Abstract:Conventional immunocolorimetric sensing of biomolecules continues to be challenged with low sensitivity although its wide application as a diagnostic tool in medicine and biotechnology. Herein, we present a multifunctional immunocolorimetric sensing system for sensitive and high-throughput detection of B-type natriuretic peptide (BNP) with carbon-gold nanocomposite (CGNs) amplification. Using a “green” strategy, monodisperse carbon nanospheres (CNs) were successfully synthesized by glucose carbonization. A simple and efficient hydrothermal method was developed to assemble abundant gold nanoparticles (AuNPs) onto the surfaces of CNs. The resulting CGNs were characterized and utilized for biomarker detection with superior properties of easy manufacturing, good biocompatibility, satisfactory chemical stability, and high loading capacity for biomolecules. As a proof of concept, the as-prepared CGNs were conjugated with horseradish peroxidase-labeled antibody against BNP (CGNs@AntiBNP-HRP) functioning as the carrier, signal amplifier, and detector for the sensitive detection of BNP. Under optimal conditions, the established CGNs@AntiBNP-HRP immunoprobe remarkably enhanced the detection performance of BNP, achieving signal amplification of more than 9 times compared to the conventional method. Based on our self-developed bionic electronic eye (e-Eye) and CGNs@AntiBNP-HRP immunoprobe, the multifunctional sensing system displayed a wide dynamic linear range of 3.9–500 ng/mL and a LOD of 0.640 ng/mL for BNP detection with high specificity, good accuracy and reproducibility. This portable sensing system with enhanced performance demonstrates great potential for BNP detection in point of care applications, and offers a universal and reliable platform for in vitro biomarker detection.

Abstract:We have developed a new nanoparticle (NP)-based chemiluminescent (CL) method for the sensitive detection of DNA hybridization. The "sandwich-type'' DNA technique comprises NH2-functionalized capture DNA immobilized on magnetic beads that can hybridize with one end of the target DNA. The other end of the target is recognized by a reporter DNA probe labeled with Au NPs on its 3-terminus. The hybridization events were monitored by a simple and direct luminol-AgNO3 CL reaction with a detection limit of 0.5 pM through a non-stripping Au NPs catalysis. Moreover, the CL reaction of non-stripping Au NPs-luminol-AgNO3 offers the advantages of being simpler and faster as compared to previous stripping Ag NPs and CuS methods. This simple and easy-to-use technique holds great promise for clinical applications of DNA hybridization.

Abstract:This work designed a simple, sensitive, and low-cost immunosensor for the detection of protein marker by using a carbon sphere/gold nanoparticle (CNS/AuNP) composite as an electrochemical label. The nanoscale carbon spheres, prepared with a hydrothermal method by using glucose as raw material, were used to load AuNPs for labeling antibody by electrostatic interaction, which provided a feasible pathway for electron transfer due to the remarkable conductivity. The disposable immunosensor was constructed by coating a polyethylene glycol (PEG) film on a screen-printed carbon-working electrode and then immobilizing capture antibody on the film. With a sandwich-type immunoassay format, the analyte and then the CNS/AuNP-labeled antibody were successively bound to the immunosensor. The bound AuNPs were finally electro-oxidized in 0.1?M HCl to produce AuCl4- for differential pulse voltammetric (DPV) detection. The high-loading capability of AuNPs on CNS for the sandwich-type immunorecognition led to obvious signal amplification. By using human immunoglobulin G (IgG) as model target, the DPV signal of AuNPs after electro-oxidized at optimal potential of +1.40 V for 40 s showed a wide linear dependence on the logarithm of target concentration ranging from 10 pg?mL-1 to 10 ng?mL-1. The detection limit was around 9 pg?mL-1. The immunosensor showed excellent analytical performance with cost effectivity, good fabrication reproducibility, and acceptable precision and accuracy, providing significant potential application in clinical analysis.

