Abstract:Highlights • RORγt as a key transcription factor controls Th17 cell development. • Modulation of RORγt activity alleviates inflammatory and autoimmune diseases. • Several small-molecule RORγt inhibitors have been recently developed. • Some of RORγt inhibitors enter clinical evaluations. As a ligand-dependent transcription factor, retinoid-associated orphan receptor γt (RORγt) that controls T helper (Th) 17 cell differentiation and interleukin (IL)-17 expression plays a critical role in the progression of several inflammatory and autoimmune conditions. An emerging novel approach to the therapy of these diseases thus involves controlling the transcriptional capacity of RORγt to decrease Th17 cell development and IL-17 production. Several RORγt inhibitors including both antagonists and inverse agonists have been discovered to regulate the transcriptional activity of RORγt by binding to orthosteric- or allosteric-binding sites in the ligand-binding domain (LBD). Some of small-molecule inhibitors have entered clinical evaluations. Therefore, in current review, the role of RORγt in Th17 regulation and Th17-related inflammatory and autoimmune diseases was highlighted. Notably, the recently developed RORγt inhibitors were summarized, with an emphasis on their optimization from lead compounds, efficacy, toxicity, mechanisms of action, and clinical trials. The limitations of current development in this area were also discussed to facilitate future research. Small-molecule RORγt Antagonists Inhibit T Helper 17 Cell Transcriptional Network by Divergent Mechanisms.

Abstract:We identified three retinoid-related orphan receptor gamma t (RORγt)-specific inhibitors that suppress T helper 17 (Th17) cell responses, including Th17-cell-mediated autoimmune disease. We systemically characterized RORγt binding in the presence and absence of drugs with corresponding whole-genome transcriptome sequencing. RORγt acts as a direct activator of Th17 cell signature genes and a direct repressor of signature genes from other T cell lineages; its strongest transcriptional effects are on cis-regulatory sites containing the RORα binding motif. RORγt is central in a densely interconnected regulatory network that shapes the balance of T cell differentiation. Here, the three inhibitors modulated the RORγt-dependent transcriptional network to varying extents and through distinct mechanisms. Whereas one inhibitor displaced RORγt from its target loci, the other two inhibitors affected transcription predominantly without removing DNA binding. Our work illustrates the power of a system-scale analysis of transcriptional regulation to characterize potential therapeutic compounds that inhibit pathogenic Th17 cells and suppress autoimmunity.
The Role of Particle Morphology in Interfacial Energy Transfer in CdSe/CdS Heterostructure Nanocrystals

Abstract:An Upside of Asymmetry Advances in synthetic techniques have enabled the preparation of nanometer-scale semiconductors in a wide range of precise shapes and sizes, including core-shell morphologies that layer several different materials in the same particle. Such two-in-one motifs are promising for light-harvesting applications because they allow optically induced charge separation across the internal interface. Borys et al. (p. 1371) studied a series of rod-shaped cadmium sulfide–cadmium selenide hybrid particles using single-particle–resolved optical spectroscopy and found that smooth versus bulbous geometries produced distinct emission spectra. Further analysis of more complex, tetrapodal particles (with four arms aligned tetrahedrally) suggested that nonuniform geometries facilitate interfacial charge transfer by reducing the likelihood of electronic band misalignment. Single-particle spectroscopy suggests that non-uniform geometries favor efficient charge separation for light harvesting. Nanoscale semiconductor heterostructures such as tetrapods can be used to mimic light-harvesting processes. We used single-particle light-harvesting action spectroscopy to probe the impact of particle morphology on energy transfer and carrier relaxation across a heterojunction. The generic form of an action spectrum [in our experiments, photoluminescence excitation (PLE) under absorption in CdS and emission from CdSe in nanocrystal tetrapods, rods, and spheres] was controlled by the physical shape and resulting morphological variation in the quantum confinement parameters of the nanoparticle. A correlation between single-particle PLE and physical shape as determined by scanning electron microscopy was demonstrated. Such an analysis links local structural non-uniformities such as CdS bulbs forming around the CdSe core in CdSe/CdS nanorods to a lower probability of manifesting excitation energy–dependent emission spectra, which in turn is probably related to band alignment and electron delocalization at the heterojunction interface.

