Abstract:Regulatory T cells (Tregs) expressing TCRαβ play a critical role in the maintenance of the immune system homeostasis. Tregs express the cell surface markers CD4 and CD25 as well as the transcription factor Foxp3. Foxp3+CD4+CD25+TCRαβ+ Tregs can be generated from mouse and human CD4+CD25− T cells in vitro via TGF-β induction. As growing evidences suggest that γδ T cells also have immunoregulatory function, we have attempted to identify and characterize Foxp3+ cells in mouse and human γδ T cells. We found that freshly isolated mouse splenic γδ T cells did not express Foxp3. When mouse splenocytes were stimulated with anti-TCRγδ in the presence of TGF-β, a population of Foxp3+ γδ T cells appeared, in most of which expressed CD25 as well. Compared with CD25− γδ T cells, TGF-β induced CD25+ γδ T cells not only expressed Foxp3, but also had increased TGF-β and GITR expression. Furthermore, the TGF-β induced γδ T cells mediated a potent immunosuppressive effect on anti-CD3 stimulated T cell activation and proliferation. In contrast, although a small fraction of human peripheral blood and tumor infiltrating γδ T cells expressed Foxp3, similar culture condition with anti-TCRγδ plus TGF-β failed to generate functional human Foxp3+ γδ T cells. In conclusion, our results suggest that mouse splenic Foxp3+ γδ T cells with suppressive function can be induced by TCR and TGF-β costimulation, whereas functional human Foxp3+ γδ T cells in peripheral blood could not be generated under the same condition.

Abstract:CD4(+)Foxp3(+) regulatory T cells (Tregs) play a crucial role in preventing autoimmunity and inflammation. There are naturally-derived in the thymus (tTreg), generated extrathymically in the periphery (pTreg), and induced in vitro culture (iTreg) with different characteristics of suppressiveness, stability, and plasticity. There is an abundance of published data on neuropilin-1 (Nrp-1) as a tTreg marker, but little data exist on iTreg. The fidelity of Nrp-1 as a tTreg marker and its role in iTreg remains to be explored. This study found that Nrp-1 was expressed by a subset of Foxp3(+)CD4(+)T cells in the central and peripheral lymphoid organs in intact mice, as well as in iTreg. Nrp-1(+)iTreg and Nrp-1(-)iTreg were adoptively transferred into a T cell-mediated colitis model to determine their ability to suppress inflammation. Differences in gene expression between Nrp-1(+) and Nrp-1(-)iTreg were analyzed by RNA sequencing. We demonstrated that the Nrp-1(+) subset of the iTreg exhibited enhanced suppressive function and stability compared to the Nrp-1(-) counterpart both in vivo and in vitro, partly depending on IL-10. We found that Nrp-1 is not an exclusive marker of tTreg, however, it is a biomarker identifying a new subset of iTreg with enhanced suppressive function, implicating a potential for Nrp-1(+)iTreg cell therapy for autoimmune and inflammatory diseases.

Abstract:Foxp3+ regulatory T cells (Treg) playing a crucial role in the maintenance of immune tolerance and prevention of autoimmune diseases consist of thymus-derived naturally occurring CD4+Foxp3+ Treg cells (nTreg) and those that can be induced ex vivo with TGF-β (iTreg). Although both Treg subsets share similar phenotypes and functional characteristics, they also have potential biologic differences on their biology. The role of iTreg in regulating B cells remains unclear so far. The suppression assays of Treg subsets on activation, proliferation, and Abs production of B cells were measured using a Treg and B cell coculture system in vitro. Transwell and Ab blockade experiments were performed to assess the roles of cell contact and soluble cytokines. Treg were adoptively transferred to lupus mice to assess in vivo effects on B cells. Like nTreg, iTreg subset also directly suppressed activation and proliferation of B cells. nTreg subset suppressed B cell responses through cytotoxic manner related to expression of granzyme A, granzyme B, and perforin, whereas the role of iTreg subset on B cells did not involve in cytotoxic action but depending on TGF-β signaling. Furthermore, iTreg subset can significantly suppress Ab produced by lupus B cells in vitro. Comparison experiments using autoantibodies microarrays demonstrated that adoptive transfer of iTreg had a superior effect than nTreg subset on suppressing lupus B cell responses in vivo. Our data implicate a role and advantage of iTreg subset in treating B cell–mediated autoimmune diseases, boosting the translational potential of these findings.

