Abstract:Purpose: This paper aimed to evaluate the effects of a combination of paclitaxel and cisplatin on osteosarcoma (OS) cell lines in the presence of hyperthermia and to investigate the related mechanism.Materials and methods: Two types of OS cell lines (OS732 and MG63) were treated with paclitaxel and cisplatin in the presence of hyperthermia. The survival rate was measured by MTT assay, and the clonogenic rate was measured by a clonogenic assay. The cellular changes were observed with an inverted phase contrast microscope and a fluorescence microscope. The apoptotic effect was analysed with flow cytometry (FCM). Fas expression by the OS cell lines was measured by western blot. Fas expression in OS tissue was measured by immunohistochemistry.Results: Our study indicated that 1 h after the application of a combination of 10 mu g/mL paclitaxel and 5 mu g/mL cisplatin to OS cells at 43 degrees C, the survival rate of the OS cells was 11.96%, which was significantly lower than when either 10 mu g/mL paclitaxel (45.02%) or 5 mu g/mL cisplatin (48.69%) was applied alone (p<0.01). Additionally, the clonogenic assay demonstrated that the clonogenic survival rate in the OS cells of the combination group was lower than that in the individual groups. Moreover, the cellular changes and apoptosis rates indicated that apoptosis in the combined application group was much greater than when either drug was applied individually. Fas expression by OS cell lines was increased by the combination of paclitaxel and cisplatin under hyperthermic conditions. More importantly, our study revealed low Fas expression in OS, which better explained the up-regulation of Fas achieved by the combination of paclitaxel and cisplatin in the presence of hyperthermia.Conclusions: The combination of paclitaxel and cisplatin increases the effects of thermochemotherapy on OS cell lines, primarily through the induction of apoptosis by the up-regulation of Fas expression.

Abstract:OBJECTIVE:To evaluate the effect of hypoxia improvement in Hep-2 cell on cisplatin-induced apoptosis.METHOD:Human laryngeal squamous cell carcinoma Hep-2 cells and HIF-1α-RNAi-Hep-2 cells were cultured in normoxic, hypoxic and reoxygenation condition. The inhibition of cisplatin on cell proliferation was evaluated by MTT assay. The influence of cisplatin on cell cycle and apoptosis were detected by flow cytometry.RESULT:The inhibition of cisplatin on cell proliferation was reduced by hypoxia. After HIF-1α gene was silenced, the inhibition of cisplatin on Hep-2 cell proliferation was increased apparently, but was still interfered partly by hypoxia. Hypoxia could induce cell apoptosis decreased and enhance chemotherapeutic resistance. After reoxygenation, cell apoptosis induced by cisplatin was increased significantly (P < 0.05). HIF-1α-RNAi-Hep-2 cells under hypoxia also showed certain resistance to apoptosis but the sensitivity to cisplatin was higher than that of Hep-2 cells. When cells were returned from hypoxic condition to normoxic condition for some time, the apoptosis induced by cisplatin was increased significantly (P < 0.05).CONCLUSION:The improvement of hypoxic microenvironment with HIF-1α gene knockout could increase the sensitivity of Hep-2 cells to chemotherapy.

Abstract:Abstract Cytotoxic effects of hyperthermia combined with DDP on MKN28 and MKN45 cells were studied by MTT assay according to a nested design. The results showed: hyperthermia alone above 43 °C for 30 mins was cytotoxic; hyperthermia at temperature lower than 43 °C for 30 mins could increase sensitivity of cancer cells to DDP. The cytotoxic effect of simultaneous use of hyperthermia and DDP was more marked than that of sequential use of the 2 treatments. Hyperthermia combined with DDP could inhibit growth of human gastric adenocarcinoma cells regardless of their degree of differentiation.