Abstract:Abstract Th17 cells, a lymphocyte subpopulation that is characterized by the expression of the transcription factor “retinoic acid receptor-related orphan receptor gamma-t” (RORγt), plays an important role in the pathogenesis of autoimmune disease. The current study was set up to discover novel and non-steroidal small-molecule inverse agonists of RORγt and to determine their effects on autoimmune disease. Structure-based virtual screening (SBVS) was used to find compounds targeting RORγt. Flow cytometry was used to detect the Th17 cell differentiation. Inverse agonists were intraperitoneally administered to mice undergoing experimental autoimmune uveitis (EAU), experimental autoimmune encephalomyelitis (EAE) or type 1 diabetes. The effects of the inverse agonists were evaluated by clinical or histopathological scoring. Among 1.3 million compounds screened, CQMU151 and CQMU152 were found to inhibit Th17 cell differentiation without affecting the differentiation of Th1 and Treg lineages (both P = 0.001). These compounds also reduced the severity of EAU ( P = 0.01 and 0.013) and functional studies showed that they reduced the number of Th17 cell and the expression of IL-17(Th17), but not IFN-γ(Th1) and TGF-β(Treg) in mouse retinas. Further studies showed that these compounds may reduce the expression of p-STAT3 by reducing the positive feedback loop of IL-17/IL-6/STAT3. These compounds also reduced the impaired blood–retinal barrier function by upregulating the expression of tight junction proteins. These compounds were also found to reduce the severity of EAE and type 1 diabetes. Our results showed that RORγt inverse agonists may inhibit the development of autoimmune diseases and may provide new clues for the treatment of Th17-mediated immune diseases.

Abstract:The nuclear hormone receptor retinoic acid receptor-related-orphan-receptor-gamma t (RORγt) is the key transcription factor required for Th17 cell differentiation and for production of IL-17 family cytokines by innate and adaptive immune cells. Dysregulated Th17 immune responses have been associated with the pathogenesis of several inflammatory and autoimmune diseases such as psoriasis, psoriatic arthritis, and ankylosing spondylitis. In this article, we describe the in vitro pharmacology of a potent and selective low molecular weight RORγt inhibitor identified after a structure-based hit-to-lead optimization effort. The compound interfered with co-activator binding to the RORγt ligand binding domain and impaired the transcriptional activity of RORγt as evidenced by blocked IL-17A secretion and RORE-mediated transactivation of a luciferase reporter gene. The inhibitor effectively reduced IL-17A production by human naive and memory T-cells and attenuated transcription of pro-inflammatory Th17 signature genes, such as IL17F, IL22, IL26, IL23R, and CCR6. The compound selectively suppressed the Th17/IL-17 pathway and did not interfere with polarization of other T helper cell lineages. Furthermore, the inhibitor was selective for RORγt and did not modify the transcriptional activity of the closely related family members RORα and RORβ. Using human keratinocytes cultured with supernatants from compound treated Th17 cells we showed that pharmacological inhibition of RORγt translated to suppressed IL-17-regulated gene expression in keratinocyte cell cultures. Furthermore, in ex vivo immersion skin cultures our RORγt inhibitor suppressed IL-17A production by Th17-skewed skin resident cells which correlated with reduced human β defensin 2 expression in the skin. Our data suggests that inhibiting RORγt transcriptional activity by a low molecular weight inhibitor may hold utility for the treatment of Th17/IL-17-mediated skin pathologies.

