Abstract:A new ratiometric fluorescence strategy for sensitive and selective detection of cobalt ions (Co 2+ ) and topotecan (TOP) is proposed. The dual-probe system consists of red-emissive nitrogen and sulphur doped carbon dots (R-NS@CDs) and TOP. For Co 2+ detection, the fluorescence of R-NS@CDs at 680 nm is enhanced upon addition of Co 2+ , while complexed TOP emission peak at 545 nm remains constant. This enables ratiometric detection of Co 2+ over a range of 5.0–160.0 ng mL −1 . The mechanism of R-NS@CDs fluorescence enhancement by Co 2+ is elucidated using FTIR, fluorescence spectroscopy, zeta potential measurements, and TEM imaging. For the detection of TOP, a ratiometric probe system comprising R-NS@CDs and Co 2+ was utilized. TOP forms a complex with Co 2+ bounded to R-NS@CDs, quenching the R-NS@CDs-Co 2+ fluorescence and simultaneously the native TOP fluorescence is enhanced. This allows ratiometric quantification of TOP from 1.0-90.0 ng mL −1 . The method provides high selectivity and low detection limits of 1.51 ng mL −1 for Co 2+ and 0.37 ng mL −1 for TOP. Practical applicability is demonstrated through selective detection of Co 2+ in environmental water samples and TOP in real plasma samples. The built-in self-calibration enabled by dual analyte modulation of R-NS@CDs makes this a simple and powerful analytical approach.

A ratiometric fluorescent probe for sensitive determination of the important glycopeptide antibiotic vancomycin

Abstract:A novel sensitive and selective probe for the important antibiotic vancomycin (Van) has been synthesized by integrating a coumarin and a fluorescein as dual fluorescence reporters and a Van binding peptide D-Ala-D-Ala. Only weak green fluorescence was initially observed, which was mostly attributed to fluorescence self-quenching induced by fluorophore stacking. Upon the binding of Van with the D-Ala-D-Ala peptide, the fluorescence turned on, probably due the disaggregation of fluorophores. The intensity ratio of the dual emission bands I519/I446 exhibited an excellent linear relationship with the concentration of Van increasing from 0-20 μM in synthetic urine. The lowest detection limit was calculated to be 92.8 nM in urine, which made the probe applicable in clinically relevant concentration ranges. The synthetic probe has also shown the potential for Van detection in human serum. More interestingly, this probe has been successfully applied for in vivo imaging of Van in zebrafish. Abstract:Vancomycin is employed to treat infections caused by gram-positive bacteria. Ensuring precise vancomycin dosages is essential to avoid the emergence of bacterial resistance. In the current study, a fluorescent nanoprobe was designed for vancomycin determination in exhaled breath condensate samples. The nanoprobe was based on carbon dots (CDs) doped with nitrogen, sulfur, phosphorus, and chlorine (NSPCl-doped CDs). Vancomycin significantly reduced the fluorescence of NSPCl-doped CDs and presented a quenching process in the analytical response of the probe within a concentration range of 0.01-2.0 μg mL-1 due to forming a non-fluorescent complex. The nanoprobe's intra-day and inter-day relative standard deviations were 1.4 % and 3.2 %, respectively. This nanoprobe was successfully used to determine vancomycin in the patients receiving this drug collected from the expiratory circuit of the mechanical ventilator.

Herein, nitrogen-sulfur co-doped carbon dots (NSCDs) diluted to different concentrations, the high concentration of G-NSCDs (0.2 mg mL −1 ) with green fluorescence and the low-concentration of B-NSCDs (0.01 mg mL −1 ) with blue fluorescence were obtained. Further, it was found that the detection behaviors are different for different concentrations. The "turn-off" fluorescent sensor of G-NSCDs can specifically recognize rifampicin in a linear range of 0.2–20 μM with a detection limit of 56.6 nM employing the mutual effect of photo-induced electron transfer and dynamic quenching. Meanwhile, a "turn-off" B-NSCDs fluorescence sensor was constructed to effectively identify morin based on the internal filtration effect and static quenching, achieving a suitable linearity of 0.2–30 μM and a detection limit of 51.2 nM. Due to the strong chelation of morin and Al 3+ , a "turn-on" fluorescent probe for sequential detection Al 3+ was fabricated based on B-NSCDs-morin system, obtaining a good linearity of 0.1–2.0 μM with a satisfactory detection limit of 45.8 nM. These interesting behaviors indicate that NSCDs are expected to become novel sensing materials for the monitoring of rifampicin, morin and Al 3+ in organisms, which is beneficial for our health.

