Abstract:Objective To investigate the possibility of superparamagnetic iron-oxide(SPIO) particles as a marker of atherosclerotic plaques detected with 1.5T magnetic resonance imaging system in New Zealand white rabbit model.Methods Model of atherosclerosis were established by injuring the right carotid artery endothelium with balloon and feeding with high lipid diet.Five rabbits(group A) were intravenously injected SPIO;and the other five rabbits(group B) received the same volume of saline injection.The MR imaging was performed at 24,48 and 72 hours after injection,and then,histological study was done.Results Twenty-four hours after the intravenous injection of SPIO,3 rabbits in group A showed signal intensity loss in the right carotid artery wall on T2WI,corresponding to Perl's Prussian blue staining-positive particles in the plaques revealed by histopathological study.T2WI change and Perl's Prussian blue staining-positive particles in the plaques were not observed in any one model in group B.Conclusion It is possible to use superparamagnetic iron-oxide (SPIO) particles as a marker of atherosclerotic plaques detected with 1.5T magnetic resonance imaging system in New Zealand white rabbit model.

Abstract:Objective To investigate the protective effects of left ventricular myocardial perfusion after delivery of acidic fibroblast growth factor (aFGF) in rats with diabetic cardiomyopathy (DCM) by using ultrasound‐targeted microbubble destruction ( UTMD ) with real‐time myocardial contrast echocardiography( RT‐MCE) . Methods Among 64 male SD rats ,52 rats were randomly selected and were induced DCM by streptozotocin through intraperitoneal injecting ,the other rats as normal control group . DCM rats were randomly divided into the DCM model group ,aFGF only group ,SonoVue‐aFGF group and the SonoVue‐aFGF+ UTMD group in this study . The aFGF only group rats were injected with aFGF solution through tail vein ,the SonoVue‐aFGF group were injected with SonoVue‐aFGF solution through tail vein and SonoVue‐aFGF+ UTMD group rats were injected with SonoVue‐aFGF solution through tail vein and using UTMD simultaneously . All rats underwent conventional echocardiography and RT‐MCE exams before and 4 weeks after intervention . Left ventricular ejection fraction ( LVEF) and left ventricular fraction shortening( LVFS ) were measured by conventional echocardiography . The plateau intensity ( A ) ,initial slope of the curve (β) and myocardial blood flow ( A ×β) of left ventricular anterior wall at the papillary muscle level were measured in left ventricular short‐axis view by RT‐MCE . Results Before intervention , LVEF and LVFS in the DCM model group ,aFGF only group ,SonoVue‐aFGF group and the SonoVue‐aFGF+UTMD group were significantly lower than in the normal control group ( P <0 .01) .During 4 weeks after intervention ,LVEF and LVFS in the aFGF only group ,SonoVue‐aFGF group and the SonoVue‐aFGF+UTMD group were slightly increased than those in the DCM model group ,but no statistically significant differences were found ( P > 0 .05) ,however A and A × β in the SonoVue‐aFGF+ UTMD group were significantly increased than those in the DCM model group( P < 0 .01) . Compared with the same group before intervention ,A and A ×βin the SonoVue‐aFGF+UTMD group were higher ( P <0 .05) and these in the DCM model group were lower during four weeks after intervention ( P < 0 .05) . Conclusions Acidic fibroblast growth factor delivered by using UTMD can improve the left ventricular myocardial perfusion in diabetic cardiomyopathy rats .

