Abstract:RNA binding proteins have emerged as central players in the mechanisms of many neurodegenerative diseases. In particular, a proteinopathy of fused in sarcoma (FUS) is present in some instances of familial Amyotrophic lateral sclerosis (ALS) and about 10% of sporadic FTLD. Here we establish that focal injection of sonicated human FUS fibrils into brains of mice in which ALS-linked mutant or wild-type human FUS replaces endogenous mouse FUS is sufficient to induce focal cytoplasmic mislocalization and aggregation of mutant and wild-type FUS which with time spreads to distal regions of the brain. Human FUS fibril-induced FUS aggregation in the mouse brain of humanized FUS mice is accelerated by an ALS-causing FUS mutant relative to wild-type human FUS. Injection of sonicated human FUS fibrils does not induce FUS aggregation and subsequent spreading after injection into naive mouse brains containing only mouse FUS, indicating a species barrier to human FUS aggregation and its prion-like spread. Fibril-induced human FUS aggregates recapitulate pathological features of FTLD including increased detergent insolubility of FUS and TAF15 and amyloid-like, cytoplasmic deposits of FUS that accumulate ubiquitin and p62, but not TDP-43. Finally, injection of sonicated FUS fibrils is shown to exacerbate age-dependent cognitive and behavioral deficits from mutant human FUS expression. Thus, focal seeded aggregation of FUS and further propagation through prion-like spread elicits FUS-proteinopathy and FTLD-like disease progression.
Neuroscience
What problem does this paper attempt to address?
The problem that this paper attempts to solve is to explore whether the aggregation of Fused in Sarcoma (FUS) protein and its spread in the nervous system can trigger a disease progression similar to Frontotemporal Dementia (FTLD). Specifically, researchers injected pre - formed FUS fibrils into the brains of mice expressing human FUS protein (including wild - type and ALS - related mutant types) to observe whether these fibrils could induce FUS aggregation and whether this aggregation would spread over time in different regions of the brain. In addition, the study also explored whether this aggregation would lead to neurodegenerative lesions and the impact on the cognitive and behavioral functions of mice. ### Research Background - **FUS Protein**: FUS is an RNA - binding protein, usually located in the nucleus. However, in some patients with familial Amyotrophic Lateral Sclerosis (ALS) and about 10% of sporadic Frontotemporal Dementia (FTLD), FUS will be mis - located to the cytoplasm and form aggregates. - **Prion - like Propagation**: Recent studies have shown that proteins in certain neurodegenerative diseases (such as α - synuclein, tau protein, Aβ - peptide, etc.) can spread between cells in a prion - like manner, leading to the accumulation of pathological proteins and the progression of the disease. ### Research Methods - **Animal Model**: Use a humanized FUS mouse model, in which the mouse FUS gene is replaced by the human FUS gene (wild - type or R521H mutant type). - **Experimental Design**: Inject pre - formed FUS fibrils into the brains of 16 - month - old mice and observe the time - dependent and spatial distribution of FUS aggregation. - **Behavioral Tests**: Evaluate the behavioral and cognitive functions of mice through open - field test, rotarod test and novel object recognition test. ### Main Findings - **Induction of FUS Aggregation**: After injecting FUS fibrils, FUS aggregation can be observed at the injection site, and this aggregation gradually spreads to brain regions far from the injection site over time. - **Pathological Features**: The pathological features caused by FUS aggregation are similar to those of FTLD, including cytoplasmic deposition of FUS, accumulation of ubiquitin and p62, etc. - **Behavioral Impact**: Mice injected with FUS fibrils show an exacerbation of age - dependent cognitive and behavioral deficits. ### Conclusions - **Propagation Mechanism of FUS Aggregation**: Injection of FUS fibrils can induce FUS aggregation, and this aggregation spreads in the nervous system in a prion - like manner, ultimately leading to a disease progression similar to FTLD. - **Clinical Significance**: This finding helps to understand the mechanism of FUS in ALS and FTLD and provides new ideas for the development of treatment strategies for these diseases. Through these studies, the authors have revealed the propagation mechanism of FUS aggregation in the nervous system, providing an important scientific basis for further research and treatment of ALS and FTLD.
