Abstract:The lipopolysaccharide of capsule-deficient Haemophilus infuenzae strain Rd contains an N-acetylgalactosamine residue attached to the terminal globotriose moiety in the Hex5 glycoform. Genome analysis identified an open reading frame, HI1578, referred to as LgtD, whose amino acid sequence shows a significant level of similarity to those of a number of bacterial glycosyltransferases involved in lipopolysaccharide biosynthesis. To investigate its function, overexpression and biochemical characterization were performed. Most of the protein was obtained in a highly soluble and active form. Standard glycosyltransferase assay, high-performance liquid chromatography (HPLC), and liquid chromatography (LC)/mass spectrometry (MS) show that LgtD is an N-acetylgalactosaminyltransferase with high donor substrate specificity, and globotriose is a highly preferred acceptor substrate for the enzyme.
4-Deoxy-4-fluoro-β-D-glucopyranose

Abstract:4-Deoxy-4-fluoro-β-D-glucopyranose, C(6)H(11)FO(5), (I), crystallizes from water at room temperature in a slightly distorted (4)C(1) chair conformation. The observed chair distortion differs from that observed in β-D-glucopyranose [Kouwijzer, van Eijck, Kooijman & Kroon (1995). Acta Cryst. B51, 209-220], (II), with the former skewed toward a B(C3,O5) (boat) conformer and the latter toward an (O5)TB(C2) (twist-boat) conformer, based on Cremer-Pople analysis. The exocyclic hydroxymethyl group conformations in (I) and (II) are similar; in both cases, the O-C-C-O torsion angle is ∼-60° (gg conformer). Intermolecular hydrogen bonding in the crystal structures of (I) and (II) is conserved in that identical patterns of donors and acceptors are observed for the exocyclic substituents and the ring O atom of each monosaccharide. Inspection of the crystal packing structures of (I) and (II) reveals an essentially identical packing configuration.

Abstract:The title compound, C20H47NO6Si4, was synthesized by per-O-trimethylsilylation of N-acetyl-d-glucosamine using chlorotrimethylsilane in the presence of hexamethyldisiloxane. The trimethylsilyl group and acetamido group are located on the same side of the pyran ring, showing an α-configuration glycoside. One of the trimethylsilyl groups is disordered over two orientations, with site-occupancy factors of 0.625 (9) and 0.375 (9). In the crystal, N—H⋯O hydrogen bonds link the molecules into supramolecular chains along the a-axis direction.

Co-crystals of 3-deoxy-3-fluoro-α-D-glucopyranose and 3-deoxy-3-fluoro-β-D-glucopyranose

Abstract:3-Deoxy-3-fluoro-D-glucopyranose crystallizes from acetone to give a unit cell containing two crystallographically independent molecules. One of these molecules (at site A) is structurally homogeneous and corresponds to 3-deoxy-3-fluoro-β-D-glucopyranose, C(6)H(11)FO(5), (I). The second molecule (at site B) is structurally heterogeneous and corresponds to a mixture of (I) and 3-deoxy-3-fluoro-α-D-glucopyranose, (II); treatment of the diffraction data using partial-occupancy oxygen at the anomeric center gave a high-quality packing model with an occupancy ratio of 0.84:0.16 for (II):(I) at site B. The mixture of α- and β-anomers at site B appears to be accommodated in the lattice because hydrogen-bonding partners are present to hydrogen bond to the anomeric OH group in either an axial or equatorial orientation. Cremer-Pople analysis of (I) and (II) shows the pyranosyl ring of (II) to be slightly more distorted than that of (I) [θ((I)) = 3.85 (15)° and θ((II)) = 6.35 (16)°], but the general direction of distortion is similar in both structures [ϕ((I)) = 67 (2)° (B(C1,C4)) and ϕ((II)) = 26.0 (15)° ((C3)TB(C1)); B = boat conformation and TB = twist-boat conformation]. The exocyclic hydroxymethyl (-CH(2)OH) conformation is gg (gauche-gauche) (H5 anti to O6) in both (I) and (II). Structural comparisons of (I) and (II) to related unsubstituted, deoxy and fluorine-substituted monosaccharides show that the gluco ring can assume a wide range of distorted chair structures in the crystalline state depending on ring substitution patterns.

