Abstract:Chemodynamic therapy (CDT), which induces cell death by decomposing high levels of H2O2 in tumor cells into highly toxic ·OH, is recognized as a promising antineoplastic approach. However, current CDT approaches are often restricted by the highly controlled and upregulated cellular antioxidant defense. To enhance ·OH-induced cellular damage by CDT, a covalent organic framework (COF)-based, ferrocene (Fc)- and glutathione peroxidase 4 (GPX4) inhibitor-loaded nanodrug, RSL3@COF–Fc (2b), is fabricated. The obtained 2b not only promotes in situ Fenton-like reactions to trigger ·OH production in cells, but also attenuates the repair mechanisms under oxidative stress via irreversible covalent GPX4 inhibition. As a result, these two approaches synergistically result in massive lipid peroxide accumulation, subsequent cell damage, and ultimately ferroptosis, while not being limited by intracellular glutathione. It is believed that this research provides a paradigm for enhancing reactive oxygen species-mediated oncotherapy through redox dyshomeostasis and may provide new insights for developing COF-based nanomedicine.

Abstract:Metal‐organic framework (MOF)‐based drug delivery nanomaterials for cancer therapy have attracted increasing attention in recent years. Here, we propose an enhanced chemodynamic anti‐tumor therapy strategy by promoting the Fenton reaction by using core‐shell zeolitic imidazolate framework‐8 (ZIF‐8)@Fe3O4 as a therapeutic platform. Carboxymethyl cellulose (CMC) was used as a stabilizer of Fe3O4, which then decorated on the surface of ZIF‐8 via the electrostatic interaction and served as an efficient Fenton reaction trigger. Meanwhile, the pH‐responsive ZIF‐8 scaffold acted as a container to encapsulate the chemotherapeutic drug doxorubicin (DOX). The obtained DOX‐ZIF‐8@Fe3O4/CMC nanoparticles concomitantly accelerated DOX release and generated more hydroxyl radicals by targeting the lysosomes in cancer cells. In vitro and in vivo studies verify that the DZFC nanoparticles trigger GPX4‐dependent ferroptosis via the activation of the c‐Jun N‐terminal kinases (JNK) signaling pathway, following to achieve the chemo/ferroptosis synergistic anti‐tumor efficacy. No marked toxic effects were detected during DZFC treatment in a tumor‐bearing mouse model. This composite nanoparticle remarkably suppressed the tumor growth with minimized systemic toxicity, opening new horizons for the next generation of theragnostic nanomedicines. Abstract:Developing agents that can accurately differentiate tumors from normal healthy tissues is of utmost importance for safe cancer therapy. Active targeting has been considered as an effective technique for tumor recognition. In this work, we demonstrate a folate-functionalized nanoscale covalent organic framework (FATD nCOF) highly specific to cancer cells through active targeting of their enriched folate receptors (FRs). The FATD nCOF prepared by simple post-synthetic modification of the COF surface defeats disperses well in water and exhibits a high loading capacity for various anticancer drugs. The biocompatible FATD nCOF is selectively internalized by FR-harboring cancer cells and consequently augments the efficacy of the loaded drug, Withaferin A (Wi-A), for targeted cancer cell killing. In biomolecular mechanism studies, Wi-A-loaded FATD (FATD@Wi-A) nanocomposites show remarkably a higher rate of apoptosis in FR-enriched cancer cells. Comparative analyses of FR-positive and FR-negative tumor xenografts reveal enhanced selective antitumor activity of FATD@Wi-A nanotherapeutics. Taken together, the study findings suggest that FATD nCOF holds great promise for active targeting of tumors in vivo . Our simple yet effective technology might be valuable for creating new state-of-the-art COFs for chemical and biomedical applications.

Abstract:Drug resistance is a major challenge in antineoplastic chemotherapy, which could be possibly solved through multifunctional anti-cancer therapy. This study constructs a drug delivery system with Zn(II)-Co(II) bimetal-organic framework nanoparticles with encapsulated doxorubicin (DOX), a model chemotherapeutic molecule. Beyond conventional therapeutic mechanisms powered by DOX, the newly applied Co(II) to the drug delivery system catalyzes the generation of reactive oxygen species (ROS) through a Fenton-like process, leading to necrosis due to oxidative stress beyond threshold. The system displayed DOX-based and Co(II)-based dual mechanism anti-cancer therapy to overcome DOX-resistance while demonstrating significant cytotoxicity promotion in non-resistant cell lines, as effects of ROS were further acknowledged by following cell characteristic examinations. This bifunctional DOX-encapsulated bimetal-organic framework nanoparticle opens a new strategy to construct multiple anti-cancer mechanism drug delivery systems with great simplicity.

