Abstract:Considerable progress has been made in studying the receptive fields of the most common primate retinal ganglion cell (RGC) types, such as parasol RGCs. Much less is known about the rarer primate RGC types and the circuitry that gives rise to noncanonical receptive field structures. The goal of this study was to analyze synaptic inputs to smooth monostratified RGCs to determine the origins of their complex spatial receptive fields, which contain isolated regions of high sensitivity called "hotspots." Interestingly, smooth monostratified RGCs co-stratify with the well-studied parasol RGCs and are thus constrained to receiving input from bipolar and amacrine cells with processes sharing the same layer, raising the question of how their functional differences originate. Through 3D reconstructions of circuitry and synapses onto ON smooth monostratified and ON parasol RGCs from central macaque retina, we identified four distinct sampling strategies employed by smooth and parasol RGCs to extract diverse response properties from co-stratifying bipolar and amacrine cells. The two RGC types differed in the proportion of amacrine cell input, relative contributions of co-stratifying bipolar cell types, amount of synaptic input per bipolar cell, and spatial distribution of bipolar cell synapses. Our results indicate that the smooth RGC's complex receptive field structure arises through spatial asymmetries in excitatory bipolar cell input which formed several discrete clusters comparable with physiologically measured hotspots. Taken together, our results demonstrate how the striking differences between ON parasol and ON smooth monostratified RGCs arise from distinct strategies for sampling a common set of synaptic inputs.

Abstract:A standard circuit motif in sensory systems is the pooling of sensory information from an upstream neuronal layer. A downstream neuron thereby collects signals across different locations in stimulus space, which together compose the neuron's receptive field. In addition, nonlinear transformations in the signal transfer between the layers give rise to functional subunits inside the receptive field. For ganglion cells in the vertebrate retina, for example, receptive field subunits are thought to correspond to presynaptic bipolar cells. Identifying the number and locations of subunits from the stimulus-response relationship of a recorded ganglion cell has been an ongoing challenge in order to characterize the retina's functional circuitry and to build computational models that capture nonlinear signal pooling. Here we present a novel version of spike-triggered non-negative matrix factorization (STNMF), which can extract localized subunits in ganglion-cell receptive fields from recorded spiking responses under spatiotemporal white-noise stimulation. The method provides a more than 100-fold speed increase compared to a previous implementation, which can be harnessed for systematic screening of hyperparameters, such as sparsity regularization. We demonstrate the power and flexibility of this approach by analyzing populations of ganglion cells from salamander and primate retina. We find that subunits of midget as well as parasol ganglion cells in the marmoset retina form separate mosaics that tile visual space. Moreover, subunit mosaics show alignment with each other for ON and OFF midget as well as for ON and OFF parasol cells, indicating a spatial coordination of ON and OFF signals at the bipolar-cell level. Thus, STNMF can reveal organizational principles of signal transmission between successive neural layers, which are not easily accessible by other means.

Cone bipolar cell synapses generate transient versus sustained signals in parallel ON pathways of the mouse retina

Abstract:Parallel processing is a fundamental organizing principle in the nervous system, and understanding how parallel neural circuits generate distinct outputs from common inputs is a key goal of neuroscience. In the mammalian retina, divergence of cone signals into multiple feed-forward bipolar cell pathways forms the initial basis for parallel retinal circuits dedicated to specific visual functions. Here, we used patch-clamp electrophysiology, electron microscopy and two photon imaging of a fluorescent glutamate sensor to examine how kinetically distinct responses arise in transient versus sustained ON alpha RGCs (ON-T and ON-S RGCs) of the mouse retina. We directly compared the visual response properties of these RGCs with their presynaptic bipolar cell partners, which we identified using 3D electron microscopy reconstruction. Different ON bipolar cell subtypes (type 5i, type 6 and type 7) had indistinguishable light-driven responses whereas extracellular glutamate signals around RGC dendrites and postsynaptic excitatory currents measured in ON-T and ON-S RGCs in response to the identical stimuli used to probe bipolar cells were kinetically distinct. Anatomical examination of the bipolar cell axon terminals presynaptic to ON-T and ON-S RGCs suggests bipolar subtype-specific differences in the size of synaptic ribbon-associated vesicle pools may contribute to transient versus sustained kinetics. Our findings indicate bipolar cell synapses are a primary point of divergence in kinetically distinct visual pathways.

