Abstract:Human decomposition is a complex biological process driven by an array of variables which are not clearly understood. The medico-legal community have long been searching for a reliable method to establish the post-mortem interval (PMI) for those whose deaths have either been hidden, or gone un-noticed. To date, attempts to develop a PMI estimation method based on the state of the body either at the scene or at autopsy have been unsuccessful. One recent study has proposed that two simple formulae, based on the level of decomposition humidity and temperature, could be used to accurately calculate the PMI for bodies outside, on or under the surface worldwide. This study attempted to validate 'Formula I' [1] (for bodies on the surface) using 42 Canadian cases with known PMIs. The results indicated that bodies exposed to warm temperatures consistently overestimated the known PMI by a large and inconsistent margin for Formula I estimations. And for bodies exposed to cold and freezing temperatures (less than 4°C), then the PMI was dramatically under estimated. The ability of 'Formulae II' to estimate the PMI for buried bodies was also examined using a set of 22 known Canadian burial cases. As these cases used in this study are retrospective, some of the data needed for Formula II was not available. The 4.6 value used in Formula II to represent the standard ratio of time that burial decelerates the rate of decomposition was examined. The average time taken to achieve each stage of decomposition both on, and under the surface was compared for the 118 known cases. It was found that the rate of decomposition was not consistent throughout all stages of decomposition. The rates of autolysis above and below the ground were equivalent with the buried cases staying in a state of putrefaction for a prolonged period of time. It is suggested that differences in temperature extremes and humidity levels between geographic regions may make it impractical to apply formulas developed in one region to any other region. These results also suggest that there are other variables, apart from temperature and humidity that may impact the rate of human decomposition. These variables, or complex of variables, are considered regionally specific. Neither of the Universal Formulae performed well, and our results do not support the proposition of Universality for PMI estimation.

Abstract:The rate of detection of chromosome abnormalities in T-cell proliferations is lower than that observed in B-cell malignancies. The former frequently involve the TCR alpha/delta locus at chromosome band 14q11. We have identified a YAC encompassing 70% of the TCR alpha/delta locus, which has been used as a fluorescence in situ hybridization probe to detect chromosome rearrangements involving 14q11, both at metaphase and within interphase nuclei, in patients with a variety of T-lymphoproliferative disorders. Its use allowed detection of previously unsuspected TCR alpha/delta rearrangements in 4/13 (30%) immature T-lineage acute leukemias, including two t(10;14) and 2 minor inversion 14s. It also clarified interpretation of complex chromosome 14 abnormalities in mature T-cell proliferations (T-prolymphocytic leukemia and ataxia telangiectasia). Use of this probe will aid the detection and characterization of abnormalities involving the TCR alpha/delta locus, particularly in cases with normal or complex karyotypes and in those proliferations for which mitoses are difficult to obtain.

The validation of 'universal' PMI methods for the estimation of time since death in temperate Australian climates

Abstract:Forensic anthropologists have traditionally relied on their knowledge and experience of the decomposition stages to make an assessment of the time since death. However, recently new and empirical methods have been developed in various regions worldwide that propose to estimate the post-mortem interval (PMI) based on the observed decomposition changes alongside important taphonomic variables. Yet despite these methods being predominantly geographic specific, a number of methods have suggested they are effective universally and it is these 'universal' methods that have been inadequately test in Australia. The current study evaluated the accuracy of two of these methods in an Australian context, specifically the Greater Western Sydney region. The protocol developed by Megyesi et al. (2005) [1] was investigated because it is commonly cited in the literature and the PMI formula created by Vass (2011) [2] was also investigated because of its 'universal' claim. Between December 2014 and March 2016, two experimental trials were undertaken during the Australian summer seasons. Sixteen adult pig carcasses were left to decompose undisturbed on a soil surface common throughout the Western Sydney region and the Megyesi et al. (2005) [1] and Vass (2011) [2] methods were applied to the remains during this period. The results showed the Megyesi et al. (2005) [1] method overestimated the known PMI of remains, whereas the Vass (2011) [2] formula underestimated the time since death of the remains in these trials. The inaccuracy may be attributed to the constants which make up the variables in these formulas and they may not reflect the values of these variables in the Western Sydney region.

