Abstract:Indole-3-acetic acid (IAA) is an important plant growth promoting hormone produced as secondary metabolite by many microorganisms. The current study aims to optimize conditions for IAA production by a rhizobacterial isolate and evaluation of its effect on plant growth. S even bacterial isolated were purified from the rhizosphere of pear-tree and screened for IAA production and optimum production medium. The most potent bacterial isolate (S5) was selected for further investigations and molecularly identified using (16S rRNA). The selected isolate was identified as Bacillus velezensis strain MOST-IAA and given the accession number (ON792392.1) in National Center for Biotechnology Information (NCBI). For maximum IAA productivity, twenty one nutritional factors were screened for their significance on IAA using Placket Burman design and the significant factors were used for statistical modeling of the production conditions using Box-Behnken design. The produced model and optimization resulted in maximum IAA (56.60 μg/ml) at: 4.41 g/l, D(+) mannose; 8.66 g/l, mannitol; 0.93 g/l, tryptophan; 5.88 g/l, potassium nitrate; 1.29 g/l, ammonium chloride; 28.5 °C, temperature and 7.69, initial pH with correlation of 94.76% to the theoretical results. The produced IAA has been extracted and used in irrigation of barley seed for 10 days. Data showed significant differences among the test and control barley, especially in the length of the produced roots. Bacillus velezensis can be used to increase the soil fertility or biotechnologically used for production of IAA to be applied for plant growth improvement.

Abstract:The ability to synthesize Indole-3-acetic acid (IAA) is widely associated with the plant growth promoting rhizobacteria (PGPR). The present work deals with isolation and characterization of such bacteria from the rhizosphere of medicinal plant Stevia rebaudiana and optimization of IAA production from its isolates. The optimization of IAA production was carried out at different pH and temperature with varied carbon and nitrogen sources of culture media. Out of different isolates obtained, three of them were screened as efficient PGPRs on the basis of different plant growth promoting attributes. Isolates CA1001 and CA2004 showed better production of IAA at pH 9 (91.7 µg ml-1) and at temperature 37 °C (81.7 µg ml-1). Dextrose (1%) was found to be the best carbon source for isolate CA1001 with 104 µg ml-1 IAA production. Isolate CA 2004 showed best production of IAA 36 µg ml-1 and 34 µg ml-1 at 1.5% and 1% Beef extract as nitrogen source respectively. Isolate CA 1001 showed 32 µg ml-1 IAA production at 0.5% nicotinic acid concentration. From the current study, CA1001 and CA2004 emerged as noble alternatives for IAA production further which also resulted in root and shoot biomass generation in crop plants, hence can be further used as bio-inoculants for plant growth promotion.

Abstract:Effect of indole-3-acetic acid (IAA) on economic characters of bivoltine pure races, Baiyu and Qinfeng were investigated. Fortification of mulberry leaf with indole-3-acetic acid (IAA) has proved to be successful in improving the crop yield. The increase in economic characters suggests that IAA has stimulatory effect on cocoon yield, silk quality and fecundity rate. Hence IAA can be utilized in sericulture for the improvement of yield and has practical advantage in grainages and reeling industry.
Optimization of indole acetic acid produced by plant growth promoting fungus, aided by response surface methodology

Abstract:Indole acetic acid (IAA) is one of the prime communicator playing a chief role in the interaction between host plant and endophytes. IAA produced by the endophytes primarily contributes to plant growth and development. Here, we optimized IAA production by an endophytic fungus Diaporthe terebinthifolli GG3F6 isolated from the asymptomatic rhizome of Glycyrrhiza glabra employing response surface methodology (RSM) and exploring its effect on the host plant biology. The methodology revealed 1.1 fold increases in IAA accumulation. The maximum IAA (121.20 μg/mL) was achieved using tryptophan substrate (1 mg/mL) in Potato dextrose broth (48 g/L) adjusted to pH 12 and incubated at 35 °C for 7 days. The significantly low p-value (p < 0.0001) of the experiment propounded that the model best fits the experimental data, and the independent variables have considerable effects on the production of IAA. Morphologically, the in-vitro grown G. glabra plants showed enhanced root and shoot growth when co-cultivated with the isolated endophytic fungal strain (GG3F6) relative to the control plants. Also, the enhanced accumulation of total phenolic (10.7 %) and flavonoid (10.2 %) in the endophyte treated plants was observed. The optimization of IAA production by an endophytic fungus using (RSM) has not been reported so far. Interestingly, 2.1 fold increase in glycyrrhizin content was recorded in GG3F6 treated in-vitro host plants as compared to the control plants. This suggested a potential use of D. terebinthifolli as a biostimulator for plant and enhanced accumulation of glycyrrhizin. The study highlights the dynamic host-endophyte interaction for exploitation in agricultural and pharmaceutical applications.

