Abstract:Islands provide a great system to explore the processes that maintain genetic diversity and promote local adaptation. We explored the genomic diversity of the Balearic lizard , an endemic species characterized by numerous small insular populations with large phenotypic diversity. Using the newly available genome for this species, we characterized more than 300,000 SNPs, merging Genotype by Sequencing (GBS) data with previously published Restriction-site associated DNA sequencing (RADSeq) data, providing a dataset of 16 island populations (191 individuals) across the range of species distribution (Menorca, Mallorca, and Cabrera). Results indicate that each islet hosts a well-differentiated population (Fst=0.247±0.09), with no recent immigration/translocation events. Contrary to expectations, most populations harbor a considerable genetic diversity (mean nucleotide diversity, Pi=0.144±0.021), characterized by overall low inbreeding values (Fis<0.1). While the genetic diversity significantly decreased with decreasing islet surface, maintenance of substantial genetic diversity even in tiny islets suggests variable selection or other mechanisms that buffer genetic drift. Maximum-likelihood tree based on concatenated SNP data confirmed the existence of the two major independent lineages of Menorca and Mallorca/Cabrera. Multiple lines of evidence, including admixture and root testing, robustly placed the origin of the species in the Mallorca Island, rather than in Menorca. Outlier analysis mainly retrieved a strong signature of genome differentiation between the two major archipelagos, especially in the sexual chromosome Z. A set of proteins were target of multiple outliers and primarily associated with binding and catalytic activity, providing interesting candidates for future selection studies. This study provides the framework to explore crucial aspects of the genetic basis of phenotypic divergence and insular adaptation.

Abstract:Corsica and Sardinia are amongst the largest islands of the Western Mediterranean. Their complex geological history included belonging to the European–Iberian continental margin (close to current-day Catalonia and Provence) and varying degrees of isolation for the last 30 million years, leading to peculiar, highly endemic faunas and floras. This is especially true for their earthworm faunas, which include endemic species of several Lumbricoidea genera and the endemic family (or subfamily) Diporodrilidae. Only three species have been described for the morphologically unique Diporodrilus , but there exists evidence for wide morphological variability within them and the existence of several species-level genetic lineages within Corsica. This work aimed to investigate the genetic diversity and phylogenetic relationships between the genetic lineages of Diporodrilus from Corsica and Sardinia (based on the sequences of 5 mito-nuclear markers), to perform an integrative systematics revision combining species delimitation techniques and morphological data, and to obtain a time-calibrated phylogeny of Diporodrilus and other Corso-Sardinian Lumbricoidea. Within 15 populations of the morphospecies Diporodrilus omodeoi and Diporodrilus pilosus across Corsica and Sardinia, 10 species-level genetic lineages were detected. Phylogenetic independence, high genetic divergence and morphological differences provided the support for the description of five new pseudocryptic species: Diporodrilus rotundus sp. nov., Di. jorgei sp. nov., Di. minor sp. nov., Di. meridionalis sp. nov. and Di. telti sp. nov. Time-calibrated phylogenetic inference estimated the age for genus Diporodrilus at 65.9 Mya; even if other Corso-Sardinian genera were significantly younger, all of them presented deep divergences predating the break-off of the microplate from the continent. The almost threefold increase in the known diversity of Diporodrilus stresses the need for comprehensive earthworm diversity inventories in both Sardinia and Corsica, and for studies on their ecological role and conservation status. The close correspondence between some geological and cladogenetic events suggest that the distribution of Corso-Sardinian earthworms could be used to inform standing geological controversies.