Abstract:Abstract. Al-Karami MA, Al-Rufaie MM. 2022. Colorimetric estimation of vancomycin using modified silver nanoparticles and implementing it on various types of pharmaceutical perpetrations. Nusantara Bioscience 14: 70-77. The current research proposes a simple, quick, and sensitive method for determining the pure state of vancomycin (Van) and several of its medicinal formulations. The antibiotic (Van) is an effective reduction agent for diluting the ore mineral salt from silver nitrate (Ag+) to silver nanoparticles. Polyvinylpyrrolidone was used as a stabilizer in a medium using sodium hydroxide as a base. The highest absorption of the vancomycin oxidation-reduction product occurs at 440 nm. The calibration curve is measured, and the following data is generated, indicating that the Beer-Lambert Law is followed within the concentration range of 2.5-70 parts per million. Sandal's molar absorptivity was 2.0043 x 104 L/mol.cm, and its sensitivity was 0.0723 g/cm2. The correlation coefficient was 0.9987, and the standard deviation rate was 0.402%. The biological influence of several Gram-positive and Gram-negative bacteria was researched, and the efficacy of the samples prepared on these bacteria has shown to be promising.

Abstract:As novel fluorescent nanomaterials, carbon dots have attracted increasing research attention because of their simple synthesis, robust fluorescence, low toxicity, and easy functionalisation. Previous research was focused on preparing carbon dots from biomass and chemical materials; however, most of these carbon dots exhibited blue fluorescence. Moreover, the fluorescence quantum yield was generally low, significantly limiting their application in biological imaging. To broaden the application scope of carbon dots, this study prepared long-wavelength emitting nano-carbon dots that exhibited increased quantum yield. Novel N-doped yellow fluorescent nano-carbon dots (Y-CDs) were synthesised via a hydrothermal method using L-tartaric acid and urea as the precursors. The Y-CDs had a high quantum yield (15.9%) and demonstrated photostability at various pHs, temperatures, and ionic strengths. The Y-CDs could detect cobalamin effectively and selectively, showing a linear relationship between fluorescence intensity and cobalamin concentration. The related coefficient was 0.997, and the detection limit was 2.101 μmol/L. In addition, the Y-CDs were successfully used as an imaging probe for MDA-MB-231 cells. Therefore, the Y-CDs developed in this study can be used for cobalamin detection and cell imaging.

Abstract:A novel ratiometric fluorescence probe was developed for the determination of azithromycin (AZM) and sulfide ions based on the differential modulation of red emissive carbon dots (R-N@CDs) and blue emissive carbon dots (B-NS@CDs). The addition of sulfide anion selectively quenched the red emission of R-N@CDs while the blue emission of B-NS@CDs unaffected. Upon subsequent introduction of AZM to this R-N@CDs@sulfide system, the quenched red fluorescence was restored. Comprehensive characterization of the CDs was performed using UV-Vis, fluorescence, FTIR spectroscopy, XPS, and TEM. The proposed method exhibited excellent sensitivity and selectivity, with limits of detection of 0.33 μM for AZM and 0.21 μM for sulfide. Notably, this approach enabled direct detection of sulfide without requiring prior modulation of the CDs with metal ions, as is common in other reported methods. The ratiometric probe was successfully applied for the determination of AZM in biological fluids and sulfide in environmental water samples with high selectivity. This work presents the first fluorometric method for the detection of AZM in biological fluids.

Abstract:Vancomycin is the first-line drug for infections of methicillin-resistant Staphylococcus aureus (MRSA) and multi-drug-resistant bacteria. The effective therapeutic concentration range of vancomycin is narrow, so it's essential to implement vancomycin therapeutic drug monitoring. However, conventional detection methods have disadvantages of expensive equipment, complicated operation, or poor reproducibility. Herein, a fluorescent sensing platform initiated by an allosteric probe was constructed for simple and sensitive monitoring of vancomycin at a low cost. The key point of this platform is the well-designed allosteric probe, which comprises an aptamer and a trigger sequence. When vancomycin exists, the combination of vancomycin and the aptamer will lead to a conformational change of the allosteric probe, thus exposing the trigger sequence. The trigger can react with the molecular beacon (MB) to generate fluorescent signals. In addition, the allosteric probe combined with hybridization chain reaction (HCR) was applied to develop an amplified platform, the linear range is from 0.5 μg mL −1 to 50 μg mL −1 with the limit of detection (LOD) of 0.26 μg mL −1 . Most importantly, this allosteric probe-initiated sensing platform shows good detection ability in human serum samples, and it also indicates great correlation and accuracy compared with HPLC. The present simple and sensitive allosteric probe-initiated platform has the potential to support the therapeutic drug monitoring of vancomycin, which is of great significance to promote the rational use of antibiotics in clinics.