Abstract:Objective To determine whether low frequency ultrasound mediated microbubbles destruction (UMMD) could inhibit VX2 orthotopic hepatic tumor growth in rabbit models.Methods Twenty-four New Zealand white rabbits were implanted with VX2 tumor in left hepatic lobe to establish a homograft rabbit model of liver neoplasms in situ,which were randomly divided into four groups(6 rabbits in each group):group A (intravenous saline only),group B (intravenous microbubbles only),group C (intravenous saline+ low frequency focused ultrasound exposure),and group D (intravenous microbubbles+low frequency focused ultrasound exposure).After 3 days consecutive treatment,tumor volume(TV),and peak intensity (PI) were monitored by conventional ultrasound and contrast enhanced ultrasound(CEUS) on 0,1,7,14 and 21 days after treatment.The rabbits were euthanized at the end of the experiment.Tumor tissues were evaluated by HE stain.Results The parameters of TV and PI of each tumor had no significant difference among four groups before treatment(all P>0.05).TV had no significant difference among four groups on 1 day after treatment(all P>0.05);PI in group C and group D were significantly lower than those in group A and group B (all P<0.05).TV and PI in group D were significantly smaller than those in other groups on 7,14,21 days after treatment(all P<0.05).There was no statistical difference in TV and PI among group A,group B and group C(all P>0.05).The pathological changes of necrosis tissue,hemorrhagic damage of microvessel and thrombosis were observed in the tumors of group D only,whereas these changes occurred rarely in other groups.Conclusions UMMD can inhibit the growth of VX2 hepatic tumors in rabbits,and be used as a promising novel therapeutic strategy to liver neoplasms.

Abstract:A characteristic feature of atherosclerosis is its diffuse involvement of arteries across the entire human body. Bone marrow cells (BMC) can be simultaneously transferred with therapeutic genes and magnetic resonance (MR) contrast agents prior to their transplantation. Via systemic transplantation, these dual-transferred BMCs can circulate through the entire body and thus function as vehicles to carry genes/contrast agents to multiple atherosclerosis. This study was to evaluate the feasibility of using in vivo MR imaging (MRI) to monitor BMC-mediated interleukin-10 (IL-10) gene therapy of atherosclerosis.For in vitro confirmation, donor mouse BMCs were transduced by IL-10/lentivirus, and then labeled with a T2-MR contrast agent (Feridex). For in vivo validation, atherosclerotic apoE(-/-) mice were intravenously transplanted with IL-10/Feridex-BMCs (Group I, n = 5) and Feridex-BMCs (Group II, n = 5), compared to controls without BMC transplantation (Group III, n = 5). The cell migration to aortic atherosclerotic lesions was monitored in vivo using 3.0T MRI with subsequent histology correlation. To evaluate the therapeutic effect of BMC-mediated IL-10 gene therapy, we statistically compared the normalized wall indexes (NWI) of ascending aortas amongst different mouse groups with various treatments.Of in vitro experiments, simultaneous IL-10 transduction and Feridex labeling of BMCs were successfully achieved, with high cell viability and cell labeling efficiency, as well as IL-10 expression efficiency (≥90%). Of in vivo experiments, MRI of animal groups I and II showed signal voids within the aortic walls due to Feridex-created artifacts from the migrated BMCs in the atherosclerotic plaques, which were confirmed by histology. Histological quantification showed that the mean NWI of group I was significantly lower than those of group II and group III (P<0.05).This study has confirmed the possibility of using MRI to track, in vivo, IL-10/Feridex-BMCs recruited to atherosclerotic lesions, where IL-10 genes function to prevent the progression of atherosclerosis.
Radiofrequency-Enhanced Vascular Gene Transduction and Expression for Intravascular Mr Imaging-Guided Therapy: Feasibility Study in Pigs