Abstract:The decomposition process of human remains can be used to estimate the post-mortem interval (PMI), but decomposition varies due to many factors. Temperature is believed to be the most important and can be connected to decomposition by using the accumulated degree days (ADD). The aim of this research was to develop a decomposition scoring method and to develop a formula to estimate the PMI by using the developed decomposition scoring method and ADD.A decomposition scoring method and a Book of Reference (visual resource) were made. Ninety-one cases were used to develop a method to estimate the PMI. The photographs were scored using the decomposition scoring method. The temperature data was provided by the Royal Netherlands Meteorological Institute. The PMI was estimated using the total decomposition score (TDS) and using the TDS and ADD. The latter required an additional step, namely to calculate the ADD from the finding date back until the predicted day of death.The developed decomposition scoring method had a high interrater reliability. The TDS significantly estimates the PMI (R 2 = 0.67 and 0.80 for indoor and outdoor bodies, respectively). When using the ADD, the R 2 decreased to 0.66 and 0.56.The developed decomposition scoring method is a practical method to measure decomposition for human remains found on land. The PMI can be estimated using this method, but caution is advised in cases with a long PMI. The ADD does not account for all the heat present in a decomposing remain and is therefore a possible bias.

Abstract:Environmental and individualistic variables affect the rate of human decomposition in complex ways. These effects complicate the estimation of the postmortem interval (PMI) based on observed decomposition characteristics. In this work, we develop a generative probabilistic model for decomposing human remains based on PMI and a wide range of environmental and individualistic variables. This model explicitly represents the effect of each variable, including PMI, on the appearance of each decomposition characteristic, allowing for direct interpretation of model effects and enabling the use of the model for PMI inference and optimal experimental design. In addition, the probabilistic nature of the model allows for the integration of expert knowledge in the form of prior distributions. We fit this model to a diverse set of 2,529 cases from the GeoFOR dataset. We demonstrate that the model accurately predicts 24 decomposition characteristics with an ROC AUC score of 0.85. Using Bayesian inference techniques, we invert the decomposition model to predict PMI as a function of the observed decomposition characteristics and environmental and individualistic variables, producing an R-squared measure of 71%. Finally, we demonstrate how to use the fitted model to design future experiments that maximize the expected amount of new information about the mechanisms of decomposition using the Expected Information Gain formalism.

Estimating postmortem interval for human cadavers in a sub-tropical climate using UV-Vis-near-infrared Spectroscopy

Abstract:Estimating postmortem interval (PMI) of surface found skeletal remains is challenging. This novel study used UV-Vis-NIR spectroscopy to scan soil collected from cadaver decomposition islands (CDIs) ranging from 15- to 963-d postmortem and control soils. A decomposition product spectra model (DPS model) was constructed by deducting the control soil spectra from the CDI soil spectra for the estimation of postmortem indices: PMI (d), ADD4 , ADD10 , and ADD20 . The DPS model (n = 55) was calibrated and subjected to a full cross-validation. Calibration R2 and RPD for the DPS model ranged from 0.97 to 0.99 and from 6.1 to 9.9, respectively, for the four postmortem interval indices. Validation R2 and RPD for the DPS model ranged from 0.73 to 0.80 and from 1.9 to 2.2, respectively. The DPS model estimated postmortem intervals for three test CDIs in a clay soil under perennial grassland (test set 1; n = 3) and six CDIs in a sandy soil under a loblolly pine forest (test set 2; n = 6). Test set 1 had PMI prediction ranges from -69 to -117 days, -796 to +832 ADD4 , +552 to +2672 ADD10 , and -478 to -20 ADD20 of observed PMI. Test set 2 PMI prediction ranged from -198 to -65 days, -9923 to +2629 ADD4 , -6724 to +1321 ADD10 , and -2850 to +540 ADD20 of observed PMI. Test set 2 had poor predictions for two CDIs, for all measures of postmortem indices resulting in discussion of sampling depth, effect of body mass index (BMI), and scavenging.

