Abstract:SUMMARY In this paper, a novel contour-tracking control method of an unknown planar object by lateral force regulation for wheeled mobile robot navigation is presented. The robot is required to follow the contour of an unknown object toward the goal position. Based on mobile robot dynamic equations, a force-control algorithm is proposed to maintain constant contact with a planar object. Measured contact force from an object is used not only to regulate a contact force in a lateral direction, but also to control the orientation angle of the robot to avoid collision with an object. Simulation and experiment of contour-tracking tasks of a wheeled mobile robot are conducted. Experimental results show that the contact force is well-regulated, and the robot arrives at the goal position successfully.

Abstract:Employing double-isotope derivative techniques, noradrenaline and adrenaline have been determined in plasma and in urine and dopamine in urine in 21 patients with essential hypertension as well as in 32 controls. Plasma noradrenaline rose with age in both groups of subjects. No differences were observed in plasma noradrenaline and plasma adrenaline in the resting supine position and in urinary excretion of noradrenaline and dopamine in hypertensive patients as compared to control subjects. Urinary excretion of adrenaline was somewhat lower in the hypertensives than in the controls. Treatment with alprenolol, a beta-adrenergic blocking agent, did not influence noradrenaline and adrenaline in plasma in the basal state or the urinary excretion of the three catecholamines. The combined treatment with alprenolol and hydralazine was followed by a significant rise in plasma noradrenaline. It is concluded that the adrenergic activity evaluated by circulating catecholamines is normal in most patients with essential hypertension.

Abstract:Although the primary photochemical events in the facultative photoheterotrophic purple bacterium Rhodobacter sphaeroides are now well understood, comparatively little is known about how their photosynthetic apparatus is assembled. Here we present a proteomic analysis of the intracytoplasmic membrane (ICM) assembly process during adaptation to lowered light intensity, in which the size of the photosynthetic units is greatly expanded by addition of the light-harvesting 2 (LH2) peripheral antenna complex. When the isolated ICM-derived chromatophore vesicles were subjected to clear native gel electrophoresis (CNE), four pigmented bands appeared; the top and bottom bands contained the reaction center - light-harvesting 1 (RC-LH1) core complex and LH2 peripheral antenna, respectively, while the two bands of intermediate migration contained associations of the LH2 and core complexes. Proteomic analysis revealed a large array of other proteins associated with the CNE gel bands - in particular, several F(1)F(O)-ATP synthase subunits gave unexpectedly high spectral counts, given the inability to detect this coupling factor, as well as the more abundant cytochrome bc (1) complex, by atomic force microscopy. Significant levels of general membrane assembly factors were also found, as well as numerous proteins of unknown function including high counts for RSP6124 that were correlated with LH2 levels. When combined with further AFM and spectroscopic studies, these proteomic approaches are expected to provide a much-improved understanding of the overall assembly process.

Abstract:We present a simple but accurate preselection method based on Schwarz integral estimates to determine the significant elements of the exact exchange matrix before its evaluation, thus providing an asymptotical linear-scaling behavior for non-metallic systems. Our screening procedure proves to be highly suitable for exchange matrix calculations on massively parallel computing architectures, such as graphical processing units, for which we present a first linear-scaling exchange matrix evaluation algorithm.

Abstract:In cyanobacteria the outer membrane, plasma membrane and thylakoid membrane represent three structurally and functionally distinct membranes. Common themes are emerging from studies of thylakoid membranes which show that the major functional components, including Photosystem I, Photosystem II, cytochrome b6f, NADH dehydrogenase, cytochrome oxidase, and ATP synthase, exist as multisubunit complexes within the membrane. These integral membrane complexes traverse the 4–5 nm lipid bilayer and extend into the stromal and lumenal spaces-thus virtually doubling the membrane thickness. In situ it appears that cytochrome b6f, cytochrome oxidase, and Photosystem II preferentially occur as dimers and Photosystem I as trimers. Determination of the density of the supramolecular complexes per thylakoid area and the spatial relationships among them can provide useful insight into membrane functional. Calculations suggest that at least 50% of the intramembrane area is occupied by photosystems I and II, cytochrome b6f, and CF0 of ATP synthase alone. Topographic mapping of the membranes can be pursued with gold-antibody labeling with a resolution of approximately 15–20 nm. Studies on complexes of the outer membrane, the plasma membrane, and the composition and influence of the peptidoglycan layer are still scarce and should be extended. The uniqueness of cyanobacterial membrane structure continues to emerge. Differences in the structure of the outer wall layers have been noted when compared to other eubacteria; however, essential thylakoid features are also found in prochlorophytes and eucaryotic algae oxygenic plants. These features emphasize that cyanobacteria and prochlorophytes should be considered a distinctive grouping of the eubacteria.
The assembly and organisation of photosynthetic membranes in Rhodobacter sphaeroides