Abstract:The title compound, C(13)H(24)O(11).4H(2)O, (I), crystallized from water, has an internal glycosidic linkage conformation having phi' (O5(Gal)-C1(Gal)-O1(Gal)-C4(All)) = -96.40 (12) degrees and psi' (C1(Gal)-O1(Gal)-C4(All)-C5(All)) = -160.93 (10) degrees , where ring-atom numbering conforms to the convention in which C1 denotes the anomeric C atom, C5 the ring atom bearing the exocyclic hydroxymethyl group, and C6 the exocyclic hydroxymethyl (CH(2)OH) C atom in the betaGalp and betaAllp residues. Internal linkage conformations in the crystal structures of the structurally related disaccharides methyl beta-lactoside [methyl beta-D-galactopyranosyl-(1-->4)-beta-D-glucopyranoside] methanol solvate [Stenutz, Shang & Serianni (1999). Acta Cryst. C55, 1719-1721], (II), and methyl beta-cellobioside [methyl beta-D-glucopyranosyl-(1-->4)-beta-D-glucopyranoside] methanol solvate [Ham & Williams (1970). Acta Cryst. B26, 1373-1383], (III), are characterized by phi' = -88.4 (2) degrees and psi' = -161.3 (2) degrees , and phi' = -91.1 degrees and psi' = -160.7 degrees , respectively. Inter-residue hydrogen bonding is observed between O3(Glc) and O5(Gal/Glc) in the crystal structures of (II) and (III), suggesting a role in determining their preferred linkage conformations. An analogous inter-residue hydrogen bond does not exist in (I) due to the axial orientation of O3(All), yet its internal linkage conformation is very similar to those of (II) and (III).

Abstract:Total synthesis of 4d-deoxy Lewisx pentasaccharide is described. O-(2,3,6-Tri-O-benzoyl-4-deoxy-α-d-xylo-hexopyranosyl)trichloroacetimidate was condensed with a diol of glucosamine to give regioselectively a β1→4 linked disaccharide, which was further fucosylated to a protected deoxy Lewisx trisaccharide. A highly stereo- and regioselective glycosylation of this trisaccharide onto a lactoside diol was performed to provide a pentasaccharide in excellent yield. After deprotection, this pentasaccharide afforded the target oligosaccharide.

Abstract:The crystal structure of 2NdCl3·galactitol·14H2O has been determined. The crystal system is triclinic, space group: −1, with unit-cell dimensions: a=9.736(2), b=10.396, c=8.027 Å; α=108.05(3), β=92.68(3), γ=88.44(3)°, V=771.6(3) Å3, Z=2. Each Nd atom is coordinated to nine oxygen atoms, three from the alditol and six from water molecules, with NdO distances from 2.461 to 2.552 Å. The seventh water molecule is hydrogen-bonded by the hydroxyl hydrogen on O-1 (O-1H-11⋯O-10, 2.639 Å). The FT-IR spectra of 2NdCl3·galactitol·14H2O and 2PrCl3·galactitol·14H2O are analogous, and show that Pr and Nd have the same coordination mode. The IR results are consistent with the crystal structures.

