Abstract:The objective of the present study was to determine whether the mitochondrial calcium uniporter plays a role in the cardioprotection induced by ischemic preconditioning (IPC). Isolated rat hearts were subjected to 30 min of regional ischemia by ligation of the left anterior descending artery followed by 120 min of reperfusion. IPC was achieved by two 5-min periods of global ischemia separated by 5 min of reperfusion. IPC reduced the infarct size and lactate dehydrogenase release in coronary effluent, which was associated with improved recovery of left ventricular contractility. Treatment with ruthenium red (RR, 5 microM), an inhibitor of the uniporter, or with Ru360 (10 microM), a highly specific uniporter inhibitor, provided cardioprotective effects like those of IPC. The cardioprotection induced by IPC was abolished by spermine (20 microM), an activator of the uniporter. Cyclosporin A (CsA, 0.2 microM), an inhibitor of the mitochondrial permeability transition pore, reversed the effects caused by spermine. In mitochondria isolated from untreated hearts, both Ru360 (10 microM) and RR (1 microM) decreased pore opening, while spermine (20 microM) increased pore opening which was blocked by CsA (0.2 microM). In mitochondria from preconditioned hearts, the opening of the pore was inhibited, but this inhibition did not occur in the mitochondria from hearts treated with IPC plus spermine. These results indicate that the mitochondrial calcium uniporter is involved in the cardioprotection conferred by ischemic preconditioning.
[Mechanism of cardioprotection induced by noninvasive limb ischemic preconditioning]

Abstract:Objective: To verify the inhibitory effect of mitochondrial calcium uniporter in remote preconditioning-induced cardioprotection. Methods: By occlusion and reperfusion of left anterior descending artery, the rat hearts were subjected to 30 min regional ischemia and 120 min reperfusion in vivo. Thus the ischemic reperfusion model was established. The rats were randomly assigned to undergo one of the following maneuvers: (1) remote preconditioning; (2) ruthenium red (an inhibitor of mitochondrial calcium uniporter); (3) spermine or SB202190 (an opener of mitochondrial calcium uniporter). Remote preconditioning was elicited by three cycles of 5 min of right femoral artery occlusion interspersed with 5 min of reperfusion. The mean arterial blood pressure, heart rate and lactate dehydrogenase released in plasma were measured during reperfusion but the infarct size was measured after reperfusion. Results: In comparison with I/R group, remote preconditioning limited infarct size [(20.4 +/- 2.5)% vs (51.0 +/- 6.0)%] and lactate dehydrogenase release [(271 +/- 9) U/L vs (339 +/- 39)U/L] during reperfusion. On the contrary, spermine or SB202190 attenuated the reduction of infarct size and lactate dehydrogenase release induced by remote preconditioning. The group of spermine was [(40.8 +/- 9.2)% vs (20.4 +/- 2.5)%] and [(383 +/- 43) U/L vs (271 +/- 9) U/L] while the group of SB202190 was [(44.3 +/- 6.8)% vs (20.4 +/- 2.5)%] and [(356 +/- 26) U/L vs (271 +/- 9) U/L]. Conclusion: Inhibition of mitochondrial calcium uniporter opening is involved in the remote preconditioning-induced cardioprotection.

Abstract:The objective of the present study was to determine whether the mitochondrial calcium uniporter plays a role in cardioprotection by ischemic preconditioning (IPC). We found that both IPC and inhibition of the mitochondrial calcium uniporter during reperfusion improved recovery of left ventricular developed pressure, maximal rise velocity and end-diastolic pressure, and reduced infarct size and lactate dehydrogenase release. These protective effects were attenuated by activating the mitochondrial calcium uniporter. We conclude that the mitochondrial calcium uniporter is involved in the cardioprotection of ischemic preconditioning.