Abstract:PML/RARα fusion protein is the oncogenic driver in acute promyelocytic leukemia (APL). While most APL cases are cured by PML/RARα targeting therapy, relapse and resistance can occur due to drug-resistant mutations. Here we report that thermal stress destabilizes PML/RARα protein, including clinically identified drug-resistant mutants. AML1/ETO and TEL/AML1 oncofusions show similar heat shock susceptibility. Mechanistically, mild hyperthermia stimulates aggregation of PML/RARα in complex with nuclear receptor corepressors leading to ubiquitin mediated degradation via SIAH2 E3 ligase. Hyperthermia and arsenic therapy destabilize P/R via distinct mechanisms and are synergistic in primary patient samples and in vivo including 3 refractory APL cases. Collectively, our results suggest that by taking advantage of a biophysical vulnerability of PML/RARα, thermal therapy may improve prognosis in drug-resistant or otherwise refractory APL. These findings serve as a paradigm for therapeutic targeting of fusion oncoprotein associated cancers by hyperthermia. Statement of Significance: Hyperthermia destabilizes oncofusion proteins including PML/RARα and acts synergistically with standard arsenic therapy in relapsed and refractory acute promyelocytic leukemia. Abstract:Objective:To observe the synergistic effect of hyperthrnia and cisplatin on human lung adenocarcinoma cell line H1299.Methods:The cells treated 42 degree for 2 hours or not were detected cisplatin sensitivity with CCK-8 assay. After 42 degree for 2 hours, ciplatin 10μg/ml for 24 hours or the combination with both sides, the inhibition ratio was detected wtih CCK-8 assay and flow cytometry analyzed DNA contents in every phase of cell proliferation, and we observed their synergistic effect on cytotoxicity and cell cycel.Results:After hyperthermia, the IC_ 50 values to cisplatin decreased from 9.51μg/ml to 6.07μg/ml. CCK-8 assay showed hyperthermia and ciplatin had synergistic cell killing effect, and FCM also showed hyperthermia had significant effect on cell cycle with increase of G0/G1 phase population and reduction of S phase population.Conclusion:Hyperthermia can significantly increase the sensitivity of H1299 cells to cisplatin.
[Pretreatment with Cisplatin Plus Hyperthermia Kills Different Tumor Cells but Preserves the Erythrocyte Function in the Intra-Operative Blood Salvage in Vitro].

Abstract:OBJECTIVE:To investigate the effects of cisplatin plus hyperthermia on erythrocytes and killing human hepatocarcinoma (HepG2), gastro carcinoma (SGC7901) and colonic carcinoma (SW620) cells in the intra-operative blood salvage from cancer surgery in vitro.METHODS:HepG2, SGC7901 or SW620 cells were mixed into the aliquot of erythrocyte concentrated from each intra-operative blood salvage of 30 patients subjected to gastrointestinal cancer surgery. The mixture cells were divided into the following groups (n = 30): A group (37 °C); B group (42 °C); C, D, E groups (50, 100, or 200 µg/ml DDP); F, G, H, I groups (42 °C, 25, 50, 100, or 200 µg/ml DDP). After treating for 60 min, tumor cells and erythrocytes were separated by density gradient centrifugation. The Na(+)-K(+)-ATPase activity, cell count, osmotic fragility, and blood gas variables were determined in erythrocytes. Cell viability and colony formation were determined in tumor cells.RESULTS:Compared with A [(0.30 ± 0.08) µmol Pi/10(7)/h], the Na(+)-K(+)-ATPase activity was significantly decreased in E, H and I groups [(0.24 ± 0.07), (0.25 ± 0.06) and (0.24 ± 0.07) µmol Pi/10(7)/h] (P < 0.05). Extra-erythrocytic K(+) in E, H and I groups [(2.16 ± 0.37), (2.16 ± 0.38) and (2.56 ± 0.50) mmol/L] were significantly increased compared with A group [(1.53 ± 0.43) mmol/L] (P < 0.05). Compared with A group, osmotic fragility in E, H and I groups was significantly increased (P < 0.05). Among B, C, D, E, F, G groups, only in G group colony formations of HepG2, SGC7901, and SW620 (0% ± 0%, 0% ± 0% and 0.01% ± 0.01%) at 14 d were completely inhibited (P < 0.01) compared with A group (78.54% ± 7.83%, 72.28% ± 6.58% and 66.69% ± 6.69%).CONCLUSION:Pretreatment with cisplatin (50 µg/ml) plus hyperthermia (42 °C) for 60 min in vitro might be an effective strategy to clear tumor cells contamination but preserve erythrocytes, which is worthy to be optimized and used in the intra-operative blood salvage in cancer surgery.
Hyperthermia enhances the efficacy of chemotherapeutic drugs in heat-sensitive cells through interfering with DNA damage repair

