Abstract:Two methacrylate monomers, oligo(ethylene glycol) methyl ether methacrylate (OEGMA; MW=300gmol−1, poly(ethylene glycol) (PEG) side chains of average length n=4.5) and 2-methacryloyloxyethyl phosphorylcholine (MPC; MW=295gmol−1), were grafted from silicon wafer surfaces via surface-initiated atom transfer radical polymerization. The grafted surfaces were used as model PEG and phosphorylcholine surface systems to allow comparison of the effectiveness of these two motifs in the prevention of plasma protein adsorption and platelet adhesion. It was found that at high graft density fibrinogen adsorption from plasma on the poly(MPC) and poly(OEGMA) surfaces for a given graft chain length was comparable and extremely low. At low graft density, poly(OEGMA) was slightly more effective than poly(MPC) in resisting fibrinogen adsorption from plasma. Flowing whole blood experiments showed that at low graft density the poly(OEGMA) surfaces were more resistant to fibrinogen adsorption and platelet adhesion than the poly(MPC) surfaces. At high graft density, both the poly(MPC) and poly(OEGMA) surfaces were highly resistant to fibrinogen and platelets. Immunoblots of proteins eluted from the surfaces after contact with human plasma were probed with antibodies against a range of proteins, including the contact phase clotting factors, fibrinogen, albumin, complement C3, IgG, vitronectin and apolipoprotein A-I. The blot responses were weak on the poly(MPC) and poly(OEGMA) surfaces at low graft density and zero at high graft density, again indicating strongly protein resistant properties for these surfaces. Since the side chains of the poly(OEGMA) are about 50% greater in size than those of poly(MPC), the difference in protein resistance between the poly(MPC) and poly(OEGMA) surfaces at low graft density may be due to the difference in surface coverage of the two graft types.

Abstract:Janus polymerization is a novel and efficient way to synthesize diblock and multiblock copolymers in one step by using Lu(OTf)(3) and propylene epoxide as a catalytic system. By modifying the epoxide initiator, which is located at the block junction with various functional groups, the possibility for future topological design is provided. Herein, 2-bicyclo[2.2.1]hept-5-en-2-yl oxirane (NB-EO) is used as an alternative for propylene epoxide to synthesize a poly(THF-co-CL)-b-PCL diblock copolymer featuring a norbornene group at this position. This also results in multiblock [poly(THF-co-CL)-b-PCL](m) copolymers carrying multiple norbornene moieties through Janus polymerization. Subsequent ring-opening metathesis copolymerization (ROMP) of the resulting poly(THF-co-CL)-b-PCL macromonomers with norbornenyl-terminated polysarcosine (PSar-NB) allows the facile preparation of heterograft molecular polymer brushes (MPBs). The MPBs feature three heterografts of PCL, P(CL-co-THF) and PSar, potentially equipping these structures with biocompatibility, biodegradability, semi-crystallinity, and amphiphilicity. A phase separation is observed after annealing in both TEM and AFM analysis. Due to their amphiphilic nature, the MPBs also undergo self-assembly into micelles in aqueous solution. Such materials combining polypeptoids, polyesters, and polyethers segments are expected to attract wide attention in drug delivery applications.
Short Poly(ethylene Glycol) Block Initiation of Poly(l‐lactide) Di‐block Copolymers: a Strategy for Tuning the Degradation of Resorbable Devices