Abstract:The crystal structure of LaCl3·galactitol·6H2O has been determined. It is monoclinic. The space group is P21/n with unit cell dimensions a=10.5091(7), b=12.5366(7), c=14.4420(10)Å, β=90.974(2)°, V=1902.4(2)Å3 and Z=4. Each La3+ ion in the unit cell is coordinated to 10 oxygen atoms, three from O1, O2 and O3 of one alditol, three from O4, O5 and O6 of another alditol and four from water molecules, with La–O distances from 2.5099 to 2.6916Å. The other two water molecules are hydrogen-bonded. FT-IR spectrum of LaCl3·galactitol·6H2O is apparently different from that of other lanthanide–galactitol complexes. It is consistent with the differences between their crystal structures. Both the IR result and the crystal structure of LaCl3·galactitol·6H2O complex show that it has the different coordination mode compared with other lanthanide complexes.
Methyl 4- O - b - D -galactopyranosyl a - D -glucopyranoside (methyl a -lactoside)

Abstract:Methyl -lactoside, C13H24O11, (I), is described by glycosidic torsion angles ' (O5galÐC1galÐO1galÐC4glc) and (C1galÐ O1galÐC4glcÐC5glc), which have values of ÿ93.52 (13) and ÿ144.83 (11) , respectively, where the ring atom numbering conforms to the convention in which C1 is the anomeric C atom and C6 is the exocyclic hydroxymethyl (±CH2OH) C atom in both residues. The linkage geometry is similar to that observed in methyl -lactoside methanol solvate, (II), in which ' is ÿ88.4 (4) and is ÿ161.3 (4) . As in (II), an intermolecular O3glcÐH O5gal hydrogen bond is observed in (I). The hydroxymethyl group conformation in both residues is gauche±trans, with torsion angles !gal (O5galÐ C5galÐC6galÐO6gal) and !glc (O5glcÐC5glcÐC6glcÐO6glc) of 69.15 (13) and 72.55 (14) , respectively. The latter torsion angle differs substantially from that found for (II) [ÿ54.6 (2) ; gauche±gauche]. Cocrystallization of methanol, which is hydrogen bonded to O6glc in the crystal structure of (II), presumably affects the hydroxymethyl conformation in the Glc residue in (II).

Abstract:4-(4'-Ethoxyphenyl)phenyl-beta-O-D-glucoside (1) was synthesized. A perfect crystal of I was obtained from all acetonitrile solution. In a mixture of water/1,4-dioxane (8/2, v/v), a gel formed at first and then collapsed into a needlelike crystal. The crystal and the gel were characterized by X-ray diffraction, UV-vis absorption, circular dichroism, Fourier transform infrared spectroscopy, and electron microscopy. It was found that the crystal formed from the gel had the same diffraction pattern Lis that of the single crystal from the acetonitrile solution, Suggesting identical molecular packing. In a monoclinic cell containing two molecules, molecules took a "T" arrangement with an antiparallel direction. In contrast, the diffraction pattern of xerogel from water/1,4-dioxane (8/2, v/v) displayed a d-spacing ratio of 1:1/2:1/3 (2.39, 1.18, and 0.78 nm), indicating an interdigitated bilayer structure. A molecular packing model from gel to crystal where molecular sheets would insert into each other with a glide movement was suggested, On the basis of the understanding of the deformation mechanism, ail additive was introduced to obtain a stable gel.

Abstract:The crystal structure of β‐d‐galactofuranosidase belonging to glycoside hydrolase family 2 was determined in complex with a mechanism‐based potent inhibitor, d‐iminogalactitol. The inhibitor was bound to the enzyme with an integral hydrogen bond network. The extensive interactions in the active site allowed discrimination between β‐d‐galactofuranoside and α‐l‐arabinofuranoside substrates. d‐Galactofuranose (Galf) is widely distributed in glycoconjugates of pathogenic microbes. β‐d‐Galactofuranosidase (Galf‐ase) from Streptomyces sp. JHA19 (ORF1110) belongs to glycoside hydrolase (GH) family 2 and is the first identified Galf‐specific degradation enzyme. Here, the crystal structure of ORF1110 in complex with a mechanism‐based potent inhibitor, d‐iminogalactitol (Ki = 65 μm) was solved. ORF1110 binds to the C5–C6 hydroxy groups of d‐iminogalactitol with an extensive and integral hydrogen bond network, a key interaction that discriminates the substrates. The active site structure of ORF1110 is largely different from those of β‐glucuronidases and β‐galactosidases in the same GH2 family. A C‐terminal domain of ORF1110 is predicted to be a carbohydrate‐binding module family 42 that may bind Galf. The structural insights into Galf‐ase will contribute to the investigation of therapeutic tools against pathogens.

