Abstract:A tumor is a complex tissue comprised of heterogeneous cell subpopulations which exhibit substantial diversity at morphological, genetic and epigenetic levels. Under the selective pressure of cancer therapies, a minor treatment-resistant subpopulation could survive and repopulate. Therefore, the intra-tumor heterogeneity is recognized as a major obstacle to effective treatment. In this paper, we propose a stochastic clonal expansion model to simulate the dynamic evolution of tumor subpopulations and the therapeutic effect at different times during tumor progression. The model is incorporated in the CES webserver, for the convenience of simulation according to initial user input. Based on this model, we investigate the influence of various factors on tumor progression and treatment consequences and present conclusions drawn from observations, highlighting the importance of treatment timing. The model provides an intuitive illustration to deepen the understanding of temporal intra-tumor heterogeneity dynamics and treatment responses, thus helping the improvement of personalized diagnostic and therapeutic strategies.
Computing Algorithm for an Equilibrium of the Generalized Stackelberg Game

Abstract:The $1-N$ generalized Stackelberg game (single-leader multi-follower game) is intricately intertwined with the interaction between a leader and followers (hierarchical interaction) and the interaction among followers (simultaneous interaction). However, obtaining the optimal strategy of the leader is generally challenging due to the complex interactions among the leader and followers. Here, we propose a general methodology to find a generalized Stackelberg equilibrium of a $1-N$ generalized Stackelberg game. Specifically, we first provide the conditions where a generalized Stackelberg equilibrium always exists using the variational equilibrium concept. Next, to find an equilibrium in polynomial time, we transformed the $1-N$ generalized Stackelberg game into a $1-1$ Stackelberg game whose Stackelberg equilibrium is identical to that of the original. Finally, we propose an effective computation procedure based on the projected implicit gradient descent algorithm to find a Stackelberg equilibrium of the transformed $1-1$ Stackelberg game. We validate the proposed approaches using the two problems of deriving operating strategies for EV charging stations: (1) the first problem is optimizing the one-time charging price for EV users, in which a platform operator determines the price of electricity and EV users determine the optimal amount of charging for their satisfaction; and (2) the second problem is to determine the spatially varying charging price to optimally balance the demand and supply over every charging station.

Abstract:Standard of care in metastatic cancers typically applies the Maximum Tolerable Dose (MTD) of treatment or treatment combinations, either continuously or in repeated identical cycles, until unacceptable toxicity, cancer progression, or cure. Cure is rare due to fast-evolving therapy resistance. I will explain how game theory can help us not only understand why MTD fails but also design the first evolutionary anti-cancer therapies—therapies that anticipate and forestall the evolution of treatment-induced resistance in cancer cells. Such therapies can be designed through Stackelberg evolutionary games (SEGs), i.e., games between a rational leader (here, a physician) and evolutionary followers (here, evolving cancer cells). I will demonstrate how data science and machine learning can empower game theory to design evolutionary anti-cancer therapies and how evolutionary therapies lead to a better quality and quantity of patients' lives. A Model Predictive Stackelberg Solution to Orbital Pursuit-Evasion Game

Abstract:In this paper, we investigate analytical numerical iterative strategies for the pursuit-evasion game involving spacecraft with leader-follower information. In the proposed problem, the interplay between two spacecraft gives rise to a dynamic and real-time game, complicated further by the presence of J2 perturbation. The primary challenge lies in crafting control strategies that are both efficient and applicable to real-time game problems within a nonlinear system. To overcome this challenge, we introduce the model prediction and iterative correction technique proposed in model predictive static programming, enabling the generation of strategies in analytical iterative form for nonlinear systems. Subsequently, we proceed by integrating this model predictive framework into a simplified Stackelberg equilibrium formulation, tailored to address the practical complexities of leader-follower pursuit-evasion scenarios. Simulation results validate the effectiveness and exceptional efficiency of the proposed solution within a receding horizon framework.
A Mathematical Tumor Model with Immune Resistance and Drug Therapy: An Optimal Control Approach

