Abstract:Rhubarb, a well-known Chinese herbal medicine, is also used in Europe and other places of the world. Anthraquinones derivatives are thought to be the major active components. A pressurized liquid extraction (PLE) and capillary zone electrophoresis (CZE) separation were developed for simultaneous determination of five anthraquinones including aloe-emodin, emodin, chrysophanol, physcion, and rhein in Rhubarb. The effects of the experimental variables on PLE and CZE have been optimized. The optimum conditions of PLE were: solvent, methanol; temperature, 140 degrees C; particle size, 0.13-0.2 mm; static extraction time, 5 min; pressure, 1500 psi; and one extraction. The best separation of the five anthraquinones could be obtained using 50 mm borate buffer (pH 8.2) containing 25% isopropyl alcohol and 25% acetontrile as modifier, while the separation voltage was 25 kV and the temperature was at 20 degrees C. The method developed is accurate, simple, and reproducible, and could be used for quality control of Rhubarb and its medical preparations.

Abstract:Abstract Rheum emodi is a perennial herb and an important medicinal plant, with anthraquinones and flavonoids as its main bioactive compounds. However, there is little knowledge about the biosynthetic pathway of anthraquinones in rhubarbs. In this study, we qualitatively and quantitatively assessed 62 pharmacological metabolites in rhubarb using dynamic multiple reaction monitoring (dMRM) of triple-quadrupole mass spectrometry (QqQ-MS), including 21 anthraquinones, 17 flavonoids, 6 stilbenes, 12 gallate esters, 3 tannins, and 3 others. Besides, the metabolomics results showed significant differences among all the 60 metabolites, except for gallic acid and piceatannol- O -β-glucoside. The combined transcriptome data of R. palmatum L . (RPL) and R. officinale Baill. (ROB) showed that 21,691 unigenes were annotated in the metabolic pathways. Taken together, 17 differentially expressed genes (DEGs) were associated with the anthraquinone biosynthetic pathway. Additionally, a significant correlation between anthraquinone peak intensity and DEG expression level existed, validating that DEGs contribute to the anthraquinone biosynthetic pathway. RT-qPCR results showed that the cluster-14354.38156 gene may catalyze the O-methylation of emodin to produce physcion. This study provides a useful resource for further studies on secondary metabolism in rhubarb and the combination analysis of transcriptome and metabolome, which can help with the discovery of enzyme genes involved in metabolite biosynthesis.

[Induction of Hairy Roots and Anthraquinone Production in Rheum Palmatum].

Abstract:OBJECTIVE:To study the induction of hairy roots and anthraquinone production in Rheum palmatum.METHOD:Leaf blades, hypocotyls and petioles of R. palmatum were infected by Agrobacterium rhizogenes LBA9402 and A. rhizogenes R1601, respectively.RESULT:Hairy roots were induced by two strains of A. rhizogenes, and LBA9402 showed stronger infective ability than R1601. Three clones of hairy roots were cultured. DH7a was induced by R1601, DH5a and DH5c were induced by LBA9402. DH7a grew faster than DH5a and DH5c, and all of them grew faster obviously than normal root (NOR). There were significant differences in anthraquinone composition and content among four kinds of roots. Emodin physcion and chrysophanol were predominant anthraquinone in DH5a, DH5c and NOR respectivly. Aloe-emodin content was the lowest in all root cultures compared with other four anthraquinones.CONCLUSION:The in vitro culture system of the established hairy roots laid a foundation for mass production of anthraquinone by hairy root culture.
[Anthraquinone Production and Analysis in the Hairy Root Cultures of Rheum Palmatum L].