Glycosylation of a lactoside diol with this trisaccharide provided a pentasaccharide, which after deprotection, afforded the target pentasaccharide.

Abstract:A fucoidan, obtained from the hot-water extract of the brown seaweed, Hizikia fusiforme, was separated into five fractions by DEAE Sepharose CL-6B and Sepharose CL-6B column chromatography. All five fractions contained predominantly fucose. mannose and galactose and also contained sulfate groups and uronic acid. The fucoidans had MWs front 25 to 950 kDa. The structure of fraction F32 was investigated by desulfation, carboxyl-gruop reduction, partial hydrolysis, methylation analysis and NMR spectroscopy. The results Showed that the sugar composition of F32 was mainly fucose, galactose, mannose, xylose and glucuronic acid: sulfate was 21.8%, and the MW was 92.7 kDa. The core of F32 was mainly composed of alternating units of -> 2)-alpha-D-Man(1 -> and 4)-beta-D-GlcA(1 ->, with a minor portion of -> 4)-beta-D-Gal(1 -> units. The branch points were at C-3 of -> 2)-Man-(1 ->, C-2 of -> 4)-Gal-(1 -> and C-2 of -> 6)-Gal-(1 ->. About two-thirds of the fucose units were at the nonreducing ends, and the remainder were (1 -> 4)-, (1 -> 3)- and (1 -> 2)-linked. About two-thirds of xylose units were at the nonreducing ends, and the remainder were (1 -> 4)-linked. Most of the mannose units were (1 -> 2)-linked, and two-thirds of them had a branch at C-3. Galactose was mainly (1 -> 6)-linked. The absolute configurations of the sugar residues were alpha-D-Manp, alpha-L-Fucp, alpha-D-Xylp, beta-D-Galp and beta-D-GlcpA. Sulfate groups in F32 were at C-6 of -> 2,3)-Man-(1 ->, C-4 and C-6 of -> 2)-Man-(1 ->, C-3 of -> 6)-Ga1-(1 ->. C-2. C-3 or C-4 of fucose, while some fucose had two sulfate groups. Structure and mechanism of l-fucose isomerase from Escherichia coli

Abstract:The three-dimensional structure of L-fucose isomerase from Escherichia coli has been determined by X-ray crystallography at 2.5 A resolution. This ketol isomerase converts the aldose L-fucose into the corresponding ketose L-fuculose using Mn2+ as a cofactor. Being a hexamer with 64,976 Da per subunit, L-fucose isomerase is the largest structurally known ketol isomerase. The enzyme shows neither sequence nor structural similarity with other ketol isomerases. The hexamer obeys D3 symmetry and forms the crystallographic asymmetric unit. The strict and favorably oriented local symmetry allowed for a computational phase extension from 7.3 A to 2.5 A resolution. The structure was solved with an L-fucitol molecule bound to the catalytic center such that the hydroxyl groups at positions 1 and 2 are ligands of the manganese ion. Most likely, L-fucitol mimics a bound L-fucose molecule in its open chain form. The protein environment suggests strongly that the reaction belongs to the ene-diol type.