About 70%CD4+CD25+CD127- T cells expressed FoxP3 of two groups and the positive rate of FoxP3 had no significantly difference between two groups [(67.43±6.41)% vs. (69.37±5.15)%, P>0.05]. Conclusions For the liver transplantation patients and the health individuals, CD39+Treg may be the main cell population for immunomodulatory effects and can be regarded as one surface marker to identify Treg cell. Compared with the healthy control group, CD4+CD25+CD39+Treg cells are significantly reduced in the peripheral blood of liver transplantation patients.

Abstract:γδ T cells represent a relevant proportion of T lymphocytes that express T cell receptors (TCRs) encoded by the γ and δ T cell receptor genes. Whereas the most circulating γδ T cell type, Vδ2 T cells, has been described and studied intensively, the potential role of Vδ1 T cells remains largely unclear. Here, we identified that expanded peripheral blood Vδ1 T cells stimulated with anti-human TCR Vδ1 monoclonal antibody (mAb) in vitro predominantly expressed forkhead box p3 (Foxp3). In addition, these cells also expressed other regulatory T cell-related molecules, such as CD25, glucocorticoid-induced TNFR family-related protein and cytotoxic T lymphocyte-associated protein-4 (CTLA-4), and held the potent capacity for the production of transforming growth factor beta 1 (TGF-β1). These autocrine and/or paracrine TGF-β1 could bind TGF-β1 receptors on Vδ1 T cells and induce sustained Foxp3 expression. Moreover, Foxp3 expression coincided with high CD25 expression. CD25 high Vδ1 T cells exhibited stronger suppression on CD4 + T cell proliferation compared with TGF-β1-induced CD25 high CD4 + regulatory T cells. Therefore, our phenotypic and functional analyses first demonstrate the potential regulatory property of anti-human TCR Vδ1 mAb-activated Vδ1 T cells. These results will broaden our understanding about the role of peripheral blood Vδ1 T cells under physical and pathological conditions.

Abstract:We have previously identified a human CD8+HLA-DR+ regulatory T cell subset with the ability to suppress proliferation of autologous PBMCs responder cells through cell contact and CTLA-4 co-inhibitory molecule. The present study characterizes the complete phenotype of CD8+HLA-DR+ Treg cells which showed great similarities with classical CD4+ cells expressing forkhead box P3 (FOXP3). The shared features included the expression of programmed cell death protein 1 (PD-1), T-cell immunoreceptor with Ig and ITIM domains (TIGIT), C-C chemokine receptor type 4 and 5 (CCR4 and CCR5), low expression of CD127, and a memory and effector-like phenotype. CD8+HLA-DR+ Treg-induced suppression on CD8+ responder T cells was abrogated by an anti-PD1 neutralizing antibody. Anti-PD-1 did not abrogate the suppressor effect induced on responder CD4+ T cells. In addition, CD8+HLA-DR+ Treg induced a preferential death on responder CD8+ T cells. This effect was not reversed by PD-1 neutralization. After activation, most CD8+HLA-DR+ Treg acquire programmed death-ligand 1 (PD-L1) expression. Interestingly, PD-L1 may induce apoptosis through CD80 expressed on activated CD8+ responder T cells. After PBMCs stimulation, CD8+HLA-DR+ Treg cells showed an increased frequency of IFN-γ and TNFα positive cells and higher degranulation. These data strongly argue against CD8+HLA-DR+ Treg being exhausted cells. Overall, the data presented in this study indicate that CD8+HLA-DR+ Treg and CD4+FOXP3+ Treg share phenotypic and functional features, which may provide cues to similar involvements in the control of antitumor immune responses and autoimmunity.

Abstract:Regulatory T (T reg) cells are critical regulators of immune tolerance. Most T reg cells are defined based on expression of CD4, CD25, and the transcription factor, FoxP3. However, these markers have proven problematic for uniquely defining this specialized T cell subset in humans. We found that the IL-7 receptor (CD127) is down-regulated on a subset of CD4+ T cells in peripheral blood. We demonstrate that the majority of these cells are FoxP3+, including those that express low levels or no CD25. A combination of CD4, CD25, and CD127 resulted in a highly purified population of T reg cells accounting for significantly more cells that previously identified based on other cell surface markers. These cells were highly suppressive in functional suppressor assays. In fact, cells separated based solely on CD4 and CD127 expression were anergic and, although representing at least three times the number of cells (including both CD25+CD4+ and CD25−CD4+ T cell subsets), were as suppressive as the “classic” CD4+CD25hi T reg cell subset. Finally, we show that CD127 can be used to quantitate T reg cell subsets in individuals with type 1 diabetes supporting the use of CD127 as a biomarker for human T reg cells.

