Abstract:The Cucurbit[n]uril (CB[n]) structure, with two electrostatically negative portals, facilitates ion‐dipole interactions with cationic species, demonstrating excellent binding affinities. A cyclic voltammetry (CV) study revealed that the presence of CB[n] with free heavy metal‐ions (HM(II)) ions in solution causes a slight shift in peak potentials and a decrease in the amount of free heavy metal ions, supporting host‐guest interactions. One of the major concerns worldwide is the rapid increase in water pollution with hazardous heavy metals. The Cucurbit[n]uril (CB[n]) structure provides an inner hydrophobic cavity accessible through two electrostatically negative portals with polar carbonyl groups, allowing for charged ion–dipole interaction between cationic species with excellent binding affinities. In this work we selected CB[5], and CB[7] for the titrimetric and electrochemical evaluation of the heavy metal complexes formed with Copper(II), Cadmium(II) and Lead(II). The host–guest interaction was found by either attenuation of the redox peaks, the formation of a new peak, or the slight shift in the redox peaks of the heavy metal ions. Titration data were obtained for CB[n] with the heavy metal solutions to determine the molar coefficient of binding. The stoichiometry of the complexes was 1 : 1 by Job′s plot. We calculated the binding constants using the Benesi–Hildebrand method achieving relatively good binding constants (K) ranging from (1.80±0.14)×105–(4.98±0.13)×105 M−1. The results showed that the complexes of Cadmium(II) and Lead(II) with both CB[5] and CB[7] are highly stable in aqueous media. Copper(II) was able to bind to the smaller sized portal of CB[5], whereas with CB[7] the heavy metal ion could not form a favourable complex. This phenomenon suggests that the CB[5] is an ideal host at forming stable host–guest interactions than their larger homologue.

Barriers for Extrusion of a Guest from the Interior Binding Cavity of a Host: Gas Phase Experimental and Computational Results for Ion-Capped Decamethylcucurbit[5]uril Complexes

Abstract:Factors affecting the extrusion of guests from metal ion-capped decamethylcucurbit[5]uril (mc5) molecular container complexes are investigated using both collision-induced dissociation techniques and molecular mechanics simulations. For guests without polar bonds, the extrusion barrier increases with increasing guest volume. This is likely because escape of larger guests requires more displacement of the metal ion caps and, thus, more disruption of the ion-dipole interactions between the ion caps and the electronegative rim oxygens of mc5. However, guests larger than the optimum size for encapsulation displace the ion caps prior to collision-induced dissociation, resulting in less stable complexes and lower dissociation thresholds. The extrusion barriers obtained for guests with polar bonds are smaller than those obtained for similarly sized guests without polar bonds. This is likely because the partial charges on the guest allow electrostatic interactions with the ion cap and rim oxygens of mc5 during extrusion, thus stabilizing the extrusion transition state and reducing the extrusion barrier. Results from this study demonstrate simple principles to consider for designing host-guest complexes with specific guest-loss behaviors. Similar trends are observed between the experimental and computational results, demonstrating that molecular mechanics simulations can be used to approximate the relative stability of mc5 molecular container complexes and likely those of other similar complexes.

Abstract:Atomic-level understanding of the dynamic feature of host–guest interactions remains a central challenge in supramolecular chemistry. The remarkable guest binding behavior of the Cucurbiturils family of supramolecular containers makes them promising drug carriers. Among Cucurbit[n]urils, Cucurbit[8]uril (CB8) has an intermediate portal size and cavity volume. It can exploit almost all host–guest recognition motifs formed by this host family. In our previous work, an extensive computational investigation of the binding of seven commonly abused and structurally diverse drugs to the CB8 host was performed, and a general dynamic binding picture of CB8-guest interactions was obtained. Further, two widely used fixed-charge models for drug-like molecules were investigated and compared in great detail, aiming at providing guidelines in choosing an appropriate charge scheme in host-guest modelling. Iterative refitting of atomic charges leads to improved binding thermodynamics and the best root-mean-squared deviation from the experimental reference is 2.6 kcal/mol. In this work, we focus on a thorough evaluation of the remaining parts of classical force fields, i.e., the bonded interactions. The widely used general Amber force fields are assessed and refitted with generalized force-matching to improve the intra-molecular conformational preference, and thus the description of inter-molecular host–guest interactions. The interaction pattern and binding thermodynamics show a significant dependence on the modelling parameters. The refitted system-specific parameter set improves the consistency of the modelling results and the experimental reference significantly. Finally, combining the previous charge-scheme comparison and the current force-field refitting, we provide general guidelines for the theoretical modelling of host–guest binding.

