Abstract:Antibodies are powerful tools for the therapy and diagnosis of various diseases. In addition to conventional hybridoma-based screening, recombinant antibody-based screening has become a common choice; however, its application is hampered by two factors: 1) screening starts only after Ig gene cloning and recombinant antibody production, and 2) the antibody is composed of paired chains, heavy and light, commonly expressed from two independent expression vectors. Here, we introduce a method for the rapid screening of recombinant monoclonal antibodies by establishing a Golden Gate-based dual-expression vector and expression of membrane-bound antibodies. Using this system, we demonstrated the efficient isolation of influenza cross-reactive antibodies with high affinity from mouse germinal center B cells over 4 days. This system is particularly useful for isolating therapeutic or diagnostic antibodies (e.g., during foreseen pandemics).

What problem does this paper attempt to address?
### What problem does this paper attempt to solve? This paper aims to develop a new genotype-phenotype association antibody screening system to rapidly and efficiently screen for high-affinity, broadly reactive antibodies. Specifically, the research team established a dual expression vector based on Golden Gate cloning technology and combined it with an in vivo expression method of membrane-bound antibodies to achieve rapid antibody screening. #### Main Issues: 1. **Limitations of traditional antibody screening methods**: Traditional hybridoma technology and clone screening methods are time-consuming and technically demanding, limiting the discovery of efficient antibodies. 2. **Efficiency issues in recombinant antibody screening**: Recombinant antibody screening requires independent cloning of heavy and light chain genes, followed by co-expression and purification, which is cumbersome and time-consuming. #### Solutions: 1. **Membrane-bound antibody expression system**: By establishing a dual expression vector based on Golden Gate cloning technology, the integrated expression of heavy and light chain genes is achieved, simplifying the screening process. 2. **Screening method compatible with NGS technology**: Utilizing the membrane-bound antibody expression system, the function of the antibody is directly associated with its gene sequence, allowing for rapid enrichment of interesting clones through flow cytometry. 3. **Efficient screening of broadly reactive antibodies**: This system was used to screen for broadly reactive antibodies against the influenza virus in an experimental mouse model, demonstrating its potential in therapeutic and diagnostic applications. ### Conclusion: The new antibody screening system proposed in this study not only simplifies the antibody screening process but also significantly improves screening efficiency, particularly valuable in addressing potential future pandemics.
