Abstract:The intestinal digestive enzyme alpha-glucosidase plays a vital role in the carbohydrate digestion. One anti-diabetic therapeutic approach reduces the postprandial glucose level by the inhibition of the alpha-glucosidase enzyme. The present study aimed to investigate the anti-diabetic activity of extract of Ailanthus altissima bark, in vitro. Three different plant extracts were formulated by using distilled water, methanol, and a mixture of methanol and water (70:30) to compare the best effect of the extract, while distilled water and metformin were used as negative and positive controls. Four different concentrations of plant extract in all solvents were used to find percentage inhibition of alpha-glucosidase. Maximum percentage inhibition was examined against 1.5 mL of all solvent extracts but aqueous methanolic plant extract showed maximum73% inhibition that was comparable to standard drug inhibition (80%). One-way ANOVA was followed for statistical analysis to find out the P-value based on which most significant and insignificant were declared. Aqueous methanolic plant extract showed the most significant results because the value of p 0.05. The assay results suggest that the presence of bioactive compounds are responsible for the versatile medicinal properties against diabetes, the aqueous methanolic extract exhibit a dose-dependent increase in inhibitory effect on alpha-glucosidase enzyme up to 73%. The current study proves aqueous methanolic extract of A. altissima bark has anti-diabetic activity in vitro and is a strong candidate for postprandial control of blood glucose level.

Abstract:We hypothesized that some medicinal herbs and food plants commonly used in the management of diabetes can reduce glucose peaks by inhibiting key carbohydrate hydrolyzing enzymes. To this effect, extracts of Antidesma madagascariense (AM), Erythroxylum macrocarpum (EM), Pittosporum senacia (PS), and Faujasiopsis flexuosa (FF), Momordica charantia (MC), and Ocimum tenuiflorum (OT) were evaluated for α-amylase and α-glucosidase inhibitory effects based on starch-iodine colour changes and PNP-G as substrate, respectively. Only FF and AM extracts/fractions were found to inhibit α-amylase activity significantly ( P < 0.05 ) and coparable to the drug acarbose. Amylase bioassay on isolated mouse plasma confirmed the inhibitory potential of AM and FF extracts with the ethyl acetate fraction of FF being more potent ( P < 0.05 ) than acarbose. Extracts/fractions of AM and MC were found to inhibit significantly ( P < 0.05 ) α-glucosidase activity, with IC 50 comparable to the drug 1-deoxynojirimycin. In vivo studies on glycogen-loaded mice showed significant ( P < 0.05 ) depressive effect on elevation of postprandial blood glucose following ingestion of AM and MC extracts. Our findings tend to provide a possible explanation for the hypoglycemic action of MC fruits and AM leaf extracts as alternative nutritional therapy in the management of diabetes.
Attenuation of Helicteres isora L. bark extracts on streptozotocin-induced alterations in glycogen and carbohydrate metabolism in albino rats

Abstract:The present study was undertaken to assess the effect of Helicteres isora L. on four important enzymes of carbohydrate metabolism (glucokinase [GK], hexokinase [HK] phosphofructokinase [PFK] and fructose-1, 6-bisphosphatase [FBP]) along with glycogen content of insulin-dependent (skeletal muscle and liver) and insulin-independent tissues (kidneys and brain) in streptozotocin (STZ; 60 mg/kg)-induced model of diabetes for 30 days. Administration of bark extracts (100, 200 mg/kg) for 30 days led to decrease in plasma glucose levels by approximately 9.60% and 22.04% and 19.18% and 33.93% on 15th and 30th day, respectively, of the experiment. Liver and two-kidney weight expressed as percentage of body weight significantly increased in diabetics (P < 0.05) versus normal controls. Renal glycogen content increased by 10 folds while hepatic and skeletal muscle glycogen content decreased by 75% and 68% in diabetic controls versus controls. H. isora did not affect glycogen content in any tissue. The decreased activities of PFK, GK, FBP and HK in diabetic controls were 40%, 50%, 50% and 60% and bark extract of H. isora partially corrected this alteration. The efficacy of the bark extract was comparable with Tolbutamide, a well-known hypoglycemic drug.

