Abstract:Improved production methods of starter cultures, which constitute the most important element of probiotic preparations, were investigated. The aim of the presented research was to analyse changes in the viability of Lactobacillus. acidophilus and Bifidobacterium bifidum after stabilization (spray drying, liophilization, fluidization drying) and storage in refrigerated conditions for 4 months. The highest numbers of live cells, up to the fourth month of storage in refrigerated conditions, of the order of 10(7) cfu/g preparation were recorded for the B. bifidum DSM 20239 bacteria in which the N-Tack starch for spray drying was applied. Fluidization drying of encapsulated bacteria allowed obtaining a preparation of the comparable number of live bacterial cells up to the fourth month of storage with those encapsulated bacteria, which were subjected to freeze-drying but the former process was much shorter. The highest survivability of the encapsulated L. acidophilus DSM 20079 and B. bifidum DSM 20239 cells subjected to freeze-drying was obtained using skimmed milk as the cryoprotective substance. Stabilization of bacteria by microencapsulation can give a product easy to store and apply to produce dried food composition.
Measurement of particle diameter of Lactobacillus acidophilus microcapsule by spray drying and analysis on its microstructure

Abstract:Lactobacillus acidophilus , as a probiotic, is widely used in many functional food products. Microencapsulation not only increases the survival rate of L. acidophilus during storage and extends the shelf-life of its products, but also optimal size microcapsule makes L. acidophilus have an excellent dispersability in final products. In this paper, L. acidophilus was microencapsulated using spray drying (inlet air temperature of 170°C; outlet air temperature of 85–90°C). The wall materials used in this study were β-cyclodextrin and acacia gum in the proportion of 9:1 (w/w), and microcapsules were prepared at four levels of wall materials (15, 20, 25 and 30% [w/v]) with a core material concentration of 6% (v/v). The microcapsule diameters were measured by Malvern’s Mastersizer-2000 particle size analyzer. The results showed that the particle diameters of microcapsule were mostly within 6.607 μm and 60.256 μm and varied with 2.884–120.226 μm (the standard smaller microcapsule designated as <350 μm). Through comparison of microcapsule size and uniformity with different concentration of wall materials, we concluded that the optimal concentration of wall material was 20% (w/v), which gave microcapsule with a relatively uniform size (averaging 22.153 μm), and the number of surviving encapsulated L. acidophilus was 1.50 × 10 9 c.f.u./ml. After 8 weeks storage at 4°C, the live bacterial number was above 10 7 c.f.u./ml, compared with unencapsulated L. acidophilus , 10 4 –10 5 c.f.u./ml. Through the observation of scanning electron microscopy, we found that the shapes of microcapsule were round and oval, and L. acidophilus cells located in the centre of microcapsule.
Microencapsulation of Lactobacillus paracasei by spray freeze drying

Abstract:This study evaluates the implementation of a new process: spray freeze drying (SFD), to produce dry micro-capsules of Lactobacillus paracasei with high viability. The study concentrated on determining the survival of the cells, encapsulated in a matrix of maltodextrin and trehalose. SFD was compared with the conventional bulk freeze drying (BFD). Overall it was shown that SFD is a successful method to generate dry micro-capsules of probiotic cells with high viability (>60%). The spraying stage did not affect the viability of the bacteria. In the freezing stage, high osmotic pressures originated by elevated trehalose concentrations, helped preserving the cells viability. It was also found that the lower the maltodextrin molecular weight, the larger the beads volume and solids concentration, the higher is the bacteria survival during the freezing and drying stages. In the drying stage, trehalose concentration was also the critical factor that increased final probiotic viability.

Abstract:Probiotic health benefits are strain specific and are dose dependent. Hence, administering the correct strains, at the recommended doses is essential to achieve probiotic health benefits. Reliable methods are needed to facilitate probiotic strain identification and enumeration. Plate count methods are the most commonly used methods for probiotic enumeration. However, these methods are time-consuming, laborious, highly variable, and non-specific. Here, we developed a real-time PCR method for enumeration of a commonly used strain, Lacticaseibacillus rhamnosus GG. The method utilizes PMAxx as a viability dye to enumerate viable cells only. Optimization of viability treatment showed that PMAxx at a final concentration of 50 μM was effective in inactivating DNA from dead cells, and that bead beating for 5 min at 3000 rpm was effective in liberating DNA. The assay demonstrated high efficiency between 93 and 102%, with R2 values > 0.99. The assay showed high precision with relative standard deviation (RSD%) below 2.3%. Assay performance was compared to a plate count method in which there was a strong correlation between both methods (Pearson r = 0.8443). This method offers a 10 × shorter time for results and a higher precision compared to plate count methods. Furthermore, this method enables specific enumeration of L. rhamnosus GG in multi-strain products, which is not possible to achieve using plate count methods. This novel method facilitates faster and more accurate enumeration of L. rhamnosus GG as a raw ingredient as well as in finished products which enables better quality assurance and efficacy of probiotics for consumers.