Abstract:Recent AFM data demonstrate that mature photosynthetic membranes of R. sphaeroides are composed of rows of dimeric RC-LH1-PufX complexes with some LH2 complexes 'sandwiched' between these rows of core complexes, and others in discrete LH2-only domains which might form the light-responsive complement of the LH2 antenna. The present work applies membrane fractionation, radiolabelling and LDS-PAGE techniques to investigate the response of R. sphaeroides to lowered light intensity. The kinetics underlying this adaptation to low light conditions were revealed by radiolabelling with the bacteriochlorophyll (bchl) biosynthetic precursor, delta-aminolevulinate, which allowed us to measure only the bchls synthesised after the light intensity shift. We show that (1) the increase in LH2 antenna size is mainly restricted to the mature ICM membrane fraction, and the antenna composition of the precursor upper pigmented band (UPB) membrane remains constant, (2) the precursor UPB membrane is enriched in bchl synthase, the terminal enzyme of the bchl biosynthetic pathway, and (3) the LH2 and the complexes of intermediate migration in LDS-PAGE exhibit completely different labelling kinetics. Thus, new photosynthetic complexes, mainly LH2, are synthesised and assembled at the membrane initiation UPB sites, where the LH2 rings pack between the rows of dimeric cores fostering new LH2-LH1 interactions. Mature membranes also assemble new LH2 rings, but in this case the 'sandwich' regions between the rows of core dimers are already fully occupied and the bulk antenna pool is the favoured location for these new LH2 complexes.
Oligomeric characterization of the photosynthetic apparatus of Rhodobacter sphaeroides R26.1 by nondenaturing electrophoresis methods

Abstract:Blue and colorless native gel electrophoresis in combination with LC-ESI-MS/MS are powerful tools in the analysis of protein networks in biological membranes. We used these techniques in the present study to generate a comprehensive overview on a proteome-wide scale of intracytoplasmic membrane (ICM) associated proteins in order to investigate the native supramolecular organization of Rhodobacter sphaeroides R26.1 photosynthetic apparatus. The results obtained were compared with past proteomic data, as well as with models for the topology of photosynthetic membranes as derived from previously published atomic force microscopy studies. We identified 52 proteins organized in 10 different multiprotein complexes. We were able to demonstrate the existence of different oligomeric states for the integral membrane pigment-protein complexes dedicated to bacterial photosynthesis. Specifically, we found dimers and trimers, as well as supercomplexes of light-harvesting (LH) 2 at very high molecular weights (around 10,000 kDa). We recovered the monomeric form of the photochemical reaction center (RC), as well as the monomer and dimer of the reaction center-light harvesting 1-PufX (RC-LH1-PufX) complex. Curiously, no type of LH1 complex was detected. Lastly, ATP synthase and cytochrome bc(1) complexes were only recovered in their monomeric states. Purified ICM vesicles were shown to be rich in newly discovered gene products, including three proteins with unknown functions (RSP_2125, RSP_3238, RSP_6207), a possible alkane hydroxylase and a spheroidene monooxygenase. Other multiprotein complexes were found to be localized in the ICM, including succinate dehydrogenase in trimeric form and sarcosine oxidase in two different aggregation states. These findings contribute to the growing body of evidence that the bacterial ICM is a specialized bioenergetic membrane hosting, not only photosynthesis, but many other critical activities.