Abstract:The objective of the present investigation was to study the antidiabetic and diuretic potential of Anogeissus latifolia (A. latifolia) bark in experimental rats. The A. latifolia bark was extracted with hydro-alcoholic solvent by cold extraction method. Acute toxicity study was performed according to OECD 425 guidelines for hydro-alcoholic extracts of A. latifolia bark (ALBE). The dose of 150 mg/kg p.o. and 300 mg/kg, p.o. of ALBE was selected for further studies. Animals were prepared diabetic by administration of alloxan (120 mg/kg, i.p.). The albino rats were divided in to five groups for oral glucose tolerance test (OGTT) and alloxan induced anti diabetic model with six animals in each group. Diabetic animals were treated with hydro-alcoholic extract of A. latifolia bark for 20 days. The blood glucose level was estimated according to standard procedures. Diuretic activity hydro-alcoholic extracts of A. latifolia was evaluated by Lipshitz method. The result shows that hydro-alcoholic extract from bark of Anogeissus latifolia 300 mg/kg (ALBE-II) shown significant hypoglycemic activity as compared to glibenclamide and diabetic group. The ALBE does not exhibit significant diuretic activity which is considered as positive marker in diabetic phenomena. Hence in present study extract of A. latifolia posses antidiabetic activity. This study may be benchmark in future to use of this drug scientifically.

Abstract:α-amylase inhibitors present in pancreatic region has an operative strategy by controlling the breakdown of starch and helps to minimize the post-prandial hyperglycemia levels. In this study, vegetative (leaf) part of herbal plant Erythrina subumbrans (Hassk.) was assessed for anti-diabetic activity. Aqueous ethanol (80 %) extract was prepared in the different concentration (10, 20, 40, 80, 160, and 320 μg/ml). Acarbose was used as a standard and treated in similar way as that of sample. Control samples were also prepared without standard and sample solutions. A known volume of α-amylase solution was added (0.1mg/mL) was added to standard, sample, control solutions which were preincubated at 37 °C for 15 minutes. Further, known volume of starch solution was added and incubated for 60 min to initiate the reaction. Hydrochloric acid (HCl) and iodine reagent was added to the test tubes and absorbance was measured at 580 nm in UV-Vis spectrophotometer. A strong pancreatic amylase inhibitory activity (>50 %) was obtained from aqueous ethanolic extract with IC50 (half maximal inhibitory concentration) value of 23 μg/ml against standard acarbose with IC50 value of 27 μg/ml. The values endorse Erythrina subumbrans (Hassk.) for further experiments on their potential for managing Diabetes.

Abstract:In many reaction-diffusion processes, ranging from biochemical networks, catalysis, to complex self-assembly, the spatial distribution of the reactants and the stochastic character of their interactions are crucial for the macroscopic behavior. The recently developed mesoscopic Green's Function Reaction Dynamics (GFRD) method enables efficient simulation at the particle level provided the microscopic dynamics can be integrated out. Yet, many processes exhibit non-trivial microscopic dynamics that can qualitatively change the macroscopic behavior, calling for an atomistic, microscopic description. We propose a novel approach that combines GFRD for simulating the system at the mesoscopic scale where particles are far apart, with a microscopic technique such as Langevin dynamics or Molecular Dynamics (MD), for simulating the system at the microscopic scale where reactants are in close proximity. This scheme defines the regions where the particles are close together and simulated with high microscopic resolution and those where they are far apart and simulated with lower mesoscopic resolution, adaptively on the fly. The new multi-scale scheme, called MD-GFRD, is generic and can be used to efficiently simulate reaction-diffusion systems at the particle level.