Abstract:The current range of medical applications of resorbable polyesters could be hugely expanded if more effective strategies for tailoring degradation rate were available. Block copolymerisation with poly(ethylene glycol) (PEG) has been shown to reduce degradation times; however, to date, this has relied on the addition of PEG to short lengths of polyester. This results in copolymers with high fractions of PEG and low molecular weights, reducing the potential range of applications. Furthermore, there has been no systematic study of the relative lengths of the blocks. In this work, we employed short hydroxyl-functionalised methoxy-terminated mPEG to initiate the synthesis of poly(L-lactide) (PLLA), resulting in controlled di-block copolymers with short mPEG blocks and long PLLA blocks. A controlled series of polymers was made with PLLA lengths (60 < M-n (kg mol(-1)) < 200) and mPEG lengths (550 < M-n (g mol(-1)) < 5000) giving very low mPEG content (0.1-1.5 wt%). We found that, despite the low fraction of mPEG, water uptake and the rate of hydrolytic degradation, k, increased. Significantly, k for the polymers was dependent only on the presence of mPEG, and was little affected by mPEG length or PLLA length in the ranges studied. Moreover, mass loss began in all polymers when Mn of the polymer fell below a threshold of about 20 kg mol(-1) and depended on both the initial molecular weight of PLLA and the presence (but not the length) of mPEG. Short-chain mPEG therefore provides a new route for targeted, temporal control of resorbable polyesters for biomedical devices. (C) 2018 The Authors. Abstract:Biodegradable macroporous hydrogels were prepared using a biodegradable crosslinker based on chitosan derivatives. The chitosan crosslinker was synthesized through the amidating reaction of amino groups in the chitosan oligosaccharide (CSO) chains and carboxyl groups in acrylic acid (AA) under the catalysis of 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimidehydro-chloride (EDC) and N-hydroxysuccinimide (NHS). The amidating reaction is able to provide polymerizable carbon-carbon double bonds for CSO. The chemical structure of the resulting compound chitosan oligosaccharide-graft-acrylic acid (CSO-g-AA) was characterized by Fourier transform infrared spectroscopy (FTIR) and H-1 nuclear magnetic resonance (H-1 NMR) spectroscopy. The content of double bonds was measured by titration analysis. The measurement showed a grafting ratio of AA on CSO of about 3.5% (on average two acryoyl groups per CSO-g-AA chain) when the applied molar ratio of AA to CSO was 1.0. Polyacrylamide (PAM) cryogels were synthesized by the free radical polymerization of acrylamide using a modified freezing polymerization method crosslinked by CSO-g-AA. The obtained cryogels were macroporous hydrogels which could be degraded into solutions of linear polymer chains when treated with appropriate enzymes. Then we used snailase as a model enzyme to study the biodegradation process. The degradation was monitored by morphological studies using a confocal laser scanning microscope (CLSM) and scanning electron microscope (SEM), mechanical strength, and swelling ratio. The duration of the degradation process was adjustable from one month to two months when using different concentrations of snailase solutions.

Abstract:In tissue engineering (TE), the support structure (scaffold) plays a key role necessary for cell adhesion and proliferation. The protein constituents of the extracellular matrix (ECM), such as collagen, its derivative gelatine, and elastin, are the most attractive materials as possible scaffolds. To improve the modest mechanical properties of gelatine, a strategy consists of crosslinking it, as naturally occurs for collagen, which is stiffened by the oxidative action of lysyl oxidase (LO). Here, a novel protocol to crosslink gelatine has been developed, not using the commonly employed crosslinkers, but based on the formation of imine bonds or on aldolic condensation reactions occurring between gelatine and properly synthesized copolymers containing amine residues via LO-mediated oxidation. Particularly, we first synthesized and characterized an amino butyl styrene monomer (5), its copolymers with dimethylacrylamide (DMAA), and its terpolymer with DMAA and acrylic acid (AA). Three acryloyl amidoamine monomers (11a–c) and their copolymers with DMAA were then prepared. A methacrolein (MA)/DMAA copolymer already possessing the needed aldehyde groups was finally developed to investigate the relevance of LO in the crosslinking process. Oxidation tests of amine copolymers with LO were performed to identify the best substrates to be used in experiments of gelatine reticulation. Copolymers obtained with 5, 11b, and 11c were excellent substrates for LO and were employed with MA/DMAA copolymers in gelatine crosslinking tests in different conditions. Among the amine-containing copolymers, that obtained with 5 (CP5/DMMA-43.1) afforded a material (M21) with the highest crosslinking percentage (71%). Cytotoxicity experiments carried out on two cell lines (IMR-32 and SH SY5Y) with the analogous (P5) of the synthetic constituent of M21 (CP5/DMAA) had evidenced no significant reduction in cell viability, but proliferation promotion, thus establishing the biocompatibility of M21 and the possibility to develop it as a new scaffold for TE, upon further investigations.

Abstract:Abstract In tissue engineering, cell origin is important to ensure outcome quality. However, the impact of the cell type chosen for seeding in a biocompatible matrix has been less investigated. Here, we investigated the capacity of primary and immortalized fibroblasts of distinct origins to degrade a gelatin/alginate/fibrin (GAF)-based biomaterial. We further established that fibrin was targeted by degradative fibroblasts through the secretion of fibrinolytic matrix-metalloproteinases (MMPs) and urokinase, two types of serine protease. Finally, we demonstrated that besides aprotinin, specific targeting of fibrinolytic MMPs and urokinase led to cell-laden GAF stability for at least several days. These results support the use of specific strategies to tune fibrin-based biomaterials degradation over time. It emphasizes the need to choose the right cell type and further bring targeted solutions to avoid the degradation of fibrin-containing hydrogels or bioinks.