Abstract:Multiple sclerosis, inflammatory bowel disease and organ transplant rejection are related to Th17 cell development and inflammatory respond. RORγt, a specific transcription factor regulating Th17 cell differentiation, is a pivotal target for the treatment of diseases. However, the clinical application of RORγt inverse agonists reported so far has been hindered due to limited efficacy and toxic side effects. Plant-derived natural products with drug-like properties and safety are wide and valuable resources for candidate drug discovery. Herein, structure-based virtual screening was used to find out 2′,4′-Dihydroxy-2,3-dimethoxychalcone (DDC), a chalcone derivative rich in plants and food, located in the binding pocket of RORγt and targeted to inhibit RORγt activity. DDC repressed murine Th17 differentiation and promoted Treg differentiation remarkably in a dose-dependent manner. In addition, DDC treatment improved experimental autoimmune encephalomyelitis recovery, ameliorated experimental colitis severity, and prevented graft rejection significantly. Mechanically, DDC indirectly stabilized Foxp3 expression by inhibiting RORγt activity and the expression of its target gene profile in vitro and in vivo, which realized its regulation of Th17/Treg balance. In conclusion, our study provides a scientific basis that DDC, as an inverse agonist of RORγt with simple structure, rich sources, low cost, high efficiency, and low toxicity, has great potential for the development of a novel effective immunomodulator for the treatment of Th17-mediated inflammatory diseases.

Abstract:Retinoid-related orphan receptor γt (RORγt), a vital transcription factor for the differentiation of the pro-inflammatory Th17 cells, is essential to the inflammatory response and pathological process mediated by Th17 cells. Pharmacological inhibition of the nuclear receptor RORγt provides novel immunomodulators for treating Th17-driven autoimmune diseases and organ transplant rejection. Here, we identified 2,2′,4′-trihydroxychalcone (TDC), a natural chalcone derivant, binds directly to the ligand binding domain (LBD) of RORγt and inhibited its transcriptional activation activity. Using three mice models of Th17-related diseases, it was found that the administration of TDC effectively alleviated the disease development of experimental autoimmune encephalomyelitis (EAE), experimental colitis, and skin allograft rejection. Collectively, these results demonstrated TDC targeting RORγt to suppress Th17 cell polarization, as well as its activity, thus, indicating the potential of this compound in treating of Th17-related autoimmune disorders and organ transplant rejection disorders.

Abstract:Significance Because of its essential role in driving IL-17 production, the orphan nuclear receptor RAR-related orphan receptor gamma t (RORγt) represents a potential therapeutic target for autoimmune diseases. Here, we present evidence that 7β, 27-dihydroxycholesterol (7β, 27-OHC) and 7α, 27-OHC are RORγt agonist ligands and may serve as potential endogenous RORγt ligands in promoting the differentiation of mouse and human CD4 + Th17 cells. These findings may not only facilitate rational design of RORγt modulators but also, provide insights into additional targets for inhibiting IL-17 production.

Abstract:In a screen for small-molecule inhibitors of retinoid acid-related orphan receptor gamma (ROR gamma), we fortuitously discovered that a class of aryl amide compounds behaved as functional activators of the interleukin 17 (IL-17) reporter in Jurkat cells. Three of these compounds were selected for further analysis and found to activate the IL-17 reporter with potencies of similar to 0.1 mu M measured by EC50. These compounds were shown to directly bind to ROR gamma by circular dichroism-based thermal stability experiments. Furthermore, they can enhance an in vitro Th17 differentiation process in human primary T cells. As ROR gamma remains an orphan nuclear receptor, discovery of these aryl amide compounds as functional agonists will now provide pharmacological tools for us to dissect functions of ROR gamma and facilitate drug discovery efforts for immune-modulating therapies.

Abstract:Th17 cells are potent mediators in autoimmune diseases, and RORgt is required for their development. Recent studies have shown that ROR gamma t(+) Treg cells in the gut regulate intestinal inflammation by inhibiting effector T cell function. In the current study, we report that ROR gamma t(+) Treg cells were also found in lymph nodes following immunization. Not only distinct from intestinal ROR gamma t(+) Treg cells in their transcriptomes, peripheral ROR gamma t(+) Treg cells were derived from Foxp3(+) thymic Treg cells in an antigen-specific manner. Development of these ROR gamma t(+) Treg cells, coined T regulatory 17 (Tr17) cells, depended on IL-6/Stat3 signaling. Tr17 cells showed suppressive activity against antigen-specific effector T cells in vitro. In addition, Tr17 cells efficiently inhibited myelin-specific Th17-cell-mediated CNS auto-inflammation in a passive EAE model. Collectively, our study demonstrates that Tr17 cells are effector Treg cells that potentially restrict autoimmunity.
Reversing SKI–SMAD4-mediated Suppression is Essential for T H 17 Cell Differentiation