Abstract:PURPOSE: To evaluate the feasibility of radiofrequency (RF)-enhanced vascular gene transduction and expression by using a magnetic resonance (MR) imaging heating guidewire as an intravascular heating vehicle during MR imaging-guided therapy.MATERIALS AND METHODS: The institutional committee for animal care and use approved the experimental protocol. The study included in vitro evaluation of the use of RF energy to enhance gene transduction and expression in vascular cells, as well as in vivo validation of the feasibility of intravascular MR imaging-guided RF-enhanced vascular gene transduction and expression in pig arteries. For in vitro experiments, approximately 104 vascular smooth muscle cells were seeded in each of four chambers of a cell culture plate. Next, 1 mL of a green fluorescent protein gene (gfp)-bearing lentivirus was added to each chamber. Chamber 4 was heated at approximately 41 degrees C for 15 minutes by using an MR imaging-heating guidewire connected to a custom RF generator. At day 6 after transduction,,the four chambers were examined and compared at confocal microscopy to determine the efficiency of gfp transduction and expression. For the in vivo experiments, a lentivirus vector bearing a therapeutic gene, vascular endothelial growth factor 165 (VEGF- 165), was transferred by using a gene delivery balloon catheter in 18 femoral-iliac arteries (nine artery pairs) in domestic pigs and Yucatan pigs with atherosclerosis. During gene infusion, one femoral-iliac artery in each pig was heated to approximately 41 degrees C with RF energy transferred via the intravascular MR imaging-heating guidewire, while the contralateral artery was not heated (control condition). At day 6, the 18 arteries were harvested for quantitative Western blot analysis to compare VEGF165 transduction and expression efficiency between RF-heated and nonheated arterial groups.RESULTS: Confocal microscopy showed gfp expression in chamber 4 that was 293% the level of expression in chamber 1 (49.6% +/- 25.8 vs 16.8% +/- 8.0). Results of Western blot analysis showed VEGF-165 expression for normal arteries in the RF-heated group that was 300% the level of expression in the nonheated group (70.4 arbitrary units [au] +/- 107.1 vs 23.5 au +/- 29.8), and, for atherosclerotic arteries in the RF-heated group, 986% the level in the nonheated group (129.2 au +/- 1003 vs 13.1 au +/- 4.9).CONCLUSION: Simultaneous monitoring and enhancement of vascular gene delivery and expression is feasible with the MR imaging-heating guidewire. Inhibited Atherosclerotic Plaque Formation by Local Administration of Magnetically Labeled Endothelial Progenitor Cells (epcs) in a Rabbit Model.

Abstract:Purpose: To investigate whether atherosclerosis can be prevented by magnetically labeled endothelial progenitor cells (EPCs) in rabbits.Materials anti methods: EPCs derived from rabbit periphery blood were labeled with a superparamagnetic iron oxide (SPIO) agent Fe2O3-poly-L-lysine (Fe2O3-PLL). Rabbit atherosclerosis was induced by high-cholesterol-diet following balloon injury via catheterization of right common carotid artery (RCCA). Fe2O3-PLL labeled EPCs (2 x 10(6)) and media were allowed to interact with the RCCA for 25 min in EPC-treated rabbits (n = 14) and control rabbits (n = 7) animals respectively. MRI was performed with a 1.5 T-magnet to measure RCCA signal intensity (SI) and caliber at week 1, 2, 3, 6,12, and 15 with animals euthanized in groups for histopathology.Results: In EPC-treated rabbits, T-2-weighted MRI showed SI loss in RCCA at week I and 2 followed by normalization after week 3. MRI outcomes corresponded well to findings of Prussian blue staining. MRI at week 6,12 and 15 showed little stenosis of RCCA in EPC-treated rabbits, but moderate to severe stenoses in control rabbits. Histology at week 15 revealed significantly thinner RCCA wall (277.62 mu m vs. 382.95 mu m, P= 0.026), greater internal diameter (913.33 mu m vs. 789.64 mu m, P= 0.037) and smaller plaque (398.60 mm(2) vs. 597.70 mm(2), P = 0.047) in EPC-treated rabbits relative to control rabbits.Conclusion: Atherosclerosis at RCCA was inhibited by SPIO-labeled EPCs, which was depicted with a clinical MRI scanner over 2 weeks after cell administration, suggesting that EPCs may play a role in restoration of endothelial injury and prevention of atherosclerosis. Application of Targeted Nano-Bubble Ultrasound Contrast Agent in the Detection of Arterial Intima Inflammation in Rats