Abstract:ABSTRACT There are conflicting data about the frequency and role of regulatory T cells (Tregs) during the course of HIV infection. Peripheral blood of a large cohort of HIV-infected patients (n = 131) at different stages of disease, including 15 long-term nonprogressors and 21 elite controllers, was analyzed to determine the frequency and phenotype of Tregs, defined as CD4+, CD25high, CD127low, FoxP3high cells. A significantly increased relative frequency of Tregs within the CD4+ compartment of HIV+ patients compared to that of healthy controls (P < 0.0001) was observed. Additionally, the relative frequency of Tregs directly correlated with HIV viral load and inversely with CD4+ counts. However, the absolute Treg number was reduced in HIV-infected patients versus healthy controls (P < 0.0001), with the exception of elite controllers (P > 0.05). The loss of absolute Treg numbers coincided with rising markers of immune activation (P < 0.0006). The initiation of antiviral therapy significantly increased absolute Treg numbers (P < 0.0031). We find that the expression of CD39, a newly defined ectonucleotidase with immunomodulatory functions on Tregs, correlated with progressive HIV disease, HIV viral load, and immune activation. Of note, when tested in peripheral blood mononuclear cells of healthy volunteers, the in vitro capacity to suppress T-cell proliferation was limited to CD4+, CD25high, CD39+ T cells. Interestingly, Tregs of elite controllers exhibited not only the highest expression of CCR5, CTLA-4, and ICOS but also the lowest level of CD39. The data presented here reconcile the seemingly contradictory results of previous studies looking at Tregs in HIV and highlight the complexity of Treg-mediated immunoregulation during human viral infections.

Abstract:Abstract Forkhead box P3 (FOXP3) is a well-known molecular marker for regulatory T cells (Tregs) and is considered as a master regulatory transcription factor for the development, maturation and function of Tregs. Mutations in FOXP3 lead to severe immune deficiencies and cancer in human and mice. Administration of Foxp3-expressing Tregs drastically reduced the disease severity in animal models of diabetes, asthma and multiple sclerosis giving rise to hope for therapeutic options. It is not clear whether cell types other than Treg express FOXP3. While investigating TGFβ-mediated signaling in the myeloid lineage, we discovered a subset of myeloid derived suppressor cells (Gr-1+CD11b+ cells or MDSCs) that express FOXP3. Indeed these cells were CD3 negative suggesting that they are not of T cell lineage. Upon further investigation, we observed that these FOXP3 expressing myeloid cells are present in different tissues of mouse. Moreover, we found that they are elevated under pathological conditions. We are currently identifying the molecular function and mechanisms of Foxp3 expression in the myeloid subsets. Our work will provide new insight into the FOXP3 research area beyond regulatory T cells, and might reveal its role in regulating the immunosuppressive activity of MDSCs. Isolation and Identification of CD4+CD25+ Regulatory T Cells

Abstract:Objective:Regulatory T cells are the hotpoint of immunology for recent years. In this study the preparation of high purity regulatory T cells (Treg) was investisated and the phenotypic and functional characteristics of innate Treg was identified. Methods:The phenotype of 3-subpopulation of CD4+/CD4+CD25+/ CD4+CD25-T cells derived from normal murine SPL were analyzed by FACS. CD+CD25+Tregs were isolated by two-step magnetic cell sorting (MACS) and the Treg-specific transcription factor Foxp3,intracellular CTLA-4 and surface antigen CD127 were identified by FACS. Results:①These three CD4+T cell populations defined by varying levels of CD25 expression exhibited marked differences in a number of some intracellular or surface antigens. The CD4+CD25+T cells expressed higher levels of forkhead box P3 ( Foxp3 ),cytotoxic T lymphocyte associated antigen 4 ( CTLA-4),glucocorticoid-induced tumor necrosis factor receptor (GITR )and CD69 (P0.01) and lower levels of CD28 (P0.05) than the CD4+ or CD4+CD25-subsets. These innate CD4+CD25+ Tregs were typically confined within the CD45RBlow T cell subset. ②After CD4+CD25+ T cells were isolated from murine splenocytes by 2-step magnetic cell sorting,90.19% (range,87.29%～92.04%) of the cells expressed both CD4 and CD25,and the yield was 1%～1.2%. Foxp3 was preferentially expressed by CD4+CD25+T cells (66.6±1.033%),whereas CD4+ or CD4+CD25-subpopulation contained low level of Foxp3 (23.97±1.997%,16.78±3.24%). CTLA-4 was expressed by a significantly higher proportion of cells in CD4+CD25+ population (77.37±1.67%) as compared to CD4+ (21.97±2.39%) or CD4+CD25-(18.04±3.16) subsets. The isolated CD4+CD25+ T cells also expressed the low intensity of CD127. Conclusion:These three innate CD4+T-cell populations defined by varying levels of CD25 expression in mice exhibited marked differences in a number of other intracellular or surface antigens,such as Foxp3,CTLA-4,GITR,CD45RB,CD69,CD28. Cells isolated from murine SPL products by 2-step magnetic cell separation protocol (negative selection and positive selection) were highly enriched in CD4+T cells with phenotypic and functional characteristics of innate CD4+CD25+Treg.