Abstract:Cucurbit[8]uril (CB[8]) heteroternary complexes display certain characteristics making them well-suited for molecular level adhesives. In particular, careful choice of host-guest binding pairs enables specific, fully reversible adhesion. Understanding the effect of the environment is also critical when developing new molecular level adhesives. Here we explore the binding forces involved in the methyl viologen·CB[8]·naphthol heteroternary complex using single-molecule force spectroscopy (SMFS) under a variety of conditions. From SMFS, the interaction of a single ternary complex was found to be in the region of 140 pN. Additionally, a number of surface interactions could be readily differentiated using the SMFS technique allowing for a deeper understanding of the dynamic heteroternary CB[8] system on the single-molecule scale.
Rapid Estimation of Binding Constants for Cucurbit[8]uril Ternary Complexes Using Electrochemistry

Abstract:Supramolecular complexes are of fundamental interests in biomedicines and adaptive materials, and thus facile methods to determine their binding affinity show usefulness in the design of novel drugs and materials. Herein, we report a novel approach to estimate the binding constants KG2 of cucurbit[8]uril-methyl viologen-based ternary complexes (CB8-MV2+-G2) using electrochemistry, achieving high precision (±0.03) and practical accuracy (±0.32) in logKG2 and short measurement time (<10 min). In particular, we have uncovered a linear correlation (R2 > 0.8) between the reduction potential of CB8-MV2+-G2 ternary complexes and their reported binding constants from isothermal titration calorimetry, which allow a calibration curve to be plotted based on 25 sample complexes. Mechanistic investigation using experimental and computational approaches reveals that this correlation stems from the dynamic host-guest exchange events occurring after the electron transfer step. Binding constants of unknown ternary complexes, where G2 = hydrocarbons, were estimated, illustrating potential applications for sparsely soluble second guests.

Abstract:A strong interaction called cation-pi bonding, which we named because it occurs between aromatics and divalent metal cations, has been successfully differentiated from the normal cation-pi intermolecular interactions. Our findings were based on the B3LYP/6-311++G(d,p) calculations and Morokuma decomposition analyses on the complexes formed by substituted benzenes with alkaline metal and alkaline earth metal ions. In comparison with the common cation-pi intermolecular interaction, the cation-pi bond in the complexes of either Be2+ or Mg2+ with the aromatics has its own characteristics: (a) short bond lengths, (b) very strong binding strength, (c) significant nonelectrostatic interaction that constitutes more than 50% of the total binding strength, (d) obvious cation-pi orbital interaction, and (e) special orbital interaction pattern that only the pi orbitals of the aromatics interact with the s, p(x), and p(y) atomic orbitals of metal cations for forming bonding MOs. While the electrostatic interaction is significantly affected by the nature of the substituents attached to the benzene, the nonelectrostatic interaction and orbital interaction are not. Furthermore, the total binding strength and electrostatic interaction are well correlated with the Hammett electronic parameters. This structural and thermochemical information is highly useful in identifying cation-pi bonds. Moreover, they are equally helpful for modifying current force fields in reproducing this unusual chemical bond that is commonly encountered in both chemical and biological systems.

Abstract:Cucurbit[8]uril-mediated complexes are widely investigated due to their high binding affinity and diverse binding modes. However, the dynamic variations observed among different binding modes have yet to be fully understood, and current instrumental analyses impose temporal and spatial restrictions on the elucidation of instantaneous intra-complex motions. In this work, we performed molecular dynamics (MD) simulations and quantum chemical calculations on a CB[8]-diarylviologen complex system. Our simulations successfully monitored instantaneous intra-complex motions across multiple binding species using solvent accessible surface area (SASA) as the descriptor, which for the very first time provided a direct proof of the pseudostatic behavior. Additionally, we found that enhanced interaction cooperation resulted in the 2:2 complex being more energy-favorable than the 1:1 counterpart despite the same stoichiometry. These findings offer insights into the variable dynamics of host–guest complexes and the crucial role of interaction cooperation in controlling binding pathways, shedding a light on engineering supramolecular systems with greater precision.