Abstract:Hairy root culture of the medicinal plant Rheum palmatum L. was established by genetic transformation with Agrobacterium rhizogenes. The effects of various media with different pH on growth of the hairy roots and biosynthesis of free anthraquinones were investigated. The experimental results showed that MS agar medium with pH 5.5-5.8 is suitable for growth of the hairy roots. Dark condition is favourable and 62.5-fold increase in fresh weight was reached within a culture period of 25 days. Auxin (0.1 mg.L-1 IAA) activated the hairy root growth but inhibited the biosynthesis of free anthraquinones. About 28% of the total free anthraquinones was released into the liquid medium from the rhubarb hairy roots.
Evaluation of anti-diabetic and alpha glucosidase inhibitory action of anthraquinones from Rheum emodi

Abstract:Rheum emodi is used as a culinary plant across the world and finds an eminent role in the Ayurvedic and traditional Chinese systems of medicine. The plant is known to principally contain 1,8-dihydroxyanthraquinones (DHAQs) like rhein, aloe emodin, emodin, chrysophanol and physcion that possess diverse pharmacological and therapeutic actions. The present work deals with developing a platform technology for isolation of these DHAQs and evaluating their anti-diabetic potential. Herein, we report the anti-hyperglycemic activity and alpha glucosidase (AG) inhibitory actions of five isolated DHAQs from R. emodi. All the five isolated DHAQs showed good anti-hyperglycemic activity with aloe emodin exhibiting maximum lowering of blood glucose in an oral glucose tolerance test. However, on evaluation of the AG inhibitory potential of the DHAQs only emodin exhibited potent intestinal AG inhibition (93 ± 2.16%) with an IC50 notably lower than acarbose. Subsequent kinetic studies indicated a mixed type of inhibition for emodin. In vivo studies using oral maltose load showed almost total inhibition for emodin when compared to acarbose. Molecular docking studies revealed the presence of an allosteric topographically distinct 'quinone binding site' and showed that interaction with Ser 74 occurs exclusively with emodin, which is vital for AG inhibition. The net benefit from the glucose lowering effect and mixed type inhibition by emodin would enable the administration of a small dosage that is safe and non-toxic in the case of prolonged use in treating diabetes.
Comparison of three different extraction methods and HPLC determination of the anthraquinones aloe-emodine, emodine, rheine, chrysophanol and physcione in the bark of Rhamnus alpinus L. (Rhamnaceae)

Abstract:Introduction: Rhamnus alpinus L. (Rhamnaceae), a traditional plants in the flora of the Abruzzo region, is known to contain active anthraquinone secondary metabolites. However, the content of anthraquinones varies among R. alpinus samples depending on collection season and site. Thus, using simple, reliable and accurate analytical methods for the determination of anthraquinones in R. alpinus extracts allows comparative study of different methods of extraction. Objective: After a partial validation of an HPLC method for the simultaneous determination of five anthraquinones, aloe-emodine, rheine, emodine, chrysophanol and physcione, in the bark of R. alpinus, we compared three different methods of extraction. Methodology: Anthraquinones were extracted from the bark of R. alpinus using different techniques (methanol maceration, ultrasonic and supercritical CO(2) extraction). Separation and quantification of anthraquinones were accomplished using a reversed-phase C(18) column with the mobile phase of H(2)O-methanol (40 : 60, v/v, 1% formic acid) at a wavelength of 254 nm. The qualitative analyses were also achieved at wavelength of 435 nm. Results: All calibration curves were linear over the concentration range tested (10-200 mM) with the determination coefficients >or=0.991. The detection limits (S/N = 3) were 5 mM for each analytes. All five anthraquinones were found in the samples tested at concentrations reported in experimental data. Conclusion: The described HPLC method and optimised extraction procedure are simple, accurate and selective for separation and quantification of anthraquinones in the bark of R. alpinus and allow evaluation of the best extraction procedure between the tested assays.

Abstract:Rheum officinale Baill. is an important traditional Chinese medicinal herb, its dried roots and rhizomes being widely utilized to cure diverse diseases. However, previous studies mainly focused on the active compounds and their pharmacological effects, and the molecular mechanism underlying the biosynthesis of these ingredients in R. officinale is still elusive. Here, we performed comparative transcriptome analyses to elucidate the differentially expressed genes (DEGs) in the root, stem, and leaf of R. officinale. A total of 236,031 unigenes with N50 of 769 bp was generated, 136,329 (57.76%) of which were annotated. A total of 5884 DEGs was identified after the comparative analyses of different tissues; 175 and 126 key enzyme genes with tissue-specific expression were found in the anthraquinone, catechin/gallic acid biosynthetic pathway, respectively, and some of these key enzyme genes were verified by qRT-PCR. The phylogeny of the PKS III family in Polygonaceae indicated that probably only PL_741 PKSIII1, PL_11549 PKSIII5, and PL_101745 PKSIII6 encoded PKSIII in the polyketide pathway. These results will shed light on the molecular basis of the tissue-specific accumulation and regulation of secondary metabolites in R. officinale, and lay a foundation for the future genetic diversity, molecular assisted breeding, and germplasm resource improvement of this essential medicinal plant.

