6.40 respectively, P<0.05). Kaplan-Meier survival analysis of MMP-2 mRNA expression did not show significant difference in all 67 cases, but revealed an association of the expression of MMP-2 mRNA, uPA, and uPA- R mRNA with worse prognosis (P = 0.0083, 0.0160, and 0.0094, respectively). Correlation between MMP2 expression in lung cancer tissues and clinical parameters: a retrospective clinical analysis

Abstract:Abstract Background Matrix metalloproteinase 2 (MMP2) has been found to be related to malignant tumors; the aim of this study was to investigate the correlation between MMP2 expression in lung cancer tissues and clinical parameters of lung cancer. Methods The expression of MMP2 in lung cancer tissues and in adjacent non-malignant tissues was tested by immunohistochemistry. The correlation between the expression of MMP2 and clinical parameters of lung cancer was analyzed by Kaplan-Meier curve and multiple regression analysis. Results The expression of MMP2 was higher in lung cancer tissues than that in adjacent non-malignant tissues ( p = 0.002). Increased MMP2 was associated with low differentiation ( p = 0.022), tumor size ( p = 0.032), lymph node metastasis ( p < 0.001), advanced stage ( p = 0.002). The post-surgical survival time in patients with high MMP2 expression was shorter than that in patients with low MMP2 expression ( p = 0.001). High expression of MMP2 ( p = 0.006) and advanced stage ( p = 0.003) were independent prognostic indicators for survival of lung cancer patients. Conclusions Increased MMP2 correlates with malignant biological behavior of lung cancer and it could be a potential biomarker for diagnosis and prognosis of the disease.

Abstract:e14116 Background: The high mobility group A2 (HMGA2) protein regulates the expression of a cohort of genes by either enhancing or suppressing their transcription. The change of HMGA2 mRNA or protein levels has been reported to correlate with malignant tumor phenotypes. Therefore, we hypothesized that the expression level of HMGA2 is related to cancer survival and serves as prognostic biomarkers for clinical outcome of colorectal cancers (CRCs). Methods: 191 consecutive CRCs who received surgical treatment in second affiliated hospital of Zhejiang University from 1999 to 2004 were entered into this outcome study. In addition, 89 CRCs from City of Hope were collected as validation set. Patients were periodically followed up for survival. The IHC study was conducted on tissue arrays to determine the expression of HMGA2 proteins. The Kaplan-Meier analysis and COX proportional hazard model were employed to conduct uni- and multi- variant survival analysis. Results: The IHC results displayed that the expression of HMGA2 could be seen significant higher in malignant tumor section in comparing corresponding normal colon epithelial cells. Univariate and multivariate logistic analysis indicated that the nuclear expression of HMGA2 is significantly related to distant metastasis among 191 CRCs (p < 0.05). Survival analysis revealed that the higher nuclear staining of HMGA2 is significantly related to poor survival of CRCs (p < 0.05). The adjusted hazard proportional ratios (HRs) are 1.81 (95% CI 1.04-3.12) and 1.86 (95% CI 1.08-3.22) for overall and progress-free survival respectively. Further analysis indicated that HMAG2 impacts the survival on CRCs at III-IV stage, rather than I-II stage. The HRs of HMGA2 at stage III-IV is 1.92 (95% CI 1.02-3.58) for overall survival, and 2.03 (95% CI 1.07-3.80) for progress-free survival. It was validated on stage III-IV CRCs from City of Hope (HRs=1.80 for overall survival, HRs=2.05 for progress-free survival). Conclusions: HMGA2 might play a critical role in promoting the invasion and metastasis phenotype of colorectal cancer. Overexpression of HMGA2 significantly impacts the survival on advanced CRCs. Therefore, HMGA2 may serve as a potential prognostic marker for predicting outcome of CRCs.