Abstract:Symbiogenesis overshadows the importance of other eukaryogenetic processes. By working on the endosymbiotic cellular heredity in its entirety, it transformed the eukaryotic world. This mini-review strived to produce a concise account of symbiogenetic heredity of membranes in eukaryotes. Symbiogenesis integrated the endosymbiotic alpha-proteobacterium and cyanobacterium with the host, by utilising almost all the major prokaryotic components of membranes and protein translocation machinery along with a lot of eukaryotic inventions. It beautifully compartmentalized the eukaryotic cell by putting the prokaryotic membranes in continuity with the eukaryotic membranes and produced a whole spectrum of membrane topologies. Topogenesis of symbiogenetic hereditary membranes produced cell organelles with a diversity of metabolic capabilities. Development of protein translocation system manifests real ingenuity of symbiogenetic processes which integrates the working of entire compliment of cellular organelles. Abstract:A cell-free translation system from the facultatively photoheterotrophic bacterium Rhodobacter capsulatus is described. Synthesis of two proteins of the bacterium's photosynthetic apparatus (light-harvesting complex B870 alpha and beta) was performed by SP6 polymerase transcription of the subcloned genes, isolation of the mRNA and translation in vitro using a cell-free extract of R. capsulatus cells. The integration of these proteins in vitro into added intracytoplasmic membrane vesicles (ICM) is demonstrated. Without addition of ICM approximately 70% of the synthesized B870 proteins were soluble. If, however, ICM were present during synthesis, the majority of the soluble protein was found to associate with the membranes. The membrane-associated polypeptides could be solubilized only by detergent treatment but could not be extracted by treatment at alkaline pH (Na2CO3), suggesting that the proteins had been firmly inserted into the lipid bilayer. Moreover, the B870 alpha and beta proteins that integrated in vitro into ICM were also found to associate with pigment ligands and to assemble into a native reaction center/B870 complex. The native conformation of this complex isolated from ICM by Triton fractionation was demonstrated by microspectral analysis of the bound pigments.

Radiofrequency ablation of small renal masses as an alternative to nephron-sparing surgery: preliminary results

Abstract:Introduction Radical endoscopic minimal-invasive treatment methods, such as thermal ablation, are sought as an alternative to standard radical surgical treatment of kidney neoplasms. We analysed patients who could be qualified for radical treatment due to T1a renal tumour. Material and methods Twenty-three patients out of 129 who underwent radiofrequency thermal ablation of kidney tumours in the years 2003-2010 were analysed. The inclusion criteria were age below 70 years, lack of major comorbidities (ASA score 1, 2), and competent contralateral kidney. In all cases tumour size was below 4 cm. All patients were followed up with computed tomography (CT) and ultrasonography (USG) every 6 months for 3 years. Results In 20 patients kidney tumour was biopsied before radiofrequency ablation (RFA) and 10 of these biopsies were positive and revealed cancer. Six patients required additional treatment due to recurrence visible in CT – 3 with a positive biopsy result, 1 with negative and 2 without biopsy. Three of them were treated with a second session of RFA, 1 with radical nephrectomy and 2 with partial nephrectomy. No disease dissemination was observed and all patients who received additional treatment remain disease free. Conclusions The RFA can be safely used in selected patients with T1a tumour as an alternative to partial nephrectomy. Careful follow-up is required after thermal ablation and allows early detection and successful treatment of recurrences.
Revealing the Dynamics of Thylakoid Membranes in Living Cyanobacterial Cells

Abstract:Cyanobacteria are photosynthetic prokaryotes that make major contributions to the production of the oxygen in the Earth atmosphere. The photosynthetic machinery in cyanobacterial cells is housed in flattened membrane structures called thylakoids. The structural organization of cyanobacterial cells and the arrangement of the thylakoid membranes in response to environmental conditions have been widely investigated. However, there is limited knowledge about the internal dynamics of these membranes in terms of their flexibility and motion during the photosynthetic process. We present a direct observation of thylakoid membrane undulatory motion in vivo and show a connection between membrane mobility and photosynthetic activity. High-resolution inelastic neutron scattering experiments on the cyanobacterium Synechocystis sp. PCC 6803 assessed the flexibility of cyanobacterial thylakoid membrane sheets and the dependence of the membranes on illumination conditions. We observed softer thylakoid membranes in the dark that have three-to four fold excess mobility compared to membranes under high light conditions. Our analysis indicates that electron transfer between photosynthetic reaction centers and the associated electrochemical proton gradient across the thylakoid membrane result in a significant driving force for excess membrane dynamics. These observations provide a deeper understanding of the relationship between photosynthesis and cellular architecture.
Proteomic characterization of the Rhodobacter sphaeroides 2.4.1 photosynthetic membrane: identification of new proteins