Abstract:Tumor-specific antibodies or ligands were connected to the surface of nano-bubbles to form a targeted nano-bubble ultrasound contrast agent (UCA), which can accumulate in tumor tissues, enhance tumor tissue visualization, and realize extravascular disease detection and ultrasound molecular imaging. In this research, the positive and negative charges were attracted to promote the self-assembly connection between the targeted vascular endothelial growth factor (VEGF) antibody and the envelope surface of the nano-bubble, thereby obtaining a tumor-specific targeted nano-bubble UCA. Then, from the basic characterization, in vivo and in vitro ultrasound contrast performance analysis, a rat model of arterial intima inflammation in vivo was constructed. 16 Wistar rats were screened and divided into a control group and a contrast-enhanced ultrasound group. The imaging performance of the targeted molecules was analyzed by preparing an UCA. in vitro contrast-enhanced ultrasound found that the contrast intensity of self-made targeted nano-bubbles was greatly affected by concentration, but there was no linear relationship between the two. in vivo experiments were performed to observe rat liver contrast. The results showed that the contrast intensity and contrast time of the targeted nano-bubbles in vivo were greatly affected by the dose, and the stability in vivo was lower than the stability in vitro . Immunohistochemical tests found that P-selectin was expressed in large amounts in the intima of damaged blood vessels. Compared with ordinary contrast agents, the prepared targeted nano-UCA after modeling can enhance the video intensity of the inner membrane ( P <0.05) and prolong the imaging time ( P <0.05). It suggested that the contrast agent can specifically bind to P-selectin on the surface of vascular endothelial cells, and it was expected to be used for the detection of early inflammatory lesions in atherosclerotic diseases.

Development of an Intrabiliary MR Imaging-monitored Local Agent Delivery Technique: A Feasibility Study in Pigs

Abstract:PURPOSETo investigate the feasibility of using magnetic resonance (MR) imaging to monitor intrabiliary delivery of motexafin gadolinium (MGd) into pig common bile duct (CBD) walls.MATERIALS AND METHODSAnimal studies were approved by the Institutional Animal Care and Use Committee. Initially, human cholangiocarcinoma cells were treated with various concentrations of MGd, a compound serving as a T1-weighted MR imaging contrast agent, chemotherapy drug, and cell marker. These cells were then examined by means of confocal microscopy to confirm the intracellular uptake of MGd. In addition, an MGd/trypan blue mixture was locally infused into CBD walls of six cadaveric pigs using a microporous balloon catheter. CBDs of six pigs were infused with saline to serve as controls. Ex vivo T1-weighted MR imaging of these CBDs was performed. For in vivo technical validation, the microporous balloon catheter was placed in the CBD by means of a transcholecytic access to deliver MGd/trypan blue into CBD walls of six living pigs. T1-weighted images were obtained with both a surface coil and an intrabiliary MR imaging guidewire, and contrast-to-noise ratios of CBD walls before and after MGd/trypan blue infusions were compared in the two groups by means of paired t test, with subsequent histologic analysis to confirm the penetration and distribution of the MGd/trypan blue agent into CBD walls.RESULTSIn vitro experiments confirmed uptake of MGd by human cholangiocarcinoma cells. The ex vivo experiments demonstrated the penetration of MGd/trypan blue into the CBD walls. The in vivo experiment confirmed the uptake of MGd/trypan blue, showing an increased contrast-to-noise ratio for the CBD after administration of the mixture, compared with images obtained prior to MGd/trypan blue administration (11.6 ± 4.2 [standard deviation] vs 5.7 ± 2.8; P = .04). Histologic results depicted the blue dye stains and red fluorescence of MGd in CBD walls, confirming the imaging findings.CONCLUSIONIt is feasible to use MR imaging to monitor the penetration of locally delivered MGd into pig CBD walls.
Microsphere as a contrast agent/gene vector in ultrasound imaging-based vascular gene delivery