Abstract:TGFβ signalling regulates T helper 17 (TH17) cell differentiation by reversing SKI–SMAD4-mediated suppression of RORγt, revealing a potential therapeutic target for treating TH17-related diseases. Yisong Wan and colleagues describe a novel mechanism by which transforming growth factor beta (TGFβ) regulates differentiation of T helper 17 (TH17) cells. TGFβ and interleukin-6 (IL-6) are required for induction of the master transcription factor RORγt, but too much TGFβ is inhibitory. In this study the authors show that SMAD4—a key signal transducer activated by TGFβ receptor ligation—forms a complex with the transcriptional repressor SKI, and together they suppress transactivation of RORγt and TH17 cell differentiation. T helper 17 (TH17) cells are critically involved in host defence, inflammation, and autoimmunity1,2,3,4,5. Transforming growth factor β (TGFβ) is instrumental in TH17 cell differentiation by cooperating with interleukin-6 (refs 6, 7). Yet, the mechanism by which TGFβ enables TH17 cell differentiation remains elusive. Here we reveal that TGFβ enables TH17 cell differentiation by reversing SKI–SMAD4-mediated suppression of the expression of the retinoic acid receptor (RAR)-related orphan receptor γt (RORγt). We found that, unlike wild-type T cells, SMAD4-deficient T cells differentiate into TH17 cells in the absence of TGFβ signalling in a RORγt-dependent manner. Ectopic SMAD4 expression suppresses RORγt expression and TH17 cell differentiation of SMAD4-deficient T cells. However, TGFβ neutralizes SMAD4-mediated suppression without affecting SMAD4 binding to the Rorc locus. Proteomic analysis revealed that SMAD4 interacts with SKI, a transcriptional repressor that is degraded upon TGFβ stimulation. SKI controls histone acetylation and deacetylation of the Rorc locus and TH17 cell differentiation via SMAD4: ectopic SKI expression inhibits H3K9 acetylation of the Rorc locus, Rorc expression, and TH17 cell differentiation in a SMAD4-dependent manner. Therefore, TGFβ-induced disruption of SKI reverses SKI–SMAD4-mediated suppression of RORγt to enable TH17 cell differentiation. This study reveals a critical mechanism by which TGFβ controls TH17 cell differentiation and uncovers the SKI–SMAD4 axis as a potential therapeutic target for treating TH17-related diseases.
Evaluation of the activity of cardiac glycosides on RORγ and RORγT nuclear receptors

Abstract:Cardiac glycosides, derived from plants and animals, have been recognized since ancient times. These substances hinder the function of the sodium-potassium pump within eukaryotic cells. Many reports have shown that these compounds influence the activity of nuclear receptors. Thus, we assessed the effects of various cardiac glycosides at nontoxic concentrations on RORγ and RORγT. RORγT is a crucial protein involved in the differentiation of Th17 lymphocytes. Sixteen analyzed cardiac glycosides exhibited varying toxicities in HepG2 cells, all of which demonstrated agonistic effects on RORγ, as confirmed in the RORγ-HepG2 reporter cell line. The overexpression of both the RORγ and RORγT isoforms intensified the effects of these compounds. Additionally, these glycosides induced the expression of G6PC, a gene regulated by RORγ, in HepG2 cells. Subsequently, the effects of two endogenous cardiac glycosides (marinobufagenin and ouabain) and the three most potent glycosides (bufalin, oleandrin, and telecinobufagenin) were evaluated in Th17 primary lymphocytes. All of these compounds increased the expression of the IL17A, IL17F, IFNG, and CXCL10 genes, but they exhibited varying effects on GZMB and CCL20 expression. Molecular docking analysis revealed the robust binding affinity of cardiac glycosides for the ligand binding domain of the RORγ/RORγT receptors. Thus, we demonstrated that at nontoxic concentrations, cardiac glycosides have agonistic effects on RORγ/RORγT nuclear receptors, augmenting their activity. This potential can be harnessed to modulate the phenotype of IL17-expressing cells (e.g., Th17 or Tc17 lymphocytes) in adoptive therapy for combating various types of cancer.