DIFFERENCES IN PHYSICAL ACTIVITY LEVELS IN SCHOOL-BASED CONTEXTS – INFLUENCE OF GENDER, AGE, AND BODY WEIGHT STATUS

Abstract:The purposes of this study were to compare the objectively measured physical activity levels across physical education, school playtime and extra-curricular sport in secondary school students, as well as to examine the influence of gender, age, and body weight status on adolescents’ objective physical activity levels in these contexts. A hundred and two secondary school students (age 12.53±1.56 years) participated in the present study. Students’ objective physical activity levels were estimated by the multi-sensor device SenseWear Pro2 Armband during a physical education, school playtime and extra-curricular sport session. The result of the one-way multivariate analysis of covariance indicated statistically significant higher values of steps/ min, average metabolic equivalent of task, and time spent in moderate-to-vigorous physical activity in extracurricular sport than in physical education and school playtime (p<.001). Additionally, metabolic equivalent of task values were statistically significantly greater in physical education than in school playtime (p<.001). On the other hand, the results of the two-way multivariate analyses of covariance showed an influence of gender and age on physical activity levels in these contexts (p<.001), whilst for body weight status no influence was found (p>.05). This preliminary evidence could inform and guide future policy regarding the promotion of physical activity among adolescents in these important school-based contexts. Since physical activity engagement in compulsory physical activity contexts, such as physical education lessons, is clearly insufficient, parents, teachers and institutions should encourage students to achieve health-related physical activity levels during their free time.
Precisely correlated firing in cells of the lateral geniculate nucleus

Abstract:SIMPLE cells within layer IV of the cat primary visual cortex are selective for lines of a specific orientation. It has been proposed that their receptive-field properties are established by the pattern of connections that they receive from the lateral geniculate nucleus (LGN) of the thalamus1–5. Thalamic inputs, however, represent only a small proportion of the synapses made onto simple cells6–8, and others have argued that corticocortical connections are likely to be important in shaping simple-cell response properties9–11. Here we describe a mechanism that might be involved in selectively strengthening the effect of thalamic inputs. We show that neighbouring geniculate neurons with overlapping receptive fields of the same type (on-centre or off-centre) often fire spikes that are synchronized to within 1 millisecond. Moreover, these neurons often project to a common cortical target neuron where synchronous spikes are more effective in evoking a postsynaptic response. We propose that precisely correlated firing within a group of geniculate neurons could serve to reinforce the thalamic input to cortical simple cells.

Abstract:Although many details remain unknown, several positive statements can be made about the laminar distribution of primate frontal eye field (FEF) neurons with different physiological properties. Most certainly, pyramidal neurons in the deep layer of FEF that project to the brainstem carry movement and fixation signals but clear evidence also support that at least some deep-layer pyramidal neurons projecting to the superior colliculus carry visual responses. Thus, deep-layer neurons in FEF are functionally heterogeneous. Despite the useful functional distinctions between neuronal responses in vivo, the underlying existence of distinct cell types remain uncertain, mostly due to methodological limitations of extracellular recordings in awake behaving primates. To substantiate the functionally defined cell types encountered in the deep layer of FEF, we measured the biophysical properties of pyramidal neurons recorded intracellularly in brain slices issued from macaque monkey biopsies. Here, we found that biophysical properties recorded in vitro permit us to distinguish two main subtypes of regular-spiking neurons, with, respectively, low-resistance and low excitability vs. high-resistance and strong excitability. These results provide useful constraints for cognitive models of visual attention and saccade production by indicating that at least two distinct populations of deep-layer neurons exist.