Abstract:The purpose of this study was to test the feasibility of using a novel biodegradable microsphere as a contrast agent/gene vector in ultrasound imaging-based vascular gene delivery. We first encapsulated green fluorescent protein (GFP) gene into the microspheres and validated the echogenic property of GFP-plasmid/microspheres with intramuscular injection. Then, the GFP-plasmid/microspheres were transferred into the arteries of pigs via a catheter-based delivery procedure under ultrasound imaging. A strong echogenic signal reflected from the microspheres indicated the destination of genes delivered, whereas functional gene expression was confirmed by immunohistochemical examination. Microspheres can be feasible contrast agents/gene vectors in ultrasound imaging-based vascular gene delivery in vivo.
3.0-T Mr Imaging of Intracoronary Local Delivery of Motexafin Gadolinium into Coronary Artery Walls

Abstract:Purpose: To develop a technique with clinical 3.0-T magnetic resonance (MR) imaging to delineate local contrast agent distribution in coronary artery walls for potential molecular MR imaging-guided local gene or drug therapy of atherosclerotic coronary artery disease.Materials and methods: This animal protocol was approved by the institutional animal care and use committee and was in compliance with the Guide for the Care and Use of Laboratory Animals. For in vitro confirmation, human arterial smooth muscle cells (SMCs) were used to determine capability of SMCs in uptake of motexafin gadolinium (MGd) and its optimal dose. For ex vivo evaluation, a 2-mL mixture of MGd and trypan blue was locally infused into coronary artery walls of six cadaveric pig hearts with MR monitoring and an MR imaging guidewire, surface coils, or both. For in vivo validation, the balloon catheter was placed into coronary arteries of seven living pigs, and the MGd and trypan blue mixture was infused into arterial walls with MR guidance. Signal-to-noise ratio (SNR) and contrast-to-noise ratio (CNR) of coronary artery walls were recorded by using different coils between pre-and postcontrast infusion, with subsequent histologic confirmation. Paired Student t tests were used to compare average SNRs and CNRs of arterial walls before and after contrast agent infusion with different coils.Results: SMCs could take up MGd with the optimal concentration at 150 mu mol/L. Average SNR with the MR imaging guidewire and surface coil combination was significantly higher than that with the MR imaging guidewire only or with surface coils only (P < .05), and average SNR and CNR of postinfusion MR imaging was significantly higher than that of preinfusion MR imaging (P < .05). Histologic analysis was used to confirm successful intracoronary infiltration of MGd and trypan blue within coronary artery walls.Conclusion: MR imaging can be used to delineate locally infused contrast agent distribution in coronary artery walls. Contrast Imaging and Gene Delivery Through the Combined Use of Novel Cationic Liposomal Microbubbles and Ultrasound in Rat Carotid Arteries

Abstract:Introduction Lipid-coated cationic microbubbles represent a new class of agents with both diagnostic and therapeutic applications. The main goal of this study was to evaluate the efficiency of gene transfer through the combined use of microbubbles and ultrasound in rat carotid arteries. Furthermore, we assessed whether the cationic liposomal microbubbles could allow long-term enhanced imaging, comparing with SonoVue®. Material and methods Normal rat carotid arteries were imaged after intravenous bolus injections of 0.5 ml/kg of two contrast agents (SonoVue® and the cationic liposomal microbubbles). Forty Sprague-Dawley rats were divided into 5 groups according to ultrasound parameters and were treated with or without microbubbles. All rats were sacrificed after being transfected for 2 days. The level of protein expression was determined by western blot analysis. Results The enhancing time of self-made microbubbles was much longer than that of SonoVue® in rat carotid arteries (p < 0.05). The results of the western blot analysis revealed that the expression of SR-BI DNA in the carotid artery was highest in the SR-BI + US/CLM group (p < 0.05). Conclusions These results suggest that the novel cationic liposomal microbubbles enhance image quality over a longer period than does SonoVue®. Additionally, the combination of ultrasound and this new type of microbubble can act synergistically to increase SR-BI DNA transfection.
Ultra-Early Diagnosis of Acute Myocardial Infarction in Rats Using Ultrasound Imaging of Hollow Double-Layer Silica Nanospheres

