Abstract:Copper (Cu) belongs to a class of heavy metals that play an essential part as a cofactor in cellular metabolism, participating in the metabolism of the respiratory chain and iron. It becomes harmful to fish above a certain concentration when its concentration reaches that of the biological requirement. Fish blood is prone to contamination and xenobiotic may be used as toxicity indexes for its alternation. This study was conducted to demonstrate the effect of copper sulfate (CuSO4) on hematological and immunological parameters of African catfish with or without garlic extract incorporation into fish feed. Materials and methods: The experiments were carried out in four groups comprising 20 fish in each group, African catfish, Clarias gariepinus. The results declared signs of anemia in the copper sulfate treated group that represented in a significant decrease of RBCs, Hb content, PCV%, MCV, WBCs, lymphocytes, eosinophils, and immunological parameters, while a significant increase in MCHC values, heterophils, and monocytes when compared with the corresponding control group. For garlic extract supplemented groups either alone or in combination with copper sulfate treatment, there was a significant increase in RBCs, Hb, PCV%, MCV, WBCs and modulating the deranged differential leukocyte count and phagocytic activity as well as a serum level of nitric oxide, lysozyme activity and IgM due to copper toxicity. The hematology and immunity of C Gariepinus can be affected negatively by CuSO4, causing numerous health and well-being disruptions. The introduction of garlic into fish feed has been found to enhance hematological parameters and immune-competent cell activity in this species, with increasing infection tolerance indicating the enhanced efficacy of garlic against the immunologic effects of CuSO4 in fish.

Abstract:Copper is a mineral that organisms need to stay healthy on one hand and on the other hand prolonged overexposure may result into various dangerous implications. The present study was outlined to assess various deleterious effects being caused to the edible fish C. punctatus exposed to an environmentally relevant concentration of copper (ERCC). For this purpose, well-acclimatized fish were classified into four groups. Group I was maintained as control while groups II, III, and IV were exposed to the ERCC (0.85 mg/L), 10% increase in ERCC (0.935 mg/L) and 20% increase in ERCC (1.02 mg/L), respectively, for 15, 30, 45 and 60d. A significant (p <0.05) reduction in Hb% and RBC count while escalation in WBC counts was documented after all exposure periods and the maximum change was recorded after the longest exposure period of 60 days, respectively. Enzymatic antioxidants viz., superoxide dismutase (SOD), catalase (CAT), glutathione reductase (GR) and lipid peroxidation (LPO) levels were significantly (p <0.05) raised and non-enzymatic antioxidant, i.e., glutathione (GSH) activity was reduced in all exposed groups in a dose-dependent manner. Severe histopathological modifications were observed in the treated fish’s gills, muscles, and kidneys. The outcomes of the present investigation substantiate that ERCC induces toxicity in fish at various levels and warn about the possible repercussions of increased ERCC in the near future as the concentration of copper continuously rises with the ever-increasing pollution.
Effect of Chinese Parsley Coriandrum Sativum on Cadmium Binding to Proteins from the Liver and Kidney of Rainbow Trout Oncorhynchus Mykiss

Abstract:Previous reports have shown that solid-form feed mixture with added Chinese parsley (CP) had no influence on either the growth or health of rainbow trout, and the cadmium (Cd) taken up by the fish was, remarkably, reduced by 12-48% in comparison with the control group. In this study, soluble fractions from the liver and kidney underwent gel filtration, and changes of Cd, Cu, and Zn concentrations over time for each fraction, and the relationship between the form of Cd that accumulated in tissues and its toxicity, were measured. A nontoxic metallothionein fraction (MTF) of 7 kDa and a high molecular weight fraction (HMF) of about 60 kDa were isolated from the soluble fractions. The induction of nontoxic MT-Cd was 1.6 times higher (p<50.05) in the CP added group than in the control group. Moreover, Cd present in the HMF from the CP addition group also increased from the 3rd week and was 1.7 times higher by the 12th week (p<0.05), even though the HMF was reported to hardly or slightly bind to Cd. These findings suggest that the interaction with several ingredients in CP may contribute to bind metallothionein (MT) and reduce the toxicity of harmful heavy metals, without effect on fish physiology.
Effects of short-term exposure to the heavy metal, nickel chloride (Nicl2) on gill histology and osmoregulation components of the gray mullet, Mugil cephalus

