Abstract:Halofuginone (stenorol) has been used as an effective anticoccidial reagent for decades but very little is known about its mode of action. In this study, chickens were inoculated with Eimeria tenella oocysts on 14-day-old and medicated with halofuginone at days 0, 1, 2, 3, 4, 5 and 6 post inoculations (groups 0, 1, 2, 3, 4, 5 and 6, respectively). Chickens in group 7 were taken as challenge-unmedicated control and in group 8 unchallenged-unmedicated control. The survival rate, body weight gains (BWG), oocysts production, cecal scores, bloody diarrhea and histological examinations were analyzed to evaluate the anticoccidial efficacy of halofuginone and to initially elucidate its mechanisms. Results showed that halofuginone which acted as a coccidiostatic can significantly enhance the BWG, and decrease both the oocyst shedding and cecal destruction caused by E. tenella infection. The histological slide examination noted that halofuginone was effective when provided 0-2 days post inoculation but only partially effective when applied 3-7 days post infection. The second-generation schizonts treated with halofuginone appeared vacuolated and degenerated. It is concluded that halofuginone can inhibit the parasite's invasion of host cecal hypothetical cell at the early stages of life cycle and later disturb the parasite's development by vacuolation of the schizonts. The resulting abnormal schizonts could not divide into schizoites and were eventually eliminated by the host's immune response.

Abstract:In this study, four series of dihydroartemisinin derivatives were designed, synthesized, and evaluated for anti-toxoplasma gondii activity, and calculated the selectivity index (SI). It was the higher the SI, the better the effect of this compound against Toxoplasma gondii. Our goal was to filter out compounds that were bigger SI than the lead compound. The compound with the highest SI was selected for the anti-toxoplasmosis test in mice in vivo. Among the synthesized compounds, the (3R,5aS,6R,8aS, 9R,12R,12aR)-3,6,9-trimethyl-decahydro-12H-3,12-epoxy [1,2]di-oxepino [4,3 -i]isochromen-10-yl (te-rt-butoxycarbonye-L-alaninate (A2) exhibited the most potent anti-T. gondii activity and low cytotoxicity (SI: 6.44), yielding better results than the lead compound DHA (SI: 1.00) and the clinically used positive-control drug spiramycin (SI: 0.72) in vitro. Furthermore, compound A2 had better growth inhibitory effects on T. gondii in vivo than spiramycin did and significantly reduced the number of tachyzoites in the peritoneal cavity of mice (P < 0.01). The evaluation of the data generated in the T. gondii mouse infection model indicates that compound A2 treatment was a good inhibitor of T. gondii in vivo and that it was effective in relieving the liver damage induced by T. gondii. In addition, the results of a docking study revealed that A2 could become a better T. gondii calcium-dependent protein kinasel (TgCDPK1) inhibitor. For this reason, compound A2 has potential as an anti-parasitic drug. Further studies are required to elucidate the mechanism of the action of compound A2, as well as to develop drug delivery systems for patients.

Abstract:A liquid and a powder made from a herbal complex consisting of Uncariae Ramulus cum Uncis, Agrimoniae Herba, Sanguisorbae Radix, Eclipta Prostrate Herba, Pulsatillae Radix, Sophorae Flavescentis Radix, Rehmanniae Radix and Glycyrrhizae Radix were studied for their anticoccidial activities in chickens. Chickens were administered with herbal liquid, powder, diclazuril or without medication during the study and challenged with oocysts of Eimeria tenella. Results indicated that the birds medicated showed less bloody faeces than those without medication. The intestinal lesion was mild in the chicks medicated with herbal liquid without significantly different lesion score when compared with uninfected chicks. The birds with medication had significantly higher body weight gains than birds without medication. Therefore, the herbal complex used in this study was effective against E. tenella infection in chickens.

