Abstract:Cancer pain is a global public health problem. The mechanism of cancer pain is complex, and opioid analgesics, which are widely used clinically, have obvious addiction and side effects, which seriously affect patients’life functions and may aggravate their anxiety, depression and other negative emotions. Acupuncture has a history of thousands of years in China, and acupuncture analgesia has been confirmed by many studies. This study investigated whether electroacupuncture can alleviate abnormal pain in bone cancer pain (BCP) mouse models and its possible central mechanism. A bone cancer pain model was established by injecting Lewis lung cancer cells into the left femoral cavity of adult male mice. Mechanical paw withdrawal threshold was tested baseline before surgery and 1, 4, 7, 10, 14 and 21 days after surgery. On day 21, behaviours related to depression emotions were tested. After the behaviours, the femurs were removed to observe pathological changes, the neck was broken and brain tissue was collected from the basal lateral amygdala (BLA) area for subsequent Western Blot and ELISA experiments were performed to verify the expression of (stimulator of interferon genes, STING) STING/NF- B pathway proteins and the expression of inflammatory factors. Immunofluorescence of Ionized calcium-binding adapter molecule-1 (Iba-1) and STING in the basal lateral amygdala (BLA) brain region was also performed. The results show that electroacupuncture can increase the pain threshold of the bone cancer pain model and alleviate the depressive-like emotional phenotype. Electroacupuncture inhibited the expression of STING/NF- B pathway proteins, activation of microglia and release of inflammatory factors in the basal lateral amygdala (BLA) area. Therefore, this study shows that electroacupuncture may relieve bone cancer pain by regulating microglial activation and inflammatory factor release through the STING/NF- B pathway.

What problem does this paper attempt to address?
The problem that this paper attempts to solve is the treatment dilemma of Bone Cancer Pain (BCP). Specifically, the researchers explored whether Electroacupuncture (EA) can reduce abnormal pain in a mouse model of bone cancer pain and relieve depression - like emotional behaviors by regulating the STING/NF - κB signaling pathway. ### Research Background and Problems 1. **Complexity of Bone Cancer Pain**: - Bone cancer pain is a global public health problem, and its mechanism is very complex. - Opioid painkillers commonly used in clinical practice have obvious addictiveness and side effects, which seriously affect patients' quality of life and mental health and may lead to negative emotions such as anxiety and depression. 2. **Application of Electroacupuncture Therapy**: - Acupuncture has a history of thousands of years in China, and many studies have shown that acupuncture has an analgesic effect. - Electroacupuncture, as a modern acupuncture technique, has been widely used in clinical practice and has gradually been incorporated into the cancer treatment guidelines of the National Comprehensive Cancer Network (NCCN) in the United States. 3. **Research Objectives**: - To explore whether electroacupuncture can relieve pain sensitivity and depression - like behaviors in a mouse model of bone cancer pain. - To study the effects of electroacupuncture on the STING/NF - κB signaling pathway, microglial cell activation, and inflammatory factor release. ### Main Findings 1. **Analgesic Effect of Electroacupuncture**: - Electroacupuncture treatment significantly increased the mechanical pain threshold of mice in the bone cancer pain model and reduced pain sensitivity. - Electroacupuncture also increased the sucrose consumption of model mice, indicating that it also has an improvement effect on depression - like behaviors. 2. **Mechanism of Electroacupuncture**: - Electroacupuncture inhibited the expression of proteins related to the STING/NF - κB signaling pathway, including STING, P - TBK1, TBK1, P - NF - κB, and NF - κB. - Electroacupuncture reduced microglial cell activation and the release of inflammatory factors in the Basal Lateral Amygdala (BLA) region, especially the level of IL - 1β was significantly reduced. ### Conclusion The results of this study show that electroacupuncture reduces bone cancer pain and depression - like emotions by regulating the STING/NF - κB signaling pathway, inhibiting microglial cell activation, and reducing the release of inflammatory factors. This finding provides new ideas and potential treatment strategies for the treatment of bone cancer pain. ### Formulas and Charts - **Statistical Analysis**: - Data are expressed as mean ± standard error of the mean (mean ± SEM). - An independent - sample t - test was used for two - group comparisons, and a one - way ANOVA was used for comparisons of three or more groups. - If the homogeneity of variance was satisfied, LSD test or Bonferroni correction was used for pairwise comparisons; if not, Tamhane test or non - parametric Kruskal - Wallis test was used for multi - group comparisons. - **Western Blot Analysis**: - Protein samples were separated on a 10% glycine SDS - PAGE gel, with electrophoresis conditions of 80 V for 30 minutes and then adjusted to 110 V for 60 minutes. - Proteins were transferred to a PVDF membrane, washed with TBST solution, and then blocked with Quick Blocking Buffer at room temperature for 30 minutes. - Primary antibodies (such as anti - STING, anti - P - TBK1, anti - TBK1, anti - NF - κB, anti - P - NF - κB, etc.) were added and incubated overnight at 4°C. - After washing, goat anti - rabbit HRP - labeled secondary antibodies were added and incubated at room temperature for 2 hours. - Results were observed and recorded using a chemiluminescence system (ECL). - **Immunofluorescence Staining**: - Mouse hearts were perfused successively with 0.9% NaCl solution and 4% PFA solution, and brain tissues were taken and fixed for 24 hours. - After tissue gradient dehydration, the sections were rinsed with PBS, and Iba - 1 primary antibody was added and incubated overnight at 4°C. - After washing, fluorescent secondary antibodies (such as Rabbit anti - Goat IgG HL(Alex488)) were added and incubated at 37°C for 1 - 2 hours. - After washing again, nuclei were stained with DAPI.