Abstract:Background: CD39 and CD73 are two novel cell surface markers of CD25(high)Foxp3(+) regulatory T-cells (Tregs). Concordant expression of these two ectoenzymes not only discriminate Tregs from other cell populations, but also generates pericellular adenosine, which has been reported to suppress proliferation of activated T effector (Teff) cells. Because it is currently unclear whether human ectoenzymes (CD39/CD73) are involved in the impaired suppressive activity of Tregs in psoriasis, we examined the frequencies and phenotypes of CD39/CD73-expressing Tregs and related receptor adenosine receptor 2A (A(2A)R) in peripheral blood of patients with different types of psoriasis. Methods: Peripheral blood mononuclear cells (PMBC) were prepared from patients with three different types of psoriasis (psoriasis vulgaris, pustular psoriasis and erythrodermic psoriasis). CD4(+) cells were separated from PBMC by negative selection on midiMACS columns, and the frequencies and phenotypes of CD39 and CD73 expressing Tregs, and A(2A)R expressing Teff were all determined by flow cytometry analysis. Blood from healthy volunteers served as controls. Results: The expression of single CD73(+) Tregs was markedly reduced (approximately 50%) in psoriasis vulgaris, compared to normal controls. In pustular psoriasis, the mean numbers of CD39(+) Tregs and A(2A)R(+) Teff was significantly lower than in normal controls. Among three different types of psoriasis, CD39 expression was strikingly reduced in the blood Treg population of pustular psoriasis patients. Decreased CD73+ Tregs levels were observed in psoriasis vulgaris compared to pustular psoriasis and erythrodermic psoriasis. Conclusions: The differences in the expression of CD39(-) and CD73(-) Tregs may be a factor in the pathogenesis of psoriasis.
Expression and Clinical Significance of Regulatory T Cells in Patients with Colon Cancer

Abstract:Objective To investigate the contents of regulator T cells(Treg) in patients with colon cancer,and explore the expression and clinical significance of FoxP3 gene in these Treg cells.Methods The levels of Treg cells in peripheral blood of 15 preoperative patients with colon cancer were determined using flow cytometry,the expression of FoxP3 gene in these Treg cells was detected with RT-PCR techniques,and the results were analyzed statistically in comparison with that of 15 healthy volunteers.Results The percentage of Treg cells in total CD4+T cells isolated from the patients with colon cancer was(11.12±0.57)%,which was higher than that from healthy volunteers(8.77±0.66)%(P0.05);The FoxP3 gene expression of Treg cells in the patients with colon cancer((0.84±0.02)%)was significantly higher than that in healthy volunteers(0.76±0.03)%(P0.05).Conclusions The number of Treg cells in patients with colon cancer was higher than that in normal people and FoxP3 gene expression of Treg cells was increased,which indicated that FoxP3 genes mould play a pivotal role in function regulation of Treg cells,and thereby affected the antitomor autoimmunity function of the patients with colon cancer.