Abstract:Fibrin gels, prepared from fibrinogen and thrombin, the key proteins involved in blood clotting, were among the first biomaterials used to prevent bleeding and promote wound healing. The unique polymerization mechanism of fibrin, which allows control of gelation times and network architecture by variation in reaction conditions, allows formation of a wide array of soft substrates under physiological conditions. Fibrin gels have been extensively studied rheologically in part because their nonlinear elasticity, characterized by soft compliance at small strains and impressive stiffening to resist larger deformations, appears essential for their function as haemostatic plugs and as matrices for cell migration and wound healing. The filaments forming a fibrin network are among the softest in nature, allowing them to deform to large extents and stiffen but not break. The biochemical and mechanical properties of fibrin have recently been exploited in numerous studies that suggest its potential for applications in medicine and bioengineering.

Abstract:A library of composite polymer networks (CPNs) were formed by combining Pluronic F127, as the primary gelator, with a range of di-acrylate functionalised PEG polymers, which tune the rheological properties and provide UV crosslinkability. A coarse-grained sol–gel room temperature phase diagram was constructed for the CPN library, which identifies PEG-dependent disruption of micelles as leading to liquefication. Small angle X-ray scattering and rheological measurements provide detailed insight into; (i) micelle-micelle ordering; (ii) micelle-micelle disruption, and; (iii) acrylate-micelle disruption; with contributions that depend on composition, including weak PEG chain length and end group effects. The influence of composition on 3D extrusion printability through modulation of the cohesive/hydrophobic interactions was assessed. It was found that only micelle content provides consistent changes in printing fidelity, controlled largely by printing conditions (pressure and feed rate). Finally, the hydrogels were shown to be UV photo-crosslinkable, which further improves fidelity and structural integrity, and usefully reduces the mesh size. Our results provide a guide for design of 3D-printable CPN inks for future biomedical applications.

Abstract:Covalent cross-linking is a common strategy to improve the mechanical properties of biological polymers. The most prominent field of application of such materials is in medicine, for example, in the form of bioprinting, drug delivery, and wound sealants. One biological polymer of particular interest is the blood clotting protein fibrinogen. In the natural process, fibrinogen polymerizes to fibrous hydrogel fibrin. Although the material shows great potential, its costs are very high due to the required enzyme thrombin. Recently, we introduced several approaches to trigger a thrombin-free fibrillogenesis of fibrinogen to a fibrin-like material. Inspired by the natural pathway of blood clotting in which covalent cross-linking stabilizes the clot, this "pseudofibrin" is now developed even further by covalently cross-linking the fibers. In particular, the effect of inexpensive glutaraldehyde on fiber morphology, rheological properties, and irreversible gel dissolution is investigated. Additionally, new insights into the reaction kinetics between fibrinogen and glutaraldehyde are gained. It could be shown that the fibrous structure of pseudofibrin can be retained during cross-linking and that glutaraldehyde significantly improves rheological properties of the hydrogels. Even more important, cross-linking with glutaraldehyde can prevent dissolution of the gels at elevated temperatures.