Abstract:We present a competition model of cancer tumor growth that includes both the immune system response and drug therapy. This is a four-population model that includes tumor cells, host cells, immune cells, and drug interaction. We analyze the stability of the drug-free equilibria with respect to the immune response in order to look for target basins of attraction. One of our goals was to simulate qualitatively the asynchronous tumor-drug interaction known as “Jeffs phenomenon.” The model we develop is successful in generating this asynchronous response behavior. Our other goal was to identify treatment protocols that could improve standard pulsed chemotherapy regimens. Using optimal control theory with constraints and numerical simulations, we obtain new therapy protocols that we then compare with traditional pulsed periodic treatment. The optimal control generated therapies produce larger oscillations in the tumor population over time. However, by the end of the treatment period, total tumor size is smaller than that achieved through traditional pulsed therapy, and the normal cell population suffers nearly no oscillations.

The Contribution of Evolutionary Game Theory to Understanding and Treating Cancer

Abstract:Evolutionary game theory mathematically conceptualizes and analyzes biological interactions where one's fitness not only depends on one's own traits, but also on the traits of others. Typically, the individuals are not overtly rational and do not select, but rather inherit their traits. Cancer can be framed as such an evolutionary game, as it is composed of cells of heterogeneous types undergoing frequency-dependent selection. In this article, we first summarize existing works where evolutionary game theory has been employed in modeling cancer and improving its treatment. Some of these game-theoretic models suggest how one could anticipate and steer cancer's eco-evolutionary dynamics into states more desirable for the patient via evolutionary therapies. Such therapies offer great promise for increasing patient survival and decreasing drug toxicity, as demonstrated by some recent studies and clinical trials. We discuss clinical relevance of the existing game-theoretic models of cancer and its treatment, and opportunities for future applications. Moreover, we discuss the developments in cancer biology that are needed to better utilize the full potential of game-theoretic models. Ultimately, we demonstrate that viewing tumors with evolutionary game theory has medically useful implications that can inform and create a lockstep between empirical findings and mathematical modeling. We suggest that cancer progression is an evolutionary competition between different cell types and therefore needs to be viewed as an evolutionary game.
Solving Strongly Convex and Smooth Stackelberg Games Without Modeling the Follower

Abstract:Stackelberg games have been widely used to model interactive decision-making problems in a variety of domains such as energy systems, transportation, cybersecurity, and human-robot interaction. However, existing algorithms for solving Stackelberg games often require knowledge of the follower's cost function or learning dynamics and may also require the follower to provide an exact best response, which can be difficult to obtain in practice. To circumvent this difficulty, we develop an algorithm that does not require knowledge of the follower's cost function or an exact best response, making it more applicable to real-world scenarios. Specifically, our algorithm only requires the follower to provide an approximately optimal action in response to the leader's action. The inexact best response is used in computing an approximate gradient of the leader's objective function, with which zeroth-order bilevel optimization can be applied to obtain an optimal action for the leader. Our algorithm is proved to converge at a linear rate to a neighborhood of the optimal point when the leader's cost function under the follower's best response is strongly convex and smooth.

Abstract:Optimizing strategic decisions (a.k.a. computing equilibrium) is key to the success of many non-cooperative multi-agent applications. However, in many real-world situations, we may face the exact opposite of this game-theoretic problem -- instead of prescribing equilibrium of a given game, we may directly observe the agents' equilibrium behaviors but want to infer the underlying parameters of an unknown game. This research question, also known as inverse game theory, has been studied in multiple recent works in the context of Stackelberg games. Unfortunately, existing works exhibit quite negative results, showing statistical hardness and computational hardness, assuming follower's perfectly rational behaviors. Our work relaxes the perfect rationality agent assumption to the classic quantal response model, a more realistic behavior model of bounded rationality. Interestingly, we show that the smooth property brought by such bounded rationality model actually leads to provably more efficient learning of the follower utility parameters in general Stackelberg games. Systematic empirical experiments on synthesized games confirm our theoretical results and further suggest its robustness beyond the strict quantal response model.
Efficient Stackelberg Strategies for Finitely Repeated Games