Abstract:Withania frutescens L. is a wild perennial woody plant used by the local population for diverse therapeutic purposes. This work aims to study for the first time the potential inhibitory effect of this plant hydroethanolic extract on α-amylase and α-glucosidase activities using in vitro methods and its antidiabetic and antihyperglycemic activities using alloxan-induced diabetic mice as a model for experimental diabetes. Two doses were selected for the in vivo study (200 and 400 mg/kg) and glibenclamide, a well-known antidiabetic drug (positive control) in a subacute study (28 days) where the antihyperglycemic activity was also assessed over a period of 12 h on diabetic mice. The continuous treatment of diabetic mice with the extract of Withania frutescens for 4 weeks succeeded to slowly manage their high fasting blood glucose levels (after two weeks), while the antihyperglycemic test result revealed that the extract of this plant did not control hyperglycemia in the short term. No toxicity signs or death were noted for the groups treated with the plant extract, and it shows a protective effect on the liver and kidney. The in vitro assays demonstrated that the inhibition of alpha-amylase and alpha-glucosidase might be one of the mechanisms of action exhibited by the extract of this plant to control and prevent postprandial hyperglycemia. This work indicates that W. frutescens have an important long term antidiabetic effect that can be well established to treat diabetes.

Abstract:Background: Diabetes mellitus is a metabolic disorder associated with relative or absolute insulin deficiency or resistance, characterized by hyperglycemia. Modern prescriptions such as pioglitazone have better therapeutic potential, but its side effects and financial burden for developing countries have motivated the researchers to find alternative natural drugs to compete hyperglycemia in patients with diabetes. The present study was conducted to explore the therapeutic potential of selected medicinal plants for the treatment of diabetes as an alternative to allopathic medicines. Method: In present study, hydroalcoholic extracts of Curcuma longa, Lavandula stoechas, Aegle marmelos, and Glycyrrhiza glabra and their polyherbal preparation (PHP) as compound drug were investigated for their antihyperglycemic potential in alloxan-induced diabetic mice. The study subjects (mice) were divided into different groups as normal control, diabetic control, pioglitazone treated (standard drug), test groups (plant extract treated 50, 100, and 150 mg/kg body weight), and PHP-treated group. Blood glucose concentration of all the study animals was determined by Glucose strip test. Qualitative phytochemical analysis of all the plant extracts was also performed following standard methods. Result: It was investigated that treatment of alloxan-induced diabetic mice with hydroalcoholic extracts of studied medicinal plants showed significant (P < .05) effects on fasting blood glucose levels (from baseline to normal range) in a manner comparable to that of the reference drug, pioglitazone (1 mg/kg body weight intraperitoneal). The tested plant extracts significantly (P < .05) reduced the glucose concentration in blood of diabetes-induced mice in a dose-dependent manner. Conclusion: It could be concluded that studied medicinal plants have antihyperglycemic activity. The study findings favor the use of traditional herbal medicinal practices for the management of diabetes that might due to the presence of bioactive phytoconstituents in plants. However, larger studies are required to identify, isolate, and characterize the bioactive phytoconstituents responsible for antihyperglycemic activity of studied medicinal plants.

Abstract:Using HPLC-PDA and HRMS analysis, five compounds p -coumaric acid, sinapic acid, quercetin, trans -ferulic and gallic acid were identified in seeds of Amomum dealbatum Roxb. The GC-MS analysis identified 1-dodecanol, phenol, 3,5-bis(1,1-dimethylethyl), Oleic Acid and 1-Heptacosanol which possess anti-diabetic properpties. A bioassay-guided technique was used to determine the degree of inhibition that A. dealbatum seeds crude methanol extract and its most active sub-fraction had against the α-glucosidase and Helicobacter pylori urease enzymes. In the Rat L6 myoblast cell line, glucose absorption through the GLUT4 transporter of most active subfraction (EASF80) was examined. According to a molecular docking investigation, these compounds strongly interacted with the GLUT4 transporter, H pylori and α-glucosidase enzyme. Sinapic acid interacted most strongly with the H. pylori urease enzyme while gallic acid interacted with both the α-glucosidase enzyme and the GLUT4 transporter. Additionally, a molecular docking simulation study was carried out to recognise the stability of the complexes.