Abstract:The precise release of drugs is essential to improve cancer therapeutic efficacy. In this work, a tandem responsive strategy was developed based on a triple-layered metal-organic framework (MOF) hybrid. The MOF nanoprobe was stepwise fabricated with a telomerase-responsive inner, a pH-sensitive MOF filling and H2O2-responsive coordination complex shell of Fe3+ and eigallocatechin gallate (EGCG). In the tumor microenvironment, the shell was dissociated by endogenous H2O2 and simultaneously produced highly reactive hydroxyl radicals by a Fenton reaction. Meanwhile, the released EGCG could downregulate the expression of P-glycoprotein responsible for drug resistance. After the dissociation of the framework by protons, telomerase could trigger the release of the drug from the DNA duplex on the exposed inner shell. By integrating confined drug release, inhibited efflux pump and chemodynamic therapy, the all-in-one chemotherapy strategy was identified with enhanced therapeutic efficacy in drug-resistant cancer cells.

Abstract:Covalent organic framework (COF) receives great attention in biomedical applications due to its variable compositions and ordered structures. However, its targeted design to achieve desirable physiological functions especially for cancer treatments remains elusive. Herein, PEGylated COF with tumor-specific TKD peptide modification is uniformly coated on photothermal mesoporous carbon nanospheres via polyethyleneimine-mediated interface polymerization to construct a multifunctional core-shell nanoparticle (OPCPT). Physicochemical studies demonstrate near infrared (NIR)-blocking ability of the crystalline COF shells under physiological conditions, whereas COF is degraded under the acidic tumor microenvironments (TME). Subsequently, the nanoparticle charge is reversed and the COF monomers can produce O-1(2)/O-2. As a result, OPCPT, activated in the TME due to the shell dissociation, penetrates deeply into tumors through positive charge-mediated/lysosome rupture-mediated transcytosis and recovers its NIR-heating potential for tumor-specific photothermal therapy. Moreover, the TME-triggered O-1(2) significantly depresses the lysosome autophagy via membrane destruction, and selectively damages the mitochondria to promote the cytochrome C release-activated apoptosis and ATP deficiency-inhibited tumor metastasis. Particularly, this unique O-2 generation mechanism relieves the tumor hypoxia upon the reactive oxygen species therapy and downregulates hypoxia-inducible factor and its downstream proteins, which all contribute to augmented tumor therapy. The findings represent a remarkable unveiling of the potential of COF-based nanomaterials for extended biomedical applications.

Abstract:Due to the limited concentration of hydrogen peroxide (H 2 O 2 ) in the tumor microenvironment, peroxidase (POD)-mimicking nanozyme-mediated catalytic therapy usually cannot completely eliminate tumor issues. In this study, we report an H 2 O 2 -responsive nanozyme constructed by loading 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) into POD-like Fe-porphyrin covalent organic frameworks (COFs) (Fe-DhaTph). The resulting ABTS@Fe-DhaTph can function as a multifunctional therapeutic nanoagent for combination therapy of catalytic therapy, photothermal therapy (PTT), and photodynamic therapy (PDT). In the tumor microenvironment, ABTS@Fe-DhaTph can catalyze the decomposition of endogenous H 2 O 2 to produce highly cytotoxic hydroxyl radicals (·OH) for in situ tumor catalytic therapy. Meanwhile, the loaded ABTS is oxidized by H 2 O 2 with the assistance of ABTS@Fe-DhaTph, and the resulting oxABTS, with strong near-infrared absorbance, can enable the H 2 O 2 -dependent PTT. Besides Fe-DhaTph-induced PDT, the generated oxABTS with glutathione (GSH)-depletion ability can synergistically enhance the generation of reactive oxygen species, thereby improving catalytic therapy and PDT. To the best of our knowledge, ABTS@Fe-DhaTph is the first COF-based nanozyme to serve as a cascade-amplified synergistic therapeutic nanoagent for cancer treatment.