Microwave-assisted Synthesis of Highly Luminescent N- and S-co-doped Carbon Dots As a Ratiometric Fluorescent Probe for Levofloxacin

Abstract:Uniform N- and S-co-doped carbon dots (NSCDs) with fluorescence quantum yields of up to 64% were synthesized via a one-step microwave-assisted method. Ammonium citrate and L-cysteine act as precursors, and synthesis is completed in 2.5 min using a 750 W microwave oven to give a 62% yield. The NSCDs show bright blue fluorescence (with excitation/emission peaks at 353/426 nm) and have narrow size distribution. On exposure to levofloxacin (LEV), the emission maximum shifts to 499 nm. This effect was used to design ratiometric (2-wavelength) assays for LEV. The fluorometric method (based on measurement of the fluorescence intensity ratio at 499 and 426 nm) has a detection limit of 5.1 μg·L−1 (3σ/k) and a linear range that extends from 0.01 to 70 mg·L−1. The method was applied to the determination of LEV in three kinds of spiked water samples and has recoveries in the range from 98.6 to 106.8%. The fluorescent probe described here is highly selective and sensitive.

Abstract:The combination of fluorescent probe and colorimetric technique has become one of the most powerful analytical methods due to the advantages of visualization, minimal measurement errors and high sensitivity. Hence, a novel dual-modality sensing probe with both colorimetric and fluorescent capabilities was developed for detecting cobalt ions (Co2+) based on homocysteine mediated silver nanoparticles and rhodamine 6G derivatives probe (AgNPs-Hcy-Rh6G2). The fluorescence of the AgNPs-Hcy-Rh6G2 probe turned on due to the opening of the Rh6G2 spirolactam ring in the presence of Co2+ by a catalytic hydrolysis. The fluorescent intensity of probe is proportional to Co2+ concentration in the range of 0.10-50 μM with a detection limit of 0.05 μM (S/N = 3). More fascinatingly, the color of AgNPs-Hcy-Rh6G2 probe changed from colorless to pink with increasing Co2+ concentration, which allowing colorimetric determination of Co2+. The absorbance of AgNPs-Hcy-Rh6G2 probe is proportional to Co2+ concentration in the range from 0.10 to 25 μM with a detection limit of 0.04 μM (S/N = 3). This colorimetric and fluorescent dual-modal method exhibited good selectivity, and reproducibility and stability, holding great potential for real samples analysis in environmental and drug field.

Abstract:Abstract Background Carbon dots, CDs, have excellent photoluminescence properties, good biocompatibility, low toxicity and good light stability. The optical, magnetic and electronic properties of CDs make them a hugely relevant tool to be used in pharmaceutical analysis, bioimaging, drug delivery, and other fields. The fluorescence of carbon nanodots makes it suitable for assay of some nitrogenous compounds of high pharmaceutical interest. In this work, we develop simple, fast and green spectrophotometric methods for quantification of Azithromycin and Rasagiline mesilate using synthesized fluorescent CDs from garlic peels. Results The spectrometric methods depend on stoichiometric reactions of both drugs with fluorescent CDs. Carbon dots exhibit a declared absorption peak λ max at 238 nm and potent fluorimetric emission at λ em 528 nm, upon excitation at λ ex 376 nm. Drugs’ concentrations in ppm are efficiently calculated using Stern–Volmer Equation. Decrease in fluorescence (Δ F = F o − F ) and the F -ratio values are linearly correlated to molar concentration of each quencher (drug). A significant linear diminish in the dots’ measured absorbance and fluorimetric emission values was observed. Validation of all the developed methods was according to the ICH guidelines. Conclusions In a new way, this work successfully indicates, spectrometric methods for rapid detection of two non-fluorophoric nitrogenous compounds using potent carbon nanodots. Consequently, these green developed methods offer several benefits as simplicity, ease of quantification, accuracy and precision that encourage the application of the developed methods in routine analysis of Azithromycin and Rasagiline mesilate in quality control laboratories as analytical tool.
Sulfur and nitrogen co-doped carbon quantum dots as fluorescent probes for the determination of some pharmaceutically-important nitro compounds