Abstract:IUGR can develop, either in response to a reduced IGP or in response to a deficient transplacental supply of O2 and nutrients. Particularly, insight in the pathogenesis of extrinsic IUGR is important for the clinician to offer him tools for a more causal treatment. Extrinsic IUGR is usually preceded by a gradually developing uteroplacental insufficiency. Uteroplacental hypoperfusion may represent the common starting point for extrinsic IUGR and maternal systemic symptomatology (PIH, (pre)eclampsia). In addition, it may be the common endpoint of two different (subclinical) pathways: defective trophoblast differentiation ("primary") and decompensation of early-pregnancy circulatory maladaptation ("secondary"). From a theoretical point of view, the primary pathway may develop at a slower rate. Therefore, the primary pathway is more likely to result in extrinsic IUGR. In contrast, the secondary pathway assumes decompensation of an initially maladapted maternal circulation. This implies a highly variable rate of development and with it, severity of clinical symptomatology. It also indicates that the the pathogenesis, eventually leading to manifest vascular disease, is superimposed on a pre-existent inadequacy in maternal hemodynamics, renal function and/or volume homeostasis. It is understood that intertwining of these two pathways is common. Unfortunately, our current knowledge, about placentation and the concomitant early-pregnancy adjustments is poor. Current research of trophoblast differentiation and of the concomitant maternal end-organ effects in normal and pathologic pregnancy, is likely to increase our understanding of the first-trimester phenomena preceding IUGR soon.

Abstract:Abstract Indole acetic acid (IAA) is a phytohormone which one of the important compounds for plant growth and development, affecting as inducing cell elongation, cell enlargement and cell division. IAA synthesized both in the plants and microorganisms. For microorganisms, production of phytohormone IAA is one of the essential criteria for plant growth promotion. The aim of the present study was to isolate and screen IAA producing bacteria from peat land area in West Kalimantan and optimized some culture conditions for maximum IAA production. The production of IAA was determined in TSB and YEMB liquid medium and the optimization of IAA production in culture media was carried out at different of incubation time, L-Tryptophan concentrations, carbon and nitrogen sources, pH and temperature. The result revealed that maximum IAA production of Bacillus siamensis was achieved at 96 hours of incubation. The YEMB medium supplemented with 250 ppm of tryptophan was obtained as the best medium for IAA production (9.89 μg/ml). This bacterium produced optimum levels of IAA in the presence of sucrose (10.23 μg/ml) and tryptone (16.61 μg/ml) as carbon and nitrogen sources respectively. In the physical parameters, the maximum production of IAA was recorded at T 35°C (8.42 μg/ml) and a pH of 8.0 (13.29 μg/ml).

Abstract:Abstract Serratia plymuthica UBCF_13 is one of the bacteria that can increase plant growth [plant growth-promoting bacteria] by producing IAA [Indole-3-Acetic Acid]. S. plymuthica UBCF_13 is a strain of Andalas University Biotechnology Laboratory Collection which can produce IAA and increase the growth of Solanaceae plants. Optimization of culture media needs to be analyzed to increase IAA production on UBCF_13. Optimization can be done by adding tryptophan as a precursor, using various types of media, adding wall affecting agents, and certain metal ions. In this study, optimization was carried out by testing the type of media [TSB, NB, YM, and King’s B], adding tryptophan [0, 100, 200, 300 μg/mL], differences in pH [5, 6, 7, 8], giving wall affecting agent [SDS 0.1 μg / mL, EDTA 0.1 μg/mL], and metal ions [Ca, Fe, K, Mg at a concentration of 0.05% and 0.1%]. The highest IAA production was obtained in the combination treatment of YM media and tryptophan 300 μg/mL. Meanwhile, the treatment of differences in pH and wall affecting agents did not have a significant effect on the increase in the production of IAA UBCF_13. Testing of metal types on IAA production showed that calcium was able to increase the production of IAA UBCF_13 by 12-14 μg/mL. Serratia plymuthica UBCF_13 produced the highest IAA on YM media combined with the addition of 300 μg/mL of tryptophan and 0.1% calcium ion.