Abstract:CD4⁺CD25⁺Foxp3⁺ T lymphocytes, known as regulatory T cells or T(regs), have been proposed to be a lineage of professional immune suppressive cells that exclusively counteract the effects of the immunoprotective "helper" and "cytotoxic" lineages of T lymphocytes. Here we discuss new concepts on the mechanisms and functions of T(regs). There are several key points we emphasize: 1. Tregs exert suppressive effects both directly on effector T cells and indirectly through antigen-presenting cells; 2. Regulation can occur through a novel mechanism of cytokine consumption to regulate as opposed to the usual mechanism of cytokine/chemokine production; 3. In cases where CD4⁺ effector T cells are directly inhibited by T(regs), it is chiefly through a mechanism of lymphokine withdrawal apoptosis leading to polyclonal deletion; and 4. Contrary to the current view, we discuss new evidence that T(regs), similar to other T-cells lineages, can promote protective immune responses in certain infectious contexts (Chen et al., 2011; Pandiyan et al., 2011). Although these points are at variance to varying degrees with the standard model of T(reg) behavior, we will recount developing findings that support these new concepts.

Expression and significance of CD39 on regulatory T cells in the peripheral blood of patients following liver transplantation

Abstract:Objective To study the clinical significance of CD39 on regulatory T (Treg) cells in the peripheral blood (PB) of patients following liver transplantation and to evaluate the relationship between the levels of CD39+ Treg cells in the PB and acute rejection.Methods A prospective study was conducted to compare the CD39+ Treg cells from 76 liver transplant patients with those coming from 20 age-matched healthy individuals.The PB samples were collected within one year at different time points post-transplant.Blood samples and liver biopsies were collected at the time when acute rejection was diagnosed.The percentages of CD39 within the CD4+ CD25+ T cells were measured by using flow cytometry.The liver transplant patients were classified into two groups:the rejection group which consisted of 17 patients who an episode of acute rejection,and the non-rejection group consisted of the remaining 59 patients who had no acute rejection episodes.The percentages of CD39 within the CD4 + CD25 + T cells and the inhibition function of the CD39+ Treg cells were compared between the two liver transplant groups.Results The percentages of CD39 within the CD4+ CD25+ cells were significantly lower in the rejection group during acute rejection as compared to the non-rejection group (P ＜ 0.05).The percentages of CD39 within the CD4 + CD25 + cells were negatively correlated with the Rejection Activity Index (r =-0.86,P ＜ 0.05).The inhibition rate regarding the CD4+ CD25+ CD39+ Treg cells in patients with acute rejection was significantly lower than those without rejection (P ＜ 0.05).Conclusions The percentages of CD39 within the CD4+ CD25+ T cells were significantly lower in the rejection group during acute rejection and were negatively correlated with the RAI.The inhibition rate regarding the CD4+ CD25+ CD39+ Treg cells in patients with acute rejection was significantly lower than those without rejection.

Abstract:Regulatory T (Treg) cells expressing the transcription factor forkhead box P3 (Foxp3) play a requisite role in the maintenance of immunological homeostasis and prevention of peripheral self-tolerance breakdown. Although Foxp3 by itself is neither necessary nor sufficient to specify many aspects of the Treg cell phenotype, its sustained expression in Treg cells is indispensable for their phenotypic stability, metabolic fitness, and regulatory function. In this review, we summarize recent advances in Treg cell biology, with a particular emphasis on the role of Foxp3 as a transcriptional modulator and metabolic gatekeeper essential to an effective immune regulatory response. We discuss these findings in the context of human inborn errors of immune dysregulation, with a focus on FOXP3 mutations, leading to Treg cell deficiency. We also highlight emerging concepts of therapeutic Treg cell reprogramming to restore tolerance in the settings of immune dysregulatory disorders.

Abstract:Regulatory T cells (T-regs) are specialized in immune suppression and play a dominant role in peripheral immune tolerance. T-reg cell lineage development and function maintenance is determined by the forkhead box protein 3 (FoxP3) transcriptional factor, whose activity is fine-tuned by its post-translational modifications (PTMs) and interaction partners. In this review, we summarize current studies in the crystal structures, the PTMs and interaction partners of FoxP3 protein, and discuss how these insights may provide a roadmap for new approaches to modulate T-reg suppression, and new therapies to enhance immune tolerance in autoimmune diseases.
A study on the expression and function of extracellular adenosine metabolism molecules CD39 and CD73 in CD4+ CD25high Foxp3+ regulatory T cells