Abstract:Cancers arise from stem cells in adult tissues and the cells that make up a cancer reflect the same stem cell → progeny → differentiation progression observed in normal tissues. All adult tissues are made up of lineages of cells consisting of tissue stem cells and their progeny (transit-amplifying cells and terminally differentiated cells); the number of new cells produced in normal tissue lineages roughly equals the number of old cells that die. Cancers result from maturation arrest of this process, resulting in continued proliferation of cells and a failure to differentiate and die. The biological behavior, morphological appearance, and clinical course of a cancer depend on the stage of maturation at which the genetic lesion is activated. This review makes a comparison of cancer cells to embryonic stem cells and to adult tis sue stem cells while addressing two basic questions: (1) Where do cancers come from?, and (2) How do cancers grow? The answers to these questions are critical to the development of approaches to the detection, prevention, and treatment of cancer.
Efficacy of Artemisia annua against Coccidiosis in Broiler Chickens: A Field Trial

Abstract:(1) Background: Various studies on artemisinin and its derivatives have shown that Artemisia annua may be of therapeutic interest for different diseases, including chicken coccidiosis. This study aimed to evaluate the effects of Artemisia annua on farm-reared broiler chickens by analyzing both the anticoccidial efficacy and its effect on the intestinal microbiota of poultry. (2) Methods: The experiment was performed within three houses on a broiler chicken farm located in Romania. House 1 was the experimental group and received a diet with an addition of A. annua. Houses 2 and 4 were the control groups and received anticoccidials. The prophylactic efficacy of A. annua against coccidiosis was evaluated by recording the weight gain, feed conversion rate, number of oocysts per gram of feces, lesion score, and mortality rate. (3) Results: The chickens fed with A. annua showed a decreasing trend in the number of oocysts per gram of faeces, and their lesion score was 80% lower than in the control group. The weight gains of the chickens treated with A. annua was lower, whilst the feed conversion rate was better than in controls. (4) Conclusions: Artemisia annua showed promising results in the prophylaxis of coccidiosis. Overall, the broiler chickens that received A. annua presented promising zootechnical performances and medical data related to coccidiosis and gut microbiota.

Abstract:Coccidiosis, an acute epidemic intestinal disease of poultry, is caused by the parasitic protozoan genus Eimeria, with Eimeria tenella being the most pathogenic spp. Novel approaches are required to address the limitations of current treatments for this disease. We investigated the effects of eight plant extracts and essential oils and their mixture on Eimeria tenella as potential treatments for coccidial infection. The anticoccidial effects of non-toxic concentrations of Punica granatum L. (0.005 mg/mL), Plantago asiatica L. (0.780 mg/mL), Bidens pilosa L. (0.390 mg/mL), Acalypha australis L. (0.390 mg/mL), Pteris multifida Poir (0.050 mg/mL), and Portulaca oleracea L. sp. Pl. (0.050 mg/mL) extracts; Artemisia argyi Levl. et Vant. (0.010 μL/mL) and Camellia sinensis (L.) O. Ktze (0.050 μL/mL) essential oils; and their mixture (0.500 mL/mL) on Eimeria tenella were determined using cell viability assays, flow cytometry, and in vivo studies. The eight plant extracts and essential oils and their mixture inhibited Eimeria tenella sporozoites from invading chicken embryo fibroblast cells in vitro. The extract and essential oil mixture improved the feed conversion ratio and body weight gain, reduced fecal oocyst excretion, substantially reduced the mortality of Eimeria tenella-infected chickens, and reduced Eimeria tenella-induced cecal damage in vivo. The results suggest that the extract and essential oil mixtures inhibit Eimeria tenella invasion both in vitro and in vivo, demonstrating their potential as anticoccidial agents.