Abstract:Cucurbiturils (CBs), the pumpkin-shaped macrocycles, are suitable hosts for an array of neutral and cationic species. A plethora of host-guest complexes between CBs and a variety of guest molecules has been studied. However, much remains unknown, even in the complexation of very simple guests such as metal cations. In the computational study herein, DFT molecular modeling has been employed to investigate the interactions of a series of trivalent metal cations (Al3+, Ga3+, In3+, La3+, Lu3+) to cucurbit[n]urils and to evaluate the main factors controlling the host-guest complexation. The thermodynamic descriptors (Gibbs energies in the gas phase and in a water environment) of the corresponding complexation reactions have been estimated. This research is a logical continuation of an earlier study on the interaction between CB[n]s and a series of biologically essential mono- and divalent metal cations (Na+/K+ and Mg2+/Ca2+, respectively).
An Ab Initio Investigation of the Structure and Alkaline Earth Divalent Cation Selectivity of 18-Crown-6

Abstract:We present an ab initio, quantum mechanical study of 18-crown-6 (18c6) and its interaction with the alkaline earth dications Mg2+, Ca2+, Sr2+, Ba2+, and Ru2+. Geometries, binding energies, and binding enthalpies are evaluated at the restricted Hartree−Fock (RHF) and second-order Møller−Plesset perturbation (MP2) levels of theory using the 6-31+G* basis set and relativistic effective core potentials. The affinity of 18c6 for the dications is considerable, ranging from 150−300 kcal mol-1. The cation−18c6 interaction arises principally from the electrostatic interaction of the dication with the nucleophilic ether backbone and from the polarization of the crown ether by the electric field of the dication. Whereas Ba2+ selectivity is observed for 18c6 in aqueous environments, our calculations clearly show that the crown ether in fact binds Mg2+ most strongly in gas phase. Thus, solvation effects appear to strongly influence cation selectivity. Indeed, Ba2+ selectivity is recovered when we consider the competition of the solvent and 18c6 molecules for the dications using a simple cation exchange reaction.

Abstract:Electrochemical properties of cationic complexes of supramolecular cavitands from the cucurbit[n]uril (CBn s ) family are studied. Based on the analysis of a complex of systematic data on these compounds, basic regularities characterizing their adsorption behavior are formulated, in particular, the extremely high surface activity and unusually wide adsorption potential region over which the adsorption layer structure is transformed upon changes of the electrode charge value and sign. The varying of concentrations of reactants involved in the complex-formation reaction (the cavitand and inorganic cation) in the studied systems is shown to change significantly the observed picture of adsorption phenomena. The comparative analysis of adsorption data for systems containing supramolecular complexes of cucurbiturils with different cations can back qualitative estimating of their strength constants. It has been established by example of systems containing inclusive complexes of the СВ7 cavitand with organic cation (the cation of 3,3′-diethylthiocarbocyanine iodide dye) and with organic molecules (super-tough complexes with 1-hydroxyadamantan and ferrocene, the strength constants K of 10 9 –10 10 М –1 by order of magnitude), that including of neutral organic molecule into the cavitand cavity does not prevent the formation of an exclusive complex with sodium cation and that such a complex is not formed in the presence of organic cation in the cavitand cavity. It was demonstrated by example of the cobaltocene(Сос)/cobaltocenium(Сос + ) redox-system studied in the solutions added with СВ7 and СВ7-free ones that the formation of adsorption layer of the Сос + ⊂ СВ7 supramolecular complex particles preceding an electrode process leads to changing of the electrode reaction rate and mechanism.

Abstract:The ion size matters: the structures of corannulene monoanions crystallized with Cs(+) and Rb(+) ions in the presence of [18]crown-6 reveal the intrinsic binding preferences of alkali metals and allow evaluation of the bowl deformation caused by negative charge distribution and metal binding. The large cesium cation coordinates exclusively to the concave face of C(20) H(10)(-), whereas the smaller rubidium cation exhibits convex binding.

