Abstract:OBJECTIVE:To investigate effects of oxidative stress on human embryonic stem cells (hESC) and their spontaneously differentiated fibroblastic progenies (at passage 5).BACKGROUND DATA:In ischemic disease models, high levels of free radicals and reactive oxygen species are critical factors in decreasing survivability and engraftment of transplanted/transfused cells. Hence, it is imperative to characterize response of hESC and their differentiated progenies to oxidative stress.METHODS:Oxidative stress was induced either by (i) varying durations (0 to 40 min) of photodynamic treatment (diode laser, 664 nm, 30 mW) in the presence of 10 microM methylene blue as a photosensitizer, or by (ii) exposure to varying concentrations of hydrogen peroxide (0 to 50 microM) for a fixed duration of 40 min. Additionally, the effects of heat shock and mild oxidative stress preconditioning on oxidative stress response was also investigated.RESULTS:Consistently higher survivability (MTT assay) of hESC was observed compared to their differentiated fibroblastic progenies, upon exposure to equivalent levels of oxidative stress. Further experiments demonstrated that heat-shock pretreatment (42 degrees C for 90 min) did not enhance the resistance of either hESC or their differentiated progenies to oxidative stress (photodynamic treatment), but in fact had a slightly detrimental effect on their survivability upon subsequent exposure to oxidative stress. Similarly, preconditioning of both undifferentiated hESC and their differentiated progenies with low levels of oxidative stress also did not enhance cellular survivability upon subsequent exposure to much higher levels of oxidative stress induced by photodynamic treatment.CONCLUSIONS:Undifferentiated hESC are intrinsically more resistant to oxidative stress compared to their spontaneously differentiated fibroblastic progenies.
Effects of High Glucose on the Activity of GSH-PX,the Expressions of NOS and ICAM-1 in Human Umbilical Vein Endothelial Cells

Abstract:Objective To investigate the effects of high glucose on human umbilical vein endothelial cells (ECV304). Methods ECV304 cells were cultured in DMEM medium with or without high glucose, followed by the classification into control group and high glucose group.The activity of GSHPX in supernatant as detected by spectrophotometry assay, the mRNA expression of ICAM-1 of the cells was determined by RTPCR, meanwhile the expressions of eNOS and iNOS in the cells were observed by immunocytochemistry. Results Compared with control group,the activity of GSHPX was significantly decreased,while the protein level of eNOS and iNOS and ICAM1 mRNA expression were found to be markedly increased( P 0.01). Conclusions Acute high glucose stimulation may down regulate the activity of GSHPX and promote the expressions of ICAM1 mRNA and eNOS、iNOS protein in vascular endothelial cells.These changes play an important role in diabetic vascular complications in the early phase.
High Glucose And Free Fatty Acids Induce Endothelial Progenitor Cell Senescence Via Pgc-1 Alpha/Sirt1 Signaling Pathway

Abstract:The objective of the research was to investigate the function of endothelial progenitor cells (EPCs) in the conditions of high glucose and lipids, which has been widely used to mimic the metabolic disorder that occurs in type 2 diabetic mellitus, and further to verify the role of PGC-1 alpha and SIRT1, cellular energy metabolism regulators, in the process of senescence of EPCs with these combined stimuli. Circulating EPCs were incubated in absence or presence of high glucose (25mM), FFA (200 mu M) or both. EPCs senescence was assessed by beta-galactosidase staining, EPCs telomerase activity was measured by telomeric repeat amplification protocol assay, in vitro angiogenesis assay and MTT assays were performed to assess angiogenesis and proliferation ability of EPCs. The results showed that combined stimuli inhibited EPCs reendothelialization ability in vitro, accelerated EPCs senescence and decreased the telomerase activity. Meanwhile, with combined stimuli, the expression of PGC-1 increased whereas SIRT1 expression decreased in EPCs accompanied by activation of P53/P21 signaling pathway. Conversely, transfection of EPCs with PGC-1-siRNA rescued EPCs premature senescence and up-regulated SIRT1 and decreased P53/P21 expression, correlating closely with the down-regulation of PGC-1 alpha itself. In addition, the combined stimuli induced up-regulation of PGC-1 alpha expression was partly mediated by ROS and P38 signaling pathway. Overall, the data presented here identify PGC-1 as a potent negative regulator of EPCs' senescence under combined stimuli, which is partly mediated by SIRT1/P53/P21 signaling pathway.
Effects of glucose concentration fluctuation on proliferation,apoptosis and activity of endothelial nitric oxide synthase of endothelial progenitor cells