Abstract:Regenerative medicine is an interdisciplinary field aiming at restoring pathologically damaged tissues and whole organs by cell transplantation in combination with proper supporting scaffolds. Gelatine-based ones are very attractive due to their biocompatibility, rapid biodegradability, and lack of immunogenicity. Gelatine-based composite hydrogels, containing strengthening agents to improve their modest mechanical properties, have been demonstrated to act as extracellular matrices (ECMs), thus playing a critical role in "organ manufacturing". Inspired by the lysyl oxidase (LO)-mediated process of crosslinking, which occurs in nature to reinforce collagen, we have recently developed a versatile protocol to crosslink gelatine B (Gel B) in the presence or absence of LO, using properly synthesized polystyrene- and polyacrylic-based copolymers containing the amine or aldehyde groups needed for crosslinking reactions. Here, following the developed protocol with slight modifications, we have successfully crosslinked Gel B in different conditions, obtaining eight out of nine compounds in high yield (57–99%). The determined crosslinking degree percentage (CP%) evidenced a high CP% for compounds obtained in presence of LO and using the styrenic amine-containing (CP5/DMAA) and acrylic aldehyde-containing (CPMA/DMAA) copolymers as crosslinking agents. ATR-FTIR analyses confirmed the chemical structure of all compounds, while optical microscopy demonstrated cavernous, crater-like, and labyrinth-like morphologies and cavities with a size in the range 15–261 μm. An apparent density in the range 0.10–0.45 g/cm3 confirmed the aerogel-like structure of most samples. Although the best biodegradation profile was observed for the sample obtained using 10% CP5/DMAA (M3), high swelling and absorption properties, high porosity, and good biodegradation profiles were also observed for samples obtained using the 5–10% CP5/DMAA (M4, 5, 6) and 20% CPMA/DMAA (M9) copolymers. Collectively, in this work of synthesis and physicochemical characterization, new aerogel-like composites have been developed and, based on their characteristics, which fit well within the requirements for TE, five candidates (M3, M4, M5, M6, and M9) suitable for future biological experiments on cell adhesion, infiltration and proliferation, to confirm their effective functioning, have been identified.

Abstract:Reversible addition–fragmentation chain transfer polymerization with a low-temperature initiator can be used to synthesize multifunctional block copolymers for polyplex formation. Utilizing three methacrylate monomers (triethylene glycol methyl ether methacrylate (MEO3MA), pyridyldisulfideethyl methacrylate (PDSM), N,N-dimethylethylenediamine methacrylate (DMAEMA)) and an azide bearing chain transfer agent for polymerization, the resulting triblock-copolymers possess two orthogonally reactive functionalities (a block with reactive disulfide side chains and an azide end-group) and two additional functional blocks for i) solubilization and ii) polyplex formation. Thereby, the MEO3MA block provides solubility in aqueous media and the DMAEMA block, having a tertiary amine in the side chain, allows polyplex formation with polyanionic biomacromolecules like DNA or RNA. Due to the reactive disulfide block the polyplexes can be (reversibly) crosslinked with dithiols. Because of the manifold of possibilities to modify the pDNApolyplexes, this polymers system is an interesting candidate for active-targeting and codelivery approaches.
DTIE, a novel core promoter element that directs start site selection in TATA-less genes

Abstract:The transcription start site (TSS) determines the length and composition of the 5′ UTR and therefore can have a profound effect on translation. Yet, little is known about the mechanism underlying start site selection, particularly from promoters lacking conventional core elements such as TATA-box and Initiator. Here we report a novel mechanism of start site selection in the TATA- and Initiator-less promoter of miR-22, through a strictly localized downstream element termed DTIE and an upstream distal element. Changing the distance between them reduced promoter strength, altered TSS selection and diminished Pol II recruitment. Biochemical assays suggest that DTIE does not serve as a docking site for TFIID, the major core promoter-binding factor. TFIID is recruited to the promoter through DTIE but is dispensable for TSS selection. We determined DTIE consensus and found it to be remarkably prevalent, present at the same TSS downstream location in ≈20.8% of human promoters, the vast majority of which are TATA-less. Analysis of DTIE in the tumor suppressor p53 confirmed a similar function. Our findings reveal a novel mechanism of transcription initiation from TATA-less promoters.