Cardioprotection of Ischemic Postconditioning Against Ischemia/reperfusion Injury in Isolated Rat Hearts

Abstract:AIM:To investigate the protective role of postconditioning in myocardial ischemia/reperfusion in rats and its mechanisms.METHODS:Cardiac contractility was analyzed by the Langendorff method.Infarct size was determined by dual staining with triphenyltetrazolium chloride and Even's blue dye,and the cardiac arrhythmia was evaluated.postconditioning was conducted by 3 cycles of 30 s ischemia followed by 30 s of reperfusion at the beginning of subsequent persistent reperfusion.RESULTS:Left ventricular systolic pressure(LVSP)and maximal rise rate of ventricular pressure(+dp/dtmax)were higher during reperfusion in postconditioning group compared with control.postconditioning reduced the infarct size in ischemia/reperfusion rat hearts.The cardiac arrhythmia score was decreased in postconditioning group in the first 10 min of reperfusion followed by ischemia compared to control group.postconditioning had similar cardioprotective effect as preconditioning.5-HD,a selective mitochondrial ATP-sensitive potassium channel(mitoKATP)inhibitor,blocked the amelioration of contract function provided by postconditioning.It also abolished the protective effect of postconditioning on cardiac arrhythmia score and infarct size.CONCLUSION:The results show that postconditioning has cardioprotective effect and attenuates reperfusion injury in ischemic heart.The effect might be partly through the activation of mitoKATP channel.
Inhibition of Mitochondrial Permeability Transition Pore Opening Mediated by Kappa‐opioid Receptor in Cardioprotection by Remote Preconditioning

Abstract:The objective of the present study was to investigate role of kappa-opioid receptor in cardioprotection by limb remote preconditioning and possible mechanism. Remote preconditioning (RPC) was induced by 3 cycles of 5 min right femoral artery occlusion followed by 5 min of reperfusion. Myocardial ischemia /reperfusion (I/R) was achieved by ligation of left anterior descending coronary artery of male Sprague-Dawley rats for 30 min and then loosened as reperfusion for 120 min. The opening of the mitochondrial permeability transition pore was monitored with fluorescent calcein-AM in isolated ventricular myocytes. RPC significantly decreased the infarct size induced by I/R, and this effect was attenuated by both naloxone (3 mg/kg), a non-specific inhibitor of opioid receptors and nor-BIN (10 mg/kg), a specific kappa-opioid receptor inhibitor. Cyclosporine A (10mg/kg), a specific inhibitor of mitochondrial permeability transition pore, blocked the effect of naloxone and nor-BIN. Atractyloside (5mg/kg), a specific activator of the pore, attenuated the effect of RPC. In isolated calcein loaded ventricular myocytes, U-50488H (100 microM), a specific activator of kappa-opioid receptor, decreased the pore opening induced by calcium (200 microM). In conclusion, the activation of cardiac kappa-opioid receptor is involved in the cardioprotection by remote preconditioning and the mitochondrial permeability transition pore may be in the post-receptor pathway following the activation of cardiac kappa-opioid receptor in remote preconditioning induced cardioprotection. Abstract:AIMTo determine mechanisms of cardioprotection induced by combination angiotensin-converting enzyme inhibitors (ACEI) with subthreshold preconditioning after activation of mitochondrial ATP-sensitive potassium (mitoK(ATP)) channel.METHODSThe Langendorff model of isolated rat heart was used. The time of the onset of uncoupling, the activities of sarcolemmal Na+/K+ -ATPase and Ca2+/Mg2+ -ATPase were measured.RESULTSThe subthreshold preconditioning (2 min of ischemia and 10 min reperfusion) or captopril (an ACEI) alone did not protect hearts against injury of sustained ischemia. However combination captopril with subthreshold preconditioning increased LVDP. Pretreatment hearts with mitoK(ATP) channel inhibitor 5-HD abolished the protection effect. Combination captopril with subthreshold preconditioning delayed the onset of uncoupling, and enhanced the activities of sarcolemmal Na+/K+ ATPase and Ca2+/Mg2+ -ATPase in ischemia/reperfusion hearts. But 5-HD cancelled these cardioprotection effects.CONCLUSIONCombination ACEI with subthreshold preconditioning delays the onset of cellular uncoupling induced by acute ischemia, and promotes the stability of sarcolemmal ion channels, in which activation of the mitoK(ATP) channels may be involved.
Activation of Mitochondrial ATP-sensitive Potassium Channels Delays Ischemia-Induced Cellular Uncoupling in Rat Heart.