Abstract:Chemodrug resistance is a major reason accounting for tumor recurrence. Given the mechanistic complexity of chemodrug resistance, molecular inhibitors and targeting drugs often fail to eliminate drug-resistant cancer cells, and sometimes even promote chemoresistance by activating alternative pathways. Here, by exploiting biochemical fragility of high-level but dynamically balanced cellular redox homeostasis in drug-resistant cancer cells, we design a nanosized copper/catechol-based metal-organic framework (CuHPT) that effectively disturbs this homeostasis tilting the balance toward oxidative stress. Within drug-resistant cells, CuHPT starts disassembly that is triggered by persistent consumption of cellular glutathione (GSH). CuHPT disassembly simultaneously releases two structural elements: catechol ligands and reductive copper ions (Cu+). Both of them cooperatively function to amplify the production of intracellular radical oxidative species (ROS) via auto-oxidation and Fenton-like reactions through exhausting GSH. By drastically heightening cellular oxidative stress, CuHPT exhibits selective and potent cytotoxicity to multiple drug-resistant cancer cells. Importantly, CuHPT effectively inhibits in vivo drug-resistant tumor growth and doubles the survival time of tumor-bearing mice. Thus, along with CuHPT's good biocompatibility, our biochemical, cell biological, preclinical animal model data provide compelling evidence supporting the notion that this copper-based MOF is a predesigned smart therapeutic against drug-resistant cancers through precisely deconstructing their redox homeostasis.

Abstract:The development of stimuli-responsively degradable porous carriers for both controlled drug release and high biosafety is vitally important to their clinical translation, but still challenging at present. A new type of porphyrin-iron metal organic framework (Fe-MOF) nanocrystals is engineered here as acid-degradable drug carrier and hydrogen donor by the coordination between porphyrin and zero-valence Fe atom. Fe-MOF nanocrystals exhibit excellent acid-responsive degradation for H2 generation and simultaneous release of the loaded drug for combined hydrogen-chemotherapy of cancer multidrug resistance (MDR) and metastasis and for local hydrogen eradication of the off-target induced toxic side effects of the drug to normal cells/tissues. Mechanistically, released H2 assists chemotherapeutic drug to efficiently inhibit cancer metastasis by immunoactivating intratumoral M1-phenotype macrophages and consequently downregulating the expression of metastasis-related matrix metalloproteinase-2 (MMP-2) and can also downregulate the expressions of both P-glycoprotein (P-gp) protein and adenosine triphosphate (ATP) in MDR cancer cells to sensitize chemotherapeutic drug for enhanced damage to mitochondria and DNA. High anti-MDR/antimetastasis efficacies and high biocompatibility endow Fe-MOF nanocrystals and the Fe-MOF-based nanomedicine with high potential for clinical translation.

Tumor microenvironments self-activated nanoscale metal-organic frameworks for ferroptosis based cancer chemodynamic/photothermal/chemo therapy

Abstract:Ferroptosis, as a newly discovered cell death form, has become an attractive target for precision cancer therapy. Several ferroptosis therapy strategies based on nanotechnology have been reported by either increasing intracellular iron levels or by inhibition of glutathione (GSH)-dependent lipid hydroperoxidase glutathione peroxidase 4 (GPX4). However, the strategy by simultaneous iron delivery and GPX4 inhibition has rarely been reported. Herein, novel tumor microenvironments (TME)-activated metal-organic frameworks involving Fe & Cu ions bridged by disulfide bonds with PEGylation (FCSP MOFs) were developed, which would be degraded specifically under the redox TME, simultaneously achieving GSH-depletion induced GPX4 inactivation and releasing Fe ions to produce ROS via Fenton reaction, therefore causing ferroptosis. More ROS could be generated by the acceleration of Fenton reaction due to the released Cu ions and the intrinsic photothermal capability of FCSP MOFs. The overexpressed GSH and H2O2 in TME could ensure the specific TME self-activated therapy. Better tumor therapeutic efficiency could be achieved by doxorubicin (DOX) loading since it can not only cause apoptosis, but also indirectly produce H2O2 to amplify Fenton reaction. Remarkable anti-tumor effect of obtained FCSP@DOX MOFs was verified via both in vitro and in vivo assays.
Dual-responsive metal organic framework for electrically-enhanced cascade catalytic tumor therapy