Abstract:Anthraquinones exhibit a significant group of natural and synthetic quinone derivatives because of their biological activities and industrial applications. Rhamnaceae is one of the families known to contain different kinds of anthraquinones. In this study, it was aimed to quantify rhein, emodin, chrysophanol and physcion in fruits of Rhamnus petiolaris Boiss. & Balansa belonging to Rhamnaceae by solid phase extraction and high performance liquid chromatography with ultraviolet detection. The anthraquinones were separated using a C18 analytical column. Gradient elution was performed using a mobile phase consisted of 0.1% o-phosphoric acid solution and methanol. Analytes were detected at 254 nm. Calibration curves were prepared in the range of 0.25-5.00 μg/mL for rhein, chrysophanol, physcion, 1.00-50.00 μg/mL for emodin. Limits of detection and quantification were between 0.07-0.11 and 0.20-0.34 μg/mL, respectively. Relative standard deviations were ≤ 5.78% in repeatability and intermediate precision studies. Accuracy was determined as relative mean error (8.17-12.06%). Extraction was achieved by maceration with acetone and ethanol, followed by hydrophilic-lipophilic balance solid phase extraction. Recoveries were between 96.2 and 109.6%. The developed and validated method was successfully performed to quantify rhein, emodin, chrysophanol and physcion in R. petiolaris fruit extracts. Only physcion was not detected above limit of detection.

Abstract:High-performance liquid chromatography (HPLC) and capillary electrophoresis (CE) were compared to simultaneously determine and separate 11 anthraquinones from rhubarb, including emodin, chrysophanol, rhein and their glucosides, aloe-emodin, sennoside A, and sennoside B. A UV-diode array detector (DAD) at 254 nm with a gradient elution of acetonitrile/water (method A: 0 min 6:94, 12 min 12:88, 15 min 20:80, 40 min 25:75, 53 min 55:45, 55 min 100:0; method B: 0 min 5:95, 2 min 15:85, 5 min 20:80, 12 min 25:75, 15 min 50:50, 19 min 98:2) at 28(+/-1) degrees C (method A) and 30-60 degrees C (method B) in HPLC or with 0.03 M borate buffer (pH 10.0) containing 25% (v/v) acetonitrile with 0.002 M 2,6-di-O-methyl-beta-cyclodextrin (CD) and 0.005 M alpha-CD in CE effectively detected this separation in 25 min. The detection limits of anthraquinones from rhubarb were in the 0.02-0.2 microg/mL and 0.1-0.8 microg/mL ranges for HPLC and CE, respectively. The established HPLC and CE methods are suitable for quantitative determination of emodin, chrysophanol, aloe-emodin, emodin-1-beta-d-glucoside, emodin-8-beta-d-glucoside, chrysophanol-1-beta-d-glucoside, chrysophanol-8-beta-d-glucoside, and rhein-8-beta-d-glucoside.

Comparative transcriptome analysis and HPLC reveal candidate genes associated with synthesis of bioactive constituents in Rheum palmatum complex