Abstract:Objective To observe the effects of glucose concentration fluctuations on proliferation,apoptosis and activity of endothelial nitric oxide synthase(eNOS)of endothelial progenitor cells(EPCs)derived from human peripheral blood.Methods Peripheral blood mononuclear cells were isolated from peripheral blood by Ficoll density gradient centrifugation.After cultured for 7 days in M199 medium containing rh-VEGF(30 ng/mL),rh-b-FGF(6 ng/mL)and 10% Fetal Bovine Serum,the attached cells were identified and verified to be EPCs.The cells were synchronized and then stimulated with glucose(5.5 mmol/L[normal control group],30 mmol/L[fixed concentration group],and 20 or 40 mmol/L with a mean of 30 mmol/L and a fluctuation interval of 3 h[fluctuation group])for different time periods(6,12,24,48 and 96 h).The proliferation of cells was examined by Microplate Spectrophotometer,the rate of apoptosis by Flow Cytometry,and the activity of eNOS by Luminescence Microplate Spectrophotometer.Results Compared with the fixed concentration group,the rate of proliferation and activity of eNOS in fluctuation group were significantly decreased(P<0.05),the rate of apoptosis in the fluctuation group was significantly higher.The rates of proliferation and apoptosis and the differences of eNOS activity were correlated with the stimulation time periods in both fixed and fluctuation groups(rproliferation=0.937,rapoptosis=0.927,reNOS=0.951,all P<0.05).Conclusion Glucose concentration fluctuation as an independent factor can promote the apoptosis of EPCs,depress the proliferation of EPC,and inhibit the activity of eNOS.EPCs have a poor adaptability.
Rosiglitazone Via Upregulation of Akt/Enos Pathways Attenuates Dysfunction of Endothelial Progenitor Cells, Induced by Advanced Glycation End Products

Abstract:Background and purpose:Advanced glycation end products (AGEs) and endothelial progenitor cells (EPCs) play key roles in pathogenesis of diabetes-related vascular complications. AGEs can induce dysfunction in EPCs. The peroxisome proliferator-activated receptor-gamma (PPAR gamma) agonists are widely used in the treatment of type 2 diabetes, and it remains unknown if they could attenuate EPC dysfunction induced by AGEs.Experimental approach:EPCs isolated from healthy adults were cultured with various concentrations of AGEs (0, 50, 100 and 200 mg center dot L-1) with or without rosiglitazone (10 nM), antibody for the receptors for AGE-human serum albumin (anti-receptor for advanced glycation end products (RAGE); 50 mu g center dot mL-1), phosphatidylinositol-3-kinase (PI3K) inhibitor (LY294002, 5 mu M), nitric oxide (NO) synthase inhibitor (L-NG-nitro-arginine methyl ester (L-NAME), 100 mu M) or sodium nitroprusside (SNP, 25 mu M). Proliferation, apoptosis, cell adhesion, migration and NO production in EPCs were assessed, and expressions of endothelial NO synthase (eNOS) and Akt were determined.Key results:Number, proliferation/migration capacities, eNOS and Akt phosphorylation as well as NO synthesized by EPCs were increased by rosiglitazone and reduced by AGEs. AGEs promoted while rosiglitazone reduced EPC apoptosis. The AGE-induced effects were significantly ameliorated by pre-incubation with rosiglitazone, RAGE antibody and SNP. The beneficial effects of rosiglitazone could be blocked by pretreatment with L-NAME and LY294002.Conclusions and implications:The PPAR gamma agonist rosiglitazone increased EPC function and attenuated EPC dysfunction induced by AGEs via upregulating the Akt-eNOS signal pathways of EPCs.
PKC/NADPH Oxidase Are Involved in the Protective Effect of Pioglitazone in High Homocysteine-Induced Paracrine Dyfunction in Endothelial Progenitor Cells.