Abstract:Accurate estimation of the postmortem interval (PMI) is crucial in forensic medico-legal investigations to understand case circumstances (e.g. narrowing down list of missing persons or include/exclude suspects). Due to the complex decomposition chemistry, estimation of PMI remains challenging and currently often relies on the subjective visual assessment of gross morphological/taphonomic changes of a body during decomposition or entomological data. The aim of the current study was to investigate the human decomposition process up to 3 months after death and propose novel time-dependent biomarkers (peptide ratios) for the estimation of decomposition time. An untargeted liquid chromatography tandem mass spectrometry–based bottom-up proteomics workflow (ion mobility separated) was utilized to analyse skeletal muscle, collected repeatedly from nine body donors decomposing in an open eucalypt woodland environment in Australia. Additionally, general analytical considerations for large-scale proteomics studies for PMI determination are raised and discussed. Multiple peptide ratios (human origin) were successfully proposed (subgroups < 200 accumulated degree days (ADD), < 655 ADD and < 1535 ADD) as a first step towards generalised, objective biochemical estimation of decomposition time. Furthermore, peptide ratios for donor-specific intrinsic factors (sex and body mass) were found. Search of peptide data against a bacterial database did not yield any results most likely due to the low abundance of bacterial proteins within the collected human biopsy samples. For comprehensive time-dependent modelling, increased donor number would be necessary along with targeted confirmation of proposed peptides. Overall, the presented results provide valuable information that aid in the understanding and estimation of the human decomposition processes.

Abstract:Establishing time elapsed for unattended death scenes is crucial in formulating a timeline of events facilitating death investigations. However, traditional postmortem interval (PMI) methods rely on visually evaluating physical atrophy and are closely influenced by both biotic and abiotic variables associated with carrion. During the bloat stage, carrion produces a characteristic landscape known as the cadaver decomposition island (CDI), through the propagation of fluids rich in decomposition by-products. Here, attenuated total reflectance- Fourier transform infrared spectroscopy (ATR-FT-IR) was employed as a non-invasive, low input, low preparation interface for determining PMI from simulated Mus musculus burial gravesoil. Furthermore, understanding the influences of environmental and inter-individual differences in gaining accurate PMI is important to validate, prior to implementation to enhance the current forensic toolkit. It is documented that the presence, type and weight of clothing interferes with progression through decomposition. The subsequent impacts of clothing material (cotton, polyester, viscose) on CDI footprint development will be reflected in the biological and chemical characteristics of this ecosystem. Principal component analysis (PCA) of the IR spectra showed two clusters of samples corresponding to control and gravesoil. PC loadings plot showed that the 3100 - 1000 cm-1 spectral range attributed for over 95% of the variance. Bands within this range are ascribed to the presence of lipids, proteins and volatile organic compounds (VOCs) as byproducts of mammalian decomposition. Overall, presence and fabric type impacted decomposition, spectral CDI detection and grave discrimination. This study questions the efficacy of proxy size and microcosm design in conducting applicable forensic research in lieu of taphonomy facilities or ethical constraints.

Abstract:Accurate determination of the post-mortem interval (PMI) is critical in forensic casework. Most studies conclude that PMI determinations are dependent on local climatic and geographical factors. Despite this, there is little data in an Australian environment outside of entomological studies. In the absence of insect data on or around the remains, alternative methods are required. Anthropological methods of observing and scoring the extent of decomposition at the time of discovery provide only broad estimates of PMI. Microbial succession is responsible for these observable soft tissue changes, particularly in early and late decomposition. The aim of this study was to combine anthropological and microbiological methods to provide data for determining PMI in a temperate Australian climate. Microbial DNA was isolated from skin and cavity swabs and used to perform 16S rRNA metagenomic analysis of pooled DNA samples to allow the choice of target taxa for qPCR. qPCR indicated significant changes in microbial communities with a dominant population of Gammaproteobacteria at early time points giving way to Firmicutes and Bacteroidetes near the end of the experiment. Expressing qPCR data as a ratio of Lactobacillales/Enterobacteriaceae provided data that could be useful in determining early and late decomposition. The genus Psychrobacter was identified as a good indicator of late decomposition in winter conditions. qPCR analysis, with further refinements, could be part of an accurate quantitative method of determining PMI.