Abstract:Abstract Indole-3-acetic acid (IAA) is the most common plant hormone of the auxin class and regulates various plant growth processes. The present study investigated IAA production by the basidiomycetous yeast Rhodosporidiobolus fluvialis DMKU-CP293 using the one-factor-at-a-time (OFAT) method and response surface methodology (RSM). IAA production was optimized in shake-flask culture using a cost-effective medium containing 4.5% crude glycerol, 2% CSL and 0.55% feed-grade l -tryptophan. The optimized medium resulted in a 3.3-fold improvement in IAA production and a 3.6-fold reduction in cost compared with those obtained with a non-optimized medium. Production was then scaled up to a 15-L bioreactor and to a pilot-scale (100-L) bioreactor based on the constant impeller tip speed (V tip ) strategy. By doing so, IAA was successfully produced at a concentration of 3569.32 mg/L at the pilot scale. To the best of our knowledge, this is the first report of pilot-scale IAA production by microorganisms. In addition, we evaluated the effect of crude IAA on weed growth. The results showed that weed ( Cyperus rotundus L.) growth could be inhibited by 50 mg/L of crude IAA. IAA therefore has the potential to be developed as a herbicidal bioproduct to replace the chemical herbicides that have been banned in various countries, including Thailand.

Abstract:Background: Indole-3-acetic acid (IAA) is produced by microorganisms and plants via either tryptophan-dependent or tryptophan-independent pathways. Herein, we investigated the optimisation of IAA production by Streptomyces fradiae NKZ-259 and its formulation as a plant growth promoter to improve economic and agricultural development. Results: The maximum IAA yield achieved using optimal conditions was 82.363 μg/mL in the presence of 2 g/L tryptophan after 6 days of incubation. Thin-layer chromatography (TLC) and high-performance liquid chromatography (HPLC) analysis of putative IAA revealed an RF value of 0.69 and a retention time of 11.842 min, comparable with the IAA standard. Regarding product formulation, kaolin-based powder achieved a suspension rate of 73.74% and a wetting time of 80 s. This carrier exhibited good shelf life stability for NKZ-259, and the cell population did not decrease obviously over 4 months of storage at 4 °C. In vivo analysis of plant growth promotion showed that tomato seedlings treated with kaolin powder containing NKZ-259 cells displayed a significant increase in root and shoot length of 7.97 cm and 32.77 cm, respectively, and an increase in fresh weight and dry weight of 6.72 g and 1.34 g. Compared to controls, plant growth parameters were increased almost it two-fold. Conclusion: Optimising the culture conditions resulted in an almost four-fold increase in IAA secretion by NKZ-259 cells. The results clearly demonstrate that S. fradiae NKZ-259 holds great potential for plant growth promotion and IAA production. Furthermore, kaolin-based powder is an effective carrier for NKZ-259 cells and may be useful for commercial applications.

Abstract:A phytohormone is a molecule that controls the development of plants. Auxin and cytokines, which may be acquired as synthetic compounds or produced by microorganisms, which promote plant growth, are employed to enhance crop yield. Indole-3-Acetic Acid (IAA) is a key phytohormone that controls a number of physiological processes in plants. Indole-3-acetic acid (IAA), the most prevalent endogenous auxin, is involved in stem elongation and root formation. Auxin levels are usually higher in the rhizosphere. The possibility of rhizosphere bacteria to encourage direct plant development has been recognised as a crucial component of auxin (IAA) production. They promote the growth of lateral roots, which increases the surface area available for nutrition absorption and enhances nutrient and water absorption from the soil. The objective of this research is to isolate, characterise, and identify bacteria that produce indole acetic acid in rhizospheric soil. Bacterial colonies were isolated using the serial dillution technique on nutrient agar medium from rhizospheric soil samples of a rice crop. Five rhizospheric bacterial isolates (RIPB-1 to RIPB-30) were identified as effective Indole acetic acid makers after qualitative screening. The quantity of Indole acetic acid produced by five bacterial strains was tested for up to 120 hours. The amount of Indole acetic acid they could generate ranged from 1 to 11.2 g/ml. The bacterial strain RIPB-20 (11.2 g/ml) generated the highest Indole acetic acid after 72 hours of incubation, followed by RIPB-14. Based on morphological, physiological, and biochemical features, the bacterial isolate RIPB-20 was tentatively identified as Bacillus sp. Finally, the finding shows that the bacteria, which have capacity to produce Indole acetic acid, are good biofertilizer inoculants for growth of plant.
Identification and combinatorial engineering of indole-3-acetic acid synthetic pathways in Paenibacillus polymyxa

