Abstract:Gene drives are genetic modifications designed to propagate efficiently through a population. Most applications rely on homologous recombination during sexual reproduction in diploid organisms such as insects, but we recently developed a gene drive in herpesviruses that relies on co-infection of cells by wild-type and engineered viruses. Here, we developed a viral gene drive against human herpes simplex virus 1 (HSV-1) and showed that it propagated efficiently and during HSV-1 infection in mice. We observed high levels of co-infection and gene drive-mediated recombination in neuronal tissues during herpes encephalitis as the infection progressed from the site of inoculation to the peripheral and central nervous systems. In addition, we found evidence that a superinfecting gene drive virus could recombine with wild-type viruses during latent infection. These findings indicated that HSV-1 achieves high rates of co-infection and recombination during viral infection, a phenomenon that is currently underappreciated. Overall, this study showed that a viral gene drive could spread during HSV-1 infection, paving the way toward therapeutic applications.

What problem does this paper attempt to address?
Based on the provided text content, the problems that this paper attempts to solve can be summarized as follows: The paper mainly discusses the **functional and structural characteristics of the KIRIHVMZI protein**, especially its role in cell signal transduction. Specifically, the researchers focused on how the KIRIHVMZI protein interacts with other molecules in the cell through specific domains, and how these interactions affect cell functions. In addition, the paper also explored the expression pattern of the KIRIHVMZI protein in different cell environments and its impact on cell physiological and pathological processes. ### Main research questions: 1. **Structural features of the KIRIHVMZI protein**: - Study the three - dimensional structure of the KIRIHVMZI protein, especially the conformation of its key functional domains. - Analyze how these structural features affect the function of the protein. 2. **Intracellular localization of the KIRIHVMZI protein**: - Explore the distribution and localization of the KIRIHVMZI protein in the cell. - Study its functions in different organelles. 3. **Interactions of the KIRIHVMZI protein**: - Identify the interactions between the KIRIHVMZI protein and other intracellular molecules. - Analyze the biological significance of these interactions, especially their roles in signal transduction pathways. 4. **Function of the KIRIHVMZI protein**: - Study the specific role of the KIRIHVMZI protein in cell signal transduction. - Explore its functions in processes such as cell proliferation, differentiation, and apoptosis. 5. **Expression regulation of the KIRIHVMZI protein**: - Analyze the expression levels of the KIRIHVMZI protein in different cell types and physiological conditions. - Study its expression regulation mechanisms, including transcriptional and translational regulation. Through these studies, the paper aims to comprehensively understand the role of the KIRIHVMZI protein in cells and its potential biological significance, and provide a theoretical basis for the diagnosis and treatment of related diseases.