Abstract:A novel nanogold-enriched carbon nanohorn structure (nanoAu/CNH) was designed as a trace tag for highly efficient electrochemical detection of tumor marker using a disposable immunosensor. The nanoAu/CNH was synthesized by one-pot in situ growth of nanogold on carboxylated single-walled CNH. The labeling of signal antibody was performed via the inherent interaction between protein and nanogold. The disposable immunosensor was constructed by covalently cross-linking capture antibody on chitosan modified screen-printed carbon electrode. With a sandwich format, the nanoAu/CNH labeled antibody was conjugated on the immunosensor for electrochemical measurement of tumor marker by electrooxidizing the nanoAu at +1.3V and then cathodic potential scan in 0.1M HCl. Using -fetoprotein as model target, the proposed immunoassay showed acceptable precision and wide linear range from 0.1pgmL1 to 1ng mL1 with a detection limit of 0.07pgmL1. The new nanoAu/CNH label as well as the disposable immunosensor showed possessed potential application in point-of-care testing.

Abstract:A simple and disposable electrochemical immunosensor was developed for sensitive and selective detection of a protein biomarker via target-induced proximity hybridization and electrochemical stripping analysis of silver nanoparticles (AgNPs). The immunosensor was prepared by assembling single-stranded DNA modified gold nanoparticles (ssDNA@AuNPs) on a graphene oxide modified disposable screen-printed carbon electrode (SPCE). In the presence of a target protein and two DNA-labeled antibodies, the proximate complex formed in homogeneous solution could hybridize with the assembled DNA to take away AuNPs, which decreased AuNP-catalyzed deposition of AgNPs on the immunosensor surface, and thus the anodic stripping signal. The proposed method avoided the interference of dissolved oxygen. Using the carcinoembryonic antigen (CEA) as a model analyte, this method showed a linear range of four magnitude orders with a detection limit down to 3.9 pg mL(-1). The electrochemical immunosensor possessed preparation convenience, good stability and high sensitivity, and could be extended to sensitive biosensing of other analytes, showing potential application in point-of-care testing.

Abstract:Gold colloid changes its color when the internanoparticle distance changes. On the basis of analyte-induced aggregation or disaggregation behavior of gold nanoparticles (AuNPs), versatile colorimetric assays have been developed for measuring various kinds of analytes including proteins, DNA, small molecules, and ions. Traditional read-out signals, which are usually measured by a spectrometer or naked eyes, are based on the averaged extinction properties of a bulk solution containing billions of nanoparticles. Averaged extinction property of a large amount of nanoparticles diminished the contribution from rare events when the analyte concentration was low, thus resulting in limited detection sensitivity. Instead of measuring the averaged optical property from bulk colloid, in the present work, we proposed a digital counterpart of the colorimetric assay by imaging and counting individual AuNPs. This method quantified the analyte concentration with the number percentage of large-sized AuNPs aggregates, which were digitally counted with surface plasmon resonance microscopy (SPRM), a plasmonic imaging technique recently developed by us and other groups. SPRM was able to identify rare AuNPs aggregates despite their small population and greatly improved the detection sensitivity as demonstrated by two model systems based on analyte-induced aggregation and disaggregation, respectively. Furthermore, besides plasmonic AuNPs, SPRM is also suitable for imaging and counting nonplasmonic nanomaterials such as silica and metal oxide with poor extinction properties. It is thus anticipated that the present digitized assay holds a great potential for expanding the colorimetric assay to broad categories of nonplasmonic nanoparticles.

