Abstract:Seasonal spermatogenesis in fish is driven by spermatogonial stem cells (SSCs), which undergo a complex cellular process to differentiate into mature sperm. In this study, we characterized spermatogenesis in the large yellow croaker (Larimichthys crocea), a marine fish of significant commercial value, based on a high-resolution single-cell RNA-seq atlas of testicular cells from three distinct developmental stages- juvenile, adult differentiating and regressed testes. We detailed continuous developmental trajectory of spermatogenic cells, from spermatogonia to spermatids, elucidating the molecular events involved in spermatogenesis. We uncovered dynamic heterogeneity in cellular compositions throughout the annual reproductive cycle, accompanied by strong molecular signatures within specific testicular cells. Notably, we identified a distinct population of SSCs and observed a critical metabolic transition from glycolysis to oxidative phosphorylation, enhancing our understanding of the biochemical and molecular characteristics of SSCs. Additionally, we elucidated the interactions between somatic cells and spermatogonia, illuminating the mechanisms that regulate SSCs development. Overall, this work enhances our understanding of spermatogenesis in seasonal breeding teleost and provides essential insights for the further conservation and culture of SSCs.

What problem does this paper attempt to address?
The main problem that this paper attempts to solve is to deeply understand the development process of spermatogonial stem cells (SSCs) and their dynamic heterogeneity in spermatogenesis in the large yellow croaker (*Larimichthys crocea*), a seasonal - breeding fish, through single - cell transcriptome sequencing technology. Specifically, the research aims to: 1. **Reveal the characteristics of spermatogonial stem cells (SSCs)**: Through high - resolution single - cell RNA sequencing (scRNA - seq), identify and characterize testicular cells of the large yellow croaker at different developmental stages, especially the molecular features and states of spermatogonial stem cells (SSCs). 2. **Analyze the dynamic process of spermatogenesis**: Describe in detail the continuous development trajectory from spermatogonia to sperm, as well as the molecular events involved in this process, especially the cell composition changes at different stages of the reproductive cycle and the strong molecular signals within specific testicular cells. 3. **Discover metabolic transitions**: Observe the key metabolic transition of spermatogonial stem cells (SSCs) from glycolysis to oxidative phosphorylation, which helps to understand the biochemical and molecular characteristics of SSCs. 4. **Explore the interactions between somatic cells and spermatogonia**: Elucidate the interaction mechanisms between somatic cells (such as Sertoli cells and Leydig cells) and spermatogonia to regulate the development of SSCs. 5. **Provide a basis for protecting and culturing SSCs**: Through these findings, provide important theoretical bases for the further protection and culturing of fish SSCs, thereby promoting the sustainable aquaculture of the large yellow croaker and the protection of germplasm resources. In summary, this research not only enhances the understanding of the spermatogenesis mechanism in seasonal - breeding fish, but also provides valuable data and insights for the research of fish reproductive biology.