Abstract:The gray mullet, Mugil cephalus is an inshore and bottom-feeding fish species of Oman sea. Therefore, the gray mullet may be more exposed to heavy metal contamination, as the toxic impacts of heavy metals mullet has been reported in various studies. This study was conducted to evaluate the toxic effects of the heavy metal, nickel (as NiCl2) on osmoregulation of the gray mullet by measuring blood biochemicals, hormones, minerals and gill histology. Fish (10 fish/tank) were experimentally exposed to NiCl2 at three environmentally relevant concentrations of 5, 10 and 15 μg/l for 96 h. Then, fish were challenged with seawater (35 mg/l) for a period of 120 min. The samples (blood and gill tissue) were collected After 96 exposure to NiCl2 and during salinity challenge (30, 60 and 120 min post challenge). The plasma levels of cortisol and glucose significantly increased in NiCl2-exposed fish. In addition, cortisol increased in all experimental groups 30 min after salinity challenge and then returned gradually to the same levels as the control at 120 min post salinity challenge (PSC). The triiodothyronine (T3) and thyroxine (T4) levels significantly decreased in response to 10 and 15 μg/l NiCl2. In all groups, the thyroid hormones significantly elevated at 30 min PSC. After 30 min PSC, T3 levels in all NiCl2-exposed fish and T4 in the treatment, 10 μg/l NiCl2 remained unchanged throughout the salinity challenge. In the treatment, 5 μg/l NiCl2, T4 levels were recovered at 120 min PSC and reached the same levels as the control. Exposure of fish to high concentrations of NiCl2 and salinity stress increased the lactate levels. However, lactate levels in 5 and 10 μg/l NiCl2 groups were recovered at 120 min PSC and reached the same levels as the control. Furthermore, plasma protein increased in response to 10 and 15 μg/l NiCl2. At 30 PSC, the protein levels decreased in control and 5 μg/l NiCl2 group, while it remained unchanged in fish exposed to 10 and 15 μg/l NiCl2 throughout the salinity challenge. Exposure of fish to NiCl2 disrupted the electrolyte (Na+, Cl-) balance both before and after salinity challenge, which may be due to gill lesions induced by the heavy metal and following alternations in gill permeability. However, fish in 5 μg/l NiCl2 re-established the ionic balance in the blood at the end of salinity challenge period. The malondialdehyde (MDA) levels significantly increased in response to 10 and 15 μg/l NiCl2. The MDA levels returned to the same levels as the control group at 120 min PSC. The results of the present study showed that nickel-induced toxicity (especially at high concentrations) can reduce the osmoregulation capabilities of mullet. However, fish are able to recover from the toxic effects over time, if contamination be eliminated.
Effect of Chinese Parsley Coriandrum Sativum and Chitosan on Inhibiting the Accumulation of Cadmium in Cultured Rainbow Trout Oncorhynchus Mykiss

Abstract:Toxic heavy metals such as cadmium (Cd) accuulate in cultured fish from solid-form feed mixtures. In this paper, two natural ingredients, supposed to inhibit accumulation of heavy metals in the fish body, were investigated to develop a functional feed mixture for healthy fish. Three test diets: (i) #1, regular diet as negative control; (ii) #2, #1 +2% lyophilized Chinese parsley Coriandrum sativum (CP); and (iii) #3, #1 +3% chitosan (CT), were each fed to 50 rainbow trout Oncorhynchus mykiss once a day, at the constantrate of 1.0–1.2% of body weight for 12 weeks. No undesirable effect on fish growth was observed by an addition of either CP or CT to regular feed. Then, 10 mg Cd was added to 1 kg each of the three test diets and they were named as: (i) #4, #1 +Cd; (ii) #5, #2 +Cd; and (iii) #6, #3 +Cd, respectively. The fish given positive a control feed (diet #4) for 3 weeks accumulated Cd in the liver and kidney at approximately 0.52 mg/kg. However, Cd in ordinary muscle was found to be just above the detection limit. Upon accumulation of Cd, three groups of 50 fish were each fed test diets #4, #5, and #6 for 12 weeks at the same feed rate described above. The concentrations of Cd accumulated in the liver, kidney, and ordinary muscle of the fish were measured every 3 weeks. Contrary to the positive control fish fed diet #4, those fish fed test diets #5 and #6 accumulated 20–30% and 25–40% less Cd in the liver, respectively. Test diets were a little less effective for inhibition of Cd uptake in the kidney than that observed in the liver. These results suggest that addition of CP or CT to fish feed might be used to decrease toxic heavy metal accumulation in fish.