Abstract:Epoxy‐functionalized microgels are used as reactive fillers to improve mechanical properties of fibrin hydrogels, slow down their degradation, and ensure efficient loading and temperature‐triggered release of drug. Tissue engineering is a steadily growing field of research due to its wide‐ranging applicability in the field of regenerative medicine. Application‐dependent mechanical properties of a scaffold material as well as its biocompatibility and tailored functionality represent particular challenges. Here the properties of fibrin‐based hydrogels reinforced by functional cytocompatible poly(N‐vinylcaprolactam)‐based (PVCL) microgels are studied and evaluated. The employment of temperature‐responsive microgels decorated by epoxy groups for covalent binding to the fibrin is studied as a function of cross‐linking degree within the microgels, microgel concentration, as well as temperature. Rheology reveals a strong correlation between the mechanical properties of the reinforced fibrin‐based hydrogels and the microgel rigidity and concentration. The incorporated microgels serve as cross‐links, which enable temperature‐responsive behavior of the hydrogels, and slow down the hydrogel degradation. Microgels can be additionally used as carriers for active drugs, as demonstrated for dexamethasone. The microgels' temperature‐responsiveness allows for triggered release of payload, which is monitored using a bioassay. The cytocompatibility of the microgel‐reinforced fibrin‐based hydrogels is demonstrated by LIVE/DEAD staining experiments using human mesenchymal stem cells. Abstract:Murine monoclonal antibodies (MAbs) directed against serotypes 1, 2, 8, and 9 of Serpulina hyodysenteriae and strain B256 of Serpulina innocens were produced and characterized. A serological classification of 96 field strains of S. hyodysenteriae and 28 field strains of S. innocens isolated from pigs showing clinical signs of swine dysentery was performed by rapid dot enzyme-linked immunosorbent assay (ELISA) with the MAbs. The results indicated that the majority of the field strains of S. hyodysenteriae (69%) belonged to serotypes 8, 1, and 9, whereas only 31% of the S. innocens strains were recognized by MAb 9H7, indicating the presence of antigenic heterogeneity among S. innocens isolates. Rapid dot ELISA with type-specific MAbs was found to be specific, sensitive, and easy to perform and thus to be suitable for routine serotyping of S. hyodysenteriae and S. innocens isolates. This is the first report of MAbs being used for serotyping clinical isolates of S. hyodysenteriae and S. innocens.
Well-defined synthetic copolymers with pendent aldehydes form biocompatible strain-stiffening hydrogels and enable competitive ligand displacement

Abstract:Dynamic hydrogels are attractive platforms for tissue engineering and regenerative medicine, but there are limited polymer platforms amenable to the formation of these dynamic materials. Most architectures reported are based either on telechelic synthetic polymers or side-chain modified natural polymers; synthetic versions of side-chain modified polymers are noticeably absent. Dynamic hydrogels are key tools for their ability to mimic key extracellular (ECM) mechanical properties like strain stiffening and stress relaxation, while enabling enhanced processing characteristics like injectability, 3D printing, and self-healing. Currently, the limited chemical design space is hindering progress. To facilitate access to new classes of dynamic hydrogels, we report the straightforward synthesis of a water-soluble copolymer with a tunable fraction of pendent aldehyde groups (12–64%) using controlled radical polymerization, and their formation into hydrogel biomaterials with dynamic crosslinks. We found the polymer synthesis to be well-controlled with the determined reactivity ratios consistent with blocky gradient microarchitecture. Subsequently, we observed fast gelation kinetics with imine-type crosslinking. We were able to vary hydrogel stiffness from ≈ 2–20 kPa and tune the onset of strain-stiffening towards a biologically relevant regime (critical stress ≈ 10 Pa). Moreover, we used fluorescence resonance energy transfer (FRET) to demonstrate ligand displacement along the copolymer backbone via competitive binding and highlighted the cytocompatibility of our system using human dermal fibroblasts. The combination of highly tunable composition, stiffness, and strain-stiffening, in conjunction with temporal control over ligand exchange/presentation, positions these cytocompatible copolymers as a powerful synthetic platform for the rational design of next generation synthetic biomaterials.

Abstract:Controlling and guiding cell behavior requires scaffolding materials capable of programming the three-dimensional (3-D) extracellular environment. In this study, we devised a new self-assembling peptide template for synthesizing nanofibrous hydrogels containing cell-responsive ligands. In particular, the insertion of a matrix metalloproteinase-2 (MMP-2) labile hexapeptide into the self-assembling building blocks of arginine-alanine-aspartate-alanine (RADA) was investigated. A series of peptides, varied by the position of the MMP-2 hexapeptide substrate and the length of RADA blocks, were prepared by parallel synthesis. Their self-assembling capabilities were characterized and compared by circular dichroism spectroscopy and dynamical mechanical analysis. Among all the different insertion patterns, the sequence comprising a centrically positioned MMP-2 substrate was flanked with three RADA units on each side self-assembled into a hydrogel matrix, with mechanical properties and nanofiber morphology comparable to the native material built with (RADA)4 alone. Exposure of the new gel to MMP-2 resulted in peptide cleavage, as confirmed by mass spectroscopy, and a decrease in surface hardness, as detected by nanoindentor, indicating that the enzyme mediated degradation was localized to the gel surface. The new design can be used for introducing biological functions into self-assembling peptides to create scaffolding materials with potential applications in areas such as tissue engineering and regenerative medicine.