Abstract:We study Stackelberg equilibria in finitely repeated games, where the leader commits to a strategy that picks actions in each round and can be adaptive to the history of play (i.e. they commit to an algorithm). In particular, we study static repeated games with no discounting. We give efficient algorithms for finding approximate Stackelberg equilibria in this setting, along with rates of convergence depending on the time horizon $T$. In many cases, these algorithms allow the leader to do much better on average than they can in the single-round Stackelberg. We give two algorithms, one computing strategies with an optimal $\frac{1}{T}$ rate at the expense of an exponential dependence on the number of actions, and another (randomized) approach computing strategies with no dependence on the number of actions but a worse dependence on $T$ of $\frac{1}{T^{0.25}}$. Both algorithms build upon a linear program to produce simple automata leader strategies and induce corresponding automata best-responses for the follower. We complement these results by showing that approximating the Stackelberg value in three-player finite-horizon repeated games is a computationally hard problem via a reduction from balanced vertex cover.

Zero-Sum Stochastic Stackelberg Games

Abstract:Zero-sum stochastic games have found important applications in a variety of fields, from machine learning to economics. Work on this model has primarily focused on the computation of Nash equilibrium due to its effectiveness in solving adversarial board and video games. Unfortunately, a Nash equilibrium is not guaranteed to exist in zero-sum stochastic games when the payoffs at each state are not convex-concave in the players' actions. A Stackelberg equilibrium, however, is guaranteed to exist. Consequently, in this paper, we study zero-sum stochastic Stackelberg games. Going beyond known existence results for (non-stationary) Stackelberg equilibria, we prove the existence of recursive (i.e., Markov perfect) Stackelberg equilibria (recSE) in these games, provide necessary and sufficient conditions for a policy profile to be a recSE, and show that recSE can be computed in (weakly) polynomial time via value iteration. Finally, we show that zero-sum stochastic Stackelberg games can model the problem of pricing and allocating goods across agents and time. More specifically, we propose a zero-sum stochastic Stackelberg game whose recSE correspond to the recursive competitive equilibria of a large class of stochastic Fisher markets. We close with a series of experiments that showcase how our methodology can be used to solve the consumption-savings problem in stochastic Fisher markets.

An Optimal Control Strategy for Mathematically Modeling Cancer Combination Therapy

Abstract:While the use of combination therapy is increasing in prevalence for cancer treatment, it is often difficult to predict the exact interactions between different treatment forms, and their synergistic/antagonistic effects on patient health and therapy outcome. In this research, a system of ordinary differential equations is constructed to model nonlinear dynamics between tumor cells, immune cells, and three forms of therapy: chemotherapy, immunotherapy, and radiotherapy. This model is then used to generate optimized combination therapy plans using optimal control theory. In-silico experiments are conducted to simulate the response of the patient model to various treatment plans. This is the first mathematical model in current literature to introduce radiotherapy as an option alongside immuno- and chemotherapy, permitting more flexible and effective treatment plans that reflect modern therapeutic approaches.

Monte Carlo Optimization for Solving Multilevel Stackelberg Games

Abstract:Stackelberg games originate where there are market leaders and followers, and the actions of leaders influence the behavior of the followers. Mathematical modelling of such games results in what's called a Bilevel Optimization problem. There is an entire area of research dedicated to analyzing and solving Bilevel Optimization problems which are often complex, and finding solutions for such problems is known to be NP-Hard. A generalization of Stackelberg games is a Multilevel Stackelberg game where we may have nested leaders and followers, such that a follower is, in turn, a leader for all lower-level players. These problems are much more difficult to solve, and existing solution approaches typically require extensive cooperation between the players (which generally can't be assumed) or make restrictive assumptions about the structure of the problem. In this paper, we present a stochastic algorithm to approximate the local equilibrium solutions for these Multilevel games. We then construct a few examples of such Multilevel problems, including: a) a nested toll-setting problem; and b) an adversarial initial condition determination problem for Robust Trajectory Optimization. We test our algorithm on our constructed problems as well as some trilevel problems from the literature, and show that it is able to approximate the optimum solutions for these problems within a reasonable error margin. We also provide an asymptotic proof for the convergence of the algorithm and empirically analyze its accuracy and convergence speed for different parameters. Lastly, we compare it with existing solution strategies from the literature and demonstrate that it outperforms them.