Abstract:BackgroundMatrix metalloproteinase 2 (MMP2) is a collagenase, which aids tumor growth and invasion by digesting the extracellular matrix surrounding the tumor tissue. Our study examined MMP2 expression in various stages of melanoma progression and tested the prognostic significance of MMP2 expression. We also analyzed the correlation between p-Akt status and MMP2 expression in melanoma patients.MethodsTissue microarray (TMA) and immunohistochemistry were employed to study the expression of MMP2. A total of 482 melanoma (330 primary and 152 metastatic) tumor biopsies and 149 nevi biopsies (49 normal and 100 dysplastic nevi) were used for the analysis. MMP2 expression was correlated with melanoma progression. Kaplan-Meier survival curve and multivariate Cox regression analysis were applied to verify the prognostic significance of MMP2 expression. The correlation between MMP2 and p-Akt expression was analyzed in 92 cases which were common in the present and the previous study on p-Akt expression.ResultsStrong MMP2 expression is significantly increased in primary (25 %) and metastatic melanoma (43 %) compared to normal (5 %) and dysplastic nevi (10 %). Patients with strong MMP2 had significantly poorer survival compared to those with negative-to-moderate MMP2 expression. MMP2 expression could predict the patient survival independent of tumor thickness and ulceration. Furthermore, in our cohort study MMP2 expression was associated with p-Akt status and patient survival.ConclusionsStrong MMP2 staining is associated with worse survival of melanoma patients and is an independent molecular prognostic factor for primary melanoma.

Abstract:Objective Research the correlation between expression of MMP-2 in colorectal cancer and patho- logical factors,or biological behaviors in colorectal cancer.Methods The expression of MMP-2 in 68 cases with col- orectal cancer were detected by immunohistochemical staining.Results The expression of MMP-2 in colorectal can- cer was not correlated with tumor region and histologic type,but related with depth of tumor invasion and metasta- sis.The frequency of positive cases in patients with colorectal cancer with of lymph,node metastasis was significantly higher than that in patients without lymph node metastasis(P0.025).Conclusion The detection of the expres- sion of MMP-2 in colorectal cancer was very important in adopting adjuvant chemotherapy and judging biological be- haviors.
Expression of MMP-9 and WAVE3 in Colorectal Cancer and Its Relationship to Clinicopathological Features

Abstract:Purpose To investigate matrix metalloproteinase 9 (MMP-9) and WAVE3 expression in human colorectal cancer (CRC) and to evaluate their clinical significance. Methods We first performed real-time PCR to evaluate mRNA expression of MMP-9 and WAVE3 in 21 pairs of fresh CRC samples matched with adjacent normal mucosa. Then, MMP-9 and WAVE3 proteins were evaluated by immunohistochemistry on CRC tissue microarrays which included 216 CRC specimens and corresponding normal colorectal mucosa, and their correlation with clinicopathological factors and overall survival after surgery was evaluated. Results Both real-time PCR and immunohistochemistry evaluation have demonstrated that MMP-9 and WAVE3 were over-expressed in colorectal cancer tissues compared with normal mucosa ( p < 0.001). MMP-9 expression was significantly higher in patients with low-grade differentiation and distant metastasis ( p = 0.003 and p = 0.005, respectively), and patients with MMP-9-positive expression had a poorer prognosis ( p = 0.008). However, patients with WAVE3-positive expression had a better prognosis ( p = 0.039) and particularly favorable prognostic factors, including non-lymph node metastasis, non-distant metastasis, and early TNM stage ( p = 0.029, 0.021, and 0.003, respectively). In addition, MMP-9-negative/WAVE3-positive patients had the best overall survival ( p = 0.021). In multivariate survival analysis, MMP-9 expression and combined expression status of MMP-9/WAVE3 were identified as independent prognostic factors for CRC ( p = 0.046 and p = 0.019, respectively). Conclusions Combined analysis of MMP-9 and WAVE3 has a significant value for assessing prognosis of CRC patients after surgery.
Prognostic values of tumoral MMP2 and MMP9 overexpression in breast cancer: a systematic review and meta-analysis