Abstract:AIM:To test the hypothesis that cellular uncoupling induced by myocardial ischemia is mediated by activation of mitochondrial ATP-sensitive potassium channels (mitoKATP).METHODS:Rat hearts were perfused on a Langendorff apparatus and subjected to 40-min ischemia followed by 30-min reperfusion (I/R). Changes in cellular coupling were monitored by measuring whole-tissue resistance.RESULTS:(1) In hearts subjected to I/R, the onset of uncoupling started at (13.3+/-1.0) min of ischemia; (2) Ischemic preconditioning (IPC) delayed the onset of uncoupling until (22.7+/-1.3) min. Blocking mitoKATP channels with 5-hydroxydecanoate (5-HD) before the IPC abolished the uncoupling delay [(12.6+/-1.6) min]; (3) Calcium preconditioning (CPC) had the same effect as IPC. And this effect was reversed by blocking the mitoKATP channel again. In the CPC group the onset of uncoupling occurred after (20.6+/-1.3) min, and this was canceled by 5-HD [(13.6+/-0.8) min]; (4) In hearts pretreated with the specific mitoKATP channel opener diazoxide before sustained ischemia, the onset was delayed to (18.4+/-1.4) min; (5) 5-HD canceled the protective effects of diazoxide (12.6+/-1.0) min; and both the L-type Ca2+ channel inhibitor verapamil and the free radical scavenger N-(2-mercaptopropionyl)glycine, reduced the extended onset time induced by diazoxide [to (13.3+/-1.8) min and (13.4+/-2.1) min, respectively].CONCLUSION:IPC and CPC delay the onset of cellular uncoupling induced by acute ischemia in rat heart, and the underlying mechanism involves activation of the mitoKATP channels.
[Mitochondrial calcium uniporter participates in TNF-alpha induced cardioprotection in isolated rat hearts subjected to ischemia/reperfusion]

Abstract:Aim: To investigate whether mitochondrial calcium uniporter participates in the cardioprotection of tumor necrosis factor alpha (TNFalpha) pretreatment in isolated rat hearts subjected to ischemia/reperfusion. Methods: Isolated perfused rat hearts were subjected to 30 min regional ischemia (occlusion of left anterior descending artery) and 120 min reperfusion. The infarct size, coronary flow (CF) and lactate dehydrogenase (LDH) release during reperfusion were measured. The mitochondria of the heart were isolated and suspended in the swelling buffer for measurement of absorbance at 520 nm. Results: Pretreatment with TNFa at 10 U/ml for 7 min followed by 10 min washout reduced the infarct size and LDH release, and improved the recovery of CF during reperfusion. Administration of spermine (20 micromol/L), an opener of mitochondrial calcium uniporter, for 10 min during early reperfusion attenuated the reduction of infarct size and LDH release, and improvement of CF induced by TNFalpha. In isolated mitochondria of the heart pretreated with TNFalpha, the absorbance at 520 nm decreased less than that of mitochondria without TNFalpha pretreatment. Administration of spermine (50 micromol/L) attenuated the change of the absorbance induced by TNFalpha. Conclusion: The findings indicate that TNFalpha protects myocardium against ischemia/reperfusion injury via inhibiting mitochondrial calcium uniporter opening as well as mitochondrial permeability transition pore opening.

Abstract:Aims: To investigate the role of the mitochondrial Ca2+-activated K+ channel in cardioprotection induced by limb remote ischemic preconditioning.Main methods: Male Sprague-Dawley rats (250-300 g) were randomized into control, ischemia/reperfusion (I/R), remote ischemic preconditioning (RPC), NS1619 (a specific mitochondrial Ca2+-activated K+ channel opener), and RPC + paxilline (a specific mitochondrial Ca2+-activated K+ channel inhibitor) groups. RPC was induced by 4 cycles of 5 min of ligation followed by 5 min of reperfusion of the left femoral artery. Myocardial I/R was achieved by ligation of the left anterior descending coronary artery for 30 min, followed by 120 min of reperfusion. Infarct size was determined by 2,3,5-triphenyltetrazolium chloride staining, the hemodynamics were monitored, and lactate dehydrogenase (LDH) levels in the coronary effluent, manganese superoxide dismutase (Mn-SOD) content in mitochondria and mitochondrial membrane potential were measured spectrophotometrically. The ultrastructure of cardiomyocyte mitochondria was assessed by electron microscopy.Key findings: NS1619 (10 mu M) improved heart function, decreased infarct size, reduced LDH release, maintained mitochondrial structural integrity and mitochondrial membrane potential, and increased the mitochondrial content of Mn-SOD to the same degree as RPC treatment. However, paxilline (1 mu M) eliminated the cardioprotective effect conferred by RPC.Significance: The mitochondrial Ca2+-activated K+ channel participates in the myocardial protection by limb remote ischemic preconditioning. [Involvement of Potassium Channel in Hemin-Induced Cardioprotection in Rat Hearts].