Abstract:The genetic structure and dispersal dynamics of reptile populations are profoundly influenced by natural processes and human activities. While natural dispersal is shaped by species' characteristics and paleogeographical features, human-mediated translocations have become increasingly prevalent, posing ecological challenges. Mitochondrial genetic markers have been pivotal in untangling invasion pathways for various species. Our study focuses on the Dalmatian Algyroides, Algyroides nigropunctatus (Duméril & Bibron, 1839), a lizard species endemic to the Balkan Peninsula, where recent observations in the Apulian region of Italy suggest an introduced population. Genetic analyses employing two mtDNA markers (16S and ND4 genes) elucidate the phylogenetic relationships of the Dalmatian Algyroides and trace the geographic origin of the introduced population. Our findings reveal areas in western Greece and southwestern Albania as the most probable areas of the source population, while we identify two previously undetected geographical lineages in the native range, highlighting the complex evolutionary history of the species in the region. Additionally, indications of potential glacial refugia and post-glacial dispersal patterns shed more light on the species' demographic dynamics.

Genomic and mitochondrial evidence of ancient isolations and extreme introgression in the four-lined snake

Abstract:Comparing mitochondrial and genomic phylogenies is an essential tool for investigating speciation processes, because each genome carries different inheritance properties and evolutionary characteristics. Furthermore, mitonuclear discordance may arise from ecological adaptation, historic isolation, population size changes, and sex-biased dispersal. Closely related taxa are expected to experience gene flow; however, this may not be true for insular populations or populations isolated in refugia. The four-lined snake Elaphe quatuorlineata has a fragmented distribution, separating populations of the Italian and Balkan Peninsulas, whereas several insular Aegean populations of significantly smaller body size (Cyclades island group and Skyros Island, Greece) are currently considered distinct subspecies. We constructed the species-tree phylogeny of this species utilizing genome-wide single nucleotide polymorphisms and a gene-tree based on complete cytochrome b sequences, aiming to detect convergence and discrepancies between biparentally and maternally inherited genomes. Population structuring, phylogenetic patterns and migration events among geographically defined lineages supported our hypothesis of isolation in multiple sub-refugia. Where biogeographical barriers did not restrict subsequent dispersal, extensive genetic exchange occurred between mainland Balkan populations. This process has led to the mitochondrial sweep of an ancestral mitolineage that survived only in peripheral (East Greece) and insular populations (North Cyclades and Skyros). The Central Cyclades represent an ancient lineage for both molecular markers that emerged almost 3.3 Mya. Considering their distinct morphology, insular E. quatuorlineata populations should be the future focus of an extensive sampling, especially since the mitonuclear discordance observed in this species could be related to ecological adaptations, such as the island-dwarfism phenomenon.

Abstract:We present a comprehensive population genomic study of 16 populations of the endemic lizard of the Balearic Islands, Podarcis lilfordi. By mapping GBS and RAD‐Seq SNP data to the newly available genome for this species, we found considerable level of genetic diversity and low inbreeding depression for most populations. We reconstructed a scenario of colonization within and among the major archipelagos, robustly placing the origin of this species in the main Mallorca Island. Abstract:A combined approach based on karyology and DNA taxonomy allowed us to characterize the taxonomic peculiarities in 10 Mesoamerican lizard species, belonging to six genera and five families, inhabiting two Biosphere Reserve in Chiapas, Mexico: La Sepultura Biosphere Reserve, and Montes Azules Biosphere. The karyotypes of four species, Phyllodactylus sp. 3 (P. tuberculosus species group) (2n = 38), Holcosus festivus (Lichtenstein et von Martens, 1856) (2n = 50), Anolis lemurinus Cope, 1861 (2n = 40), and A. uniformis Cope, 1885 (2n = 29-30) are described for the first time, the last one showing a particular X1X1X2X2/X1X2Y condition. In Aspidoscelis deppii (Wiegmann, 1834) (2n = 50) and Anolis capito Peters, 1863 (2n = 42), we found a different karyotype from the ones previously reported for these species. Moreover, in A. capito, the cytogenetic observation is concurrent with a considerable genetic divergence (9%) at the studied mtDNA marker (MT-ND2), which is indicative of a putative new cryptic species. The skink Scincella cherriei (Cope, 1893), showed high values of genetic divergence (5.2% at 16S gene) between the specimens from Montes Azules and those from Costa Rica and Nicaragua, comparable to the values typical of sister species in skinks. A lower level of genetic divergence, compatible with an intraspecific phylogeographic structure, has been identified in Lepidophyma flavimaculatum Duméril, 1851. These new data identify taxa that urgently require more in-depth taxonomic studies especially in these areas where habitat alteration is proceeding at an alarming rate.
What story does geographic separation of insular bats tell? A case study on Sardinian rhinolophids

