Abstract:We report a synthesis of bicyclo[2.1.1]hexanes via an intramolecular formal (3+2) cycloaddition of allylated cyclopropanes bearing a 4-nitrobenzimine substituent. Both activated and unactivated alkenes are tolerated in the transformation. The bicyclic imine products are prone to photo-induced ring opening, allowing for the epimerization of C5-stereogenic compounds.

Abstract:A new approach to directly access bicyclo[2.1.1]hexanes (BCHs) from easily prepared enol silyl ethers and bicyclo[1.1.0]butanes (BCBs) through formal [2π+2σ] cycloaddition was developed involving two-electron mediated process promoted by commercially available Lewis acids. This new reaction tolerated a wide range of enol silyl ethers and BCBs. In addition, a scale-up experiment and the synthetic transformations of the cycloadducts further highlighted the synthetic utility.

Abstract:A tandem coupling-ketooxygenation reaction of readily accessible enediyne-carboxylic compounds with inner alkynes has been developed that utilizes the PdCl2/CuBr2 catalytic system under an O2 atmosphere and assembles a class of isoindolinones and o-acylbenzoic acids. The two oxygen atoms are regioselectively incorporated into enediyne units at the 1- and 6-positions from atmospheric molecular oxygen and H2O, respectively, during the present process. This study uncovered a formal [4C + 2C] benzannulation-diketonization of enediynes and alkynes via a coupling and decoupling strategy.

Abstract:The first intramolecular (4+3) cycloaddition of pyrroles with epoxy enolsilanes as the electrophiles was developed and used to generate optically-enriched cycloadducts containing the nortropane substructure in good yields. Using this pyrrole cycloaddition as the key step, we achieved the asymmetric synthesis of a nortropane compound bearing the BCDEF ring structure common to the ClassII galbulimima alkaloids.

Abstract:To study the intramolecular Diels-Alder cycloadditon of 2H-chromen-2-one as a diene, a series of chiral N-allyl-N-benzylamides that contain a 2H-chromen-2-one moiety were designed for the synthesis of benzo[f]isoindol-1-ones via an intramolecular Diels-Alder and a subsequent retro-Diels-Alder cycloaddition with the expulsion of CO2. Both the yield (80%-89%) and absolute stereocontrol of the tandem reaction were high when an electron-withdrawing group was attached to the dienophile. The double bond in the styrene substructure remained in the products could be further derivatized by dihydroxylation. Abstract:A highly regio- and diastereoselective visible light-promoted [2 + 2] cycloaddition between readily available 1,4-dihydropyridines and olefins has been developed. This strategy is operationally simple and atom-economical and enables the construction of strained polysubstituted 2-azabicyclo[4.2.0]octanes with three all-carbon quaternary centers with good functional group tolerance. These products can be easily converted to various structurally unique derivatives. The primary mechanistic studies demonstrated that the reaction proceeds through an energy transfer pathway.

Abstract:A metal-free, TfOH (1.1 equiv)-mediated formal cycloisomerization of easily prepared allene-alkynylbenzenes to give pyrrolidines and cyclopentanes derivatives was developed. This reaction is initiated by the generation of allylic cation from allene, followed by alkyne's reaction with the allylic cation, to give a vinyl cation, which is finally intercepted by the triflate (TfO) anion. This cycloisomerization can be further tuned to become an acid-mediated intramolecular formal [4+3] cycloaddition by using 10 equiv of TfOH (The excess acid was used to promote the Friedel-Crafts reaction of the acid mediated cycloisomerization products). The present system can also be applied to synthesized F-incorporated products by using HBF4 or Me3OBF4 as the fluoro source.

Abstract:A cobalt-diphosphine catalyst has been found to promote a formal [3 + 2] cycloaddition reaction between a cyclopropanol and an allene via cyclopropanol ring opening, which affords a 3-alkylidenecyclopentanol derivative with high regio- and diastereoselectivities. The reaction tolerates mono substituted, 1,1-disubstituted, and 1,3-disubstituted allenes and various functional groups. The reaction is proposed to proceed through carbometalation of the allene with a cobalt homoenolate followed by intramolecular carbonyl allylation of the resulting allylcobalt species.