Abstract:Abstract Background Breast cancer (BC) is a leading cause of cancer-related death in females worldwide. Previous studies have demonstrated that matrix metalloproteinases (MMPs) play key roles in metastasis and are associated with survival in various cancers. The prognostic values of MMP2 and MMP9 expression in BC have been investigated, but the results remain controversial. Thus, we performed the present meta-analysis to investigate the associations between MMP2/9 expressions in tumor cells with clinicopathologic features and survival outcome in BC patients. Methods Eligible studies were searched in PubMed, Web of Science, EMBASE, CNKI and Wanfang databases. The associations of MMP2/9 overexpression in tumor cells with overall survival (OS), disease-free survival (DFS) and recurrence-free survival (RFS) were assessed by hazard ratio (HR) and 95% confidence interval (CI). The associations of MMP2/9 overexpression with clinicopathological features were investigated by calculating odds ratio (OR) and 95% CI. Subgroup analysis, sensitivity analysis, meta-regression, and analysis for publication bias were performed. Results A total of 41 studies comprising 6517 patients with primary BC were finally included. MMP2 overexpression was associated with an unfavorable OS (HR = 1.60, 95% CI 1.33 –1.94, P < 0.001) while MMP9 overexpression predicted a shorter OS (HR = 1.52, 95% CI 1.30 –1.77, P < 0.001). MMP2 overexpression conferred a higher risk to distant metastasis (OR = 2.69, 95% CI 1.35–5.39, P = 0.005) and MMP9 overexpression correlated with lymph node metastasis (OR = 2.90, 95% CI 1.86 – 4.53, P < 0.001). Moreover, MMP2 and MMP9 overexpression were both associated with higher clinical stage and histological grade in BC patients. MMP9 overexpression was more frequent in patients with larger tumor sizes. Conclusions Tumoral MMP2 and MMP9 are promising markers for predicting the prognosis in patients with BC.

Abstract:ABSTRACT Objective:To measure the expression levels of matrix metalloproteinase 2 (MMP2) and its inhibitor, tissue inhibitor of metalloproteinase 2 (TIMP2), and to discuss their relationship with the colorectal neoplasia. Methods: RTPCR technique was used to measure the levels of MMP2 mRNA and TIMP2 mRNA in 12 cases of colon carcinoma and 20 cases of rectal carcinoma (involved 4 recurrent cases). Results: The MMP2 mRNA was expressed in 27/32 tumor tissues and 18/32 normal tissues. For the TIMP2, the rates were 29/32 and 25/32, respectively. The levels of MMP2 mRNA in tumor tissues were negatively correlated with the ages of patients. Eleven cases had the higher level of MMP2 mRNA in normal tissue than in tumor tissue, and the incidence in colon carcinoma was much higher than that in rectal carcinoma (18/32,16/32,P0.05). Conclusion: The biological behaviours of colorectal cancer changed with the ages of patients which may be correlated with the expression levels of the MMP2 in cancer tissue. The biological behaviours of the colon cancer were different from those of rectal cancer.
Prognostic Value of Matrix Metalloproteinases (MMP-2 and MMP-9) in Patients with Lymph Node-Negative Breast Carcinoma

Abstract:Twenty-five to thirty percent of patients with node-negative breast cancer are expected to relapse following surgery, therefore great efforts have been made to identify new prognostic markers that could be useful in defining patients for additional therapy. The expression of MMP-2 and MMP-9 has been associated with high potential of metastasis in several human carcinomas including breast cancer. In the present study we examined the prognostic value of immunoreactive MMP-2/MMP-9 protein in 270 consecutive lymph node negative cases who received radical mastectomy or modified radical mastectomy. Among the patients, 211 cases received adjuvant endocrine therapy and/or adjuvant chemotherapy. Using immunohistochemical assay, we found that 56.7% of the resected tumors were positive for MMP-2 whereas 59.6% of the samples were positive for MMP-9. χ 2 test demonstrated a significant direct association between MMP-2 and MMP-9 ( p < 0.001); positive immunostaining of MMP-2 was significantly related to higher tumor grade ( p < 0.001) and larger tumor size ( p =0.012); positive immunostaining of MMP-9 was significantly related to higher tumor grade ( p =0.002). In univariate analysis, using Cox-proportional hazard model we found MMP-2, MMP-9 and the co-expression of MMPs (MMP2/MMP9) were significantly associated with patients' relapse free survival ( p =0.016, 0.015 and 0.013 respectively) but not overall survival ( p =0.122, 0.320 and 0.091 respectively). Log-rank test also showed that MMP-2, MMP-9 or the co-expression of MMP2/MMP9 was unfavorable prognostic factor for relapse free survival but not overall survival. In subgroup analysis, we found MMPs were more prognostic for patients with no adjuvant treatment than for patients with adjuvant therapy. In multivariate analysis, using Cox-proportional hazard model we found co-expression of MMPs, larger tumor size and higher tumor grade were unfavorable for relapse free survival ( p =0.038, 0.007 and 0.015 for each). We concluded that MMP-2 and MMP-2 are unfavorable prognostic factors in breast cancer patients. They might be potential predictive factor for adjuvant systemic therapy. The co-expression of MMP-2 and MMP-9 has significantly prognostic value in node-negative patients.
PRL-3 and MMP9 Expression and Epithelial-Mesenchymal Transition Markers in Circulating Tumor Cells from Patients with Colorectal Cancer: Potential Value in Clinical Practice.

