Abstract:We show that myofibrils of insect flight and leg muscle contain a doublet of polypeptides with apparent molecular weights of 700K (K = 10(3) Mr) (Hmp I) and 600K (Hmp II), respectively. In Locusta migratoria high ionic strength extraction solubilizes only Hmp II, which is readily purified in native form. It probably reflects a proteolytic derivative of the non-extractable Hmp I. On the basis of its viscosity radius and sedimentation coefficient, Hmp II has a molecular weight of 600K and seems to consist of a single polypeptide chain. The highly asymmetric structure of the molecule is confirmed by rotary shadowing. The flexible rods have a uniform diameter of 3-4 nm and an average length of 260 nm. Polyclonal antibodies show cross-reactivity between Hmp II and its putative precursor Hmp I. We discuss the similarities and differences between the larger titin I/titin II of vertebrate sarcomeric muscle and the smaller Hmp I/Hmp II of invertebrate muscle and conclude that the latter may reflect a mini-titin. In line with the smaller length, immunoelectron microscopy locates the insect mini-titin to the I band and a very short portion of the A band only, while vertebrate titin is known to connect the Z band to the M band. Mini-titin has also been purified from several other insects including Drosophila. Immunofluorescence microscopy on frozen sections shows that mini-titin is present in the sarcomeric muscles of various species from different invertebrate phyla. These include Annelida, Nematomorpha, Plathelmintha, Nemertea and Nematoda like Ascaris lumbricoides and Caenorhabditis elegans. This wide-spread occurrence of invertebrate mini-titin is confirmed by immunoblotting experiments.
Titin in insect spermatocyte spindle fibers associates with microtubules, actin, myosin and the matrix proteins skeletor, megator and chromator

Abstract:Titin, the giant elastic protein found in muscles, is present in spindles of crane-fly and locust spermatocytes as determined by immunofluorescence staining using three antibodies, each raised against a different, spatially separated fragment of Drosophila titin (D-titin). All three antibodies stained the Z-lines and other regions in insect myofibrils. In western blots of insect muscle extract the antibodies reacted with high molecular mass proteins, ranging between rat nebulin (600-900 kDa) and rat titin (3000-4000 kDa). Mass spectrometry of the high molecular mass band from the Coomassie-Blue-stained gel of insect muscle proteins indicates that the protein the antibodies bind to is titin. The pattern of staining in insect spermatocytes was slightly different in the two species, but in general all three anti-D-titin antibodies stained the same components: the chromosomes, prophase and telophase nuclear membranes, the spindle in general, along kinetochore and non-kinetochore microtubules, along apparent connections between partner half-bivalents during anaphase, and various cytoplasmic components, including the contractile ring. That the same cellular components are stained in close proximity by the three different antibodies, each against a different region of D-titin, is strong evidence that the three antibodies identify a titin-like protein in insect spindles, which we identified by mass spectrometry analysis as being titin. The spindle matrix proteins skeletor, megator and chromator are present in many of the same structures, in positions very close to (or the same as) D-titin. Myosin and actin also are present in spindles in close proximity to D-titin. The varying spatial arrangements of these proteins during the course of division suggest that they interact to form a spindle matrix with elastic properties provided by a titin-like protein.

Abstract:This article explores a strategy of mutual adaptation between the information system and the business process, facilitated by the iterative and incremental mechanisms of modern SW engineering frameworks. The concept of mutual adaptation implies that the real innovation of an information systems project is not the software, but the working solution after implementation. Too little is known about this phenomenon: What are the dynamics of mutual adaptation? To what extent is it possible to control? Which organisational mechanisms facilitate it? Should planned mutual adaptation be part of an information systems development project, or is it outside the reasonable scope of such projects? The article builds on a longitudinal case study from a large public auditing organisation that developed a new audit process in parallel with the construction of a new information system to support it. The project is described through seven iterations as it experiments with different organisational mechanisms to achieve mutual adaptation. The dynamics of mutual adaptation is described by a window of opportunity, where both the information system and the business process are malleable. Copyright © 2005 John Wiley & Sons, Ltd.