Abstract:Alpha amylases cleave the starch like molecules and led to increase in blood sugar level in diabetic patients then in order to manage it the inhibition action is carried out by their inhibitors. The α-amylase inhibitors catalyses the process of endo-hydrolysis of α (1→ 4) glycosidic linkage present in starch which ultimately led to decrease in the starch hydrolysis process. It also affects the carbohydrate absorption in the intestine. In 1945, the first alpha amylase inhibitor was reported from beans. It has already been isolated from various plant species namely, Hibiscus sabdariffa.L, Geranium pratense L and Cedrus libani A. Rich with inhibition percentage of 100, 43 and 31 respectively. The studies have already been reported on inhibitors and their interactions with human pancreatic alpha amylases are made .In-vitro and in vivo tests has already been done by various researchers which showed a significant drop in blood sugar level of a normoglycemic mice after an extract from the leaves of Bersama abbyssinica was tested for its potential effect as anti-diabetic.

Abstract:Abstract Background α-Amylase catalyses the hydrolysis of starch and ultimately producing glucose. Controlling the catalytic activity of this enzyme reduces glucose production in the postprandial stage, which could be a therapeutic benefit for people with diabetes. This study was conducted to evaluate α-amylase inhibition for utilizing the crude extracts of some medicinal plants traditionally used in Nepal for the treatment of diabetes and its related complications. Methods Microtiter plate approach has been used to assess inhibitory activities of in vitro α-amylase of methanolic extracts of thirty-two medicinal plants. A starch tolerance test was used in rats to investigate the in vivo study of the methanolic extract concerning glibenclamide as the positive control. Results Acacia catechu , Dioscorea bulbifera , and Swertia chirata exhibited inhibitory activity against α-amylase and with IC 50 values; 49.9, 296.1, and 413.5 μg/mL, respectively. Kinetics study revealed that all the extracts displayed a mixed type of inhibition pattern, with K i values ranging from 26.6–204.2 μg/mL. Free radical scavenging activity was again re-examined and found prominent in extracts of A. catechu . Likewise, A. catechu and S. chirata showed significant reduction of blood glucose concentration up to 30 min after oral dose of 250 mg/kg (F (4, 20) = 4.1, p = .048), and (F (4, 20) = 4.1, p = .036), respectively. Conclusions Enzymatic assay for α-amylase inhibition using extracts was successfully evaluated. Also, the in-vitro and in-vivo study model revealed that medicinal plants could be a potent source of α-amylase inhibition. So, they could serve as potential candidates for future drug development strategies for curing diabetes with minimal or no adverse side effects.

Abstract:Ethnopharmacological relevance Hodgsonia heteroclita has been known as an important traditionally consumed medicinal plant of North-East India known to have antidiabetic properties. This study aims to investigate the effects of the ethanolic fruit extract of Hodgsonia heteroclita against hyperglycemia and hyperlipidemia by using streptozotocin (STZ) treated diabetic mice. Materials and methods The fruits of H. heteroclita were collected from the various parts of Kokrajhar district, Assam India (Geographic coordinates: 26°24′3.85′′ N 90°16′22.30′′ E). Basic morphological evaluations were carried out by the Botanical Survey of India, Eastern circle, Shillong, who also certified and identified the plant. Hexane, chloroform, and ethanolic extracts of the fruit of H. heteroclita were investigated for α-amylase inhibition assay as a rapid screening tool for examining anti-diabetic activity. The efficacy of ethanolic extract at a dose of 100, 200, and 300 mg/kg body weight was tested for 21 days in STZ-induced diabetic mice. The body weight, fasting plasma glucose and serum lipids, and hepatic glycogen levels were measured in experimental animals to examine the antihyperglycemic and antihyperlipidemic efficacy of the extract. Both HPTLC and LC-MS analysis was performed to examine the phyotochemicals present in the ethanolic extract of H. heteroclita . Results It has been observed that treatment with the ethanolic extract dose-dependently reduced the plasma glucose levels, total cholesterol, low density lipoprotein-cholesterol, very low-density lipoprotein-cholesterol, triglyceride, and increased the body weight, liver glycogens and high-density lipoprotein-cholesterol in STZ treated diabetic mice. HPTLC demonstrated the presence of triterpene compounds and LC-MS analysis revealed the presence Cucurbitacin I, Cucurbitacin E, and Kuguacin G as the triterpene phytoconstituents. Conclusion The present study demonstrated that ethanolic fruit extract of H. heteroclita improved both glycemic and lipid parameters in mice model of diabetes.