Abstract:Background: Hyperthermic intraperitoneal chemotherapy (HIPEC) has been shown to be clinically effective, but the mechanisms by which hyperthermia enhances the sensitivity of cells to chemotherapeutic drugs has not yet been elucidated.Methods: To identify the key molecules involved in thermochemotherapy, this study used mass spectrometry (MS)-based quantitative proteomics technology to analyze the effects of thermochemotherapy on the heat-sensitive ovarian cancer cell line A2780. We divided the A2780 cell line into four groups, one group served as blank control, and the other three groups were stimulated by oxaliplatin, stimulated by hyperthermia at 42 ℃, and stimulated by hyperthermia combined with oxaliplatin. Samples were then collected for tandem mass tag (TMT) labeling, high-performance liquid chromatography fractionation, and MS-based quantitative proteomics for analysis The differentially expressed proteins were quantitatively compared and identified, and Gene Ontology (GO) assessment and cluster analyses were performed. Finally, the above MS results were verified again by Western blotting experiments.Results: A total of 349 differentially expressed proteins were identified between cells treated with chemotherapy alone (group B) and cells treated with a combination of chemotherapy and hyperthermia (group D). There were 145 upregulated proteins and 204 downregulated proteins. Among the top 20 proteins with significantly different expression levels, nearly two-thirds were involved in DNA damage repair. These proteins were subsequently verified by Western blot analysis. Indeed, consistent with MS data, the expression of the RBL1 protein was significantly upregulated in cells treated with thermochemotherapy (group D) compared to cells treated with chemotherapy alone (group B).Conclusions: In heat-sensitive ovarian cancer cells, the damage repair of tumor cell DNA is disturbed by hyperthermia, making it unable to fully repair when damaged by chemotherapeutic drugs. As a result, hyperthermia enhances the efficacy of chemotherapeutic drugs. <i>RBL1</i>, as a tumor suppressor gene, may be associated with the repair of DNA damage, and thus it may be a key target for hyperthermia to enhance the sensitivity of thermosensitive cells to chemotherapeutic drugs.

Abstract:Tumors with homologous recombination (HR) deficiency are particularly responsive to PARP inhibitors, however strategies to improve the sensitivity of epithelial ovarian carcinoma (EOC) with sufficient HR abilities still need to be deeply explored. In the present study, we firstly validated that hyperthermia (HT) changed diverse genes and signal pathways related to HR and oxidative stress in HR proficient EOC cells. HT impaired HR efficiency through inhibiting Olaparib (Olap) induced RAD51 foci formation in EOC cells, which was independent of the expression level of RAD51. Combination therapy of HT and Olap synergistically induced oxidative stress and oxidative DNA damage of EOC cells. Furthermore, we revealed that HT and Olap synergistically aggravated double-strand breaks of DNA in EOC cells. Conclusively, our findings confirmed that HT could synergistically enhance HR proficient EOC cells' sensitivity to PARP inhibitor through impairing HR efficiency and increasing oxidative stress.