Abstract:Competition may lead to changes in a species' environmental niche in areas of sympatry and shifts in the niche of weaker competitors to occupy areas where stronger ones are rarer. Although mainland Mediterranean (Rhinolophus euryale) and Mehely's (R. mehelyi) horseshoe bats mitigate competition by habitat partitioning, this may not be true on resource-limited systems such as islands. We hypothesize that Sardinian R. euryale (SAR) have a distinct ecological niche suited to persist in the south of Sardinia where R. mehelyi is rarer. Assuming that SAR originated from other Italian populations (PES)--mostly allopatric with R. mehelyi--once on Sardinia the former may have undergone niche displacement driven by R. mehelyi. Alternatively, its niche could have been inherited from a Maghrebian source population. We: a) generated Maxent Species Distribution Models (SDM) for Sardinian populations; b) calibrated a model with PES occurrences and projected it to Sardinia to see whether PES niche would increase R. euryale's sympatry with R. mehelyi; and c) tested for niche similarity between R. mehelyi and PES, PES and SAR, and R. mehelyi and SAR. Finally we predicted R. euryale's range in Northern Africa both in the present and during the Last Glacial Maximum (LGM) by calibrating SDMs respectively with SAR and PES occurrences and projecting them to the Maghreb. R. mehelyi and PES showed niche similarity potentially leading to competition. According to PES' niche, R. euryale would show a larger sympatry with R. mehelyi on Sardinia than according to SAR niche. Such niches have null similarity. The current and LGM Maghrebian ranges of R. euryale were predicted to be wide according to SAR's niche, negligible according to PES' niche. SAR's niche allows R. euryale to persist where R. mehelyi is rarer and competition probably mild. Possible explanations may be competition-driven niche displacement or Maghrebian origin.

Abstract:Italy hosts a large number of endemic freshwater fish species due to complex geological events which promoted genetic differentiation and allopatric speciation. Among them, the South European roach Sarmarutilus rubilio inhabits various freshwater environments in three different ichthyogeographic districts. We investigated the genetic diversity of S. rubilio using two different mitochondrial markers (COI and CR), aiming to define its relationship with other similar taxa from the Balkan area and, from a phylogeographic perspective, test the effects of past hydrogeological dynamics of Italian river basins on its genetic structure and demographic history. Our analysis highlighted a marked genetic divergence between S. rubilio and all other roach species and, among Italian samples, revealed the existence of three deeply divergent geographic haplogroups, named A, B and C. Haplogroup C likely corresponds to a new putative cryptic species and is located at the northern border of the South European roach range; haplogroup B is restricted to Southern Italy; and haplogroup A is widespread across the entire range and in some sites it is in co-occurrence with C or B. Their origin is probably related to the tectonic uplifting of the Apuan Alps in the north and of the Colli Albani Volcano in the south during the Pleistocene, which promoted isolation and vicariance followed by secondary contacts.

Abstract:The day gecko Phelsuma borbonica displays marked differences in coloration and occupies a variety of habitats across its very small range on the volcanic oceanic island of Reunion in the southwestern Indian Ocean. Such differences might reflect geographic and ecological divergence and thus be associated with an underlying genetic differentiation. We examined the partitioning of genetic diversity in Phelsuma borbonica across Reunion at a very small spatial scale (< 10 km) using mtDNA and nuDNA sequences, along with microsatellite genotype data. This study provides an account of the evolutionary history of Phelsuma borbonica , along with a quantification of effective population sizes, which constitute important information for the conservation of this endangered species, which may represent a previously unrecognized case of incipient speciation. We found that populations of Phelsuma borbonica are genetically strongly differentiated, highlighting historically low levels of gene flow. Phylogenetic analyses suggest that Phelsuma borbonica reached its present range through two parallel southward expansions from an ancestral population located in the northern part of the island.