Abstract:Abstract The superior colliculus is a midbrain structure that plays important roles in visually guided behaviors in mammals. Neurons in the superior colliculus receive inputs from retinal ganglion cells but how these inputs are integrated in vivo is unknown. Here, we discovered that high-density electrodes simultaneously capture the activity of retinal axons and their postsynaptic target neurons in the superior colliculus, in vivo. We show that retinal ganglion cell axons in the mouse provide a single cell precise representation of the retina as input to superior colliculus. This isomorphic mapping builds the scaffold for precise retinotopic wiring and functionally specific connection strength. Our methods are broadly applicable, which we demonstrate by recording retinal inputs in the optic tectum in zebra finches. We find common wiring rules in mice and zebra finches that provide a precise representation of the visual world encoded in retinal ganglion cells connections to neurons in retinorecipient areas.

Cone-Opponent Ganglion Cells in the Primate Fovea Tuned to Non-Cardinal Color Directions

Abstract:A long-standing question in vision science is how the three cone photoreceptor types – long (L), medium (M) and short (S) wavelength sensitive – combine to generate our perception of color. Hue perception can be described along two opponent axes: red-green and blue-yellow. Psychophysical measurements of color appearance indicate that the cone inputs to the red-green and blue-yellow opponent axes are M vs. L+S and L vs. M+S, respectively. However, the “cardinal directions of color space” revealed by psychophysical measurements of color detection thresholds following adaptation are L vs. M and S vs. L+M. These cardinal directions match the most common cone-opponent retinal ganglion cells (RGCs) in the primate retina. Accordingly, the cone opponency necessary for color appearance is thought to be established in cortex. However, small populations with the appropriate M vs. L+S and L vs. M+S cone-opponency have been reported in large surveys of cone inputs to primate RGCs and their projections to the lateral geniculate nucleus (LGN), yet their existence continues to be debated. Resolving this long-standing open question is necessary because a complete account of the cone-opponency in the retinal output is critical for efforts to understand how downstream neural circuits process color. Here, we performed adaptive optics calcium imaging to longitudinally, noninvasively measure foveal RGC light responses in the living macaque ( Macaca fascicularis ) eye. We confirm the presence of L vs. M+S and M vs. L+S neurons with non-cardinal cone-opponency and demonstrate that cone-opponent signals in the retinal output are more diverse than classically thought. Significance Statement Retinal ganglion cells (RGCs) provide the only source of visual information to the brain and thus make up the building blocks for downstream visual processing. Accordingly, a complete account of how RGCs compare the cone photoreceptors is critical for understanding color vision. The cone inputs to the most common primate RGCs establish “cardinal directions” in color space that can explain color detection, but not color appearance. We identified foveal RGCs with non-cardinal cone opponency matching the signed cone inputs to red, green, blue and yellow. While this correlation does not imply causation, it opens new directions for understanding the neural processing of color. Our results indicate that the spectral information in the retinal output is more diverse than classically thought.

Abstract:Summary In retinal neurons, morphology strongly influences visual response features. Ganglion cell (GC) dendrites ramify in distinct strata of the inner plexiform layer (IPL) so that GCs responding to light increments (ON) or decrements (OFF) receive appropriate excitatory inputs. This vertical stratification prescribes response polarity and ensures consistent connectivity between cell types, whereas the lateral extent of GC dendritic arbors typically dictates receptive field (RF) size. Here, we identify circuitry in mouse retina that contradicts these conventions. A2 amacrine cells are interneurons understood to mediate “cross-over” inhibition by relaying excitatory input from the ON layer to inhibitory outputs in the OFF layer. Ultrastructural and physiological analyses show, however, that some A2s deliver powerful inhibition to OFF GC somas and proximal dendrites in the ON layer, rendering their inhibitory RFs smaller than their dendritic arbors. This OFF pathway, avoiding entirely the OFF region of the IPL, challenges several tenets of retinal circuitry.