Abstract:At the Department of Food Technology at Fulda University of Applied Sciences different methods for microencapsulation of Lactobacillus reuteri DSM 20016 were investigated. The aim of these studies was to develop a process to stabilize the probiotic bacteria for storage and to prevent them from the gastric conditions, to ensure that a satisfactory amount of the probiotics could reach their target location, the human intestine. Drying processes like spray drying and freeze drying were tested as well as fluidized bed granulation with optional Wurster coating using different auxiliary materials. As encapsulation material maltodextrine, sweet whey powder or gummi arabicum were used. The coating experiments were performed with an aqueous shellac solution. In the performed studies the fluidized bed bottom spray granulation with an additional Wurster coating turned out to be an encouraging procedure to keep the probiotics in a stable form resistant against gastric conditions. The survival rate in the simulated gastro-intestinal passage could be increased up to the sevenfold amount of the untreated bacteria.
Microencapsulated Lactobacillus rhamnosus GG powders: relationship of powder physical properties to probiotic survival during storage

Abstract:Freeze-dried commercial Lactobacillus rhamnosus GG (LGG) were encapsulated in an emulsion-based formulation stabilized by whey protein and resistant starch and either spray-dried or freeze-dried to produce probiotic microcapsules. There was no difference in loss of probiotics viability after spray drying or freeze drying. Particle size, morphology, moisture sorption, and water mobility of the powder microcapsules were examined. Particle size analysis and scanning electron microscopy showed that spray-dried LGG microcapsules (SDMC) were small spherical particles, whereas freeze-dried LGG microcapsules (FDMC) were larger nonspherical particles. Moisture sorption isotherms obtained using dynamic vapor sorption showed a slightly higher water uptake in spray-dried microcapsules. The effect of water mobility, as measured by nuclear magnetic resonance (NMR) spectroscopy, at various water activities (a(w) 0.32, 0.57, and 0.70) and probiotic viability during storage at 25 °C was also examined. Increasing the relative humidity of the environment at which the samples were stored caused an increase in water mobility and the rate of loss in viability. The viability data during storage indicated that SDMC had better storage stability compared to FDMC. Although more water was adsorbed for spray-dried than freeze-dried microcapsules, water mobility was similar for corresponding storage conditions because there was a stronger water-binding energy for spray-dried microcapsule. This possibly accounted for the improved survival of probiotics in spray-dried microcapsules.

Abstract:BACKGROUND Exercise-induced bronchoconstriction (EIB) is of particular importance in children with asthma. It is an important measure of asthma control and should be monitored by exercise testing. However, exercise testing puts a large demand on health-care resources and is therefore not widely used in routine monitoring of pediatric asthma control. The fractional concentration of exhaled nitric oxide (FeNO) also reflects uncontrolled asthma. We hypothesized that FeNO may be used for prescreening of asthmatic children to exclude those with good asthma control unlikely to have EIB, thereby reducing the need for exercise testing. OBJECTIVE The aim of this study was to estimate the value of FeNO as a predictor of EIB in asthmatic children. METHODS Stable outpatient asthmatic school children performed standard exercise challenge tests and measurement of FeNO. RESULTS FeNO and response to a standardized submaximal exercise test on the treadmill were measured in 111 school children with asthma. EIB could be excluded with a probability of 90% in asthmatic children with FeNO levels < 20 parts per billion (ppb) without current inhaled corticosteroid treatment, and < 12 ppb in children with current inhaled corticosteroid treatment. CONCLUSION Measurement of FeNO is a simple, and time- and resource-efficient tool that may be used to screen for EIB testing and therefore optimizes the resources for exercise testing in pediatric asthma monitoring.
Determination of the viability and release profile of compression coated probiotic cells using fluorescent staining and flow cytometry