Abstract:We have cloned and sequenced a novel Bombyx mori gene that encodes a protein having a high degree of homology with other known troponin C (TnC) proteins. The amino acid sequence, DX[DN]X[DSG]X 6 E, a highly conserved putative Ca 2+ -binding motif found in loops within the globular domains of previously identified TnC proteins, is also present in BmTnC. We have expressed and purified to homogeneity a His-tagged BmTnC fusion protein having a molecular weight of approximately 21.6 kDa. We have used this purified fusion protein to produce polyclonal antibodies against BmTnC for Western blot analyses. These analyses have revealed that BmTnC is expressed in the larval head, the Malpighian tubule, the epidermis, the testis, and the gut, as has been confirmed by immunohistochemistry. In addition, real-time reverse transcription/polymerase chain reaction has shown that BmTnC mRNA levels differ substantially among these tissues. Our findings indicate that BmTnC is selectively expressed in the muscular tissues of the silkworm, including portions of the head, the Malpighian tubule, the body wall, and the gut.
Prokaryotic expression of the Nilaparvata lugens flightin gene and its differential expression in different life stages

Abstract:Flightin, located in thick filament, was originally found in the indirect flight muscle of Drosophila melanogaster. It is important in these muscles both for structural integrity and sarcomere function. Prior to this study, differences in the muscle components of macropterous and brachypterous adults of the brown planthopper( BPH, Nilaparvata lugens Stl) had not been studied. We amplified the flightin gene from the cDNA of macropterous female adults and inserted it into the expression vector pGEX-6P-1 for fusion expression with gultathione S transferase( GST). The recombinant vector was then inserted into Escherichia coli Rosseta. After optimization of the expression conditions at different temperatures and concentrations of IPTG,high level of expression of flightin was achieved. The soluble expressed GST-flightin was purified and used to immunise rabbits for preparing a polyclonal antibody against flightin. Using the prepared antibody,we searched for flightin expression in different developmental stages and in two-winged forms. We found that flightin was uniquely expressed in the adult stage and only expressed in macropterous female adults. No positive bands were detected in eggs,nymphs and brachypterous female adults. This study provides a basis for the study of wing muscle development and wing dimorphism,as well as for research into the interaction of flightin with other proteins in the BPH.
Cloning and Expression of Tachypleus Tridentatus Factor C

Abstract:Factor C is an endotoxin-sensitive, intracellular serine protease zymogen which initiates the coagulation cascade system in the horseshoe crab hemolymph. The special lipopolysaccharide (LPS) binding activation of FC makes it a potential drug for anti-LPS treatment and has a high commercial value. Based on the sequence of reported FC from Japan horseshoe crab, two pairs of primers were designed. The total RNA was extracted from amebocytes of Chinese Tachypleus tridentatus and the cDNA was separated into two parts and were cloned using RT-PCR, respectively. FCs from different geographical areas showed high homology in sequence. The whole FC cDNA was cloned into pET-28a (+) containing T7 promoter and recombinant expression plasmid pET-FC was constructed. The recombinant plasmid was transformed into E. coli BL21 (DE3). Recombinant FC was expressed as inclusion body when the expression strain was induced with 1 mmol/L IPTG. Refolded recombinant FC was confirmed to be active by bacteriostatic assay in vitro. The results of Western blot also suggested the recombinant FC may be able to cleave itself partly and produced an extra immunoblot band.
The N-terminal sequence of paratropomyosin binding fragments from beta-connectin

Abstract:In order to clarify the position where paratropomyosin binds to connectin in the A-I junction region of a sarcomere, chicken beta-connectin was digested by Staphylococcus aureus V8 protease under denaturing conditions and the digested peptides were electrophoretically separated. Five peptides, 150-kDa, 100-kDa, 70-kDa, and 43-kDa fragments, were simultaneously detected by biotinylated paratropomyosin and an anti-connectin monoclonal antibody. The N-terminal sequence of the 43-kDa fragment was found to be YQFRVYAVNK, similar to the sequence of 7556-7565 amino acids in the I51 fibronectin type 3 domain that was located at the A-I junction region of human cardiac titin/connectin. Therefore, we propose that paratropomyosin binds to the 43-kDa fragment from beta-connectin at the A-I junction region in both living muscle and in muscle immediately postmortem, and the N-terminus of the 43-kDa fragment is localized in the I51 domain.
Locusta migratoria flight muscle troponin partially activates thin filament in a calcium-dependent manner