Abstract:Increasing evidence suggests that EPCs improve neovascularization and endothelial regeneration via the production of paracrine factors. VEGF and IL-8 are major cytokines involved in EPC-mediated angiogenesis and re-endothelialization. In our previous studies, Hcy impaired EPC migratory and adhesive activities. We devised this study to determine whether Hcy could affect the expression and secretion of VEGF and IL-8 from EPCs. We found that high levels of Hcy (100-500 μM) decreased the EPC-mediated protein secretion and mRNA expression of VEGF and IL-8. Moreover, PIO, a PPARγ agonist, has been suggested to regulate EPC adhesion, migration, survival. In this study, PIO normalized the production of these cytokines by EPCs stimulated with Hcy. These effects of Hcy and PIO were primarily mediated by PKC and ROS via NADPH oxidase. We further confirmed this mechanism via knockdown of the NADPH oxidase subunits p67phox and Nox2. Furthermore, the PPARγ inhibitor GW9662 was not observed to abrogate the beneficial effect of PIO, indicating that PIO protected EPC paracrine function against Hcy in a PPARγ-independent manner.
High Glucose Conditions Suppress the Function of Bone Marrow-Derived Endothelial Progenitor Cells Via Inhibition of the Enos-Caveolin-1 Complex.

Abstract:The present study aimed to reveal how high glucose affects rat bone marrow-derived endothelial progenitor cells in vitro. Total mononuclear cells of bone marrow were obtained, cultured in endothelial cell growth medium-2 and identified by fluorescence microscopy. Using immunofluorescence, endothelial progenitor cells were identified by expression of VEGFR‑2 as well as CD133 and were further characterized as those adherent cells which were double positive by Dil‑ac‑LDL uptake and FITC-UEA-1 lectin binding. The attached cells were collected and glucose was added to the culture medium at various final concentrations (11.1, 33.3 and 55.5 mmol/l). Proliferation, migration and in vitro angiogenic ability of endothelial progenitor cells were measured. The change in mRNA and protein levels of caveolin-1 and endothelial nitric oxide synthase (eNOS) were examined; in addition, nitric oxide (NO) levels in the cell medium were measured. Based on the results, with increasing glucose concentration in the medium, proliferation, migration and in vitro angiogenic capacity of the cells were reduced, whereas mRNA and protein levels of caveolin-1 gene increased gradually. The mRNA levels of the eNOS gene did not differ, but the protein expression was reduced and NO levels in the culture medium declined. In conclusion, high glucose conditions are detrimental to the function of bone marrow‑derived endothelial progenitor cells in vitro, probably by damaging the eNOS-caveolin-1 complex, which results in the reduction of NO synthesis, eventually leading to the impaired function of endothelial progenitor cells.
Nadph Oxidase Activation Played A Critical Role in the Oxidative Stress Process in Stable Coronary Artery Disease

Abstract:Objectives: The study was designed to investigate the oxidative stress levels of endothelial progenitor cells (EPCs) in stable coronary artery disease (CAD) and to explore the underlying mechanisms of NADPH oxidase activation and subsequent EPCs dysfunction. Methods: EPCs were isolated from patients with stable CAD (n=50) and matched healthy volunteers (n=50). NADPH oxidase activation was detected by measuring the expression of each subunit using western blotting and qPCR analyses and the membrane translocation of p47(phox) using immunofluorescence. The in vivo angiogenesis capacity was evaluated using immunofluorescence by transplanting EPCs into a rat hind limb ischemia model. The PKC inhibitor GO-6983 was used to determine the role of PKC in NADPH oxidase activation. Results: Oxidative stress level was increased and the in vivo angiogenesis capacity was impaired in EPCs obtained from CAD subjects with the activation of NADPH oxidase. P47(phox) membrane translocation increased in CAD group vs controls. These effects were resolved by NADPH oxidase inhibition. Up-regulation of PKC alpha/beta 2 was found in EPCs from CAD subjects, PKC inhibition GO-6983 could reduce the expression and activity of NADPH oxidation. Conclusions: NADPH oxidase activation via p47(phox) membrane translocation played a critical role in the initiation and progression of CAD, and the PKC alpha/beta 2 signaling pathway might be involved.
Rosiglitazone ameliorates the dysfunction of endothelial progenitor cells induced by advanced glycation end products