Abstract:Aim Previous studies have shown that compression coating improves die storage stability of freeze-dried Lactobacillus acidophilus, although this stability is related to the degree of cell injury, which in turn is related to the compression pressure used. Compression coating has also been found to improve the survival of freeze dried L acidophilus during exposure to simulated gastric fluid (SGF). The aim of the present work is to monitor and refine the release profile of a compression coated L. acidophilus formulation, with targeted release at the terminal ileum and beginning of the colon in the human gastrointestinal tract. Methods. Dissolution studies were performed using a phosphate buffer with a pH of 2 and 6.8, to simulate gastric fluid and intestinal fluid (SIF), respectively. Cell viability was monitored using a Microcyte flow cytometer, together with traditional dilution plating (CFU/mL). Results. Results show that an enteric coating material, Eudragit L100-55, is both suitable for compression coating, and enhancing the survival of cells when exposed to SIR Pectin USP 100 has also been shown to promote targeted release of the cells, via its swelling properties upon hydration. Conclusion. This work shows the importance of encapsulation with respect to cell physiological state including viability during exposure to SGF, and highlights the potential of flow cytometry as a rapid technique for the monitoring and optimisation of probiotic cell physiology during processing conditions.
Optimization of spray-drying process parameters for microencapsulation of three probiotic lactic acid bacteria selected by their high viability rate in sucrose and fructose levels and high temperatures

Abstract:Microencapsulation is an efficient way to increase the survival rate of probiotics against harsh conditions. In this study, three probiotic strains (Lactiplantibacillus plantarum subsp. plantarum strain W2 (LP4), Lactiplantibacillus pentosus strain XL640 (LPE1), and Limosilactobacillus fermentum strain W8 (LF2)), isolated from shalgam and gilaburu, were microencapsulated with spray drying and process conditions [maltodextrin concentration (MC, 10–30%) and inlet air temperature (IAT, 110–130 °C)] were optimized by central composite rotatable design of response surface methodology. The results indicated that the predicted IAT and MC values for the maximum powder yield and viability were 123.21 °C and 22.76%, 130.37 °C and 19.49%, and 127.94 °C and 10.00% for LF2, LP4 and LPE1, respectively. At these conditions, bacterial viability ranged from 10.27 to 10.33 log colony-forming units per gram (cfu/g), while the powder yield values for the encapsulation of the bacteria were between 43.38% and 50.97%. Furthermore, MC was the most significant factor for the powder yield of LF2, LPE1, and viability of LPE1. Encapsulation efficiency values higher than 92.77% demonstrated the efficiency of spray drying for the protection of selected strains. The microcapsules produced at the optimum points had moisture content between 5.30 and 5.96%. SEM images showed that the microcapsules were in spherical shape. In conclusion, the results confirmed that the selected probiotics were successfully microencapsulated with spray drying with high powder yield, viability, and encapsulation efficiency levels and these features could reveal the potential of the encapsulated probiotic strains to be used in high-sugar foods.

Abstract:Abstract Background Probiotic Enterococcus strains of human origin were microencapsulated by spray drying using whey protein and maltodextrin as an encapsulating agent. The obtained encapsulates were characterized for stability, viability, and physiological properties. Results The microcapsules were prepared from probiotic Enterococcus strains that were previously isolated from human vagina and infants’ meconium. The microcapsules revealed similar particle sizes and morphologies. The highest hygroscopicity was observed in the microcapsules produced with strain E. rivorum S22C (0.17 ± 1.15) g water/kg powder/min. E. canintestini S18A revealed highest dissolution time in water (703 ± 2 s). The DSC thermogram revealed excellent thermal stability of all microcapsules. The physicochemical and morphological characteristics of the microcapsules were acceptable with regard to residual water content, particle mean size, and thermophysical properties and storage stability under room temperature conditions, with a low inactivation rate of Enterococcus strains. All the microcapsules revealed the recommended count of probiotic cells, low moisture content with low water activity. Observation under a scanning electron microscope revealed spherical-shaped partially collapsed structures measuring between 9 and 14 μm with surface concavities. Conclusions The microcapsule probiotic strains of Enterococcus microencapsulated by spray drying using whey protein and maltodextrin revealed properties of acceptable standards. These strains can have future potential as developing probiotic animal feed and food industry.

Abstract:This study introduces an optimized integration of flow cytometry and fluorescence in situ hybridization (Flow-FISH) as an approach for the specific enumeration of gram-positive bacteria in probiotic products, overcoming the limitations of conventional methods. The enhanced Flow-FISH technique synergizes the rapid and automated capabilities of flow cytometry with the high specificity of FISH, facilitating the differentiation of viable cells at the species level within probiotic blends. By analyzing lyophilized samples of Lacticaseibacillus rhamnosus , Lactiplantibacillus plantarum , and Bifidobacterium animalis subsp. lactis , and a commercial product, the study highlights the optimized Flow-FISH protocol's advantages, including reduced hybridization times to 1.5 h and elimination of centrifugation steps. Comparative evaluations with the widely accepted enumeration methods plate count and Live/Dead (L/D) staining were conducted. The study revealed that Flow-FISH produces higher viable cell counts than plate count, thereby challenging the traditional "gold standard" by highlighting its predisposition to underestimate actual viable cell numbers. Against L/D staining, Flow-FISH achieved comparable results, which, despite the different foundational premises of each technique, confirms the accuracy and reliability of our method. In conclusion, the optimized Flow-FISH protocol represents a significant leap forward in probiotic research and quality control. This method provides a rapid, robust, and highly specific alternative for the enumeration of probiotic bacteria, surpassing traditional methodologies. Its ability to enable a more detailed and reliable analysis of probiotic products paves the way for precise quality control and research insights, underscoring its potential to improve the field significantly.