Abstract:Timely diagnosis of acute myocardial infarction (AMI) strongly impacts the survival rate of patients. The authors report the development of a two-shell hollow silica contrast agent useful for ultrasound (US) imaging, which is able to provide ultra-early diagnosis of AMI. To target the characterization of fast blood flow and high blood pressure in the heart, two shells of hollow silica are adopted with opposite polarities, which assemble based on amino and perfluorodecyl silanes. The external amino silane facilitates the attachment of disease-targeted groups, while the internal perfluorodecyl silane provides great US imaging contrast. The material also possesses superior water dispersity, controllable morphology, low toxicity, and biodegradability both in vitro and in vivo, thus promoting its applications in the ultra-early diagnosis of AMI in rats, and is particularly useful for delineation of myocardial necrosis sites.

Abstract:Atherosclerotic disease is a leading cause of morbidity and mortality in developed countries, and oxidized LDL (OxLDL) plays a key role in the formation, rupture, and subsequent thrombus formation in atherosclerotic plaques. In the current study, anti-mouse OxLDL polyclonal antibody and nonspecific IgG antibody were conjugated to polyethylene glycol-coated ultrasmall superparamagnetic iron oxide (USPIO) nanoparticles, and a carotid perivascular collar model in apolipoprotein E-deficient mice was imaged at 7.0 Tesla MRI before contrast administration and at 8 h and 24 h after injection of 30 mg Fe/kg. The results showed MRI signal loss in the carotid atherosclerotic lesions after administration of targeted anti-OxLDL-USPIO at 8 h and 24 h, which is consistent with the presence of the nanoparticles in the lesions. Immunohistochemistry confirmed the colocalization of the OxLDL/macrophages and iron oxide nanoparticles. The nonspecific IgG-USPIO, unconjugated USPIO nanoparticles, and competitive inhibition groups had limited signal changes (p < 0.05). This report shows that anti-OxLDL-USPIO nanoparticles can be used to directly detect OxLDL and image atherosclerotic lesions within 24 h of nanoparticle administration and suggests a strategy for the therapeutic evaluation of atherosclerotic plaques in vivo.
Construction of CNA35 Collagen-Targeted Phase-Changeable Nanoagents for Low-Intensity Focused Ultrasound-Triggered Ultrasound Molecular Imaging of Myocardial Fibrosis in Rabbits

Abstract:Myocardial fibrosis plays an important role in the development of heart failure and malignant arrhythmia, which potentially increases the incidence of sudden cardiac death. Therefore, early detection of myocardial fibrosis is of great significance for evaluating the prognosis of patients and formulating appropriate treatment strategies. Late gadolinium-enhanced magnetic resonance imaging is considered as the currently effective strategy for noninvasive detection of myocardial fibrosis, but it still suffers from some critical issues. In this work, multifunctional CNA35-labeled perfluoropentane nanoparticles (CNA35-PFP NPs) have been elaborately designed and constructed for molecular imaging of fibrotic myocardium based on ultrasound imaging. These as-constructed CNA35-PFP NPs are intravenously infused into rabbit circulation with an animal model of myocardial infarction. Especially, these targeted CNA35-PFP NPs with nanoscale size could efficiently pass through the endothelial cell gap and adhere to the surface of fibroblasts in the fibrotic myocardium. Importantly, followed by low-intensity focused ultrasound irradiation on the myocardium, these intriguing CNA35-PFP NPs could transform from liquid into gaseous microbubbles, which further significantly enhanced the ultrasound contrast in the fibrotic area, facilitating the detection by diagnostic ultrasound imaging. Therefore, this work provides a desirable noninvasive, economical, and real-time imaging technique for the assessment of cardiac fibrosis with diagnostic ultrasound based on the rational design of liquid-to-gas phase-changeable nanoplatforms.

