Abstract:Diabetic retinopathy (DR), a leading cause of vision impairment and blindness, is characterized by abnormal retinal vascular changes due to chronic hyperglycemia. The signaling pathway, essential for vascular growth and remodeling, has emerged as a key therapeutic target, though its molecular mechanisms and interactome remain largely unclear. Through a protein-centric approach, we identified a novel lncRNA and named it . lncRNA regulates endothelial cell migration, proliferation, tube formation, and permeability by modulating ER-calcium homeostasis and cytoskeleton dynamics. In zebrafish, modulation led to angiogenic defects, which were rescued by human orthologue. Our molecular studies further revealed that negatively regulates protein via ubiquitin-mediated degradation. Notably, expression is upregulated in the blood of DR patients and downregulated in endothelial DR cell models. Overexpression of restored endothelial permeability and VE-cadherin surface expression. These findings establish as a novel regulator of protein turnover, with potential therapeutic implications for diabetic retinopathy.

What problem does this paper attempt to address?
The main issue this paper attempts to address is the problem of vascular dysfunction in Diabetic Retinopathy (DR). Specifically, researchers have discovered a new long non-coding RNA (lncRNA) through a proteomics approach, named TAAL (Tie1-associated angiogenic lncRNA). TAAL plays a significant role in regulating endothelial cell migration, proliferation, tube formation, and permeability, which are closely related to vascular abnormalities in diabetic retinopathy. ### Main Issues: 1. **Vascular Dysfunction in Diabetic Retinopathy**: - Diabetic retinopathy is a leading cause of vision impairment and blindness, characterized by retinal vascular abnormalities due to chronic hyperglycemia. - The Tie1 signaling pathway plays a critical role in vascular growth and remodeling, but its molecular mechanisms and interaction networks remain unclear. 2. **Function of TAAL lncRNA**: - Researchers identified a new lncRNA through a proteomics approach and named it TAAL. - TAAL affects endothelial cell migration, proliferation, tube formation, and permeability by regulating endoplasmic reticulum calcium homeostasis and cytoskeletal dynamics. 3. **Interaction Between TAAL and Tie1 Protein**: - The study found that TAAL negatively regulates the stability of Tie1 protein through ubiquitin-mediated degradation. - In a zebrafish model, regulation by TAAL led to vascular defects, while the human homolog of TAAL could rescue these defects. 4. **Expression of TAAL in Diabetic Retinopathy**: - The study found that TAAL expression is upregulated in the blood of diabetic retinopathy patients, while it is downregulated in endothelial cell models. - Overexpression of TAAL can restore endothelial cell permeability and surface expression of VE-cadherin. ### Summary: This paper reveals the important role of TAAL lncRNA in regulating vascular function in diabetic retinopathy, particularly through its interaction with the Tie1 protein. These findings provide new potential targets for the treatment of diabetic retinopathy.