Abstract:Retinal ganglion cells comprise about 10 morphological types that also differ functionally. To determine whether functional differences might arise partially from differences in excitatory input, we quantified the distributions of ribbon contacts to four mammalian ganglion cell types [brisk‐transient (BT), brisk‐sustained (BS), local edge (LE), directionally selective (DS)], comparing small vs. large and “sluggish” vs. “brisk.” Cells in guinea pig retina were filled with fluorescent dye, immunostained for synaptic ribbons, and reconstructed with their ribbon contacts by confocal microscopy. False‐positive contacts were corrected by performing the same analysis on processes that lack synapses: glial stalks and rod bipolar axons. All types shared a domed distribution of membrane that was well fit by a Gaussian function (R2 = 0.96 ± 0.01); they also shared a constant density of contacts on the dendritic membrane, both across each arbor and across cell types (19 ± 1 contacts/100 μm2 membrane). However, the distributions of membrane across the retina differed markedly in width (BT > DS ≈ BS > LE) and peak density (BS > DS > LE > BT). Correspondingly, types differed in peak density of contacts (BS > DS ≈ LE > BT) and total number (BS ≈ BT > DS > LE). These differences between cell types in spatial extent and local concentration of membrane and synapses help to explain certain functional differences. J. Comp. Abstract:Synchronized firing is an efficient way for retinal ganglion cells (RGCs) to encode visual stimuli In the present study, we studied synchronized activities among RGCs in response to natural movie and pseudo-random checker-board flickering The results showed that nearby RGCs tended to fire synchronously much more frequently than expected by chance, in response to both stimuli Under our experimental conditions, synchronous groups could contain three or more cells in response to natural movie; but activities were more often observed between pair-wise cells in response to checker-board flickering The correlation index calculated between neuron pairs did not have any significant tendency of increase or decrease when natural movie stimulation was lasted; however, it tended to increase when pseudo-random checker-board flickering stimulation was lasted.
A presynaptic source drives differing levels of surround suppression in two mouse retinal ganglion cell types

Abstract:Abstract In early sensory systems, cell-type diversity generally increases from the periphery into the brain, resulting in a greater heterogeneity of responses to the same stimuli. Surround suppression is a canonical visual computation that begins within the retina and is found at varying levels across retinal ganglion cell types. Our results show that heterogeneity in the level of surround suppression occurs subcellularly at bipolar cell synapses. Using single-cell electrophysiology and serial block-face scanning electron microscopy, we show that two retinal ganglion cell types exhibit very different levels of surround suppression even though they receive input from the same bipolar cell types. This divergence of the bipolar cell signal occurs through synapse-specific regulation by amacrine cells at the scale of tens of microns. These findings indicate that each synapse of a single bipolar cell can carry a unique visual signal, expanding the number of possible functional channels at the earliest stages of visual processing.

Abstract:Abstract We consider a model of basic inner retinal connectivity where bipolar and amacrine cells interconnect and both cell types project onto ganglion cells, modulating their response output to the brain visual areas. We derive an analytical formula for the spatiotemporal response of retinal ganglion cells to stimuli, taking into account the effects of amacrine cells inhibition. This analysis reveals two important functional parameters of the network: (1) the intensity of the interactions between bipolar and amacrine cells and (2) the characteristic timescale of these responses. Both parameters have a profound combined impact on the spatiotemporal features of retinal ganglion cells’ responses to light. The validity of the model is confirmed by faithfully reproducing pharmacogenetic experimental results obtained by stimulating excitatory DREADDs (Designer Receptors Exclusively Activated by Designer Drugs) expressed on ganglion cells and amacrine cells’ subclasses, thereby modifying the inner retinal network activity to visual stimuli in a complex, entangled manner. Our mathematical model allows us to explore and decipher these complex effects in a manner that would not be feasible experimentally and provides novel insights in retinal dynamics.