Abstract:TH17 cells and their associated cytokines act on resident parenchymal cells within tissues, thereby setting the stage for chronic inflammation. This realization, together with the finding that TH17 cell development is reciprocally linked to that of T regulatory (Treg) cells, has revolutionized the way T cell-mediated immune pathology is viewed, and challenged the long-standing binary view of T cell differentiation (i.e., TH1/TH2), thereby opening exciting new opportunities to treat auto-immune inflammation. Much effort is now placed on understanding how TH17 cells are restrained through endogenous mechanisms; the goal being to negatively regulate TH17 development or function in clinical disease settings. The TH1 and TH2 cytokines, IFNγ and IL-4, as well as IL-27 and IL-10, all repress TH17 cell differentiation. TGFβ signaling, which supports TH17 differentiation in some contexts, can also strongly induce expression of the signature regulatory T cell transcription factor, Foxp3, which in turn cripples TH17 differentiation through direct antagonism of the TH17-specific orphan nuclear receptor RORγt. Emerging evidence also suggests that TH17 cells are both inherently unstable and uniquely sensitive to metabolic stress. Here, we discuss some of the key molecular features of TH17 cell development and highlight examples of cell-intrinsic and cell-extrinsic pathways that negatively influence TH17 differentiation, the latter of which could be exploited for therapeutic application.

Abstract:Recent evidence indicates that toll-like receptor (TLR) 2 and 4 are involved in the pathogenesis of dilated cardiomyopathy (DCM), but the exact mechanisms of their actions have not been elucidated. We explored the therapeutic potential of blocking TLRs in mice with established cardiomyopathy. Cardiomyopathy was generated by a single intraperitoneal injection of doxorubicin (10 mg/kg). Two weeks later, the mice were treated with TLR2 or TLR4 neutralizing antibody. Blocking TLR2, but not TLR4, activity not only reduced mortality, but also attenuated doxorubicin-induced cardiac dysfunction by 20% and inhibited myocardial fibrosis. To determine the differential effects of blocking TLR2 and TLR4 in chronic cardiomyopathy, mice were injected with doxorubicin (3.5 mg/kg) once a week for 8 weeks, followed by treatment with TLR2 or TLR4 neutralizing antibody for 40 days. Blocking TLR2 activity blunted cardiac dysfunction by 13% and inhibited cardiac fibrosis, which was associated with a significant suppression of myocardial inflammation. The underlying mechanism involved interrupting the interaction of TLR2 with its endogenous ligands, resulting in attenuation of inflammation and fibrosis. In contrast, blocking TLR4 exacerbated cardiac dysfunction and fibrosis by amplifying inflammation and suppressing autophagy. Our studies demonstrate that TLR2 and TLR4 play distinct roles in the progression of doxorubicin-induced DCM. TLR4 activity is crucial for the resolution of inflammation and cardiac fibrosis, while blocking TLR2 activity has therapeutic potential for the treatment of DCM.
Inhibiting the Th17/IL-17A-related inflammatory responses with digoxin confers protection against experimental abdominal aortic aneurysm