Abstract:Content of bioactive constituents is one of the most important characteristics in Rheum palmatum complex. Increasing ingredient content through genetic breeding is an effective strategy to solve the contradiction between large market demand and resource depletion, but currently hampered by limited understanding of metabolite biosynthesis in rhubarb. In this study, deep transcriptome sequencing was performed to compare roots, stems, and leaves of two Rheum species (PL and ZK) that show different levels of anthraquinone contents. Approximately 0.52 billion clean reads were assembled into 58,782 unigenes, of which around 80% (46,550) were found to be functionally annotated in public databases. Expression patterns of differential unigenes between PL and ZK were thoroughly investigated in different tissues. This led to the identification of various differentially expressed genes (DEGs) involved in shikimate, MEP, MVA, and polyketide pathways, as well as those involved in catechin and gallic acid biosynthesis. Some structural enzyme genes were shown to be significantly up-regulated in roots of ZK with high anthraquinone content, implying potential central roles in anthraquinone synthesis. Taken together, our study provides insights for future functional studies to unravel the mechanisms underlying metabolite biosynthesis in rhubarb. Assessment of the genetic fidelity of true-to-type regenerants of medicinal plant Rheum emodi using RAPD and ISSR molecular markers

Abstract:Rheum emodi commonly known as rhubarb is mainly found in Northern Himalayas. It is a valuable medicinal plant having major pharmacological activities such as antimicrobial, anticancer, antioxidant, anti-inflammatory and is used extensively as purgative, stomachic and astringent tonic and improves gastro related problems. This herb is used by the local communities for medicinal as well as common eating purpose. This leads to its immense decline in its natural habitat and now this herb falls under threatened species and demands conservation. In this prospective, an efficient in vitro propagation method from callus culture has been achieved using leaf explants excised from the juvenile plant of R. emodi. Murashige and Skoog (MS) basal medium was used for regeneration procedure with different concentration of phytohormones. Maximum frequency of callus formation (84.44±0.27%) was observed in MS+36.19μM (2, 4-D) in combination with 11.10μM (BAP). The highest percentage of adventitious shoot regeneration was observed as 75.56±0.27% and the maximum number of shoots per explant that is 3.67±0.27 was achieved on MS basal medium containing BAP (35.5 μM) and Kn (11.61 μM). The maximum frequency of rooting was observed in MS full strength media + IAA (28.55 μM) + BAP (8.88 μM). The highest frequency of roots per shoot was observed as 5.0±0.47 with an average root length of 11±1.25mm. For ascertaining the clonal fidelity, 20 ISSR markers and 15 RAPD markers were assayed and employed to validate the true-to-type regenerants of Rheum emodi. Out of 15 RAPD and 20 ISSR markers, 7 markers and 15 markers produced distinct, clear and scorable bands with an average of 4.5 bands and 4.4 bands per marker respectively among the tissue cultured progenies. For each primer, the banding pattern was uniform and comparable to mother plant and showed about 99% homology. All the markers produce the monomorphic bands and no variation was detected among the micropropagated plants. Thus, the analysis of ISSR and RAPD patterns revealed that the bands were shared by both in vitro raised plants and parent clump confirming the genetic stability. DNA based molecular markers have proved to be versatile tools in diverse fields of biology. These markers proved to be model tools for routine analysis of clonal fidelity of micropropagated plants prior to commercialization.

Abstract:Anthraquinones are phenolic compounds known best for their laxative activity. Rheum australe D. Don (Indian rhubarb, Polygonaceae) is a prominent medicinal plant with anthraquinones as the bioactive compounds, among a few others. The online literature search was carried out to collect data on the phytochemistry and pharmacological activity of anthraquinones, particularly those isolated from R. australe. Anthraquinones are commonly found in the genera of Rheum, Senna, Aloe, Frangula, and Rubia, which can be quantitatively and qualitatively determined using both conventional and advanced analytical methods. The anthraquinones of R. australe were found in free and glycosidic forms, which were best extracted by the microwave-assisted extraction method. Various chromatographic techniques were commonly conducted to isolate the pure compounds. In addition to its laxative activity, anthraquinones of R. australe also showed potential antibacterial, cytotoxic, and antioxidant properties.

Abstract:Objective: To study the chemical constituents in the root of Rheum wittrochii. Method: Chromatography and spectroscopic analysis were used to isolate and elucidate the chemical constituents in the plant. Result: Six compounds, namely chrysophanol, physcion, emodin, chrysophanol-8-O-β-D-glucopyranoside, physcion-8-O-β-D-glucopyranoside and β-sitosterol, have been isolated from petroleum ether and ethyl acetate fractions. Conclusion: All the above-cited compounds were isolated from the title plant for the first time, and anthraquinone compounds are some of the major constituents of R. wittrochii.

