Abstract:OBJECTIVE To synthesize resveratrol analogues,and evaluate their estrogen-like activity in vitro.METHODS The target compounds(1-9) were prepared from substituted benzyl bromide via phosphorylation,condensatiom and demethylation,then their estrogen-like activities on sensitive human mammary cancer(MCF-7) cell were determined by MTT assay. RESULTS All target compounds were confirmed by mp and 1HNMR. All resveratrol analogues had different estrogenic activity.The results of relative proliferative effect(RPE) suggested that resveratrol,2,4 were full-agonists of estrogen receptor;3,7,8,9 were partial-agonists of estrogen receptor,1,5,6 were weakly-agonists of estrogen receptor.Of the total,2 showed the strongest effect in promoting MCF-7 cell growth.CONCLUSION It was the first report that resveratrol analogues had estrogen-like activity and they could be a new source of estrogenic agent.

Abstract:Receptors are proteinous macromolecules which remain in the apo form under normal/unliganded conditions. As the ligand approaches, there are specific stereo-chemical changes in the apo form of the receptor as per the stereochemistry of a ligand. Accordingly, a series of substituted dimethyl-chroman-based stereochemically flexible and constrained Tamoxifen analogs were synthesized as anti-breast cancer agents. The synthesized compounds 19a-e, 20a-e, 21, and 22a-e, showed significant antiproliferative activity against estrogen receptor-positive (ER + , MCF-7) and negative (ER - , MDA MB-231) cells within IC 50 value 8.5–25.0 μM. Amongst all, four potential molecules viz 19b , 19e , 22a , and 22c , were evaluated for their effect on the cell division cycle and apoptosis of ER + and ER - cancer cells (MCF-7 & MDA MB-231cells), which showed that these compounds possessed antiproliferative activity through triggering apoptosis. In-silico docking experiments elucidated the possible affinity of compounds with estrogen receptors-α and -β.

Abstract:Utilizing molecular modeling techniques and structure-activity relationship data from the literature, a series of 2- and 3-substituted chromones and related heterocycles have been designed and synthesized as retinoic acid receptor ligands. The compounds were prepared using known coupling reactions and Wittig-Homer-Emmons reaction conditions. These compounds were then evaluated for affinity to retinoic acid receptor subtypes. Several of the compounds reported herein were found to bind with moderate affinity to the target receptors.[GRAPHICS]

Abstract:Two series (14a–d and 21a–h) of novel spin-labeled combretastatin derivatives were synthesized and evaluated for cytotoxicity against four tumor cell lines (K562, SGC-7901, Hela and HepG-2). Simultaneously, a representative compound 21a was selected to investigate the antitumor mechanisms of these synthetic compounds. The results indicated that some of the compounds showed significant cytotoxicity against four tumor cell lines in vitro and were more active than etoposide, a clinically available anticancer drug. Among the newly synthesized compounds, 21a, 21b and 21c displayed the greatest cytotoxicity against three tested tumor cell lines (HEPG-2, BGC-832 and Hela), with IC50 values ranging from 0.15 to 1.05μM, compared with values of 0.014–0.403μM for 3-amino-deoxycombretastatin A-4 (3). In addition, the mechanistic analysis revealed that compound 21a effectively interfered with tubulin dynamics to prevent mitosis in cancer cells, leading to cell cycle arrest and, eventually, dose dependent apoptosis.

Abstract:Aromatase inhibitors are among the most effective treatment of the breast cancer. Aromatase catalyzes estrogen biosynthesis, which is a long‐term cause of breast cancer. Current study describes the synthesis, purification of 26 new fluorinated and non‐fluorinated thiourea derivatives of desloratadine (5), and their aromatase inhibition activity, cytotoxicity against cancer cell line (MDA‐MB‐231). Compounds 7v and 7l exhibited a significant anti‐aromatase activity, while compounds 7a, 7g‐h, 7m and 7u were also significant active against MDA‐MB‐231 cell line. Furthermore, the molecular docking studies revealed that active compounds form key interactions with the crucial amino acid of aromatase active site including TRP224, LEU477, CYS437, ALA438, MET374, ARG115, ILE305, and PHE221, which are responsible for the binding interaction of aromatase. All analogues were new, except 7b and 7k and also lacked cytotoxicity BJ human fibroblasts, with the exception of 5 and 7x. This selectivity makes this series particularly interesting for further studies.