Abstract:Copper, like iron and zinc, is one of the most essential trace elements for organisms. Different forms of copper have distinctive and specific uses. For example, copper oxide nanoparticles (CuO-NP) are widely used in the world as a nanomaterial. Copper sulphate (CuSO4) is worldwide used as a fungicide in agriculture and as an algaecide in aquaculture. Nowadays, the increasing use of these chemicals raises concerns regarding their potential effects on the health of aquatic organisms and ecological risks. Therefore, in the present research, toxic effects of CuSO4 and CuO-NP, alone and in combination, were evaluated using biochemical markers (plasma biochemical and gill and liver oxidative stress) in freshwater fish, Oreochromis niloticus. The fish were exposed to 0.05 mg/L CuSO4, CuO-NP, and CuSO4+CuO-NP for 4 and 21 days. Especially at 21 days, CuSO4 and CuO-NP, alone and combined, generally increased plasma alkaline phosphatase, aspartate aminotransferase, alanine aminotransferase, lactate dehydrogenase, cortisol, glucose, creatinine, blood urea nitrogen, and tissue malondialdehyde while they decreased plasma total protein, and tissue superoxide dismutase, catalase, glutathione-S-transferase, glutathione reductase, and glutathione. Consequently, our results illustrate that CuSO4 and CuO-NP have similar toxic effects in fish, however, co-exposure of CuO-NP and CuSO4 is more toxic than effects of these chemicals alone.

Abstract:Abstract During present study, the copper (Cu) mediated oxidative stress was measured that induced DNA damage by concentrating in the tissues of fish, Catla catla (14.45±1.24g; 84.68±1.45mm) (Hamilton,1822). Fish fingerlings were retained in 5 groups for 14, 28, 42, 56, 70 and 84 days of the exposure period. They were treated with 2/3, 1/3, 1/4 and 1/5 (T1-T4) of 96h lethal concentration of copper. Controls were run along with all the treatments for the same durations. A significant (p 70 > 42 > 84 > 28 > 14 days while CAT activity exhibited an inverse relationship with the increase in Cu concentration. POD activity showed a significant rise with an increase in Cu exposure duration. Comet assay exhibited significant DNA damage in the peripheral erythrocytes of Cu exposed C. catla. Among four exposure concentrations, 2/3rd of LC50 (T1) caused significantly higher damage to the nuclei compared to control. Increased POD and SOD activity, as well as a decrease in CAT activity in response to Cu, demonstrates the involvement of a protective mechanism against reactive oxygen species (ROS), whereas increased ROS resulted in higher DNA damage. These above-mentioned molecular markers can be efficiently used for the biomonitoring of aquatic environments and conservation of edible fish fauna.

Abstract:Although fish feed supplemented with Chinese parsley (CP), Coriandrum sativum, does not affect the health of rainbow trout Oncorhynchus mykiss, it has the remarkable effect of reducing the uptake of Cd (12–17% compared with controls without CP). Here, we measured the change in the amount of metallothionein (MT) in the liver and kidney using gel permeation chromatography-high-performance liquid chromatography (TSK GEL SW 3000 column). The increase in MT content over time in the Cd-fed groups corresponded to the change in Cd accumulation. Gel permeation was performed with soluble fractions from the fish liver and kidney, and changes in the Cd, Cu, and Zn concentrations over time for each fraction, and the relationship between the form of Cd that accumulated in tissues and its toxicity were measured. A nontoxic MT fraction of 7,000 Da and a high-molecular-weight fraction of approximately 60,000 Da were isolated from the soluble fractions. The induction of nontoxic MT–Cd was higher in the CP-supplemented groups than in the control groups. The synergetic actions of several compounds in CP may contribute to bind MT, thereby preventing accumulation of harmful heavy metals and essential metals in fish.