Abstract:To evaluate the treatment effects of combined therapy using Doxorubicin and radiofrequency hyperthermia and expound the mechanism of local Chemotherapy under the heat-stress condition in vitro and in a rabbit VX2 liver tumor model Institutional Animal Care and Use Committee approved all studies. In vitro experiments VX2 cell lines cultured and were randomly assigned to four treatment groups, namely. Group A, phosphate-buffered saline (n = 6); group B, 5 μΜ Doxorubicin alone; hroup C, 42°C culture for 20 min (n = 6); and group D, 5 μΜ doxorubicin plus under 42°C culture for 20 min (n = 6). The viability and doxorubicin uptake of VX2 tumor cells line were assayed by flow cytometer and fluorescence microscopy at 24h after different treatment. Western blot was used to evaluate the Hsp70 expression. Tumor tissues were harvested from our Prestudy of rabbit liver VX2 tumor model. Fluorescence microscopy observed the distribution of doxorubicin, Hsp70 activity was analyzed by Western blot and Immunohistochemistry. P <0.05 was considered statistically significant. Radiofrequency hyperthermia-induced apoptosis and necrosis in VX2 cells in a mechanism of enhancing doxorubicin dose in cell and Hsp 70 expression. Fluorescence microscopy showed a further increase doxorubicin concentration in tumor cells and the penetration depth in tumor tissue. The western blot result showed Hsp70 expression in the combined group was significantly greater than in the control groups (mean ± SEM: 0.15 ± 0.03 vs. 0.64 ± 0.13 vs. 0.83 ± 0.10 vs. 1.28 ± 0.13, P <0.05). In the tumor tissue, immunohistochemistry staining results showed the significant increase of Hsp70-positive signaling was observed in RFH combined doxorubicin group than control group (P <0.01). Significantly increased of the tumor apoptosis and necrosis also found in the combined group with Ki-67 and TUNEL staining (P < .001). Radiofrequency hyperthermia mediate chemotherapy-induced VX2 cell killing regulated cell death mechanisms include enhance penetration of doxorubicin and HSP70 interacting at several points on apoptotic and immune signaling pathways, leads to inhibition of apoptosis of cell death at the HCC tumor ablation margin.
RESEARCH ON COMBINATION OF CISPLATIN WITH HYPERTHERMIA AGAINST HEPATOCELLULAR CARCINOMA CELL LINE SMMC-7721 IN VITRO

Abstract:Objectives This experiment aims to investigate whether concurrent exposure of human hepatocellular carcinoma cells SMMC-7721 to cisplatin combined with hyperthermia will increase anticancer effects.Methods In our study,the MTT assay has been developed for quantitative evaluation of the proliferation of SMMC-7721 and cytotoxicity of cisplatin with or without hyperthermia.Flow cytometric analyses were used for the assessment of apoptosis rates after treatment.Results The combination treatments induced significantly higher apoptosis and cytotoxicity than exposure to single 43 ℃ hyperthermia or cisplatin.Conclusion While either hyperthermia or cisplatin can induce apoptosis and anti-proliferation effect,they may offer synergistic benefits when used concurrently.
The Role of Hyperthermia in Potentiation of Anti-Angiogenic Effect of Cisplatin and Resveratrol in Mice Bearing Solid Form of Ehrlich Ascites Tumour

Abstract:The aim of this study was to investigate the therapeutic potential of resveratrol in combination with cisplatin on the inhibition of tumour angiogenesis, growth, and macrophage polarization in mice bearing the solid form of an Ehrlich ascites tumour (EAT) that were exposed to whole-body hyperthermia treatment. In addition, we investigated whether a multimodal approach with hyperthermia and resveratrol could abolish cisplatin resistance in tumour cells through the modulation of histone deacetylase (HDAC) activity and levels of heat shock proteins (HSP70/HSP90) and contribute to the direct toxicity of cisplatin on tumour cells. The tumour was induced by injecting 1 × 106 EAT cells subcutaneously (sc) into the thighs of Balb/c mice. The mice were treated with resveratrol per os for five consecutive days beginning on day 2 after tumour injection and/or by injecting cisplatin intraperitoneally (ip) at a dose of 2.5 mg/kg on days 10 and 12 and at a dose of 5 mg/kg on day 15. Immediately thereafter, the mice were exposed to systemic hyperthermia for 15 min at a temperature of 41 °C. The obtained results showed that the administration of resveratrol did not significantly contribute to the antitumour effect of cisplatin and hyperthermia, but it partially contributed to the immunomodulatory effect and to the reduction of cisplatin toxicity and to a slight increase in animal survival. This treatment schedule did not affect microvessel density, but it inhibited tumour growth and modulated macrophage polarization to the M1 phenotype. Furthermore, it abolished the resistance of tumour cells to cisplatin by modulating HDAC activity and the concentration of HSP70 and HSP90 chaperones, contributing to the increased lifespan of mice. However, the precise mechanism of the interaction between resveratrol, cisplatin, and hyperthermia needs to be investigated further.