Abstract:To reduce the endometrial toxicity and improve the efficacy of current selective estrogen receptor modulators used in breast cancer treatment by enhancing ERβ selectivity, inspired by active resveratrol oligomer, a series of analogs (5a–f, 6a–b, 7a–d, 8a–d) were designed, synthesized and biologically evaluated. Among them, the chiral indone analog (2R,3R)-8a exhibited best antiproliferative activity against both breast cancer cell lines (MDA-MB-231 and MCF-7) and better safety profile on uterus than tamoxifen. Analog (2R,3R)-8a demonstrated good binding affinity and selectivity toward ERβ, which was further proved by both molecular docking and radiometric competitive binding assay. Other studies for (2R,3R)-8a also have been explored including cell cycle and apoptosis evaluation and in vitro metabolic stability studies. These results demonstrated that (2R,3R)-8a could be a promising lead compound for future exploration of selective ERβ anti-breast cancer agents.
Integrated "3+1" oxorhenium(V) complexes as estrogen mimics

Abstract:The diagnosis and staging of breast cancer could be improved by the development of imaging radiopharmaceuticals that provide a noninvasive determination of the estrogen receptor (ER) status of tumor cells. Toward this goal, we have synthesized a number of integrated "3+1" oxorhenium(V) complexes designed to mimic estradiol and a class of nonsteroidal estrogens, the tetrahydrochrysenes (THC). The monodentate component of the estradiol mimic is a p-hydroxyphenethyl thiol ligand with ethyl substituents at the benzylic and homobenzylic positions. Model complexes of this ligand were easily made, but steric hindrance of the secondary thiol prevented the formation of the complex with the disubstituted ligand. The three "3+1" oxorhenium(V) complexes prepared to mimic the THC class mimics represent the first pyridinedithiol rhenium complexes of their kind to be made. These complexes are quite stable to air and moisture. The target tridentate ligand was prepared from chelidamic acid, and the VT NMR of the rhenium complex displays interesting fluxional behavior. The binding affinities of these complexes for the estrogen receptor are low, and their lipophilicities are rather high. Nevertheless, our findings provide a further refinement of our understanding of ligand structure-binding affinity correlations for the estrogen receptor.

Abstract:A series of versatile phthalocyanine precursors, consisting of gem-dibromovinyl phthalonitrile derivatives, were synthesized and characterized by various spectroscopic techniques. The assigned structures were further supported by X-ray crystallographic techniques. The crystal structures showed primarily C-H···N, C-H···Br, C-Br···N and π=π interactions. The overall intermolecular interactions in the structures were quantified and fully described by Hirshfeld surface analysis. In addition, energy-frame calculations were used to analyse three-dimensional topology of the crystal packing. The dispersion energy framework of all compounds was shown to be dominant over the electrostatic framework.

Abstract:The modification of the 7-position in the estradiol structure has drawn significant attention from pharmacologists. In this paper, we synthesize various amine derivatives of estradiol, functionalized with a side chain at the 7-position. The anti-tumor activities of target compounds were evaluated using MTT assay. As the side chain is alkyl amides or halogen atoms substituted alkyl amine, the compounds exhibit excellent activity, with short chains being more active than long chains. Additionally, we studied the antitumor mechanism of the 7-substituted estradiol amide compounds. Compounds 9o can effectively inhibit the proliferation and migration of MCF-7 cells and induce early apoptosis in breast cancer tumors under certain concentration conditions.

Abstract:The influence of electrolytes upon plant and animal cells has been studied by a great number of observers; and the literature of the subject has been so well reviewed by Falk (1923) .that it requires no detailed summary here. Abstract:A new synthetic route leading to the titled compounds by three steps from estriol (I) has been developed. When estriol 3-benzoate (IIa) and methyl acetobromoglucuronate were stirred in benzene in the presence of silver carbonate, the condensation reaction occurred at C-16 and C-17 yielding two isomeric glucuronide derivatives (IIIa, IVa) in approximately equal amount. Upon alkaline hydrolysis these two were readily converted into the desired estriol monoglucuronides (V, VI). The structures of IIIa and IVa were further confirmed by transforming into the 16, 17-ketol derivatives (VII, VIII) by Jones oxidation. The yield and composition of the product in Koenigs-Knorr reaction were significantly influenced by the solvent as was shown in Table I. Application of the present method to the preparation of estriol-6, 7-3H 16-and 17-glucuronides has also been described.

Abstract:Starting from 3-methoxyestra-1,3,5(10),16-tetraene-17-carbaldehydes of natural (13β) and epimeric (13α) series, a series of isomeric 3-hydroxy-17-hydroxymethylestra-1,3,5(10)-trienes, including those containing 16α,17α-annulated cyclopropane and cyclohexane ring D', were prepared using the Corey-Chaykovsky and Diels-Alder reactions followed by reduction-demethylation with diisobutylaluminum hydride and hydrogenation. Target compounds showed antiproliferative effects on MCF-7 breast cancer cells to varying degrees superior to that on MCF-10A cells, in low micromolar concentrations. The ER α -mediated luciferase reporter gene assay demonstrated that obtained steroids without an additional carbocycle or with a cyclopropane 16α,17α-annulated carbocycle are effective ER α activators. In this test, steroids of the natural configuration showed high activity at both 10 nM and 100 nM concentrations, whereas 13α-steroids showed a strong dose-dependent effect, surpassing their natural counterparts at a concentration of 100 nM. The 13β-steroid bearing additional 16α,17α-cyclohexane ring had low activity in the test. A simple docking approach using AutoDock Vina was used as a test for a preliminary assessment of the estrogenicity of the compounds. The scope of its applicability and limitations were shown using examples of synthesized molecules.