Abstract:The visual image transmitted by the retina to the brain has long been understood in terms of spatial filtering by the center-surround receptive fields of retinal ganglion cells (RGCs). Recently, this textbook view has been challenged by the stunning functional diversity and specificity observed in ~40 distinct RGC types in the mouse retina. However, it is unclear whether the ~20 morphologically and molecularly identified RGC types in primates exhibit similar functional diversity, or instead exhibit center-surround organization at different spatial scales. Here, we reveal striking and surprising functional diversity in macaque and human RGC types using large-scale multi-electrode recordings from isolated macaque and human retinas. In addition to the five well-known primate RGC types, 18-27 types were distinguished by their functional properties, likely revealing several previously unknown types. Surprisingly, many of these cell types exhibited striking non-classical receptive field structure, including irregular spatial and chromatic properties not previously reported in any species. Qualitatively similar results were observed in recordings from the human retina. The receptive fields of less-understood RGC types formed uniform mosaics covering visual space, confirming their classification, and the morphological counterparts of two types were established using single-cell recording. The striking receptive field diversity was paralleled by distinctive responses to natural movies and complexity of visual computation. These findings suggest that diverse RGC types, rather than merely filtering the scene at different spatial scales, instead play specialized roles in human vision.

Abstract:Various retinal ganglion cells (RGCs) show regional adaptations, for instance, to increase visual acuity. However, for many RGC types, it is not known how they vary in their signalprocessing properties across the retina. In the mouse retina, sustained ON alpha (sONα) RGCs were found to have differences in morphology and receptive field sizes along the nasotemporal axis, and temporal sONα RGCs are likely to play a role in visually guided hunting. Thus, we hypothesised that this cell type also exhibits regional adaptations on the level of dendritic signal processing and that these adaptations are advantageous for prey capture. Here, we measured dendritic signals from individual sONα RGCs at different locations in the whole-mount mouse retina using two-photon microscopy. We measured both postsynaptic Ca signals at the dendrites of individual RGCs and presynaptic glutamate signals from bipolar cells (BCs). We found that temporal sONα RGC dendrites exhibit, in addition to the expected sustained-ON signals with only weak surrounds, signals with strong surround suppression, which were not present in nasal sONα RGCs. This difference was also present in the excitatory presynaptic inputs from BCs, suggesting a presynaptic origin. Finally, using population models in an encoder-decoder paradigm, we showed that these adaptations might be beneficial for detecting crickets in hunting behaviour.

The main results are: (1) the population RGCs tend to fire in synchrony far more frequently than expected by chance during both NM and CB stimulation; (2) more synchronous groups could be formed and each group contains more neurons under NM than CB stimulation; (3) the individual neurons also participate in more groups and have more distinct partners in NM than CB stimulation. All these results suggest that the synchronized firings in RGCs are more extensive and diverse, which may account for more effective information processing in representing the natural visual environment.

Abstract:Unlike the exhaustive determination of cell types in the retina, key populations in the lateral geniculate nucleus of the thalamus (LGN) may have been missed. Here, we have begun to characterize the full range of extracellular neuronal responses in the LGN of awake monkeys using multi-electrodes during the presentation of colored noise visual stimuli to identify any previously overlooked signals. Extracellular spike waveforms of single units were classified into seven distinct classes, revealing previously unrecognized diversity: four negative-dominant classes that were narrow or broad, one triphasic class, and two positive-dominant classes. Based on their mapped receptive field (RF), these units were further categorized into either magnocellular (M), parvocellular (P), koniocellular (K), or non-RF (N). We found correlations between spike shape and mapped RF and response characteristics, with negative and narrow spiking waveform units predominantly associated with P and N RFs, and positive waveforms mostly linked to M RFs. Responses from positive waveforms exhibited shorter latencies, larger RF sizes, and were associated with larger eccentricities in the visual field than the other waveform classes. Additionally, N cells, those without an estimated RF, were consistently responsive to the visually presented mapping stimulus at a lower and more sustained rate than units with an RF. These findings suggest that the LGN cell population may be more diverse than previously believed.