Abstract:The present study was designed to test the influence of montmorillonite (MMT) on cadmium accumulation in different tissues of Carassius auratus. 180 normal healthy C auratus were randomly divided into four groups. They were fed with normal diet, normal diet supplemented with MMT (0.5%), high-cadmium (cadmium chloride, 120 mg/kg) diet and high-cadmium diet supplemented with MMT (0.5%). After 60 days, cadmium concentration in several tissues were measured by eletrothermic atomic absorption spectrophotometry, metallothionein contents were determined using the Cd-chelex assay and histopathological alterations of kidney were observed. MMT inhibited cadmium accumulation in kidney, intestine. liver, spleen, blood, gills, spermary, ovary, brain, bone and muscles of C. auratus. Metallothionein levels in liver, intestine and muscle decreased and pathological changes of kidney were reduced by MMT addition. Thus, MMT protected tissues of C. auratus from cadmium accumulation and damage. (C) 2008 Published by Elsevier B.V.
Copper-induced Genotoxicity, Oxidative Stress, and Alteration in Transcriptional Level of Autophagy-associated Genes in Snakehead Fish Channa punctatus

Abstract:Copper (Cu) is an essential and important trace element for some significant life processes for most organisms. However, an excessive amount of Cu can be highly toxic. The present study was conducted to elucidate the oxidative stress–induced alteration in transcriptional level of autophagy-related genes in the liver and kidney tissue of fish Channa punctatus after treatment with three different sublethal concentrations of CuSO 4 for 28 days. All the studied enzymatic and non-enzymatic oxidative stress markers viz. superoxide dismutase-SOD, catalase-CAT, glutathione peroxidase-GPx, glutathione reductase-GR, and glutathione-GSH showed an increase in their activity levels in the treated groups in a dose-dependent manner. Particularly SOD and CAT have shown a significant hike in activity levels. ROS levels in blood cells increased significantly ( p < 0.05) in all the treated groups, i.e., Group II-1/20 th of 96 h-LC 50 (0.2 mg/L), Group III-1/10 th of 96 h-LC 50 (0.4 mg/L), and Group IV-1/5 h of 96 h-LC 50 (0.8 mg/L) of Cu 2+ in a dose-dependent manner as compared to control (Group I). The upregulation in mRNA levels of autophagy-related genes Microtubule-associated protein 1 light chain 3 (LC3), Gamma-aminobutyric acid receptor-associated protein precursor (Gabarap), and Golgi-associated ATPase enhancer of 16 kDa (GATE16), autophagy-related 5 (ATG5) was observed while mammalian target of rapamycin (mTOR) showed downregulation in the liver and kidney tissue of fish. The decrease in mTOR and increase in ATG5 gene expression projects autophagic vesicle formation due to oxidative stress. There was significant induction in micronuclei (MN) frequency in all the treated groups. The highest frequency of MN induced by Cu 2+ was recorded in Group IV after 28 days of the exposure period. Thus, it can be concluded that the available information about Cu 2+ -induced oxidative stress–mediated autophagy in the liver and kidney of fish C. punctatus remains largely unclear to date, so to fill the aforesaid gap, the present study was undertaken, which gives an insight for the mechanisms of autophagy induced by Cu 2+ in fish.

Copper Nanoparticles Mitigate the Growth, Immunity, and Oxidation Resistance in Common Carp (Cyprinus carpio)

Abstract:The present investigation aimed to evaluate the influence of copper nanoparticles (Cu-NPs) on the growth, immunity, and oxidation resistance of common carp (3.02 ± 0.01 g, initial mean weight ± S.E.). Five groups of fish fed diets with Cu-NPs at 0, 0.5, 1, 2, and 4 mg/kg for 8 weeks. The results suggested that Cu-NPs in diets increased the growth performance and reduced FCR with linear and quadratic model (P < 0.05). Also, common carp fed Cu-NPs showed increased carcass protein, lipid, and ash contents in a dose-dependent manner (P < 0.05). The Cu accumulation in the carcass, liver, muscle, and gills increased by Cu-NPs and showed the maximum at 4 mg Cu-NPs/kg (P < 0.05). No significant alterations were found in the blood variables due to Cu-NP supplementation except for the Hb, RBCs, total protein, albumin, and globulin levels which showed the highest level in 2 mg/kg (P < 0.05). IgM level, phagocytic, lysozyme, SOD, CAT, and GPX activities were boosted by Cu-NPs with decreased malondialdehyde (MDA) content (P < 0.05). Based on regression analysis, the requirement of dietary Cu-NPs for common carp was estimated to be 2.19 to 2.91 mg/kg diet.