Abstract:The troponin (Tn) complex, the sensor for Ca2+ to regulate contraction of striated muscle, is composed of three subunits, that is, TnT, TnI and TnC. Different isoforms of TnI and TnC are expressed in the thorax dorsal longitudinal muscle (flight muscle) and the hind leg extensor tibiae muscle (jump muscle) of the migratory locust, Locusta migratoria. The major Tn complexes in the flight muscle and the jump muscle are Tn-171 (TnT1/TnI7/TnC1) and Tn-153 (TnT1/TnI5/TnC3), respectively. Here, we prepared a number of recombinant Tn complexes and the reconstituted thin filaments, and investigated their regulation on thin filament. Although both Tn-171 and Tn-153 regulate thin filament in a Ca2+ -dependent manner, the extent of Ca2+ activation mediated by Tn-171 was significantly lower than that by Tn-153. We demonstrated that TnC1 and TnC3, rather than TnI5 and TnI7, are responsible for the different levels of the thin filament activation. Mutagenesis of TnC1 and TnC3 shows that the low level of TnC1-mediated thin filament activation can be attributed to the noncanonical residue Leu60 in the EF-hand-II of TnC1. We therefore propose that, in addition to Ca2+ , other regulatory mechanism(s) is required for the full activation of locust flight muscle.
The Structural Features of Skeletal Muscle Titin Aggregates

Abstract:—Titin is a multidomain protein of striated and smooth muscles of vertebrates. The protein consists of repeating immunoglobulin-like (Ig) and fibronectin-like (FnIII) domains, which are β-sandwiches with a predominant β-structure, and also contains disordered regions. In this work, the methods of atomic force microscopy (AFM), X-ray diffraction, and Fourier transform infrared spectroscopy were used to study the morphology and structure of aggregates of rabbit skeletal muscle titin obtained in two different solutions: 0.15 M glycine-KOH, pH 7.0 and 200 mM KCl, 10 mM imidazole, pH 7.0. According to AFM data, skeletal muscle titin formed amorphous aggregates of different morphologies in the above two solutions. Amorphous aggregates of titin formed in a solution containing glycine consisted of much larger particles than aggregates of this protein formed in a solution containing KCl. The "KCl-aggregates" according to AFM data had the form of a "sponge"-like structure, while amorphous "glycine-aggregates" of titin formed "branching" structures. Spectrofluorometry revealed the ability of "glycine-aggregates" of titin to bind to the dye thioflavin T (TT), and X-ray diffraction revealed the presence of one of the elements of the amyloid cross β-structure, a reflection of ~4.6 Å, in these aggregates. These data indicate that "glycine-aggregates" of titin are amyloid or amyloid-like. No similar structural features were found in "KCl-aggregates" of titin; they also did not show the ability to bind to thioflavin T, indicating the non-amyloid nature of these titin aggregates. Fourier transform infrared spectroscopy revealed differences in the secondary structure of the two types of titin aggregates. The data we obtained demonstrate the features of structural changes during the formation of intermolecular bonds between molecules of the giant titin protein during its aggregation. The data expand the understanding of the process of amyloid protein aggregation.