Disorder and conformational analysis of methyl β-D-galactopyranosyl-(1→4)-β-D-xylopyranoside

Abstract:Methyl β-D-galactopyranosyl-(1→4)-β-D-xylopyranoside, C(12)H(22)O(10), (II), crystallizes as colorless needles from water with positional disorder in the xylopyranosyl (Xyl) ring and no water molecules in the unit cell. The internal glycosidic linkage conformation in (II) is characterized by a φ' torsion angle (C2'(Gal)-C1'(Gal)-O1'(Gal)-C4(Xyl)) of 156.4 (5)° and a ψ' torsion angle (C1'(Gal)-O1'(Gal)-C4(Xyl)-C3(Xyl)) of 94.0 (11)°, where the ring atom numbering conforms to the convention in which C1 denotes the anomeric C atom, and C5 and C6 denote the hydroxymethyl (-CH(2)OH) C atoms in the β-Xyl and β-Gal residues, respectively. By comparison, the internal linkage conformation in the crystal structure of the structurally related disaccharide, methyl β-lactoside [methyl β-D-galactopyranosyl-(1→4)-β-D-glucopyranoside], (III) [Stenutz, Shang & Serianni (1999). Acta Cryst. C55, 1719-1721], is characterized by φ' = 153.8 (2)° and ψ' = 78.4 (2)°. A comparison of β-(1→4)-linked disaccharides shows considerable variability in both φ' and ψ', with the range in the latter (~38°) greater than that in the former (~28°). Inter-residue hydrogen bonding is observed between atoms O3(Xyl) and O5'(Gal) in the crystal structure of (II), analogous to the inter-residue hydrogen bond detected between atoms O3(Glc) and O5'(Gal) in (III). The exocyclic hydroxymethyl conformations in the Gal residues of (II) and (III) are identical (gauche-trans conformer).

Abstract:Crystal structures and FT-IR spectra of metal ion–galactitol (C6H14O6, the ligand here abbreviated as L) complexes: 2LaCl3·C6H14O6·10H2O and SrCl2·C6H14O6 complexes are reported. Crystal data of lanthanide chlorides (La3+, Nd3+, Sm3+, Eu3+, Tb3+)–galactitol complexes and alkaline earth chlorides (Ca2+, Sr2+)–galactitol complexes published earlier are summarized. Unlike other lanthanide ion–galactitol complexes (2MCl3·C6H14O6·14H2O), lanthanum ions give rise to two different structures: LaCl3·C6H14O6·6H2O (LaL1) and 2LaCl3·C6H14O6·10H2O (LaL2). Sr2+–galactitol complexes also crystallized with two structures: SrCl2·C6H14O6·4H2O (SrL1) and SrCl2·C6H14O6 (SrL2). These metal ions thus give different coordination structures with galactitol. The crystal structures and FT-IR spectra of lanthanide ion and alkaline earth ion–galactitol complexes were integrated to interpret the coordination modes of different metal ions. Similar IR spectra demonstrate the same coordination modes of the complexes.
Inhibition of Lactose Crystallisation in the Presence of Galacto-Oligosaccharide

Abstract:The stabilization of lactose in the form of amorphous (i.e. non-crystalline form) is the basic requirement to maintain the quality of relevant food and pharmaceutical products. The physiochemical properties of amorphous lactose mixed with galacto-oligosaccharide (GOS) were investigated. Water sorption, glass transition temperature, and crystallisation behaviour of lactose in the present of GOS (1:1 w/w) were measured at various water activity (0.11-0.75 a(w), 25 degrees C) and lactose mutarotation was also evaluated. All of them were compared with the physiochemical properties of trehalose-lactose (1:1 w/w). The addition of GOS to lactose increased the hygroscopicity of the mixture, as well as slightly increased the glass transition temperature compared to lactose alone. The critical water activity (at 0.68 a(w)) of lactose crystallisation was increased by the addition of GOS as compared to that of trehalose-lactose (1:1 w/w) (at 0.58 a(w)) or lactose alone (at 0.44 a(w)). The dramatical inhibition of lactose crystallisation with a lower crystallisation kinetic constant and the alternation of lactose crystal forms in the presence of GOS was observed as compared to the crystallisation behaviour of trehalose-lactose (1:1 w/w) and pure lactose at 0.68 and 0.75 a(w), 25 degrees C. Without affecting its T-g, the significantly delayed crystallisation of lactose in GOS-lactose mixture (1:1 w/w) was more likely due to the change of lactose mutarotation. As comparing to trehalose that is an effective inhibitor, GOS has a stronger ability to prevent lactose from crystallisation in hydrous matrices.
Membrane-bound sorbitol dehydrogenase is responsible for the unique oxidation of D-galactitol to L-xylo-3-hexulose and D-tagatose in Gluconobacter oxydans