Administration of vitamin E and C enhances immunological and biochemical responses against toxicity of silver nanoparticles in grass carp (Ctenopharyngodon idella)

Abstract:The aim of the current study was to evaluate the toxic effect of silver nanoparticles (Ag-NPs) on biochemical biomarkers, immune responses, and the curative potential effects of vitamin C and E on grass carp. Fish (n = 420) with an average initial body weight of 8.045 ± 0.13 g were shifted to glass aquaria (36 x 18 x 18 inches, filled with 160-L tap water) in triplicates. Aquaria were randomly designated as A, B, C, D with alone Ag-NPs (Control (0), 0.25, 0.50, 0.75 mg/L) and E, F, G with Ag-NPs + Vit. C + Vit. E (0.25+0.25+0.25, 0.50+0.50+0.50, 0.75+0.75+0.75 mg/L). NPs particles were administrated viz, oral and intravenous routes for 7 days. The results indicated that both routes had non-significant effect, but levels of Ag-NPs had significant effect. Treatments C, D and G showed significant decrease in levels of RBC, HGB and HCT except for WBC and NEUT levels, which significantly increased. ALT, ALP, AST, urea, and creatinine showed significant increase in activity in the C, D, and G groups. CAT, SOD decreased significantly in all Ag-NPs alone groups, while significantly increased with vitamin E and C. LYZ, TP, ALB, GLB showed significant low activity in the B, C, and D groups while significantly high activity in the E, F, and G groups. Cortisol, glucose and triglycerides showed significant increase in the B, C, and D groups, while E, F, and G groups showed significant low levels of triglycerides, COR, and GLU. Cholesterol level was same across all treatment groups. In conclusion, vitamin E and C as powerful antioxidants protect the fish against Ag-NPs except high dose level of 0.75mg/L, while 0.25mg/L of Ag-NPs was presumably safe for C . idella .

Abstract:Copper (Cu) is a common contaminant in aquatic environments, which could cause physiological dysfunction in aquatic organisms. However, few studies have comprehensively examined the impact of copper toxicity in freshwater fish over the past decade. In this research, the oxidative stress, liver transcriptome, intestinal microbiota, and histopathology of common carp (C. carpio) in response to Cu exposure were studied, by exposing juvenile carp to 0.2 mg/ml Cu2+ for 30 days. The results revealed that Cu2+ could induce significant changes in malondialdehyde (MDA) content and antioxidant enzyme (superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx)) activity. The changes in antioxidant enzyme activities indicate that Cu can induce oxidative stress by generating reactive oxygen species (ROS) content. RNA-seq analysis of the liver identified 1069 differentially expressed genes (DEGs) after treatment with 2.0 mg/L Cu2+. Among the DEGs, 490 genes were upregulated and 579 genes were downregulated. GO functional enrichment analysis revealed that Cu could affect the fatty acid biosynthetic process, carnitine biosynthetic process, and activity of carboxylic acid transmembrane transporter. Meanwhile, the most significantly enriched KEGG pathway also included the lipid metabolism pathway. In addition, Cu2+ exposure increased bacterial richness and changed bacterial composition. At the phylum level, we found that the ratio of Bacteroidetes to Firmicutes was increased in the treatment carps, which can regulate intestinal epithelium function and reduce inflammation and immune responses. At the genus level, the abundances of 11 genera were significantly altered after exposure to Cu2+. The altered composition of the microbial community caused by Cu exposure may play a useful role in compensation of the intestinal lesions by Cu exposure. Furthermore, we found that Cu2+ exposure could cause histological alterations such as structural damage to the liver and intestines. The results of this research contribute to a better understanding of mechanisms related to Cu toxicity in fish.
The alleviative effects of green synthesized copper oxide nanoparticles against oxidative stress, hepato-renal alterations, and immune suppression induced by Staphylococcus aureus infection in Clarias gariepinus

Abstract:Copper oxide nanoparticles (CuONPs) have attracted much interest due to their catalytic and anti-bacterial activity. The present study exhibits that Moringa oleifera synthesized CuONPs (MS-CuONPs) might provide a promising anti-bacterial substitute for conventional antibiotics to treat Staphylococcus aureus infection in African catfish. MS-CuONPs showed an in vitro anti-bacterial activity (21.33 ± 0.80 mm inhibition zone) against S. aureus . One hundred-twenty fish (average weight: 120 ± 5.36 g) were haphazardly allocated into four groups in triplicates for 10 days (control, MS-CuONPs, S. aureus , and MS-CuONPs + S. aureus ), where a concentration of 0.1 mg/L of MS-CuONPs was used. The S. aureus infection induced a marked reduction in immunological (lysozyme, complement 3, and nitric oxide), protein profile (total protein, total globulins, and albumin), and hepatic antioxidant (total antioxidant capacity, superoxide dismutase, and glutathione peroxidase) indices as well as a higher mortality rate (46.67%). Moreover, an apparent elevation in the values of tumor necrosis factor-alpha (TNF-α), hepatic malondialdehyde (MDA), stress biomarkers (cortisol and glucose), and hepato-renal indicators (aspartate aminotransferase, alkaline phosphatase, alanine aminotransferase, creatinine, and urea) were the consequences of S. aureus infection. Interestingly, treatment of S. aureus- infected fish with MS-CuONPs improved these parameters. According to these outcomes, MS-CuONPs as a water additive can protect fish against the negative impacts of S. aureus and can be used a promise anti-bacterial agent.