Binding properties of the calcium-activated F2 isoform of Lethocerus troponin C

Abstract:While in most muscles contraction is triggered by calcium effluxes, insect flight muscles are also activated by mechanical stretch. We are interested in understanding the role that the troponin C protein, usually the calcium sensor, plays in stretch activation. In the flight muscles of Lethocerus, a giant water bug often used as a model system, there are two isoforms of TnC, F1 and F2, present in an approximately 10:1 ratio. F1 TnC is responsible for activating the muscle following a stretch, whereas F2 TnC produces a sustained contraction, the magnitude of which depends on the concentration of Ca(2+) in the fiber. We have previously shown that F1 TnC binds only one Ca(2+) ion in its C-terminal domain and that interaction with troponin H, the insect ortholog of troponin I, is insensitive to Ca(2+). Here, we have studied the effect of Ca(2+) and Mg(2+) on the affinities of the interaction of F2 TnC with troponin H peptides. We show that the presence of two Ca(2+) ions, one in each of the globular domains, increases the affinity for TnH by at least 1 order of magnitude. The N lobe has a lower affinity for Ca(2+), but it is also sensitive to Mg(2+). The C lobe is insensitive to Mg(2+) as previously demonstrated by mutations of the individual EF-hands. The interaction with TnH seems also to have significant structural differences from that observed for the F1 TnC isoform. We discuss how our findings could account for stretch activation.
Identification of Two Distinct Molt-Inhibiting Hormone-Related Peptides from the Giant Tiger Prawn Penaeus Monodon

Abstract:: Six peptides belonging to the crustacean hyperglycemic hormone (CHH) family were isolated from extracts of the sinus glands of the giant tiger prawn Penaeus monodon by reverse-phase high-performance liquid chromatography. These were designated Pem-SGP-A to Pem-SGA-F (Pem, Penaeus monodon; SGP, sinus gland peptide) and their amino-terminal amino acid sequences were analyzed. Five of the 6 peptides (Pem-SGP-A, -B, -D, -E, and -F) exhibited similar amino acid sequences to those of CHH peptides that had been characterized previously from the same prawn species by the other research groups, while 1 peptide (Pem-SGP-C) exhibited a novel sequence. Pem-SGP-C showed sequence similarity to known putative molt-inhibiting hormones (MIHs), particularly to an MIH from the kuruma prawn Penaeus japonicus, and less similarity to the CHHs determined thus far. Two similar complementary DNAs encoding Pem-SGP-C were cloned and found to encode very similar but distinct peptides, which were named Pem-SGP-C1 and Pem-SGA-C2. The open reading frame of each cDNA consisted only of a signal peptide and an MIH-like peptide. We also cloned 2 corresponding genes, both of which consisted of 3 exons and 2 introns. Analyses by reverse transcriptase polymerase chain reaction demonstrated that both Pem-SGP-C1 and Pem-SGP-C2 transcripts were detected only in cDNA synthesized using total RNA from the eyestalk but not in that from brain, thoracic ganglia, abdominal ganglia, abdominal muscle, hepatopancreas, or heart tissue of P. monodon.
In vitro motility of native thin filaments from Drosophila indirect flight muscles reveals that the held-up 2 TnI mutation affects calcium activation

Abstract:A procedure for the isolation of regulated native thin filaments from the indirect flight muscles (IFM) of Drosophila melanogaster is described. These are the first striated invertebrate thin filaments to show Ca-regulated in vitro motility. Regulated native thin filaments from wild type and a troponin I mutant, held-up-2, were compared by in vitro motility assays that showed that the mutant troponin I caused activation of motility at pCa values higher than wild type. The held-up2 mutation, in the sole troponin I gene (wupA) in the Drosophila genome, is known to cause hypercontraction of the IFM and other muscles in vivo leading to their eventual destruction. The mutation causes substitution of alanine by valine at a homologous and completely conserved troponin I residue (A25) in the vertebrate skeletal muscle TnI isoform. The effects of the held-up 2 mutation on calcium activation of thin filament in vitro motility are discussed with respect to its effects on hypercontraction and dysfunction. Previous electron microscopy and 3-dimensional reconstruction studies showed that the tropomyosin of held-up 2 thin filaments occupies positions associated with the so-called 'closed' state, but independently of calcium concentration. This is discussed with respect to calcium dependent regulation of held-up-2 thin filaments in in vitro motility.
Identification and characterization of troponin genes in Locusta migratoria