Abstract:Aim Recent literature is unveiling more complex responses of biodiversity to Pleistocenic climatic oscillations than those derived from initial models based solely on southern glacial macrorefugia. Here, we zoom in to delve into the genetic diversity and population structure of Podarcis lusitanicus, a lizard species native to the north‐western Iberian Peninsula. Location North‐western Iberian Peninsula. Taxon Podarcis lusitanicus. Methods Utilising a comprehensive multilocus approach, we explored the species' phylogeography and demographic history at a very fine spatial scale and timeframe. Concurrently, we evaluated both current and historical habitat suitability for the species, linking it to the observed demographic trends. We examined DNA sequences from two mitochondrial (cytb and ND4) and three nuclear genes (ACM4, MC1R and PDC) in 230 specimens collected from 21 populations spanning most of the species' distribution range. Results Our findings corroborate cytonuclear discordances in Podarcis, with nuclear genes failing to identify the mitochondrial clades in P. lusitanicus. Despite its separation from other Podarcis species approximately 4 million years ago, its diversification occurred much more recently, around 0.4 million years ago, resulting in three monophyletic lineages that are geographically coherent. This pattern suggests that some lineages of P. lusitanicus might have gone extinct after the split from the common ancestor. Pleistocene climatic oscillations profoundly influenced the demographic history of the species. During warmer periods, such as the Last Interglacial, populations expanded and came into secondary contact, whereas during the Last Glacial Maximum, populations contracted and became isolated in multiple micro‐refugia. The subsequent Mid‐Holocene warming facilitated population recovery, range expansion and lineage admixture. Main Conclusions This study underscores the intricate interplay between isolation and dispersal at a microscale, driven by Pleistocene climatic fluctuations, which have profoundly shaped the genetic structure and diversity of Iberian terrestrial biodiversity and may remain overlooked by biogeographic studies at coarser scales.

Abstract:Abstract The common practice of using subspecies as conservation targets raises the question of whether efforts are focused on preserving conspicuous components of the species’ phenotypic variability rather than evolutionarily significant units. To address this question, in this study we performed a comprehensive morphological and genetic assessment on all the subspecies of wall lizard described for the Aeolian Archipelago (Italy) to determine whether they represent distinct evolutionary lineages and/or discrete phenotypic partitions. Further, we applied a monophyly test to 70 subspecies belonging to seven wall lizard species occurring in Italy, based on our results and on previous phylogeographic studies. We found that none of the Aeolian subspecies represents a distinct evolutionary lineage, despite some morphological differentiation of island populations across the archipelago, suggesting a very recent origin of island populations and of the observed phenotype variation. Across seven wall lizard species, tests revealed that lizard subspecies rarely (< 9% of cases) match evolutionary units. This study demonstrates that intraspecific taxonomy of wall lizards is a poor predictor of phylogeographic partitions and evolutionary units, and therefore of limited use (if not dangerous) for defining conservation and management units. A better approach would be relying on the integration of genomic and phenotypic data to assess the evolutionary significance and conservation value of phenotypic and genetic units within species.

The geography and timing of genetic divergence in the lizard Phrynocephalus theobaldi on the Qinghai-Tibetan plateau

Abstract:The Qinghai-Tibetan Plateau (QTP) represents one of the earth's most significant physical features and there is increasing interest in the historical generation of biodiversity within this region. We hypothesized that there should be clear geographically coherent genetic structuring within one of the world's highest altitude lizards, Phrynocephalus theobaldi, due to considerable historical population fragmentation in this environment. This was tested using a major mitochondrial DNA (mtDNA) survey and sequencing of two nuclear markers (AME and RAG-1) from P. theobaldi, from across the southern QTP. A Bayesian method (BPEC) was used to detect four geographically structured mtDNA clusters. A Bayesian phylogenetic tree, together with associated dating analyses, supported four corresponding evolutionary lineages with a timing of 3.74-7.03 Ma for the most basal P. theobaldi split and Pliocene splits of 2.97-5.79 Ma and 2.40-5.39 Ma in the two daughter lineages. Himalayan uplift and changes in the Jilong basin may have contributed to these divergences, but uplift of the Gangdese mountains is rejected due to its timing. The nuclear markers appeared to be sorted between the four mtDNA groups, and species delimitation analyses supported the four phylogeographical groups as candidate species. The study contributes to our understanding of biodiversity on the QTP.