Abstract:Abstract In this study, highly fluorescent sulfur and nitrogen co-doped carbon quantum dots (SN-CQDs) were synthesized by a simple one-pot hydrothermal method using thiosemicarbazide and citric acid as starting materials. Various spectroscopic and microscopic techniques were applied to characterize the prepared SN-CQDs. The synthesized SN-CQDs’ maximum fluorescence emission was obtained at 430 nm after excitation at 360 nm. Rifampicin (RFP), tinidazole (TNZ), ornidazole (ONZ), and metronidazole (MNZ) all quantitatively and selectively quenched the SN-CQDs’ native fluorescence, which was the base-for their-spectrofluorimetric estimation without the need for any tedious pre-treatment steps or high-cost instrumentation. SN-CQDs demonstrated a “turn-off” fluorescence response to RFP, TNZ, ONZ, and MNZ over the ranges of 1.0–30.0, 10.0–200.0, 6.0–200.0, and 5.0–100.0 μM with detection limits of 0.31, 1.76, 0.57, and 0.75 μM and quantitation limits of 0.93, 5.32, 1.74, and 2.28 μM respectively. The suggested method was successfully used to determine the investigated drugs in their commercial dosage forms. The method was further extended to their determination in spiked human plasma samples, with satisfactory mean % recoveries (99.44–100.29) and low % RSD values (< 4.52). The mechanism of fluorescence quenching was studied and discussed. The suggested method was validated in accordance with ICH recommendations.

Abstract:This study reports the successful synthesis of optically stable red-emitting carbon dots (R-CDs) through a solvothermal method, using glycine as the carbon source and o-phenylenediamine as the nitrogen-doping agent. The R-CDs exhibit long-wavelength emission characteristics with optimal excitation and emission wavelengths of 533 nm and 600 nm, respectively, and a quantum yield of 26.7 %. The results demonstrate that R-CDs possess excellent salt resistance and photostability. The R-CDs display bright fluorescence emission and show a sensitive response to ceftazidime (CF). Leveraging these properties, a fluorescent probe based on R-CDs was developed for the sensitive determination of CF. The fluorescence quenching intensity of this system exhibits a good linear relationship with CF concentration in the range of 0–0.7 mmol/L. The linear equation is ( F 0 - F )/ F 0 =0.9564 C CF (mmol/L)+0.0089, with a linear correlation coefficient ( R 2 ) of 0.9945. The detection limit is 4.9 μmol/L, with recovery rates ranging from 94.2 % to 100.5 % and relative standard deviations between 2.2 % and 3.2 %. This work provides a theoretical basis for the detection of CF using red-emitting carbon dots and demonstrates promising potential for practical applications.

Abstract:In this study, dual-emissive carbon dots (CDs) were prepared using p-phenylenediamine (pPDA) and phytic acid (PA) precursors via a one-pot-hydrothermal method. The photophysical, morphological, and structural characterization of CDs was carried out using absorption, fluorescence, Fourier transform infrared (FT-IR) spectroscopy, nuclear magnetic resonance (NMR), and high-resolution transmission electron microscopy (HR-TEM) analysis. The as-prepared CDs displayed dual-fluorescence peaks at 525 and 620 nm upon excitation at 450 nm. The CDs showed good photostability and exhibited solvent-dependent fluorescence properties. The solvatochromic behavior of CDs was utilized to detect water content in organic solvents. Furthermore, the dual-emissive property of CDs was utilized for the sequential detection of ferric (Fe3+) and pyrophosphate ions (PPi) by a fluorescence turn-off-on mechanism. The proposed assay showed appreciable fluorescence response toward Fe3+ and PPi with high selectivity and good tolerance for common interfering ions. The potential practical application of the CD probe was ascertained by carrying out the fluorometric detection of PPi to affirm the loop-mediated isothermal amplification (LAMP) reaction specific for Mycobacterium tuberculosis (negative and positive clinical samples).

Abstract:Background: Carbon quantum dots (CQDs) have gained much interest recently for being efficient probes. Their cost-effectiveness, eco-friendliness, and unique photocatalytic activities made them distinctive alternatives to other luminescent approaches like fluorescent dyes and luminous derivatization. Meanwhile, delafloxacin (DLF) is a recently approved antibacterial medicine. DLF has been authorized for the treatment of soft-tissue and skin infections as well as pneumonia. Therefore, new eco-friendly, cost-effective, and sensitive tools are needed its estimation in different matrices. Results: In the proposed study, green copper and nitrogen carbon dots (Cu-N@CDs) were synthesized from a green source (plum juice with copper sulphate). Cu-N@CQDs were then characterized using multiple tools including X-ray photon spectroscopy (XPS), FTIR and UV-VIS spectroscopy, Zeta potential measurements, High-resolution transmission electron microscopy (HRTEM), and fluorescence spectroscopy. After gradually adding DLF, the developed quantum dots' fluorescence was significantly enhanced within the working range of 0.5-100.0 ng mL-1. The limits of detection and quantification were 0.08 and 0.27 ng mL-1, respectively. The accuracy of the proposed method ranged from 96.00 to 99.12 % in recovery%, when recovered from milk and plasma samples. Significance: Cu-N@CDs were utilized and validated for selectively determining DLF in several matrices including pharmaceutical forms, human plasma and in milk samples using spectrofluorimetric technique. The bio-analytical method is simple and could be used in content uniformity testing as well as in therapeutic drug monitoring in human plasma.