Abstract:A new approach is taken to estimating the time of death based on cadaver temperatures. The predictions are obtained by using numerical simulation that can be performed in a case-by-case scenario. Such a method enables time-of-death predictions for persons of any size and weight and in any thermal environment. An added advantage of the method is that it is not dependent upon an identification of the individual tissue layers and regions. Rather, a homogeneous tissue model is used, and the results that are obtained agree closely with the results of prior estimation methods and also with a prior published case study. Among the results presented in this study are various convective environments in both air and water (or a combination for a partially submerged body). The orientation of the body (face down vs face up) is investigated. It is found that when a body is face up, its body core temperature is more responsive to the ambient conditions, compared with a face-down orientation, at least for some partial-submergence depths. The method disclosed here can also be used to handle situations, where the environmental conditions are changing (such as diurnal temperature variations, variation in cloudy or sunny skies, etc.). Current nomogram methods are not able to handle such timewise variations.

Abstract:The near ubiquitous presence of numerical simulation has made case‐specific calculations of body temperatures following death possible so that accurate calculations of body temperatures can provide valuable information for estimating the time of death and can aid in forensic investigations. Here, a computational approach is described that has been validated against multiple, independent, and experimental investigations. The approach only requires one subjective input parameter (the heat transfer coefficient). A simple algorithm serves as a guidepost to the selection of this parameter. The algorithm incorporates clothing and the space in which the body is housed. Heat transfer coefficients that range from h = 2 W/m2/°C for bodies that are heavily clothed to h = 9 W/m2/°C for bodies that are nude (in air). The method also requires setting of ambient temperature conditions (ambient temperature)—however, that input is often available. The paucity of inputs makes this technique remarkably easy to employ. The new method is also able to calculate cadaver cooling rates for situations where the cadaver is in a timewise or spatially changing thermal environment (diurnal temperature variations, bodies partially submerged in water, changes to weather, insolation, etc.). Results from the present calculations are compared with a large body of measurements from the literature; it was found that the predictions and measurements were in excellent agreement, regardless of the ambient temperature conditions and the nature of the clothing of the body. This new calculation approach can be used with reasonable accuracy for determining cadaver cooling and time since death.

Abstract:Introduction: The accurate estimation of postmortem interval (PMI), the time between death and discovery of the body, is crucial in forensic science investigations as it impacts legal outcomes. PMI estimation in extremely cold environments becomes susceptible to errors and misinterpretations, especially with prolonged PMIs. This study addresses the lack of data on decomposition in extreme cold by providing the first overview of decomposition in such settings. Moreover, it proposes the first postmortem microbiome prediction model for PMI estimation in cold environments, applicable even when the visual decomposition is halted. Methods: The experiment was conducted on animal models in the second-coldest region in the United States, Grand Forks, North Dakota, and covered 23 weeks, including the winter months with temperatures as low as −39°C. Random Forest analysis models were developed to estimate the PMI based either uniquely on 16s rRNA gene microbial data derived from nasal swabs or based on both microbial data and measurable environmental parameters such as snow depth and outdoor temperatures, on a total of 393 samples. Results: Among the six developed models, the best performing one was the complex model based on both internal and external swabs. It achieved a Mean Absolute Error (MAE) of 1.36 weeks and an R2 value of 0.91. On the other hand, the worst performing model was the minimal one that relied solely on external swabs. It had an MAE of 2.89 weeks and an R2 of 0.73. Furthermore, among the six developed models, the commonly identified predictors across at least five out of six models included the following genera: Psychrobacter (ASV1925 and ASV1929), Carnobacterium (ASV2872) and Pseudomonas (ASV1863). Discussion: The outcome of this research provides the first microbial model able to predict PMI with an accuracy of 9.52 days over a six-month period of extreme winter conditions.