Abstract:This work explores the biological evaluation of novel cyanomethyl vinyl ether derivatives as antiproliferative agents. Tubulin, crucial to microtubule structure and function, is a target for cancer therapies. In vitro cytotoxicity assessments revealed significant activity in SKOV3 ovarian carcinoma cells and A549 lung carcinoma cells. Structure-Activity Relationship (SAR) analysis indicated that the E isomer and specific substitutions influenced the biological activity. Computational assays predicted favorable ADME properties, highlighting potential as anticancerous agents. Molecular docking studies demonstrated that compound 12E , with the E geometry of the double bond and fused polyaromatic rings such as phenanthrene, has robust interaction with tubulin, suggesting enhanced stability due to diverse amino acid interactions. Comparative spatial distributions with colchicine further indicated potential mechanistic similarities.

Abstract:Phenotypic analysis was performed on six mutants of Saccharomyces cerevisiae deleted in one of the following open reading frames (ORFs), located on chromosome II: YBR254c, YBR255w, YBR257w, YBR258c, YBR259w and YBR266c. Disruption of the ORFs was carried out in the diploid strain FY1679 using the kanMX4 marker flanked by short sequences homologous to the target locus. Tetrad analysis following sporulation of the heterozygous disruptants showed that YBR254c and YBR257w are essential genes. YBR257w was later characterized and renamed POP4, its gene product being involved in 5.8S rRNA and tRNA processing (Chu et al., 1997). The tetrad analysis performed for the heterozygous disruptant for YBR266c showed that two of the four viable spores gave colonies of smaller size, reflecting a slower growth rate. Growth analysis of the disruptant haploids confirmed this defect at 30°C and also at 15°C. However, complementation tests failed to confirm that the deletion of YBR266c is responsible for this growth defect. Growth analysis of the disruptant haploid ybr255wΔ showed a slower growth rate on YPD and minimal medium at 15°C. Finally, no phenotypic effect could be detected associated to the disruption of ORFs YBR258c and YBR259w. Copyright © 1999 John Wiley & Sons, Ltd.

Abstract:3'-Methoxy-E-diethylstilbestrol (2), with the structural and original similarities to 2-methoxyestradiol (2-ME2, 1), was synthesized and screened against HUVEC and a series of human cancer cell lines including RL95-2, SKOV-3, MCF-7 and T-47D in vitro. The configuration of the title compound was determined via the single crystal X-ray diffraction of its benzoyl-ester derivative (10). The fact that 3'-methoxy-E-diethylstilbestrol and its analogues (8 and 11) showed potential antiangiogenesis and anti-tumor activities at a close level, whereas its ester derivative (10) did not display any cytotoxic activities on all the screening cell lines indicated that the core scaffold of 3'-methoxy-3,4-diphenylhexane and the exposed hydroxyl-groups in the structures are essential pharmacophores for their anti-tumor activities.
Exploration and biological evaluation of 20‐vinyl pregnenes: A step forward toward selective modulators of the estrogen receptor α signaling for breast cancer treatment

Abstract:A structurally novel selective modulator of the estrogen receptor α signaling pathway based on the 20‐keto‐pregnene scaffold was discovered in a series of 20‐keto‐Δ21,22 steroids. The lead compound was selective against breast cancer cells, including 4‐hydroxytamoxifen‐resistant cells. It acts as an effective modulator of the ERK, cyclin D1, and CDK4 pathways and as a potent selective agent blocking estrogen receptor α. A series of D‐ring modified steroids bearing a vinyl ketone pendant were synthesized and evaluated for antiproliferative activity against breast cancer cell line and cytochromes P450. The lead compound, 21‐vinyl 20‐keto‐pregnene (2f) (IC50 = 2.4 μM), was shown to be a promising candidate for future anticancer drug design, particularly against estrogen receptor α (ERα)‐positive breast cancer. The lead compound was found to have a significant effect on the signaling pathways in parental and 4‐hydroxytamoxifen‐resistant cells. Compound 2f modulated the ERK, cyclin D1, and CDK4 pathways and blocked the expression of ERα, the main driver of breast cancer growth. Compound 2f significantly reduced 17β‐estradiol‐induced progesterone receptor expression. Accumulation of cleaved poly(ADP‐ribose) polymerase in cells treated with compound 2f indicated induction of apoptosis. The selectivity analysis showed that lead compound 2f produces no significant effects on cytochromes P450, CYP19A1, CYP21A2, and CYP7B1.