[Effect of Wt P53 Gene Transfection on Sensitivity of Hyperthermia in C6 Glioma Cells].

Abstract:OBJECTIVE:To investigate the effect of wt p53 gene transfection on the sensitivity of hyperthermia in C6 glioma.METHODS:The recombinant eukaryotic expressive vector pCMV-p53 plasmids were extracted, identified and transmitted to C6 glioma cells by stable transfection. The biological characteristics were compared among C6/p53 (+) cells, thermo-treated C6/p53 (+) cells, thermo-treated C6 cells, thermotolerance C6 cells and control C6 cells. Transplanted glioma was treated by laser interstitial thermotherapy (LITT) in tumor-bearing nude mice.RESULT:The wt p53 gene segments were extracted and correctly identified by restriction enzyme (Hind and EcoR). The optimal transfection concentration of plasmid to liposome was 1:6, neo gene expressed stably in C6 cells transfected with positive and blank plasmid. Thermotolerance C6 cell growth was normal in vitro, growth of C6/p53 (+) cells was inhibited. Growth of hyperthermia treated C6 cells was inhibited significantly 12 h after thermal treatment, cell proliferation activity of C6/p53 (+) cells was suppressed significantly 6 h after hyperthermia treatment. Apoptosis was observed in C6/p53 (+), thermo-treat C6 and thermo-treat C6/p53 (+) cells. The apoptosis ratio of hyperthermia treated C6/p53(+) cells was increased by 30 folds compared with that of control groups. Study in nude mouse model demonstrated lower positive inoculation rate in groups of C6/p53 (+) cells (57 %), hyperthermia treated C6 cells (75 %) and hyperthermia treated C6/p53(+) cells (20 %). C6/p53(+) LITT group showed the highest antitumor effect, compared with C6 LITT group(P<0.05). However, thermotolerance C6 LITT group had the lowest effect.CONCLUSION:Transfection of wt p53 gene can increase the sensitivity of hyperthermia in glioma cells.

Abstract:The safe use of intraoperative blood salvage (IBS) in cancer surgery remains controversial. Here, we investigated the killing effect of cisplatin combined with hyperthermia on human hepatocarcinoma (HepG2) cells and erythrocytes from IBS in vitro. HepG2 cells were mixed with concentrated erythrocytes and pretreated with cisplatin (50, 100, and 200 μg/ml) alone at 37 °C for 60 min and cisplatin (25, 50, 100, and 200 μg/ml) combined with hyperthermia at 42 °C for 60 min. After pretreatment, the cell viability, colony formation and DNA metabolism in HepG2 and the Na(+)-K(+)-ATPase activity, 2,3-diphosphoglycerate (2,3-DPG) concentration, free hemoglobin (Hb) level, osmotic fragility, membrane phosphatidylserine externalization, and blood gas variables in erythrocytes were determined. Pretreatment with cisplatin (50, 100, and 200 μg/ml) combined with hyperthermia (42 °C) for 60 min significantly decreased HepG2 cell viability, and completely inhibited colony formation and DNA metabolism when the HepG2 cell concentration was 5×10(4) ml(-1) in the erythrocyte (P<0.01). Erythrocytic Na(+)-K(+)-ATPase activity, 2,3-DPG level, phosphatidylserine externalization, and extra-erythrocytic free Hb were significantly altered by hyperthermia plus high concentrations of cisplatin (100 and 200 μg/ml) (P<0.05), but not by hyperthermia plus 50 μg/ml cisplatin (P>0.05). In conclusion, pretreatment with cisplatin (50 μg/ml) combined with hyperthermia (42 °C) for 60 min effectively eliminated HepG2 cells from IBS but did not significantly affect erythrocytes in vitro.
Orthotopic Esophageal Cancers: Intraesophageal Hyperthermia-enhanced Direct Chemotherapy in Rats