Abstract:Coccidiosis is an important cause of digestive system disorders in birds, especially the poultry. The aim of this study was to evaluate the in-vitro anticoccidial effects of the different fractions of Chromolaena odorata extracts on sporulated Eimeria tenella oocysts. Sporulation inhibition bioassay was used to evaluate the activity of crude methanol fraction of Chromolaena odorata extracts; n-hexane, ethyl acetate, and methanol fractions of Chromolaena odorata on sporulation of Eimeria tenella, which was used to cause coccidiosis. in the birds following oral inoculation of experimental broiler birds with oocysts. Post-mortem examination of dead birds showed lesions consistent with Eimeria tenella. The fractions of the Chromolaena odorata were examined for the oocysticidal activities. Amprolium an anticoccidial drug was used as control. The fractions were used in 4 doses: 0 mg/ml; 0.1 mg/ml; 0.2 mg/ml and 0.4 mg/ml. The control drug was used in corresponding doses. The highest oocysticidal activity of tested plant extracts and fractions was recorded. The n-hexane fraction of Chromolaena odorata gave the highest oocysticidal activity. This oocysticidal activity was noticed at lower concentrations but activity was found to be dose-dependent. The highest oocysticidal activity for the n-hexane fraction of Chromolaena odorata was at 65.67±2.08%. Amprolium showed no oocysticidal activity at all concentrations. However, other fractions of the Chromolaena odorata showed different levels of oocysticidal activities. The methanol fractions were secondary to the n-hexane while the ethyl acetate was the least. In conclusion, this study provides the basis for using Chromolaena odorata, especially the n-hexane fraction as preventive agents against coccidiosis and suggests their use as a feed additive in the control of coccidiosis.

Abstract:This work investigated the preventive effect of Artemisia annua L. dried leaves supplied as a botanical coccidiostat to two broiler genotypes reared in a Danish free-range system in a factorial experiment (two genotypes and ± supplement of dried A. annua leaves). The genotypes White Bresse L40, a pure slow-growing line, and Kosmos 8 Ross, a hybrid genotype with medium growing characteristics, were used. Broilers were raised indoor until 29-days-old and kept free of parasites. Twelve groups of 30 randomly selected broilers were placed in the range forming three replicates for each treatment combination. The paddocks were cultivated with a mix of grass and clover. A separate group of broilers was naturally infected with Eimeria spp. oocysts and five animals nominated as "seeders" were introduced to the above mentioned 12 groups, 10 days after its formation, with each group consisting of 35 animals per plot. This infection strategy was meant to imitate the transmission pathway observed at farm level. Ten individual birds from each of the 12 groups, in total 120 animals of mixed sex, were monitored twice weekly for 30 days for oocysts excretion. PCR of pooled faecal samples, oocyst morphology and localization upon necropsy were used to identify the Eimeria species involved in the infection. In general, broilers from both genotypes in the range coped well with a coccidia infection caused by Eimeria acervulina and Eimeria maxima as no clinical symptoms, or deaths, were reported during the experiment. In general, broilers supplemented with A. annua dried leaves showed a significantly (p<0.05) reduced number of excreted oocysts during the infection with no interaction to genotype. Females generally had a significantly higher shedding of oocysts than males (p<0.05). The overall body weight gain and the daily weight gain when infection was subdued showed a three-way interaction among genotype, sex and treatment - accounted mainly for the fact that Kosmos females responded positively to the Artemisia treatment while Kosmos males responded negatively, and only minor differences were found between sexes for the White Bresse genotype. In conclusion, supply of A. annua dried leaves as a botanical coccidiostat significantly reduced oocyst output in free ranged broilers and thus may form part of a strategy to prevent commercial losses.

Abstract:This study aims to assess the ability of essential oils (EOs) to destroy Eimeria oocyst in vitro using microscopic counting and 273 nm absorbing material release. A screening for the ability of ten EOs to destroy Eimeria oocyst was carried out in liquid medium. Out of these ten, artemisia, tea tree, thyme and clove EOs were identified as being the most effective. The treatment of Eimeria oocyst with these EOs leads to their lysis as shown by the release of substances absorbing at 273 nm. These results were obtained after approximately three hours contact. Four EOs were proven to destroy Eimeria oocysts in a few hours at low concentration. This destructive effect is a consequence of their lysis. This work is a preliminary contribution aiming to develop a new generation of natural efficient agents for destroying Eimeria oocyst to fight coccidiosis in broiler chicken.
Ponazuril: Clinical efficacy, ultrastructure, and histopathology studies of in vivo anticoccidial action against Eimeria tenella