Stackelberg vs. Nash in the Lottery Colonel Blotto Game

Abstract:Resource competition problems are often modeled using Colonel Blotto games. However, Colonel Blotto games only simulate scenarios where players act simultaneously. In many real-life scenarios, competition is sequential, such as cybersecurity, cloud services, Web investments, and more. To model such sequential competition, we model the Lottery Colonel Blotto game as a Stackelberg game. We solve the Stackelberg equilibrium in the Lottery Colonel Blotto game in which the first mover's strategy is actually a solution to a bi-level optimization problem. We develop a constructive method that allows for a series of game reductions. This method enables us to compute the leader's optimal commitment strategy in a polynomial number of iterations. Furthermore, we identify the conditions under which the Stackelberg equilibrium aligns with the Nash equilibria. Finally, we show that by making the optimal first move, the leader can improve utility by an infinite factor compared to its utility in the Nash equilibria. We find that the player with a smaller budget has a greater incentive to become the leader in the game. Surprisingly, even when the leader adopts the optimal commitment strategy, the follower's utility may improve compared to that in Nash equilibria.

First, the mathematical model covering immune cells and tumor cells considering the effects of chemotherapy drugs and immune agents. Second, the bounded optimal control problem covering is transformed into solving Hamilton-Jacobi-Bellman (HJB) equation considering the actual constraints and infinite-horizon performance index based on minimizing the amount of medication administered. Finally, approximate optimal control strategy is acquired through iterative-dual heuristic dynamic programming (I-DHP) algorithm avoiding dimensional disaster effectively and providing optimal treatment scheme for clinical applications.
The prisoner’s dilemma as a cancer model

Abstract:Tumor development is an evolutionary process in which a heterogeneous population of cells with different growth capabilities compete for resources in order to gain a proliferative advantage. What are the minimal ingredients needed to recreate some of the emergent features of such a developing complex ecosystem? What is a tumor doing before we can detect it? We outline a mathematical model, driven by a stochastic Moran process, in which cancer cells and healthy cells compete for dominance in the population. Each are assigned payoffs according to a prisoner’s dilemma evolutionary game where the healthy cells are the cooperators and the cancer cells are the defectors. With point mutational dynamics, heredity, and a fitness landscape controlling birth and death rates, natural selection acts on the cell population and simulated ‘cancer-like’ features emerge, such as Gompertzian tumor growth driven by heterogeneity, the log-kill law which (linearly) relates therapeutic dose density to the (log) probability of cancer cell survival, and the Norton–Simon hypothesis which (linearly) relates tumor regression rates to tumor growth rates. Optimizing adaptive cancer therapy: dynamic programming and evolutionary game theory

Abstract:Recent clinical trials have shown that the adaptive drug therapy can be more efficient than a standard MTD-based policy in treatment of cancer patients. The adaptive therapy paradigm is not based on a preset schedule; instead, the doses are administered based on the current state of tumor. But the adaptive treatment policies examined so far have been largely ad hoc. In this paper we propose a method for systematically optimizing the rules of adaptive policies based on an Evolutionary Game Theory model of cancer dynamics. Given a set of treatment objectives, we use the framework of dynamic programming to find the optimal treatment strategies. In particular, we optimize the total drug usage and time to recovery by solving a Hamilton-Jacobi-Bellman equation based on a mathematical model of tumor evolution. We compare adaptive/optimal treatment strategy with MTD-based treatment policy. We show that optimal treatment strategies can dramatically decrease the total amount of drugs prescribed as well as increase the fraction of initial tumour states from which the recovery is possible. We also examine the optimization trade-offs between the total administered drugs and recovery time. The adaptive therapy combined with optimal control theory is a promising concept in the cancer treatment and should be integrated into clinical trial design.