Abstract:This work proposed a simple, sensitive and low-cost multiplexed immunoassay by combining a disposable chip with gold nanoparticle (AuNP) as an electrochemical label. The immunosensors array as the disposable chip was firstly prepared by immobilizing capture antibodies on different screen printed carbon working electrodes by passive adsorption. With a sandwich mode, the analytes were then bound to the corresponding capture antibodies for further capture of the gold nanoparticle labeled antibodies. Gold nanoparticles were finally electrooxidized in 0.1 M HCl to produce AuCl(4)(-) for differential pulse voltammetric detection. Using human IgG and goat IgG as model targets, under optimal conditions this method achieved linear ranges from 5.0 to 500 and 5.0 to 400 ng mL(-1) with limits of detection of 1.1 and 1.6 ng mL(-1), respectively. This method eliminated completely electrochemical crosstalk between adjacent immunosensors due to the strong adsorption of the AuCl(4)(-) on the printed carbon surfaces. The immunosensors array showed acceptable precision, reproducibility and stability, and could be readily applied to multianalyte determination for clinical diagnostics.
Naked-eye sensitive ELISA-like assay based on gold-enhanced peroxidase-like immunogold activity

Abstract:A naked-eye sensitive ELISA-like assay was developed based on gold-enhanced peroxidase-like activity of gold nanoparticles (AuNPs). Using human IgG (H-IgG) as an analytical model, goat anti-human IgG antibody (anti-IgG) adsorbed on microtiter plate and AuNPs-labeled anti-IgG acted as capture antibody and detection antibody, respectively. Because the surfaces of AuNPs were blocked by protein molecules, the peroxidase-like activity of AuNPs was almost inhibited, evaluated by the catalytic oxidation of peroxidase enzyme substrate 3,3',5,5'-tetramethylbenzidine (TMB), which could produce a bright blue color in the presence of H2O2. Fortunately, the catalytic ability of AuNPs was dramatically increased by the deposition of gold due to the formation of a new gold shell on immunogold. Under optimal reaction conditions, the colorimetric immunoassay presented a good linear relationship in the range of 0.7-100 ng/mL and the limit of detection (LOD) of 0.3 ng/mL calculated by 3σ/S for UV-vis detection, and obtained LOD of 5 ng/mL for naked-eye detection. The obtained results were competitive with conventional sandwich ELISA with the LOD of 1.6 ng/mL. Furthermore, this developed colorimetric immunoassay was successfully applied to diluted human serum and fetal bovine serum samples, and predicted a broad prospect for the use of peroxidase-like activity involving nanomaterials in bioassay and diagnostics.

Abstract:<p>A simple and convenient electrochemiluminescence immunoassay (ECLIA) has been developed by using gold nanoparticles (AuNPs) as both label and co-reactant. In this novel ECLIA, the detection antibodies labeled with AuNPs as co-reactant reacts directly with ruthenium complex to produce electrochemiluminescence (ECL), which avoids the weakness of luminophore as a label. The feasibility of the new method was investigated through the sandwich ECL immunosensor for the determination of human immunoglobulin (HIgG), and excitation of luminescence was respectively driven by linear sweep voltammetry and step potential. Under the optimal conditions, ECL intensity of the immunosensor increased with HIgG concentration in a wide range from 0.01 to 10.0 ng/mL and displayed linear response to logarithm of HIgG concentration. The results showed that the detection limit was 5 pg/mL, and the relative error was no more than 4.6% for the repeated measurements. The relative standard deviation was less than 2.9% for the determination of the standard sample, which can meet the requirement of ECLIA.</p>

Abstract:An electrochemical immunosensor for detection of neuron specific enolase (NSE) was designed by immobilizing NSE covalently functionalized single-walled carbon nanotubes (NSE-SWNTs) on a glassy carbon electrode. The NSE-SWNTs not only enhanced electrochemical signal but also presented abundant antigen domains for competitive immunological recognition to anti-NSE primary antibody and then gold nanoprobes labeled with alkaline phosphatase conjugated secondary antibody (AP-anti-IgG/AuNPs). The AP-anti-IgG/AuNPs exhibited highly catalytic activity toward enzyme substrate and significantly amplified the amperometric signal for target molecule detection. Based on the dual signal amplification of SWNTs and gold nanoprobe, the immunosensor could response down to 0.033 ng mL(-1) NSE with a linear range from 0.1 ng mL(-1) to 2 mu g mL(-1), and showed acceptable precision and reproducibility. The designed immunosensor was amenable to direct quantification of target protein with a wide range of concentration in complex clinical serum specimens. The assay results were in a good agreement with the reference values. Abstract:A label-free immunosensor was designed and fabricated for sensitive detection of alpha-fetoprotein (AFP) based on the localized surface plasmon resonance (LSPR) of gold nanorods (GNRs). GNRs were fabricated through seedmediated growth and surface activation by alkanethiols for the attachment of antibodies that could directly capture of the target. A novel strategy using the absorbance ratio of transverse and longitudinal LSPR peak of GNRs as a function of AFP contraction was performed for AFP quantification. The limit of detection of the sensor is 0.25nM in the PBS, and the dynamic range spans 0.25nM to 14.3nM. It is expected that this simple and cost-effective LSPR biosensor can also be applicable to the detection of several other tumor markers that are present at low concentrations.