Abstract:Background: Paenibacillus polymyxa is a typical plant growth-promoting rhizobacterium (PGPR), and synthesis of indole-3-acetic acid (IAA) is one of the reasons for its growth-promoting capacity. The synthetic pathways of IAA in P. polymyxa must be identified and modified. Results: P. polymyxa SC2 and its spontaneous mutant SC2-M1 could promote plant growth by directly secreting IAA. Through metabonomic and genomic analysis, the genes patA, ilvB3, and fusE in the native IPyA pathway of IAA synthesis in strain SC2-M1 were predicted. A novel strong promoter P04420 was rationally selected, synthetically analyzed, and then evaluated on its ability to express IAA synthetic genes. Co-expression of three genes, patA, ilvB3, and fusE, increased IAA yield by 60% in strain SC2-M1. Furthermore, the heterogeneous gene iaam of the IAM pathway and two heterogeneous IPyA pathways of IAA synthesis were selected to improve the IAA yield of strain SC2-M1. The genes ELJP6_14505, ipdC, and ELJP6_00725 of the entire IPyA pathway from Enterobacter ludwigii JP6 were expressed well by promoter P04420 in strain SC2-M1 and increased IAA yield in the engineered strain SC2-M1 from 13 to 31 μg/mL, which was an increase of 138%. Conclusions: The results of our study help reveal and enhance the IAA synthesis pathways of P. polymyxa and its future application.
Environmental Factors Modulating Indole-3-Acetic Acid Biosynthesis by Four Nitrogen Fixing Bacteria in a Liquid Culture Medium

Abstract:This study evaluated the effects of some environmental conditions on IAA biosynthesizing capacity of four nitrogen fixing bacteria, namely Paenibacillus cineris TP-1.4, Bacillus megaterium MQ-2.5, Klebsiella pneumoniae OM-17.2, and Pseudomonas boreopolis CP-18.2. Carbon source, pH, NaCl, and tryptophan supplement treatments were set to investigate the effects of those environmental factors on IAA synthesis. The IAA synthesizing capacity of bacterial strains in liquid medium was measured spectroscopically following incubation by Salkowski's reagent method. The results showed that, under the sucrose amendment, the IAA concentrations produced by all four bacterial strains were significantly higher than those of the other four carbon source added treatments. Two of the four bacterial strains produced the highest yield of IAA in liquid medium at pH 7 (TP-1.4 and OM-17.2), whereas pH 8 was optimum for the other two strains (MQ-2.5 and CP-18.2). The MQ-2.5 strain could synthesize IAA fairly well in up to 5% NaCl and produced the highest amount of IAA with 1% NaCl. Furthermore, IAA synthesizing capability of tested bacterial strains increased sharply along with increasing tryptophan content in culture medium except for the TP-1.4 strain. From the current study, these isolates emerged as possible alternatives for future IAA production for plant growth and yield enhancement. Hence, they have a great potential to be used as bio-inoculants for plant growth promotion in eco-friendly and sustainable agriculture.

Abstract:Endophytic fungi are known to produce indole-3-acetic acid (IAA), which can stimulate plant growth. Twenty-seven isolates of endophytic fungi were isolated from Coffea arabica in northern Thailand. Only one isolate (CMU-A109) produced IAA in vitro. This isolate was identified as Colletotrichum fructicola based on morphological characteristics and molecular phylogenetic analysis of a combined five loci (internal transcribed spacer of ribosomal DNA, actin, β-tubulin 2, chitin synthase and glyceraldehyde-3-phosphate dehydrogenase genes). Identification of a fungal IAA production obtained from indole 3-acetamide (IAM) and tryptophan 2-monooxygenase activity is suggestive of IAM routed IAA biosynthesis. The highest IAA yield (1205.58±151.89 μg/mL) was obtained after 26 days of cultivation in liquid medium supplemented with 8 mg/mL L-tryptophan at 30°C. Moreover, the crude fungal IAA could stimulate coleoptile elongation of maize, rice and rye. This is the first report of IAA production by C. fructicola and its ability to produce IAA was highest when compared with previous reports on IAA produced by fungi.