Abstract:Abstract Aims (Ultra) Small superparamagnetic iron oxide nanoparticles, (U)SPIO, are widely used as magnetic resonance imaging contrast media and assumed to be safe for clinical applications in cardiovascular disease. As safety tests largely relied on normolipidaemic models, not fully representative of the clinical setting, we investigated the impact of (U)SPIOs on disease-relevant endpoints in hyperlipidaemic models of atherosclerosis. Methods and results RAW264.7 foam cells, exposed in vitro to ferumoxide (dextran-coated SPIO), ferumoxtran (dextran-coated USPIO), or ferumoxytol [carboxymethyl (CM) dextran-coated USPIO] (all 1 mg Fe/mL) showed increased apoptosis and reactive oxygen species accumulation for ferumoxide and ferumoxtran, whereas ferumoxytol was tolerated well. Pro-apoptotic (TUNEL+) and pro-oxidant activity of ferumoxide (0.3 mg Fe/kg) and ferumoxtran (1 mg Fe/kg) were confirmed in plaque, spleen, and liver of hyperlipidaemic ApoE−/− (n = 9/group) and LDLR−/− (n = 9–16/group) mice that had received single IV injections compared with saline-treated controls. Again, ferumoxytol treatment (1 mg Fe/kg) failed to induce apoptosis or oxidative stress in these tissues. Concomitant antioxidant treatment (EUK-8/EUK-134) largely prevented these effects in vitro (−68%, P < 0.05) and in plaques from LDLR−/− mice (−60%, P < 0.001, n = 8/group). Repeated ferumoxtran injections of LDLR−/− mice with pre-existing atherosclerosis enhanced plaque inflammation and apoptosis but did not alter plaque size. Strikingly, carotid artery plaques of endarterectomy patients who received ferumoxtran (2.6 mg Fe/kg) before surgery (n = 9) also showed five-fold increased apoptosis (18.2 vs. 3.7%, respectively; P = 0.004) compared with controls who did not receive ferumoxtran. Mechanistically, neither coating nor particle size seemed accountable for the observed cytotoxicity of ferumoxide and ferumoxtran. Conclusions Ferumoxide and ferumoxtran, but not ferumoxytol, induced apoptosis of lipid-laden macrophages in human and murine atherosclerosis, potentially impacting disease progression in patients with advanced atherosclerosis.

ICAM-1-carrying targeted nano contrast agent for evaluating inflammatory injury in rabbits with atherosclerosis

Abstract:Abstract To investigate the feasibility of using ICAM-1-targeted nano ultrasonic contrast to evaluate the degree of inflammatory injury at different stages in the abdominal aorta of rabbits with atherosclerosis (AS). Twenty-five experimental rabbits were assigned to five groups: the control group (A); the week-4 after modeling group (B); the week-8 after modeling group (C); the week-12 after modeling group (D); the week-16 after modeling group (E). All groups were given 2D ultrasonography, conventional ultrasonic contrast (SonoVue), and ICAM-1-targeted nano ultrasonic contrast, respectively. Signal intensity of vascular perfusion was evaluated. Signal intensity of ICAM-1-targeted nano ultrasonic contrast was substantially enhanced and prolonged in the vascular wall of the abdominal bubble aorta increased in B, C, D, and E groups (all P < 0.05). A positive linear correlation between intensity and the expression of ICAM-1 (r = 0.895, P < 0.001). The intensity of outer membrane was enhanced from week 4 to week 12, and both the intima-media membrane and outer membrane were enhanced with double-layer parallel echo at week 16, which was in line with the progression of atherosclerotic plaque inflammatory injury. ICAM-1-targeted nano contrast agent would be possibly a novel non-invasive molecular imaging method for plaque inflammatory injury and site high expression of specific adhesion molecules in early atherosclerotic lesions.