Abstract:Purpose: To determine the feasibility of using intraesophageal radiofrequency (RF) hyperthermia to enhance local chemotherapy in a rat model with orthotopic esophageal squamous cancers.Materials and Methods: The animal protocol was approved by the institutional animal care and use committee and the institutional review board. Human esophageal squamous cancer cells were transduced with luciferase lentiviral particles. Cancer cells, mice with subcutaneous cancer esophageal xenografts, and nude rats with orthotopic esophageal cancers in four study groups of six animals per group were treated with (a) combination therapy of magnetic resonance imaging heating guidewire-mediated RF hyperthermia (42 degrees C) plus local chemotherapy (cisplatin and 5-fluorouracil), (b) chemotherapy alone, (c) RF hyperthermia alone, and (d) phosphate-buffered saline. Bioluminescent optical imaging and transcutaneous ultrasonographic imaging were used to observe bioluminescence signal and changes in tumor size among the groups over 2 weeks, which were correlated with subsequent histologic results. The nonparametric Mann-Whitney U test was used for comparisons of variables.Results: Compared with chemotherapy alone, RF hyperthermia alone, and phosphate-buffered saline, combination therapy with RF hyperthermia and chemotherapy induced the lowest cell proliferation (relative absorbance of formazan: 23.4% +/- 7, 44.6% +/- 7.5, 95.8% +/- 2, 100%, respectively; P < .0001), rendered the smallest relative tumor volume (0.65 mm(3) +/- 0. 15, P < .0001) and relative bioluminescence optical imaging photon signal (0. 57 x 10(7) photons per second per square millimeter +/- 0.15, P < .001) of mice with esophageal cancer xenografts, as well as the smallest relative tumor volume (0.68 mm(3) +/- 0.13, P < .05) and relative photon signal (0.56 X 10(7) photons per second per square millimeter +/- 0.11. P < .001) of rat orthotopic esophageal cancers.Conclusion: Intraesophageal RF hyperthermia can enhance the effect of chemotherapy on esophageal squamous cell cancers. (C) RSNA, 2016

Abstract:Objective To observe the effects and mechanisms of hyperthermia therapy combined with chemotherapy on multidrug-resistant human lung adenocarcinoma cell line A549/DDP, it will provide theory evidence in thermochemotherapy for clinic. Methods A549 or A549/DDP treated with different temperature were detected of sensitivity to cisplatin with CCK-8 assay. After hyperthermia, A549/DDP cell cycle was analyzed with flow cytometry and ERCC1mRNA expression was detected using RT-PCR. Results Parent human lung adenocarcinoma cell A549 was more sensitive to cisplatin than drug-resistant cell line A549/DDP. Treated with different temperature, drug sensitivity of A549/DDP to cisplatin all increased. Hyperthermia had significant effect on cell cycle with increase of G2/M phase population and reduction of G0/G1 phase population, and thermochemotherapy was more notable(P<0.01). After hyperthermia, the expression of ERCC1 mRNA did not change. Conclusion Hyperthermia can significantly enhance the sensitivity to cisplatin in human multidrug-resistant lung cancer cell line A549/DDP. Thermochemotherapy has synergistic effects of inhibiting cell proliferation. The major synergistic mechanisms of thermochemotherapy are that they can induce cell cycle arrest?inhibit cell proliferation ,but have no correlation with gene ERCC1.