Biological and Hypoglycemic Effects of a Polyherbal Extract on Alloxanized Diabetic Rats

Abstract:The current study investigates the antioxidant, antidiabetic, hepatoprotective, and nephroprotective potentials of a polyherbal mixture containing the methanolic extracts of seeds from Nigella sativa, Cicer arietinum, Silybum marianum, and Citrullus colocynthis and the rhizome of Zingiber officinale. The polyherbal extract (PHE) showed significant total phenolic contents (187.17 GAE/g), ferric reducing power (28%), and radical-scavenging activity (86.16%). The PHE also showed a substantial hypoglycemic effect in alloxan-induced diabetic rats by reducing the blood glucose level of the PHE-treated rats (-48.64%) and increasing the insulin level (107.5%) as compared with the diabetic control group. Likewise, an increase in high-density lipoprotein (HDL) contents (22.95%) with an associated decrease in low-density lipoprotein (LDL) levels (-43.93%) was also noted. A significant decrease in serum levels of liver marker enzymes, e.g., SGPT (-36%), SGOT (-31%), and serum ALP (-12%), was also observed as compared with the standard drug-treated group. Based on the findings of the study, it may be suggested that PHE helps ameliorate the severity of diabetes as a herbal remedy and might be employed in nutra-pharmaceuticals, replacing synthetic antidiabetic compounds.

Abstract:BACKGROUND: Carbohydrate digestive enzymes play a major role in the management of the postprandial hyperglycemia. A chronic hyperglycemia can lead to serious health problems due to excessive production of several reactive oxygen species. Therefore, the inhibition of carbohydrate digestive enzyme and the use of antioxidant natural product can be an important strategy to control the glycaemia level and prevent against the complication of diabetes.AIM: The study aims to perform a phytochemical analysis, antioxidant activity, inhibitory effect on α -amylase, α -glucosidase (in vitro and in vivo) and the intestinal glucose absorption in Wistar rats of Artemisia campestris aqueous extract (AcAE) and hydro-ethanolic extract (AcEE).RESULTS: The test of total phenolic content, show that the AcAE has the highest quantity of polyphenol (44.65 ± 0.54 μ g GAE/mg extract) compared to the AcEE (31.7 ± 0.53 μ g GAE/mg extract) significantly. The amount of flavonoid and condensed tannins content in AcAE is 24.41 ± 3.57 μ g QrE/mg extract, 14.31 ± 5.26 μ g CE/mg respectively. The AcAE has also exhibit a great antioxidant activity in DPPH-scavenging and Ferric reducing antioxidant power assay (FRAP) compared to AcEE with an IC 50 = 0.355 ± 0.057 mg/mL and IC 50 = 0.269 ± 0.025 mg/mL. However, in a β -carotene bleaching assay the AcEE has the highest effect with an IC 50 = 0.319 ± 0.097 mg/mL. The both extract of Artemisia campestris L. (250 mg/kg) decreased postprandial hyperglycemia in the normal and alloxane diabetic rats in a very significant manner after starch or sucrose administration as an α -amylase and α -glucosidase substrate respectively. This result is confirmed in vitro by a remarkable inhibitory effect on α -amylase digestive enzymes by an IC 50 = 1.259 ± 0.128 mg/mL and IC 50 = 0.602 ± 0.072 mg/mL receptively for AcAE and AcEE. For the α -glucosidase enzyme, the both extracts significantly inhibit α -glucosidase activity compared to the control and they are almost similar to each other. Using a jejunum perfusion technique (in situ), Artemisia campestris L. decrease the intestinal D-glucose absorption activity significantly compared to the control and comparable to the Phlorizin used as a positive control by an amount of glucose absorbed equal a 6.53 ± 0.57, 5.34 ± 0.64 and 4.71 ± 0.24 mg/10 cm/h, for AcAE, AcEE and Phlorizin respectively.CONCLUSIONS: These results showed that the Artemisia campestris L. has highest phenolic content, antioxidant activity and demonstrated a postprandial anti-hyperglycemic effect via the inhibiting of the carbohydrate digestive enzyme ( α -amylase and α -glucosidase) and the intestinal glucose absorption.