Abstract:Objective To investigate whether peroxisome proliferator-activated receptor-gamma(PPARγ)agonists rosiglitazone can ameliorate the dysfunction of endothelial progenitor cells(EPCs)induced by advanced glycation end products(AGEs).Methods EPCs isolated from healthy adults were cultured with various concentrations of advanced glycation end product-human serum albumin(AGE-HSA)(0,50,100 and 200 mg/L)for 24 h in the absence or presence of rosiglitazone(10 nmol/L),anti-RAGE antibody(50 mg/L).We also included a blank control group.The proliferation,apoptosis,cell adhesion,migration,NO production and vascular cell adhesion molecule(VCAM)-1 secretion by EPCs were assessed.Results AGE-HSA at 200 mg/L significantly impaired the proliferation(reduced by [37.7±6.3]% compared with control,P<0.05),adherent([35.6±1.5]/HPF vs.[20.2±1.3]/HPF,P<0.05),and migration([28.6±2.8]/HPF vs.[15.0±2.1]/HPF,P<0.05)of EPCs,and aggravated the apoptosis([4.6±0.8]% vs.[15.2±1.0]%,P<0.05)of EPCs.Rosiglitazone significantly reversed the impairment effect of AGE-HSA on the proliferation(increased to [89.3±5.9]%,P<0.05),adherent(increased to [31.2±2.6]/HPF,P<0.05)and migration(increased to [21.6±3.1]/HPF,P<0.05)of EPCs,and ameliorated the apoptosis(reduced to [6.1±0.9]%,P<0.05)of EPCs.Rosiglitazone also increased the production of NO([16.2±1.0]μmol/L vs.[10.2±0.5] μmol/L,P<0.05)and reduced the secretion of VCAM-1([5.2±0.04] μg/L vs.[6.2±0.1] μg/L,P<0.05)of EPCs.Conclusion PPARγ agonist rosiglitazone ameliorates the dysfunction of endothelial progenitor cells induced by advanced glycation end products.

Abstract:Oxidative stress is considered to be a critical factor in diabetes-induced endothelial progenitor cell (EPC) dysfunction, although the underlying mechanisms are not fully understood. In this study, we investigated the role of high mobility group box-1 (HMGB-1) in diabetes-induced oxidative stress. HMGB-1 was upregulated in both serum and bone marrow-derived monocytes from diabetic mice compared with control mice. In vitro, advanced glycation end productions (AGEs) induced, expression of HMGB-1 in EPCs and in cell culture supernatants in a dose-dependent manner. However, inhibition of oxidative stress with N-acetylcysteine (NAC) partially inhibited the induction of HMGB-1 induced by AGEs. Furthermore, p66shc expression in EPCs induced by AGEs was abrogated by incubation with glycyrrhizin (Gly), while increased superoxide dismutase (SOD) activity in cell culture supernatants was observed in the Gly treated group. Thus, HMGB-1 may play an important role in diabetes-induced oxidative stress in EPCs via a positive feedback loop involving the AGE/reactive oxygen species/HMGB-1 pathway.
Effect of High Glucose-Induced Oxidative Stress on Paraoxonase 2 Expression and Activity in Caco-2 Cells

Abstract:(1) Background: Hyperglycemia leads to several biochemical and physiological consequences, such as the generation of advanced glycation end products (AGEs) and reactive oxygen species (ROS), which are involved in the development of several human diseases. Intestinal cells are continuously exposed to pro-oxidants and lipid peroxidation products from ingested foods, and also to glyco-oxidative damage. It has been reported that free radical generation may be linked to the development of inflammation-related gastrointestinal diseases. (2) Methods: The effects of high glucose (HG) treatment (50 mM) were assessed in terms of free radical production, lipid peroxidation, and AGEs formation. Furthermore, the expression and the antiapoptotic and antioxidant activity of the paraoxonase-2 (PON2) enzyme in intestinal cells has been investigated. (3) Results: Caco-2 cells treated with media supplied with high glucose (HG) (50 mM) showed, with respect to physiological glucose concentration (25 mM), an increase in ROS production, lipid peroxidation, and AGEs formation. Moreover, a lower PON2 expression and activity in HG-treated cells was related to activation of the apoptotic pathways. (4) Conclusions: Our results demonstrated that high glucose concentrations triggered glyco-oxidative stress in intestinal cells; the downregulation of PON2 could result in a higher oxidative stress and might contribute to intestinal dysfunction.

Biphasic Response of Endothelial Progenitor Cell Proliferation Induced by High Glucose and Its Relationship with Reactive Oxygen Species.