Abstract:In spite of the biological importance of the binding of Zn2+, Ca2+, and Mg2+ to carboxylate anions, previous experimental and computational studies have reached conflicting conclusions regarding the corresponding binding affinities. Here, we report the first use of Raman multivariate curve resolution (Raman-MCR) vibrational spectroscopy to obtain self-consistent free and bound metal acetate spectra and one-to-one binding constants, without the need to invoke any a priori assumptions regarding the shapes of the corresponding vibrational bands. The experimental results, combined with classical molecular dynamics simulations with a force field effectively accounting for electronic polarization via charge scaling and ab initio simulations, indicate that the measured binding constants pertain to direct (as opposed to water separated) ion pairing. The resulting binding constants do not scale with cation size, as the binding constant to Zn2+ is significantly larger than that to either Mg2+ or Ca2+, although Zn2+ and Mg2+ have similar radii that are about 25% smaller than Ca2+. Remaining uncertainties in the metal acetate binding free energies are linked to fundamental ambiguities associated with identifying the range of structures pertaining non-covalently bound species.
Noncovalent Interactions in Supramolecular Complexes: A Study on Corannulene and the Double Concave Buckycatcher

Abstract:Stimulated by the recent observation of {\pi}-{\pi} interactions between C60 and corannulene subunits in a molecular tweezer arrangement (J. Am. Chem. Soc. 2007, 129, 3842), a density functional theory study was performed to analyze the electronic structure and properties of various noncovalent corannulene complexes. The theoretical approach is first applied to corannulene complexes with a series of benchmark molecules (CH4, NH3, and H2O) using several new-generation density functionals. The performance of nine density functionals, illustrated by computing binding energies of the corannulene complexes, demonstrates that Zhao and Truhlar's MPWB1K and M05-2X functionals provide energies similar to that obtained at the SCS-MP2 level. In contrast, most of the other popular density functionals fail to describe this noncovalent interaction or yield purely repulsive interactions. Further investigations with the M05-2X functional show that the binding energy of C60 with corannulene subunits in the relaxed molecular receptor clip geometry is -20.67 kcal/mol. The results of this calculation further support the experimental interpretation of pure {\pi}-{\pi} interactions between a convex fullerene and the concave surfaces of two corannulene subunits.
Salt Solutions of Molecular Containers of Cucurbiturils: Relationship Between the Characteristics of Cationic Complexes and Adsorbate Layers with Their Participation

Abstract:An analysis of the impedance data for the Hg-electrode/salt solutions interface of molecular containers of cucurbiturils containing their supramolecular cationic complexes was carried out. The need to study the properties of such systems is determined by their practical use in the field of supramolecular chemistry, medicine, biochemistry, and nanotechnology. The formation of two types of adsorption layers and a significant change in the adsorption behavior with a change in the initial concentrations of the system components and the electrode potential were established. It was shown that the observed changes are due to a change in the concentration of free molecules of cucurbiturils not involved in complex formation. The high sensitivity of the impedance measurement data to this parameter found this way made it possible to establish the corresponding quantitative dependences for systems with known values of the strength constants for complexes of inorganic cations with cucurbiturils. It was shown that for systems close in nature, this opens up the possibility of finding previously unknown strength constants of cationic cucurbiturils complexes when analyzing impedance measurement data. The work showed the applicability of the proposed method for determining the strength constants of supramolecular cationic cucurbiturils complexes, which can be an alternative to more complex and time-consuming methods.

Abstract:PM3 and B3LYP/3-21g* calculations were performed on cu-curbit[7] uril and its complex with protonated 2, 6-bis(4,5dihydro-1H-imidazol-2yl)naphthalene. The results agreed well with the experimental observations. It indicated that in addition to van der Weals interaction and hydrophobic effect, hydrogen bonding is also an important driving force for the molecular recognition of cucurbiturils.

