Abstract:Polarized vesicular trafficking directs specific receptors and ion channels to cilia, but the underlying mechanisms are poorly understood. Here we describe a role for DLG1, a core component of the Scribble polarity complex, in regulating ciliary protein trafficking in kidney epithelial cells. Conditional knockout of in mouse kidney caused ciliary elongation and cystogenesis, and cell-based proximity labelling proteomics and fluorescence microscopy showed alterations in the ciliary proteome upon loss of DLG1. Specifically, the retromer-associated protein SDCCAG3, IFT20 and polycystin-2 (PC2) were reduced in cilia of DLG1 deficient cells compared to control cells. This phenotype was recapitulated and rescuable by re-expression of wildtype DLG1, but not a Congenital Anomalies of the Kidney and Urinary Tract (CAKUT)-associated DLG1 variant, p.T489R. Finally, biochemical approaches and Alpha Fold modelling suggested that SDCCAG3 and IFT20 form a complex that associates, at least indirectly, with DLG1. Our work identifies a key role for DLG1 in regulating ciliary protein composition and suggests that ciliary dysfunction of the p.T489R DLG1 variant may contribute to CAKUT.

What problem does this paper attempt to address?
The problems that this paper attempts to solve mainly focus on the following aspects: 1. **Explore the role of DLG1 in primary cilia biogenesis and function**: Through conditional knockout of the Dlg1 gene in mouse kidneys and experiments on cultured renal epithelial cells, researchers hope to understand the specific role of DLG1 in regulating cilia length and composition. 2. **Clarify the impact of DLG1 on cilia protein composition in renal epithelial cells**: Using cell - based proximity - labeling proteomics techniques and fluorescence microscopy techniques, researchers aim to reveal how DLG1 deletion affects changes in the cilia proteome, especially the localization and expression level changes of key proteins such as SDCCAG3, IFT20 and polycystin - 2 (PC2) in cilia. 3. **Investigate the subcellular localization of DLG1 in renal epithelial cells and its indirect regulatory mechanism for cilia length**: Research has found that DLG1 is mainly located in the lateral plasma membrane of polarized renal epithelial cells, rather than directly in the cilia - centrosome axis. Therefore, researchers further explore how DLG1 indirectly regulates cilia length through its position in the lateral plasma membrane. 4. **Verify the necessity of DLG1 for the cilia targeting of SDCCAG3 and IFT20**: Through immunofluorescence microscopy analysis and experiments with stable expression of mCherry - DLG1, researchers have verified the important role of DLG1 in promoting the targeting of SDCCAG3 and IFT20 to primary cilia. 5. **Evaluate the role of DLG1 in regulating the cilia targeting of PC2**: Given the known roles of SDCCAG3 and IFT20 in promoting the transport of PC2 to primary cilia, researchers further explore whether DLG1 affects the cilia targeting of PC2 by regulating these proteins. In summary, this paper aims to deeply understand the specific role of DLG1 in primary cilia biogenesis and function, especially the regulatory mechanisms of cilia protein composition and length in renal epithelial cells, and the potential connection between these mechanisms and congenital kidney and urinary tract anomalies (CAKUT).