Abstract:Objective: T helper 17 cells and interleukin-17A have been implicated in the progression of abdominal aortic aneurysm (AAA). Retinoic acid-related orphan receptor gamma thymus, the master transcription factor of T helper 17 cell differentiation, is selectively antagonized by digoxin. However, the effect of antagonizing retinoic acid-related orphan receptor gamma thymus on AAA has not been investigated. Approach and results: We used human aortic sample analysis and 2 different experimental AAA models: (a) Angiotensin II (Ang II)-induced ApoE(-/-) male mice (Ang II/APOE model) and (b) porcine pancreatic elastase perfusion C57BL/6 mice (porcine pancreatic elastase/C57 model). In the Ang II/APOE model, all mice (n=80) were divided into 4 groups: sham group (saline+0.5% dimethyl sulfoxide treatment), control group (Ang II+0.5% dimethyl sulfoxide treatment), low-dose group (Ang II+low-dose digoxin, 20 μg/d per mouse), and high-dose group (Ang II+high-dose digoxin, 40 μg/d per mouse). All treatments began on day 0 after surgery. Efficacy was determined via aortic diameter and systolic blood pressure measurements, histopathology and protein expression, and flow cytometry analysis when euthenized. Human aortic tissue analysis showed that both interleukin-17A and retinoic acid-related orphan receptor gamma thymus increased in AAA tissues. The low-dose and high-dose groups had AAA incidences of 60% and 35%, respectively, compared with 70% in the control group. The T helper 17- and interleukin-17A-related inflammatory responses were dose-dependently attenuated by digoxin treatment. Digoxin was also highly effective in the porcine pancreatic elastase/C57 model. Conclusions: Digoxin attenuates experimental AAA progression in a model-independent manner. Antagonizing retinoic acid-related orphan receptor gamma thymus activity by digoxin may become a novel strategy for nonsurgical AAA treatment.
Short-term Pharmacological Inhibition of MyD88 Homodimerization by a Novel Inhibitor Promotes Robust Allograft Tolerance in Mouse Cardiac and Skin Transplantation.

Abstract:Background. Most strategies for antirejection and tolerance induction in clinical transplantation have focused on modifying adaptive immunity, it is unclear whether pharmacological suppressing the innate immune systemcan promote transplant tolerance. Methods. We inhibited innate immunity by using our self-generated inhibitor of myeloid differentiation factor 88 (MyD88), TJ-M2010-5, and investigated its therapeutic effects and mechanisms in cardiac and skin transplant models. Results. TJ-M2010-5 directly and indirectly interacted with the Toll/IL-1R domain of MyD88, inhibiting MyD88 homodimerization. In vitro, TJ-M2010-5 inhibited maturation of dendritic cells, which suppressed nuclear translocation of NF-kappa B and Tcell activation. In vivo, short-term (10 days) monotherapy of TJ-M2010-5 resulted in long time survival of 50% of the cardiac allografts, and longer-term (14 days) combination treatment of TJ-M2010-5 with CD154 mAb resulted in survival of 29% of skin allografts, which outperformed far more than CsA did and stimulated the proliferation of CD4(+)CD25(+)FoxP3(+) Regulatory T cells in recipient mice. Conclusions. Pharmacological inhibition of MyD88 signaling by this novel inhibitor TJ-M2010-5 shows a powerful antirejection effect, which may have therapeutic potential in clinical transplantation. The inhibition of both innate and adaptive immunity may be necessary for tolerance induction in nonsolid organs.

Abstract:Retinoid-related orphan receptor gamma t (ROR gamma t) is a member of the nuclear receptor family that is specifically expressed in T cell compartments. ROR gamma t regulates the development of T cells in the thymus and the differentiation of effector T cells in the periphery. During T cell development, ROR gamma t enhances CD4(+)CD8(+) double positive thymocyte survival by upregulating Bcl-x(L). In the periphery, ROR gamma t regulates IL-17 production and dictates the differentiation of pro-inflammatory T helper 17 (T(H)17) cells that play a critical role in inflammatory conditions and autoimmunity. ROR gamma t-deficient T cells fail to differentiate into T(H)17 cells, whereas forced expression of ROR gamma t is sufficient to induce naive T cells to produce IL-17. T(H)17 cells are believed to be the major inflammatory cells in autoimmune diseases. Therefore, inhibition of ROR gamma t activity could potentially alleviate the symptoms associated with the T(H)17-dependent inflammatory autoimmune diseases. ROR gamma t is thus potentially an excellent therapeutic target for the intervention of inflammatory autoimmunity.