Abstract:Objective:To investigate the effect and mechanism of hyperthermia on sensitivity of Navelbine. Methods: The working concentrations of NVB against human pulmonary adenocarcinoma cells(A549) were determined by MTT assay that has been developed for quantitative evaluation of the proliferation of cells. Then hyperthermia and chemotherapy were used singly or concurrently and the cell survival rates were obtained at 24h. The anticancer effect was evaluated by Veleriote method. The changes in cell cycle of cells and the assessment of apoptosis rates after treated for 42℃ hyperthermia and NVB were analysed by flow cytometry (FCM). Results: The concentration was defined as that of IC10-IC20 at 24h. It turned out to be 10ug/ml. Both single 42℃ hyperthermia and single chemotherapy suppressed the proliferation of A549 cells(P0.05); Combination of 42℃ hyperthermia and chemotherapy suppressed proliferation and killed more cells than single chemotherapy and single hyperthermia(P0.01).Flow cytometry showed that 42℃ hyperthermia resulted in progressive decrease in cells in S phase; Single chemical drug arrested cell cycle progression in G2/M phase; Compared with the group of single chemotherapy, the proportion of S phase cells of the group of combined treatment of 42℃ hyperthermia and chemotherapy decreased while the proportions of G2/M phase cells increased. The apoptosis rates of the four groups (control group, hyperthermia group, chemotherapy group and combination group)were 1.5%,7.9%,13.9% and 29.1% respectively. Conclusion: 42℃ hyperthermia can obviously enhance the cytotoxicity of NVB; The anti-tumor mechanism may be related to disturbance of cell cycle.
Comparison of Either Cisplatin or Oxaliplatin with Hyperthermia Against Human Gastric Cancer Cell Line BGC-823

Abstract:Objective:To compare the anticancer effect of cisplatin or oxaliplatin combined with hyperthermia against human undifferentiated gastric adenocarcinoma cell line BGC-823. Methods:The working concentrations of cisplatin and oxaliplatin against BGC-823 were determined by MTT assay that has been developed for quantitative evaluation of the proliferation of cells. Then the chemotherapy or hyperthermia, singly or concurrently, were used and the cell survival rates were obtained. The anticancer effect was evaluated and compared by Veleriote method at 24 h or 48 h. Results:The working concentrations were defined as the concentration causing 20%～30% inhibition rate. They turned out to be 1μg/ml and 5μg/ml of cisplatin and oxaliplatin, respectively. Exposing to 43℃ hyperthermia for 1 hour did not show obvious inhibition to BGC-823 at 24 h or 48 h(P0.05). The chemotherapies with cisplatin 1μg/ml or oxaliplatin 5μg/ml did not exhibit obvious inhibition to BGC-823 at 24 h (P0.05), but they did at 48 h(P 0.01). The combination of cisplatin or oxaliplatin with 43℃ hyperthermia for 1 hour showed obvious inhibition to BGC-823 at 24 h and 48 h.(P0.05). And the effect of chemohyperthermia was amplified as the time prolonged(P0.05). There was no difference between the two drugs at 24 h and 48 h when used singly. The inhibitory effect of cisplatin was much higher than that of the oxaliplatin when used with hyperthermia. Judged with the Veleriote method at 24 h and 48 h, there was obvious synergism of cisplatin 1μg/ml combined with hyperthermia. Oxaliplatin combined with hyperthermia exhibited subaddictive effect at 24 h and synergism at 48 h. Conclusion:The combination of cisplatin or oxaliplatin and 43℃ hyperthermia for 1 hour!showed subaddictive effect or synergism to the inhibition of human undifferentiated gastric adenocarcinoma cell line BGC-823,and cisplatin is much better than oxaliplatin when combined with hyperthermia.