Abstract:Sicily is one of the main islands of the Mediterranean Sea, and it is characterized by a variety of archaeological records, material culture and traditions, reflecting the history of migrations and populations' interaction since its first colonization, during the Paleolithic. These deep and complex demographic and cultural dynamics should have affected the genomic landscape of Sicily at different levels; however, the relative impact of these migrations on the genomic structure and differentiation within the island remains largely unknown. The available Sicilian modern genetic data gave a picture of the current genetic structure, but the paucity of ancient data did not allow so far to make predictions about the level of historical variation. In this work, we sequenced and analyzed the complete mitochondrial genomes of 36 individuals from five different locations in Sicily, spanning from Early Bronze Age to Iron Age, and with different cultural backgrounds. The comparison with coeval groups from the Mediterranean Basin highlighted structured genetic variation in Sicily since Early Bronze Age, thus supporting a demic impact of the cultural transitions within the Island. Explicit model testing through Approximate Bayesian Computation allowed us to make predictions about the origin of Sicanians, one of the three indigenous peoples of Sicily, whose foreign origin from Spain, historically attributed, was not confirmed by our analysis of genetic data. Sicilian modern mitochondrial data show a different, more homogeneous, genetic composition, calling for a recent genetic replacement in the Island of pre-Iron Age populations, that should be further investigated.

Abstract:<h3>Background</h3><p>The Pleistocene cyclic sea-level fluctuations are thought to have markedly affected the distribution and genetic architecture of Atlanto-Mediterranean biota. Despite the acknowledged key role played by these historical events in shaping population genetic structure of marine species, little is still known about the processes involved in shaping the spatial distribution of genetic variation within intertidal species. We intended in this study to reconstruct the phylogeography of a common and widely distributed coastal species across the East Atlantic and Mediterranean Sea (the warty crab <em>Eriphia verrucosa</em>), aiming to unravel potential microevolutionary processes likely involved in shaping its genetic polymorphism. For this purpose, a total of 155 specimens of <em>E. verrucosa</em> from 35 locations across the entire distribution range were analyzed by comparing a 453 basepairs region of the mitochondrial gene cytochrome oxidase subunit 1 (Cox1).</p><h3>Results</h3><p>Our results unveiled the prevalence of high genetic connectivity among East Atlantic and Mediterranean populations, with noticeable genetic distinctiveness of the peripheral population from the Azores. Spatio-temporal patterns of genetic diversification and demographic history allowed retrieving genetic imprints of late Pleistocene vicariant event across the Gibraltar Strait followed by subsequent postglacial expansion events for both the East Atlantic and Mediterranean regions. Integrative evidences from the outcomes of comparison of regional genetic diversification, as well as evolutionary and biogeographic histories reconstructions, support the existence of potential glacial refugia for <em>E. verrucosa</em> in the East Atlantic and western Mediterranean. Our results also revealed low levels of genetic variability along with recent demographic and spatial expansion events for eastern Mediterranean warty crabs, suggesting that the eastern areas within the distribution range of the species might have been recently colonized from putative glacial refugia.</p><h3>Conclusions</h3><p>These findings provide new insights into the phylogeography and evolutionary history of a common but poorly studied Atlanto-Mediterranean decapod species. Specifically, they contribute to the understanding of the impact of historical processes on shaping contemporary population genetic structure and diversity in intertidal marine species.</p>