Abstract:Lactobacillus rhamnosus GG (LGG) and Lactobacillus acidophilus NCFM (LNCFM) were encapsulated in alginate microgel particles (microbeads) by a novel dual aerosols method. The encapsulated probiotics in microbead gel matrix were further stabilized in maltodextrin solids by either spray or freeze-drying to form probiotic microcapsule powders. The free cells of probiotics were also sprayed and freeze-dried in maltodextrin only without microgel encapsulation. After rehydration of microgel-encapsulated powder, gel particles regained their shape. There was no difference in the loss of viability between encapsulated and unencapsulated probiotics during spray drying or freeze-drying. For LNCFM, spray-dried bacteria with or without gel encapsulation exhibited less death (3.03 and 3.07 log CFU/g reduction, respectively) than those of freeze-dried bacteria (4.36 and 4.89 log CFU/g reduction, respectively) after 6 months storage at 4 °C. The same trend was also observed in spray-dried LGG without gel encapsulation which showed 5.87 log CFU/g reduction in viability after 6 months at 4 °C; however, freeze-dried LGG without gel encapsulation exhibited a rapid reduction in viability of 5.91 log CFU/g within just 2 months. Gel-encapsulated LGG which was freeze-dried exhibited less death (3.32 log CFU/g reduction) after 6 months at 4 °C. This work shows that spray drying results in improved subsequent probiotic survivability compared to freeze-drying and that alginate gel encapsulation can improve the survivability following freeze-drying in a probiotic-dependent manner.

Abstract:Limosilactobacillus fermentum NCU001464 is a probiotic bacterium with anti-hyperuricemia potential, however, its viability decreases during food processing, storage and in the gastrointestinal tract. This study aims to microencapsulate Limosilactobacillus fermentum NCU001464 by spray-drying with Maillard reaction products of gelatin and xylooligosaccharides, heated for various times. Properties of powders and bacterial vitality change during producing, storage and simulated digestion were evaluated. The microcapsules had a low moisture content and the encapsulation appeared effective, however, under scanning electron microscope, cracks were visible on the surface of microcapsules with longer-time heat-treated Maillard reaction products. Heat treatment for 60 min resulted in the highest bacterial viability and adhesion capacity to intestinal epithelial cells after gastrointestinal digestion. After 10 weeks storage at 4 degrees C and 25 degrees C, the viable count of MRP-microencapsulated cells was 9.00-6.80 Log CFU/g, more than the recommended dose of 6 Log CFU/mL.
Microencapsulation of Probiotic Lactobacillus helveticus with Different Wall Materials by Spray Drying

Abstract:The present study seeks to evaluate different matrixes (chitosan, gelatin – bloom 189, gelatin – bloom 246, gum Arabic, and maltodextrin) to encapsulate Lactobacillus helveticus (LH 091), as well as optimal spray-drying conditions. L. helveticus displayed a survival rate close to 89%, with their viability above the minimum level of 107 CFU/g at point of delivery, after microencapsulation in a gum Arabic-maltodextrin (1:1) matrix followed by spray-drying (air in at 70 to 80oC, a feed rate of 0.09 mL/min, and a pressure of 0.10 bar). Microcapsules containing probiotic displayed a water activity of 0.494, rounded edges and an average size of 2.6 μm. The differential scanning calorimetry and thermogravimetry pointed to the thermal stability of the microparticles with encapsulated probiotics. These favorable properties of the probiotic microparticles make them suitable for incorporation into functional food.

Abstract:Microencapsulation of probiotic bacteria with fruit juice can be a good alternative for new probiotic foodstuff production mainly for people with galactosemia and lactose intolerance. The aim of this study was to encapsulate three probiotic bacteria, namely Lactobacillus rhamnosus, Lactobacillus casei and Lactobacillus plantarum, with a 40:60% (w/w) mixed juice of acerola and ciriguela by spray drying at air inlet temperature of 140 °C, 0.60 L/h feed flow rate and 10% (w/w) maltodextrin, as well as to evaluate the influence of maltodextrin dextrose equivalent (DE) value (5, 10 and 15) on microcapsules' properties. Powders produced were characterized in terms of cell viability counts before and after microencapsulation, physicochemical property analyses and sensory properties. Although spray drying caused a reduction of 2 decimal logarithm units in the number of viable cells, all powders had probiotic cell counts higher than that required for probiotic foods (106 CFU/g). All the three maltodextrins tested were feasible to obtain powders with satisfactory probiotic viability, physicochemical and sensory properties, thereby demonstrating that they are promising materials to microencapsulate probiotic bacteria in fruit juices.