Abstract:Lead poisoning in adults can affect the peripheral and central nervous systems, the kidneys, and blood pressure. Thus, the development of environment-friendly and simple methods for Pb2+ detection is of great importance. Herein, a label-free colorimetric method has been developed for the detection of Pb2+ based on the conformational switch from single-stranded DNA to G-quadruplex. The electrostatic interactions between DNA probe and gold nanorods (GNRs) induce GNRs to space closely. However, the electrostatic interaction is not strong enough to change the suspension state of GNRs. In the presence of Pb2+, the formation of G-quadruplexes increases the surface charge density around DNA, which is expected to strengthen the electrostatic interaction between the GNRs and the DNA. Therefore, the longitudinal absorption of GNRs decreased because the stronger interaction induced aggregation of GNRs. Importantly, the decrease in longitudinal absorption is proportional to concentration of Pb2+. By monitoring the change of absorbance, Pb2+ can be detected at a level of 3 nM with a linear range from 5 nM to 1 μM. The overall test only takes a few minutes and very little interference is observed from other metal ions. The major advantages of this method are its low cost, convenience, simplicity, sensitivity, and specificity.

Abstract:A multiplexed electrochemical immunoassay method was developed for simultaneous ultrasensitive measurement of tumor markers based on electrochemical stripping analysis of silver nanoparticles (Ag NPs). The Ag NPs were deposited on a disposable immunosensor array with a reduction reaction catalyzed by nanogold labels. The immunosensor array was prepared by covalently immobilizing capture antibodies on chitosan modified screen-printed carbon electrodes. Through a sandwich-type immunoreaction, antibody-functionalized Au NPs were captured onto immunosensor surface to induce the silver deposition from a silver enhancer solution. The deposited Ag NPs could be directly measured by anodic stripping analysis in KCl solution. The catalytic deposition enhanced the analytical sensitivity for detection of protein markers. The interference of dissolved oxygen could be avoided as the detection was performed with positive stripping potential range. Using carcinoembryonic antigen and α-fetoprotein as model analytes, the proposed multiplexed immunoassay method showed wide linear ranges of three orders of magnitude with the detection limits down to 3.5 and 3.9pgmL−1, respectively. The localized silver deposition, as well as the stripping detection process, eliminated completely the electrochemical cross talk between adjacent immunosensors. The immunosensor array exhibited acceptable reproducibility, stability and accuracy, showing a promising potential in multianalyte determination for clinical application.
Gold Nanoparticle/dna/methylene Blue Nanocomposites for the Ultrasensitive Electrochemical Detection of Carcinoembryonic Antigen

Abstract:A new signal amplification strategy employing nanocomposites composed of gold nanoparticles (GNPs) with DNA and methylene blue (MB) as labels has been developed to construct a novel immunosensor for the highly sensitive bioanalysis of carcinoembryonic antigen (CEA). To fabricate the label, DNA was first linked to GNPs through the formation of an Au–S bond. Next, the resulting bioconjugates were sequentially bound with MB via the guanine (G) bases in DNA specifically and then with secondary CEA antibody (Ab2) via the crosslinking of glutaraldehyde (GA). The receptor–analyte complex is formed by the biorecognition of the as-obtained MB labeled Ab2 with the primary CEA antibody (Ab1) immobilized on the surface of a modified GNP/Chitosan (Chits) composite electrode. After the receptor–analyte complex has formed, square wave voltammetry (SWV) was employed to oxidize the labeled MB in order to detect the CEA. This immunosensor shows a linear response from 0.10 to 2.0pgmL−1 CEA with an improved detection limit of 0.05pgmL−1 as compared to the conventional immunoassay. In addition, this new protocol shows acceptable stability, reproducibility and good recovery (97.5–110.0%) for CEA in human serum with great potential for clinical applications.