Abstract:Burkholderia pyrrocinia JK-SH007 is a plant growth-promoting bacteria (PGPB), that can promote the growth of poplar and other trees, and, production of the plant hormone indole-3-acetic acid (IAA) is one of the reasons for this effect. Therefore, the aims of this study were to evaluate the effect of the external environment on the synthesis of IAA by B. pyrrocinia JK-SH007 and to perform a functional analysis of its IAA synthesis pathway. In this study, IAA and its synthetic intermediates indole-3-acetamide (IAM), indole-3-pyruvic acid (IPyA), tryptamine (TAM), and indole-3-acetonitrile (IAN) were detected in B. pyrrocinia JK-SH007 fermentation broth by high-performance liquid chromatography and tandem mass spectrometry (HPLC-MS/MS), and these indolic compounds were also found in the cell-free extraction of B. pyrrocinia JK-SH007, but the genomic analysis of B. pyrrocinia JK-SH007 indicated that IAA biosynthesis was mainly through the IAM and TAM pathways. The effects of L-tryptophan (L-Trp), temperature and pH on the synthesis of IAA were investigated, and the results showed that L-Trp exerted a significant effect on IAA synthesis and that 37°C and pH 7 were the optimal conditions IAA production by B. pyrrocinia JK-SH007. In addition, the protein expression of tryptophan 2-monooxygenase and indoleacetamide hydrolase, which are the key enzymes of the indole acetamide-mediated IAA synthesis pathway, was analyzed, and their activity was verified by substrate feeding experiments. The results revealed the existence of an IAA synthesis pathway mediated by IAM and indicated that this pathway plays a role in B. pyrrocinia JK-SH007. This study lays the foundation for further exploration of the specific pathway and mechanism of IAA synthesis in B. pyrrocinia JK-SH007.

Abstract:Indole acetic acid (IAA) as a plant hormone, was one of the valuable products of anaerobic fermentation. However, the enriching method remained unknown. Moreover, whether zero valent iron (ZVI) could enhance IAA production was unexplored. In this work, IAA producing bacteria Klebsiella (63 %) was enriched successfully. IAA average production rate and concentration were up to 3 mg/L/h and 56 mg/L. With addition of 1 g/L ZVI, IAA average production rate and concentration was increased for 2 and 3 folds. Mechanisms indicated ZVI increased Na + K + -ATP activity and electron transport activity for 2 folds and 1 fold. Moreover, macro transcription determined indole pyruvate pathway activity like primary-amine oxidase, indole pyruvate decarboxylase and aldehyde dehydrogenase were increased for 146 %, 187 %, and 557 %, respectively. Therefore, ZVI was suitable for enhancement IAA production from mixed culture anaerobic fermentation.

Abstract:Indole‐3‐acetic acid (IAA) is an essential phytohormone that controls a variety of plant growth mechanisms. Bacteria can produce IAA to stimulate plant growth, with its production influenced by the culture conditions. Serratia plymuthica UBCF_13 is recognized as an IAA‐producing bacterium, exhibiting maximum IAA production in a yeast medium comprising yeast extract, sucrose, K2HPO4, MgSO4, NaCl, and CaCO3. However, prior studies optimizing individual inorganic salt components indicated minimal impact on IAA synthesis within this medium. This study aimed to eliminate the unnecessary inorganic salt components and the medium was then applied to investigate the IAA biosynthesis pathway and the plant growth‐promoting assay. The elimination assay consisted of yeast sucrose medium devoid of K2HPO4, MgSO4, NaCl, or CaCO3, and yeast sucrose medium containing only MgSO4 and CaCO3. Various indole compounds were then added to the revised medium composition to investigate the IAA biosynthesis pathway of UBCF_13 using high‐performance liquid chromatography (HPLC). Furthermore, the effect of UBCF_13 culture supernatant, cultivated in the new medium, on chili plant growth was evaluated. The highest IAA production (138.8 μg/mL) was observed in the yeast sucrose with CaCO3 and MgSO4 (elimination of K2HPO4 and NaCl). The presence of indole‐3‐acetamide (IAM) compound from the medium extracts, supplemented with multiple indole compounds, revealed that UBCF_13 may use the IAM pathway. The application of UBCF_13 supernatant enhanced the shoot, root length, fresh weight, and germination time of chili seeds by 37.7%, 49.3%, 204.3%, and 38.6%, respectively. This study demonstrated that eliminating K2HPO4 and NaCl provided a new culture medium composition conducive to IAA production by UBCF_13. Moreover, the UBCF_13 extract has the potential to promote plant growth.