Abstract:Chemodynamic Therapy (CDT) based on Fenton reaction that can produce highly toxic reactive oxygen species to kill cancer cells has attracted considered attention. However, the effect of the CDT is limited in specific conditions (e.g. pH and H 2 O 2 content) in tumor microenvironment (TME). In this paper, a glucose oxidase (GOx) and Fe 3+ -loaded metal organic framework system (@Fe 3+ ), to enhance the cascade reaction of the therapy, was developed for combined starvation therapy and CDT with the assistance of electrolysis. The @Fe 3+ in the tumor microenvironment was decomposed and thus released the Fe 3+ and GOx. The electrolysis procedure, can not only trigger the Fe 3+ and Fe 2+ cyclic reactions, also provided O 2 for the oxidation and decomposition of glucose to generate H 2 O 2 . This resulted in the improvement of the pH in TME for Fenton reaction, to enhance the CDT treatment effect. Meanwhile, glucose and O 2 were consumed by GOx to achieve the effect of blocking nutrients. Compared with other control groups, the @Fe 3+ under electrolysis showed the considerable inhibitory effect of tumor cells, and the apoptosis rate even reached 80%. Our work presents an ingenious nanoenzyme for Fenton reactions that achieves a self-supplying strategy under electrolysis to improve anti-tumor efficacy.

Abstract:Efforts have been made to improve the therapeutic efficiency of tumor treatments, and metal–organic frameworks (MOFs) have shown excellent potential in tumor therapy. Monotherapy for the treatment of tumors has limited effects due to the limitation of response conditions and inevitable multidrug resistance, which seriously affect the clinical therapeutic effect. In this study, we chose to construct a multiple cascade synergistic tumor drug delivery system MIL−101(Fe)−DOX−TCPP−MnO2@PDA−Ag (MDTM@P−Ag) using MOFs as drug carriers. Under near−infrared (NIR) laser irradiation, 5,10,15,20−tetrakis(4−carboxyphenyl)porphyrin (TCPP) and Ag NPs loaded on MDTM@P−Ag can be activated to generate cytotoxic reactive oxygen species (ROS) and achieve photothermal conversion, thus effectively inducing the apoptosis of tumor cells and achieving a combined photodynamic/photothermal therapy. Once released at the tumor site, manganese dioxide (MnO2) can catalyze the decomposition of hydrogen peroxide (H2O2) in the acidic microenvironment of the tumor to generate oxygen (O2) and alleviate the hypoxic environment of the tumor. Fe3+/Mn2+ will mediate a Fenton/Fenton−like reaction to generate cytotoxic hydroxyl radicals (·OH), while depleting the high concentration of glutathione (GSH) in the tumor, thus enhancing the chemodynamic therapeutic effect. The successful preparation of the tumor drug delivery system and its good synergistic chemodynamic/photodynamic/photothermal therapeutic effect in tumor treatment can be demonstrated by the experimental results of material characterization, performance testing and in vitro experiments.

Abstract:Nanomaterials with intrinsic enzyme-like activities (nanozymes) have gained significant attention in cancer catalytic therapy; however, developing metal-free nanozymes with multivariant enzyme-like activity as the "all-rounder" for cancer therapy remains challenging. Herein, a covalent organic framework (COF) derived carbon-based nanozyme is rationally devised to achieve synergistic catalytic therapy and second near-infrared (NIR-II) photothermal therapy of cancer. The developed nanozyme possesses multivariant enzyme-like activities, including oxidase (OXD)-like, catalase (CAT)-like, and peroxidase (POD)-like catalytic activities, which enables the nanozyme to produce adequate reactive oxygen species (ROS) for cancer cell killing. Furthermore, the nanozyme showed excellent photothermal converting activity that could kill cancer cells upon NIR-II laser irradiation, owing to the strong NIR-II absorption capacity of carbon-based materials. It is also worth noting that the nanozyme exhibited cytotoxicity specifically in tumor tissue profiting from the discrepant H2O2 level between tumor and normal tissue and the spatiotemporal controllability of laser irradiation. This work may inspire further development of intelligent nanozymes in biological applications across broad therapeutic and biomedical fields.