Abstract:Gap junctions are channels that allow for direct transmission of electrical signals between cells. However, the ability of one cell to be impacted or controlled by other cells through gap junctions remains unclear. In this study, heterocellular coupling between ON α retinal ganglion cells (α-RGCs) and displaced amacrine cells (ACs) in the mouse retina was used as a model. The impact of the extent of coupling of interconnected ACs on the synchronized firing between coupled ON α-RGC-AC pair was investigated using the dopamine 1 receptor (D1R) antagonist-SCH23390 and agonist-SKF38393. It was observed that the synchronized firing between the ON α-RGC-ACs pairs was increased by the D1R antagonist SCH23390, whereas it was eradicated by the agonist SKF38393. Subsequently, the signaling drive was investigated by infecting coupled ON α-RGC-AC pairs with the channelrhodopsin-2(ChR2) mutation L132C engineered to enhance light sensitivities. The results demonstrated that the spikes of ON α-RGCs (without ChR2) could be triggered by ACs (with ChR2) through the gap junction, and vice versa. Furthermore, it was observed that ON α-RGCs stimulated with 3-10 Hz currents by whole cell patch could elicit synchronous spikes in the coupled ACs, and vice versa. This provided direct evidence that the firing of one cell could be influenced by another cell through gap junctions. However, this phenomenon was not observed between OFF α-RGC pairs. The study implied that the synchronized firing between ON α-RGC-AC pairs could potentially be affected by the coupling of interconnected ACs. Additionally, one cell type could selectively control the firing of another cell type, thereby forcefully transmitting information. The key role of gap junctions in synchronizing firing and driving cells between α-RGCs and coupled ACs in the mouse retina was highlighted.NEW & NOTEWORTHY This study investigates the role of gap junctions in transmitting electrical signals between cells and their potential for cell control. Using ON α retinal ganglion cells (α-RGCs) and amacrine cells (ACs) in the mouse retina, the researchers find that the extent of coupling between ACs affects synchronized firing. Bidirectional signaling occurs between ACs and ON α-RGCs through gap junctions.

Abstract:The basic plan of the retina is conserved across vertebrates, yet species differ profoundly in their visual needs 1 . Retinal cell types may have evolved to accommodate these varied needs, but this has not been systematically studied. Here we generated and integrated single-cell transcriptomic atlases of the retina from 17 species: humans, two non-human primates, four rodents, three ungulates, opossum, ferret, tree shrew, a bird, a reptile, a teleost fish and a lamprey. We found high molecular conservation of the six retinal cell classes (photoreceptors, horizontal cells, bipolar cells, amacrine cells, retinal ganglion cells (RGCs) and Müller glia), with transcriptomic variation across species related to evolutionary distance. Major subclasses were also conserved, whereas variation among cell types within classes or subclasses was more pronounced. However, an integrative analysis revealed that numerous cell types are shared across species, based on conserved gene expression programmes that are likely to trace back to an early ancestral vertebrate. The degree of variation among cell types increased from the outer retina (photoreceptors) to the inner retina (RGCs), suggesting that evolution acts preferentially to shape the retinal output. Finally, we identified rodent orthologues of midget RGCs, which comprise more than 80% of RGCs in the human retina, subserve high-acuity vision, and were previously believed to be restricted to primates 2 . By contrast, the mouse orthologues have large receptive fields and comprise around 2% of mouse RGCs. Projections of both primate and mouse orthologous types are overrepresented in the thalamus, which supplies the primary visual cortex. We suggest that midget RGCs are not primate innovations, but are descendants of evolutionarily ancient types that decreased in size and increased in number as primates evolved, thereby facilitating high visual acuity and increased cortical processing of visual information.