Abstract:OBJECTIVE:To investigate the effects of heme oxygenase 1 inducer hemin on protection of ischemia-reperfusion injury in rats and its mechanisms.METHODS:The Langendorff model of isolated rat heart was used; the left anterior descending coronary artery was occluded for 30 min and subsequently reperfused for 2 h. Then the ventricular function and infarct size were measured.RESULT:Hemin preconditioning prevented the increase in LVEDP, decrease in LVDP and +/- dp/dt(max) in the isolated ischemia-reperfusion rat hearts. The leakage of LDH and CK in the coronary effluent was significantly declined in hemin-treated rat hearts. And the infarct size was also reduced. Administration of a blocker of mitochondrial ATP-sensitive potassium channel (mitoK(ATP)) 5-HD (5 mg/kg) before hemin preconditioning increased the LVEDP, and reduced the LVDP and +/- dp/dt(max). The leakage of LDH and CK in the coronary effluent and the infarct size were also increased compared with only hemin-treated rat hearts. Pretreatment of the rats with a blocker of sarcolemmal ATP-sensitive potassium channel (sarcK(ATP)) HMR-1098 (6 mg/kg) before hemin preconditioning also abolished the protective effect. Infusion of paxilline (1 micromol/L), a blocker of calcium activated potassium channel (K(Ca)) for 10 min before ischemia/reperfusion led to larger infarct size and poorer myocardial performance as compared with the hemin group. The leakage of LDH and CK in the coronary effluent was also increased.CONCLUSION:Both mitoK(ATP)and sarcK(ATP)channels activation are required for the delayed cardioprotection induced by hemin. The opening of K(Ca) channels-dependent mechanism may be involved in the protection.
Ischemic Preconditioning Delays Ischemia-Induced Cellularelectrical Uncoupling in Rat Heart

METHODS: Adult rat hearts perfused on a Langendorf f apparatus were subjected to 40 min ischemia followed by 30 min reperfusion. C han ges in coupling were monitored by measuring whole-tissue resistance. RES ULTS: IP C delayed the onset of uncoupling campared to ischemic control; Blocking mitoK ATP channels before the IPC protocol abolished the delay of uncoupling. The specif ic mitoK ATP channel opener diazoxide mimicked the protective effect of IPC . The delay induced by diazoxide was reduced by 5-HD, L-type Ca 2+ channel inhibitor verapamil and a free radical scavenger N-(2-mercaptopropionyl)glycine. CONCLUSIONS: IPC delays the onset of cellular electrical uncoup ling induced by acute ischemia, in which activation of the mitoK ATP channe ls may be involved.
[Myocardial Protective Effect of Acetylcholine Against Ischemia/reperfusion Injury and Its Underlying Mechanism].

Abstract:OBJECTIVE:To determine whether the caidioprotection of acetylcholine (ACh) against ischeniia/reperftision (I/R) injury is re-kited to mitochondrial permeability transition pore (MEW) and mitochondrial AW-sensitive potassium channel (mitoK(ATP)).METHODS:Male Sprague-Dawley rats were used for Langendorif isolated bean perkision. The hearts were subjected to global ischemia for 30 mm followed by 120 rein of reperfusion and the left ventricular hemodynaniic parameters were measured. Formazan, a product of 2,3, 5-triphenyl-tetrazolium chloride (TTC), which is proportional to myocardial viability, was measured at 490 nm, and the level of lactate dehydrogenase (LDH) in the coronary effluent was measured to evaluate the cardiac injury.RESULTS:The pretreatment with ACh (0.1 mol/L, 5 mm) before I/R markedly increased myocardial formazan content, reduced LDH release, improved the recovery of the left veritficular developed pressure, +/- dP/dtmax, and rate pressure product (left ventricular developed pressure multiplied by hean rate) and attenuated the decrease of coronary flow during reperfusion. The opener of MPTP, atiractyloside (20 mmoL/L) or the inhibitor of mitoK(ATP), 5-hydroxydecanoate (100 micromol/L) abolisbed the beneficial effect of ACh.CONCLUSION:In the isolated rat bean, ACh protects myocardium against ischemia/reperfusion injury via inhibiting the opening of MPTP and increasing the opening of mitoKATP in heart.
[Angiotensin-Converting Enzyme Inhibitors Potentiate Subthreshold Preconditioning Through NO and Mitok(atp) Channel.].