Abstract:Development of ferroptosis-inducible nanoplatforms with high efficiency and specificity is highly needed and challenging in tumor ferrotherapy. Here, we demonstrate highly effective tumor ferrotherapy using iron (II)-based metal-organic framework (Fe ss MOF) nanoparticles, assembled from disulfide bonds and ferrous ions. The as-prepared Fe ss MOF nanoparticles exhibit peroxidase-like activity and pH/glutathione-dependent degradability, which enables tumor-responsive catalytic therapy and glutathione depletion by the thiol/disulfide exchange to suppress glutathione peroxidase 4, respectively. Upon PEGylation and Actinomycin D (ActD) loading, the resulting Fe ss MOF/ActD-PEG nanoplatform induces marked DNA damage and lipid peroxidation. Concurrently, we found that ActD can inhibit Xc − system and elicit ferritinophagy, which further boosts the ferrotherapeutic efficacy of the Fe ss MOF/ActD-PEG. In vivo experiments demonstrate that our fabricated nanoplatform presents excellent biocompatibility and a high tumor inhibition rate of 91.89%.

A Catalase‐Like Metal‐Organic Framework Nanohybrid for O 2 ‐evolving Synergistic Chemoradiotherapy

Abstract:Tumor hypoxia, the "Achilles' heel" of current cancer therapies, is indispensable to drug resistance and poor therapeutic outcomes especially for radiotherapy. Here we propose an in situ catalytic oxygenation strategy in tumor using porphyrinic metal-organic framework (MOF)-gold nanoparticles (AuNPs) nanohybrid as a therapeutic platform to achieve O2 -evolving chemoradiotherapy. The AuNPs decorated on the surface of MOF effectively stabilize the nanocomposite and serve as radiosensitizers, whereas the MOF scaffold acts as a container to encapsulate chemotherapeutic drug doxorubicin. In vitro and in vivo studies verify that the catalase-like nanohybrid significantly enhances the radiotherapy effect, alleviating tumor hypoxia and achieving synergistic anticancer efficacy. This hybrid nanomaterial remarkably suppresses the tumor growth with minimized systemic toxicity, opening new horizons for the next generation of theranostic nanomedicines.
Tumor-targeted supramolecular catalytic nanoreactor for synergistic chemo/chemodynamic therapy via oxidative stress amplification and cascaded Fenton reaction

Abstract:Utilizing intratumoral iron mediated Fenton reaction to induce cancer cells destruction, chemodynamic therapy (CDT) is an emerging tumor-specific therapeutic strategy. However, the CDT efficacy is severely restrained by insufficient endogenous H2O2 and catalytic ferrous ions in tumor site. Herein, based on innovative small molecule prodrug design concept, an acid-labile ferrocene-modified cinnamaldehyde prodrug (Fc-CA) was first synthesized. Skillfully combining ROS-generator CA with Fenton catalyst Fc in its molecular structure, Fc-CA could synchronously elevate H2O2 level and catalytic iron amount for enhanced CDT efficacy. Then, hydrophobic Fc-CA was efficiently encapsulated into hydrophilic cyclodextrin-modified hyaluronic acid conjugate (HA-CD) via host-guest interactions, to spontaneously form pH/redox dual-responsive supramolecular catalytic nanoreactor (HA-CD/Fc-CA NPs). The nanoreactor with high drug loading and stability, excellent catalytic activity and tumor specificity could preferentially accumulate in tumor tissues via EPR effect and CD44 receptor-mediated internalization. Through the cleavage of acid-labile hydrazone bond under acidic lysosome environments, the nanoreactor would be dissociated and release pre-protected CA for reinforced oxidation therapy and amplified H2O2 level, which would in turn react with Fc through cascaded Fenton reaction, to generate highly cytotoxic •OH for enhanced cancer killing in a positive feedback process. Both in vitro and in vivo results verified excellent cascade-amplifying therapeutic performance and favorable biocompatibility of this nanoreactor. This study provides an innovative strategy for exploiting versatile organic therapeutic nanoplatforms, simultaneously activating tumor-specific oxidative stress amplification and cascaded enhanced Fenton reaction for superior cancer therapy with excellent biosafety.