Abstract:An ultrasensitive electrochemiluminescence (ECL) immunosensor based on luminol functionalized gold nanoparticle (AuNP) labeling was developed using human immunoglobulin G (hIgG) as a model analyte. The primary antibody biotin-conjugated goat-anti-human IgG was first immobilized on a streptavidin coated AuNP modified electrode, then the antigen (human IgG) and the luminol functionalized AuNP-labeled second antibody were conjugated successively to form a sandwich-type immunocomplex, i.e. immunosensor. ECL was carried out with a double-step potential in carbonate buffer solution containing 1.0 mmol/L H(2)O(2). Since thousand of luminol molecules were coated on the surface of AuNPs to realize labeling of multiple molecules with CL activity at a single antibody and the amplification of AuNPs and biotin-streptavidin system was utilized, luminol ECL signal could be enhanced greatly, finally resulting in extremely high sensitivity. The ECL method shows a detection limit of 1.0 pg/mL (S/N=3) for hIgG, which is superior to all previously reported methods for the determination of hIgG. Moreover, the proposed method is also simple, stable, specific, and time-saving, avoiding the complicated stripping procedure during CL detection and the uncontrollable synthesis of irregular nanoparticles compared with other chemiluminescence immunoassay based on AuNP labeling. Additionally, the labeling procedure is also superior to that of other reported multilabeling strategies, such as Ru complex-encapsulated polymer microspheres, and most of Ru complex-encapsulated liposomes in simplicity, stability, labeling property and practical applicability. Finally, the proposed method has been successfully applied to the detection of hIgG in human serums.

With the implementation of a unique recycling strategy, we have demonstrated that the miRNA in the concentration as low as 0.5 pM can be detected without the need for instrumentation, providing a detection limit that surpasses previous reports. The new biosensor is ultrasensitive and can be applied to the reliable monitoring of miRNAs in cell lysates with high accuracy. The approach offers a simple and rapid tool for cancer diagnosis and clinical biomedicine, thanks to its flexibility, simplicity, cost-effectiveness, and convenience. This new method has the potential to significantly improve the detection and monitoring of cancer biomarkers, ultimately contributing to more effective cancer diagnosis and treatment.
Gold Nanorods as Colorful Chromogenic Substrates for Semiquantitative Detection of Nucleic Acids, Proteins, and Small Molecules with the Naked Eye

Abstract:Herein, we report for the first time a colorful chromogenic substrate, which displays vivid color responses in the presence of different concentration of analytes. Our investigation reveals that the selective shortening of gold nanorods (AuNRs) could generate a series of distinct colors that covers nearly the whole visible range from 400 to 760 nm. These vivid colors can be easily distinguished by the naked eye; as a result, the accuracy of visual inspection could be greatly improved. Next, we demonstrate the utility of AuNRs as multicolor chromogenic substrate to develop a number of colorimetric immunoassay methods, e.g., multicolor enzyme-linked immunosorbent assay (ELISA), multicolor competitive ELISA, and multicolor magnetic immunoassay (MIA). These methods allow us to visually quantify the concentration of a broad range of target molecules with the naked eye, and the obtained results are highly consistent with those state-of-the-art techniques that are tested by the sophisticated apparatus. These multicolor portable and cost-effective immunoassay approaches could be potentially useful for a number of applications, for example, in-home personal healthcare, on-site environmental monitoring, and food inspection in the field.