Abstract:In this study, the effect of high glucose (HG) on endothelial progenitor cell (EPC) proliferation and its relationship with cyclins and reactive oxygen species (ROS) were investigated. Mouse EPCs were isolated from bone marrow using a magnetic activated cell-sorting system and cultured in the presence or absence of HG (30 mmol/l). We found that in the early stage of incubation (3 days), HG promoted cell proliferation, and increased the expressions of cdk2 and cyclin E, while in the late stage of culture (7 days) it inhibited cell proliferation and decreased the expressions of cdk2, cyclin E, and proliferating cell nuclear antigen (PCNA). Moreover, on the third day after incubation, HG significantly inhibited the apoptosis of EPCs, while in the late stage it markedly activated caspase-3 and promoted apoptosis. ROS generation in cells and maleic dialdehyde level in medium were significantly increased in HG group on the seventh day, whereas the expressions of superoxide dismutase and glutathione levels decreased. Tempol, a membrane-permeable radical scavenger, significantly inhibited ROS production in EPCs and partially reversed the HG-mediated inhibition of EPCs proliferation on the seventh day. We hypothesize that in the HG environment, the biphasic response of EPC proliferation may be related to the generation of ROS, which causes modulation of cyclins and cell cycle effect.
GLP-1 Inhibits High-Glucose-Induced Oxidative Injury of Vascular Endothelial Cells

Abstract:The aim of this work was to evaluate the effects of glucagon-like peptide-1 (GLP-1) on high-glucose-induced oxidative stress and investigate the possible mechanisms underlying this process. We measured reactive oxygen species (ROS) production, cell apoptosis, the expression of NOX4 and its subunits, and p47phox translocation in human umbilical vein endothelial cells (HUVECs). An experimental type 2 diabetes model was induced using streptozotocin in male Sprague-Dawley rats. Fasting blood glucose (FBG), fasting insulin (FINS), total cholesterol (TC), triglycerides (TGs), and free fatty acid (FFA) were measured. Histomorphological analysis of the aorta was performed using hematoxylin-eosin staining. NOX4 and VCAM-1 expression in the aorta was measured. We found that high-glucose-induced ROS production and apoptosis were inhibited by GLP-1 treatment. High glucose caused upregulation of NOX4, p47phox, and Rac-1 and translocation of p47phox but had no effect on the cells pretreated with GLP-1. Furthermore, in the diabetic group, FBG, FINS, TG, TC, and FFA were increased, and NOX4 and VCAM-1 levels were also elevated. However, GLP-1 attenuated all these changes. GLP-1 ameliorated high-glucose-induced oxidative stress by inhibiting NOX4, p47phox, and Rac-1 expression and translocation of p47phox, suggesting its clinical usefulness in diabetic vascular complications.

Abstract:Uncoupling proteins (UCPs) are mitochondrial transporters present in the inner membrane of mitochondria. They belong to the family of anion mitochondrial carriers. UCPs could act as proton carriers activated by metabolites and create a shunt between complexes of the respiratory chain and ATP synthase. The increased leakiness of the mitochondrial inner membrane to protons may be to minimize superoxide production by limiting the maximum Delta mu(H+). The purpose of this study was to detect UCP expression in retinal capillary cells and their modification in high levels of glucose. The role of reactive oxygen species (ROS) of mitochondria and UCPs in pathogenesis of diabetic retimipathy was investigated. Bovine retinal capillary endothelial cells and pericytes were cultured with selective culture media, respectively. Passage cells were cultured in three different glucose concentrations (5, 23, 30 mM) until passage four. ROS changes in mitochondria of these cells in different glucose concentrations were detected with scanning laser confocal microscopy (SLCM). The mitochondria membrane potential (Delta psi), cell death rate and apoptosis rate were measured with flowing cytometry. UCP expression in retinal capillary cells was detected by immunocytochemistry. Expression and modification of MnSOD and uncoupling proteins (UCPs) in different concentrations of glucose were detected by means of semi-quantitative RT-PCR. ROS in mitochondria of both endothelial cells and pericytes increased as the glucose concentration of media increased. Alp and cell death rate of endothelial cells increased also. ROS was correlated to Delta psi and cell death rate positively in endothelial cells. No difference in Delta psi and cell death rate among different glucose levels was found in pericytes. Apoptosis rate of endothelial cells and pericytes in high glucose levels was higher than that in lower glucose levels. UCP1 and UCP2 were expressed in cultured retinal capillary cells whereas UCP3 was not. At high levels of glucose, expression of UCP1, UCP2 and MnSOD increased to accommodate ROS production compensatively. The compensative mechanism disappeared when glucose concentration was too high (30 mM). The results of this study showed that increasing mitochondrial ROS could be induced by high glucose concentration. Those proteins related to antioxidation mechanism, such as MnSOD and UCPs, could exert compensative action to a certain extent. Hydrogen Peroxide Regulates Glucose‐regulated Protein 78 Expression Via a Cyclooxygenase‐2 Dependent Mechanism