Abstract:Objective In this study,the lipid-coated cationic microbubble which can load DNA effectively was prepared.The duration of enhanced imaging and the efficiency of gene transfer on Sprague-Dawley rat carotid artery was assessed,comparing with SonoVue.The purpose of this study is to offer a novel gene carrier,which can lead a long-time enhanced imaging.Methods Cationic liposomal microbubbles were performed using the lyophilization method.Its appearance,mean diameter,PH and zeta potential were detected.The enhancement effects and duration of the cationic liposomal microbubbles and SonoVue on Sprague-Dawley rat carotid artery were compared.Forty Sprague-Dawley rats were divided into 5 groups according to different ultrasound parameters with or without microbubbles: (1) Control group; (2) SR-BⅠ group; (3) SR-BⅠ+US/SonVue group; (4) SR-BⅠ+CLM group; (5) SR-BⅠ+US/CLM group.The US parameters were performed as follows: frequency: 1 MHz,duty: 50%; intensity: 2.0 W/cm2,time: 15 min.All the rats were killed after being transfected for 2 days.Tissue for western blot analysis was obtained from the rats'carotid artery performed by US.Results The mean diameter range of lipid-coated cationic microbubbles was 0.532-5.16 μm and the mean diameter was 1.17 μm.pH of the microbubbles was 7.34, and the zeta potential was (8.10±0.8) mV.Contrast imaging showed that the novel microbubbles had obviously longer duration of enhancement than SonoVue.The results of western blot analysis showed that the expression of SR-BⅠ in the carotid artery was highest in the SR-BⅠ+US/CLM group.Conclusions The self-made cationic liposomal microbubbles loading SR-BⅠ could be prepared easily.Furthermore,these results suggest that the novel cationic liposomal microbubbles had a longer enhancing time than that of SonoVue,and the combination of ultrasound and the novel microbubbles could have a synergistic effect to increase SR-BⅠ transfection.This method could offer a new strategy for gene therapy of atherosclerosis.

Abstract:Objective: To investigate the possibility of improving the delivery of vascular endothelial growth factor (VEGF) gene to the myocardium in rats by using ultrasound-mediated microbubble destruction (UMMD). Methods: Fifteen male Wistar rats underwent left anterior descending coronary artery ligation in this study. The rats were divided into three groups 3 days after ligation. Ultrasound microbubble vectors (UMVs) attaching to pcD2VEGF121 gene were injected into the tail vein of rats with or without simultaneous echocardiographic microbubble destruction in two groups. The third group was used as control group. VEGF protein expression and formation of new blood vessels were evaluated by immunohistochemical technique during autopsy on 15 rats at 2 weeks after gene transformation. Microvascular density (MVD) in the area with myocardial infarction was counted under a microscope. Results: VEGF protein expression and MVD in the ischemic myocardium were higher in the rats receiving UMMD than in the group that did not receive UMMD. Conclusion: UMMD is a noninvasive method to effectively improve the delivery of targeted genes to the heart. (C) 2004 Elsevier Inc. All rights reserved.

Abstract:In this study, we aimed to prepare a neovascularization-relevant inflammatory cytokine-targeted ultrasound contrast agent and apply it in the ultrasound imaging of atherosclerotic plaque. An interleukin-8 (IL-8) monoclonal antibody was conjugated to SonoVue microbubbles using the N-succinimidyl-3-(2-pyridyldithio)propionate cross-linking method. Then, a prepared IL-8-targeted contrast agent was used for contrast-enhanced ultrasound (CEU) to detect rabbit abdominal aorta atherosclerotic plaque and to investigate the imaging characteristics of atherosclerotic plaque with the contrast agent. We found that an IL-8 monoclonal antibody can be successfully coupled to SonoVue microbubbles with stable biological characteristics. CEU with this IL-8-targeted contrast agent can increase the atherosclerotic plaque detection sensitivity, with stronger echo, so that three more plaques were detected compared with using non-targeted SonoVue microbubbles. Thus, an inflammatory cytokine-targeting ultrasound contrast agent carrying IL-8 monoclonal antibody can provide unique advantages for researching the characteristics of atherosclerotic plaque.