Abstract:AIM OF THE STUDY: The present study was designed to investigate the effects of the EtOH and H(2)O extracts of Selaginella tamariscina (Beauv.) Spring on hyperglycemia in diabetic rats and HepG2 cells, and to confirm the active fractions of EtOH extract in HepG2 cells.MATERIALS AND METHODS: HepG2 cells and type II diabetic rats induced by low-dose streptozotocin (STZ) and high-fat diet (HFD) were used to evaluate the hypoglycemic effect of EtOH and H(2)O extracts of Selaginella tamariscina. HepG2 cells were used to evaluate the promotive effect of different fractions of EtOH extract obtained from a polyamide column on glucose utilization.RESULTS: The results in HepG2 cells indicated that the EtOH extract had a better hypoglycemic effect than the H(2)O extract. The results in diabetic rats indicated that both EtOH extract and H(2)O extract were able to ameliorate the fasting blood glucose (FBG) level and improve oral glucose tolerance (OGTT). Total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-c), free fatty acids (FFA), tumor necrosis factor-α (TNF-α), alanine aminotransferase (ALT), aspartate aminotransferase (AST), blood urea nitrogen (BUN) and malondialdehyde (MDA) levels in serum were lowered. High density lipoprotein (HDL-c), insulin and superoxide dismutase (SOD) levels in serum were elevated as well as the hepatic glycogen content in diabetic rats. Compared with H(2)O extract, the effects of EtOH extract were more marked. The 80% ethanol fraction exhibited a stronger hypoglycemic effect than the aqueous and 50% ethanol fractions, but the 95% ethanol fraction did not show any appreciable effects in HepG2 cells.CONCLUSIONS: The results suggested that the EtOH extract had a better hypoglycemic effect than the H(2)O extract; the 80% ethanol fraction from polyamide column had a strong hypoglycemic activity in HepG2 cells.

Abstract:Zingiber officinale (ginger,II), Citrullus colocynthis (CC, III) and Anethum graveolens (dill, IV) are functional medicinal plants containing beneficial compounds and some evidence exists on their anti-diabetic, anti-inflammatory and hypolipidemic activities. The objective of this study was to evaluate the antihyperglycemic and antihyperlipidemic potential of various extracts of these plants with different polarities in experiments on streptozotocin-induced diabetic rats. Test animals (n = 112) were randomly divided into control, diabetic, diabetic+chloroform extract, diabetic+carbon tetrachloride extract, diabetic+hydroalcoholic extract, and diabetic+aqueous extract groups. Rats were made diabetic by a single injection of streptozotocin (STZ, 60 mg/kg). The plant extracts were daily administered (100 mg/kg, i.p.) for 3 weeks. Serum glucose and lipid levels were measured at different weeks. All extracts of ginger and dill as well as carbon tetrachloride and hydroalcoholic extracts of CC significantly reduced serum glucose level in diabetic groups. In addition, some plants extracts approppriately and significantly improved serum lipid levels including triglyceride, total cholesterol, HDLcholesterol, and LDL cholesterol in diabetic rats. According to extract polarities and components, all ginger extracts exerted hypoglycemic effect, however, hydroalcoholic extract showed the best potential to appropriately modify serum lipids in diabetic condition. In addition, hydroalcoholic extracts of C. colocynthis and A. graveolens had the best anti-diabetic potential which could be related to better extraction of bioactive components by polar or non-polar organic and aqueous solvents from these medicinal herbs.