Abstract:The Lygodactylus tolampyae complex includes several deep genetic lineages of small diurnal geckos from the West and North West of Madagascar whose taxonomy is largely unsolved. We sequenced DNA fragments of one mitochondrial and four nuclear-encoded genes for up to 70 samples across the entire known range of these geckos. We find as many as 11 mitochondrial lineages differentiated by >4% pairwise distances in the 16S rRNA gene fragment, with >9% pairwise distance for the majority of lineage comparisons. Many of these lineages were concordantly differentiated in all of the nuclear-encoded genes without any haplotype sharing, despite the syntopic occurrence of some of them. We therefore hypothesize that the complex contains seven candidate species, but a comprehensive taxonomic resolution is complicated by various hindrances. These include incomplete sampling, with two lineages each known only from a single specimen, and one further lineage with no voucher specimens available for examination. Further hurdles are the probably lost holotype of L. tolampyae and its imprecise type locality, as well as the apparent lack of any morphological differentiation between the majority of the genetic lineages. Based on a survey of historical literature and the travel routes of the original collector, A. Grandidier, we conclude that the provenance of the holotype of L. tolampyae is likely in the wider Morondava area in the West and assign the sole candidate species from this area to this name. We then proceed to describe three species that represent separate genetic lineages for all markers studied: Lygodactylus morii sp. nov., a species common in Ankarafantsika National Park and several nearby sites in the North West; L. herilalai sp. nov., a species occurring in close syntopy with L. morii in Ankarafantsika without any signal of genetic admixture; and L. schwitzeri sp. nov. from Sahamalaza Peninsula in the North West. This leaves three more lineages without a name and with the need to gather additional samples, two from Namoroka National Park and one from other sites in the North West. We confirm the L. tolampyae complex to be an apparently rare example of truly cryptic reptile species in Madagascar, where even detailed morphological examination does not reveal morphological differences among lineages that are clearly evolutionarily independent and require recognition as distinct species due to their co-occurrence without admixture.

Abstract:The processes shaping population dynamics of benthic marine invertebrates with non-planktotrophic larvae are still poorly understood but have seen a renewed interest in applying integrative taxonomic approaches. We used mitochondrial and microsatellite (SSR-GBAS) data to estimate connectivity across islands and seamounts in the central North Atlantic Azores Archipelago in five species of the bryozoan genus Reteporella Busk, 1884. Discordant patterns were inferred between datasets, which might be due to methodological constraints related to the application of multilocus approaches based on amplification to multiple species or due to interspecific introgression in deep waters. A divergent cryptic ecotype of Reteporella atlantica (Busk, 1884) was found in shallow waters, likely resulting from ecologically-driven incipient speciation, posing new questions regarding the role of bathymetrical zonation as a promoter of differentiation. The occurrence of ecologically-driven differentiation and potential interspecific introgression in other bryozoans should be considered, both with potentially important evolutionary and biogeographical consequences. The discovery of incipient species, prompted by ecological factors, calls for the need to consider marine invertebrates when developing conservation strategies in oceanic insular ecosystems.

Abstract:In this study, we investigate the phylogeographic patterns of Dolichophis species in the Aegean region, aiming to elucidate their genetic diversity and putative historical colonisation routes through mitochondrial and nuclear DNA data. Our findings revealed distinct phylogeographic patterns: D. caspius exhibited a higher level of haplotypes within two shallow mitochondrial lineages, contrasting with D. jugularis, which displayed lower genetic variability in the area. Additionally, we identified evidence showing possible human-mediated historical translocation of D. caspius populations to Karpathos from the Balkans mainland. The mitochondrial variability in D. jugularis remained relatively uniform across southwestern Anatolia and Dodecanese, except for Rhodes Island. The evidence from mitochondrial and nuclear data confirming the previously described morphological differentiation of the Rhodes snakes, and thus the name D. j. zinneri Cattaneo, 2012, described on the island, could be applied to this isolated population. This result addresses the first genetic view on the long-standing taxonomic uncertainties regarding the subspecies status of Rhodes D. jugularis. Our results also raise questions regarding possible historical hybridisations between D. caspius and D. jugularis in the Dodecanese islands, prompting the need for further investigation using extensive field studies and genomic approaches. Ultimately, the Aegean islands, particularly Kos and Rhodes, seem to be important sites for the evolution of these colubrid snakes and their historical dynamics.