Abstract:The Rhodobacter sphaeroides intracytoplasmic membrane (ICM) is an inducible membrane that is dedicated to the major events of bacterial photosynthesis, including harvesting light energy, separating primary charges, and transporting electrons. In this study, multichromatographic methods coupled with Fourier transform ion cyclotron resonance mass spectrometry, combined with subcellular fractionation, was used to test the hypothesis that the photosynthetic membrane of R. sphaeroides 2.4.1 contains a significant number of heretofore unidentified proteins in addition to the integral membrane pigment-protein complexes, including light-harvesting complexes 1 and 2, the photochemical reaction center, and the cytochrome bc(1) complex described previously. Purified ICM vesicles are shown to be enriched in several abundant, newly identified membrane proteins, including a protein of unknown function (AffyChip designation RSP1760) and a possible alkane hydroxylase (RSP1467). When the genes encoding these proteins are mutated, specific photosynthetic phenotypes are noted, illustrating the potential new insights into solar energy utilization to be gained by this proteomic blueprint of the ICM. In addition, proteins necessary for other cellular functions, such as ATP synthesis, respiration, solute transport, protein translocation, and other physiological processes, were also identified to be in association with the ICM. This study is the first to provide a more global view of the protein composition of a photosynthetic membrane from any source. This protein blueprint also provides insights into potential mechanisms for the assembly of the pigment-protein complexes of the photosynthetic apparatus, the formation of the lipid bilayer that houses these integral membrane proteins, and the possible functional interactions of ICM proteins with activities that reside in domains outside this specialized bioenergetic membrane.

Abstract:In bacteria and eukaryotes alike, proper cellular physiology relies on robust subcellular organization. For the phototrophic purple nonsulfur bacteria (PNSB), this organization entails the use of a light-harvesting, membrane-bound compartment known as the intracytoplasmic membrane (ICM). Here we show that ICMs are spatially and temporally localized in diverse patterns among PNSB. We visualized ICMs in live cells of 14 PNSB species across nine genera by exploiting the natural autofluorescence of the photosynthetic pigment bacteriochlorophyll (BChl). We then quantitatively characterized ICM localization using automated computational analysis of BChl fluorescence patterns within single cells across the population. We revealed that while many PNSB elaborate ICMs along the entirety of the cell, species across as least two genera restrict ICMs to discrete, nonrandom sites near cell poles in a manner coordinated with cell growth and division. Phylogenetic and phenotypic comparisons established that ICM localization and ICM architecture are not strictly interdependent and that neither trait fully correlates with the evolutionary relatedness of the species. The natural diversity of ICM localization revealed herein has implications for both the evolution of phototrophic organisms and their light-harvesting compartments and the mechanisms underpinning spatial organization of bacterial compartments. IMPORTANCE Many bacteria organize their cellular space by constructing subcellular compartments that are arranged in specific, physiologically relevant patterns. The purple nonsulfur bacteria (PNSB) utilize a membrane-bound compartment known as the intracytoplasmic membrane (ICM) to harvest light for photosynthesis. It was previously unknown whether ICM localization within cells is systematic or irregular and if ICM localization is conserved among PNSB. Here we surveyed ICM localization in diverse PNSB and show that ICMs are spatially organized in species-specific patterns. Most strikingly, several PNSB resolutely restrict ICMs to regions near the cell poles, leaving much of the cell devoid of light-harvesting machinery. Our results demonstrate that bacteria of a common lifestyle utilize unequal portions of their intracellular space to harvest light, despite light harvesting being a process that is intuitively influenced by surface area. Our findings therefore raise fundamental questions about ICM biology and evolution.

Abstract:Cyanobacteria and chloroplasts of algae and plants harbor specialized thylakoid membranes that convert sunlight into chemical energy. These membranes house photosystems II and I, the vital protein-pigment complexes that drive oxygenic photosynthesis. In the course of their evolution, thylakoid membranes have diversified in structure. However, the core machinery for photosynthetic electron transport remained largely unchanged, with adaptations occurring primarily in the light-harvesting antenna systems. Whereas thylakoid membranes in cyanobacteria are relatively simple they become more complex in algae and plants. The chloroplasts of vascular plants contain intricate networks of stacked grana and unstacked stroma thylakoids. This review provides an in-depth view of thylakoid membrane architectures in phototrophs, and the determinants that shape their forms, as well as presenting recent insights into the spatial organization of their biogenesis and maintenance. Its overall goal is to define the underlying principles that have guided the evolution of these bioenergetic membranes.