Abstract:Aim: This study aimed to investigate the oxidative stress, genotoxicity and histopathology after sub-lethal exposure of chromium (Cr6+) and copper (Cu+2) to common food fish Channa punctatus. Metal exposure to fishes of River Ganga is a very common phenomenon due to their ubiquitous nature and through discharge of domestic and industrial waste into the river water. Methodology: An in-vitro experiment was performed to study ill effects of metallic stressors on fish. Fishes were divided into seven groups. Group I served as control, Group II, III, IV as 1/20th ,1/10th and 1/5th of 96 hr-LC50 of Cr6+. Group V, VI and VII were similarly treated with Cu at 1/20th, 1/10th and 1/5th of 96 hr LC50 of Cu+2, respectively. All the treatments were done for 15, 30 and 45 days of exposure period. Genotoxicity was evaluated by micronuclei (MN) induction. Enzymatic and non-enzymatic oxidative stress markers, including reactive oxygen species (ROS), superoxide dismutase (SOD), catalase (CAT), glutathione (GSH), glutathione reductase (GR), and glutathione peroxidase (GPx) were evaluated. Histopathological studies of liver and kidney tissue were also performed. Results: Stress biomarkers were more profound in fishes of chromium treated groups in comparison to copper. Particularly, SOD and CAT showed a significant hike in the activity levels. ROS levels in blood cells increased significantly (p < 0.05) in all the treated groups (Group II, III, and IV) in a dose dependent manner as compared with control (Group I). There was significant induction in micronuclei (MN) frequency in all the treated groups for both the metals. The highest frequency of MN induction was recorded in Group IV after 96 hr of the exposure period.Significant histological alterations were observed in liver and kidney of treated fish, changes were more pronounced in chromium treated fish than copper. Interpretation: Acute exposure of chromium and copper induces ROS and generates oxidative stress mediated genotoxicity and histopathological perturbations in the prime organs of fish.Cr and Cu induces the changes in the production and accumulation of enzymatic antioxidant system for the regulation and scavenging of ROS induced by Cr and Cu. Abstract:The effects of cadmium, hydroxylated multi-walled carbon nanotubes, and their mixture on metal accumulation and antioxidant defenses were studied using the goldfish Carassius auratus as the test organism. The fish were exposed to 0.1 mg/L Cd, 0.5 mg/L OH-MWCNTs, or 0.1 mg/L Cd+0.5 mg/L OH-MWCNTs for 3 and 12 days. Then, the Cd concentration was determined in the gill, liver and muscle. Moreover, hepatic antioxidant enzyme activity (superoxide dismutase, catalase and glutathione peroxidase), glutathione level and malondialdehyde content were also measured. A continuous accumulation of Cd was observed throughout the experimental period. Cd accumulation in tissues occurred in the following order: gill>liver>muscle at 3 days and liver>gill>muscle at 12 days. The concentrations of Cd in the livers of fish exposed to the combination of Cd+OH-MWCNTs were significantly higher than those in fish exposed to either single chemical after 12 d of exposure. Meanwhile, the mixture evoked severe oxidative stress in the exposed fish, as indicated by significant inhibition of SOD, CAT and GPx activity, a remarkable decrease in GSH level, and simultaneous elevation of MDA content. These results suggested that the effect of the combined factors on metal accumulation and oxidative stress biomarkers was more obvious than that of single factors at longer exposure durations.
Effect of unilateral nephrectomy in rabbits on pharmacokinetics of salicylate.