Abstract:This study was designed to investigate the effect of hydrogen peroxide on the expression of endoplasmic reticulum stress marker glucose‐regulated protein 78 (GRP78) in endothelial cells and reveals the possible role of cyclooxygenase in this effect. The porcine endothelial cell line was cultured in 1640 medium. Western blot and immunocytochemistry were used to detect the expression of GRP78. The caspase‐12 activity was analyzed with the immune fluorescence method. The results showed that after the endothelial cells were incubated with 250 μM of hydrogen peroxide for 12 h, apoptosis increased, which was antagonized by the cyclooxygenase‐2 inhibitor nimesulide or the nonselective cyclooxygenase inhibitor aspirin, but not by the cyclooxygenase‐1 inhibitor piroxicam. The expression of GRP78 was induced in endothelial cells after exposure to hydrogen peroxide for 12 h. The overexpression of GRP78 was inhibited by nimesulide and aspirin, but not by piroxicam. There are no significant differences in caspase‐12 activity among all groups. The present study provides evidence that hydrogen peroxide induced GRP78 overexpression in endothelial cells by a mechanism involving cyclooxygenase‐2‐dependent pathway. © 2010 Wiley Periodicals, Inc. Abstract:Ergothioneine (Egt), 2-mercapto-L-histidine betaine (ESH), is a dietary component acting as antioxidant and cytoprotectant. In vitro studies demonstrated that Egt, a powerful scavenger of hydroxyl radicals, superoxide anion, hypochlorous acid and peroxynitrite, protects vascular function against oxidative damages, thus preventing endothelial dysfunction. In order to delve the peculiar oxidative behavior of Egt, firstly identified in cell free-systems, experiments were designed to identify the Egt oxidation products when endothelial cells (EC) benefit of its protection against high-glucose (hGluc). HPLC-ESI-MS/MS analyses revealed a decrease in the intracellular GSH levels and an increase in the ophthalmic acid (OPH) levels during hGluc treatment. Interestingly, in the presence of Egt, the decrease of the GSH levels was lower than in cells treated with hGluc alone, and this effect was paralleled by lower OPH levels. Egt was also effective in reducing the cytotoxicity of H2O2 and paraquat (PQT), an inducer of superoxide anion production, showing a similar time-dependent pattern of GSH and OPH levels, although with peaks occurring at different times. Importantly, Egt oxidation generated not only hercynine (EH) but also the sulfonic acid derivative (ESO3H) whose amounts were dependent on the oxidative stress employed. Furthermore, cell-free experiments confirmed the formation of both EH and ESO3H when Egt was reacted with superoxide anion. In summary, these data, by identifying the EH and ESO3H formation in EC exposed to hGluc, highlight the cellular antioxidant properties of Egt, whose peculiar redox behavior makes it an attractive candidate for the prevention of oxidative stress-associated endothelial dysfunction during hyperglycemia.

Abstract:Diabetes-associated long-term hyperglycaemia leads to oxidative stress-mediated fibrosis in different tissues and organs. Endothelial-to-mesenchymal-transition (EndMT) appears to play a role in diabetes-associated fibrotic conditions. Here, we investigate whether EndMT is implicated in the diabetic retinopathy fibrotic process and evaluate the possibility that resveratrol could counteract EndMT by inhibiting high glucose (HG)-induced increases in ROS. Primary Human Retinal Endothelial Cells (HRECs) were either pre-treated for 24 h with 1 μM resveratrol or left untreated, then glucose (30 mM) was applied at 3-day intervals for 10 days. qRT-PCR and ELISA were used to detect mRNA or protein expression of endothelial markers (CD31, CDH5, vWF) or mesenchymal markers (VIM, αSMA and collagen I), respectively. Intracellular ROS levels were measured with carboxy-DCFDA, while NOX-associated ROS levels were evaluated using the NADPH-specific redox biosensor p47-roGFP. Treatment of HRECs with HG increased intracellular ROS levels and promoted phenotype shifting towards EndMT, evidenced by decreased expression of endothelial markers concomitant with increased expression of mesenchymal ones. HG-induced EndMT appears to be mediated by NADPH-associated ROS generation as pre-treatment of HRECs with resveratrol or the NADPH inhibitor, diphenyleneiodonium chloride (DPI), attenuated ROS production and EndMT transition, suggesting that the effect of resveratrol on HG-induced ROS occurs via down-regulation of NADPH oxidase. It is worth noting that resveratrol or Chelerythrine, a Protein kinase C (PKC) inhibitor, reduce ROS and EndMT in HG-exposed cells, suggesting that NADPH activation occurs via a PKC-dependent mechanism. Taken together, our results provide the basis for a resveratrol-based potential protective therapy to prevent diabetic-associated complications.