Abstract:Interspecific hybridisation can be consequential for rare and insular endemic species. The Critically Endangered Aeolian wall lizard, , severely declined due to interactions with the invasive Italian wall lizard, . The largest population of survives on a narrow peninsula (Capo Grosso) that is mildly connected to the island of Vulcano, which has been entirely invaded by . Recent observation of individuals with an intermediate phenotype raised concern over the risk that hybridisation might swamp this last stronghold. We genetically characterised lizards from Vulcano using genome-wide SNPs, considering individuals showing multiple phenotypes (native, invasive, and “intermediate”). Hybridisation rate was low (∼3%), with just two F1 hybrids and two backcrosses. However, pure showed extremely low genetic diversity, a very small effective population size, and a low N /N ratio. Management strategies are urgently needed to control invasive species and maintain the genetic diversity of .

Cytogenetic Characterization of a Population of Acanthodactylus lineomaculatus Duméril and Bibron, 1839 (Reptilia, Lacertidae), from Southwestern Morocco and Insights into Sex Chromosome Evolution

Abstract:Acanthodactylus lineomaculatus is now regarded as an ecotype of A. erythrurus with which it has been recently synonymized. Despite the wide range of A. erythrurus, karyological data for this species are scarce and limited to classical cytogenetic studies carried out in individuals from only 2 locations (central Spain and Spanish enclave of Melilla on the northwestern Mediterranean Moroccan coast). Here, for the first time, we cytogenetically characterized individuals of A. lineomaculatus from the southwestern Moroccan Atlantic coast with the aim to increase the karyological knowledge of this wide-ranging species and to assess if any chromosomal changes can be found in this ecotype in comparison to other populations of this species. The diploid number of the individuals investigated is 2n = 38 which is typical of most lacertids. Active NORs were located telomerically in a medium-small pair of chromosomes, and no inactive NORs were detected. C-banding revealed an intensely heterochromatic W chromosome composed of AT-rich (centromere and long arm telomeric region) and GC-rich (most of the long arm) regions, with extended interstitial telomeric sequences. These telomere-like repeats occupy the GC-rich heterochromatin of the W. The DNA composition of the W represents a trait distinguishing A. lineomaculatus (southwestern Morocco) from A. erythrurus from Spain that possess a DAPI-positive (AT-rich) W chromosome. In conclusion, these results add further evidence to the remarkable karyotype conservation in lacertid lizards, although differences in NOR location and in W chromosome structure among populations could suggest an incipient speciation mediated by chromosome changes in this wide-ranging lizard species.

Abstract:Thermoregulation is essential for ectotherms but its relative cost, especially under ongoing climate change, depends on the thermal quality of habitats. Populations at the warm margin of a thermal niche could be negatively affected by environmental temperatures that approach the limits of a species' thermoregulation capacity. This study aims to define the thermal niche of the Bedriaga's rock lizard, Archaeolacerta bedriagae , a rock-dwelling species endemic to the Corsica and Sardinia islands (western Mediterranean Sea), and to investigate its thermoregulation effectiveness at the warm edge of its niche. We collected data on climate, body temperature, and microhabitat temperature throughout the species' range to characterize its thermal niche. We found that A. bedriagae does not occupy the entire climatic space available across its distribution range; rather, it selects temperate climates. Remarkably, thermoregulation effort increases when the habitat thermal quality decreases towards warmer sites. Populations at the warm edge of the thermal niche show the best thermoregulation effectiveness, but they are also more sensitive to the effects of climate change as they may already be at (or beyond) the species' maximum thermal capacity under the current conditions. We observed such a pattern at the extreme hot side of the thermal niche. This study provides key information on the thermoregulatory response of A. bedriagae to ongoing climate change that can be useful to identify populations facing a higher extinction risk either currently or in the near future.