Abstract:Troponin complex comprises three subunits, namely troponin C (TpnC), troponin I (TpnI) and troponin T (TpnT), and regulates the contraction of striated muscle. We found that the locust Locusta migratoria genome has one TpnT gene (LmTpnT), one TpnI gene (LmTpnI) and three TpnC genes (LmTpnC1, LmTpnC2 and LmTpnC3). Through alternative splicing, LmTpnT and LmTpnI potentially encode two and eight isoforms, respectively. The flight muscle and the jump muscle of L. migratoria express an identical LmTpnT isoform, but different LmTpnC isoforms and LmTpnI isoforms. LmTpnC2 and LmTpnC3 both contain highly conserved residues essential for calcium binding in the EF-hand II and IV, thus belonging two-site isoform. LmTpnC1 contains non-conserved substitutions in the EF-hand II and all highly conserved residues for calcium binding in the EF-hand IV. Mutagenesis and tyrosine fluorescence spectroscopic analysis show that both the EF-hand II and IV of LmTpnC1 can serve as calcium-binding site. Therefore, all three LmTpnC isoforms belong to two-site isoform. This is in contrast to the situation in the insect with asynchronous flight muscle, which expresses both one-site isoform and two-site isoform of TpnC. Those results suggest that the origination of insect asynchronous flight muscle is associated with the emergence of one-site isoform of TpnC.
Early Divergence of the C-Terminal Variable Region of Troponin T Via a Pair of Mutually Exclusive Alternatively Spliced Exons Followed by a Selective Fixation in Vertebrate Heart

Abstract:Troponin T (TnT) is the thin filament anchoring subunit of troponin complex and plays an organizer role in the Ca2+-regulation of striated muscle contraction. From an ancestral gene emerged ~ 700 million years ago in Bilateria, three homologous genes have evolved in vertebrates to encode muscle type-specific isoforms of TnT. Alternative splicing variants of TnT are present in vertebrate and invertebrate muscles to add functional diversity. While the C-terminal region of TnT is largely conserved, it contains an alternatively spliced segment emerged early in C. elegans, which has evolved into a pair of mutually exclusive exons in arthropods (10A and 10B of Drosophila TpnT gene) and vertebrates (16 and 17 of fast skeletal muscle Tnnt3 gene). The C-terminal alternatively spliced segment of TnT interfaces with the other two subunits of troponin with functional significance. The vertebrate cardiac TnT gene that emerged from duplication of the fast TnT gene has eliminated this alternative splicing by the fixation of an exon 17-like constitutive exon, indicating a functional value in slower and rhythmic contractions. The vertebrate slow skeletal muscle TnT gene that emerged from duplication of the cardiac TnT gene has the exon 17-like structure conserved, indicating its further function in sustained and fatigue resistant contractions. This functionality-based evolution is consistent with the finding that exon 10B-encoded segment of Drosophila TnT homologous to the exon 17-encoded segment of vertebrate fast TnT is selectively expressed in insect heart and leg muscles. The evolution of the C-terminal variable region of TnT demonstrates a submolecular mechanism in modifying striated muscle contractility and for the treatment of muscle and heart diseases.

Abstract:Molt-inhibiting hormone (MIH) activity was tested in terms of the inhibitory activity of ecdysteroid secretion by cultured Y-organs of the crayfish, Procambarus clarkii. Several neuropeptides were separated from a crude extract of sinus glands of P. clarkii by means of reverse-phase high-performance liquid chromatography (HPLC) and tested for MIH activity. An extremely high level of activity was found in one fraction in reverse-phase HPLC, suggesting that the novel neuropeptide in this fraction may be a P. clarkii MIH.
Analysis of Titin in Red and White Muscles: Crucial Role on Muscle Contractions Using a Fish Model