Synthesis and Antitumor Evaluation of Novel Hybrids of Phenylsulfonylfuroxan and Estradiol Derivatives

Abstract:Abstract Fifteen novel furoxan‐based nitric oxide (NO) releasing hybrids of estradiol derivatives were synthesized and evaluated in vitro anti‐proliferative activity in MDA‐MB‐231, A2780, Hela and HUVEC cell lines. Most of them displayed potent anti‐proliferative effects. Among the compounds, 4‐bromo‐3‐((phenylsulfonyl)‐1,2,5‐oxadiazole 2‐oxide)‐oxy‐propoxy‐estradiol (11 b) exhibited the best activity with IC50 values of 3.58–0.0008 μM. Preliminary pharmacological studies showed that 11 b induced apoptosis and hardly affected the cell cycle of MDA‐MB‐231 cell line. NO‐releasing capacity and inhibition of ERK/MAPK pathway signaling might explain the potent antineoplastic activity of these compounds. The preliminary structure‐activity relationship (SAR) showed that steroidal scaffolds with a linker in 3‐position were favorable moieties to evidently increase the bioactivities of these hybrids. Overall, these results implied that 11 b merited to be further investigated as a promising anti‐cancer candidate.

Abstract:In this study, twelve 3,4‐dihydropyrimidines derivatives were synthesized through Biginelli multi‐component reaction. The efficacy of these compounds against MCF‐7, A549, and HeLa cells was evaluated using the MTT method. The results showed that designed derivatives were more effective against A549 cancer cells than MCF‐7 and HeLa cells. Compound 5l (bearing 4‐Cl‐phenyl at C4 of 3, 4‐dihydropyrimidin‐2(1H)‐one ring) was the most potent analogue (A549: 18.65±1.87μM, HeLa: 26.59±2.71 μM, MCF‐7: 31.82±2.64 μM). The presence of an electron‐withdrawing group with optimum lipophilicity at the C4 position of the phenyl ring increased the cytotoxic effect. The flow cytometry findings indicated that compound 5l induced apoptosis in A549 cancer cells in a dose‐dependent manner. Eg5 and AKT1 were selected as molecular modeling target by applying pharmacology network analyses. The molecular docking results indicated that both enantiomers of compound 5l had significant interactions with key residues in both Eg5 (Gly117 and Glu116) and AKT1 (Ala123 and Glu121) active sites. However, MD simulation revealed that the R enantiomer had a more stable complex and a higher binding affinity to the Eg5 enzyme active site than the S‐enantiomer. The affinity of 5l (R enantiomer) to Eg5 was predicted more than AKT1.

Synthesis, characterization, and anticancer activity of 4,6-dichloropyrimidine chalcone hybrids and their mechanistic interaction studies with HSA: In-silico and spectroscopic investigations

Abstract:s The pyrimidine scaffold is often considered as one of the most important pharmaceutically active chemicals. Pyrimidine functionalized with a halogen group becomes an appropriate precursor for several structural variations in the synthesis of pyrimidine-based compounds. Herein, an effort was made to design and synthesize a series of 4, 6-dichloropyrimidine chalcone derivatives of biological and pharmaceutical interest. These compounds showed anticancer potential against A549 cells under In vitro conditions observed through MTT assay. Among all the tested compounds 1c-thiourea and 1 d-urea displayed highest cytotoxic potential with IC 50 value of 91.36 μM and 128.14 μM. Further, we studied the interaction of these compounds (1c-thiourea and 1d-urea) with the human serum albumin (HSA) using UV–Vis., fluorescence emission, and far-UV-CD spectroscopic techniques. The steady state fluorescence studies show quenching in the fluorescence emission intensity of HSA in the presence of 1c-thiourea and 1d-urea compounds, which infers the binding affinity of HSA with our synthesized compounds. The observed thermodynamic parameter, i.e., Gibbs free energy (∆ G o ) of the system suggests that the binding process is spontaneous in nature. In addition, structural changes in the secondary structure of HSA induced by 1c-thiourea and 1d-urea were studied using far UV-CD. The α-helical content of HSA is increased in the presence of both the ligands indicating alterations in the secondary structure of protein. Molecular docking was performed to assess the binding affinity of all these compounds with HSA, and the results suggest strong binding with HSA through H-bonding and hydrophobic interactions. The current study provides vital information regarding the interaction of HSA with the synthesized compounds, which may provide a new path for designing novel potent 4, 6-dichloropyrimidine chalcone derivatives for use in the pharmaceutical and biomedical fields.