Abstract:Rhubarb, a famous traditional Chinese medicine, shows a wide range of physiological activities and pharmacological benefits. Rhubarb anthraquinones are perceived as the pharmacologically active compounds of Rhubarb, and understanding metabolism of them is crucial to assure safety and effectiveness of clinical application. In this study, the pharmacokinetics, tissue distribution and excretion of five rhubarb anthraquinones (aloe-emodin, rhein, emodin, chrysophanol, physcion) were systematically investigated after oral administration of rhubarb extract to rats.An HPLC method was developed and validated for quantitation of five rhubarb anthraquinones in rat plasma, tissues, urine and faeces to investigate the Pharmacokinetic characteristics. The results showed that the proposed method was suitable for the quantification of five anthraquinones in plasma, tissue and excreta samples with satisfactory linear (r > 0.99), precision (<10%) and recovery (85.12-104.20%). The plasma concentration profiles showed a quick absorption with the mean Tmax of 0.42-0.75 h and t1/2 of 6.60-15.11 h for five anthraquinones. The analytes were widely distributed in most of the tissues. Approximately 0.13-10.59% and 28.47-81.14% of five anthraquinones were recovered in urine and faeces within 132 h post-dosing, which indicated the major elimination route was faeces excretion.In summary, this study lays a foundation for elucidating the pharmacokinetic rule of rhubarb anthraquinone and the important data can provide reliable scientific resource for further research.

Determination of anthraquinones in Rhamnus purshiana using high-performance liquid chromatography coupled to diode array detector and simple ultraviolet spectroscopic analysis

Abstract:A new method based on Ultraviolet spectrophotometry was developed and compared with that based on high-performance liquid chromatography for the determination and quantification of anthraquinones in the extracts of Rhamnus purshiana bark. A validated quantitative analysis of cascaroside A, cascaroside B, emodin, and aloe-emodin in these herbal products has been previously performed using high-performance liquid chromatography coupled with a diode array detector. In the high-performance liquid chromatography analysis, all the anthraquinones showed satisfactory regression (r2 > 0.98) within the test ranges, and the recovery was in the range of 94-117%. The limits of detection and quantification were 0.008-0.010 and 0.029-0.035 μg/mL, respectively. Hierarchical cluster analysis and principal component analysis showed differences in the anthraquinones determined from herbal samples. Subsequently, a simple and low-cost ultraviolet spectrophotometric methodology for the quantitative analysis of the same compounds in the extracts was applied, and all the contents were determined. A paired t-test confirmed that there were no significant differences between the two methods. Our results revealed that the developed method is simple and provides the ability to discriminate and control the quality of anthraquinones in herbal products.

Abstract:A sophisticated LC-ESI-MS (n) method was utilized for the analysis of the phenolic compounds in five unofficial RHEUM species for the first time. These species belong to the section deserticola Maxim. of the Polygonaceae family, including Rheum racemiferum Maxim., R. nanum Siev. ex Pall., R. delavayi Franch., R. sublanceolatum C. Y. Cheng et T. C. Kao and R. pumilum Maxim. A total of 101 phenolic compounds, including sennosides, anthraquinones, stilbenes, glucose gallates, naphthalenes, procyanidins and chromones were identified or tentatively characterized based on their UV and mass spectral data from the methanol extracts. These chemical constituents were compared with those of an official species ( R. palmatum l.). The results showed that most sennosides and procyanidin derivatives were only present in R. palmatum rather than in the unofficial species; stilbenes and chromones were only found in the sect. deserticola species; while anthraquinones, glucose gallates and naphthalenes could be detected in all studied species. For anthraquinone glycosides, only emodin O-glucosides were detected in the sect. deserticola species. Moreover, remarkable difference was observed among unofficial species. These results may be useful for the quality control of rhubarb in order to guarantee its clinical effects, and may also be helpful for the further clarification of the taxonomic relationship of the plants in the sect. deserticola.
Effect of Various Media on Quality of Hairy Root Clones of Rheum Palmatum