Abstract:The facile intramolecular [2 + 2 + 2] homo-Diels-Alder cycloadditions of oxanorbornadienedicarboxylates and analogues have been investigated by theoretical calculations using B3LYP and M06-2X density functional methods and experimental confirmation. The oxanorbornadienedicarboxylates formed from furans and but-2-ynedioates undergo the resulting intramolecular [2 + 2 + 2] cycloaddition in a concerted but asynchronous fashion, requiring energy barriers of about 30 kcal/mol to construct five- and three-membered rings simultaneously. Bridgehead substitutents have little influence on the regioselectivity, whereas 5-substitutents involving steric hindrance or electron-acceptor groups are predicted to attenuate the cycloaddition at the substituted side. Furthermore, the linker length, unsaturated bonds, and bridge-ring size are very sensitive to the cyclization rate. Additionally, aza- and norbornadienedicarboxylates demonstrate less reactivity, while thionorbornadienedicarboxylates show more reactivity with the challenge of their synthesis. The intermolecular version was also evaluated in comparison with the intramolecular version. Finally, our experimental tests verified the calculational prediction of the regioselectivity and reactivity.

Abstract:Herein, a B(C6F5)3-catalyzed formal (n+3) (n = 5,6) cycloaddition of bicyclo[1.1.0]butanes (BCBs) with imidazolidines/hexahydropyrimidines is described. The reaction provides a modular, atom-economical and efficient strategy to two libraries of synthetically challenging medium-bridged rings, 2,5-diazabicyclo[5.1.1]nonanes and 2,6-diazabicyclo[6.1.1]decanes, in moderate to excellent yields. This reaction also features simple operation, mild reaction conditions and broad substrate scope. A scale-up experiment and various synthetic transformations of products further highlight the synthetic utility. Control experiments support that the reaction mechanism involves a nucleophilic addition of imidazolidines/hexahydropyrimidines to B(C6F5)3-activated BCBs, succeeded by an intramolecular cyclization. As we know, this methodology represents the first high order (n+3) (n > 3) cycloaddition of BCBs to saturated bicyclo[n.1.1]alkanes. We anticipate that this report will promote the exploration of BCB-based high order cycloaddition chemistry to access diverse challenging medium-bridged rings.

Abstract:Palladium and acid cooperative catalysis is presented as a strategy for the [3 + 2] cycloaddition of acceptor/donor-type diazo compounds with terminal alkynes. The [3 + 2] cycloaddition of α-aryl-α-diazoacetates with terminal alkynes proceeded smoothly to produce 2,3,5-trisubstituted furans with high yields. This synthesis method provided a direct and efficient pathway to prepare furan ring-containing organosilane and organoboron reagents. A possible mechanism involving thermoinduced single electron transfer of allylic amines and radical cycloaddition was proposed. Abstract:1H-1,2,3-Triazolium scaffolds are used in a variety of fields including supramolecular chemistry, organometallic chemistry, and organocatalysis. In this study, an oxidative [3 + 2] cycloaddition of triazenes and trisubstituted dichloroalkenes was developed. The reaction proceeded under mild conditions to afford functionalized chloro-1H-1,2,3-triazolium salts. The reaction was successfully performed on a gram scale. Moreover, a chloro-1H-1,2,3-triazolium salt could be employed for nucleophilic aromatic substitution with various nucleophiles.

Abstract:ABSTRACT An efficient approach to functionalized 4,6-disubstituted-and 4,6,6-trisubstituted-1,3-oxazinan-2-ones skeleton has been developed through the reaction of semicyclic N, O-acetals 4a and 4b with 1,1-disubstituted ethylenes 5 or 8. As a result of such a [4 + 2] cycloaddition process, 4,6,6-trisubstituted-1,3-oxazinan-2-ones 6aa, 6af-6au, 7ba, 7bf-7bw and 6,6-spiro containing 1,3-oxazinan-2-ones 9ad, 9ae, 10ba-10bg were obtained in 36%-96% yields and with moderate to excellent diastereoselectivities. In addition, the synthesis of (±)-norallosedamine 12 could be conveniently achieved from the cycloadduct 7bf.