Abstract:Ciprofloxacin (CIP) plays an important role in many physiological processes. Previously reported methods for CIP detection have many drawbacks and so it is crucial to develop highly sensitive methods to detect CIP. In this study, a dual-emission ratiometric fluorescent probe (SF-CQDs/CuAg NCs) of a nanocomposite of carbon quantum dots (SF-CQDs) and copper-silver bimetallic nanoclusters (CuAg NCs) was developed for sensitive and selective detection of CIP. The fluorescent signal of CuAg NCs was quenched while the fluorescence intensity of SF-CQDs was enhanced in the presence of CIP. The working principle of the fluorescent probe SF-CQDs/CuAg NCs is mainly based on the complexation of CIP with CuAg NCs to free SF-CQDs. The quenching of CuAg NCs by CIP arises from the chelation between the metal and the 1-oxo substituent and the adjacent carboxyl group of CIP. The fluorescence recovery of SF-CQDs is attributing to the release of SF-CQDs from SF-CQDs/CuAg NCs and the hydrogen bonding interaction between SF-CQDs and CIP. The ratiometric fluorescence measurement of SF-CQDs/CuAg NCs has been employed to analyze CIP with good sensitivity and selectivity. The working range of CIP is 5.0–200.0 μM and the detection limit is 3.3 μM. More importantly, the fluorescence of SF-CQDs/CuAg NCs can be detected intuitively via a smartphone which can be applied to determine CIP.

Abstract:A pathogenic bacterium has its own mechanisms for not only pathogenic attack but also exogenous invasion defense, in which the bacterial cell wall is the front line of attack and defense. We developed a biochemical lanthanide-encoding approach to quantify the uncanonical d-amino acid (d-X) that was edited in a small proportion into the terminal acyl-d-Ala-d-X of nascent peptidoglycan UDP-MurNAc-pentapeptides in the bacterial cell wall. This approach overcomes the difficulties regarding quantification and accuracy issues encountered by the popular optical imaging and traditional high-performance liquid chromatography-based methods. Newly synthesized azide-d-Leu and ketone-d-Met were used together with alkynyl-d-Ala for their metabolic assembly and then bioorthogonally encoded by the correspondingly fabricated DBCO-DOTA-Gd, H2NO-DOTA-Eu, and azide-DOTA-Sm tags. This approach allows direct quantification of the d-X in situ in the cell wall using 158Gd, 153Eu, and 154Sm species-unspecific isotope dilution inductively coupled plasma mass spectrometry, avoiding any tedious and complex "cell-broken" pretreatment procedures that might induce racemization of the d-X. The obtained site-specific and accurate in situ information about the d-X enables quantitative monitoring of the bacterial response when Staphylococcus aureus meets vancomycin, showing that the amounts of azide-d-Leu and ketone-d-Met assembled are more important after determining the structure- and composition-dependent bacterial antibiotic resistance mechanisms. In addition, we found that the combined use of vancomycin and d-Ala restores the efficacy of vancomycin and might be a wise and simple way to combat vancomycin intermediate-resistant S. aureus.

Abstract:This work synthesized the ethylenediamine-doped puffer fish skin carbon dots using biomass as a green precursor. Three tetracycline antibiotics (TCs) were detected fluorescence and colorimetrically using carbon dots as probes; on this basis, hydrogels were used to carry out visual identification and quantification. In the presence of TCs, the nitrogen-doped carbon dots (N-CDs) displayed a quenching impact on fluorescence because of fluorescence resonance energy transfer (FRET), dynamic quenching, and band gap transition. As a result, it was possible to identify the precise concentrations of tetracycline (TC), oxytetracycline (OTC), and chlortetracycline (CTC). The assay has great biocompatibility, which was supported by cytotoxicity, a wide linear range (0.5–500 M), a low detection limit (0.15 M), and outstanding selectivity. With a high recovery rate, the approach has been successfully used to identify TCs in food and water samples such milk, honey, and tap water.