Abstract:The pharmacokinetics of salicylate was investigated in rabbits 6 months after unilateral nephrectomy. Sodium salicylate was given through the stomach gauge once in a dose of 20 mg/kg. Pharmacokinetic calculations assuming one compartment open model have revealed that in nephrectomized rabbits area under the curve (AUC) was increased by 131%, t0.5 was prolonged from 4.46 to 5.78 h, and kel was diminished by 40%. The plasma creatinine level remained unchanged and the remaining kidney was increased by 68%.

Abstract:Copper (Cu) contamination is serious in China, with ≤2.76 mg/L in some waters. Exposure to Cu causes a high toxicity to the aquatic organisms and subsequent ecological risk. To understand fish responses to Cu exposure, we analyzed the metabonomic changes in multiple tissues (gill, liver, and muscle) of Cyprinus flammans using an nuclear magnetic resonance-based metabonomic technique. Our results showed that metabolic alterations are dose-dependent. No significant metabolic alterations in three tissues of fish are caused by 0.25 mg/L Cu. However, 1.53 mg/L Cu caused changes of energy-related metabolites and amino acids, which we suggest are due to enhanced metabolic acidosis in gill and muscle, decreased tricarboxylic acid cycle activity in muscle, increased gluconeogenesis from amino acids in liver, and improved glycogenesis in liver and muscle. The Cori cycle between liver and muscle is concurrently triggered. Furthermore, high concentration of Cu resulted in the alteration of choline metabolism such that we hypothesize that Cu induces membrane damage and detoxification of CuSO4 in gill as well as altered osmoregulation in all three tissues. Choline-O-sulfate in gill may be used as a biomarker to provide an early warning of Cu exposure in C. flammans. Moreover, Cu exposure caused alterations of nucleoside and nucleotide metabolism in both gill and muscle. These findings provide a new insight into the metabolic effects of Cu exposure on C. flammans and highlight the value of metabonomics in the study of metabolic metal disturbance in fish.

Abstract:This study evaluated the effects of chronic exposure to copper nanoparticles (Cu-NPs) and waterborne copper (CuSO4) on the reproductive system of yellow catfish (Pelteobagrus fulvidraco). Juvenile yellow catfish were exposed to 100 and 200 μg Cu/L Cu-NPs and 100 μg Cu/L CuSO4 for 42 days. The results showed clear reproductive defects in both female and male yellow catfish in the 200 μg Cu/L Cu-NPs and 100 μg Cu/L CuSO4 groups. Exposure to Cu-NPs or CuSO4 inhibited folliculogenesis and vitellogenesis in the ovaries, and spermatogenesis in the testes, accompanied by elevation of the apoptotic signal. Ultrastructural observations also revealed damaged organelles of gonadal cells in both testes and ovaries. Most of the hypothalamic-pituitary-gonadal (HPG) axis genes examined and serum sex steroid hormones tended to be downregulated after Cu exposure. Metabolomic analysis suggested that gonadal estradiol level is sensitive to Cu-NPs or CuSO4. The heat map of gonadal metabolomics suggested a similar effect of 200 μg Cu/L Cu-NPs and 100 μg Cu/L CuSO4 in both the ovaries and testes. Additionally, metabolomics data showed that the reproductive toxicity due to Cu-NPs and CuSO4 may occur via different metabolic pathways. Cu-NPs tend to dysregulate the metabolic pathways of sphingolipid and linoleic acid metabolism in the ovary and the biosynthesis of amino acids and pantothenate and CoA in the testis. Overall, these findings revealed the toxicological effects of Cu-NPs and CuSO4 on the HPG axis and gonadal metabolism in yellow catfish.

Abstract:Copper bioavailability in the tissues of goldfish and antioxidant defenses in the liver of fish were investigated in vivo following 40 days of exposure to different species of copper solutions at different concentrations. Copper seemed to be more concentrated in the gill than in the liver after Cu(2+) and copper-ethylenediaminetetraacetate (Cu-EDTA) exposure. The addition of EDTA markedly affected copper adsorption in the gill but had no significant effects in the liver. We also found that in the fish exposed to the concentration of Cu(2+) below the Chinese national fishery water quality standard of 0.01 mg/L, superoxide dismutase (SOD) and glutathione S-transferase (GST) activities in liver were stimulated and catalase (CAT) and Se-dependent glutathione peroxidase (Se-GPx) activities were inhibited after 40 days of exposure. Compared to copper exposure, the addition of EDTA did not affect SOD and GST significantly. However, CAT and GPx activity with Cu(2+) exposure were significantly different from those with Cu-EDTA exposure.