Abstract:Ponazuril, a novel antiprotozoal drug in the class of triazine, has shown a promising application on apicomplexan infections in poultry and livestock. However, the effect and mechanism of action of ponazuril against Eimeria tenella (E. tenella) are unclear. The efficacy against E. tenella was initially studied by administering different doses of ponazuril in drinking water. The treated stage and site of ponazuril on E. tenella were observed through ultrastructural and histopathological analyses. Chicks were orally treated with a dose of 15 mg/kg body weight of ponazuril at different endogenous stages of E. tenella post-infection. According to the clinical study, the values of anticoccidial indices (ACI) were 157.0, 162.3, 196.9, 194.5, and 190.9, respectively, when the ponazuril was administered in drinking water at doses of 5, 10, 20, 40, and 50 mg/L for two consecutive days after infection. Among them, the 20 mg/L ponazuril group showed the best anticoccidial effect, which was superior to that of the toltrazuril treatment group, with an ACI value of 191.7. Histological analysis indicated that ponazuril effectively relieved cecal lesions, and decreased the number of merozoites. Transmission electron micrographs (TEM) observed that merozoites became irregular in shape, and some apparent protrusions of the outer membrane were presented especially the second-generation merozoites. Additionally, abnormalities in the development of WFBI and WFBII in the macrogametocyte were observed, which may affect the formation of the ovule wall. Moreover, merozoites in the treated group showed uneven and marginalized chromatin and mitochondrial swelling. These results suggested ponazuril is a potential anticoccidial drug, providing information on the mechanism of anticoccidial effects.
In Vitro: The Effects of the Anticoccidial Activities of Calotropis procera Leaf Extracts on Eimeria stiedae Oocysts Isolated from Rabbits

Abstract:Hepatic coccidiosis is an infectious and mortal disease that causes global economic losses in rabbits. The research aimed to assess the efficacy of Calotropis procure leaf extracts on the inhibition of Eimeria stiedae oocysts and to determine the optimal dosage for suppressing the parasite's infective phase. In this experiment, oocyst samples per milliliter were tested, and 6-well plates (2 mL) of 2.5% potassium dichromate solution containing 102 non-sporulated oocysts on Calotropis procera leaf extracts were exposed after 24, 48, 72, and 96 h, and the treatments were as follows: a nontreated control, 25%, 50%, 100%, and 150% of C. procera for oocyst activities. In addition, amprolium was utilized as a reference drug. The Calotropis procera was analyzed by GC-Mass, and results showed that the botanical extract contained 9 chemical components that were able to inhibit the oocysts of E. stiedae at 100% and 150% concentrations by about 78% and 93%, respectively. In general, an increase in the incubation period and a greater dose resulted in a decrease in the inhibition rate. The results showed that C. procera has an effective ability, inhibitory potential, and protective effect on the coccidian oocyst sporulation of E. stiedae. It can be used in the disinfection and sterilization of poultry and rabbit houses to get rid of Eimeria oocysts.

Abstract:Coccidiosis is a widespread intestinal disease of poultry caused by a parasite of the genus Eimeria. Eimeria tenella, is one of the most virulent species that specifically colonizes the caeca, an organ which harbors a rich and complex microbiota. Our objective was to study the impact of the intestinal microbiota on parasite infection and development using an original model of germ-free broilers. We observed that germ-free chickens presented significantly much lower load of oocysts in caecal contents than conventional chickens. This decrease in parasite load was measurable in caecal tissue by RT-qPCR at early time points. Histological analysis revealed the presence of much less first (day 2pi) and second generation schizonts (day 3.5pi) in germ-free chickens than conventional chickens. Indeed, at day 3.5pi, second generation schizonts were respectively immature only in germ-free chickens suggesting a lengthening of the asexual phase of the parasite in the absence of microbiota. Accordingly to the consequence of this lengthening, a delay in specific gamete gene expressions, and a reduction of gamete detection by histological analysis in caeca of germ-free chickens were observed. These differences in parasite load might result from an initial reduction of the excystation efficiency of the parasite in the gut of germ-free chickens. However, as bile salts involved in the excystation step led to an even higher excystation efficiency in germ-free compared to conventional chickens, this result could not explain the difference in parasite load. Interestingly, when we shunted the excystation step in vivo by infecting chickens with sporozoites using the cloacal route of inoculation, parasite invasion was similar in germ-free and in conventional chickens but still resulted in significantly lower parasite load in germ-free chickens at day 7pi. Overall, these data highlighted that the absence of intestinal microbiota alters E. tenella replication. Strategies to modulate the microbiota and/or its metabolites could therefore be an alternative approach to limit the negative impact of coccidiosis in poultry.