Abstract:Alpha-glucosidase (maltase, sucrase, isomaltase and glucoamylase) activities which are involved in carbohydrate metabolism are present in human intestinal maltase-glucoamylase (MGAM) and sucrase-isomaltase (SI). Hence, these proteins are important targets to identify drugs against postprandial hyperglycemia thereby for diabetes. To find natural-based drugs against MGAM and SI, Artocarpus heterophyllus leaf was explored for MGAM and SI inhibition in in vitro and in silico. A. heterophyllus leaf aqueous active fraction (AHL-AAF) was prepared using Soxhlet extraction followed by silica column chromatography. The phytoconstituents of AHL-AAF were determined using LC-MS/MS. AHL-AAF showed dose-dependent and mixed inhibition against maltase (IC50 = 460 μg/ml; Ki = 300 μg/ml), glucoamylase (IC50 = 780 μg/ml; Ki = 480 μg/ml), sucrase (IC50 = 900 μg/ml, Ki = 504 μg/ml) and isomaltase (IC50 = 860 μg/ml, Ki = 400 μg/ml). AHL-AAF phytoconstituents interaction with N-terminal (Nt) and C-terminal (Ct) subunits of human MGAM and SI was analyzed using induced-fit docking, molecular dynamics (MD), and free energy calculation. In docking studies, rhamnosyl hexosyl methyl quercetin (RHMQ), P-coumaryl-O-16-hydroxy palmitic acid (PCHP), and spirostanol interacted with active site amino acids of human MGAM and SI. Among these RHMQ stably interacted with all the subunits (Nt-MGAM, Ct-MGAM, Nt-SI and Ct-SI) whereas PCHP with Ct-MGAM and Nt-SI during MD analysis. In molecular docking, the docking score of RHMQ with NtMGAM, CtMGAM, NtSI and CtSI was -8.48, -12.88, -11.98 and -11.37 kcal/mol. The docking score of PCHP for CtMGAM and NtSI was -8.59 and -8.4 kcal/mol, respectively. After MD simulation the root mean square deviation (RMSD) and root mean square fluctuation (RMSF) values further confirmed the stable protein-ligand interaction. The RMSD value of all the complexes were around 2.5 Å and the corresponding RMSF values were also quite low. In MM/GBSA analysis, the involvement of Van der Waals and lipophilic energy in the protein/ligand interactions are understood. Further binding free energy for Nt-MGAM-PCHP, Nt-MGAM-RHMQ, Nt-SI-PCHP, Nt-SI-RHMQ, Ct-MGAM-PCHP, Ct-MGAM-RHMQ and Ct-SI-RHMQ complexes was found to be -24.94, -46.60, -46.56, -44.48, -40.3, -41.86 and -19.39 kcal/mol, respectively. Altogether, AHL-AAF showed inhibition of α-glucosidase activities of MGAM and SI. AHL-AAF could be further studied for its effect on diabetes in in vivo.

Abstract:The inhibition of carbohydrate hydrolyzing enzymes such as α-amylase can be an important strategy in the postprandial blood glucose level in patients with type II diabetes, plants contains different chemical constituents with potential for inhibition of α-amylase and hence may be used as therapeutic. Nine types of plants include (Clove, Lemon, Chamomile, Radish, Ginger, Black seed, pomegranate, Beetroot and Garlic) were used in this study to select the optimum plant material that inhibited α- amylase enzyme. Raphanus raphanistrum was chosen, it had the highest inhibition activity of the enzyme (95.5%). Also sodium phosphate buffer (0.2 M, pH 7.5) was selected as a best extraction buffer of plants inhibitor with inhibition activity 83%. The optimum extraction ratio represented by 1:20 (w:v) after 90 min, it was given 96.8% enzyme inhibition activity. Antioxidant activities of plants were performed using DPPH free radical scavenging assay, and lemon had a highest by 94.4%. This confirms that the plant might protect cells from oxidative damage, resulting in certain diseases. The antimicrobial properties of lemon, garlic, clove have been proven the highest effective against selected human pathogens such as Staphylococcus aureus, E. coli and Enterococcus sp. Also total phenolic content (TPC) of lemon and garlic were estimated spectrophotometrically according to the Folin-Ciocalteu procedure and calculated as gallic acid equivalents (GAE), lemon revealed the highest value compared with garlic.