Abstract:Background:To investigate the underlying molecular mechanisms of radiofrequency hyperthermia (RFH)-enhanced direct chemotherapy of pancreatic cancers.Method:Rat ductal PaCa cell line DSL-6A/C1 and orthotopic pancreatic cancers of Lewis rats were divided into four study groups with various treatments: i) phosphate-buffered saline (PBS) as a control; ii) RFH alone; iii) intratumoral chemotherapy alone (gemcitabine); and (iv) combination therapy of gemcitabine plus intratumoral RFH at 42 °C for 30 min. In the in-vitro confirmation experiments, the viability and apoptosis of DSL-6A/C1 cells in each treatment group were evaluated using cell live/dead staining, flow cytometry, and Western blot. In the in vivo validation experiments, related proteins were evaluated by immunohistochemistry (IHC) staining of tumors.Results:Of the in-vitro experiments, the lowest cell viability and more apoptotic cells were shown in the group with combination therapy compared to other treatments. Western blot data showed elevated Bax/Bcl-2, Caspase-3, and HSP70 expressions in DSL cells with combination therapy, compared to other treatments. Of the in vivo experiments, IHC staining detected the significantly increased expressions of HSP70, IL-1β, TNF-ɑ, Bax, and Caspase-3 in pancreatic cancer tissues of the animal group treated by combination therapy of gemcitabine with RFH.Conclusion:Molecular imaging-guided interventional RFH can significantly enhance the chemotherapeutic effect on pancreatic cancers via potential molecular mechanisms of up-regulating Bax/caspase-3-dependent apoptosis pathways.

Abstract:OBJECTIVE:To observe the cytotoxic effect of thermo-chemotherapy with cisplatin on osteosarcoma OS-732 cell line and to explore its mechanism.METHODS:The osteosarcoma OS-732 cell line was treated with different temperature, cisplatin alone and 43 degrees C +cisplatin for 1 h, respectively and the in vitro cytotoxic effect was observed with MTT assay. The cell cycle and the apoptotic rate were analyzed with flow cytometry (FCM); the cellular apoptosis was observed also with electron microscope.RESULT:The cytotoxicity index increase markedly as the temperature elevated or the concentration of cisplatin increased. While treated with 43 degrees C hyperthermia and cisplatin simultaneously, the cytotoxicity index increased to 72.37%;the cell cycle of the treated OS-732 cells line was changed with marked increase in S phase and decreasing in G(2)/M phase. The apoptotic rate increased markedly with the highest of 56.47%. Electron microscope showed the characteristic apoptotic alterations.CONCLUSION:Hyperthermia with cisplatin enhance cytotoxicity on osteosarcoma OS-732 cell line and the induced cell apoptosis may be one of the mechanisms of enhanced cytotoxicity by thermo-chemotherapy.

Abstract:Gemcitabine (2′,2′-difluoro-2′-deoxycytidine;dFdC) is a first-line chemotherapy drug for pancreatic cancer. Recently, a synergistic anti-tumor treatment of dFdC and hyperthermia has achieved good clinical results, but there are few reports on the molecular mechanism influenced by hyperthermia. This study is an initial exploration of the effects of hyperthermia on changes in the concentration of dFdC and its metabolites in pancreatic cancer cells. The aim is to provide a theoretical basis for clinical detection and pharmacokinetic research. PANC-1 cells at logarithmic growth phase were used as the experimental object. The MTT assay was performed to determine the half maximal inhibitory concentration (IC50) of dFdC. After PANC-1 cells were cultured in DMEM medium containing IC50dFdC and treated with hyperthermia at 41 °C or 43 °C, changes in the concentration of dFdC, 2′,2′-difluorodeoxyuridine (dFdU) and difluorodeoxycytidine triphosphate (dFdCTP) in the cells were tested using an optimized reverse phase high-performance liquid chromatography (RP-HPLC) protocol. We found that 41 °C and 43 °Chyperthermia gave rise to a decrease in dFdC and dFdU content. At 41 °C, the levels respectively fell to 9.28 and 30.93% of the baseline, and at 43 °C, to 24.76 and 57.80%, respectively. The dFdCTP content increased by 21.82% at 41 °C and 42.42% at 43 °C. The two heat treatments could alter the mechanism of dFdC metabolism in PANC-1 cells. The effect of 43 °C hyperthermia is more significant. Our observations may be instrumental to explaining the higher anti-tumor efficacy of this combination therapy.