Abstract:Microencapsulation is an up-and-coming technology for maintaining the viability of probiotics. However, the effect of core-to-wall ratios and ratios of polysaccharides on the protection of the Lactiplantibacillus plantarum 299v strain has not been deeply discussed. Lyophilization of the Lp. plantarum 299v strain was conducted, and different core-to-wall ratios and ratios of maltodextrin (MD) and resistant starch (RS) were applied. Results demonstrated that the content of MD and RS had an influence on the yield and bulk density in both core-to-wall ratios (1:1 and 1:1.5). In addition, samples coated with a core-to-wall ratio of 1:1.5 had significantly higher viability than those coated with a core-to-wall ratio of 1:1. Moreover, samples coated with core-to-wall ratios of 1:1 and MD:RS 1:1, as well as core-to-wall ratios of 1:1.5 and MD:RS 3:1, had the highest cell number after simulated gastric fluid and simulated intestinal fluid testing, respectively. Furthermore, the optimal formulation for the application of microencapsulated Lp. plantarum 299v in apple juice (serving as a functional beverage) is listed as follows: core-to-wall ratios of 1:1 and MD:RS 1:1, with the fortification method, and stored at 4 °C. After 11 weeks of storage, the cell count was 8.28 log (CFU/mL). This study provided a strategy for Lp. plantarum 299v to achieve high viability in long-term storage and provides an application in functional apple beverages.

Abstract:Probiotic Lactobacillus acidophilus LA1 was encapsulated using spray drying with concentrated skim milk with total solids (17%), and maltodextrin (25%), and gum acacia(15%) as carriers. The encapsulated probiotics were assessed for viability following drying at an inlet temperature of 170°C ±3°C and an outlet temperature of 80°C ± 3°C. The viable countenumerated after drying was 108 log cfu/g. The prepared probiotic adjunct was further assessed for viability through in vitro acidand bile salt exposure assay. The results indicated that the cellcount decreased at lower pH (1, 2) and high incubation periods(1 hr and 2 hr). A similar trend was observed with bile. However, they showed better survivability for different incubation periods at low pH and in high bile salt concentrations. The spray-dried probiotic adjunct was kept for storage at three different temperatures i.e., 8 °C, 25 °C, and 37 °C for eight weeks, and assessed weekly for cell viability. An inverse relationship between cell survival and storage temperature was observed during storage.
The viability of complex coacervate encapsulated probiotics during simulated sequential gastrointestinal digestion affected by wall materials and drying methods

Abstract:The objective of this study was to investigate the impact of protein type (sodium caseinate and pea protein isolate) and protein to sugar beet pectin mixing ratio (5 : 1 and 2 : 1) on complex coacervate formation, as well as the impact of the finishing technology (freeze-drying and spray-drying) for improving the viability of encapsulated Lactobacillus rhamnosus GG (LGG) in complex coacervates during simulated sequential gastrointestinal (GI) digestion. The physicochemical properties of LGG encapsulated microcapsules in liquid and powder form were evaluated. The state diagram and ζ-potential results indicated that pH 3.0 was the optimum pH for coacervate formation in the current systems. Confocal laser scanning microscopy (CLSM), viscoelastic analysis, and Fourier transform infrared spectroscopy (FTIR) confirmed that the gel-like network structure of the complex coacervates was successfully formed between the protein and SBP at pH 3.0 through electrostatic interaction. In terms of physiochemical properties and viability of LGG encapsulated in the microcapsule powder, the drying method played a crucial role on particle size, microstructure and death rate of encapsulated LGG during simulated sequential GI digestion compared to protein type and biopolymer mixing ratio. For example, the microstructure of spray-dried microcapsules exhibited smaller spherical particles with some cavities, whereas the larger particle size of freeze-dried samples showed a porous sponge network structure with larger particle sizes. As a result, spray-dried LGG microcapsules generally had a lower death rate during simulated sequential gastrointestinal digestion compared to their freeze-dried counterparts. Among all samples, spray-dried PPI-SBP microcapsules demonstrated superior performance against cell loss and maintained more than 7.5 log CFU per g viable cells after digestion.