Abstract:Abstract Background Time of death estimation in humans for the benefit of forensic medicine has been successfully approached by Henssge, who modelled body cooling based on measurements of Marshall and Hoare. Thereby, body and ambient temperatures are measured at the death scene to estimate a time of death based on a number of assumptions, such as initial body temperature and stable ambient temperature. While so far, practical use of the method resorted to paper print outs or copies of a nomogram using a ruler, increasingly, users are interested in computer or mobile device applications. We developed a computational solution that has been available online as a web accessible PHP program since 2005. From that, we have received numerous requests not so much to detail our code but to explain how to efficiently approximate the solution to the Henssge equation. Methods To solve Henssge’s double exponential equation that models physical cooling of a body, it is sufficient to determine a difference term of the equation that will be close to zero for the correct time of death using a discrete set of all sensible possible solutions given that the modelled time frame has practical upper limits. Best post-mortem interval approximation yields minimal difference between equation terms Results The solution is approximated by solving the equation term difference for a discrete set of all possible time of death intervals that are sensibly found, and by then determining the particular time of death where equation term difference is minimal. Conclusions The advantage of a computational model over the nomogram is that the user is also able to model hypothermia and hyperthermia. While mathematically impossible to solve in a straightforward way, solutions to the Henssge equation can be approximated computationally.

Abstract:Microbial succession has been suggested to supplement established postmortem interval (PMI) estimation methods for human remains. Due to limitations of entomological and morphological PMI methods, microbes are an intriguing target for forensic applications as they are present at all stages of decomposition. Previous machine learning models from soil necrobiome data have produced PMI error rates from two and a half to six days; however, these models are built solely on amplicon sequencing of biomarkers (e.g., 16S, 18S rRNA genes) and do not consider environmental factors that influence the presence and abundance of microbial decomposers. This study builds upon current research by evaluating the inclusion of environmental data on microbial-based PMI estimates from decomposition soil samples. Random forest regression models were built to predict PMI using relative taxon abundances obtained from different biological markers (bacterial 16S, fungal ITS, 16S-ITS combined) and taxonomic levels (phylum, class, order, OTU), both with and without environmental predictors (ambient temperature, soil pH, soil conductivity, and enzyme activities) from 19 deceased human individuals that decomposed on the soil surface (Tennessee, USA). Model performance was evaluated by calculating the mean absolute error (MAE). MAE ranged from 804 to 997 accumulated degree hours (ADH) across all models. 16S models outperformed ITS models (p = 0.006), while combining 16S and ITS did not improve upon 16S models alone (p = 0.47). Inclusion of environmental data in PMI prediction models had varied effects on MAE depending on the biological marker and taxonomic level conserved. Specifically, inclusion of the measured environmental features reduced MAE for all ITS models, but improved 16S models at higher taxonomic levels (phylum and class). Overall, we demonstrated some level of predictability in soil microbial succession during human decomposition, however error rates were high when considering a moderate population of donors.

Abstract:Death is a multifaceted process wherein each individual cell and tissue has a metabolic homeostasis and a time of functional cessation defined by the dying process as well as by intrinsic and extrinsic factors. Decomposition is physiologically associated with the release of different types of volatile organic compounds (VOCs), and these form volaboloma mortis. The main purpose of this study was to record the volabolomic fingerprint produced by volatile molecules during the physiological decomposition process of human tissue and muscle cells. The volatile chemical signature has important implications for an open issue in forensics and pathology, namely the estimation of the postmortem interval (PMI), which decreases in accuracy with the passage of time. Volatile metabolites emitted from human tissues and muscle cells at 0, 24, 48, and 72 h were recorded in real time with an electronic nose sensor device. The key findings were the continuous sampling of VOCs emitted from tissues and cells. These showed a common behavior as time progressed; particularly, after 48 h the distributions became dispersed, and after 72 h they became more variable. Volabolomic fingerprinting associated with time progression relevant to the study of PMIs was reconstructed. Additionally, there may be broader applications, such as in dog training procedures for detecting human remains, and perhaps even for studying scavenger and insect attractants.