[N-myc status of neuroblastoma from bone marrow cells].

Abstract:The analysis of the N-myc gene in bone marrow specimens at the time of initial diagnosis and as well at the time of relapse from patients with Neuroblastoma stage IV and bone marrow infiltration could give some informations about the N-myc status of these patients. In stage IV neuroblastoma patients with bone marrow infiltration an estimation of the N-myc gene amplification should be attempted, if otherwise no information about the tumor content of the N-myc gene could be gathered. In our investigation we could demonstrate a Southern-blot-analysis of 27 bone marrow specimens with respect to the N-myc gene status which correlated qualitatively well to the N-myc amplification detected later on in the corresponding tumor tissue. In six cases the tumor infiltrated bone marrow showed a clear amplification of the N-myc gene. Because of the contamination by non malignant cells in bone marrow there was a quantitative difference in the calculated N-myc gene copies between the examined bone marrow specimens and corresponding tumor tissue.
Biosynthesis of the photosynthetic membranes of Rhodopseudomonas sphaeroides

Abstract:The steady-state biosynthesis of the photosynthetic membrane (ICM) of Rhodopseudomonas sphaeroides has been reviewed. At moderate light intensities, 500 ft-c, preexisting ICM serves as the insertion matrix for newly synthesized membrane components. Whereas the bulk of the membrane protein, protein-pigment complexes, and pigments are inserted into preexisting ICM throughout the cell cycle, phospholipid is transferred from outside the ICM to the ICM only at the time of cell division. Because the site of cellular phospholipid synthesis is the cytoplasmic membrane, these results infer that despite the physical continuity of cytoplasmic membrane and ICM, there must exist between these membranous domains a "barrier" to the free diffusion of cellular phospholipid. The cyclical alternation in protein to phospholipid ratio of the ICM infers major structural and functional alternations, such as changes in the protein to lipid ratio of the membrane, specific density of the membrane, lipid structure within the membrane, and the rate of cyclic electron flow. When biochemical studies are correlated with detailed electron microscopic investigations we can further conclude that the number of photosynthetic units within the plane of the membrane can vary by nearly a factor of two over the course of the cell cycle. The average physical size of the photosynthetic units is constant for a given light intensity but inversely proportional to light intensity. The distribution of photosynthetic unit size classes within the membrane can be interpreted as suggesting that the "core" of the photosynthetic unit (reaction center plus fixed antenna complex) is inserted into the membrane coordinately as a structural entity. The variable antenna complex is, on the other hand, inserted independent of the "core" and randomly associates with both old and new core complexes. Finally, we conclude that there is substantial substructure to te distribution of photosynthetic units within the ICM, ie, they are highly ordered and exist in a defined spatial orientation to one another.

Abstract:Photosynthetic organisms rely on a network of light-harvesting protein-pigment complexes to efficiently absorb sunlight and transfer excitation energy to reaction center proteins for charge separation. In photosynthetic purple bacteria, these complexes are embedded in the cell membrane, where lipid composition affects their clustering and inter-complex energy transfer. However, the lipid bilayer's impact on intra-complex excitation dynamics is less understood. Recent experiments compared photo-excitation dynamics in detergent-isolated light harvesting complex 2 (LH2) to LH2 embedded in membrane discs mimicking the biological environment, revealing differences in spectra and intra-complex energy transfer rates. We use available quantum chemical and spectroscopy data to develop a complementary theoretical study on the excitonic structure and intra-complex energy transfer kinetics of the LH2 from photosynthetic purple bacteria Rhodoblastus acidophilus in two conditions: LH2 in a membrane environment and detergent-isolated LH2. Dark excitonic states crucial for B800-B850 energy transfer within LH2 are found to be more delocalised in the membrane model. Using non-perturbative and generalised Förster calculations, it is shown that the increased quantum delocalisation leads to a B800 to B850 transfer rate 30% faster than in the detergent-isolated complex, consistent with experimental results. We identify the main energy transfer pathways in each environment and show how differences in the B800 to B850 transfer rate stem from changes in LH2's electronic properties when embedded in the membrane. By considering quasi-static variations of electronic excitation energies in LH2, we show that the broadening of the B800 to B850 transfer rate distribution is affected by lipid composition. We argue that the variation in broadening could indicate a speed-accuracy trade-off, common in biological systems.