Abstract:Background: Gluconobacter oxydans, is used in biotechnology because of its ability to oxidize a wide variety of carbohydrates, alcohols, and polyols in a stereo- and regio-selective manner by membrane-bound dehydrogenases located in periplasmic space. These reactions obey the well-known Bertrand-Hudson's rule. In our previous study (BBA-General Subjects, 2021, 1865:129740), we discovered that Gluconobacter species, including G. oxydans and G. cerinus strain can regio-selectively oxidize the C-3 and C-5 hydroxyl groups of D-galactitol to rare sugars D-tagatose and L-xylo-3-hexulose, which represents an exception to Bertrand Hudson's rule. The enzyme catalyzing this reaction is located in periplasmic space or membrane-bound and is PQQ (pyrroloquinoline quinine) and Ca2+-dependent; we were encouraged to determine which type of enzyme(s) catalyze this unique reaction. Methods: Enzyme was identified by complementation of multi-deletion strain of Gluconobacter oxydans 621H with all putative membrane-bound dehydrogenase genes. Results and conclusions: In this study, we identified this gene encoding the membrane-bound PQQ-dependent dehydrogenase that catalyzes the unique galactitol oxidation reaction in its 3'-OH and 5'-OH. Complement experiments in multi-deletion G. oxydans BP.9 strains established that the enzyme mSLDH (encoded by GOX0855-0854, sldB-sldA) is responsible for galactitol's unique oxidation reaction. Additionally, we demonstrated that the small subunit SldB of mSLDH was membrane-bound and served as an anchor protein by fusing it to a red fluorescent protein (mRubby), and heterologously expressed in E. coli and the yeast Yarrowia lipolytica. The SldB subunit was required to maintain the holo-enzymatic activity that catalyzes the conversion of D-galactitol to L-xylo-3-hexulose and D-tagatose. The large subunit SldA encoded by GOX0854 was also characterized, and it was discovered that its 24 amino acids signal peptide is required for the dehydrogenation activity of the mSLDH protein. General significance: In this study, the main membrane-bound polyol dehydrogenase mSLDH in G. oxydans 621H was proved to catalyze the unique galactitol oxidation, which represents an exception to the Bertrand Hudson's rule, and broadens its substrate ranges of mSLDH. Further deciphering the explicit enzymatic mechanism will prove this theory.
De Novo Biosynthesis of Difucosyllactose by Artificial Pathway Construction and α1,3/4-Fucosyltransferase Rational Design in Escherichia coli

Abstract:Difucosyllactose (DFL) is a significant and plentiful oligosaccharide found in human breast milk. In this study, an artificial metabolic pathway of DFL was designed, focusing on the de novo biosynthesis of GDP-fucose from only glycerol. This was achieved by engineering Escherichia coli to endogenously overexpress genes manB, manC, gmd, and wcaG and heterologously overexpress a pair of fucosyltransferases to produce DFL from lactose. The introduction of α-1,2-fucosyltransferase from Helicobacter pylori (FucT2) along with α-1,3/4-fucosyltransferase (HP3/4FT) addressed rate-limiting challenges in enzymatic catalysis and allowed for highly efficient conversion of lactose into DFL. Based on these results, molecular modification of HP3/4FT was performed based on computer-assisted screening and structure-based rational design. The best-performing mutant, MH5, containing a combination of five mutated sites (F49K/Y131D/Y197N/E338D/R369A) of HP3/4FT was obtained. The best strain BLC09-58 harboring MH5 yielded 45.81 g/L of extracellular DFL in 5-L fed-batch cultures, which was the highest titer reported to date.