Abstract:The aim of the present study was to determine whether angiotensin-converting enzyme inhibitors (ACEI) could contribute to the protective effects of preconditioning, and to explore its underlying mechanism. The Langendorff model of isolated rat heart was used. Cardiac contractility and lactate dehydrogenase (LDH) in the coronary effluent were measured, and infarct area of hearts after 30 min of ischemia followed by 120 min of reperfusion was analyzed. We found that: (1) The subthreshold preconditioning (2 min of ischemia and 10 min of reperfusion), captopril (an ACEI with sulfhydryl groups) or perindoprilate (an ACEI without sulfhydryl groups) alone did not protect the hearts from being injured by 30 min of ischemia and 120 min of reperfusion. (2) However, the combination of captopril or perindoprilate with subthreshold preconditioning could decrease left ventricular end-diastolic pressure (LVEDP), increase left ventricular developed pressure (LVDP) and coronary flow compared with the subthreshold preconditioned group. The combination treatments also inhibited the release of LDH from ischemia/reperfusion hearts, and reduced the infarct area in ischemic heart after 2 h of reperfusion (P<0.05). (3) By using NOS inhibitor L-NAME (100 mumol/L) before combined administration of ACEI with subthreshold preconditioning, the protection effect triggered by the combination treatment was significantly reduced. Pretreatment of the hearts with mitochondrial ATP-sensitive potassium (mitoK(ATP)) channel inhibitor 5-HD (100 mumol/L) also abolished the protection effect (P<0.05). (4) Subthreshold preconditioning, captopril or perindoprilate alone could enhance the NO content in coronary effluent (P<0.05), but the combination of captopril or perindoprilate with subthreshold preconditioning could further augment the NO content compared with the subthreshold preconditioned group (P<0.05). The results indicate that ACEIs with or without sulfhydryl groups may potentiate the subthreshold preconditioning to trigger cardiac protection effect against the ischemia/reperfusion injury. This protection effect in the heart is possibly mediated by the generation of NO and the activation of mitoK(ATP) channel.
CARDIOPROTECTIVE EFFECTS OF NOS AND PKC UNDER HEMIN INDUCED ISCHEMIA/REPERFUSION INJURY IN RAT HEARTS

Abstract:AIM:Whether hemin, a heme oxygenase 1 (HO-1) inducer, reduces ischemia/reperfusion injury and whether NO synthase (NOS) and PKC are involved in the cardioprotective effects were investigated in the present study.METHODS:The Langendorff model of isolated rat heart was used. The ventricular function, infarct size, LDH and CK during ischemia/reperfusion period were also observed.RESULTS:(1) After intraperitoneal injection of hemin (50 mg/kg) for 24 h, COHb concentration in rat blood enhanced. He-min preconditioning prevented the increase in LVEDP, decrease in LVDP and +/- dP/dt(max) in the isolated ischemia/reperfusion (ischemia for 30 main and subsequent reperfusion for 2 h) rat hearts. The leakage of LDH and CK in the coronary effluent was significantly declined in hemin-treated rat hearts. And the infarct size was als reduced. (2) By using an inhibitor of NOS NG-nitro-L-arginine methyl ester before the administration of hemin could inhibit the protection induced by hemin. (3) Administration of an inhibitor of protein kinase C chelerythrine (1 mg/kg) before hemin preconditioning could also abolish the cardioprotection induced by hemin.CONCLUSION:These data suggest that the involvement of NO synthase and protein kinase C have been implicated in hemin-induced delayed cardioprotection in isolated rat hearts.