Abstract:A second-generation Faraday-strip array detector has been coupled to an inductively coupled plasma Mattauch-Herzog geometry mass spectrograph, thereby offering simultaneous acquisition of a range of mass-to-charge ratios. The second-generation device incorporates narrower, more closely spaced collectors than the earlier system. Furthermore, the new camera can acquire signal on all collectors at a frequency greater than 2 kHz and has the ability to independently adjust the gain level of each collector. Each collector can also be reset independently. With these improvements, limits of detection in the hundreds of picograms per liter for metals in solution have been obtained. Some additional features, such as a broader linear dynamic range (over 7 orders of magnitude), greater resolving power (up to 600), and improved isotope ratio accuracy were attained. In addition, isotope ratio precision as low as 0.018% RSD was achieved.

Abstract:Abstract The aim of this study was to investigate, under conditions similar to commercial broiler production, the effect of the herbal extract blend (HE) at a quantity of 1 g per kg feed (200 mg of each herbal extract, Allium sativum, Salvia officinalis, Echinacea purpurea, Thymus vulgaris and Origanum vulgare), used individually or in combination with mannan oligosaccharide (MOS; 1 g per kg feed) or chitosan (3 ml containing 2% deacetylated chitin per kg feed) on the performance parameters of broiler chickens, the results of the slaughter analysis, litter moisture and the number of oocysts excreted in feces. The experiment was conducted on 4,500 broiler chickens of both sexes kept in straw-bedded pens. Chickens were randomly assigned to 5 experimental treatments with 5 replicates (pens) of 180 birds. The experimental design included negative and positive (diclazuril, 1 mg per kg feed) control groups. The examined herbal extract blend used individually during natural exposure to the coccidia improved, compared to the negative control diet, the performance parameters to a greater extent than coccidiostat, lowered the litter moisture content and reduced the oocyst output. Combined dietary supplementation with a herbal extract blend of chitosan or mannan oligosaccharide did not result in further improvement.

Abstract:Ionophorous antibiotics have been popularly used in the treatment of avian coccidiosis. Tissue residue of these antibiotics may be found in poultry, we have sought safe alternative anticoccidial herbal materials for the control of avian coccidiosis. Efficacy of extracts from 15 different herbs, including Bupleurum chinese DC, Sophora flavescens Aiton, and Artemisia annua Linne was tested against Eimeria tenella. One-day-old broiler chicks were infected with a USDA reference book of E. tenella, and administered various herbal extracts. Survival rates, lesion scores, body weight gains, bloody diarrhea, and oocysts excretions were investigated at the first and the second week after infection. Bloody diarrhea in the S. flavescens and Sinomenium acutum treated groups was milder than that of the other infected groups. Survival rates in the groups treated with Ulmus macrocarpa (100%), Pulsatilla koreana, Torilis japonica, Artemisia asiatica and S. flavescens (90%) were higher than that of the infected control group (70%). Lesion scores in the groups treated with U. macrocarpa (1.40+/-1.14) or Pulsatilla koreana (1.60+/-1.82) were significantly lower than those of the infected control group (3.00+/-1.10). During the first week after infection, the weight gains in the groups treated with Quisqualis indica (232.9+/-43.5 g), S. flavescens (214.4+/-46.1 g) and S. acutum (211.3+/-29.4 g) were significantly higher than the infected control group (172.4+/-17.6 g). In conclusion, the data of the survival rates, bloody diarrhea symptoms, lesion scores, body weight gains and oocyst excretions indicate that the extract of S. flavescens was the most effective. P. koreana, S. acutum, U. macrocarpa and Q. indica were also effective. Further research on the above herbal materials will be carried out by the authors by chemical analysis of the extracts.