Abstract:Studies were conducted to find efficient Indole acetic acid producing strains and assessing the effectiveness of combined dose of azolla (dried) and IAA producers on plant growth. Out of ten isolates, only two isolates, viz, F1 and G2 were found to be potent IAA producers and were used for further experiments. It was found that the isolate F1 and G2 produced maximum amount of IAA at 0.9% tryptophan and incubation for 72 h at room temperature. The optimum pH for IAA production for isolates F1 and G2 was found to be at pH 9. The isolates were also assessed for the presence of other plant growth promoting activity, such as hydrogen cyanide production, oxalate solubilization, phosphate solubilization and zinc solubilization. Effect of IAA producers on seed germination was done by pot experiments carried out into sets. Isolates F1 and G2 were found efficient for plant growth promotion in desired aspects. Effect of combined dose of azolla and IAA producers on seed germination was assessed by pot experiments. For this, dried powder of azolla was added during sowing of culture treated seeds. No further culture supply was given to the seeds. Azolla has high protein content. Slow protein releasing property of azolla can be useful to provide constant supply of the precursor (tryptophan) for IAA synthesis by isolate. In addition to the plant exudates, azolla may also give constant supply of tryptophan. From all the assessed samples, the isolates FI and G2 showed efficient growth of plants in all aspects. G2 was found to be more potent than F1. So as to assess the compatibility of isolate in natural conditions, field studies were done. Four plots designed having individual size 9 × 5 m, were used. It was found that plots 3 (only culture) and 4 (culture with azolla) showed better plant growth as compared to others. On the basis of IAA producing ability of G2, pot experiments and field studies, it was concluded that isolate G2 can be used alone or in conjunction with azolla as a biofertilizer to get better plant growth.Graphical abstractOverview of process involving combined dose of azolla and IAA producer.

Indole-3-Acetic Acid Biosynthesis Pathways in the Plant-Beneficial Bacterium Arthrobacter pascens ZZ21

Abstract:Arthrobacter pascens ZZ21 is a plant-beneficial, fluoranthene-degrading bacterial strain found in the rhizosphere. The production of the phytohormone indole-3-aectic acid (IAA) by ZZ21 is thought to contribute to its ability to promote plant growth and remediate fluoranthene-contaminated soil. Using genome-wide analysis combined with metabolomic and high-performance liquid chromatography-mass spectrometry (HPLC-MS) analyses, we characterized the potential IAA biosynthesis pathways in A. pascens ZZ21. IAA production increased 4.5-fold in the presence of 200 mg·L-1 tryptophan in the culture medium. The transcript levels of prr and aldH, genes which were predicted to encode aldehyde dehydrogenases, were significantly upregulated in response to exogenous tryptophan. Additionally, metabolomic analysis identified the intermediates indole-3-acetamide (IAM), indole-3-pyruvic acid (IPyA), and the enzymatic reduction product of the latter, indole-3-lactic acid (ILA), among the metabolites of ZZ21, and subsequently also IAM, ILA, and indole-3-ethanol (TOL), which is the enzymatic reduction product of indole-3-acetaldehyde, by HPLC-MS. These results suggest that the tryptophan-dependent IAM and IPyA pathways function in ZZ21.

Abstract:The aim of this research was to examine the diversity of indigenous plant growth promoting rhizobacteria from paddy soil and to obtain a superior isolate that can support the growth and vigor of rice plants. This research was conducted in the Laboratory of Agronomy and Horticulture, Faculty of Agriculture, Jenderal Soedirman University from July to September 2017. The bacteria were isolated from paddy soil rhizosphere originated from rice production centers of Banyumas Regency i.e. Kebasen, Rawalo, Patikraja, Jatilawang, and Karangwangkal. Results showed that indigenous PGPR from paddy soil were able to produce Indol Acetic Acid (IAA) in the range 0.05-5.40 ppm, but did not have the ability to solubilize phosphate. Plant Growth Promoting Rhizobacteria (PGPR) inoculation in rice seedlings was able to increase seed vigor, plant height, root length, and seed germination. Based on morphological and biochemical characters, PGPR isolates were identified as Bacillus sp., Pseudomonas sp., Streptococcus sp., and Staphylococcus sp.