Abstract:Objective: To develop and implement an online application for the numerical method of determination of the postmortem interval and its limiting faults, caused by errors of measurement processes, based on the Marshall-Hoare law of cooling and dual thermometry of the corpse's core. Material and methods: The numerical search for a minimum of two variables' target function, obtained from the system of non-linear Marshall-Hoare equations, reflecting the dual thermometry of the corpse's core was performed. The code of online program was written in Python 3 programming language. Results: The method for determination of the postmortem interval, based on iterative algorithm of non-linear optimization, allowing by dual thermometry of the corpse to consider the uniqueness of its cooling conditions, as well as to calculate the degree of uncertainty of computational estimations, caused by the impact of measurement errors, has been developed. The method is implemented in the format of the Warm Bodies Z online application. Conclusions: The developed numerical method and its online application are recommended for determining of the postmortem interval.
An economical and efficient method for postmortem DNA sampling in mass fatalities

Abstract:In mass fatality events, the need to identify large numbers of deceased persons using DNA can be a significant drain on already overburdened forensic practitioners, both in the field setting and the laboratory. The laboratory may be required to extract DNA from a variety of postmortem sample types, family or direct reference samples related to the missing, and perform matching of these results in a short period of time. While most forensic institutions are well equipped to handle both family and direct reference samples, postmortem samples such as bone or heterogeneous tissue samples can be difficult for labs to analyze. We have devised an easily deployable, efficient, and inexpensive method for collecting postmortem DNA samples on commercially available DNA preservation cards ("FTA®" cards). FTA® cards are already widely used in forensic labs and are convenient for shipping due to their small volume and stability at room temperature. We evaluated the suitability of a protocol involving swabbing of incisions made on cadavers and sample deposition onto FTA® cards over various postmortem intervals and under different environmental conditions. Each trial took place during a different point in the calendar year to evaluate the effects of seasonal weather patterns and temperature on decomposition, DNA yield, and rates of DNA degradation. To further account for the effects of seasonality (temperature and humidity), the progression of body decomposition was recorded following the Total Body Score (TBS) method [1]. DNA degradation was assessed either through STR amplification of 1.2 mm FTA punches or DNA extraction from 3.0 mm punches followed by real-time PCR quantification and STR amplification and genotyping. No consistent relationship was observed between postmortem interval and DNA degradation. Instead, the TBS score, which captures the stage of body decomposition, was shown to correlate well with DNA quantity. A TBS of 15 and below consistently yielded strong partial or full profiles (20 STR loci and Amelogenin using the PowerPlex 21 System) from all individuals from either 1.2 mm or 3.0 mm punches. Transfer of sample swabs to FTA cards is shown to be a simple and effective method for both field and laboratory operations over a range of conditions that can be evaluated by field forensic practitioners based on a body decomposition score. The approach could be beneficially integrated into mass fatality response plans.
Identifying Factors to Help Improve Existing Decomposition-Based PMI Estimation Methods

Abstract:Accurately assessing the postmortem interval (PMI) is an important task in forensic science. Some of the existing techniques use regression models that use a decomposition score to predict the PMI or accumulated degree days (ADD), however, the provided formulas are based on very small samples and the accuracy is low. With the advent of Big Data, much larger samples can be used to improve PMI estimation methods. We, therefore, aim to investigate ways to improve PMI prediction accuracy by (a) using a much larger sample size, (b) employing more advanced linear models, and (c) enhancing models with factors known to affect the human decay process. Specifically, this study involved the curation of a sample of 249 human subjects from a large-scale decomposition dataset, followed by evaluating pre-existing PMI/ADD formulas and fitting increasingly sophisticated models to estimate the PMI/ADD. Results showed that including the total decomposition score (TDS), demographic factors (age, biological sex, and BMI), and weather-related factors (season of discovery, temperature history, and humidity history) increased the accuracy of the PMI/ADD models. Furthermore, the best performing PMI estimation model using the TDS, demographic, and weather-related features as predictors resulted in an adjusted R-squared of 0.34 and an RMSE of 0.95. It had a 7% lower RMSE than a model using only the TDS to predict the PMI and a 48% lower RMSE than the pre-existing PMI formula. The best ADD estimation model, also using the TDS, demographic, and weather-related features as predictors, resulted in an adjusted R-squared of 0.52 and an RMSE of 0.89. It had an 11% lower RMSE than the model using only the TDS to predict the ADD and a 52% lower RMSE than the pre-existing ADD formula. This work demonstrates the need (and way) to incorporate demographic and environmental factors into PMI/ADD estimation models.