Convex-Concave Zero-sum Markov Stackelberg Games

Abstract:Zero-sum Markov Stackelberg games can be used to model myriad problems, in domains ranging from economics to human robot interaction. In this paper, we develop policy gradient methods that solve these games in continuous state and action settings using noisy gradient estimates computed from observed trajectories of play. When the games are convex-concave, we prove that our algorithms converge to Stackelberg equilibrium in polynomial time. We also show that reach-avoid problems are naturally modeled as convex-concave zero-sum Markov Stackelberg games, and that Stackelberg equilibrium policies are more effective than their Nash counterparts in these problems.

Abstract:Stackelberg evolutionary game (SEG) theory combines classical and evolutionary game theory to frame interactions between a rational leader and evolving followers. In some of these interactions, the leader wants to preserve the evolving system (e.g. fisheries management), while in others, they try to drive the system to extinction (e.g. pest control). Often the worst strategy for the leader is to adopt a constant aggressive strategy (e.g. overfishing in fisheries management or maximum tolerable dose in cancer treatment). Taking into account the ecological dynamics typically leads to better outcomes for the leader and corresponds to the Nash equilibria in game-theoretic terms. However, the leader's most profitable strategy is to anticipate and steer the eco-evolutionary dynamics, leading to the Stackelberg equilibrium of the game. We show how our results have the potential to help in fields where humans try to bring an evolutionary system into the desired outcome, such as, among others, fisheries management, pest management and cancer treatment. Finally, we discuss limitations and opportunities for applying SEGs to improve the management of evolving biological systems. How to use transcriptomic data for game-theoretic modeling of treatment-induced resistance in cancer cells? A case study in patient-derived glioblastoma organoids

Abstract:Game theory is a powerful theory to model strategic decision-making, but also complex biological systems, such as cancer. In the past decades, game-theoretical models have helped to understand cancer, its response to various treatments, and to design better therapies. However, to fully utilize the potential of game-theoretical modelling in designing better anti-cancer therapies, we need more information on cancer population (ecological) and strategy (evolutionary) dynamics in response to treatment. Here we explore how transcriptomics data can be utilized as inputs of game-theoretic models for predicting evolutionary response to irradiation using patient-derived glioblastoma organoids. For that purpose, we utilize both supervised and unsupervised methods to disentangle cell mixtures from publicly available datasets and identify proportions of relevant cancer cell types in patient-derived glioblastoma organoids treated with radiotherapy. We then fit these proportions to the replicator dynamics, the most classic evolutionary game dynamics, to predict both transient evolutionary dynamics and evolutionary stable cell proportions in these organoids. Hereby we demonstrate a methodology that can be utilized to design evolutionary therapies, i.e. therapies that anticipate and prevent evolution of therapy resistance in cancer cells. Our predictions in glioblastoma suggest that hypoxia is the most important factor in identifying short-term response to irradiation, while this seems much less relevant for the long-term response. Further, we conclude that supervised methods are the best way to estimate cancer evolutionary dynamics when therapy resistance is a qualitative evolutionary trait. We provide a pipeline for designing better therapies through testing evolutionary responses in patient-derived organoids, while in parallel the ecological response can be tracked through serum biomarkers and imaging in the corresponding patients.

Algorithmic asymptotic analysis: extending the arsenal of cancer immunology modeling

Abstract:The recent advances in cancer immunotherapy boosted the development of tumor-immune system models aiming to provide mechanistic understanding and indicate more efficient treatment regimes. However, the complexity of such models, their multi-scale dynamics and their overparameterized character renders them inaccessible for wide utilization. In this work, the dynamics of a fundamental model formulating the interactions of tumor cells with natural killer cells, CD8$^+$ T cells and circulating lymphocytes is examined. It is first shown that the long-term evolution of the system towards high-tumor or tumor-free equilibria is determined by the dynamics of an initial \emph{explosive stage} of tumor progression. Focusing on this stage, the algorithmic Computational Singular Perturbation methodology is employed to identify the underlying mechanisms confining the system's evolution towards the equilibrium and the governing slow dynamics along them. It is shown that these insights are preserved along different tumor-immune system and patient-dependent realizations. Utilizing the obtained mechanistic understanding, a novel reduced model is constructed in an algorithmic fashion, which accurately predicts the dynamics of the system during the explosive stage and includes half of the parameters of the detailed model.