Antioxidant properties of D-limonene and its nanoemulsion form enhance its anticoccidial efficiency in experimentally infected broilers with Eimeria tenella: an in vitro and in vivo study

Abstract:The excessive use of conventional medications to treat coccidiosis has led to concerns regarding drug residues in tissues and the emergence of multidrug resistance. Essential oils with anti-inflammatory and antioxidant activities may also have anticoccidial effects. The present study investigated the efficacy of D-limonene and its nanoemulsion form against Eimeria tenella in chickens. An in vitro study was conducted to evaluate the sporulation inhibitory effects of D-limonene on Eimeria tenella oocysts. Five D-limonene concentrations (0.625, 1.25, 2.5, 5, and 10% v/v) were tested alongside positive (10% formalin) and negative (2.5% potassium dichromate) controls. Each ELISA plate well was inoculated with 1200 unsporulated oocysts and incubated at 30 °C for 24, 48, and 72 h. Subsequently, samples were microscopically examined to assess sporulation inhibition and calculate the percentage of sporulated oocysts. For the in vivo study, 125 eight-day-old broiler chicks were divided into five groups of 25 birds each. The control negative group remained uninfected and untreated. The control positive group was challenged with 5 × 104 sporulated Eimeria tenella oocysts. The diclazuril group received 0.2 mg/kg diclazuril in their diet two days prior to, and until 10 days post infection. The D-limonene (DL) and D-limonene nanoemulsion (DLN) groups were challenged with 5 × 104 sporulated E. tenella oocysts at 18 days of age and administered 150 mg/L of their respective treatments in drinking water from day eight until the end of the experiment. Results from the in vitro study demonstrated that D-limonene suppressed oocyst sporulation by 50.83% at its highest concentration of 10%. In the in vivo study, both DL and DLN treated groups exhibited a significant reduction in oocyst output per gram of feces (OPG), along with increased body weight and decreased parasite stages in the cecal tissue. Furthermore, these treatments were associated with elevated levels of antioxidant enzymes such as glutathione peroxidase (GPX), catalase (CAT), and superoxide dismutase (SOD), accompanied by a decrease in malondialdehyde (MDA) and nitric oxide (NO) levels. Particularly, DLN treatment remarkably increased the number of goblet cells. In conclusion, D-limonene and its nanoemulsion represent promising alternatives for managing coccidiosis in poultry. They not only effectively control parasites but also promote intestinal health and boost antioxidant defenses.
Anticoccidial effects of herbal extracts on Eimeria tenella infection in broiler chickens: in vitro and in vivo study

Abstract:Safe alternative anticoccidial drug to chemical feed additives are herbal extracts, because they don’t results to tissue residue and drug resistance. In order to evaluate the effects of herbal extracts to control avian coccidiosis, 180 one-day-old broiler chickens were randomly divided into nine equal groups, as follows: (1) Biarum bovei (2) Nectaroscordum tripedale( 3) Dorema aucheri (4) Cichorium intybus (5) Prangos ferulaceae (6) diclazuril (7) Artemisia absinthium (8) infected control (9) uninfected control (each contains two groups). Administration of herbal extracts and supplementation of diclazuril was began 2 days before challenge and lasted for the duration of the experiment. The chicks of all the groups except uninfected control group were inoculated orally with sporulated oocysts (3 × 103 oocysts of Eimeria tenella) on the day 22 of age. The criteria employed were: body weight, feed conversion ratio, blood in feces, survival rate, lesion scoring, number of oocyst output per gram feces and histopathological changes. For histopathological evaluation, on day 12 post inoculation three birds from each group were randomly selected and humanly sacrificed. N. tripedale and diclazuril revealed better results in terms of growth performance, lesion score, extent of bloody diarrhea and oocyst count as compared to other herbal extracts. The increase in the severity of lesions was observed in groups of D. aucheri, A. absinthium, B. bovei, P. ferulaceae, C. intybus, diclazuril and N. tripedale, respectively. In conclusion, the current study showed that herbal extracts were effective in control of coccidiosis caused by the E. tenella infection.
Anti-Eimeria tenella activity of Ethanamizuril in vitro and in vivo