Abstract:Aim: To investigate the role and mechanism of mitochondrial calcium uniporter (MCU) in myocardial hypoxia/reoxygenation injury. Methods: Isolated rat hearts were perfused with Langendorff apparatus. The hypoxia/reoxygenation injury was achieved by ligation of left anterior coronary artery for 30 min followed by release of ligation for 120 min. The left ventricular developed pressure (LVDP), the maximum rise/fall rate of left ventricular pressure (+/- dP/dt(max)), and the left ventricular end-diastolic pressure (LVEDP) were recorded. Activities of lactate dehydrogenase (LDH) in coronary effluent and reactive oxygen species (ROS) of myocardial mitochondria were spectrophotometrically assayed. Infarct size was determined by TTC staining method. Results: Compared with the hypoxia/reoxygenation (H/R) group, ruthenium red (RR, 5 micromol/L), given at the on set of reoxygenation, significantly improved the contractile function of left ventricle, decreased the myocardial infarct size, alleviated the production of ROS in myocardial mitochondria and LDH release in coronary effluent. Spermine (20 micromol/L), given at the onset of reoxygenation, enhanced the production of ROS in the mitochondria and LDH release in coronary effluent at 5, 20 and 30 min of reoxygenation, however, there were no significant differences of ventricular contractile parameters and infarct size between groups subjected to hypoxia/reoxygenation with or without spermine treatment. Co-treatment of ROS scavenger N-2-mercaptopropionyl glycine (1 mmol/L) with spermine abolished the effect of spermine. Conclusion: Inhibition of mitochondrial calcium uniporter may refrain heart from hypoxia/reoxygenation injury via decreasing the production of ROS in heart mitochondria.

Abstract:Time-resolved resonance-enhanced Stokes and anti-Stokes Raman spectra of the thiocyanate dimer radical anion, (SCN)2•-, prepared by pulse radiolysis in water, have been obtained and interpreted in conjunction with theoretical calculations to provide detailed information on the molecular geometry and bond properties of the species. The structural properties of the radical are used to develop a molecular perspective on its thermochemistry in an aqueous solution. Twenty-nine Stokes Raman bands of the radical observed in the 120-4200 cm-1 region are assigned in terms of the strongly enhanced 220 cm-1 fundamental, weakly enhanced 721 cm-1, and moderately enhanced 2073 cm-1 fundamentals, their overtones, and combinations. Calculations by range-separated hybrid density functionals (ωB97x and LC-ωPBE) support the spectroscopic assignments of the 220 cm-1 vibration to a predominantly SS stretching mode and the features at 721 cm-1 and 2073 cm-1 to CS and CN symmetric stretching modes, respectively. The corresponding bond lengths are 2.705 (±0.036) Å, 1.663 (±0.001) Å, and 1.158 (±0.002) Å. A first order anharmonicity of 1 cm-1 determined for the SS stretching mode suggests a convergence of vibrational states at an energy of ∼1.5 eV, using the Birge-Sponer extrapolation. This value, estimated for the radical confined in solvent cage, compares well with the calculated gas-phase energy of 1.26 ± 0.04 eV required for the radical to dissociate into SCN• and SCN- fragments. The enthalpy of dissociation drops to 0.60 ± 0.03 eV in water when solvent dielectric effects on the radical and its dissociation products upon S-S bond scission are incorporated in the calculations. No frequency shift or spectral broadening was observed between light and heavy water solvents, indicating that the motion of solvent molecules in the hydration shell has no perceptible effect on the intramolecular dynamics of the radical. The Stokes and anti-Stokes Raman frequencies were found to be identical within the experimental uncertainty, suggesting that the frequency difference between the thermally relaxed and spontaneously created vibrational states of (SCN)2•- in water is too small to be observable.
Protective Effects of Remote Ischemic Preconditioning in Rat Hindlimb on Ischemia- Reperfusion Injury

Abstract:Three cycles of remote ischemic pre-conditioning induced by temporarily occluding the bilateral femoral arteries (10 minutes) prior to 10 minutes of reperfusion were given once a day for 3 days before the animal received middle artery occlusion and reperfusion surgery. The results showed that brain infarct volume was significantly reduced after remote ischemic pre-conditioning. Scores in the forelimb placing test and the postural reflex test were significantly lower in rats having undergone remote ischemic pre-conditioning compared with those who did not receive remote ischemic pre-conditioning. Thus, neurological function was better in rats having undergone remote ischemic pre-conditioning compared with those who did not receive remote ischemic pre-conditioning. These results indicate that remote ischemic pre-conditioning in rat hindlimb exerts protective effects in ischemia-reperfusion injury.