Abstract:Although small-molecule thymus-specific isoform of retinoic acid receptor-related orphan nuclear receptor γ (RORγt) antagonists suppressing interleukin (IL)-17-producing T helper (Th17) cells are widely reported, the effect of these molecules on other RORγt-expressing cells is unknown. However, a new study reports that RORγt inhibition in CD4+CD8+ thymocytes resulted in skewed T cell repertoire, contributing to a reduction in the frequency of self-reactive T cells and resistance to autoimmunity.

Abstract:ObjectiveAccumulating studies have identified self‐DNA as driving IgG anti–double‐stranded DNA (anti‐dsDNA) in lupus, though the underpinning mechanisms of this process remain largely undefined. Here, we explored the activity of transcription factor retinoic acid receptor–related orphan nuclear receptor γt (RORγt) in the differentiation and function of self‐DNA–specific follicular helper T (Tfh) cells in lupus.MethodsB6, TCRα–/–, CD4–/–, RORγtfl/flCD4Cre, RORγt+/+CD4Cre, Bcl‐6fl/flCD4Cre, Bcl‐6+/+CD4Cre, IL‐17–/–, and ICOS–/– mice were immunized with normal self‐DNA, immunogenic self‐DNA, and pathogen DNA to induce the production of Tfh cells and IgG anti‐dsDNA. Tfh cells with or without interleukin‐17 (IL‐17) were evaluated for their role in supporting the generation of IgG. NSG mice were reconstituted with immune cells and circulating DNA from human subjects for translational studies. IL‐17–positive Tfh cells were analyzed for their correlation with IgG anti‐dsDNA levels as well as their response to circulating self‐DNA in lupus patients.ResultsUnlike normal self‐DNA, immunogenic self‐DNA and pathogen DNA efficiently induced IgG responses. Immunogenic self‐DNA induced IgG in a CD4+ T cell–dependent manner, which was abrogated by RORγt deficiency. In contrast, RORγt was not required for the generation of pathogen DNA–induced IgG. Further analyses identified RORγt as essential for the differentiation and function of Tfh cells in response to immunogenic self‐DNA, assigning IL‐17 as a feature cytokine. These IL‐17–positive Tfh cells functioned independent of inducible costimulator (ICOS), critically supporting IgG generation. Targeting immunogenic self‐DNA–specific Tfh cells by RORγ knockdown and IL‐17 blockade ameliorated IgG response and lupus nephritis in a humanized mouse model. The presence of IL‐17–positive Tfh cells was associated with IgG anti‐dsDNA levels and were expanded by circulating immunogenic self‐DNA in lupus patients.ConclusionImmunogenic self‐DNA instructs ICOS‐dispensable IL‐17–positive Tfh cells via RORγt to produce an IgG anti‐dsDNA response. As such, IL‐17–positive Tfh cells are a promising therapeutic target for lupus patients.

Abstract:Relatively low-grade inflammatory of osteoarthritic joints is characterized by synovitis and a catabolic and proinflammatory state of the chondrocytes and plays an important role in osteoarthritis (OA) initiation and exacerbation. Our previous research showed cardiac glycoside compounds might be effective in OA synovitis. However, the effect of digoxin (DIG), an FDA-approved cardenolide, on inflammation inhibition of osteoarthritic joints has not been investigated. In the present study, a western blot analysis and immunofluorescence staining revealed that DIG alleviated OA synovitis by inhibiting the M1-like polarization of synovial macrophages in OA patients and collagenase-induced OA (CIOA, with considerable synovitis) mice. Subsequently, the exosomes produced by macrophages and M1-like macrophages treated with or without DIG were isolated and identified. According to miRNA sequencing analysis of these exosomes and subsequent target activity assays, we confirmed DIG controls OA inflammatory microenvironment and promotes chondrogenesis by, at least partly, downregulating the M1-like macrophage-derived exosomal miR-146b-5p/Usp3&Sox5 axis in vitro and in vivo. This research provides reliable experimental evidence supporting the clinical application of DIG as a disease-modifying drug for inflammation-associated OA. Additionally, the spectrum of diseases of inflammation controlled by DIG has been broadened, which prompting research interest in the new function of an "old" FDA-approved drug.