Abstract:The prevalence of chicken coccidiosis in the poultry industry is a significant concern, further exacerbated by the emergence of drug-resistant coccidia resulting from the indiscriminate use of medications. Ethanamizuril, a novel triazine anti-coccidial compound, has been used to combat drug resistance. Currently, it is known that Ethanamizuril acts on the second-generation merozoites and early gametogenesis stages of Eimeria. Limited information exists regarding its impact on the early merozoites and exogenous stage of Eimeria. In the present study, the anti-coccidial properties of Ethanamizuril were evaluated both in vitro and in vivo. The in vitro experiments demonstrated that Ethanamizuril effectively inhibits the sporulation of E. tenella oocysts in a dose-dependent manner and significantly reduces the sporozoite excystation rate. Furthermore, in vivo tests revealed that treatment with 10 mg/L Ethanamizuril in drinking water significantly decreased the copy number of first-generation and secondary-generation merozoites in the chicken cecum, indicating that it can inhibit the development of whole schizonts development. Moreover, treatment with Ethanamizuril demonstrated excellent protective efficacy with an anti-coccidial index (ACI) of 180, which was manifested through higher body weight gains, lighter cecal lesion, lower fecal oocyst shedding score and reduced liver index. Collectively, this study suggests that Ethanamizuril effectively treats E. tenella infection by inhibiting both endogenous and exogenous stages development.
In vivo and in vitro anticoccidial efficacy of Astragalus membranaceus against Eimeria papillata infection

Abstract:Coccidiosis is a parasitic disease of wild and domestic animals caused by Eimeria spp. Currently, several drugs are available for the control of this disease but resistance has been confirmed for all them. There is an urgent need, therefore, for the identification of new compounds as alternative treatments to control coccidiosis. Astragalus membranaceus proven to have anti-inflammatory, anti-oxidant, immunomodulating and anticancer activities. The present study was therefore undertaken to assess the in vivo and the in vitro anticoccidial activity of Astragalus membranaceus root (AMR). Mice were divided into five groups, with the first left non-infected and the second, third, fourth and fifth groups being infected with 1×103 sporulated oocysts of E. papillata. The third, fourth and fifth groups also received, respectively, an oral dose of 10, 25 and 50 mg/kg AMR suspended in physiological saline daily for five consecutive days. 50 mg/kg, was the most effective dose, inducing a significant reduction in the number of oocysts output in mice faeces (by about 57%), accompanied by a significant decrease in the number of parasitic stages in jejunal sections. Moreover, the treatment with AMR increased the number of goblet cells and upregulated the expression of its specific gene, MUC2. In addition, our study proved that AMR reduced oxidative damage since levels of TBARS decreased (indicating reduced lipid peroxidation), levels of glutathione (GSH) and glutathione peroxidase (GPX) increased, and the mRNA level of iNOS was downregulated. Also, AMR treatment revealed anti-apoptotic activity as it was able to regulate the gene expression of Bcl-2 in the jejunum of E. papillata infected mice. Finally, the in vitro study revealed that AMR significantly inhabited the oocyst sporulation in a dose dependent manner. Overall, therefore, our results indicate that AMR exhibits significant in vivo and in vitro anticoccidial effects.

Effects of Six Natural Compounds and Their Derivatives on the Control of Coccidiosis in Chickens

Abstract:Chicken coccidiosis costs the poultry industry over GBP 10 billion per year. The main method of preventing and controlling coccidiosis in chickens continues to be the use of drugs. Unfortunately, the prevalence of drug resistance in the field reduces or even eliminates the effectiveness of drugs, and drug residues in the food supply chain can also can be harmful to humans. Therefore, safe and effective anticoccidial drugs are urgently needed. Natural products have many advantages such as being safe, effective and inexpensive and are a sustainable way to control coccidiosis. In this study, the anticoccidial effects of six natural compounds were tested by Eimeria tenella infection. Oocyst production, cecum lesion, body weight gain, feed conversion ratio, and intestinal microbiota were measured. The results showed that nerolidol had a moderate effect on maintaining both body weight gain and feed conversion ratio. Silymarin and dihydroartemisinin showed significant anticoccidial effects by reducing total oocyst output. Dihydroartemisinin also significantly reduced the cecum lesion caused by Eimeria infection, but this compound may be toxic to the host at such informed doses because it decreases growth and survival rates. In addition, both silymarin and dihydroartemisinin partly restored the microbiota after challenge. This indicates that silymarin, dihydroartemisinin, and nerolidol are effective in the control of chicken coccidiosis. Our data provide basic knowledge about the anticoccidial effects of such natural compounds/derivates.