Abstract:To analyze the expression and function of molecules CD39/CD73 produced during extracellular metabolism of adenosine of CD4+ CD25highFoxp3+ regulatory T cells,15 healthy people and 10 patients with psoriasis vulgaris were selected.The percentage and attributes of CD39/CD73-expressing regulatory T cells in peripheral blood were measured by flow cytometry.Immunohistochemical staining was used to analyze expression of Foxp3,CD39 and CD73 in biopsy tissue from healthy controls.Using the flow sorting,CD4+ CD25high,CD4+ CD25highCD39+ and CD4+ CD25highCD39+ CD73+ regulatory T ceils were isolated from peripheral blood from 5 healthy controls,[3 H]-thymidine incorporation assay was performed to analyze the inhibition of CD4+ CD25-T cells proliferation by the above three different subgroups of regulatory T cells.Compared to CD4+ CD25mid and CD4+ CD25-T cells,normal healthy peripheral blood CD4+ CD25high Foxp3+ regulatory T cells showed significantly higher expression levels of CD39(P＜0.01)and less CD73(P＜0.05).In normal control biopsies,CD39+ ceils were scattered throughout the epidermis and dermis,while CD73+ cells were localized predominantly in the dermis.The inhibitory capacity of CD39+ CD73-Treg cells was significantly higher than that of the unfractionated Treg cell-pool(P＜0.01).The expression of CD73+ CD4+ CD25high Foxp3+ Tregs was markedly reduced in psoriasis vulgaris patients(P＜0.01).Therefore,the adenosine metabolism is an important part in regulatory T cells function and related to the pathogenesis of psoriasis vulgaris.

Abstract:CD4+CD25+ regulatory T cells (Treg) are instrumental in the maintenance of immunological tolerance. One critical question is whether Treg can only be generated in the thymus or can differentiate from peripheral CD4+CD25− naive T cells. In this paper, we present novel evidence that conversion of naive peripheral CD4+CD25− T cells into anergic/suppressor cells that are CD25+, CD45RB−/low and intracellular CTLA-4+ can be achieved through costimulation with T cell receptors (TCRs) and transforming growth factor β (TGF-β). Although transcription factor Foxp3 has been shown recently to be associated with the development of Treg, the physiological inducers for Foxp3 gene expression remain a mystery. TGF-β induced Foxp3 gene expression in TCR-challenged CD4+CD25− naive T cells, which mediated their transition toward a regulatory T cell phenotype with potent immunosuppressive potential. These converted anergic/suppressor cells are not only unresponsive to TCR stimulation and produce neither T helper cell 1 nor T helper cell 2 cytokines but they also express TGF-β and inhibit normal T cell proliferation in vitro. More importantly, in an ovalbumin peptide TCR transgenic adoptive transfer model, TGF-β–converted transgenic CD4+CD25+ suppressor cells proliferated in response to immunization and inhibited antigen-specific naive CD4+ T cell expansion in vivo. Finally, in a murine asthma model, coadministration of these TGF-β–induced suppressor T cells prevented house dust mite–induced allergic pathogenesis in lungs.

Abstract:Regulatory T cells (Tregs) are essential to maintain immune homeostasis by promoting self-tolerance. Reduced Treg numbers or functionality can lead to a loss of tolerance, increasing the risk of developing autoimmune diseases. An overwhelming variety of human Tregs has been described, based on either specific phenotype, tissue compartment, or pathological condition, yet the bulk of the literature only addresses CD25-positive and CD127-negative cells, coined by naturally occurring Tregs (nTregs), most of which express the transcription factor Forkhead box protein 3 (FOXP3). While the discovery of FOXP3 was seminal to understanding the origin and biology of nTregs, there is evidence in humans that not all T cells expressing FOXP3 are regulatory, and that not all Tregs express FOXP3. Namely, the activation of human T cells induces the transient expression of FOXP3, irrespective of whether they are regulatory or inflammatory effectors, while some induced T cells that may be broadly defined as Tregs (e.g., Tr1 cells) typically lack demethylation and do not express FOXP3. Furthermore, it is unknown whether and how many nTregs exist without FOXP3 expression. Several other candidate regulatory molecules, such as GITR, Lag-3, GARP, GPA33, Helios, and Neuropilin, have been identified but subsequently discarded as Treg-specific markers. Multiparametric analyses have uncovered a plethora of Treg phenotypes, and neither single markers nor combinations thereof can define all and only Tregs. To date, only the functional capacity to inhibit immune responses defines a Treg and distinguishes Tregs from inflammatory T cells (Teffs) in humans. This review revisits current knowledge of the Treg universe with respect to their heterogeneity in phenotype and function. We propose that it is unavoidable to characterize human Tregs by their phenotype in combination with their function, since phenotype alone does not unambiguously define Tregs. There is an unmet need to align the expression of specific markers or combinations thereof with a particular suppressive function to coin functional Treg entities and categorize Treg diversity.