Abstract:B-type natriuretic peptide (BNP) is a regulatory autacoid in the mammalian myocardium, whose functions play significant roles in health and disease. Previous work has identified that BNP protect myocardium through mitochondrial pathway-dependent mechanism against ischemia-reperfusion (I/R) injury. Mitochondria are both essential effectors of cardioprotection and primary targets of cardioprotective signaling. In particular, mitochondrial channel are activated to act as the major determinants of cell life and death. Since the discovery of mitochondrial calcium uniporter (MCU), MCU has been its contribution to cardiomyocytes under specific physiological or pathological conditions. The role of mitochondria and MCU, in mediating reperfusion-induced heart injury is a novel investigative area. In addition, the relationship of BNP with MCU in cardiomyocytes undergoing reperfuison is unclear. In this study, we used cultured neonatal rat cardiomyocytes to investigate the effect of BNP on MCU during reperfusion, the well-characterized pathological process of heart diseases. Our results demonstrated that treatment with BNP protected cardiomyocytes from apoptosis against I/R injury. Further investigation of underlying mechanisms revealed that BNP could partly prevent opening of mitochondrial calcium uniporter during I/R. And these mechanisms were associated with BNP-attenuated dissipation of mitochondrial membrane potential (Deltapsi(m)), generation of reactive oxygen species (ROS). BNP also increased the level of anti-apoptotic Bcl-2 protein, decreased the expressions of pro-apoptotic Bax and Smac/DIABLO. In summary, we demonstrated that BNP exerts protective functions within reperfusion by blocking mitochondrial calcium uniporter. Our findings also suggested that phosphatidylinositol 3-kinase (PI3K) dependent pathway may be involved in the actions of BNP.
Mitofusin-2 Mediated Mitochondrial Ca2+ Uptake 1/2 Induced Liver Injury in Rat Remote Ischemic Perconditioning Liver Transplantation and Alpha Mouse Liver-12 Hypoxia Cell Line Models.

Abstract:AIM To investigate the protective mechanism of mitofusin-2 (Mfn2) in rat remote ischemic perconditioning (RIC) models and revalidate it in alpha mouse liver-12 (AML-12) hypoxia cell lines. METHODS Sprague-Dawley rats were divided into three groups (n = 6 each): sham, orthotopic liver transplantation and RIC. After operation, blood samples were collected to test alanine aminotransferase and aspartate aminotransferase. The liver lobes were harvested for histopathological examination, western blotting (WB) and quantitative real-time (qRT)-PCR. AML-12 cell lines were then subjected to normal culture, anoxic incubator tank culture (hypoxia) and anoxic incubator tank culture with Mfn2 knockdown (hypoxia + Si), and data of qRT-PCR, WB, mitochondrial membrane potential (Delta psi m), apoptosis, endoplasmic reticulum Ca2+ concentrations and mitochondrial Ca2+ concentrations were collected. RESULTS Both sham and normal culture groups showed no injury during the experiment. The RIC group showed amelioration of liver function compared with the orthotopic liver transplantation group (P < 0.05). qRTPCR and WB confirmed that Mfn2-mitochondrial Ca2+ uptake 1/2 (MICUs) axis was changed (P < 0.005). In AML-12 cell lines, compared with the hypoxia group, the hypoxia + Si group attenuated the collapse of..m and apoptosis (P < 0.005). The endoplasmic reticulum Ca2+ decrease and mitochondrial Ca2+ overloading observed in the hypoxia group were also attenuated in the hypoxia + Si group (P < 0.005). Finally, qRT-PCR and WB confirmed the Mfn2-MICUs axis change in all the groups (P < 0.005). CONCLUSION Mfn2 participates in liver injury in rat RIC models and AML-12 hypoxia cell lines by regulating the MICUs pathway.
