Abstract:In a real-world environment, the brain must integrate information from multiple sensory modalities, including the auditory and olfactory systems. However, little is known about the neuronal circuits governing how odors influence and modulate sound processing. Here, we investigated the mechanisms underlying auditory-olfactory integration using anatomical, electrophysiological, and optogenetic approaches, focusing on the auditory cortex as a key locus for cross-modal integration. First, retrograde and anterograde viral tracing strategies revealed a direct projection from the piriform cortex to the auditory cortex. Next, using electrophysiological recordings of neuronal activity in the auditory cortex of awake male or female mice, we found that odors modulate auditory cortical responses to sound. Finally, we used optogenetic manipulations during electrophysiology to demonstrate that olfactory modulation in auditory cortex, specifically, odor-driven enhancement of sound responses, depends on direct input from the piriform cortex. Together, our results identify a novel role of piriform-to-auditory cortical circuitry in shaping olfactory modulation in the auditory cortex, shedding new light on the neuronal mechanisms underlying auditory-olfactory integration.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is to understand how the brain integrates auditory and olfactory information, especially in the auditory cortex, how olfactory information affects and modulates sound processing. Specifically, through anatomical, electrophysiological and optogenetic methods, the researchers explored the role of the direct projection from the piriform cortex to the auditory cortex in auditory - olfactory integration. They aimed to reveal the specific function of this neural pathway and its importance in multisensory information integration. ### Main research questions: 1. **Does a direct projection from the piriform cortex to the auditory cortex exist?** - Using retrograde and anterograde virus - tracing techniques, the researchers confirmed the direct projection from the piriform cortex (Pir) to the auditory cortex (ACx), and found that this projection mainly comes from the posterior part of the piriform cortex (pPir). 2. **How does olfactory information modulate the activity of the auditory cortex?** - By performing electrophysiological recordings in awake mice, the researchers found that olfactory stimuli can modulate the auditory cortex's response to sound. 3. **Is the direct projection from the piriform cortex to the auditory cortex involved in olfactory modulation?** - Using optogenetic techniques, the researchers further confirmed that the enhancing effect of olfactory modulation in the auditory cortex depends on the direct input from the piriform cortex. ### Research background: - **The importance of multisensory integration**: In the real - world environment, the brain must integrate information from multiple sensory modalities to optimize decision - making and behavior. The integration of the auditory and olfactory systems is crucial for animals and humans to perceive and interact with complex environments. - **Limitations of existing research**: Although some studies have shown that there are interactions between the auditory and olfactory systems, the specific neural pathways and mechanisms of these interactions remain unclear. ### Research methods: - **Retrograde and anterograde virus - tracing**: Used to confirm the direct projection from the piriform cortex to the auditory cortex. - **Electrophysiological recording**: Record the neural activity of the auditory cortex in awake mice to observe the influence of olfactory stimuli on auditory responses. - **Optogenetic manipulation**: Through optogenetic techniques, specifically activate or inhibit the projection from the piriform cortex to the auditory cortex to verify its role in olfactory modulation. ### Main findings: - **Direct projection from the piriform cortex to the auditory cortex**: Confirmed the direct projection from the piriform cortex to the auditory cortex, mainly from the posterior part of the piriform cortex. - **Olfactory modulation of auditory responses**: Olfactory stimuli can modulate the auditory cortex's response to sound, especially enhancing the response to certain sounds. - **Optogenetic verification**: Through optogenetic techniques, it was confirmed that the enhancing effect of olfactory modulation depends on the direct input from the piriform cortex to the auditory cortex. In conclusion, this study revealed the important role of the direct projection from the piriform cortex to the auditory cortex in auditory - olfactory integration, providing new insights into understanding the neural mechanisms of multisensory information integration.