Abstract:OBJECTIVE Prolonged catabolic states in type 2 diabetes (T2D), exacerbated by excess substrate flux and hyperglycemia, can challenge metabolic flexibility and antioxidative capacity. We investigated cellular responses to glucose load after prolonged fasting in T2D. RESEARCH DESIGN AND METHODS Glucose-tolerant individuals (CON, n = 10), T2D individuals with (T2D+, n = 10) and without diabetes complications (T2D−, n = 10) underwent oral glucose tolerance test before and after a 5-day fasting-mimicking diet. Peripheral blood mononuclear cells’ (PBMC) resistance to ex vivo dicarbonyl methylglyoxal (MG) exposure after glucose load was assessed. Markers of dicarbonyl detoxification, oxidative stress, and mitochondrial biogenesis were analyzed by quantitative PCR, with mitochondrial complex protein expression assessed by western blotting. RESULTS T2D+ exhibited decreased PBMC resistance against MG, while T2D− resistance remained unchanged, and CON improved postglucose load and fasting (−19.0% vs.−1.7% vs. 12.6%; all P = 0.017). T2D+ showed increased expression in dicarbonyl detoxification (mRNA glyoxalase-1, all P = 0.039), oxidative stress (mRNA glutathione-disulfide-reductase, all P = 0.006), and mitochondrial complex V protein (all P = 0.004) compared with T2D− and CON postglucose load and fasting. Citrate synthase activity remained unchanged, indicating no change in mitochondrial number. Mitochondrial biogenesis increased in T2D− compared with CON postglucose load and fasting (mRNA HspA9, P = 0.032). T2D−, compared with CON, exhibited increased oxidative stress postfasting, but not postglucose load, with increased mRNA expression in antioxidant defenses (mRNA forkhead box O4, P = 0.036, and glutathione-peroxidase-2, P = 0.034), and compared with T2D+ (glutathione-peroxidase-2, P = 0.04). CONCLUSIONS These findings suggest increased susceptibility to glucose-induced oxidative stress in individuals with diabetes complications after prolonged fasting and might help in diet interventions for diabetes management.

Abstract:PurposeMitochondrial reactive oxygen species (ROS) overproduction in capillary endothelial cells is a prerequisite for the development of diabetic nephropathy. Inhibition of xanthine oxidase, another ROS generator, ameliorates experimental diabetic nephropathy. To test the hypothesis that the initial high glucose-induced ROS production by the mitochondria activates xanthine oxidase, which afterward remains as the major source of ROS, we cultured primary human glomerular endothelial cells (GEnC) under normal or high-glucose conditions, with or without the xanthine oxidase inhibitor allopurinol.MethodsROS generation and nitric oxide synthase (NOS) activity were assessed by chemiluminescence or colorimetrically. Levels of intercellular adhesion molecule 1 (ICAM-1), p53 and phosphorylated p53 (p-p53) were assessed by western blotting.ResultsAllopurinol prevented high glucose-induced ROS generation indicating that xanthine oxidase is the major source of ROS. Allopurinol protected GEnC from endothelial dysfunction since it prevented the high glucose-induced decrease in NOS activity and increase in ICAM-1 expression. Allopurinol reduced p53 and p-p53 levels induced by high glucose suggesting an axis of xanthine oxidase-derived ROS, DNA damage, p53 stabilization and endothelial dysfunction that may contribute to the pathogenesis of diabetic nephropathy.ConclusionsAllopurinol protects GEnC from high glucose-induced ROS generation, p53 overexpression and endothelial dysfunction. These data provide a pathogenetic mechanism that supports the results of experimental and clinical studies about the beneficial effect of xanthine oxidase inhibitors on the development of diabetic nephropathy.