Abstract:Cucurbiturils are particularly interesting to chemists, because these macrocyclic molecules are suitable hosts for an array of neutral and cationic species. It is believed that the host-guest binding originated from hydrophobic interactions and ion-dipole interactions in the case of cationic guests. The fact that an elementary unit of cucurbiturils consists of two fused imidazole rings, which ionize readily, has remained largely unnoticed up to now. This work reports ionized cucurbiturils and their binding to C60 fullerene using versatile electronic-precision description. The methodology is based on density functional theory. We assert that cationization of cucurbiturils fosters C60-cucurbituril binding due to polarization of electron density in C60. Therefore, more stable host-guest complexes can be derived.

Cation−π Interactions: A Theoretical Investigation of the Interaction of Metallic and Organic Cations with Alkenes, Arenes, and Heteroarenes

Abstract:The nature of the cation−π interaction has been examined by carrying out high level ab initio calculations of both metallic (Li+,Na+,K+, and Ag+) and organic (NH4+, C(NH2)3+, and N(CH3)4+) cations with different classes of π systems, viz. alkenes (ethene), arenes (benzene), and heteroarenes (pyrrole). The calculations, which include a rigorous decomposition of the interaction energies, indicate that the interaction of these π systems with the metal cations is characterized by contributions from both electrostatic and induction energies, with the contribution of the latter being dominant. Though the contributions of dispersion energies are negligible in the cation−π complexes involving Li+ and Na+, they assume significant proportions in the complexes involving K+ and Ag+. In the π complexes of the organic cations, the repulsive exchange contributions are much larger than the attractive electrostatic contributions in the π complexes of organic cations, and thus, the contributions of both induction and dispersion energies are important. While inclusion of electron correlation is essential in obtaining accurate estimates of the dispersion energy, it also magnifies the contribution of the induction energies in the π complexes of the organic cations. This results in significant consequences in the evaluation of geometries and energies of these cation−π complexes. The major difference between the cation−π and cation−H2O complexes stems from the differences in the relative contributions of electrostatic and induction energies, a foreknowledge of which is vital in the design of ion-selective ionophores and receptors. The blue shift in the highly IR active out-of-plane CH bending mode of the π systems in these complexes is representative of the strength of the cation−π interaction.

Abstract:With the emergence of host-guest systems, a novel branch of complexation chemistry has found wide application in industries such as food, pharmacy, medicine, environmental protection and cosmetics. Along with the extensively studied cyclodextrins and calixarenes, the innovative cucurbiturils (CB) have enjoyed increased popularity among the scientific community as they possess even better qualities as cavitands as compared to the former molecules. Moreover, their complexation abilities could further be enhanced with the assistance of metal cations, which can interestingly exert a dual effect on the complexation process: either by competitively binding to the host entity or cooperatively associating with the CB@guest structures. In our previous work, two metal species (Mg2+ and Ga3+) have been found to bind to CB molecules in the strongest fashion upon the formation of host-guest complexes. The current study focuses on their role in the complex formation with three dye molecules: thiazole orange, neutral red, and thioflavin T. Various key factors influencing the process have been recognized, such as pH and the dielectric constant of the medium, the cavity size of the host, Mn+ charge, and the presence/absence of hydration shell around the metal cation. A well-calibrated DFT methodology, solidly based and validated and presented in the literature experimental data, is applied. The obtained results shed new light on several aspects of the cucurbituril complexation chemistry.

Abstract:The cation−π interaction and the hydrophobic effect are important intermolecular forces in chemistry and play major roles in controlling recognition in biological systems. We compared their relative contributions to the binding of molecular "dumbbell" guests in synthetic container hosts in water. The guests offered direct, intramolecular competition between trimethylammonium groups, −N+(CH3)3, and tert-butyl groups, −C(CH3)3, for the internal surfaces (aromatic panels) of the containers. In contrast with previous studies, the container molecules consistently preferred binding to the uncharged tert-butyl groups, regardless of the presence of anionic, cationic, or zwitterionic groups on the container peripheries. This preference is determined by solvation of the polar trimethylammonium group in water, which outcompetes the attraction between the positive charge and the π-surfaces in the container. The synthetic container complexes provide a direct measure of the relative strengths of cation−π interactions and desolvation in water. Interactions with the uncharged tert-butyl group are more than 12 kJ mol–1 more favorable than the cation−π interactions with the trimethylammonium group in these cavitand complexes.This article has not yet been cited by other publications.