Abstract:Objective:To evaluate the clinical correlation of epithelial-mesenchymal transition (EMT) with PRL-3 and MMP9 expression in the circulating tumor cells (CTCs) of patients with colorectal cancer (CRC).Materials and Methods:Between January 2016 and December 2018, the EMT phenotype-based subsets of CTCs and the expression levels of PRL-3 and MMP9 in CTCs were identified, and their clinical values in 172 patients were evaluated. The CTCs were isolated, classified, and counted using the CanPatrol™ CTC filtration system. The CTC subsets (epithelial cells, mesenchymal cells and biphenotypic cells), as well as PRL-3 and MMP9 expression, were detected by RNA in situ hybridization.Results:CTCs were detected in 93.0% (160/172) of the included patients with CRC. Positive PRL-3 and MMP9 expression in CTC and M-CTC was found in 75.0% (102/136) and 80.8% (97/120) of the patients, respectively. The proportion of patients with positive PRL-3 and MMP9 expression in M-CTC was significantly associated with distant metastasis (p<0.05). The patients with ≥6 CTCs tended to show poorer progression-free survival (PFS) and overall survival (OS) rates (p=0.016, 0.02, respectively), and the patients with ≥3 M-CTC also showed poor PFS (p=0.0013). Additionally, the patients with positive PRL-3 and MMP9 expression in CTCs had significantly poorer PFS (p=0.0024) and OS (p=0.095) than the patients with negative PRL-3 and MMP9 expression. Multivariate Cox analysis uncovered that positive PRL-3 and MMP9 expression in CTCs may be an independent prognostic factor for worse PFS.Conclusion:EMT phenotypes and CTC numbers can be used as prognostic indicators for metastasis and survival in patients with CRC, and the combination of PRL-3 and MMP9 expression in CTCs is a promising clinical marker for patients with CRC.

Abstract:Background While previous studies have indicated variability in distant metastatic potential among different mismatch repair (MMR) states in colorectal cancer (CRC), their findings remain inconclusive, especially considering potential differences across various ethnic backgrounds. Furthermore, the gene regulatory networks and the underlying mechanisms responsible for these variances in metastatic potential across MMR states have yet to be elucidated. Methods We collected 2058 consecutive primary CRC samples from the South West of China and assessed the expression of MMR proteins (MLH1, MSH2, MSH6, and PMS2) using immunohistochemistry. To explore the inconsistencies between different MMR statuses and recurrence, we performed a meta‐analysis. To delve deeper, we employed Weighted Gene Co‐expression Network Analysis (WGCNA), ClueGo, and iRegulon, pinpointing gene expression networks and key regulatory molecules linked to metastasis and recurrence in CRC. Lastly, both univariate and multivariate Cox regression analyses were applied to determine the impact of core regulatory molecules on metastasis. Results Of the samples, 8.2% displayed deficient MMR (dMMR), with losses of MLH1 and PSM2 observed in 40.8% and 63.9%, respectively. A unique 24.3% isolated loss of PMS2 without concurrent metastasis was identified, a result that diverges from established literature. Additionally, our meta‐analysis further solidifies the reduced recurrence likelihood in dMMR CRC samples compared to proficient MMR (pMMR). Two gene expression networks tied to distant metastasis and recurrence were identified, with a majority of metastasis‐related genes located on chromosomes 8 and 18. An IRF1 positive feedback loop was discerned in the metastasis‐related network, and IRF1 was identified as a predictive marker for both recurrence‐free and distant metastasis‐free survival across multiple datasets. Conclusion Geographical and ethnic factors might influence peculiarities in MMR protein loss. Our findings also highlight new gene expression networks and crucial regulatory molecules in CRC metastasis, enhancing our comprehension of the mechanisms driving distant metastasis.