Abstract:Several studies have compared molecular components between red and white skeletal muscles in mammals. However, mammalian skeletal muscles are composed of mixed types of muscle fibers. In the current study, we analyzed and compared the distributions of titin, lipid, phosphate ions, and fatty acid levels in red and white muscles using a fish model ( Tilapia ), which is rich in red and white muscles, and these are well separated. Oil-red O staining showed that red muscle had more-abundant lipids than did white muscle. A time-of-flight secondary-ion mass spectrometric (TOF-SIMS) analysis revealed that red muscle possessed high levels of palmitic acid and oleic acid, but white muscle contained more phosphate ions. Moreover, elastica-van Gieson (EVG) and Mito-Tracker green FM staining showed that collagen and elastic fibers were highly, respectively, distributed in connective tissues and mitochondria in red muscle. An electron micrographic analysis indicated that red muscle had a relatively higher number of mitochondria and longer sarcomere lengths and Z-line widths, while myofibril diameters were thicker in white muscle. Myofibrillar proteins separated by SDS-PAGE showed that the major giant protein, titin, was highly expressed in white muscle than in red muscle. Furthermore, ratios of titin to myosin heavy chain (MHC) (titin/MHC) were about 1.3 times higher in white muscle than red muscle. We postulated that white muscle is fit for short and strong contractile performance due to high levels of titin and condensed sarcomeres, whereas red muscle is fit for low intensity and long-lasting activity due to high levels of lipids and mitochondria and long sarcomeres.

Similarities and differences between frozen-hydrated, rigor acto-S1 complexes of insect flight and chicken skeletal muscles

Abstract:The structure and function of myosin crossbridges in asynchronous insect flight muscle (IFM) have been elucidated in situ using multiple approaches. These include generating "atomic" models of myosin in multiple contractile states by rebuilding the crystal structure of chicken subfragment 1 (S1) to fit IFM crossbridges in lower-resolution electron microscopy tomograms and by "mapping" the functional effects of genetically substituted, isoform-specific domains, including the converter domain, in chimeric IFM myosin to sequences in the crystal structure of chicken S1. We prepared helical reconstructions (approximately 25 A resolution) to compare the structural characteristics of nucleotide-free myosin0 S1 bound to actin (acto-S1) isolated from chicken skeletal muscle (CSk) and the flight muscles of Lethocerus (Leth) wild-type Drosophila (wt Dros) and a Drosophila chimera (IFI-EC) wherein the converter domain of the indirect flight muscle myosin isoform has been replaced by the embryonic skeletal myosin converter domain. Superimposition of the maps of the frozen-hydrated acto-S1 complexes shows that differences between CSk and IFM S1 are limited to the azimuthal curvature of the lever arm: the regulatory light-chain (RLC) region of chicken skeletal S1 bends clockwise (as seen from the pointed end of actin) while those of IFM S1 project in a straight radial direction. All the IFM S1s are essentially identical other than some variation in the azimuthal spread of density in the RLC region. This spread is most pronounced in the IFI-EC S1, consistent with proposals that the embryonic converter domain increases the compliance of the IFM lever arm affecting the function of the myosin motor. These are the first unconstrained models of IFM S1 bound to actin and the first direct comparison of the vertebrate and invertebrate skeletal myosin II classes, the latter for which, data on the structure of discrete acto-S1 complexes, are not readily available.

Abstract:The presence and distribution of alpha-actinin has been studied in several invertebrate muscle cell types. These comprised transversely striated muscle (flight muscle) from the fruit fly Drosophila melanogaster, transversely striated muscle (heart muscle) from the snail Helix aspersa, obliquely striated muscle (body wall muscle) from the earthworm Eisenia foetida, smooth muscle (retractor muscle) from H. aspersa, and smooth muscle (outer muscular layer of the pseudoheart) from E. foetida. The study was carried by means of Western blot analysis, ELISA, and immunohistochemical electron microscopy, using anti alpha-actinin antibody. Immunoreaction for a protein with the same molecular weight as that of mammalian alpha-actinin was detected in all muscle types studied, although the amount and intensity of immunoreaction varied among them. In the insect muscle, immunolabelling was found along the whole Z-line. In both the transversely striated muscle from the snail and the obliquely striated muscle from the earthworm, immunolabelling did not occupy the whole Z-line but showed discontinuous, orderly arranged patches along the Z-line course. In the two smooth muscles studied (snail and earthworm), immunolabelling was limited to small patches which did not show an apparently ordered distribution. Since it is assumed that alpha-actinin is located at the anchorage sites for actin filaments, present observations suggest that, only in the Drosophila muscle, actin filaments are parallelly arranged in all their course, whereas in the other invertebrate muscles studied these filaments converge on discontinuously distributed anchorage sites.