Abstract:Objective To study the effect of optimized media and clones on growth and anthraquinone yield in hairy root cultures of Rheum palmatum.Methods The four monoclones and four media were optimized statistically for studying the effect on the biomass accumulation and five anthraquinones yield in hairy root cultures of R.palmatum.Results Significant differences in biomass were shown among various clones and media,in which monoclone DH7a was the highest and WP medium was the best.Chrysophanol and physcion were predominant among five free anthraquinones in hairy root cultures of R.palmatum.Notable differences of total five free anthraquinones yield were observed among different clones, in which DH7a was the highest.Reciprocity analysis showed that the highest yield of five free anthraquinones is in the combinations of DH7a with B5 medium.DH5a less and DH5a the least with WP medium,DH7a and B5 medium were predominent for the yield of above five free anthraquinones with exception of DH5c which has no significant difference in four media.Conclusion The study lays the foundation for mass production of anthraquinone by hairy root culture of R.palmatum.
Determination of Anthraquinones in Different Rhubarb and Coptis Xiexin Decoctions by HPLC

Abstract:This is a study to determine 5 main compounds of rhubarb in Rhubarb and Coptidis Xiexin decoctions,using an assay method,in an attempt to compare the concentration of anthraquinones in different compatibility infusions.We developed a HPLC method,using a phenyl column(250mm×4.6mm,5μm)under an ambient temperature,and using gradient elution with mobile phase of methanol-0.01% phosphoric acid and detected at 254 nm,to determine the con- centration of 5 compounds in Rhubarb.The calibration curve of aloe-emodin,rhein,emodin,and chrysophanol indi- cating a good linearity was obtained within the range of 3.15～64.00μg/ml,and 1.70～33.92μg/ml of physcion.The RSD of 5 compounds were less than 2%.The average extraction recovery of the 5 main compounds was 102.1%,with an RSD less than 5%.The compatibility of rhubarb and coptis(scutellaris)has a significant effect on the concentration of anthraquinones.It is concluded that this method has been proved to be accurate and reliable for determining the concentration of anthraquinones in prescriptions containing Radix et rhizoma rhei.The assay results show that an in- creased or reduced scutellaris concentration in Rhubarb and Coptis Xiexin Decoction has a significant effect on the concentration of anthraquinones.

Abstract:1 The coupling of N‐formyl‐methionyl‐leucyl‐phenylalanine (fMet‐Leu‐Phe) receptor stimulation to Ca2+ mobilisation has been investigated in the human neutrophil by measuring the concentration‐effect curves for inositol 1,4,5‐trisphosphate (IP3) formation and Ca2+ mobilisation. 2 fMet‐Leu‐Phe‐dependent mobilisation of intracellular Ca2+ has been monitored in fluo‐3‐loaded human neutrophils by measuring increases in the cytoplasmic free Ca2+ concentration ([Ca2+]i) in the presence of extracellular EGTA. Fluo‐3 was used in preference to fura‐2 because it was found to be more sensitive to the high Ca2+ levels seen in stimulated neutrophils. 3 fMet‐Leu‐Phe induced a rapid mobilisation of intracellular Ca2+ (EC50 = 2.9 ± 0.1 nm) and increased [Ca2+]i to a maximum of 1286 ± 184 nm. 4 The amount of IP3 in fMet‐Leu‐Phe‐stimulated neutrophils was determined by competition with [3H]‐IP3 for a specific IP3 binding protein isolated from bovine adrenocortical microsomes. Basal IP3 levels of 13.3 ± 2.0 pmol per 107 cells were increased nearly 4 fold by maximally effective concentrations of fMet‐Leu‐Phe. 5 The EC50 for the IP3 response (95 ± 18 nm) was much higher than that for mobilisation of intracellular Ca2+, such that only a doubling in the concentration of IP3 was required to fully mobilise intracellular Ca2+. 6 As a result of this relationship IP3 production was more sensitive than Ca2+ mobilisation to inhibition by demethoxyviridin, an inhibitor of phospholipase activation.