Abstract:PURPOSE: Matrix metalloproteinase (MMP)-2 and MMP-9 are considered to play an important role in the metastasis of malignant tumors. Membrane type 1-MMP (MT1-MMP) and tissue inhibitor of metalloproteinase 2 (TIMP-2) are essential factors for the activation of pro-MMP-2. There are some reports about expressions of MMP family in relationship to clinical features of head and neck squamous cell carcinoma (SCC), but the results were not uniform and the prognostic value of their expressions remains unclear.EXPERIMENTAL DESIGN: The study group consisted of 53 Japanese patients with oral SCC of early stage (T(1-2)N(0)M(0)). Expressions of MMP-2, MMP-9, MT1-MMP, and TIMP-2 were examined using immunohistological methods on the sections of tumor biopsy samples. The intensity of MMP expression was categorized into four grades (score 0-3) by semiquantitative analysis using a computer with NIH image, and correlation between this grade and clinical aspects such as tumor recurrence, metastasis, and prognosis were examined.RESULTS: The expression score of MMP-2 correlated with that of MMP-9 (r = 0.291; P = 0.036), MT1-MMP (r = 0.286; P = 0.039), and TIMP-2 (r = 0.257; P = 0.050). Patients who developed regional lymph node and/or distant metastasis showed significantly higher scores in the expressions of MMP-9 and TIMP-2 than patients without any tumor metastases (P = 0.036 and P = 0.043, respectively). Kaplan-Meier analyses as well as univariate analyses using the Cox proportional hazards model showed that expression of MMP-9 (P = 0.0143 and P = 0.0418, respectively) and marked expression of TIMP-2 (P < 0.0001 and P = 0.0004, respectively) correlated with worse-cause-specific survival. Multivariate analysis confirmed that marked expression of TIMP-2 was the only independent factor for cause-specific death (hazard ratio, 7.543; confidence interval, 1.693-33.610; P = 0.0080).CONCLUSIONS: Expressions of MMP-9 and TIMP-2 have predictive value for tumor metastases and cause-specific survival. High expression of TIMP-2 is the most independent factor for worse prognosis in early-stage oral SCC.

Abstract:BACKGROUND: Matrix metalloproteinase-2 (MMP-2) is well known for its critical role in cell survival and cancer development. It also plays an important role in hematopoietic recovery after chemotherapy-induced myelosuppression. In this study, the authors investigated the association of MMP-2 polymorphisms with treatment efficacy and the occurrence of severe toxicity in patients with nonsmall cell lung cancer (NSCLC) who were receiving first-line, platinum-based chemotherapy. METHODS: A pharmacogenetic association study was performed in 663 Chinese patients who had inoperable stage III/IV NSCLC and were receiving first-line, platinum-based regimens. Information about objective response, progression-free survival, overall survival, grade 3 or 4 gastrointestinal toxicity (nausea/vomiting), and hematologic toxicity (neutropenia, anemia, thrombocytopenia) was available. Sixteen tag single nucleotide polymorphisms (SNPs) of MMP-2 were assessed. RESULTS: In 7 polymorphisms, significant associations were observed with the incidence of grade 3 or 4 neutropenia. The variant homozygotes of reference SNP rs12934241 exhibited the most significant effect on the risk of neutropenia, leading to an incidence rate that increased from 12.3% (for the C/C genotype) to 50% (for the T/T genotype; odds ratio, 8.33; P = 8.8 x 10(-5)). Stratified analyses indicated that rs12934241 exhibited a much stronger influence in the cisplatin-gemcitabine regimen subgroup than subgroups that received other regimens (Pinteraction = .003). Further haplotype analyses produced results that were consistent with results from single-SNP analyses. However, no significant association was observed between MMP-2 polymorphisms and treatment efficacy, including response rate, clinical benefit, progression-free survival, and overall survival. CONCLUSIONS: To the authors' knowledge, this study provides the first evidence for a predictive role of MMP-2 polymorphisms in the variability of severe chemotherapy-related neutropenia among Chinese patients with platinum-treated, advanced NSCLC.
